Abstract:
Abstract The present article addresses genre shifts in Israeli political campaign in 2013, specifically written advertisements published in newspapers, on the Internet, on stickers, in leaflets, and on billboards. I examine shifts to diverse genres (protest slogans, writing on social networks, personal conversation, a math exercise etc.), and analyze various discursive and para-linguistic strategies by means of which these shifts are implemented: use of registers and sociolects identified with particular genres; syntactical, semantic and lexical repetitions; graphic, typographical, and visual elements, etc. I show that due to the expectations of the addressees regarding the reconstructed genres and the addressing parties, genre shifts may serve to fulfill two main pragmatic-rhetorical functions: (1) Promoting messages regarding the desired conduct of the voters during the elections; (2) Self-positioning of the parties, either to strengthen their existing image among the public or to create a new, surprising one. I thus emphasize the key role played by genre shifts in the intertextual phenomenon of integrating semiotic meanings into a linguistic text; specifically in this case, integrating cultural-societal meanings into campaign advertisements.

Abstract:
The article is devoted to the analysis of the rhythm of texts of different genres: fiction novels, advertisements, scientific articles, reviews, tweets, and political articles. The authors identified lexico-grammatical figures in the texts: anaphora, epiphora, diacope, aposiopesis, etc., that are markers of the text rhythm. On their basis, statistical features were calculated that describe quantitatively and structurally these rhythm features.The resulting text model was visualized for statistical analysis using boxplots and heat maps that showed differences in the rhythm of texts of different genres. The boxplots showed that almost all genres differ from each other in terms of the overall density of rhythm features. Heatmaps showed different rhythm patterns across genres. Further, the rhythm features were successfully used to classify texts into six genres. The classification was carried out in two ways: a binary classification for each genre in order to separate a particular genre from the rest genres, and a multi-class classification of the text corpus into six genres at once. Two text corpora in English and Russian were used for the experiments. Each corpus contains 100 fiction novels, scientific articles, advertisements and tweets, 50 reviews and political articles, i.e. a total of 500 texts. The high quality of the classification with neural networks showed that rhythm features are a good marker for most genres, especially fiction. The experiments were carried out using the ProseRhythmDetector software tool for Russian and English languages. Text corpora contains 300 texts for each language.

Abstract:
Every text has at least one topic and at least one genre. Evidence for a text's topic and genre comes, in part, from its lexical and syntactic features—features used in both Automatic Topic Classification and Automatic Genre Classification (AGC). Because an ideal AGC system should be stable in the face of changes in topic distribution, we assess five previously published AGC methods with respect to both performance on the same topic–genre distribution on which they were trained and stability of that performance across changes in topic–genre distribution. Our experiments lead us to conclude that (1) stability in the face of changing topical distributions should be added to the evaluation critera for new approaches to AGC, and (2) Part-of-Speech features should be considered individually when developing a high-performing, stable AGC system for a particular, possibly changing corpus.

Abstract This paper considers issues related to the textual organization of university lectures and the degree to which they are comprised of conventionalized text-organizational schema or genre stagings. It utilizes a dataset of video recordings and transcripts of four social science lectures which were delivered by two lecturers at a university in Japan in 2003, and offers findings which demonstrate that lecturers draw on a range of ready-made genres of the type which have been described in the so-called Sydney school genre literature and combine these conventionalized genres into genre complexes, which appear not to be conventionally or predictably ordered. The paper also offers some insights into at least one aspect of the distinction between online (spontaneous/spoken) versus offline (prepared/written) language. Specifically, it shows that lectures, as instances of spoken, apparently extemporaneous language do, to a significant degree, rely on pre-fabricated and culturally recurrent textual arrangements which the speakers draw on as they spontaneously produce the current communicative event.

Abstract The purpose of this study is to support early childhood teachers’ understanding of the diversity and sophistication present in young children’s storytelling to counteract singular and tacit conceptualizations of story. Drawing on the notion of genre, this study uses systemic functional linguistics to examine rhetorical patterns in the storytelling of a group of children in multilingual and multicultural classrooms. Stories were collected in the context of a small group storytelling activity called story circles. Participation in story circles resulted in 176 stories. Children’s stories were parsed into story stages and story genres based on functional purpose and known patterns in the grammar of language. Results show that the children told stories that varied not only in length and complexity, but in choice of story genres. Further, the majority of children employed different genres of story across the four weeks of the activity. The children in this sample used storytelling as a vehicle for giving significance to everyday experience, elevating ideas and establishing relationships in the classroom context.

In most current work on genre, a set of genre categories needs to be predetermined. However, there are some cases where such predetermined genres cannot be clearly identified. Popular science, for instance, is a broad register carrying several specific purposes within it, suggesting that there are several genres of popular science, but it is unclear what these genres are. This paper introduces a linguistic approach to reveal hidden genres. For 600 written popular science texts from a variety of sources and disciplines, linguistic features were analysed using a range of computer programs and a cluster analysis conducted. The analysis produced four clusters with shared linguistic features, representing text types. The association of these text types with key features, functional relations, dominant sources, and prototypical members of each cluster helps us to induce genres on the basis of communicative purposes, a traditional criterion in identifying genres. Whether the produced text types are equivalent to genres was evaluated with a test set of data. The proposed approach achieves more than 70 % accuracy. The approach appears applicable for identifying genres of popular science and has pedagogical implications.

The purpose of this paper is to present and analyse text genres offer and authorisation/approval as unilateral text types that are mostly used for communication purposes in tourism and hospitality management. This analysis will show a specific communication model of management. The paper starts with an introduction that leads to argumentation of text within LSP and discusses the text as a tool in management of tourism and hospitality corporations and corporations in general. Further, this paper gives shortly the methodolgy of corpus description and classification of commisives. Unilateral commisives, their function and definition is discussed, and afterwards follows the analysis of text genre offer and authorisation/approval used in corporations and companies. The conclusion gives the main characteristics of unilateral text genres in corporative communication and also gives characteristics of communication models used by management. The corpus for this study includes examples of texts which are used for communication purposes in corporations. Text genres offer and authorisation/approval are characterized as unilateral text subtypes. This paper provides a linguistic analysis of the text and text genres. Every text will be analysed within three levels. Text genres are crucial for gaining success in corporate communication. Depending from the type of corporate ideology and philosophy, text genres offer and authorisation/approval show characteristics on the micro and macro structural level, they show specific vocabulary, grammatical structures and syntax in communication. Communicative function and implementation of communication in written discourse is discussed in this paper. Available sources and references on this topic are based on text linguistics and analysis of general text genres. However, little has been published and no research of text genres used in corporations (as specialized text genres) has been conducted in Croatian so far.

Abstract In the present study, we sought to clarify how differences in contextualized experience influence the performance of participants engaged in genre decision-making. Using a simple learning algorithm, we ran a series of computational simulations to model the effects that context and cue competition have on the way readers of different backgrounds make genre decisions. Next, we used the results of those simulations as predictions for our behavioural genre decision experiment. Differences in test performance were strongly influenced by the factors that have long been known to influence learning: Cue competition and its embedding in a specific context jointly modulate what gets learned and that inevitably affects later performance. We discuss our findings in the context of learning and literary genres.

