Abstract:
ABSTRACT We analyzed cryptophyte nucleomorph 18S rRNA gene sequences retained in natural Myrionecta rubra cells and plastid 16S rRNA gene and psbA sequences retained in natural cells of several Dinophysis species collected from Japanese coastal waters. A total of 715 nucleomorph sequences obtained from 134 M. rubra cells and 564 plastid 16S rRNA gene and 355 psbA sequences from 71 Dinophysis cells were determined. Almost all sequences in M. rubra and Dinophysis spp. were identical to those of Teleaulax amphioxeia, suggesting that M. rubra in Japanese coastal waters preferentially ingest T. amphioxeia. The remaining sequences were closely related to those of Geminigera cryophila and Teleaulax acuta. Interestingly, 37 plastid 16S rRNA gene sequences, which were different from T. amphioxeia and amplified from Dinophysis acuminata and Dinophysis norvegica cells, were identical to the sequence of a D. acuminata cell found in the Greenland Sea, suggesting that a widely distributed and unknown cryptophyte species is also preyed upon by M. rubra and subsequently sequestered by Dinophysis. To confirm the reliability of molecular identification of the cryptophyte Teleaulax species detected from M. rubra and Dinophysis cells, the nucleomorph and plastid genes of Teleaulax species isolated from seawaters were also analyzed. Of 19 isolates, 16 and 3 clonal strains were identified as T. amphioxeia and T. acuta, respectively, and no sequence variation was confirmed within species. T. amphioxeia is probably the primary source of prey for M. rubra in Japanese coastal waters. An unknown cryptophyte may serve as an additional source, depending on localities and seasons.

Protist–alga symbiosis is widespread in the ocean, but its characteristics and function in situ remain largely unexplored. Here we report the symbiosis of the ciliate Mesodinium rubrum with cryptophyte cells during a red-tide bloom in Long Island Sound. In contrast to the current notion that Mesodinium retains cryptophyte chloroplasts or organelles, our multiapproach analyses reveal that in this bloom the endosymbiotic Teleaulax amphioxeia cells were intact and expressing genes of membrane transporters, nucleus-to-cytoplasm RNA transporters, and all major metabolic pathways. Among the most highly expressed were ammonium transporters in both organisms, indicating cooperative acquisition of ammonium as a major N nutrient, and genes for photosynthesis and cell division in the cryptophyte, showing active population proliferation of the endosymbiont. We posit this as a “Mesodinium-farming-Teleaulax” relationship, a model of protist–alga symbiosis worth further investigation by metatranscriptomic technology.

Abstract:
Kleptoplastic mixotrophic species of the genus Dinophysis are cultured by feeding with the ciliate Mesodinium rubrum, itself a kleptoplastic mixotroph, that in turn feeds on cryptophytes of the Teleaulax/Plagioselmis/Geminigera (TPG) clade. Optimal culture media for phototrophic growth of D. acuminata and D. acuta from the Galician Rías (northwest Spain) and culture media and cryptophyte prey for M. rubrum from Huelva (southwest Spain) used to feed Dinophysis, were investigated. Phototrophic growth rates and yields were maximal when D. acuminata and D. acuta were grown in ammonia-containing K(-Si) medium versus f/2(-Si) or L1(-Si) media. Dinophysis acuminata cultures were scaled up to 18 L in a photobioreactor. Large differences in cell toxin quota were observed in the same Dinophysis strains under different experimental conditions. Yields and duration of exponential growth were maximal for M. rubrum from Huelva when fed Teleaulax amphioxeia from the same region, versus T. amphioxeia from the Galician Rías or T. minuta and Plagioselmis prolonga. Limitations for mass cultivation of northern Dinophysis strains with southern M. rubrum were overcome using more favorable (1:20) Dinophysis: Mesodinium ratios. These subtleties highlight the ciliate strain-specific response to prey and its importance to mass production of M. rubrum and Dinophysis cultures.

