Abstract:
Hypotheses: Suicide was expected to be more likely amongst released prisoners than the general population. Risk factors would include being on remand, charged or convicted with a violent offence, being released from a local prison, having had mental health problems, misused alcohol/substances, a history of suicidal behaviour, and a poor level of post-release engagement with community services. Method: This case-control study identified all suicides and probable suicides between 2000 and 2002, committed by offenders within 12 months of their release from prison in England and Wales. One matched control was recruited for each case. Suicide rates per 100,000 person-years were compared with rates in the general population using the indirectly age-standardised mortality ratio. Information on case and controls was obtained from various official databases and locally held personal records. Logistic regression modelling was used to identify key factors related with an increased risk of post-release suicide. Findings: Out of 256,920 recently released prisoners, 384 offenders committed suicide within a year of release from prison, producing a suicide rate of 150 per 100,000 person-years. Seventy nine (21%) suicides occurred within the first 28 days after release. Released males were 8 times and females 36 times more likely to commit suicide than expected in the general population. Individuals with a history of alcohol misuse, a history of self harm and a psychiatric diagnosis were identified as at greatest risk of post-release suicide. Local prisons were associated with a 2-fold increase in offenders' risk of post-release suicide and suicides were more likely amongst those in contact with mental health services. Interpretation: Recently released prisoners are at a much greater risk of suicide than the general population, especially in the first few weeks after release. The risk of suicides in recently released prisoners is approaching that observed in discharged psychiatric patients. A shared responsibility lies with the prison, probation, health and social services to develop more collaborative practices in providing services for this at-risk group.

Abstract:
Plant roots have a dynamic relationship with the surrounding soil, which forms a vital interface for the terrestrial biosphere. Without a strong interface with soil, plants could not extract the necessary resources needed for growth. As a part of a multifaceted strategy, plant roots release a variety of high and low molecular weight compounds into the soil. This exudate is believed to increase water and nutrient uptake, form the first barrier of defence, and aid in the symbiosis with fungi and bacteria. This investigation reports on the identity and biochemistry of the polysaccharides released from the roots of several crops and one basal land plant, and explores their possible functions. Crops were grown hydroponically in order to isolate the polysaccharides released by their roots. After growth, the hydroponic media were screened with a library of monoclonal antibodies (MAb). The MAbs revealed the presence of arabinogalactan-protein (AGP), extensin, xylan and xyloglucan. Signatures of these polysaccharides were also determined by monosaccharide linkage analysis. By using anion-exchange Epitope Detection Chromatography, polysaccharides released into the hydroponic medium of the crops were separated for further immunochemical analysis. This analysis demonstrated that the polysaccharides released by wheat were part of a multi-polysaccharide complex, Root Exudate Complex 1 (REC1). A similar polysaccharide complex, formed of AGP-xyloglucan (REC2) was also found to be released by liverworts, which were not previously known to secrete polysaccharides. Novel soil analytics were developed in this study to decipher the effects of polysaccharides released by roots on soil aggregate status. Tamarind seed xyloglucan, xylan from birchwood, and isolated REC1 from wheat were each demonstrated to increase the abundance of soil aggregates, with REC1 shown to be most effective. This increase in the abundance aggregates may help plants to bioengineer the rhizosphere resulting in increased uptake of resources required for growth.

