This doctoral dissertation identifies a new sub-genre in Contemporary Spanish Peninsular Literature, the Spanish Campus Novel. The impetus for research was to ascertain whether or not the genre characterized the Spanish novels dealing with university life (SpCN). The texts in question build upon the British and American Campus Novel tradition while inflecting it with issues, styles and themes particular to Spanish literature. I examined nine examples of the Spanish Campus Novel (SpCN) to determine their distinctive characteristics: Carlota Fainberg, Antonio Muñoz Molina (1999); El inquilino (1989) and La velocidad de la luz (2005), Javier Cercas; Todas las almas (1989) and Negra espalda del tiempo (1998), Javier Marias; El enigma (2002), Josefina Aldecoa; Último domingo en Londres (1997), Laura Freixas; Mimoun (1988), Rafael Chirbes; and Soy un escritor frustrado (1996), José Angel Mañas. In spite of variances in the circumstances of the protagonists, the repetition of key elements created a justification for the academic novel classification. Chapter One reviewed criticism of the Anglo academic novel and established essential characteristics of the majority of the novels: campus location, academic protagonist, satire and humor, job-insecurity, political correctness and departmental politics. I reviewed the socio-political history of the Spanish university in order to contextualize the SpCN, both its paucity and its recent emergence. Chapter Two examines the works of Antonio Muñoz Molina and Javier Cercas; their protagonists share the commonality of living and working in the US. Chapter Three considers two novels of Javier Marías and how the author plays with the both the academic novel and fiction. Chapter Four reviews the novels by Josefina Aldecoa and Laura Freixas and the manner in which stereotypical professors (sexually predatory ones) imply certain cultural mores. Chapter Five investigates the lyrical novel of Rafael Chirbes and its contribution to the campus novel. Additionally, José Angel Mañas’ bleak comedy is investigated as unique, the only novel taking place in Spain. The conclusion summarizes the novels, the identified Anglo and Spanish characteristics and contextualizes the novels within current trends in recent Spanish Peninsular fiction. Lastly, an overview of four Latin American Campus Novels is suggested for further research

Cette thèse en création est consacrée au campus novel (qu’on pourrait traduire par «roman universitaire»). Son premier volet explore la nature et la portée de ce sous-genre romanesque issu des littératures britannique et américaine. Le deuxième, les fragments romanesques Sur une clôture, constituent un court campus novel franco-ontarien, fondé sur l’expérience d’un bachelier en lettres françaises à l’Université d’Ottawa. Un retour réflexif englobant les lectures de corpus et théoriques, et le processus de création lui-même, conclut le tout. L’ensemble de la démarche s’inspire largement du roman This Side of Paradise (1920) de F. Scott Fitzgerald, et donc cette thèse présente en quelque sorte un effort de réécriture accompagné de notions théoriques et critiques. Sur une clôture, tout comme This Side of Paradise, se présente à la fois comme une fiction librement inspirée de l’expérience personnelle de son auteur et comme un commentaire social et une satire du système d’éducation, au temps présent. À cet effet, l’esthétique comme le sujet de Fitzgerald (mais aussi ceux d’extraits de plusieurs autres romans universitaires) sont repris, voire pastichés, et sont ensuite étudiés dans la rétroaction de cette thèse. Il en résulte un aperçu de la réalité contemporaine, subjective quant à son auteur, notamment du bilinguisme en Ontario, de la culture franco-ontarienne, et du sentiment (ou ressentiment) de la génération des Millénaires.

This thesis has three main sections, the first a full-length novel entitled The Gift of Death, the third the bibliography and two appendices. The second contains variously: a preface; a critical exposition/analysis of the preceding novel with subsections, considering in conceptual fashion three central themes: death considered through symbolic, ideological and other meanings; a positioning of the academy in the ‘Campus’ novel sub-genre; and a socio-cultural analysis of fiction as a field of production and associated struggles for entry determined by class, origin and periodic cultural preferences. The Gift of Death concerns a sixty-year-old’s attempt to write a novel. Procrastinating English scholar, archetypal baby-boomer Jim Dent, revisits the thwarted ambitions of youth. Inspired by novelist Sue Townsend’s death, once a friend, Jim recalls knowing other aspirant artists—writers and film-makers— living and dying in obscurity. He reflects upon a troubled past, on unsatisfactory elements of the present and the increasingly daunting task of composition. The Gift of Death reworks the tradition of the campus or varsity novel, detailing lives tied to the rhythms of the academy. The chapters explore various eccentrics whose lives Jim traces through tentative, inadequate notes. Expanding such recollections the narrative includes: schooldays; postgraduate studies and school-teaching in Leicester; a voyage to interview Basil Bunting; and friendships with oddball alcoholics writers, Cedric and Challis, never satisfied or fully recognized creatively. Finally, overwhelmed by self-doubt, Jim abandons his Sisyphean task. Reflecting upon failure, an unexpected turn of events associated with visiting Bunting emerges in the present, offering resolution of sorts. The Gift of Death’s primary themes/contexts are: self-reflexive, multi-chronic form; death, loss and mourning; the baby-boomer generation; struggling for professional entry into the field of fiction; lost provincial and local creativity; the juxtaposition of past and present; loyalty, friendship and memory; parental conflict; and finally procrastination and disappointment.

Abstract:
In view of the fact that David Lodge’s campus novels are renowned for their ability to make light of traditional gender stereotypes as well as for their purportedly liberal, pro-feminist, intertexual, dialogical and metatextual dimensions, this article seeks to explore more precisely the strategic and unavowedly political functions humour and the comic fulfil in Changing Places, Small World and Nice Work. What will be demonstrated is that Lodge’s light-hearted, tolerant and at times even progressive liberalism is little more than an effect produced by the textual surface structure. In the case of Nice Work, this discrepancy between the surface and the deep structure leads to the paradox that while voyeuristic structures and male bonding are overtly ridiculed, on a deep structural level they are effectively reaffirmed. Though Lodge’s novels are at the level of their surface structure sustained by a logic which uses the “comic mode” as a more or less subtle form to critique traditional gender stereotypes, literary conventions, the British university system and British industry, ultimately his ‘Rummidge trilogy’ reinforces an aesthetically, morally and politically conservative subtext.

Abstract:
Value of leather produced from sheep is determined by its quality and looseness is one of the quality attributes that determines the value of the leather. As of now, looseness in sheep skin can be determined only after the tanning process is done and it is a long and expensive process to treat the looseness in skins after the tanning process. An interdigital sensor based sensing system has been developed which works on the principle of sensing technique based on interaction of electric field with the materials under test. Finite element software has been used for analysis and design of sensors. It has been reported that a good correlation was found between the actual looseness values and calculated looseness values.

"Les stéréotypes sont très répandus et bien ancrés dans l'esprit des gens. Pour caractériser leurs voisins, tous les peuples disposent d'un répertoire d'étiquettes appelées "stéréotypes". La quantité de segments "préfabriqués" est importante aussi dans le discours de tous les jours. Le locuteur est toujours traversé par le discours de l'Autre, la rumeur de la Société qui sous-tendent ses énoncés. A partir d'un examen des méthodes FLE en usage au Vietnam, nous proposons un état des lieux des approches qui ont construit la stéréotypie comme objet théorique et des perspectives qui placent l'analyse de la stéréotypie dans la problématique de la construction du sens en langue, en discours et en communication. Notre but est d'essayer de réhabiliter, de reconsidérer et de reconcevoir la notion de " stéréotype " qui fut négligée et rejetée dans l'histoire de la didactique des langues-cultures. Quelques perspectives didactiques concluent cette recherche"
Stereotypes are everywhere and well anchored in people's mind. In oder to characterize their neighbors, most eople have available a repectoire of labels called "stereotypes". The amount of pre-made language segments is also important in everyday speaking. In a dialogue, the speaker is always influenced by the other, rumor of society, which underline his/her wording. Following a carreful review of FLE methods in use in Vietnam, we are proposing to assess the current state of different approaches, which contribute to stereotypy as a theoretical object, as well as various percpectives, which put the analysis of stereotypy in the problematic sense of language in the speach and in the communication. Our goal is to try to rehebilitate, reevaluate and remodel the notion of "stereotype", which has been negleted and rejeted for most of the history of the didactic of the languages- cultures. Some didactic perspectives are concluding this research

It argues that the differences in these two issues mainly emerge from the difference in the two authors&

views of art. In the thematic analysis, on the other hand, the study focuses on the recurring themes in the two authors&

novels. It argues that Camus and Beckett explore similar themes in their novels because both writers belong to the absurd tradition. In other words, although their notions of art are different, their views of the human condition are quite similar, which is reflected in the common themes they explore in their novels.