Growing evidence suggests that sexual reproduction might be common in unicellular organisms, but observations are sparse. Limited knowledge of sexual reproduction constrains understanding of protist ecology. Although Teleaulax amphioxeia and Plagioselmis prolonga are common marine cryptophytes worldwide, and are also important plastid donors for some kleptoplastic ciliates and dinoflagellates, the ecology and development of these protists are poorly known. We demonstrate that P. prolonga is the haploid form of the diploid T. amphioxeia and describe the seasonal dynamics of these two life stages. The diploid T. amphioxeia dominates during periods of high dissolved inorganic nitrogen (DIN) and low irradiance, temperature, and grazing (winter and early spring), whereas the haploid P. prolonga becomes more abundant during the summer, when DIN is low and irradiance, temperature, and grazing are high. Dimorphic sexual life cycles might explain the success of this species by fostering high genetic diversity and enabling endurance in adverse conditions.

Abstract:
Mixoplankton are protist plankton that are capable of phototrophy and phagotrophy. These organismsare increasingly recognised not just as freaks of nature, but as a substantial part of marineplankton. Most existing plankton models still assume a strict dichotomy between phototrophsand heterotrophs. Few models consider mixoplanktonic activity as a synergism of the two trophicmodes. Many different mixoplankton functional types exist on a gradient between heterotrophy andphototrophy. The cryptophyte (Teleaulax)-Mesodinium-Dinophysis (TMD) complex is a specificpredator-prey interaction of different types of mixoplankton and a good example of the complexityof mixoplankton interaction and trophodynamics. The specialist non-constitutive mixoplankton(SNCM) Mesodinium acquires its chloroplasts strictly from a specific constitutive mixoplankton(CM) cryptophyte, while the harmful algal bloom (HAB) species Dinophysis acquires its third-handchloroplasts exclusively from Mesodinium.The generic NPZ-style protist model developed here shows that mixoplankton displays dynamicsthat are distinctly different from strict heterotrophs and autotrophs in terms of growth and theway they shape their environment. In addition, there is a clear niche separation between differentmixoplankton types (general non-constitutive mixoplankton (GNCM), SNCM and CM) according tonutrient, prey and light resource availabilities indicating a niche separation of each type. Thus,considering the different mixoplankton functional types in specialised multi-organism relationshipsas they are found in the TMD-complex may be important for their understanding and accurateprediction of growth and biomass development. Currently, none of the many models of Dinophysiscapture the biological dependencies. Results from a nitrogen-based TMD model suggest thatthe timing and quantity of prey availability is crucial for the bloom dynamics of Mesodinium andDinophysis. Some CMs may only feed when phosphate is the limiting nutrient. The results ofthe variable stoichiometric “Perfect Beast” model that was configured as Teleaulax amphioxeia incombination with experimental data strongly suggest that the cryptophyte feeds on bacteria tocompensate for phosphate limitation.This work shows the importance of considering mixoplankton in ecosystem models alongsidestrict heterotrophs and autotrophs and that distinction between different mixoplankton functionaltypes matters. Mixoplankton distinctly differ in their nutrient utilisation and growth dynamics.Predator-prey interactions have different implications for mixoplankton than for heterotrophs andtheir inclusion in models could improve our understanding of the formation of harmful mixoplanktonblooms. The unique physiology of mixoplankton and their nutrient utilisation and trophic levelsneed consideration in species specific models.