This dissertation wants to understand the church history regarding the divide which resulted in the so called fundamentalist and the liberal movements in the church being birthed in identifying the reasons for the conflict between the aforementioned groups. It then presents principles of conflict transformation and management in order to understand and to help transform and manage potential confrontational situations between the aforementioned groups effectively. It further propose alternative interpretations to potential conflict-generating Bible texts by extrapolating new information from those texts in order to stir the potential of birthing a second naïve love for the discredited texts by offering new meanings to those same texts. Unfortunately, this objective can’t effectively be measured within the scope of this dissertation, but will nevertheless remain as an objective of hope.

This work aims the investigation of the several criminal suit court record text categories: types, genres and species (according to TRAVAGLIA, [2003]/2007) which are used in our society in the criminal court. Thus, it is necessary to contemplate the idea that the number of texts used in a criminal process is great and that each epoch lives a set of rules that are of its own, i.e, they do not abandon the past, they do not break up with the traditions, although they shape or discipline the human facts according to the demands of the moment. Therefore, our intention is to verify the existence of these several categories of texts, characterize them linguistically and verify in the genre sentence the reflexes, by means of linguistic marks, of those text genres which were used in criminal processes and motivated the judge to make up his decision and declare his sentence . This study was done according to the theoretical bases of the Text Linguistics and the reflections of the several text categories present in our daily routine at the criminal court, about which, lawyers manuals refer to, sometimes as criminal court documents, process or simply nominate them according to the legal terminology proposed in our criminal legal texts, which many times can lead to misunderstandings once they do not have the rigor of the juridical terminology, once the rigor of the law science depends substantially on the language which can not nominate different institutes with the same name neither can it nominate the same institutes with different names. Nevertheless, the legislator did not hold on any criteria and, being divorced of any scientific or systematic orientation, has opted for each law the empiric solutions. So, for the analyses of our text categories we have developed a qualitative study with an analytic-descriptive methodology that uses scraps of the collected corpus from 10 (ten) criminal processes collected for this investigation with the scope of analyzing and characterizing each of them with special attention to the sentence by means of the linguistic materiality of this corpus which is left behind as a clue to the motivation of the judge. Such analyses and characterizations are also based on the foundations of the Theory of the Speech Acts (AUSTIN, 1962), The Communicative Action Theory (HABERMAS, 1983, 2003), the theories about the argumentation, as well as the postulate of Discourse Communities (SWALES, 1990).
Este estudo tem como finalidade investigar as várias categorias de texto forenses criminal: tipos, gêneros e espécies (segundo TRAVAGLIA, [2003]/2007) que circulam em nossa sociedade. Destarte, é necessário contemplar a idéia de que grande é o número das categorias de texto que circulam em nossa sociedade e que cada época vive um complexo de regras que lhe são próprias, ou seja, não desprezam o passado, não rompem com as tradições, contudo, modelam ou disciplinam os fatos humanos, segundo as exigências do seu momento. Portanto pretende-se verificar a existência das várias categorias de texto presentes nos processos penais, caracterizá-las lingüisticamente e verificar no gênero sentença os reflexos, por meio das marcas lingüísticas, daqueles gêneros textuais apresentados no decorrer do processo criminal que motivam o julgador a proferir sua sentença. Norteia-se, este estudo, teoricamente pelos estudos da Lingüística Textual e pelas reflexões sobre as mais variadas categorias de texto presentes no nosso dia a dia forense, sobre as quais manuais de advogados ora se referem como peças processuais, ora como processo, ou simplesmente pela terminologia jurídica proposta na legislação, que muitas vezes leva ao erro, uma vez que não possuem um rigor da terminologia na linguagem jurídica. Haja vista, que o rigor da ciência jurídica depende substancialmente da linguagem, não devendo designar com um nome comum institutos diversos, nem institutos iguais com nomes diferentes. O legislador não abraçou, porém, nenhum critério e divorciado de qualquer preocupação científica ou sistemática, preferiu, em cada lei, as soluções meramente empíricas. Assim, para a análise das categorias de textos forenses criminais, desenvolve-se um estudo qualitativo, cuja metodologia analítico-descritiva faz recortes no corpus coletado em 10 (dez) processos criminais, analisa-se e caracteriza-se criteriosamente o gênero sentença por meio das marcas presentes na materialidade lingüística desse corpus. Tais análises e caracterizações norteiam-se, também, pelos pressupostos teóricos da Teoria dos Atos de Fala (AUSTIN, 1962), da teoria da Ação Comunicativa (HABERMAS, 1983,2003), as teorias sobre a argumentação, assim como pelo postulado teórico de Comunidades Discursivas (SWALES, 1990).

Abstract:
Bachelor thesis focuses on problems and issues of machine translation while translating texts of different genres. The theoretical part of the paper covers such topics as the notion of machine translation (MT), its most crucial mistakes and the notion of text genres in the English language. The practical part consists of the analysis of 5 different texts pointing out the most severe mistakes detected in the output of MT. Conclusions drawn after the analysis showed that MT requires further improvment and more thorough investigation.

Teorinė dalis aptaria tokias temas, kaip mašininio vertimo (MV) samprata ir šio reiškinio pačias svarbiausias klaidas, taip pat šioje dalyje aptariama teksto stiliaus samprata. Praktinėje dalyje buvo analizuojami 5 skirtingų stilių tekstai ir aptariamos pačios ryškiausios ir svarbiausios klaidos šiuose tekstuose. Išvados paaiškėjusios po tyrimo atskleidė, kad MV sistemą dar reikia tobulinti ir atlikti išsamesnius tyrimus.