L'objet de cette thèse est de mettre en évidence un des aspects les plus caractéristiques de l'évolution du roman en Afrique noire francophone : le renouvellement de la thématique et de la technique après les indépendances. Pour ce faire, nous avons effectué nos recherches dans une perspective comparatiste sur La plaie ou Malik Fall, visant la condition humaine au-delà du hic et nunc africain traditionnel, se serait prêté, selon certains critiques, à l’influence de l'humanisme camusien et occidental. S'il est vrai que l'auteur sénégalais a importé l'humanisme camusien dans la littérature française, quelle en est alors la raison ? Comment peut-on soutenir que le roman de Fall dérive de celui de Camus ? Peut-on affirmer, en raison de similitudes plus ou moins fortuites, qu'une œuvre dérive d'une autre ? Plus inquiétantes sont les questions suivantes : la littérature des colonises était-elle fatalement condamnée à s'inspirer systématiquement des littératures européennes (si tant est que camus fut un écrivain européen) qui leur fourniraient des modèles impérissables ? S'il est vrai que la littérature africaine ployait sous une relation ombilicale avec l'occident, le roman de Fall a-t-il contribué à consolider ou à entamer cette dépendance pour faciliter la naissance d'une littérature africaine libre de toute tutelle ? La première partie, descriptive, présente les repères biographiques des auteurs et l'originalité des deux romans du corpus. Nous avons fait de l'analyse des rapports de l'individu à la société le thème de la deuxième partie dont l'étude révèle la solitude et le manque de communication faisant des personnages principaux des antihéros monstrueux. Mais leur destin symbolise l'individuation humaine face à l'absurde de la vie qui fait plus ou moins désirer la mort. Ainsi montrons-nous dans la dernière partie que l'humanisme propose par camus puis par Fall est un humanisme tragique
This thesis of highlight one of the most characteristic aspects of the development of the novel in French-speaking black afrika: the renewal of themes and techniques after independence. To achieve this, we have conducted our research in a comparative perspective on La plaie where Malik Fall, viewing the human condition beyond the traditional African hic et nunc, was greatly influenced, according to some critics, by camusian and western humanism. If it is true that the Senegalese writer imported camusian humanism into African literat what was the reason for this? How can one argue that Fall's novel derives from camu's? Can one assert, on the basis of more or less fortuitous similarities, that one work derives from another? The following questions are even more worrying: was the literature of colonized peoples doomed to take its inspiration systematically from European literature (supposing Camus was a European writer) which would provide timeless models? -if it is true that African literature was labouring in an umbilical relationship with the west, did Fall's novel help consolidate or undermine this dependence and facilitate the birth of an African literature free from supervision? Part one is descriptive and sets out the biographical landmarks in the authors' lives and the original features of the novels of the corpus. Part two focuses on the theme of the relations of individuals and society, and the analysis brings the solitude and lack of communication which turn the main characters into monstrous anti-heroes. But their fate symbolizes human individuation in the face of the absurdity of life which, to a greater or lesser extent, underlies a death wish. We thus show in part three that the humanism proposed by Camus and fall is a tragic humanism

Il basso costo e la loro facilità di deposizione li rende ottimi candidati per lo sviluppo di una nuova generazione di rivelatori di radiazione ionizzante flessibile e scalabile su larga area. Altre caratteristiche avvalorano il loro possibile ruolo nei vari campi di applicazione medico, di fisica ad alte energie, sicurezza in aeroporto e installazioni nucleari o spaziali, quali la loro resistenza, leggerezza, flessibilità e la loro equivalenza ai tessuti umani in termini di assorbimento di fotoni ad alta energia. Alla base di questa tesi vi è lo studio e l’analisi di transistors organici a effetto di campo (OFET), con semiconduttore in TIPS e TIPGe -pentacene, come rivelatori di raggi X e la loro resistenza a danni al materiale e alla performance dovuti a esposizione a radiazione prolungata.

Amb el propòsit d'explorar el perfil intel·lectual de les noves generacions, es va fer una investigació amb joves mexicans, estudiants de batxillerat, a partir de la teoria de Joan Ferrés que descriu les característiques de la generació de l'espectacle, com a conseqüència de la seva exposició primerenca a la tecnologia i a un procés de socialització en el qual abunden les imatges i els estímuls sensorials que impacten en el desenvolupament cognitiu dels joves. Amb la finalitat d'explorar l'estil intel·lectual dels joves, es va fer servir com a marc de referència la teoria de les intel·ligències múltiples proposada per Howard Gardner i es van aplicar i analitzar els resultats de dos qüestionaris: un de percepció d'autoeficàcia amb relació al tipus d'intel·ligència i un altre de nivell de cultura digital. Per mitjà de la percepció d'autoeficàcia dels joves es va aconseguir determinar un perfil d'intel·ligències múltiples distintiu entre els clústers elaborats per a l'estudi, on es va trobar que els joves amb més nivell d'activitat tant digital com presencial van obtenir una autopercepció d'intel·ligències múltiples elevada en sis de les vuit intel·ligències i es van constatar perfils diferents d'activitat digital i d'intel·ligències destacades en els grups anomenats actius, passius i selectius amb relació a l'ús de tecnologia.
Con el propósito de explorar el perfil intelectual de las nuevas generaciones, se realizó una investigación con jóvenes mexicanos, estudiantes de bachillerato, a partir de la teoría de Joan Ferrés que describe las características de la generación del espectáculo, como consecuencia de su exposición temprana a la tecnología y a un proceso de socialización en el cual abundan las imágenes y los estímulos sensoriales que impactan en el desarrollo cognitivo de los jóvenes. Para explorar el estilo intelectual de los jóvenes, se tomó como marco de referencia la teoría de las inteligencias múltiples propuesta por Howard Gardner y se aplicaron y analizaron los resultados de dos cuestionarios: uno de percepción de autoeficacia en cuanto a tipos de inteligencia y otro de nivel de cultura digital. Por medio de la percepción de autoeficacia de los jóvenes se logró determinar un perfil de inteligencias múltiples distintivo entre los clústeres elaborados para el estudio, donde se encontró que los jóvenes con mayor nivel de actividad tanto digital como presencial obtuvieron una autopercepción de inteligencias múltiples elevada en seis de las ocho inteligencias y se constataron perfiles distintos de actividad digital y de inteligencias destacadas en los grupos denominados activos, pasivos y selectivos en cuanto al uso de tecnología.
In order to explore the intellectual profile of the new generations, research was carried out with young Mexican high school students. The work was based on Joan Ferrés' theory, which describes the characteristics of the generation of the spectacle, as a result of early exposure to technology and a socialization process in which images and sensory stimuli that impact the cognitive development of young people abound. To explore the intellectual style of young people, the theory of multiple intelligences proposed by Howard Gardner was taken as a reference and the results of two questionnaires were applied and analyzed. One of the questionnaires was about the perception of self-efficacy in terms of intelligence, and the other was about the level of digital culture. Through young people’s perception of self-efficacy, a multiple intelligences profile was identified from among the clusters developed for the study. This profile showed that young people with higher levels of both digital and face-to-face activity demonstrated high self-perception in six of the eight intelligences. In addition, different digital activity and outstanding intelligence profiles were found in the groups denominated as actives, passives and selectives in relation to the use of technology.