Le mixoplancton inclut les protistes planctoniques capables de phototrophie et de phagotrophie.Ces organismes sont de plus en plus reconnus comme une partie importante du plancton marin.Toutefois, la plupart des modèles mathématiques planctoniques existants supposent encoreune stricte dichotomie entre les organismes phototrophes et hétérotrophes et peu de modèlesconsidèrent l’activité mixoplanctonique comme une synergie entre les deux modes trophiques.De nombreux types fonctionnels mixoplanctoniques différents existent dans un gradient entrel’hétérotrophie et la phototrophie. Le complexe cryptophyte (Teleaulax)-Mesodinium-Dinophysis(TMD) est une interaction prédateur-proie spécifique entre différents types de mixoplancton et unbon exemple de la complexité des interactions et des relations trophodynamiques du mixoplancton.Mesodinium, mixoplancton spécialiste non constitutif (SNCM), ne peut acquérir ses chloroplastesque de cryptophytes (mixoplancton constitutif (CM)) spécifiques (tel que Teleaulax), tandis quel’espèce Dinophysis, responsable d’efflorescences algales nuisibles, acquiert ses chloroplastesexclusivement de Mesodinium. Le modèle générique de protistes, de type NPZ, développé dansce travail montre que le mixoplancton présente une dynamique nettement différente de celle deshétérotrophes et autotrophes strictes en termes de croissance et de la façon dont ils façonnentleur environnement. En outre, il existe une séparation de niches claire entre les différents typesde mixoplancton (mixoplancton généraliste non-constitutif (GNCM), SNCM et CM) en fonction dela disponibilité en lumière, en nutriments et en proies. En conséquence, la prise en compte desdifférents types fonctionnels du mixoplancton dans des relations multi-organismes spécialisées,telles qu’on les trouve dans le complexe TMD, peut être importante pour leur compréhension et laprédiction précise de leur croissance et biomasse. Actuellement, aucun des modèles existants deDinophysis ne rend compte de ces dépendances biologiques. Les résultats d’un modèle TMD basésur l’azote suggèrent que le moment et la quantité de proies disponibles sont des facteurs cruciauxpour la dynamique de Mesodinium et de Dinophysis. Certains CM peuvent se nourrir uniquementlorsque le phosphate est le nutriment limitant. Les résultats du modèle à stoechiométrie variable"Perfect Beast", qui a été configuré pour représenter Teleaulax amphioxeia sur base de donnéesexpérimentales, suggèrent fortement que le cryptophyte se nourrit de bactéries pour compenserla limitation en phosphate. Ce travail montre l’importance de prendre en compte le mixoplanctondans les modèles d’écosystème en plus des hétérotrophes et des autotrophes stricts et que ladistinction entre les différents types fonctionnels de mixoplancton est importante. Le mixoplanctonse distingue par son utilisation des nutriments et sa dynamique de croissance. Les interactionsprédateur-proie n’ont pas les mêmes implications pour le mixoplancton que pour les hétérotropheset leur prise en compte dans les modèles pourrait améliorer notre compréhension de la formationdes efflorescences nuisibles de mixoplancton. La physiologie unique du mixoplancton, sonutilisation des nutriments et ses niveaux trophiques doivent être pris en compte dans les modèlesspécifiques aux espèces.

As transformações geradas pelo culminar do clima de guerra no século XX, principalmente na segunda metade, reflectiram-se na estrutura social e, subsequencialmente, escolar que, inevitavelmente, tiveram as suas repercussões na forma como se entendia e aceitava a deficiência. Consequência de um trilho igualmente evolutivo, deparamo-nos, no actual contexto, com a crescente disseminação das novas tecnologias da informação e da comunicação. Da união destas duas realidades díspares e simultaneamente complementares, esta investigação propõe-se a estudar o impacto das soluções tecnológicas no percurso educativo, num determinado público-alvo, atendendo, assim, às suas especificidades. Neste sentido, este trabalho, através da metodologia de estudo de caso, procura conhecer os processos de participação, interacção e inclusão de alunos com doença oncológica, através da utilização de uma solução tecnológica. Esta solução tecnológica advém da responsabilidade social de uma empresa portuguesa – a Portugal Telecom – que, através da Fundação PT e com o apoio da PT Inovação, desenvolvem e disponibilizam todos os meios técnicos necessários para que os alunos com deficiência possam assistir às aulas, a partir de suas casas. Cabe também aqui referir o papel mediador do Ministério da Educação, enquanto entidade superior, a quem competem todas as responsabilidades legais de foro educativo. O presente estudo, após a análise de todos os dados recolhidos, permite concluir que a solução PT Teleaula proporciona, em diferentes graus, a participação, a interacção e a inclusão dos alunos portadores de deficiência, através dos distintos modos de que dispõe para facultar a comunicação entre a escola e os alunos. ABSTRACT: The changes generated by the culmination of the climate of war in the 20th century, especially in the second half, were reflected in the social, and subsequently educational, structure, which inevitably had repercussions in the way disability is understood and accepted. In the current context, and as a result of an equally evolutionary path, we are faced with the increasing spread of new information and communication