Abstract:
A novel protein kinase, the Esk kinase, has been isolated from an embryonal carcinoma (EC) cell line by using an expression cloning strategy. Sequence analysis of two independent cDNA clones (2.97 and 2.85 kb) suggested the presence of two Esk isoforms in EC cells. The esk-1 cDNA sequence predicted an 857-amino-acid protein kinase with a putative membrane-spanning domain, while the esk-2 cDNA predicted an 831-amino-acid kinase which lacked this domain. In adult mouse cells, esk mRNA levels were highest in tissues possessing a high proliferation rate or a sizeable stem cell compartment, suggesting that the Esk kinase may play some role in the control of cell proliferation or differentiation. As anticipated from the screening procedure, bacterial expression of the Esk kinase reacted with antiphosphotyrosine antibodies on immunoblots. Furthermore, in in vitro kinase assays, the Esk kinase was shown to phosphorylate both itself and the exogenous substrate myelin basic protein on serine, threonine, and tyrosine residues, confirming that the Esk kinase is a novel member of the serine/threonine/tyrosine family of protein kinases.

Esketamine (ESK) has been approved as a rapid-acting intranasal treatment for treatment-resistant depression (TRD). Although existing studies have investigated the efficacy of ESK in the 4-week induction phase, our knowledge about long-term ESK efficacy remains poor. The aim of this systematic review was to summarize the available data on long-term ESK efficacy for TRD. A systematic search was performed including articles in English, up to 31 March 2021. The search found 7 relevant studies, involving 1024 adult TRD patients. Continuing treatment with ESK after the 4-week induction phase may be associated with stable efficacy in relapse prevention among TRD patients. Conversely, the long-term antidepressant effectiveness upon discontinuation of ESK might be limited, although data from three studies had a moderate to high risk of bias. Overall, the results on the effectiveness of this compound in the long term are mixed. According to our findings, ESK treatment should be continued following the induction phase to reach a stable efficacy in relapse prevention, while the long-term antidepressant and anti-suicidal effects of ESK after discontinuation are inconsistent. Currently, the level of proof of ESK efficacy in long-term TRD treatment remains low and more RCTs with larger sample sizes and active comparators are needed.

Abstract. We study here the recently proposed measure of local geomagnetic activity called the IHV (Inter-Hour Variability) index calculated for the Eskdalemuir (ESK) station. It was found earlier that the ESK IHV index depicts an artificial, step-like increase from 1931 to 1932. We show here that this increase is due to the fact that the values of the magnetic field components of the ESK observatory stored at the World Data Center are two-hour running averages of hourly data stored in ESK yearbooks. Two-hour averaging greatly reduces the variability of the data which leads to artificially small values of the IHV index in 1911–1931. We also study the effect of two-hour averaging upon hourly mean and spot values using 1-minute data available for recent years, and calculate the correction factors for the early years, taking into account the weak dependence of correction factors on solar activity. Using these correction factors, we correct the ESK IHV indices in 1912–1931, and revise the estimate of the centennial change based on them. The effect of correction is very significant: the centennial increase in the ESK IHV-raw (IHV-cor) index in 1912–2000 changes from 73.9% (134.4%) before correction to 10.3% (25.3%) thereafter, making the centennial increase at ESK quite similar to other mid-latitude stations. Obviously, earlier long-term studies based on ESK IHV values are affected by the correction and need to be revised. These results also strongly suggest that the ESK yearbook data should be digitized and the hourly ESK data at WDC should be replaced by them.

Eruptive seborrheic keratoses (ESK) are rare in dermatology. They are usually inflammatory in nature and may be encountered as Leser-Trélat sign. ESK may also be simultaneously observed with hepatic angiomas, chemotherapy, segmental neurofibromatosis, HIV or erythrodermic pityriasis rubra pilaris, psoriasis, and drug eruption. ESK may be transient and self-healing. Others recede after successful treatment of the underlying disease. In some instances, seborrheic keratoses may follow an isotopic response and remain strictly restricted to sites of previous eczema, photo-exposition or tattoos. A patient with patch/plaque lesions of classic-type mycosis fungoides (MF) presented sudden ESK that were exclusively limited to the MF lesions. In conclusion, this patient combined an isotopic response and ESK.