(In)tentar hablar del barroco es pre-pararse para no parar. In-tentando tantear y siempre fallar el gesto de tocar el sentido del que habla Jean-Luc Nancy. Una falla que no implica solamente un error, una falta, sino también un juicio. Es ese juicio el que en el barroco, y esto es lo que (me) resulta más atractivo del asunto, es un juicio trastornado, extravagante; un juicio des-viado que va extremando la errancia, siguiendo todas las vías, donde fallar puede ser un hallar; encuentro e incumplimiento. Fingimiento y fallo en que la apariencia impera por sobre la estructura,que pasa a ser secundaria. Despojada de su dominio afianzado en la función, resulta arrinconada por la apariencia que se eleva y se monta en la utilidad y el orden. Esta textualidad se excribe en y desde una temporalidad, no en lo atemporal de una esencialidad que con lleva un deber ser como deuda y obligación de un telos moderno. En consecuencia, señalaré la importancia del marco, pero de un marco entendido como el lugar anfibológico desde el que leeré las obras de arte, concebido como lugar que es también un no lugar, un párergon porque está con, en y contra el ergon. Abstract:
Introduction: 23 million patients worldwide suffer from heart failure. These patients depend on cardiac research, because cardiac research enables the development of new therapeutic strategies and –targets. In cardiomyocytes, the compartmentalization of cAMP and cGMP depends on many factors. T-tubuli and PDEs are responsible for the division of cells in microdomains in which localized and specific cAMP and cGMP-signaling occurs. The aim of this thesis was to develop a method to answer the open questions that remain about the physiological and pathophysiological significance of cAMP/cGMP compartmentalization. Methods: I used the zebrafish as a model, because the transparency of zebrafish larvae enabled non-invasive fluorescent imaging in cardiomyocytes in the living animal. I cloned the Fluorescence Resonance Energy Transfer (FRET) sensors EPAC1-camps for cAMP and cGi500 for cGMP and injected them into zebrafish fertilized embryos. Then I used the F0 generation for Fluorescence Lifetime Imaging (FLIM) -FRET-measurements of cAMP and cGMP. Ca2+ is an important downstream mediator of cAMP and cGMP, because Ca2+ regulates cardiac contraction. Therefore, I also cloned the Ca2+ sensor GCaMP6 and used the dye Fluo-4 AM to include intracellular Ca2+ in the imaging. Results: The cloned sensors for cAMP, cGMP and Ca2+ were successfully injected into the zebrafish and showed expression in individual cardiomyocytes. I developed a protocol to mount the living zebrafish embryos and to measure intracellular cAMP and cGMP with FLIM-FRET in vivo with high spatial resolution. I characterized the sensors in their functionality by showing that the sensors react to changes in intracellular concentrations of cAMP and cGMP. The results of this study include evidence that zebrafish have mechanisms that lead to cAMP/cGMP compartmentalization in the absence of T-tubuli, and these mechanisms keep compartmentalization constant even under extreme cAMP or cGMP increasing drug treatment. Furthermore, I imaged intracellular Ca2+ by confocal microscopy and developed a protocol to use Fluo-4 AM for Ca2+ imaging. Conclusion: The method used in this thesis should allow the investigation of subcellular cAMP/cGMP compartmentalization and Ca2+ and to subsequently answer open questions in the field, for example whether a change of cAMP compartmentalization leads to the pathological phenotypes of cardiac disease or if a changed compartmentalization of cAMP in cardiac disease influences Ca2+ concentrations and therefore contraction. Einleitung: Weltweit sind mehr als 23 Millionen unter Herzinsuffizienz leidende Patienten auf die kardiologische Grundlagenforschung angewiesen, da diese die Voraussetzung für eine bessere Versorgung durch adaptierte und neue Behandlungswege schafft. In Kardiomyozyten hängt die Kompartimentierung von cAMP und cGMP von vielen Faktoren ab. T-Tubuli und PDEs werden unter anderem für die Aufteilung der Zellen in Mikrodomänen, in denen lokalisierte und spezifische cAMP- und cGMP-Signalgebung stattfinden kann, verantwortlich gemacht. Das Ziel dieser Arbeit war die Etablierung einer Methode, mithilfe derer offene Fragen bezüglich der physiologischen und insbesondere der pathophysiologischen Relevanz der cAMP- und cGMP Kompartimentierung beantwortet werden können. Methode: Als Modell diente der Zebrafisch, da die Transparenz von Zebrafisch Embryonen eine nicht-invasive Bildgebung von Fluoreszenz in Kardiomyozyten im lebenden Tier ermöglicht. Dafür klonierte ich die Förster Resonance Energy Transfer (FRET) -Sensoren EPAC1-camps als cAMP-Sensor und cGi500 als cGMP-Sensor und injizierte diese in befruchtete Zebrafisch Embryonen. Anschließend benutzte ich die F0-Generation für Fluorescence Lifetime Imaging (FLIM) -FRET-Messungen von cAMP und cGMP. Da Ca2+ als wichtiger downstream Mediator von cAMP und cGMP die kardiale Kontraktion reguliert, klonierte ich außerdem den Ca2+-Sensor GCaMP6 und benutzte den Farbstoff Fluo-4 AM, um intrazelluläres Ca2+ darzustellen. Ergebnisse: Die klonierten Sensoren für cAMP, cGMP und Ca2+ konnten erfolgreich in den Zebrafisch injiziert werden und zeigten alle Expression in einzelnen Kardiomyozyten. Ich entwickelte ein Protokoll, dass die Fixierung von lebenden Zebrafisch Embryonen und nachfolgender Bildgebung von cAMP und cGMP mit hoher zellulärer Auflösung mit FLIM-FRET in vivo erlaubte. Ich konnte eine funktionelle Charakterisierung der Sensoren durchführen, indem ich zeigte, dass sie auf Konzentrationsänderungen von intrazellulärem cAMP und cGMP reagieren sowie zeigen, dass Zebrafische trotz fehlender T-Tubuli eine signifikante cAMP- und cGMP Kompartimentierung aufweisen, auch unter extremen Bedingungen nach Gabe von cAMP/cGMP stimulierenden Substanzen in hoher Dosierung. Ich konnte zudem subzelluläres Ca2+ durch konfokale Mikroskopie bildgebend darstellen und entwickelte ein Protokoll, um mit Fluo-4 AM eine schnelle Möglichkeit zu haben, Ca2+ mit in die Messungen einzubeziehen. Ausblick: Die in dieser Arbeit benutzte Methode bietet eine gute Möglichkeit, subzelluläre cAMP- und cGMP-Kompartimentierung und Ca2+ zu untersuchen und damit zum Beispiel die Fragen zu beantworten, ob eine veränderte cAMP/cGMP Kompartimentierung zu Herzkrankheiten wie Hypertrophie führt oder ob eine veränderte cAMP Kompartimentierung den zellulären Ca2+ Haushalt und damit die kardiale Kontraktion beeinflusst. Darüber hinaus kann das von mir etablierte Protokoll dazu genutzt werden, mehr über cAMP, cGMP und Ca2+ während der Regeneration im Herzen zu lernen, da der Zebrafisch über ausgeprägte Regenerationsfähigkeiten verfügt

Nos referimos con este concepto a un proceso que consiste en introducir en una misma persona una forma de pensar diferente a la que este tenía hasta un momento determinado, pero como la primera nunca se elimina del todo se produce un conflicto entre ambas. El objetivo de esto es lograr que el sujeto sea provechoso para quienes educan. Para lograr esto se utiliza una herramienta demasiado cotidiana en nuestros tiempos, la tortura.

ENGLISH ABSTRACT: As existent beings, we identify with the world through our thoughts and perceptions. Man is driven to seek meaning by the very complexities and contradictions of existence. As self-conscious beings, we cannot live without a sense of awareness and understanding. Creativity allows an individual to develop a unique understanding of the nature and destiny of man. This study draws attention to writers who were able to transcend their external environment and immerse themselves in a setting where man’s individuality is fundamental to living an authentic life. Camus, Dostoevsky and Kafka made every effort to live consciously and authentically. They believed that inwardness was not to be defined by an external, social setting, but rather through an intimacy of consciousness. This awareness and unveiling of being enables us to create meaning. These authors removed their social mantles and were willing to sacrifice acceptance in the pursuit of this cause. They believed that every man has a responsibility to live an individual and authentic life. This psychological and even physical isolation is not easy, however, and often causes much suffering. Using existentialism as a framework, this thesis will focus on solitariness, suffering and creativity, all of which point to the importance of individual consciousness rather than living a life of societal pressures and conformity.
AFRIKAANSE OPSOMMING: As lewende wesens identifiseer ons onsself met die wêreld deur middel van gedagtes en waarnemings. Die mens word gedryf deur die soeke na betekenis in die kompleksiteit en teenstellings van sy bestaan. As wesens met selfkennis kan ons nie leef met ‘n gebrek aan bewustheid en begrip nie. Kreatiwiteit laat die individu toe om ‘n unieke begrip van die aard en lot van die mens te ontwikkel. Hierdi verhandeling vestig die aandag op skrywers wat verby hul uiterlike omgewings kon uitreik en hulself kon indompel in ‘n mileu waar die mens se individualiteit grondliggend is om ‘n onvervalste lewe te lei. Camus, Dostoevsky en Kafka het alles in hul vermoë gedoen om bewustelik en suiwer te lewe. Hulle het geglo dat die innerlike nie gedefinieer kan word deur die uiterlike, sosiale omgewing nie, maar eerder deur ‘n intimiteit van bewustheid. Hierdie bewustheid en openbaring van bestaan laat ons toe om betekenis te skep. Hierdie skrywers het hul sosiale mantels afgewerp en was bereid om sosiale aanvaarbaarheid op te offer in hul strewe na hierdie doelwit. Hulle het geglo dat elke mens oor ‘n individuele en onvervalste lewe beskik. Die sielkundige en selfs fisieke afsondering is egter nooit maklik nie en het dikwels groot lyding tot gevolg. Met eksistensialisme as raamwerk sal hierdie tesis focus op afsondering, lyding en kreatiwiteit.