technologies. Based on the union of these two disparate and simultaneously complementary realities, this research sets itself to study the impact of technological solutions on the educational process of a particular audience, taking into consideration its specific characteristics. Adopting a case study methodology, this project seeks to understand the participation, interaction and inclusion processes of students suffering from oncological disease, through the use of a technology solution. This technological solution emanated from the social responsibility of a Portuguese company - Portugal Telecom - which, through the PT Foundation and with the support of PT Inovação, developed and made available all the necessary tools for students with disabilities to be able to attend classes at their homes. It is important to mention here the mediating role of the Ministry of Education, as a higher entity with all the legal responsibilities in the educational forum. After analyzing all the data collected, we can conclude that the PT Teleaula solution provides opportunities, to different degrees, for the participation, interaction and inclusion of students with disabilities through the various modes it has available to establish communication between the school and students.

Este estudo teve por objetivos identificar o perfil de trabalhador representado no Telecurso 2000 e configurar de que modo os adultos-trabalhadores participantes do Telecurso se vêem representados nas teleaulas. Com vistas a atingir os objetivos da pesquisa, foram observadas 10 teleaulas de Matemática e 10 teleaulas de História do Brasil, em nível de Ensino Fundamental, examinados documentos, realizadas entrevistas e utilizada a técnica da associação livre junto a quatro trabalhadores. A caracterização do projeto Telecurso 2000 foi desenvolvida a partir de roteiro para a análise de produtos televisivos proposto por Fischer (2001). A problematização acerca das representações sociais, com base nos estudos de Moscovici (1978), buscou estabelecer um movimento entre a análise do produto midiático e a escuta dos telealunos. The purpose of this dissertation is to identify the worker’s profile that is represented in the “Telecurso 2000” [an educational program broadcast by Globo Television Network] and to discuss how the adult workers who participate in that telecourse see themselves represented in the teleclasses. In order to reach this goal, the author observed 10 teleclasses of Mathematics and 10 teleclasses of Brazilian History at the level of elementary or basic education, examined documents, carried out interviews and used the free association technique with four workers. The characterization of the “Telecurso 2000” project was developed on the basis of a question list proposed by Fischer (2001) for the analysis of TV products. The problematization of the social representations was done on the basis of Moscovici’s studies (1978) and tried to establish a movement between the analysis of the media product and the listening to the telestudents. Abstract:
Este trabalho discute e avalia a acessibilidade de um material de ensino a distância muito disseminado no Brasil. Através de observações, viu-se a necessidade de tornar esse material mais acessível às pessoas surdas que se utilizam da Língua Brasileira de Sinais (Libras). Nosso objetivo é adaptar uma Teleaula do programa Telecurso 2000, que aborda o conceito de frações para alunos surdos a fim de viabilizar o acesso deste público a esse meio de ensino a distância, uma vez que esse material é amplamente utilizado por adultos buscando certificação de conclusão da educação básica e, com as mudanças nas leis, por empresas que buscam qualificação de seus funcionários. Nossa pesquisa foi desenvolvida em três etapas. Na primeira escolhemos os participantes iniciais, três surdos da região metropolitana de São Paulo, e aplicamos à esses participantes o material como é proposto, ou seja, em seu formato original; na segunda etapa focamos nas adaptações, produzimos assim a Teleaula Adaptada (TA) e a Apostila Adaptada (AA); e na terceira etapa submetemos o material adaptado (Teleaula e Apostila) ao crivo dos participantes finais, quatro surdos residentes na cidade de Rio Claro/SP. Tivemos como aporte teórico os trabalhos de Vygotsky (1997) sobre Defectologia, Sacks (2010) abordando a educação de surdos e Nunes (2012) o ensino de Números Racionais. As adaptações que adotamos estão relacionadas principalmente a utilização da primeira língua do público alvo, a questões de apresentação e representação visual e seleção de conteúdo. Nossa pesquisa mostrou que as adaptações realizadas foram necessárias para uma melhora na compreensão do conteúdo pelos participantes surdos, entretanto não foram suficientes para que eles pudessem realizar todas as atividades propostas. A questão da língua ainda é um dos principais fatores que podem dificultar a compreensão dos conteúdos, principalmente quando as proposta envolve material impresso. Para adaptar um material para o público surdo, não basta colocar uma Janela de Libras e pensar que os problemas serão resolvidos. Vai muito além. Para garantir acessibilidade a essa ou outra modalidade de educação à diversidade de usuários, é necessário ter um olhar mais minucioso e realmente levar em consideração as potencialidades do público em questão, caso contrário, será apenas mais um material criado com a falsa ideia de ajudar.