Abstract:
Tumour-associated macrophages (TAMs) have been shown to play an important role in tumour survival and progression. Thus, high numbers of macrophages in the tumour tissue are often associated with a poor prognosis. Identification of factors responsible for recruiting macrophages to the sites of different types of tumours might help to develop more effective cancer treatment. Burkitt's lymphoma (BL) is characterised by uncontrolled cell proliferation, high rate of spontaneous apoptosis and significant macrophage infiltration. Although BL cells undergo extensive apoptosis, in situ their corpses are cleared very effectively by macrophages infiltrating the tumour. It is now widely believed that dying cells are themselves able to release chemotactic molecules to ensure macrophage chemotaxis and subsequent clearance of their site of death. Previous work carried out in this laboratory identified fractalkine/CX3CL1 (FKN) released from dying BL cells to be an important player in macrophage chemotaxis to BL. Yet, these results have also indicated that FKN may not be the only chemokine involved in this process. Following from those observations, the first part of this work focused on examination of the potential role of monocyte chemoattractant protein-1 (MCP-1) in macrophage recruitment to BL. Despite the initial promising results, careful analysis of the data obtained by various techniques led to the conclusion that MCP-1 is, probably, not expressed by BL cells. Subsequently, effort was concentrated on understanding mechanisms regulating FKN processing during cell death. The studies performed before in this laboratory identified a new form of FKN to be present in apoptotic BL cells and showed that this is the form that is, most likely, responsible for mediating macrophage migration. Here, this apoptosis-related 60 kDa FKN was found to be a likely caspase-3 cleavage product. Moreover, it was demonstrated that FKN and active caspase-3 are released together in apoptotic BL cell-derived microparticles, suggesting that the proteolytic events could take place also extracellularly. In the final results chapter the differences between BL cell lines in the way they process FKN during cell death were revealed and a new cell death-associated 55 kDa FKN was observed. Through several lines of evidence, this new form was identified to be a possible product of calpain-mediated proteolysis. To conclude, this work provides the first evidence for a possible direct participation of the two major cell death executioner proteases – caspases and calpains, in production of ‘find me’ signals for macrophages and thus, ensuring effective clearance of dying cells. These results indicate that FKN cleavage and release might be of key importance during cell death. Moreover, the studies presented here contribute to better understanding of the process of FKN secretion.

The purpose of the project is to investigate if two new types of wood biofuels (pellets) are more or less reactive than the pellets previously investigated at Växjö University. To measure the released thermal power, an isothermal calorimeter with eight channels has been used. To see how the microbial activity is influenced, the pellets have been stored under various conditions with focus on temperature and metal.</p>

Abstract:
This study asked two questions. First, to what extent can metacognitive strategies instruction increase metacognitive awareness in released-time seminary students? Second, if metacognitive awareness is increased, is this increase correlated with changes in released-time seminary students' attitudes towards scripture study, their scripture study behavior, how they perceive the quality of their study, and how much they enjoy studying the scriptures? A control group and two experimental groups were used for this study. Experimental group 1 was taught basic scripture reading strategies without metacognition. Experimental group 2 was taught metacognitive strategies related to scripture study. Students in each experimental group used these strategies for 10 consecutive class sessions. Pre- and post-survey data was collected for comparison. Statistically significant gains in metacognitive awareness were found when comparing the pre- and post-survey scores of experimental group 2. When comparing experimental group 2 to experimental group 1 and the control group, analysis of covariance (ANCOVA) demonstrated that these gains in metacognitive awareness were not significant by comparison. Students in experimental groups 1 and 2 were asked to rate themselves in the post-survey on the effort they expended utilizing the strategies presented. When students in experimental group 2 who rated themselves high for effort were isolated, an analysis of covariance yielded statistically significant gains for metacognitive awareness in comparison with the other two groups. The same analyses were performed on measures related to scripture study. While experimental group 2 showed statistically significant gains from pre-survey to post-survey, when analyzed against experimental group 1 and the control group, no significant changes were observed. This was also the case for students who rated themselves high for effort in implementing the strategies presented. The results from this study suggest that metacognitive strategies can increase metacognitive awareness in released-time seminary students when they put forth the required effort to learn them. Further research in metacognitive application to scripture study is warranted. Qualitative studies with small focus groups could be a valuable avenue of exploration in future studies.