Abstract:
Presenting and applying an ideal developmental model for the classical existential hero, or main character, provides a functional paradigm for discrimination between essentialist and existential texts. In particular it allows for degrees of fine existential differentiation amongst the hero's acts of any literary work. The paradigm does so by making it possible clearly to discern and describe the "recuperation" that a reader must do to render an "impaired" text intelligible. The paradigm covers four phases of transformational activity by the hero, more or less successfully negotiated, depending on the given work under analysis; vacillation/bad faith; crisis/arrest; abrogation/nothingness; and nihilation/project choice. Only one of the three novels so analyzed, Camus' The Stranger, contains a hero, Meursault, who is able to engage this paradigm successfully. The other two novels, not generally associated with existentialism, Heller's Something Happened and Chopin's The Awakening, reveal important and explicable variations of the model, but neither finally gives an exemplary authentic hero. The value of this paradigm is the way it functions as a dynamic heuristics, as a template, to isolate and render meaningful the dimensions of the career of each main character of these works as an "existential murderer." After an introduction of the paradigm, the thesis analyzes the tragic suicide of Mrs. Edna Pontellier, the comic infanticide of Bob Slocum, and the tragicomic homicide of Meursault.

Abstract:
This thesis writes back against the representation of the island in colonial discourse as a tabula rasa, easily possessed and inscribed. In colonial narratives island possession also emerges as explicitly gendered: the male castaway seeks to control the permissive, feminised territory of the island. It is this interpretation of the island, this gendered process of possession and inscription, that my thesis interrogates and subverts. My critical thesis is an analysis of Robinson Crusoe and three of its revisionary texts –– J M Coetzee’s Foe, Jane Gardam’s Crusoe’s Daughter and Julieta Campos’ The Fear of Losing Eurydice. I conduct a ‘ground-clearing’ exercise of Robinson Crusoe, critically remapping the island from a tabula rasa to an embodied construction of appetite and desire. My analysis of Foe and Crusoe’s Daughter explores how the texts’ women protagonists navigate the masculine terrain of Crusoe’s island, and how their authors negotiate an intertextual relationship with Robinson Crusoe. My analysis of The Fear of Losing Eurydice shows how Campos draws upon a discourse of desire to delegitimise linear narratives of colonial island possession. The desire for the island becomes a call uttered across multiple times and spaces, leading to Campos’ creation of the ‘archipelago of desire’; a motif which remaps Western insular cartographies to stress the relationality between the Caribbean, Asia, Europe, and Africa. I trace these cartographies of desire, charting the ways in which these novels disturb the terrain, and transgress the boundaries of Crusoe’s island. My novel, Bodeg, is set on a fictional island in the Arctic circle. Remote, desolate, bubbling with a molten undercurrent of menace, Bodeg is a volcanic island which has claimed the lives and minds of travellers. The novel follows Rebecca and her adult twins, Anna and Daniel, who visit the island. Rebecca is retracing the steps of an old love, Jake, whilst Anna seeks respite from a stale marriage. Daniel suffers from a mental health disorder that causes aural and visual hallucinations. Soon he becomes convinced the island is talking to him – that he understands Bodeg in a way nobody else can. But his sister and mother are too caught up in their own problems to notice. Rebecca’s search for answers produces only more questions. Meanwhile, Anna has become obsessively attracted to the strange, stormy Ferryman. Over the course of their trip the family find their lives intersecting with the island in ways that are both alluring yet deadly. In Bodeg they each find something they crave, not realising the island is wielding its own magnetic influence, steadily pulling them closer and closer and threatening to never let them go.

Constrói-se uma interpretação do processo de formação e consolidação do samba, na primeira metade do século XX, enquanto um campo, de acordo com a teoria de Pierre Bourdieu. Tendo sido originariamente sinônimo de festa improvisada, o samba é racionalizado e transformado em produto para se adequar às dinâmicas da indústria cultural. A partir desse ponto, vai se consolidando enquanto um campo no qual seus agentes disputam a legitimação. O sucesso do samba possibilitou a sua expansão, promovendo uma migração de outros artistas e públicos para esta arena. As disputas por capitais promoveram uma intensa disputa no interior do campo do samba, o que ocorreu principalmente a partir de diversas propostas de samba concorrendo entre si pela imposição do princípio legitimador nesta arena. A trajetória de Noel Rosa se coloca como um caso notável deste processo. Noel era um músico branco de classe média que entrou no campo do samba e buscou se legitimar de diversas formas. Desenvolveu uma estratégia multifacetada, na qual elogiava o malandro em determinados momentos e o atacava em outros. Na maior parte do tempo, tentou impor o rapaz folgado como alternativa ao malandro para possibilitar sua legitimação neste campo. O rapaz folgado era o malandro contido e inofensivo cujo perfil coincidiu com as propostas vitoriosas no campo do samba, que permitiram a nacionalização deste campo por volta dos anos 1940. The Thesis builds an interpretation of the process of formation and consolidation of samba in the first half of XXth century as a field, according to Pierre Bourdieu’s theory. Originally taken as a synonym of improvised party, the samba is rationalised and converted in a product to adapt itself to the dynamics of cultural industry. Then, it consolidates itself as a field in which the agents enter in a struggle for legitimation. The success of samba made possible its growth, promoting the coming of other artists and audience to this field. The search of capital promoted a intense dispute in the environment of samba as a field. This occurred mainly from diverse conceptions of samba in a struggle among themselves for the imposition of the legitimation principle in this field. Noel Rosa’s path its specially noticeable in this process. He was a middle class white musician that got into samba field looking to legitimate himself in many ways. He developed a multifaceted strategy through which he would sometimes complement the malandro and also attack him in other situations. Most of the time he tried to impose the rapaz folgado as an alternative to the malandro to make it possible his legitimating in this field. This rapaz folgado was the suppressed and harmless malandro whose profile coincided with the successful proposals in the world of samba that allowed the nationalization of its field in the 40s.

Affrontando lo studio della Letteratura Italiana risulta evidente l'omissione dai manuali, ad eccezione di Primo Levi, degli scritti di coloro che sono stati deportati tra il 1939 e il 1945 in seguito alle persecuzioni razziali. Il tempo trascorso dalla Shoah ha, invece, dimostrato la presenza di un ingente quantità di opere che costituiscono il corpus della Letteratura Concentrazionaria, ossia la letteratura dei campi di concentramento. Gli autori di questa letteratura sono poco conosciuti e considerati minori, ma la loro scrittura raggiunge spesso un'ineguagliabile altezza intellettuale, morale, stilistica. La narrazione dei loro ricordi traumatici è il risultato di precise decisioni: quali fatti raccontare, in che ordine cronologico, ma soprattutto attraverso quale forma. Questi testi concentrazionari, infatti, possono essere studiati da diversi punti di vista, quello più inusuale è la divisione per generi: racconto, autobiografia, saggio, romanzo, diario, lettera, poesia. Non sempre la distinzione fra questo o quel genere è netta, ma è molto significativo che a partire da una tragedia, quale l'Olocausto, si possa scegliere di raccontare la propria esperienza prestando attenzione al modo di esprimerla. È il primo segno che dimostra quanto anche di fronte al male l'uomo non perda il desiderio di ricercare il vero ed il bello.
Such a long time has passed since the end of the Shoah and it has become clear that there are a lot of works written in those years which now form the corpus of literature based on the experiences in concentration camps. The authors of these works are not very well-known and they are usually considered minor but their works have often reached highly intellectual, moral and stylistic results. The narration of their traumatic memories is the result of precise decisions, i.e. what to tell, in what chronological order, but especially in what forms. The texts based on the experiences in concentration camps can in fact be studied from different points of view. The most unusual is their study through genres: short story, autobiography, essay, novel, journal, letter, poetry. The distinction between one genre and another is not often clear-cut. What is interesting to underline is that in front of a tragedy, as the Holocaust was, it is possible to choose to tell one's own experience by paying special attention to the way of expressing it. This is evidence that in front of evil man does not ever lose the desire to look for truth and beauty.