Nos dias de hoje, podemos constatar o crescimento das formas de linguagem mediadas pelas tecnologias da informação e comunicação. É em torno dessa constatação e nesse contexto que esta dissertação se insere. Seu tema é a caracterização do par post/comentário à luz da noção de gêneros textuais. Nosso objetivo é refletir sobre o referido, a fim de responder os seguintes questionamentos: (a) se post e comentário são dois gêneros textuais ou partes de um mesmo fenômeno de comunicação/gênero; (b) se são gêneros, como caracterizá-los e diferenciá-los; (c) não sendo o post um novo gênero textual, que especificidades podem caracterizá-lo como ferramenta e quais os aspectos que o vinculam ao gênero comentário. A fim de respondê-los, nossa opção metodológica foi por um estudo documental de cunho qualitativo. Este consistiu na coleta de publicações feitas no perfil da pesquisadora na rede social Facebook por diferentes sujeitos, no período pré e pós eleições brasileiras de 2014, especificamente no mês de outubro. Os posts e comentários selecionados referem-se exclusivamente à temática das eleições presidenciais. Entendemos que as redes sociais têm sido grandes incentivadoras à produção escrita e, consequentemente, promotoras de atividades de compreensão leitora. E, no que concerne em particular ao tema escolhido, acolheram e fomentaram intensos debates. As reflexões sobre leitura, portanto, perpassam este trabalho porque, ao pensarmos no nascimento e uso de um gênero, pensamos, também, em sua recepção, logo, nos processos leitores envolvidos. Para analisar os dados, utilizamos a proposta de Donato (2014), discutindo os quatro pilares constituintes do gênero função comunicativa, forma, conteúdo e suporte, e também pensando em três níveis: o da produção, recepção e contextualização. Como base teórico-metodológica seguimos a perspectiva da sociocognição, calcada especialmente nos estudos de Marcuschi (2005, 2008, 2010), Koch (1996, 2003) e Koch e Elias (2008, 2013), assim como fundamentos de Bakhtin (1997) sobre o caráter sócio-histórico dos gêneros. A título de conclusão, segundo nossas análises, salientamos a interdependência dos elementos do par estudado. No entanto, defendemos a delimitação do post enquanto gênero textual eminentemente digital e não apenas uma ferramenta ou parte do gênero comentário
Today we can see the growth of language forms mediated by information and communication technologies. It is through these findings and in this context that this dissertation is introduced. Its theme is characterized by the pair post/ comments within the light of concept text genres. Our objective is to reflect on the mentioned material above in order to answer the following questions: (a) is post and comment two genres or parts of the same phenomenon of communication / genrer; (b) if these are genre, how do we then characterize them and differentiate between them; (c) if this post is not a new genre, how can we specifically characterize it as a tool and which aspects of the post tie it to the genre comment. In order to answer these questions, our methodological option was a documented study of qualitative nature. This consisted with the collection of publications from the researchers social network profile within Facebook during the period of the Brazilian elections in 2014, specifically from the month of October. The posts and selected comments refer exclusively to the topic of presidential elections. We understand that social networks have been a great incentive towards writing production and therefore promote reading comprehension activities. With regard to the particular chosen theme, we welcomed and encouraged intense debate. When reflecting on literature, we prepare this work for the birth of a new text genre and also how it is received so the reader is involved in the process. To analyze the data, we use the proposed Donato (2014), discussing the four constituent pillars of genre - communicative function, form, content and support. We also considered three levels: production, reception and context. The theoretical and methodological basis we followed the perspective of sociocognitive, based especially on studies from Marcuschi (2005, 2008, 2010), Koch (1996, 2003) and Koch and Elias (2008, 2013), as well as the foundations of Bakhtin (1997) on the socio-historical character of genres. By way of conclusion, according to our analysis, we stressed the interdependence of the pair of elements studied. However, we defend the limitation of the post as an eminently digital text genre and not just as a tool or part of a genre comment

O ensino de produção escrita de textos literários está presente em livros didáticos de língua portuguesa desde as primeiras coletâneas com os exercícios de composição, quando se propunha o texto literário como modelo para ensinar a escrever de forma elaborada. Avaliações municipais da cidade de São Paulo apontam que muitos alunos do segundo ciclo do Ensino Fundamental apresentam dificuldades na elaboração de um texto escrito. Nesse sentido, o objetivo desta dissertação é descrever e analisar as atividades didáticas de produção escrita de conto e de causo de alunos do 6º ano do ensino fundamental, procurando identificar os procedimentos linguístico-discursivos que orientam essas propostas. O corpus selecionado é o material didático da rede municipal da cidade de São Paulo - Cadernos de Apoio e Aprendizagem: Língua Portuguesa, dirigido a alunos e professores do Ensino Fundamental II, publicado em 2010 e editado pela Fundação Padre Anchieta. Três questões nortearam a análise dos dados coletados: 1) Como as propostas de produção escrita contextualizam socialmente o conto e o causo? 2) Que atividades apresentam e discutem as características discursivas e textuais do conto e do causo? 3) Como são desenvolvidas as estratégias linguísticas e discursivas da produção escrita? A fundamentação teórico-metodológica centra-se na teoria bakhtiniana, particularmente nos conceitos de enunciado, gênero do discurso e texto. O foco central da análise foi a unidade 1 do 6º ano Um pouquinho de prosa: a arte de contar histórias em contos e causos, percorrendo a constitutiva relação entre as atividades de leitura, compreensão e escrita. A partir da proposta de leitura de dez contos de diferentes culturas, escritos em várias épocas históricas, as atividades de compreensão permitem que o aluno identifique as estruturas fundamentais da narrativa e como o gênero discursivo se modifica conforme a interação dos vários interlocutores sociais, contribuindo para um entendimento da historicidade do gênero conto. Os principais recursos linguístico-discursivos destacados na construção narrativa são o uso de tempos verbais (no eixo do pretérito imperfeito e mais-queperfeito e do futuro do pretérito), dos verbos de dizer e o emprego dos pronomes. Quanto à orientação do causo, o foco aparece na apresentação de três vídeos que exploram os recursos narrativos da oralidade. Com base nesses filmes o aluno é orientado a escrever um causo observando os recursos da fala. A partir da produção escrita do causo é proposta a dramatização, momento significante de entonação expressiva, gestos, sons e expressão corporal. São recursos da oralidade que aparecem na construção do causo. A escrita é entendida como o resultado do processo do ensino. As várias atividades didáticas contribuem para o desenvolvimento da fluência escrita na medida em que os recursos narrativos são ensinados com base na materialidade linguístico-discursiva dos textos analisados.
The teaching of written production of literary texts has been part of Brazilian Portuguese school books since their very first collections, when it was proposed the literary texts as a model to teach how to write well. Municipal evaluations from São Paulo city show that many students from the second cycle of basic education have difficulties in writing a text. In this context, the aim of this research is to describe and analyse the activities of written productions of conto (Brazilian short stories) and causo (Brazilian popular tales) by students from the 6th basic education year, aiming to identifying the linguistic-discursive procedures that guide those proposals. The corpus of this research is the school material from the municipal education network of São Paulo city - Cadernos de Apoio e Aprendizagem: Língua Portuguesa, directed to students and teachers from the second cycle of basic education, published in 2010 and edited by Fundação Padre Anchieta. Three questions were the basis of the data analysis: 1) How do the written production proposals socially contextualize conto and causo? 2) Which activities present and analyse the discursive and textual features of conto and causo? 3) How are the linguistic and discursive strategies of written production developed? The theoretical-methodological literature revision is based on Bakhtin´s theory, especially in the concepts of utterances, speech genres and text. The main focus of the analysis was the first unit of the 6th basic education year: Um pouquinho de prosa: a arte de contar histórias em contos e causos, through the relationship between reading, comprehension and writing processes. Through the reading of ten different contos from different cultures, written in many historical moments, the comprehension activities allow students to identify the main structures of narrative, as well as the way this speech genre can modify itself according to the interaction of various social interlocutors. In this way, it contributes to understand the historicism of conto. The main linguistic-discursive resources in the narrative are the verbal tenses (Brazilian Portuguese kinds of past pretérito perfeito, mais-queperfeito and futuro do pretérito), the use of verbs de dizer (to say) and pronouns. Concerning the guidance for causo, the focus was three movies exploring the oral narrative resources. The student, based on the movie watched with his/her teacher, is required to write a causo regarding the speech resources. From the written production of causo, it is proposed the dramatization, a very important moment of expressive intonation, movements, sounds and body expression. These are all speech resources that appear when writing a causo. The writing process is understood as the result of the teaching process. The various school activities contribute for developing the writing skill, for the narrative resources are taught based on the linguistic-discursive materiality of analysed texts.