This thesis frames and initiates discourse about this unexplored phase of the terrorism continuum. Utilizing a grounded theory approach, we arrive at four findings: (1) we do not yet know to what extent convicted and released terrorists pose a threat; (2) convicted terrorists are treated no differently from an administrative or social standpoint from most other criminals; (3) the American public knows very little about convicted and released terrorists; and (4) there is no defined entity responsible for convicted and released terrorists. We then extrapolate the political, social, and legal implications of these findings, including whether our theoretical or structural frameworks are adequate to the threat. How might such a threat be measured or determined, by whom, and with what policy consequences? We look at existing models ranging from sex offender registries to megacommunities and existing sociological theories of terrorism as potential tools with which to address this complex and interdisciplinary issue.

The objective of this thesis was to assess the environmental fate and transport of chemicals emitted from consumer products through the development and application of modelling tools. The following hypotheses were tested: i) Multimedia fate models can be applied in a multistage assessment process to emerging chemicals when limited knowledge exists to identify the likely environmental fate and to direct further research; ii) the indoor environment acts as a source of anthropogenic substances in consumer products to the outdoor environment; and iii) chemical removal pathways in the indoor environment are important for the fate of organic chemicals in densely populated areas. The thesis shows that a structured chemical fate assessment strategy can and should be applied at early stages of the evaluation of emerging chemicals to assess their fate and to direct further research. Multimedia fate models play a key role in this strategy. The three‐solubility approach is a simple, rapid method that can be used to estimate physical‐chemical properties for use in early stage evaluation (Paper I). Emissions in the indoor environment affect the urban fate of hydrophobic organic chemicals by providing additional removal pathways and prolonging urban chemical residence times compared to outdoor emissions (Paper III). Emissions of BDE 209, DINP and DEHP to Stockholm indoor air were estimated to be 0.1, 3.4 and 290 mg/capita year, respectively (Paper IV). The contribution of emissions indoors to outdoor air pollution varies between substances. For BDE 209, emissions in the indoor environment added 38 % to the mass entering Stockholm city with inflowing air. For Sweden, the indoor environment was estimated to account for 80 % of BDE 209 emissions to outdoor air (Papers II and IV). For the phthalates, outdoor emissions and/or background inflow are the dominant sources to outdoor air pollution in Stockholm and the influence of the indoor environment is limited (Paper IV).<br><p>At the time of the doctoral defense, the following paper was unpublished and had a status as follows: Paper 4: Manuscript</p>

This is a qualitative research project about the experiences of forty-six former drug detainees who had been incarcerated because of illicit drug use. I examine their painful experiences of being arrested, imprisoned, and their experiences of post-released lives. I did the formal fieldwork in 2012. I have adopted a qualitative approach for my study. I stayed in Zhiyang for six months and Motai, another Chinese city, for one week in 2012 (both Zhiyang and Motai are not the real name of the cities). In 2013, I went back to Zhiyang again to reconfirm some of the data that I had collected. I had met forty-six former prisoners who were willing to share their stories with me. I have done semi-structured interviews with forty-three of them and participated in their formal and informal social gatherings. Behind their painful experiences, I would demonstrate, is a fundamental contradiction between the unrealistically ideal Party propaganda, which is made according to “exemplary norms”, and the everyday actual practices of the police officers and the prisoner officers. These realities are based on a variety of practical norms guided by different bureaucratic rules and regulations. Throughout the process, I will show, former drug detainees had suffered from physical pain, shame and degradation instead of being “rehabilitated”. Motivation: The resurgence of interest in controlled drug delivery reflects the increasing appreciation of the importance of local pharmacokinetics in defining the efficacy and potency of bioactive compounds. Despite promising data in vitro, growth factor use in vivo has generally failed to live up to its full potential. Without a theoretical framework to consider local pharmacology, there remains no consensus approach for improving the clinical outcomes of control released growth factors. As a result, a series of experiments incorporating cellular pharmacokinetics, computational modeling, and tissue pharmacokinetics were conducted. CellularPharmacokinetics: The effect of ligand-receptor trafficking was examined on the potency of sustained release versus bolus administration of two growth factors that differed by intracellular trafficking kinetics. Sustained delivery potency was demonstrated to be dependent on both ligand and receptor trafficking and could be predicted based on trafficking considerations.<br>(cont.) Computational Models: Computational models were conceptualized to describe the relationship between controlled-delivery, transport, receptor ligand pharmacokinetics, and tissue response. Theoretical predictions were made about potential approaches for optimizing local delivery and about the spatial correlation of tissue concentrations with tissue response. Tissue Pharmacokinetics: The roles of physiological transport forces and proteolytic constraints on control released growth factor were examined. Partitioning and convection were shown to be dominant forces in governing drug distribution. A novel method based on intramolecular fluorescence quenching was implemented to describe the proteolytic constraints of local fibroblast growth factor delivery in an ex vivo carotid explant model. Conclusion: Both experimental and theoretical models of cellular and tissue interactions of growth factors have suggested concepts that may be relevant for local growth factor delivery.<br>by David Wu.<br>Ph.D.