We investigated the role of mothers’ references to mental states and behaviors and children’s emotion situation knowledge (ESK) in a prospective, cross-cultural context. European American mothers ( n = 71) and Chinese immigrant mothers ( n = 60) and their children participated in the study. Maternal references to mental states and behaviors were assessed at Time 1 when children were three years of age. ESK was assessed when children were 3, 3.5, and 4.5 years of age. Multi-group latent growth curve analyses were used to model children’s growth in ESK over time, as well as relations between mental state language and references to behaviors on children’s trajectories. Results indicated that maternal references to mental states were associated with concurrent levels of ESK for European American children, and change over time for the Chinese immigrant children. Maternal references to behaviors were negatively associated with concurrent ESK for both groups.

Abstract:
Abstract The Wilms’ tumor oncogene protein (WT1) is an intracellular, oncogenic transcription factor that is over-expressed in a wide range of leukemias and solid cancers. RMFPNAPYL (RMF), a WT1-derived CD8 T cell HLA-A0201epitope, is a validated target for T cell-based immunotherapy. We generated a high affinity, fully human IgG1 mAb specific for the RMF/HLA-A0201 complex. The mAb shows potent anti-leukemia activity both in vitro and in vivo in mouse models. Bi-specific T cell engaging antibodies (BiTE) have been used effectively to target cell surface proteins and kill cancers. We have developed a new, potent form of the ESK mAb that is a bi-specific T cell engaging antibody (BiTE) specific for tumor cells coexpressing the intracellular oncoprotein, WT-1 and HLA A0201. ESK-BiTE and an irrelevant control BiTE were constructed with ESK1 scFv or irrelevant ScFv on one arm, and anti-CD3 ScFv fragment as the other arm. The BiTE constructs were expressed in CHO cells. Both the ESK-BiTE and the control BiTE bind human CD3 T cells. The ESK-BiTE selectively bind to leukemia cells that express both WT1 and HLA-A0201. The ESK-BiTE activated human resting T cells and EBV-specific T cells retargeting potent cytotoxicity against WT-1+ HLA A0201+ human leukemia cells in vitro. In an NSG mouse xenograft model, injection of ESK-BiTE (20 ug/ml) twice a week, following I.V. administration of 2x107 human EBV-specific T cells, once a week, significantly inhibited the growth of a previously established disseminated HLA A0201+, WT-1+ human Ph+ ALL, BV173 expressing luciferase, as measured by bioluminescence imaging. In a second NSG mouse model mice injected I.V. with an aggressive human AML, SET-2, on day 0, were treated on day 4 with ESK-Bite for 6 days consecutively at 20 ug/day together with EBV-specific T cells given twice a week. In this setting, the ESK-BiTE and T cells resulted in undetectable leukemic growth for 14 days post-leukemia inoculation, with a minimal tumor burden detected by day 18, while all control groups showed massive increases in leukemia burden by day 14. Mice bearing SET-2 leukemia, that received ESK-BiTE and T cells also showed longer survival and delayed limb paralysis. As expected, the human T cells, which were EBV-specific, did not induce signs of GVHD in mice. Our data provide evidence that ESK-BiTE is a potent and specific therapeutic agent against aggressive human leukemias expressing WT1 and HLA-A0201. This is the first study showing efficacy of a TCR-like BiTE antibody targeting an intracellular tumor antigen expressed at low density. Background: A research on mood disorder pathophysiology has hypothesized abnormalities in glutamatergic neurotransmission, by suggesting further investigation on glutamatergic N-methyl-Daspartate (NMDA) receptor modulators in treating Major Depressive Disorder (MDD). Esketamine (ESK), an NMDA receptor antagonist able to modulate glutamatergic neurotransmission has been recently developed as an intranasal formulation for treatment-resistant depression (TRD) and for rapid reduction of depressive symptomatology, including suicidal ideation in MDD patients at imminent risk for suicide. Objective: The present study aims at investigating recent clinical findings on research on the role of the glutamatergic system and ESK in treating suicidal depression in MDD and TRD. Methods: A systematic review was here carried out on PubMed/Medline, Scopus and the database on U.S. N.I.H. Clinical Trials (https://clinicaltrials.gov) and the European Medical Agency (EMA) (https://clinicaltrialsregister.eu) from inception until October 2019. Results: Intravenous infusion of ESK is reported to elicit rapid-acting and sustained antidepressant activity in refractory patients with MDD and TRD. In phase II studies, intranasal ESK demonstrated a rapid onset and a persistent efficacy in patients with TRD as well as in MDD patients at imminent risk for suicide. However, some data discrepancies have emerged in phase III studies. Conclusion: The U.S. Food and Drug Administration (FDA) granted fast track and Breakthrough Therapy Designation to Janssen Pharmaceuticals®, Inc. for intranasal ESK in 2013 for treatment-resistant depression (TRD) and in 2016 for the treatment of MDD with an imminent risk of suicide. However, further studies should be implemented to investigate the long-term efficacy and safety of intranasal ESK.