Esta dissertação pretende, mediante a análise de livros didáticos aprovados pelo PNLD 2005 - Plano Nacional do Livro Didático, trazer à luz discussões a respeito dos gêneros narrativos ficcionais nas escolas hoje. O trabalho se inicia com o levantamento e discussão das principais correntes teóricas acerca da narrativa e teorias da narratividade. A partir daí, buscando conceituar gêneros do discurso e tipos de texto, eixo norteador desta pesquisa, apresentamos elementos da discussão recente sobre o assunto entre os pesquisadores na área de ensino de língua portuguesa. Em seguida, procuramos analisar a forma como esses conceitos aparecem nos documentos oficiais - PCN e PNLD. Por fim, apresentamos a análise de duas coleções didáticas (Português: Linguagens de William Roberto Cereja e Thereza Cochar Magalhães e Português: Uma proposta para o letramento de Magda Soares) dirigidas a alunos do Ensino Fundamental II à luz das discussões anteriores. Com a discussão teórica sobre a presença dos gêneros do discurso e tipos de texto nos documentos oficiais e livros didáticos e a análise do livro didático de Português para o Ensino Fundamental II, pretendemos levantar questões sobre a importância da produção do texto narrativo ficcional na escola hoje.
This dissertation aims at discussing the narrative genres in schools today. The first part presents the main theoretical studies about narrative and narratology. By the study on discourse genres and types of text, main trait of this research, it is intended to show elements of a recent discussion among researchers in the field of teaching Portuguese. Next, it is investigated the concepts of genre and text in the official documents - PCN and PNLD. Finally, it is presented an analysis of two collections of books (Português: Linguagens de William Roberto Cereja e Thereza Cochar Magalhães e Português: Uma proposta para o letramento de Magda Soares) intended for Ensino Fundamental II students. By presenting a discussion on discourse genres and types of texts in the official documents and text books and an analysis of Portuguese text books, it is intended to raise questions on the importance of fiction writing in schools today.

O objetivo central do presente trabalho é caracterizar o gênero matéria televisiva ou VT, considerando suas propriedades sociorretóricas e funcionais, esperando fornecer subsídios para o desenvolvimento de estratégias de ensino da produção textual no âmbito do telejornalismo. Nesta pesquisa, descrevemos a referida categoria a partir da articulação de vertentes teóricas distintas. Por um lado, teorias de gêneros textuais em que estes são tratados como mediadores efetivos nas trocas simbólicas e intersubjetivas , sendo suas configurações típicas resultantes, fundamentalmente, da natureza dialógica da linguagem e dos propósitos comunicativos almejados em cada circunstância nas quais são empregados os “tipos relativamente estáveis” de enunciados. Por outro, o arcabouço conceitual e técnico oriundo do campo jornalístico, de modo mais amplo, e do telejornalismo, em específico, por meio do qual rastreamos a evolução dos padrões de textualização no jornalismo em tevê visando ao entendimento das razões – institucionais, organizacionais, midiáticas e mercadológicas – em torno dos mesmos. Da mesma perspectiva sociorretórica que norteia nosso enfoque, no tocante à teoria, também derivam nossas opções metodológicas. Assim, numa primeira etapa, analisamos um corpus contendo 88 textos de VTs, em busca de recorrências pertinentes a organização bem como a modalidades retóricas, além de estratégias e movimentos mais habituais – investigação que nos permitiu elaborar modelos de categorias subgenéricas. Esses modelos, em seguida, na segunda parte, foram submetidos à apreciação de usuários profissionais do gênero – fase em que priorizamos a investigação dos aspectos culturais, compartilhados pela comunidade discursiva em questão, relevantes para o estabelecimento da identidade e dos limites genéricos, e ainda das transgressões toleráveis. Identificamos, então, duas subcategorias genéricas: as matérias factuais e frias. Estas, para fins didáticos, passaram a ser tratadas como pólos de um contínuo, sendo as características dos exemplares do gênero matéria televisiva decorrentes da pendência para uma ou outra das extremidades. Diante dos resultados, evidenciou-se a importância de uma aproximação mais intensa entre os estudos contemporâneos das práticas textuais com universos de atuação que demandam uma produtividade eficiente e, simultaneamente, inovadora.
This work aims to characterize the television broadcast genre or VT, considering its socio-rhetorical and functional properties and willing to offer support for the development of teaching strategies on text production in news broadcasting. In this study, this category is described by articulating different theoretical perspectives. On the one hand, theories of text genres, in which these are treated as effective mediators in symbolic and intersubjective exchanges, with their typical settings resulting fundamentally from the dialogical nature of language and from the communicative purposes pursued in every circumstance in which the "relatively stable types" of statements apply. On the other hand, the conceptual and technical framework arising from the journalistic field, broadly, and in television journalism, in particular, through which the evolution of patterns of textualization in TV journalism is tracked aiming at understanding the reasons - institutional, organizational, media and marketing - around it. The same socio-rhetorical perspective that guides the approach, concerning theory, also provides the methodological choices. In the first part, a corpus containing texts of 88 VTs is analyzed in search of relevant recurrences to the organization, as well as the rhetorical modes, along with strategies and more habitual movements - research that allowed the development of models of subgeneric categories. In the second part, these models were submitted to consideration of professional users of the genre - on this stage, the investigation of the cultural aspects shared by the interested speech community was prioritized, relevant to establishing the identity and generic limits, beyond the tolerable transgressions. Then, two general subcategories were identified: the factual and the cold materials. These, for teaching purposes, are now treated as poles of a continuum, with the characteristics of the specimens of the television broadcast genre arising from pending to one or the other end. Therefore, the results showed the importance of a more intense approach between contemporary studies of textual practices and several activities that require an efficient and simultaneously innovative productivity.

Este estudo tem como objetivo propor uma ação pedagógica interdisciplinar integrada a um projeto de leitura para a 5ª série do Ensino Fundamental de uma escola da rede municipal de ensino de Caxias do Sul. Os dados obtidos através de registros de observações- participantes de reuniões pedagógicas, reuniões individuais com a coordenação e conversas informais com professores, articulados com documentos da escola, e com a resposta dos professores à apresentação da proposta sugerem que a escola apresenta aspectos favoráveis para o trabalho interdisciplinar, assim como algumas limitações para que ele aconteça na prática. A proposta de ação pedagógica busca detalhar atividades de um projeto interdisciplinar, com o apoio de referenciais teóricos para que a interdisciplinaridade aconteça na prática.
The aim of this study is to propose an interdisciplinary educational action integrated to a reading project for the 5th grade of an elementary school that is part of the municipal public school system in Caxias do Sul. The data were obtained from observation recordings in pedagogical meetings, individual meetings with the pedagogical coordinator, and informal conversations among teachers. The data have been further related to some school documents and the response by the teachers to the proposal presented. The findings suggest that the school shows some positive aspects towards interdisciplinary work, as well as some limitations that prevent it from happening adequately. The proposed action aims to detail educational activities of an interdisciplinary project with the support of a theoretical background in order to create conditions for actual interdisciplinary.