Abstract:
Tissue homeostasis is necessary for optimal organ functioning. The onset of tissue trauma compromises the homeostatic environment resulting in widespread cell death with the likelihood of exposure to invading micro-organisms. Early stage elimination of microbes and immunomodulation is co-ordinated by leukocytes of the innate immune system of which neutrophils and macrophages play a pivotal role. Leukocyte-released pro-inflammatory factors are vital in the containment of infection but bring with it a degree of collateral tissue destruction. Thus cascading stages during inflammation must be tightly regulated to bring about timely tissue regeneration and regained homeostasis. However, chronic inflammatory diseases e.g. rheumatoid arthritis highlights the existence of defective regulation at numerous stages during this transition, often leading to debilitating disease progression. Recently published findings by our research group identified the anti-inflammatory properties of α-defensin - an anti-microbial peptide released from dying human neutrophils - on stimulated macrophages. Thus the main objective of my research was to gain an understanding into the molecular actions of α-defensins which inhibit the macrophage inflammatory potential. Strong evidence supported the propensity of α-defensins to inhibit both intracellular and secreted protein synthesis, as assessed by de novo 35S-radiolabeled Methionine incorporation. Inhibition was not attributed to endoplasmic reticulum stress events, a common diagnosis in the regulation of global translation. Supporting evidence using cell-free systems identified a fundamental block in translation with the inclusion of α-defensin. Biochemical studies linked the ability of α-defensin to bind non-specifically to oligonucleotide sequences. This binding potential was also demonstrated on ribosomal RNA (rRNA), impeding its migration through electrophoretic gels. Immunocytochemical assays proposed an emerging suggestion of α-defensins in macrophages concentrated in close proximity to ribosomes around the perinuclear region. Evidence of suggested defensin/ribosome accumulation after 24hrs after treatment were attempted but to date remained unconfirmed. Attempts to determine the fate of these proposed accumulations were inconclusive, assessed by autophagy assays and ribosome semiquantitation. This thesis describes for the first time an enhanced understanding into the intracellular inhibitory mechanisms of α-defensins on macrophages and possibly other cell types. Understanding the molecular impact of α-defensins provide key insights into this novel inflammatory regulator, with the potential to be utilized in future immunotherapies.