Abstract:
Geomorphic, structural and chronological data are used to establish the late Quaternary paleoseismicity of the active dextral-oblique Northern Esk Fault in North Canterbury, New Zealand. Detailed field mapping of the preserved c. 35 km of surface traces between the Hurunui River and Ashley Head reveals variations in strike ranging from 005° to 057°. Along with kinematic data collected from fault plane striae and offset geomorphic markers along the length of the fault these variations are used to distinguish six structural subsections of the main trace, four dextral-reverse and two dextral-normal. Displacements of geomorphic markers such as minor streams and ridges are measured using differential GPS and rangefinder equipment to reveal lateral offsets ranging from 3.4 to 23.7 m and vertical offsets ranging from < 1 to 13.5 m. Characteristic single event displacements of c. 5 m and c. 2 m have been calculated for strike-slip and reverse sections respectively. The use of fault scaling relationships reveals an anomalously high displacement to surface rupture length ratio when compared to global data sets. Fault scaling relationships based on width limited ruptures and magnitude probabilities from point measurements of displacement imply earthquake magnitudes of Mw 7.0 to 7.5. Optically Stimulated Luminescence (OSL) ages from displaced Holocene alluvial terraces at the northern extent of the active trace along with OSL and radiocarbon samples of the central sections constrain the timing of the last two surface rupturing events (11.15 ±1.65 and 3.5 ± 2.8 ka) and suggest a recurrence interval of c. 5612 ± 445 years and late Quaternary reverse and dextral slip rates of c. 0.31 mm/yr and 0.82 mm/yr respectively. The results of this study show that the Northern Esk Fault accommodates an important component of the c. 0.7 – 2 mm/yr of unresolved strain across the plate boundary within the North Canterbury region and affirm the Esk Fault as a source of potentially damaging ground shaking in the Canterbury region.