In Portugal, as in other countries, a common characteristic of many of the novels published in the past twenty years is the recourse to a repertoire of literary devices that recall those of the historical novel.                 On the basis of critical studies published in the 1990’s, and in particular Fernández Prieto’s Poética de la Novela Histórica, this thesis undertakes a study of the Portuguese historical novel with the aim of defining, to the extent possible, the limits of this literary genre, and to examine in detail some of its intrinsic—and pertinent—semantic and pragmatic traits: the setting of a fictional narrative in the past, the introduction of historical persons and events and, not the least, the transtextual relationship between the fictional and the historiographic text.        Special attention is given to some of the more innovative forms of the contemporary historical novel, to highlight how by emphasising certain generic traits —that refer to the reader’s acquired knowledge— the author can compensate for the absence of others. Despite the inherent ”heterodoxy”, this device justifies grouping these novels under the general heading of the historical novel. The thesis explores the analogies between some of these innovative narrative strategies of the contemporary historical novel with transtextual practices characteristic of literature predating the 19th Century and by which a certain ”historical knowledge” is requisite for decoding the text. The thesis also establishes a set of generic traits that the historical novel shares with epic poetry. In light of these analogies, the thesis proposes a reading of Os Lusíadas as a historical poem. Notre travail porte sur le traitement de la question du mal dans des récits français publiés entre 1945 et 1962, soit entre deux procès ayant interrogé la spécificité du mal et de la culpabilité, celui de Nuremberg d’une part, celui d’Eichmann d’autre part. Ces écrits sont des récits de l’après publiés par des écrivains qui ont participé à la guerre (comme Simon ou Camus) ou qui n’en ont reçu que des échos lointains (comme Butor ou Bataille). Le fil conducteur de notre analyse s’appuie sur l’essai de Paulhan, Les Fleurs de Tarbes ou la terreur dans les lettres, lequel nous a permis d’étudier le mal non seulement dans son versant historique au prisme de la terreur nazie et de ses conséquences, mais aussi dans son versant littéraire, entendant alors le terme de terreur dans le sens métaphorique que lui accordent Paulhan et Sade avant lui. La terreur est contrebalancée en permanence dans ces écrits et dans notre travail par son opposé, la rhétorique, et engendre la constitution d’une génération de romanciers « antimodernes » qui accèdent à une modernité esthétique sans renier la tradition romanesque du XIXe. L’approche du mal en termes de terreur et de rhétorique nous permet ainsi de concevoir la spécificité du traitement du mal chez Sartre, Bataille ou dans la littérature de témoignage, mais également de réinterroger sur nouveaux frais le statut du Nouveau Roman au prisme d’une historicité souvent déniée par la critique et de marquer l’oscillation de ce groupe de romanciers entre terreur et rhétorique. Cherchant l’hermétisme, l’exception, les nouveaux romanciers sont par ailleurs en quête d’une rhétorique fédératrice, capable de refonder une communauté brisée par la guerre
Our work relates to the question of evil in French stories published between 1945 and 1962, therefore between two trials that have questioned the specificity of evil and guilt, namely the Nuremberg trial and the Eichmann trial. These stories, written after the war, were published by writers who either directly took part in the fight (like Simon or Camus), or simply had echos of it (like Butor or Bataille). The heart of our study is the essay of Jean Paulhan entitled Les Fleurs de Tarbes ou la terreur dans les lettres. This essay enabled us to analyse evil, not only in terms of History — by referring to the Nazi terror and its consequences — but also in literary and metaphorical terms, therefore considering the term ‘terror’ according to the definition of Paulhan and Sade. In both these stories and our work, terror is constantly offset by its opposite, rhetoric, which constitutes the basis for a generation of ‘anti-Modern’ writers who aim at esthetical modernity without denying the importance of the novelistic tradition of the 19th Century. The question of evil through the notions of terror and rhetoric leads us to specify the approach of evil in Sartre and Bataille’s novels, or in testimonies. These notions allow us to analyse the status of the Nouveau Roman from an historical perspective, often denied by critics, and to show the constant wavering of this group of writers between terror and rhetoric. The nouveaux romanciers are not only looking for exception, abstruseness, but they are also seeking a common rhetoric, capable of recreating a community otherwise broken by the war

Para analizar este proceso de mitologización me concentro en la concepciones de Herder, quien señala que una mitolgía es la fuente de una poética nacional. En este sentido, analizaré cómo el espacio del sertón y lo sertaneros son mitificado a través de las novelas hasta proyectar al sertón como el lugar de un origen brasilero, en un proceso de creación de un mito nacional.

In questa tesi è stato ricavato un modello matematico generalizzato della macchina asincrona con numero di fasi dispari e rotore a gabbia di scoiattolo, che tiene conto di un numero arbitrario di armoniche di campo al traferro. Tale modello ha permesso di creare, in ambiente Simulink, un blocco function generalizzato, atto a descrivere il comportamento della macchina asincrona nei diversi funzionamenti. In particolare, il modello permette di visualizzare le componenti armoniche che vengono create nello statore e nel rotore a gabbia. Tale blocco è stato utilizzato per simulare il comportamento di una macchina trifase riavvolta come nove fasi, alimentata a V/f=cost, ed è stato possibile fare un confronto tra le due, sottolineando il miglioramento che si è ottenuto da una configurazione multifase. Con l’utilizzo di una configurazione a nove fasi, è stato possibile constatare un abbattimento dell' armonica 5 e 7 delle correnti rotoriche rispetto alla configurazione trifase. Il modello ottenuto, ha permesso in poco tempo di confrontare ben 4 tipi di funzionamenti della stessa macchina, cambiando solamente un paio di parametri (numero di fasi e fattore di inclinazione). Sono state fatte infine prove sperimentali con controllo vettoriale sul motore nove fasi presente nei laboratori, andando ad analizzare gli andamenti delle correnti nei diversi transitori di velocità.

The subject of this work is analyses and interpretation of David Lodge´s campus novels. In the concrete, the diploma paper is focused on sources and elements of comicality, especially humour, satire and parody, in Lodge´s free trilogy Changing Places, Small World and Nice Work. The starting point of this diploma paper is characterisation of the genre of the campus novel, with the inclusion of the social situation in the period of the genesis of this genre, and the short outline of its representatives, with emphasis on the novel of Kingsley Amis Lucky Jim. In the following the work considers the ways of the realisation of comicality in the text and from this basis the practical part derives. The main aim is to point out the ways of usage of comic in Lodge´s campus novels and this purpose is achieved in the analysis of the particular texts.

Alison, the protagonist, is a 21-year-old cello student from Northwest Ohio who studies at the Cincinnati Conservatory of Music. Although she comes from a conservative Methodist upbringing, Alison has turned away from her beliefs and entered into a polyamorous relationship with Luke, a graduate student at the University of Cincinnati, and his wife Cheryl. Through this relationship, Alison has been able to explore her nascent bisexuality and enjoy the affection she has been denied by her reticent mother and absent father. While the members of the triad hold lofty principles about openness and trust, the practice of that relationship—and its ever-shifting unwritten rules—fail to live up to their ideals.