A importância das atividades escolares com narrativas autobiográficas é levar os estudantes a um movimento de interiorização, alimentando a imaginação de como contar aos outro sua pequena biografia. Nas autobiografias escolares, a escrita permite aos alunos tornarem-se autores de suas próprias trajetórias, e traçar o processo sócio-histórico em que viveram e vivem. Nesta dissertação, a proposta de escrita autobiográfica com duas turmas do 7º ano dos anos finais do ensino fundamental mostrou-se relevante devido a três aspectos: a) o envolvimento dos participantes de 12 anos com a criação de um texto pessoal; b) em cada relato da curta vida dos jovens há um processo da comunicação breve alimentando o texto; c) o acompanhamento da leitura do conjunto de textos permite ao leitor uma valorização de objetos, lugares e pessoa trazidos pelos alunos que contam rápidas biografias e constroem a singularidade dos percursos juvenis. Assim, o ensino de produção escrita pode ser potencializado pelo estudo dos gêneros discursivos que se voltam para o eu do aluno. O objetivo central desta dissertação é analisar a singularidade e a memória discursiva de jovens do 7º ano. Por meio de textos produzidos durante uma sequência didática, busca-se identificar os procedimentos linguístico-discursivos utilizados na estruturação dos textos, verificando as categorias de tempo e espaço empregadas na reorganização da trajetória de vida, a fim de compreender as marcas valorativas que constituem a identidade dos alunos. Desta forma, duas perguntas norteiam este estudo: 1. De que maneira linguístico-discursiva os estudantes reconstroem o tempo e o espaço? 2. Quais elementos memorialísticos emergem das narrativas autobiográficas? Como pressupostos teóricos, o conceito de gênero discursivo, de Mikhail Bakhtin e o Círculo e a noção autobiográfica na perspectiva teórica de Philippe Lejeune e de Leonor Arfuch com a finalidade de aprofundar características do gênero autobiográfico. A partir da análise do corpus organizado com a produção e reelaboração de 42 redações, os resultados indicam a importância do ato de escrever sobre si, o que possibilita ao aluno ampliar seu repertório de visões de mundo, uma vez que se abre uma possibilidade com a subjetividade, ainda de maneira breve por se tratar de jovens autores. Ao se voltarem para as suas vidas, temas singulares que guardam dentro de si se tornam matérias fundamentais para a construção de narrativas sobre perdas e deslocamentos. The importance of school activities with autobiographical narratives is to lead the students to a movement of interiorization, fueling the imagination of how to tell the others their small biography. In school autobiographies, writing allows students to become authors of their own trajectories, and to trace the socio-historical process in which they live and lived. In this dissertation, the proposal of autobiographical writing with two classes of the 7th grade of elementary school proved to be relevant due to three aspects: a) the involvement of the participants of 12 years with the creation of a personal text; b) in each account of the short life of the young there is a process of fleeting communication feeding the text; c) monitoring the reading of the set of texts allows the reader to appreciate the objects, places and person brought by the students that count quick biographies and build the uniqueness of the youth pathways. Thus, the teaching of written production can be enhanced by the study of the discursive genres that turn to the student\'s self. The main objective of this dissertation is to analyze the singularity and the discursive memory of young people of 7th grade. Through texts produced during a didactic sequence, the aim is to identify the linguistic-discursive procedures used in the structuring of texts, verifying the categories of time and space used in the reorganization of the life trajectory, to understand the value marks that constitute the identity of the students. Thus, two questions guide this study: 1. In what linguistic-discursive way do students reconstruct time and space? 2. What memorialistic elements emerge from autobiographical narratives? As theoretical assumptions, the concept of discursive genre, by Mikhail Bakhtin and the Circle and the autobiographical notion in the theoretical perspective of Philippe Lejeune and Leonor Arfuch with the purpose of deepening on characteristics of the autobiographical genre. From the analysis of the organized corpus with the production and re-elaboration of 42 essays, the results indicate the importance of the writing act on itself, which allows the student to expand his / her repertoire of world visions, since a possibility opens with subjectivity, even superficially because they are young authors. When they return to their lives, unique themes that they keep inside themselves become fundamental subjects that construct narratives like losses and displacements. This constant change is central to the identities of young people in the 21st century.

Currently, Portuguese language teaching has as a priority the study of typologies and textual genres. Therefore, some didactic materials produced in this area are guided by this study. The narrative textual type, being one of the first which the student has contact with, will be the focus of this research. The narration is present in several textual genres as stories, news, novels, reports, films and even in social networks and it is observed that there is an integration of these genres with varied types of semiosis, such as images and sounds. The Secretary of Education of the city of Rio de Janeiro has as guiding of its work a list which includes the skills and competences to be developed in elementary education in the 2nd segment. Two skills stand out throughout this teaching segment: to locate explicit information in a text and to distinguish a fact from the opinion regarding this fact. The students have presented difficulties in carrying out these activities. Thus, it is important that the student is able to identify facts in a narrative, since their exposure in this textual typology is frequent. Therefore, this research, based on the methodology of action research in the terms of Thiollent (2009), aims to develop in students the ability to retextualize the gender mythological tale to the news genre through the identification of facts. In order to achieve this goal, the proposal was elaborated based on the applications of Marcuschi (2010 [2001]) and Dell'Isola (2008). The work was carried out, first, with a 6th grade class, with students divided into groups; later, the work was repeated in the same class, in the following year, in the 7th grade, but with individual productions. In addition, both interventions applied are characterized by the study of the structure and characteristics of the textual genres tale and news. Then, a few steps were taken so that the retextualization process could happen. The methodological approach was based on the concept of a didactic sequence elaborated by Dolz, Noverraz and Schneuwly (2004) and it consisted of the reading of short stories of Greek Mythology as well as of news texts. From the reading of these tales, students should identify facts and select one that is relevant enough to be turned into news; then they would go to work on it. For a retextualization process to be successful, the information contained in the story should be retained in the produced text (news). The rewriting of the elaborated news was an activity considered to reach a suitable final text. These steps were fundamental for this work, since they collaborated for the construction and fixation of concepts and for the development of textual production competence. The results obtained were satisfactory and through them it was concluded that the process of retextualization was an excellent resource for working with the genres as well as for the appropriation of the outstanding abilities
Atualmente, o ensino de L?ngua Portuguesa tem como prioridade o estudo de tipologias e g?neros textuais. Portanto, alguns materiais did?ticos produzidos nessa ?rea s?o norteados por esse estudo. O tipo textual narrativo, por ser um dos primeiros com o qual o aluno tem contato, ser? o enfoque desta pesquisa. A narra??o se faz presente em v?rios g?neros textuais como contos, not?cias, romances, relatos, filmes e at? mesmo nas redes sociais e observa-se que h? uma integra??o desses g?neros com tipos variados de semioses, como por exemplo, imagens e sons. A Secretaria Municipal de Educa??o do Rio de Janeiro tem como norteador de seu trabalho uma lista de descritores na qual constam as habilidades e compet?ncias a serem desenvolvidas no ensino fundamental de 2? segmento. Dois descritores se destacam ao longo de todo esse segmento de ensino: localizar informa??es expl?citas em um texto e distinguir um fato da opini?o relativa a esse fato. Os alunos t?m apresentado dificuldades na realiza??o dessas atividades. Assim sendo, ? importante que o aluno saiba identificar fatos em uma narrativa, visto que a exposi??o deles nessa tipologia textual ? frequente. Portanto, esta pesquisa, com base na metodologia da pesquisa-a??o nos termos de Thiollent (2009), tem como finalidade desenvolver nos alunos a habilidade de retextualiza??o do g?nero conto mitol?gico ao g?nero not?cia por meio da identifica??o de fatos. Dessa forma, os alunos poder?o ser capazes de buscar informa??es e utiliz?-las de forma aut?noma e proficiente. A fim de alcan?ar tal objetivo, a proposta foi elaborada pautando-se nas postula??es de Marcuschi (2010 [2001]) e Dell?Isola (2008). O trabalho foi realizado, primeiramente, com uma turma de 6?ano, com os alunos divididos em grupos; posteriormente, o trabalho foi repetido na mesma turma, no ano subsequente, isto ?, no 7? ano, por?m com produ??es individuais. Ademais, ambas as interven??es aplicadas se caracterizam pelo estudo da estrutura e das caracter?sticas dos g?neros textuais conto e not?cia. Em seguida, alguns passos foram dados para que o processo de retextualiza??o pudesse acontecer. A abordagem metodol?gica foi baseada na concep??o de sequ?ncia did?tica elaborada por Dolz, Noverraz e Schneuwly (2004) e constou da leitura de contos da Mitologia Grega bem como de textos noticiosos. A partir da leitura de contos, os alunos deveriam identificar fatos e selecionar um que fosse relevante o suficiente para ser transformado em not?cia; em seguida, partiriam para a sua elabora??o. Para um processo de retextualiza??o ser bem sucedido, as informa??es contidas no conto deveriam ser mantidas no texto produzido (not?cia). A reescritura das not?cias elaboradas foi uma atividade considerada para atingir um texto final adequado. Tais etapas foram fundamentais para este trabalho, uma vez que colaboraram para a constru??o e fixa??o de conceitos e para o desenvolvimento da compet?ncia de produ??o textual. Os resultados obtidos foram satisfat?rios e atrav?s deles concluiu-se que o processo de retextualiza??o configurou-se como excelente recurso para o trabalho com os g?neros bem como para a apropria??o das habilidades destacadas.