The proposal of the Continuing Education Program of the Methodist University of São Paulo in the television format raises a discussion of two important points. Firstly, to the issue of providing adequate training of the faculty that performs this task. Secondly, to the fact that the professor is no longer alone in the production of the educational process, but is part of a greater setting which is composed by a team of professionals responsible for the technical conduction and television operationalization of the classes. The present work seeks to analyze these intersubjective relations from a multi-referential perspective in an attempt to comprehend the importance of these relations in the pedagogical process of constructing televised classes. In order to reach the ultimate goal of this investigation, the first chapter will describe the methods and approaches utilized in the examination. The second chapter will provide a theoretical panorama that accompanies the binomial relationship between Education and Communication, so that a discussion of the impact and changes made to the Educational field as a result of the influence of the information and communicational technologies is raised. The third chapter is constituted of a description of the development of implementation of the Continuing Education program of the Methodist University of Sao Paulo and of the television formulation of the classes, as well as the implications of the given format in the practice of the faculty. The fourth chapter presents the occurrences registered in the research journal, as the faculty training and respective analysis based on the multireferential approach took place, followed by the conclusion and final considerations.(AU)
Ao propor as suas teleaulas no formato televisivo, o programa de Educação a Distância da Metodista de São Paulo, problematiza a questão da capacitação docente para esta modalidade de ensino e propõe a constituição de um novo espaço pedagógico: O estúdio de TV. Entretanto, o professor não está mais sozinho na produção destes processos educativos, agora, também, faz parte a equipe responsável pela condução técnica e operacionalização televisiva destas aulas. Este trabalho objetiva fazer uma leitura destas relações intersubjetivas, a partir de uma perspectiva multirreferencial, para a compreensão da importância destas relações na construção pedagógica da teleaulas. Para tanto, no capitulo um, encontra-se as descrições dos métodos e a abordagem utilizada na pesquisa. O capitulo dois é composto por um panorama teórico em torno do binômio educação e comunicação, a fim de delinear as alterações decorrentes das tecnologias da informação e comunicação na educação. O capitulo três é constituído pela descrição da implantação do programa de Educação a distancia da Universidade Metodista, e da formatação televisiva de suas teleaulas, assim como as implicações destas na prática docente. No capitulo quatro, a descrição de ocorrências registradas no jornal de pesquisa, durante as capacitações docentes e teleaulas do programa, e as respectivas análises pautadas na abordagem adotada, apresentando por fim, as conclusões e as considerações finais.(AU)

The present study proposes an understanding of the interpersonal relationships between subjects involved in the construction and execution of the pedagogic Project of Distance Learning under an ethno methodological and multi-referential perspective of education.We propose identifying how relationships are established in Distance Learning, which its organizational agents are and how they interact in the organization of the work, since Distance Learning requires an array of professionals with different kinds of knowledge so as to achieve its goals. We used as reference Distance Learning Courses at UNESP and through the use of observation, reports, questionnaires and interviews, we intend to identify how subjects see and understand themselves in the process and how they understand and recognize other subjects in the development of activities of such course. Using as reference the steps taken in the elaboration and realization of a video-conference, we aim at recognizing the communication process within the group of technicians, managers, professors and learners in the organization, as well as analyzing connections and ruptures, the wide-range actions and the importance of its functions on a daily basis.(AU)