The Study of Interpersonal Relationship of the Personage in the Chen,Zhu-Lu’s Teenage Campus Novel Abstract In this modern society with prospect, the education is for everyone. The school enrollment rate is relatively high. As a result, the teenagers are equivalent to students of school. They spent most of their daily time on campus. Therefore, the campus has become a very significant place to influence the life of those teenagers. The teenagers interact with other on campus. Hence, the interpersonal relationship with teachers and their peers has become a very significant part of the school life. Chen, Zhu-Lu is the novelist with outstanding talent among Chinese novelists. He is likened the four heavenly kings in the Chinese novelists for teenagers. Chen’s creation often based on the campus as background of this novel. His realistic method of writing depicted the life of campus. The interpersonal relationships between the personage of teachers and the relationship between peers are his focus of his writing. This focus has drawn the attention of educational researchers. This study mainly used contextual analysis integrated with literature review to study Chen’s eight teenage novels. The purpose is to explore the teachers’ and students’ relationship and peer relationships on campus. This has revealed as the writing signature of Chen’s campus novel. The first chapter of this research is the introduction, which depicted the primary interests, purpose, coverage and framework. Also the research review discussed the formal researches done by other researchers. Therefore, the sophistication of the research can be reached. In the second chapter, Chen, Zhu-Lu’s life experience, characteristics of his personality, and his belief in creation were also discussed. Furthermore, eight novels of Chen’s are briefly introduced in this chapter. In Chapter three, factors that influenced peer’s harmony and conflicts were closely examined. Secondly, the other factors that caused conflict between peers and between teachers and students in Chen’s novel were also explored. Furthermore, the peer relationship was also closely examined. The final chapter, chapter five, the conclusion were stated as follows: (1) the realism method fully conveys social responsibility. (2) Compassion was integrated with chivalry spirits. (3) The intercept of ration and arts was also found. (4) They focus on the capture of appearance and plots. This study discovered that the relationship between teachers and students and students with peers depicted in Chen, Zhu-Lu’s teenage campus novels served valuable enlightenment both to the teachers and the teenagers. While getting along with schoolmates, conflict is evolved from friction because of different opinions, which teen novels continuously grope and develop on. This research takes seventeen texts concerning campus conflicts as samples, including The Eighteenth Emergency, Frindle, The Report, etc. to go into how children’s literature deals with teenagers’ frustrated and anxious sentiments caused from the conflicts, afterwards leads teenagers to fit themselves to growing journey. This research analyzes the texts to look into teenager s’ growing journey based on Joseph Campell’s “hero’s journey”, and find out the protagonists’ characters based on Carol S. Pearson’s “the heros-within archetype” . The study discovers that conflict issues in campus novels have three types, and they are same generation in opposing, teacher-student in opposition, and teacher-student in conversation. The first two stages emphasize teenagers’ build up their self values and adjust their relationships with people around. The third stage highlights school systems expand new subjects of debate. The conflicts frequently originate from doubts in young adults’ self values, so adults become the vital role when they are having adjustment of themselves. As a result, the research reaffirms the importance of family; not only conveys love can actually help young people beat the difficulties in their growing, but also extends this idea to adult readers. A contiguidade entre a ficção e a realidade sente-se desde os primórdios da literatura. Surgiu com as primeiras narrativas e nunca deixou de existir, tendo-se manifestado de diferentes modos ao longo do tempo. No séc. XIX, nasceu o género literário que privilegia a relação História / ficção: o romance histórico. Depois de um breve ponto da situação relativo à tradição do romance histórico na literatura europeia e portuguesa, analisamos o estatuto eclético do narrador e o modo como exerce a sua função aglutinadora da narrativa, mantendo o leitor cativo da diegese: Como lhe dispensa as informações? Que facetas assume? Como organiza a narrativa? É um narrador eclético que não abandona a narrativa e acompanha com carinho as suas personagens favoritas. A memória intertextual povoa o texto: Camões, Pessoa, Fernão Lopes, Shakespeare, o texto bíblico,… Os intertextos vêm enriquecer a narrativa através de remissões e sugestões que deixam no ar, insinuam e pedem ao leitor que faça também o seu exercício de leitura a completar o da escrita. Por outro lado, a memória vivencial é sobretudo a do Vento, entidade extremamente dinâmica, não só pela mobilidade que lhe é caraterística, mas também porque não deixa esquecer que é praticamente omnipresente e a sua memória remonta ao passado longínquo, proporcionando ao leitor uma narrativa dinâmica e cativante.
The contiguity between fiction and reality exists since the beginning of literature. It came up with the first stories and never ceased to exist, having been manifested in different ways, over time. In the 19th century, the literary genre that emphasizes the relationship between history and fiction showed up: the historical novel. After a brief report on the tradition of the historical novel in the Portuguese and European literature, we analyze the eclectic narrator's status and how he exerts his unifying role in the narrative, keeping the reader captivated by the diegesis: How does he provide the information? Which facets does he assume? How does he organize the narrative? He is an eclectic narrator who never abandons the story and accompanies with affection his favorite characters. The intertextual memory is present all over the text: Camões, Pessoa, Fernão Lopes, Shakespeare, the Bible, ... The intertexts come up to enrich the narrative with references and suggestions left in the air, insinuating and asking the reader to do also his reading task, completing the writing one. On the other hand, the main existential memory is the Wind’s, extremely dynamic, not only because of his mobility, but also because he never lets us forget that he is practically ubiquitous and his memory goes far back into the distant past, providing the reader a dynamic and enchanting narrative.

Andrew Clements is a best-selling author in America who has written well over 50 works. Clements creates picture books for children and writes novels for the adolescents. The most famous novel, which is also his first novel, is Frindle. It immediately became a best-seller right after being published and has won over 10 awards with copyright in more than 10 countries. Since the late 1960’s, American adolescent novels present a multiple appearance in the campus culture. The novel subject matters of Andrew Clements are mostly about challenging authorities. There are many topics for discussions about the campus and its humorous contents as well as the use of satire aiming at educational conditions. The writer observes the adolescents carefully and captures trifles in the campus to connect with various people and their funny stories with his proficient writing skills. What’s more, his further observation and sincere suggestions are implied in his humorous tones as well as his attempt to grant adult readers an access to the adolescents’ world and know about their thoughts and actions. The writer believes that such literary works have a positive value in the growing and enlightenment of adolescent readers and also make parents and teachers think more and be inspired in different aspects. From the comprehensive survey conducted in Taiwan, we can see that the literatures about Andrew Clements’s campus novels are quite few. The author takes the ten translated novels released in Taiwan as research category, explores the book subjects and reflects on the current education problems in Taiwan and the confusion of education for parents. The author also explores the conflicting subjects from the aspects of sociology and psychology and analyzes the writing style and character-shaping technique by using textual analysis. It also discusses the reason why and how writing styles attract readers and how to acquire inspiration from the novels. This study also provides reference opinions for campus novels writers in Taiwan as well as for future follow-up researchers.

This thesis aims to discuss the contemporary British novelist, David Lodge’s two campus novels—Changing Places: A Tale of Two Campuses, and Small World: An Academic Romance—and suggests him as a postmodern Menippean satirist by focusing on his narrative and metafictional techniques; his use of Bakhtinian definition on language and carnivalesque devices of Menippean satire. David Lodge is widely acclaimed as a writer of campus novels with a keen slant toward exploring the vice and folly in academic communities. Firstly, unlike the traditional realistic novelists, Lodge attempts to write his novels with the mixture of various narrative techniques, challenging the postmodernist concept of collage and hybridity. Secondly, Lodge not only endeavors to write the academic novel with a distinguished form but also makes an effort to diversify the traditional campus novel content—“the Cult of Oxbridge.” Thirdly, there is more zealous debate among critics about whether or not Lodge is a satirist than in all their discussions of other campus novelists combined. All these stimulating issues have helped motivate this study.

Abstract:
Tumor angiogenesis, the ability of tumor cells to stimulate blood vessel growth, is one the most critical steps of tumor progression. To support the growth of the expanding tumor, the “angiogenic switch” is turned on, which is often triggered by hypoxia (i.e., low oxygen)-mediated events such as expression of vascular endothelial growth factor (VEGF), causing normally quiescent endothelial cells to proliferate and sprout. An emerging picture of angiogenesis suggests that while governed by complex mechanisms, cell adhesion molecules (CAMs) plays a pivotal role in the regulation of angiogenesis. Our laboratory recently identified multiple previously unknown proteins including, transmembrane and immunoglobulin domain containing 1 (TMIGD1) and immunoglobulin-containing and proline-rich receptor 1 (IGPR1). Immunoglobulin-containing and cysteine-rich receptor 1 (IGCR1) represents the third remember of IGPR-1 family proteins. To investigate the expression and function of IGCR1, we have developed a rabbit polyclonal anti-IGCR1 antibody and demonstrated that IGCR1 is expressed in the endothelial cells of human blood vessels. To examine possible function of IGCR1, we have generated porcine aortic endothelial (PAE) cells over-expressing IGCR1. We demonstrate that IGCR1 expression in PAE cells inhibited cell proliferation and capillary tube formation as measured by colorimetric MTT and matrigel tube formation assays, respectively. In contrast, over-expression of IGCR1 in PAE cells inhibited cell migration as measured by wounding assay. Taken together, this study identifies IGCR1 as a novel regulator of angiogenesis. Given, angiogenesis is a highly coordinated cellular processes controlled spatially and temporally by a myriad of cell surface receptors and ligands, IGCR1 by modulating the rate of endothelial cell proliferation and migration, plays a significant role in the formation of blood vessels.