Abstract:
An engineering geomorphological investigation of the Esk River catchment has been undertaken to quantify the relationships between the valley's geomorphic evolution, the many 1-10km2 deep-seated landslides present within the catchment, and a significant flood event that inundated the lower valley in April 1938. The identification of key geomorphic processes enabled the assessment of catchment's geomorphic stability, the development of generalised models for landsliding, and the delineation of a pre-disposed zone of instability. This information is then applied to assess key geomorphic controls on the flood event. The region lies within the forearc basin of the obliquely convergent Hikurangi Margin, and is underlain by soft, gently eastward dipping Pliocene marine strata. Structurally it is dominated by its close proximity to the Mohaka Fault, as well as two westward-dipping blind thrusts beneath the valley identified in this study: the Wakarara Fault – Trelinnoe Sector, and the Eastern Patoka Fault. Evidence from seismic reflection surveys indicates that these have both been active since the early Mangapanian (2.8 - 3.2 Ma), and an analysis of stream longitudinal profiles and plan form suggests limited displacement may have taken place within the last 10,000 years. A survey of rock mass defects within a representative sample area in the centre of the valley highlights four sub-vertical joint sets; conjugate sets strike 153° and 246°, and another sub-parallel to the folding strikes 033°. These defects correlate well with lineaments identified in aerial and satellite photographs and are attributed to extension of the sediments across the top of fault-propagated folds. The generally low power streams have exploited these defects and highly incised channels now run almost exclusively along them. Deep-seated landslides occur generally within the area of folding and their extents are defined by lineaments inferred to correspond to persistent joints in the rock mass. The slides are translational, and are facilitated by up to 80m of incision - ongoing since the abandonment of an extensive terrace inferred to be Ohakean (18-10ka) in age. Basal failure surfaces commonly dip at angles as low as 6°, and a combination of tectonically induced flexural shears sub-parallel to bedding and very low shear strength tuffaceous horizons are inferred to provide planes of sufficiently low shear strength to facilitate failure. While most deep-seated landslides appear active, there is no evidence to suggest they were substantially affected by recent major tectonic (e.g. 1931 Ms7.8 Hawke's Bay Earthquake) or climatic events (e.g. 1938 c.1 :1000yr Esk Valley Storm). The headwaters of the catchment are located on the Maungaharuru Range. This rises from 500m - l300m and provides baseflow to the Esk River. Extensive deep-seated landslides dominate the eastern face of the range. These are inferred to have been triggered by the removal of lateral support at the foot of the range following significant incision and denudation in the last interglacial c.125ka. A deep-seated gravitational slope deformation is proposed to extend to 1.2km below sea level, and provide a driving mechanism for the slides. While the 1938 Esk Valley flood was primarily a result of an exceptional three day storm event, suspended sediment load was also an important factor. This is inferred to have resulted primarily from channel erosion in soft colluvial sediments on the Maungaharuru Range. Combined with significant sediment load from shallow landsliding and possible tectonic subsidence preconditioning the lower reaches of the aggradational valley, this lead to c.1 m of silt being deposited in the lower reaches of the aggradational valley. Rapid stream incision in response to uplift in 1931 and aggradation in 1938 is returning the lower reach of the river to grade and decreasing the flood hazard.

Abstract:
Geomorphological and paleoseismic studies provide insight into the fault geometry and kinematics of a series of dextral northeast striking faults, including the Porters Pass, Hawdon, Bullock Hill, and Esk faults, in the South Island of New Zealand. These faults show post-glacial offsets that are significantly larger than predicted from co-seismic displacement - surface rupture length regressions derived from empirical relationships. Geomorphological mapping reveals slip rates as high as 9 mm/year for the Hawdon fault and Bullock Hill fault over an expected fault length of c. 140 km. Surface expressions of some parts of the studied faults are obscured by glacial gravels, indicating that blind faults are present in parts of the Sourthern Alps and may be the source for a component of a reported slip deficit in North Canterbury. Concluding from comparing scaling relationship results for the individual faults I hypothesize that the Porters Pass, Hawdon, Bullock Hill and Esk faults are segments of an incipient fault system that stretches from the western tip of the Porters Pass fault to the Hope fault, east of Hanmer Springs. Considering the location, similar strike and dextral deformation mode, I suggest that this 140 km long dextral strike-slip fault system marks the southernmost extension of the Marlborough Fault System resulting from the ongoing southward propagation of the Pacific-Australian plate boundary in New Zealand's South Island.