Abstract:
The monograph is devoted to the study of the phenomenon of "discourse" from the perspective of its institutionality. The focus of research interest is sports discourse, presented in the form of a complex conceptual space with a particular genre-stylistic and pragmatic characteristics. As a material of study are sports articles, sports interviews and sports commentary, considered as genres of sports discourse, allocated according to criteria focus of the text and its function. The use of frame analysis, content analysis and conversational analysis have shown the peculiarities of representation of speech and thoughts of individuals, operating in the conditions of specific discursive practices. Abstract:
This paper proposes a temporal analysis for music metadata using a generative probabilistic model for collections the discrete datasets such as text corpora. This method is also a topic model that is used for discovering abstract topics from a collection of documents. The method is then applied to audio metadata and song lyrics extracted with Echo Nest® engine, Spotify® Lyrics Genius® API. Song data time series are generated by grouping data items by release date, genre and dominant topics (from LDA analysis). Using a technique from Network Theory we visualise how these topics, in this case, genres, are related to each other through time.

Abstract:
The submitted study addresses the topic of the current state of the opinion journalism and its genres in the Slovak periodical press. The author draws attention to the question of classification of the opinion journalism of a rational and emotional type from the genre categorization point of view and, simultaneously, reflects on its application in the present journalistic practice. This brings a certain rate of confrontation between the defined theoretical premises and their subsequent practical (non-)implementation. The main objective of the study is to clarify the presence of genres of analytical and literary opinion journalism stated by media theory in the environment of the Slovak periodicals. Presentation of the basic terminological axis and the related explication of journalism genres included in the opinion journalism constitute the secondary objectives of the paper. For the purposes of achieving the set objectives, the author uses methods of logical analysis of text in combination with discourse analysis. Similarly, she predicts the evident presence of the phenomenon of hybridization in the Slovak journalistic practice.

Abstract:
The article explores the key vocabulary of 2020 in the network space of Ukraine. Texts of journalistic, official-business style, analytical publications of well-known journalists on current topics are analyzed. Extralinguistic factors of new word formation, their adaptation to the sphere of special and socio-political vocabulary of the Ukrainian language are determined. Examples show modern impressions in the media, their stylistic use and impact on public opinion in a pandemic. New meanings of foreign expressions, media terminology, peculiarities of translation of neologisms from English into Ukrainian have been clarified. According to the materials of the online media, a «dictionary of the coronavirus era» is provided. The journalistic text functions in the media on the basis of logical judgments, credible arguments, impressive language. Its purpose is to show the socio-political problem, to sharpen its significance for society and to propose solutions through convincing considerations. Most researchers emphasize the influential role of journalistic style, which through the media shapes public opinion on issues of politics, economics, education, health care, war, the future of the country. To cover such a wide range of topics, socio-political vocabulary is used first of all – neutral and emotionally-evaluative, rhetorical questions and imperatives, special terminology, foreign words. There is an ongoing discussion in online publications about the use of the new foreign token «lockdown» instead of the word «quarantine», which has long been learned in the Ukrainian language. Research on this topic has shown that at the initial stage of the pandemic, the word «lockdown» prevailed in the colloquial language of politicians, media personalities and part of society did not quite understand its meaning. Lockdown, in its current interpretation, is a restrictive measure to protect people from a dangerous virus that has spread to many countries; isolation of the population («stay in place») in case of risk of spreading Covid-19. In English, US citizens are told what a lockdown is: «A lockdown is a restriction policy for people or communities to stay where they are, usually due to specific risks to themselves or to others if they can move and interact freely. The term «stay-at-home» or «shelter-in-place» is often used for lockdowns that affect an area, rather than specific locations». Content analysis of online texts leads to the conclusion that in 2020 a special vocabulary was actively functioning, with the appropriate definitions, which the media described as a «dictionary of coronavirus vocabulary». Media broadcasting is the deepest and pulsating source of creative texts with new meanings, phrases, expressiveness. The influential power of the word finds its unconditional embodiment in the media. Journalists, bloggers, experts, politicians, analyzing current events, produce concepts of a new reality. The world is changing and the language of the media is responding to these changes. It manifests itself most vividly and emotionally in the network sphere, in various genres and styles.

For the last three decades, the issue of translator’s gender and its representation in the target text has been actively researched in translation studies. Over the period there appeared numerous, sometimes contradicting views on markers of feminine / masculine / other types of speech, on whether the translator’s gender is revealed in the target text, and on the quality of translation depending on the translator’s gender. The present paper focuses on the translator’s gender markers in the target text. Taking into account the results of other linguists and my own observations, I consider the researched units being either definite or ambiguous markers of the translator’s gender. I want to bring to light gender differences in two Ukrainian translations (female translation by Natalia Tysovska and male translation by Viacheslav Brodovyi) of George R.R. Martin’s A Game of Thrones. The semantic, pragmatic and stylistic shifts in the target text conditioned by the translator’s gender and gender stereotypes blur the sense of the source text. Thus, such shifts should be regarded as unwanted changes and better be avoided.