Abstract:
Adenylyl Cyclases (ACs) catalyze the formation of the key universal second messenger adenosine 3’, 5’-cyclic monophosphate (cAMP) from adenosine 5’- triphosphate. Cyclic AMP participates in several signal transduction pathways and is present in bacteria and higher and lower eukaryotes including higher plants. Previous studies in plants have shown a role for cAMP in signal transduction during e.g. the cell cycle, elongation of the pollen tube and stimulation of protein kinase activity. More recently cAMP has been shown to play a role in stress responses. Interestingly, cAMP has also been shown to regulate ion transport in plant cells. Here we used a similar strategy that led to the discovery of the first guanylyl cyclase in plants that was based on the alignment of conserved and functionally assigned amino acids in the catalytic centre of annotated nucleotide cyclases from lower and higher eukaryotes, to identify a novel candidate ACs in Arabidopsis (Arabidopsis thaliana K+ Uptake 5 and 7). ATKUP5 and 7 are homologous to K+ uptake permeases (KUPs) from bacteria and high-affinity K+ transporters (HAKs) from fungi. The AC activity was investigated by recombinantly expressing the ATKUP5 and 7 AC domain in vitro and by complementation of an E. coli AC mutant (cyaA). Furthermore, ATKUP5 was tested for its ability to functionally complement a yeast mutant deficient in Trk1 and Trk2 high affinity potassium uptake transporters. Site-mutagenesis in the AC domain was used to test the effect of both functions in each other. Furthermore, ATKUP5 was characterized electrophysiologically in HEK-293 cells to characterize the nature of this transporter. The localization of the ATKUP5 in Arabidopsis was examined using a Green Fluorescent Protein (GFP) fusion with the ATKUP5 to determine whether ATKUP5 is expressed at the plasma or tonoplast membrane. Arabiodpsis thaliana of the wild type, overexpressing ATKUP5 and atkup5 mutant lines were used to examine phenotypic differences.

The main purpose of this study was to adopt the youth novels and explore the sixth-graders’ critical thinking skills on the concept of authority in campus. The researcher adopted action research and used the examples from the daily lives and youth novels by critical thinking teaching strategies. The action research process of curriculum design included two cycles. The researcher chose a sixth graders’ class in an elementary school in Taichung by purposive sampling approach. The researcher started the courses from December 2015 to March 2016, a total of 20 weeks. There were 29 students that participated in this research. The researcher collected the datas by interviews, observations, teaching reflection notes, and analyzed the data to understand the process of the implementation of action research. The researcher interpreted the pupils’ performance and attitudes as well as the researcher’s professional development and reflections. The main conclusions are as following: 1. The teacher as researcher used the youth novels and examples from the daily lives to guide and help the pupils to improve their critical thinking skills on the concept of authority in campus. 2. The researcher used the interactive dialogue teaching can improve both the teachers’ and pupils’ sills of critical thinking and concept clarification. 3. The researcher used effective cooperatice learning through group discussions to encourage pupils to think critically. 4. The researcher increased the pupils’ active exploring and curiosity for knowledge by teaching of reading youth novels. 5. While the teachers led students’ explorations, they should clarify the issues steps by steps. 6. Accoeding to the results, the participants had positive attitudes while the teacher taught the critical thinking by youth novels. Finally, the researcher discussed the findings and suggested some points for those who using the youth novels to improve the pupils’s critical thinking skills in the future studies.

Abstract:
Introduction: 23 million patients worldwide suffer from heart failure. These patients depend on cardiac research, because cardiac research enables the development of new therapeutic strategies and –targets. In cardiomyocytes, the compartmentalization of cAMP and cGMP depends on many factors. T-tubuli and PDEs are responsible for the division of cells in microdomains in which localized and specific cAMP and cGMP-signaling occurs. The aim of this thesis was to develop a method to answer the open questions that remain about the physiological and pathophysiological significance of cAMP/cGMP compartmentalization. Methods: I used the zebrafish as a model, because the transparency of zebrafish larvae enabled non-invasive fluorescent imaging in cardiomyocytes in the living animal. I cloned the Fluorescence Resonance Energy Transfer (FRET) sensors EPAC1-camps for cAMP and cGi500 for cGMP and injected them into zebrafish fertilized embryos. Then I used the F0 generation for Fluorescence Lifetime Imaging (FLIM) -FRET-measurements of cAMP and cGMP. Ca2+ is an important downstream mediator of cAMP and cGMP, because Ca2+ regulates cardiac contraction. Therefore, I also cloned the Ca2+ sensor GCaMP6 and used the dye Fluo-4 AM to include intracellular Ca2+ in the imaging. Results: The cloned sensors for cAMP, cGMP and Ca2+ were successfully injected into the zebrafish and showed expression in individual cardiomyocytes. I developed a protocol to mount the living zebrafish embryos and to measure intracellular cAMP and cGMP with FLIM-FRET in vivo with high spatial resolution. I characterized the sensors in their functionality by showing that the sensors react to changes in intracellular concentrations of cAMP and cGMP. The results of this study include evidence that zebrafish have mechanisms that lead to cAMP/cGMP compartmentalization in the absence of T-tubuli, and these mechanisms keep compartmentalization constant even under extreme cAMP or cGMP increasing drug treatment. Furthermore, I imaged intracellular Ca2+ by confocal microscopy and developed a protocol to use Fluo-4 AM for Ca2+ imaging. Conclusion: The method used in this thesis should allow the investigation of subcellular cAMP/cGMP compartmentalization and Ca2+ and to subsequently answer open questions in the field, for example whether a change of cAMP compartmentalization leads to the pathological phenotypes of cardiac disease or if a changed compartmentalization of cAMP in cardiac disease influences Ca2+ concentrations and therefore contraction. Additionally, this method can be used to learn more about cAMP, cGMP und Ca2+ during regeneration in the heart, because the zebrafish cardiomyocytes can regenerate.
Einleitung: Weltweit sind mehr als 23 Millionen unter Herzinsuffizienz leidende Patienten auf die kardiologische Grundlagenforschung angewiesen, da diese die Voraussetzung für eine bessere Versorgung durch adaptierte und neue Behandlungswege schafft. In Kardiomyozyten hängt die Kompartimentierung von cAMP und cGMP von vielen Faktoren ab. T-Tubuli und PDEs werden unter anderem für die Aufteilung der Zellen in Mikrodomänen, in denen lokalisierte und spezifische cAMP- und cGMP-Signalgebung stattfinden kann, verantwortlich gemacht. Das Ziel dieser Arbeit war die Etablierung einer Methode, mithilfe derer offene Fragen bezüglich der physiologischen und insbesondere der pathophysiologischen Relevanz der cAMP- und cGMP Kompartimentierung beantwortet werden können. Methode: Als Modell diente der Zebrafisch, da die Transparenz von Zebrafisch Embryonen eine nicht-invasive Bildgebung von Fluoreszenz in Kardiomyozyten im lebenden Tier ermöglicht. Dafür klonierte ich die Förster Resonance Energy Transfer (FRET) -Sensoren EPAC1-camps als cAMP-Sensor und cGi500 als cGMP-Sensor und injizierte diese in befruchtete Zebrafisch Embryonen. Anschließend benutzte ich die F0-Generation für Fluorescence Lifetime Imaging (FLIM) -FRET-Messungen von cAMP und cGMP. Da Ca2+ als wichtiger downstream Mediator von cAMP und cGMP die kardiale Kontraktion reguliert, klonierte ich außerdem den Ca2+-Sensor GCaMP6 und benutzte den Farbstoff Fluo-4 AM, um intrazelluläres Ca2+ darzustellen. Ergebnisse: Die klonierten Sensoren für cAMP, cGMP und Ca2+ konnten erfolgreich in den Zebrafisch injiziert werden und zeigten alle Expression in einzelnen Kardiomyozyten. Ich entwickelte ein Protokoll, dass die Fixierung von lebenden Zebrafisch Embryonen und nachfolgender Bildgebung von cAMP und cGMP mit hoher zellulärer Auflösung mit FLIM-FRET in vivo erlaubte. Ich konnte eine funktionelle Charakterisierung der Sensoren durchführen, indem ich zeigte, dass sie auf Konzentrationsänderungen von intrazellulärem cAMP und cGMP reagieren sowie zeigen, dass Zebrafische trotz fehlender T-Tubuli eine signifikante cAMP- und cGMP Kompartimentierung aufweisen, auch unter extremen Bedingungen nach Gabe von cAMP/cGMP stimulierenden Substanzen in hoher Dosierung. Ich konnte zudem subzelluläres Ca2+ durch konfokale Mikroskopie bildgebend darstellen und entwickelte ein Protokoll, um mit Fluo-4 AM eine schnelle Möglichkeit zu haben, Ca2+ mit in die Messungen einzubeziehen. Ausblick: Die in dieser Arbeit benutzte Methode bietet eine gute Möglichkeit, subzelluläre cAMP- und cGMP-Kompartimentierung und Ca2+ zu untersuchen und damit zum Beispiel die Fragen zu beantworten, ob eine veränderte cAMP/cGMP Kompartimentierung zu Herzkrankheiten wie Hypertrophie führt oder ob eine veränderte cAMP Kompartimentierung den zellulären Ca2+ Haushalt und damit die kardiale Kontraktion beeinflusst. Darüber hinaus kann das von mir etablierte Protokoll dazu genutzt werden, mehr über cAMP, cGMP und Ca2+ während der Regeneration im Herzen zu lernen, da der Zebrafisch über ausgeprägte Regenerationsfähigkeiten verfügt.