В работе рассматривается использование искусственных нейронных сетей для автоматической обработки высокоширотных ионограмм вертикального зондирования. Нейросетевые модели обучались различать типы слоев Esи F2 и определять их параметры: критические частотыобыкновенной волны для Es и F2 слоев (foEs и foF2), минимальные действующие высоты (hEs, hF2), наименьшую наблюдаемую частоту отражения от ионосферы (fmin), экранирующую частоту слоя Es (fbEs), высоту максимума слоя Esr, наибольшую частоту отражения от области F (fxI), предельную частоту рассеянной обыкновенной компоненты слоя F (foI), высоту и частоту для коэффициента M3000 F2 слоя (M3000f F2 и M3000h F2). Эффективность моделей оценивалась с помощью тестовой выборки, содержащей 30000 ионограмм. Точность определения слоёв Esи F2 наиболее эффективной модели составляет 91.1% и 91.9% соответственно. Средняя ошибка определения частот составляет от 0.27 до 0.66 МГц для разного типа частот; средняя ошибка определения высоты Es~8.5 км, высоты F2 ~30 км.

Abstract:
In this paper, a non-linear active disturbance rejection controller (NADRC) is designed and originally applied to a non-linear one-degree-of-freedom electrostatic actuator (ESA). The imperfections of micro-fabrication and micro-packaging result in the modeling uncertainties of ESA. In addition, the ESA is inherently unstable due to pull-in phenomenon. So our control goal is to overcome the pull-in instability of ESA and achieve 99.99% of its full travel range despite of the presence of uncertainty. The NADRC consists of an extended state observer (ESO) and a feedback controller. Two kinds of ESOs are developed in this paper. They are high gain ESO (HG ESO) and the ESO with Fal nonlinearity (FAL ESO). The NADRC is independent of accurate model information, and therefore is a suitable controller for the uncertain ESA. The NADRCs with two different ESOs are simulated on the nonlinear ESA. A comparison study is conducted between both ESOs in terms of control performance and stability. The simulation results demonstrate the controllers with both ESOs reach our control goal successfully. While the NADRC with HG ESO generates larger control effort than the one with FAl ESO, the former is more robust against parameter variations and disturbance than the later.

“Cuando se sospecha de la inminencia de la pérdida de algo es cuando se comienza a hablar de ese algo” (Laguillo, 1995:20). Hace más de 2.000 años los filósofos se preguntaban sobre qué ocurriría con las escuelas de filosofía si se perdiera el oficio de pensar. ¿Habría que preguntarse ahora si se está perdiendo el oficio de fotógrafo? Efectivamente, el oficio entendido como antaño ha cambiado desde la irrupción de las nuevas tecnologías. De hecho, la evolución de la técnica fotográfica se desarrolla continuamente con formas y usos profundamente distintos a los anteriores, lo que crea nuevos hábitos en la profesión. Esa superación de las cuestiones técnicas es algo que, sin duda, supone una ventaja que hay que considerar, puesto que lo que ahora empieza a diferenciar a los profesionales es más una cuestión conceptual que una destreza mecánica. Pero no se puede perder de vista que aún hoy el nuevo sistema digital fotográfico está recostado sobre todo lo que ya se hizo en el sistema argéntico. Por lo tanto, tampoco se puede afirmar que los métodos de trabajo hayan cambiado drásticamente. Este artículo no cuestiona que la fotografía digital haya supuesto un incremento de las ventajas sobre los inconvenientes del sistema anterior. Ni siquiera rebate que haya habido una superación tecnológica sobre los conocimientos técnicos de los que tan solo gozaban los maestros fotógrafos del sistema fotoquímico. Lo que se plantea en este estudio es la necesidad de defender un oficio fotográfico basado en el conocimiento profundo de la técnica para luego usarla como se quiera. Por eso, el fotógrafo actual se debe proponer que el control sobre ella resida en no dejar actuar sola a la cámara. Porque, por muy precisa y exacta que sea la construcción de su tecnología, la meta del fotógrafo es usarla con constancia hasta conocer su idiosincrasia y hacerla finalmente suya. Ese es el modo adecuado con el que lograr obtener un “negativo” idóneo que recree el clima en que se toma, en cuanto a que se tomamos eoISO),lidadó la fotografía y con el que alcanzar un buen trabajo posterior. Debido a la complejidad del asunto, se han seguido distintos métodos que han reforzado la hipótesis. Partiendo de una profunda revisión bibliográfica, de la experiencia del propio autor, realizando una comparativa entre las respuestas una batería de preguntas en torno a tres cuestiones -la esencia de la fotografía, el buen hacer y sus reglas y la irrupción del sistema digital-, mediante la técnica de los grupos nominales y en una serie de entrevistas con profesionales españoles directamente vinculados al sector fotográfico y que han vivido esta fase de tránsito entre lo fotoquímico y lo digital desde distintos canales (distribuidores, profesionales, galeristas, coleccionistas o teóricos, entre otros).