A gap exists between research on learning and research on interest. Cognitive researchers rarely consider motivational processes, and interest researchers rarely consider cognitive process. However, it is essential to consider both since achievement and interest are in fact intertwined. In this paper we (1) discuss a theoretical model that intertwines cognitive and interest development, (2) describe how that model informed the development of educational materials, and (3) report on the results of the cognitive components of a randomized research study examining the impact of text genre on learning and interest. In our prior analyses, we examined the effects of text characteristics (i.e., narrative or expository genre) on situational interest. We found that students with higher levels of prior individual interest preferred the narrative versions of text whereas students with lower levels of prior individual interest preferred the expository versions of text. In this paper, we examine the impact of text characteristics on student learning. The results of this research showed that contrary to prior research, there was no significant difference in comprehension based on text characteristics. These results provide evidence that is possible to differentiate instruction based students' prior interest without sacrificing learning outcomes.

Abstract. Tomato yellow leaf curl virus (TYLCV), a monopartitebegomovirus, is one of the most devastating viruses of cultivated tomatoes and poses increasing threat to tomato production worldwide. Because all accessions of the cultivated tomato are susceptible to these viruses, wild tomato species have become a valuable resource of resistance genes. QTL controlling resistance to TYLCV and other begomoviruses (Ty loci) were introgressed from several wild tomato species and mapped to the tomato genome. Additionally, a non-isogenic F₁diallel study demonstrated that several of these resistance sources may interact with each other, and in some cases generate hybrid plants displaying lower symptoms and higher fruit yield compared to their parental lines, while their respective resistance genes are not necessarily allelic. This suggests that pyramiding genes originating from different resistance sources can be effective in obtaining lines and cultivars which are highly resistant to begomoviruses. Molecular tools needed to test this hypothesis have been developed by our labs and can thus significantly improve our understanding of the mechanisms of begomovirus resistance and how to efficiently exploit them to develop wider and more durable resistance. Five non-allelic Ty loci with relatively major effects have been mapped to the tomato genome using molecular DNA markers, thereby establishing tools for efficient marker assisted selection, pyramiding of multiple genes, and map based gene cloning: Ty-1, Ty-2, Ty-3, Ty-4, and ty-5. This research focused on Ty-3 and Ty-4 due to their broad range of resistance to different begomoviruses, including ToMoV, and on ty-5 due to its exceptionally high level of resistance to TYLCV and other begomoviruses. Our aims were: (1) clone Ty-3, and fine map Ty-4 and Ty-5 genes, (2)introgress each gene into two backgroundsand develop semi isogenic lines harboring all possible combinations of the three genes while minimizing linkage-drag, (3) test the resulting lines, and F₁ hybrids made with them, for symptom severity and yield components, and (4) identify and functionally characterize candidate genes that map to chromosomal segments which harbor the resistance loci. During the course of this research we have: (1) found that the allelic Ty-1 and Ty-3 represent two alternative alleles of the gene coding DFDGD-RDRP; (2) found that ty-5is highly likely encoded by the messenger RNA surveillance factor PELOTA (validation is at progress with positive results); (3) continued the map-based cloning of Ty-4; (4) generated all possible gene combinations among Ty-1, Ty-3 and ty-5, including their F₁ counterparts, and tested them for TYLCV and ToMoV resistance; (5) found that the symptomless line TY172, carrying ty-5, also carries a novel allele of Ty-1 (termed Ty-1ⱽ). The main scientific and agricultural implications of this research are as follows: (1) We have developed recombination free DNA markers that will substantially facilitate the introgression of Ty-1, Ty-3 and ty-5 as well as their combinations; (2) We have identified the genes controlling TYLCV resistance at the Ty-1/Ty-3 and ty-5 loci, thus enabling an in-depth analyses of the mechanisms that facilitate begomovirus resistance; (3) Pyramiding of Ty resistance loci is highly effective in providing significantly higher TYLCV resistance.

The article clarifies of gender identity stereotypes in modern media. The main gender stereotypes covered in modern mass media are analyzed and refuted. The model of gender relations in the media is reflected mainly in the stereotypical images of men and woman. The features of the use of gender concepts in modern periodicals for women and men were determined. The most frequently used derivatives of these macroconcepts were identified and analyzed in detail. It has been found that publications for women and men are full of various gender concepts that are used in different contexts. Ingeneral, theanalysisofthe concept-maximums and concept-minimum gender and their characteristics is carried out in the context of gender stereotypes that have been forme dand function in the society, system atizing the a ctual presentations. The study of the gender concept is relevant because it reveals new trends and features of modern gender images. Taking into account the special features of gender-labeled periodicals in general and the practical absence of comprehensive scientific studies of the gender concept in particular, there is a need to supplement Ukrainian science with this topic. Gender psychology, which is served by methods of various sciences, primarily sociological, pedagogical, linguistic, psychological, socio-psychological. Let us pay attention to linguistic and psycholinguistic methods in gender studies. Linguistic methods complement intelligence research tasks, associated with speech, word and text. Psycholinguistic methods used in gender psychology (semantic differential, semantic integral, semantic analysis of words and texts), aimed at studying speech messages, specific mechanisms of origin and perception, functions of speech activity in society, studying the relationship between speech messages and gender properties participants in the communication, to analyze the linguistic development in connection with the general development of the individual. Nowhere in gender practice there is the whole arsenal of psychological methods that allow you to explore psychological peculiarities of a person like observation, experiments, questionnaires, interviews, testing, modeling, etc. The methods of psychological self-diagnostics include: the gender aspect of the own socio-psychological portrait, a gender biography as a variant of the biographical method, aimed at the reconstruction of individual social experience. In the process of writing a gender autobiography, a person can understand the characteristics of his gender identity, as well as ways and means of their formation. Socio-psychological methods of studying gender include the study of socially constructed women’s and men’s roles, relationships and identities, sexual characteristics, psychological characteristics, etc. The use of gender indicators and gender approaches as a means of socio-psychological and sociological analysis broadens the subject boundaries of these disciplines and makes them the subject of study within these disciplines. And also, in the article a combination of concrete-historical, structural-typological, system-functional methods is implemented. Descriptive and comparative methods, method of typology, modeling are used. Also used is a method of content analysis for the study of gender content of modern gender-stamped journals. It was he who allowed quantitatively to identify and explore the features of the gender concept in the pages of periodicals for women and men. A combination of historical, structural-typological, system-functional methods is also implemented in the article. Descriptive and comparative methods, method of typology, modeling are used. A method of content analysis for the study of gender content of modern gender-labeled journals is also used. It allowed to identify and explore the features of the gender concept quantitatively in the periodicals for women and men. The conceptual perception and interpretation of the gender concept «woman», which is highlighted in the modern gender-labeled press in Ukraine, requires the elaboration of the polyfunctionality of gender interpretations, the comprehension of the metaphorical perception of this image and its role and purpose in society. A gendered approach to researching the gender content of contemporary periodicals for women and men. Conceptual analysis of contemporary gender-stamped publications within the gender conceptual sphere allows to identify and correlate the meta-gender and gender concepts that appear in society.