Abstract:
The main topic of this diploma thesis resides in the comparison of the expressionist painting and the existential literature in connection with the motif of screaming. Despite the temporal and geographical distance of the two artistic movements (we are focused more profoundly in the German and Austrian expressionist painting, while the main literary work of this thesis, the existential novel The Fall comes from the great mind of a French writer, Albert Camus), the research aspires to prove the interconnection of the two movements and their common tendencies, based on the analyses of chosen themes, motifs, technics, and structure. We propose the motif of screaming as the main part of the comparison, as it constitutes an essential axis of the occidental art, as well as the crucial contact point between Expressionism and Existentialism. In spite of the bountifulness of the various interpretations this motif offers, we have chosen only selected ideas and concepts. As far as the formal structure is concerned, the work is divided into three chapters, where the first two are focused on the origins, a brief characterization of the main point of the two esthetics. Abstract:
Living organisms employ different kinds of mechanisms, to regulate the functions of genes or their products, which may help in maintaining homeostasis inside the cell or may help in fighting hostile environment in the case of pathogenic organisms. These mechanisms act at the transcriptional, post-transcriptional, translational, and post-translational levels. In order to understand the physiology of an organism, it is essential to obtain an in-depth knowledge of such mechanisms, in which several proteins participate in interlinked pathways. In this regard, the present study focuses on two such proteins: (i). the newly identified Fic (Filamentation induced by cAMP) protein; and (ii). NDK (Nucleoside Diphosphate Kinase), which had been studied for decades. Fic protein and NDK share several common features: (i). both use nucleoside triphosphate (NTPs) or nucleoside diphosphate (NDPs) or their derivatives as one of their substrates; (ii). they have been found to be involved in diverse cellular pathways, involving different types of substrates that form the second substrate of these proteins; (iii). both are ubiquitously present in all the living organisms - from bacteria to humans to plants. However, there is very little information on these proteins from mycobacterial systems, which include some major human pathogens, Mycobacterium tuberculosis and Mycobacterium leprae, which are the causative agents of Tuberculosis and Leprosy, respectively. In view of these reasons, in the present study, the structural and/or functional features of the Fic and NDK proteins from Mycobacterium tuberculosis, were analysed, as it might be of medical significance for effectively combating the pathogen. The Chapter 1 of the thesis contains the Introduction to the research work and Chapter 2 is on the overall Materials and Methods. The remaining chapters pertain to the data obtained on the structural and/or functional features of the Fic and NDK proteins from Mycobacterium tuberculosis. Chapter 3. Cloning, Expression and Purification of Mycobacterium tuberculosis Fic The role of FIC (Filamentation induced by cAMP) domain containing proteins in the regulation of many vital pathways, mostly through the transfer of NMPs from NTPs to specific target proteins (NMPylylation), in microorganisms, higher eukaryotes, and plants is emerging. In order to understand the biological role of FIC domain containing proteins in mycobacteria, the gene for the FIC domain containing protein of the human pathogen, Mycobacterium tuberculosis, MtuFic, was cloned, overexpressed, purified to homogeneity, and biochemically characterised. Neither the His-tagged nor the GST-tagged MtuFic protein, overexpressed in Escherichia coli, nor expression of Mtufic in Mycobacterium smegmatis, yielded the protein in the soluble fraction. However, the maltose binding protein (MBP) tagged MtuFic (MBP-MtuFic) could be obtained partly in the soluble fraction. Denatured-refolded protein was used for the antibody generation in mice and rabbit. The cellular localisation and secretion of MtuFic were characterised using the antibody. Chapter 4. Biochemical Characterisation of Mycobacterium tuberculosis Fic Sequence alignment with several FIC motif containing proteins, complemented with homology modeling on the FIC motif containing protein, VbhT of Bartonella schoenbuchensis as the template, showed conservation and interaction of residues constituting the FIC domain. MtuFic, possesses the critical His144 residue, in the characteristic FIC Motif, HPFREGNGRSTR (HPFxxGNGRxxR), spanning 144th to 155th residue. Site-specific mutagenesis of the His144, or Glu148, or Asn150 of the FIC motif, or of Arg87 residue that constitutes the FIC domain, or complete deletion of the FIC motif, abolished the NTP to NMP conversion activity. The activity of MtuFic was consistent with the biochemical activities hitherto reported for a variety of bacterial FIC domain containing proteins. Studies were also carried out on NMPylylation in the presence of eukaryotic proteins and eukaryotic and mycobacterial cell lysates. Although formation of NMPs from NTPs mediated by MBP-MtuFic could be detected, we could not identify any protein as the target substrate either in the human macrophage (THP1) cells or in the M. tuberculosis cells. VopSΔ30 (kind gift from Dr. Kim Orth), along with human G proteins as targets, were used as the positive controls. Various possibilities for the inability to detect a protein target substrate are discussed. Chapter 5. Transcriptional Analysis of Mycobacterium tuberculosis fic Gene (Mtufic) In parallel, in order to understand the transcriptional regulation of Mtufic, primer extension analysis was carried out. The Transcription Start Site (TSS; +1 site) of Mtufic were mapped under different growth/stress conditions, which tubercle bacilli encounter in human host. Mtufic got expressed mainly through two transcripts, T1 and T2, arising from two different transcription start sites (TSS). Putative promoter regions were cloned in a promoter probe vector, which expresses a GFP protein of very high intensity, in order to qualitatively detect the activity of the promoters. The half-life of the gfp mRNA was determined to be 4 min and therefore justifiably quantitated the Mtufic promoter activity by determining the gfp mRNA levels. The levels of Mtufic mRNA were two-fold higher under nutrient-depleted stationary phase of growth, as compared to the levels at mid-log phase. The activity of P1 and P2, as quantitated real-time using the short half-life gfpm2+ mRNA levels in Mycobacterium smegmatis transformants, showed that the activity of P2 was upregulated two-fold under nutrient-depleted stationary phase of growth, while that of P1 remained unaltered while of P1 and P2 were low under hypoxia. Co-transcription of Mtufic, with the immediate upstream gene, Rv3642c, of unknown function, was observed. Taken together, the data strongly indicated that the expression of Mtufic gets altered under nutrient-depleted and hypoxic conditions, which are the stress conditions experienced by tubercle bacilli in granuloma in tuberculosis patients. Chapter 6. Functional Characterisation of Mycobacterial FtsZ-NDK Interaction During the past few decades, our laboratory has been carrying out extensive molecular and functional studies on the cytokinetic protein, FtsZ, of different mycobacterial species, and of a variety of other mycobacterial proteins that are believed to be interacting with the cell division machinery. In this regard, in parallel to the work on MtuFic, we carried out work on the identification and characterisation of the proteins that interact with mycobacterial FtsZ. In this context, we found for the first time that the nucleoside diphosphate kinase (NDK), which can generate NTPs from ATP/GTP and NDPs, interacts with FtsZ and that the interaction was conserved across several mycobacterial species. Therefore, the FtsZ-NDK interaction was extensively characterised in vitro, using the recombinant, purified FtsZ and NDK proteins from different mycobacterial species. This novel finding on the interaction of NDK with FtsZ adds another role to NDK, namely in bacterial cell division.