Resumen: La intención es exponer las ideas que llevaron a Le Corbusier a construir su último y único proyecto urbanístico “Chandigarh”;realizar su sueño de construir sobre una ciudad constituida por aquellos elementos prefigurados desde sus libretas, fotografías y textos de sus Viajes a Oriente, 1911 y Sud América, 1929. Le Corbusier encuentra en Chandigarh su ciudad imaginada, aquella configurada en base al constructo de la razón; montañas, paisajes, árboles, cielos, lagos y ríos, etc. y para ello diseñó un mapa en tres dimensiones que contuviese esa razón fundamental de todos sus años de trabajo: “El monumento de la mano abierta” es un lugar donde superpone esa geografía construida (la idealizada con la hallada), diversos dibujos han ido otorgándole en el tiempo esa capacidad de expresar la vocación del tiempo construido . La mano - además de serlo- ya no es más un símbolo o un “signo”, es un mapa topográfico que contiene sus propósitos esenciales. Chandigarh se construye sobre esa “ciudad imaginada” – siendo sus numerosos dibujos de montañas, ríos y lagos cartografiados desde los botes, trenes, y aviones, - Le Corbusier siempre estuvo a esperaba encontrar un lugar que coincidiera y encajara con todas esas condiciones naturales- alegrías esenciales- y ese lugar fue Punjab (India); allí es cuando se eclipsan realmente por primera vez todos sus elementos configurados. Las montañas del Himalaya y el Lago Suknha más que elementos geográficos - que constituyen los límites del proyecto- son esencialmente imprescindibles para comprenderlo. El monumento de la mano abierta se configura como un recurso ideográfico en la obra de Le Corbusier. Abstract: The intention is to present the ideas that led to Le Corbusier to build its latest and unique urban project "Chandigarh", realize his dream of building on a city made up of those elements foreshadowed from his notebooks, photographs and texts of his trips to East 1911 South America 1929. Le Corbusier in Chandigarh discovers his imagined city, that set based on the construct of reason; mountains, landscapes, trees, skies, lakes and rivers, etc. and for this he designed a three-dimensional map that contained the fundamental reason for all his years of work: "The monument open hand" is a place where overlaps that built geography (the idealized with found), various drawings have been giving at the time that ability to express the vocation of time built. Besides hand the be-is no longer a symbol or a "sign" is a topographic map containing the essential purposes. Chandigarh is built on the "imagined city" - remains his numerous drawings of mountains, rivers and lakes mapped from boats, trains, and planes - Le Corbusier was always expected to find a place to coincide and fit with all those naturales- conditions essential- there was joy and Punjab (India); that's when all set items are really overshadow first. Himalaya Mountains and Lake Suknha than geographic features - which form the boundaries of the project are essentially necessary to understand it.
