Tetrahydrocurcumin, the most abundant curcumin transformation product in biological systems, can potentially be a new alternative therapeutic agent with improved anti-inflammatory activity and higher bioavailability than curcumin. In this article, we describe the synthesis and evaluation of the anti-inflammatory activities of tetrahydrocurcumin derivatives. Eleven tetrahydrocurcumin derivatives were synthesized via Steglich esterification on both sides of the phenolic rings of tetrahydrocurcumin with the aim of improving the anti-inflammatory activity of this compound. We showed that tetrahydr

Despite advances in understanding the molecular basis of tumorigenesis, diagnosis, and clinical therapies, the discovery and development of effective drugs is an active and vital field in cancer research. Tetrahydrocurcumin is a major curcuminoid metabolite of curcumin, naturally occurring in turmeric. The interest in tetrahydrocurcumin research is increasing because it is superior to curcumin in its solubility in water, chemical stability, bioavailability, and anti-oxidative activity. Moreover, the pro-oxidant effect of curcumin is reported, whereas tetrahydrocurcumin does not show any pro-oxidant effects. This contrasting behaviour of the two is attributed to their structures, because conjugation is involved only in curcumin, not in the tetrahydrocurcumin. It can b

Cancer is a complicated disease with many puzzles of tumorigenesis remaining unsolved; however, there is strong evidence regarding the link between oxidative stress and cancer. The malignant transformation of cells requires the integration of many factors, and reactive oxygen species (ROS) are dynamically implicated in each step of tumorigenesis. Moreover, ROS are involved in tumorigenesis not only within cancer cells, but also in the interplay between malignant and nonmalignant cells.
