Abstract:
Large polycyclic aromatic hydrocarbons with molecular mass exceeding 278 Da are rarely object of study, in spite of the fact that some compounds from this group are proved carcinogens. These compounds belong to the group of persistent organic compounds and they are considered as dangerous environmental contaminants. Diploma thesis is focused on the evaluation of the occurence of large PAHs in technological and waste waters produced in black pitch processing in the DEZA Valašské Meziříčí factory. The method of their analysis based on reversed-phase liquid chromatography with non-aqueous gradient; UV-VIS and fluorescence detection will be used. This method will enable the study of changes of large PAHs distribution at various stages of waste water treatment process.

This paper deals with the monitoring of water contamination and sediments in the area of Olší - Drahonín. Firstly, the water contamination released into the river Hadůvka is evaluated. Secondly, the sediments in the river are monitored and the surface water flow is evaluated. Finally, the water treatment technology and sampling is described. A description of the separation methods and choice of standard methods is outlined.

Abstract:
This thesis was focused on the possibility of secondary utilization of waste material from the wine industry to remove heavy metals from wastewater. In the theoretical part, there were discussed mainly the problems of heavy metals pollution including methods of their removal and ICP-OES detection. Furthermore, the waste material from the wine production and the possibilities of its further processing were discussed there. The primary objective of the experimental part was to characterize the material by IR analysis, optimization of adsorption conditions, isotherms construction and determination of maximum adsorption capacities of white, red and chemically modified marc for Cd, Cr, Cu, Ni and Pb. From the parameters influencing the adsorption efficiency, the adsorbent load was optimized (m/V ratio = 0,02); pH values found were 5 for Cd, Cu, Ni, Pb and 3 for Cr and the contact time to reach the equilibrium was 15 minutes for Cd, Ni, Pb and 30 minutes for Cr and Cu. Maximum adsorption capacity for heavy metals were calculated from adsorption isotherms by applying the Langmuir model and found to be 18,829 7 mg/g for Cd; 10,664 7 mg/g for Ni; 35,602 0 mg/g for Pb (modified marc with 1M NaOH); 4,678 4 mg/g for Cu and 9,629 0 mg/g for Cr. The results of the work confirm the good adsorption potential of heavy metals on marc and the positive effect of chemical modification of natural biosorbents to improve their adsorption capacity.

Abstract:
Until a few decades ago few people could imagine that the photovoltaic, solar thermal and other power based on renewable resources, will become a reality. Today people from all over the world on the contrary try at full blast derive benefit from of all possible available source. Using sunlight as a source of energy is first enforced only for small devices such as calculators for charging the battery, but now we are able to produced energy from the sun to supply people around the world. Of course it is not possible supply consumer sector plus firm only from performances renewable power supply. Therefore endeavour is derive benefit from classical energy production at the same time with others power supply. The basic components of photovoltaic and solar thermal power are panels. The panels are made of different materials in different shapes and sizes. During production, the resulting effect looks in addition to costs associated with production. For photovoltaic and solar thermal power plant requires sufficient sunlight. The sunshine has biggest intensity on south of ours planets. Therefore endeavour is build lump these power station just in stand with bigger intensity sunshine. One of them is just Cyprus, too.

Abstract:
The thesis describes time dependent fluorescence shift method and fluorescence correlation spectroscopy method (FCS) with its extensions FLCS, Z-scan FCS and dual-focus FCS applied on specific problems in DNA and lipid research. Compaction mechanism of a DNA molecule smaller than a resolution of a confocal microscope was elucidated. The process was revealed to be "all or non" for a polycation spermine as a condenser in contrast with the gradual compaction caused by a cationic surfactant. Biophysical properties of a phospholipid bilayer influenced by presence of oxidized phospholipids with truncated sn-2 chain were explored. The dynamics of hydrated functional groups in the headgroup region was proved to get faster while the hydration of the headgroup region increased. These effects are in relation with the reorientation of the short sn-2 chains observed in molecular dynamics simulations. Presence of oxidized species may also influence the lateral diffusion of the lipids - a slight increase of the diffusion coefficient was observed. Decrease of hydration and mobility in the headgroup region was found as an influence of heavy water on the phospholipid membrane. The diploma thesis deals with the determination of selected heavy metals in wastewater, while the first part describes electromigration methods and individual heavy metals. In the next part of the thesis is describe, the problems of sewage sludge and methods commonly used for determination of heavy metals in wastewater. The practical part of the thesis describes the development of the electrolyte system together with the use of computer technology. Following is a discussion of the possibility of removing heavy metals from wastewater. In conclusion, the results of heavy metal measurements, comparison with foreign literature and assessment of suitability of the method for analysis of hazardous substances at low concentrations in the environment are presented and commented.

Abstract:
In this thesis, water quality and the assessment of anthropogenic pollution in the sediments of the middle course of the Elbe River oxbow lakes Kozelská and Vrť were studied. It is widely accepted that the oxbow lakes are extremely significant ecosystems, However, a large amount of contaminated material may deposit in these lakes. The pollution probably comes from industrial sources of contamination from the second half of the 20th century. In addition, the oxbow lakes show the development of the riverbed, and contribute to the stability of the river ecosystem. The research of Lake Kozelská was chosen especially to its proximity to the chemical factory Spolana in Neratovice, which used to be the biggest source of pollution of the Elbe River. The research included bathymetric measurements, regular observations of hydrological regime, monthly analysis of chemical and physical parameters of water in the period from December 2016 to November 2017, and marginally microscopic analyses of phytoplankton and zooplankton species too. The next part of this research included grain analysis and determination of metal and arsenic concentrations in the sediment fraction of 20µm using Aqua Regia leaching and total decomposition as well. Abstract:
The aim of this dissertation thesis "Development of electrochemical detector for heavy metal monitoring" was to create sensible sensors for the determination of heavy metals in the aquatic ecosystem and which would be usable for real applications, determination of the content of various metals in waste industrial waters. For the simultaneous determination of Zn(II), Cd(II), Pb(II) and Cu(II), a carbon paste electrode modified with chromium oxide was developed, based on this modification were produced the screen printed electrodes for the simultaneous determination of said metals with good limit of detection. For the usability of printed sensors in situ, a new electrochemical cell design was designed. This proposed cell also facilitates dosing of samples and buffer necessary for electrochemical analysis. Further, for determination of the total chromium content in aqueous solutions, was introduced a new method using a silver film on a glassy carbon electrode. Finally, various methods for waste water treatment from heavy metals have been tested.