O presente trabalho propõe um entendimento sobre as relações interpessoais entre os sujeitos envolvidos na construção e execução do projeto pedagógico de Educação na modalidade a distância sob uma perspectiva etnometodológica e multirreferencial de educação. Propomos identificar como as relações se estabelecem no cotidiano da EAD, quais são seus agentes organizacionais e como estes se interagem na estruturação do trabalho, uma vez que a EAD exige uma complexidade de profissionais de diferentes saberes para a concretização de seus objetivos. Tomando como referência os cursos de educação a distância da UMESP, pretendemos, por meio de observação, relatórios, aplicação de questionários e entrevistas, identificar como cada agente se vê e se percebe no processo, e de que forma entende e reconhece o outro no desenvolvimento das atividades do referido curso. Utilizando como referência o percurso realizado na elaboração e operacionalização de uma teleaula desejamos reconhecer os processos comunicacionais entre o grupo de profissionais técnicos, dos gestores, dos docentes e discentes dentro da organização acadêmica, analisando suas conexões e rupturas, assim como a abrangência e importância de suas funções na prática diária.(AU)

A teleaula constitui um recurso pedagógico de ensino à distância. A sua implementação na prática pedagógica pressupõe uma preparação prévia do docente no sentido de proporcionar ao aluno, que por razões de saúde se encontre impossibilitado de frequentar as aulas em ambiente de sala de aula numa escola, uma educação de qualidade. A legislação portuguesa assegura o direito ao aluno que se encontra nestas circunstâncias, para concretizar a sua escolarização obrigatória através do ensino à distância. Neste estudo de caso, procedeu-se ao levantamento de necessidades de formação de professores, que lecionam numa escola do Funchal, relativamente à implementação de aulas através de teleaula. Participaram trinta e dois docentes, respondendo a um questionário disponibilizado online. Foi ainda realizada uma entrevista semiestruturada no intuito de conhecer melhor a monitorização e avaliação desta modalidade de ensino, teleaula, na Região Autónoma da Madeira. Concluiu-se que a maioria dos docentes inquiridos, quando questionados sobre a sua formação relativamente à implementação do ensino à distância através da teleaula, concordam que a sua formação é fraca, manifestando a maioria dos docentes também, uma opinião favorável quanto à relevância de um programa de formação sobre teleaula e quanto à adesão a um programa de formação sobre esta temática. Após reflexão sobre os dados recolhidos elaborou-se uma proposta de programa de formação de professores para a implementação da teleaula em escolas do 3º ciclo e secundário.
The teleclass is a pedagogical resource for distance teaching. Its implementation in the pedagogical practice presupposes a previous preparation of the teacher to provide the student, who for reasons of health is unable to attend classes in a classroom environment in a school, a quality education. The portuguese legislation ensures the right to the student who is in these circumstances, to realize their compulsory schooling through distance education. In this case study, the needs of teachers training were studied in a school in Funchal, in relation to the implementation of classes through teleclass. Thirty-two teachers participated, responding to a questionnaire made available online. A semi-structured interview was also carried out to better understand the monitoring and evaluation of this type of teaching, teleclass, in the Autonomous Region of Madeira. It was concluded that most teachers interviewed, when questioned about their training regarding the implementation of distance teaching through teleclass, agree that their training is weak, and most teachers also express a favorable opinion about the relevance of a training program on teleclass and on adherence to a training program on this subject. After reflection on the collected data, a proposal was made for a teacher training program for the implementation of teleclass in middle and high schools.