Abstract:
Mitochondria are the site of respiration and numerous other metabolic processes required for plant growth and development. Increased demands for metabolic energy are observed during different stages in the plants life cycle, but are particularly ample during germination and reproductive organ development. These activities are dependent upon the tight regulation of the expression and accumulation of various organellar proteins. Plant mitochondria contain their own genomes (mtDNA), which encode for a small number of genes required in organellar genome expression and respiration. Yet, the vast majority of the organellar proteins are encoded by nuclear genes, thus necessitating complex mechanisms to coordinate the expression and accumulation of proteins encoded by the two remote genomes. Many organellar genes are interrupted by intervening sequences (introns), which are removed from the primary presequences via splicing. According to conserved features of their sequences these introns are all classified as “group-II”. Their splicing is necessary for organellar activity and is dependent upon nuclear-encoded RNA-binding cofactors. However, to-date, only a tiny fraction of the proteins expected to be involved in these activities have been identified. Accordingly, this project aimed to identify nuclear-encoded proteins required for mitochondrial RNA splicing in plants, and to analyze their specific roles in the splicing of group-II intron RNAs. In non-plant systems, group-II intron splicing is mediated by proteins encoded within the introns themselves, known as maturases, which act specifically in the splicing of the introns in which they are encoded. Only one mitochondrial intron in plants has retained its maturaseORF (matR), but its roles in organellar intron splicing are unknown. Clues to other proteins required for organellar intron splicing are scarce, but these are likely encoded in the nucleus as there are no other obvious candidates among the remaining ORFs within the mtDNA. Through genetic screens in maize, the Barkan lab identified numerous nuclear genes that are required for the splicing of many of the introns within the plastid genome. Several of these genes are related to one another (i.e. crs1, caf1, caf2, and cfm2) in that they share a previously uncharacterized domain of archaeal origin, the CRM domain. The Arabidopsis genome contains 16 CRM-related genes, which contain between one and four repeats of the domain. Several of these are predicted to the mitochondria and are thus postulated to act in the splicing of group-II introns in the organelle(s) to which they are localized. In addition, plant genomes also harbor several genes that are closely related to group-II intron-encoded maturases (nMats), which exist in the nucleus as 'self-standing' ORFs, out of the context of their cognate "host" group-II introns and are predicted to reside within the mitochondria. The similarity with known group-II intron splicing factors identified in other systems and their predicted localization to mitochondria in plants suggest that nuclear-encoded CRM and nMat related proteins may function in the splicing of mitochondrial-encoded introns. In this proposal we proposed to (i) establish the intracellular locations of several CRM and nMat proteins; (ii) to test whether mutations in their genes impairs the splicing of mitochondrial introns; and to (iii) determine whether these proteins are bound to the mitochondrial introns in vivo.

Abstract:
Repair of DNA breaks is an essential function in plant cells as well as a crucial step in addition of modified DNA to plant cells. In addition, our inability to introduce modified DNA to its appropriate locus in the plant genome remains an important hurdle in genetically engineering crop species.We have taken a combined forward and reverse genetics approach to examining DNA double strand break repair in plants, focusing primarily on nonhomologous DNA end-joining. The forward approach utilizes a gamma-plantlet assay (miniature plants that are metabolically active but do not undergo cell division, due to cell cycle arrest) and has resulted in identification of five Arabidopsis mutants, including a new one defective in the homolog of the yeast RAD10 gene. The reverse genetics approach has identified knockouts of the Arabidopsis homologs for Ku80, DNA ligase 4 and Rad54 (one gene in what proves to be a gene family involved in DNA repair as well as chromatin remodeling and gene silencing)). All these mutants have phenotypic defects in DNA repair but are otherwise healthy and fertile. Additional PCR based screens are in progress to find knockouts of Ku70, Rad50, and Mre11, among others. Two DNA end-joining assays have been developed to further our screens and our ability to test candidate genes. One of these involves recovering linearized plasmids that have been added to and then rejoined in plant cells; plasmids are either recovered directly or transformed into E. coli and recovered. The products recovered from various mutant lines are then compared. The other assay involves using plant transposon excision to create DNA breaks in yeast cells and then uses the yeast cell as a system to examine those genes involved in the repair and to screen plant genes that might be involved as well. This award supported three graduate students, one in Israel and two in the U.S., as well as a technician in the U.S., and is ultimately expected to result directly in five publications and one Masters thesis.

Abstract:
Original objectives . 1. Test anti-fungal gene products for activity against Uncinula necator, Aspergillus niger, Rhizopus stolonifer and Botrytis cinerea. 2. For Agrobacterium transformation, design appropriate vectors with gene combinations. 3. Use biolistic bombardment and Agrobacterium for transformation of important cultivars. 4. Characterize gene expression in transformants, as well as level of powdery mildew and Botrytis resistance in foliage of transformed plants. Background The production of new grape cultivars by conventional breeding is a complex and time-consuming process. Transferring individual traits via single genes into elite cultivars was proposed as a viable strategy, especially for vegetatively propagated crops such as grapevines. The availability of effective genetic transformation procedures, the existence of genes able to reduce pathogen stress, and improved in vitro culture methods for grapes, were combined to serve the objective of this proposal. Effective deployment of resistance genes would reduce production costs and increase crop quality, and several such genes and combinations were used in this project. Progress The efficacy of two-way combinations of Trichoderma endochitinase (CHIT42), synthetic peptide ESF12 and resveratrol upon the control of growth of Botrytis cinerea and Penicillium digitatum were evaluated in vitro. All pairwise interactions were additive but not synergistic. Per objective 2, suitable vectors with important gene combinations for Agrobacterium transformation were designed. In addition, multiple gene co-transformation by particle bombardment was also tested successfully. In New York, transformation work focused on cultivars Chardonnay and Merlot, while the technology in Israel was extended to 41B, R. 110, Prime, Italia, Gamay, Chardonnay and Velika. Transgenic plant production is summarized in the appendix. Among plants developed in Israel, endochitinase expression was assayed via the MuchT assay using material just 1-5 days after co-cultivation. Plants of cv. Sugraone carrying the gene coding for ESF12, a short anti-fungal lytic peptide under the control of the double 358 promoter, were produced. Leaf extracts of two plants showed inhibition zones that developed within 48 h indicating the inhibitory effect of the leaf extracts on the six species of bacteria. X fastidiosa, the causal organism of Pierce's disease, was very sensitive to leaf extracts from ESF12 transformed plants. Further work is needed to verify the agricultural utility of ESF12 transformants. In New York, some transformants were resistant to powdery mildew and Botrytis fruit rot. Major conclusions, solutions, achievements and implications The following scientific achievements resulted from this cooperative BARD project: 1. Development and improvement of embryogenesis and tissue culture manipulation in grape, while extending these procedures to several agriculturally important cultivars both in Israel and USA. 2. Development and improvement of novel transformation procedures while developing transformation techniques for grape and other recalcitrant species. 3. Production of transgenic grapevines, characterization of transformed vines while studying the expression patterns of a marker gene under the control of different promoter as the 35S CaMV in different part of the plants including flowers and fruits. 4. Expression of anti-fungal genes in grape: establishment of transgenic plants and evaluation of gene expression. Development of techniques to insert multiple genes. 5. Isolation of novel grape specific promoter to control the expression of future antimicrobial genes. It is of great importance to report that significant progress was made in not only the development of transgenic grapevines, but also in the evaluation of their potential for increased resistance to disease as compared with the non engineered cultivar. In several cases, increased disease resistance was observed. More research and development is still needed before a product can be commercialized, yet our project lays a framework for further investigations.