Abstract:
Extracellular vesicles (EVs) are nano-sized particles constitutively released from cells into biological fluids such as blood, plasma, saliva and urine. Interestingly, these vesicles contain genetic cargoes including proteins, RNA and bioactive lipids that can be functionally delivered and affect recipient cells. Hence, EVs are postulated to be an alternative source of cell-cell communication in both normal physiological systems and pathological situations. Recently, EVs from mesenchymal stem cells have been shown to promote regeneration of injured cells. Induced pluripotent stem cells (iPSCs) are a relative newer type of stem cells that is emerging as useful tools for re-modeling diseases. However, knowledge about EVs from iPSCs is relatively sparse. In initial experiments, we successfully purified EVs from mouse iPSCs using the differential ultracentrifugation (UC) method. However, we noticed a discrepancy between total particle counts and expression of EV markers, across different EV batches. One crucial prerequisite for EV research is the availability of a standardized workflow for collection and purification of EVs from biological sources. Increasing evidence from recent studies has suggested that the original UC method is limited by several shortcomings such as low EV yield, purity and altered biophysical properties. Hence, this has led to a new wave in the development of alternative EV purification strategies. In this thesis, we start with a systematic comparison study between UC and an alternative purification protocol, size-exclusion liquid chromatography (LC) of EVs from serum-free conditions. We found that LC is better than UC in terms of overall EV yields, purity and vesicle integrity. Subsequently, we demonstrate that LC allowed for the derivation of pure EVs from complex media sources used for growing stem cells like iPSCs, which was previously impossible with UC. Lastly, we describe novel data on the characterization of EVs from pluripotent stem cell sources and discuss the possible roles of these EVs in stem cell biology.

Various methods, programs, and efforts to educate students in effective and efficient ways have been employed and studied for many years in the United States. Many teachers, administrators, and communities seek to gain a better understanding of and implement programs that will help achieve the academic goals of their respective organizations. Previous research indicates a correlation between some types of classes, programs, and characteristics of students and their academic achievement. Data indicating academic information for three hundred and fifty suburban secondary school students were collected and analyzed to support or refute previous research in this area of study. Grade point averages of these students were analyzed based on the characteristics of gender, age, core and non-core course selection, and enrollment in released-time religious education (RTRE) courses. Results indicated that selected characteristics of students were positively correlated with increased academic achievement. Such characteristics included: involvement in RTRE, the number of non-core courses taken, and the number of core courses taken. Students who were enrolled in fewer courses generally exhibited higher grade point averages. Results also indicated that the age and gender of students had little if any effect on their academic performance. (52 pages)

Community treatment of insanity acquittees is a highly controversial matter among legislators, mental health professionals and the public. At issue is the balancing of public safety concerns with least restrictive, non-punitive rehabilitation alternatives for insanity acquittees. Attempts to determine predictors of dangerousness in mentally ill offenders have produced mixed results with questionable practical value. In view of this, many mental health professionals are instead recommending that research on insanity acquittees be focused on evaluating the effectiveness of mandated community treatment programs for insanity acquittees. One area being designated for such research is the monitoring and management of psychopathology in conditionally released insanity acquittees, as mental deterioration in this population is a primary concern in assuring public safety. This thesis examined the monitoring and management of psychiatric symptomatology in a subsample of conditionally released insanity acquittees.

Hypertension (HTN) is the most common chronic disease among jail inmates. Many inmates treated for HTN while incarcerated in the Fairfax County Jail do not continue treatment when they return to their communities. Factors that contribute to discontinuing HTN management once the inmate returns to the community include homelessness, low income, and lack of access to care. The purpose of this quality improvement project was to educate inmates with HTN about community-based outreach services for HTN management and continuity of care while in the community. The outcome measured was the number of inmates who returned to jail reporting use of a community-based clinic for follow-up HTN care after their last release from jail. The project was conducted in 2 phases during a 6-month period. A pre-HTN survey questionnaire measuring HTN history and lifestyle was administered on initial incarceration. A post-HTN survey was completed when the inmates return to the jail during the 6-month period and measured adherence to post jail follow up HTN care. The findings of this quality improvement project indicate that both inmates who returned to jail in Phase 2 of the project followed up their HTN care in the community after release from jail. This project shows promise as a first step in the process of social change in planning discharge for inmates with HTN at the time of incarceration.

Objective: Bone invasion represent significant problem in managing head and neck cancers, however the mechanisms of interactions between oral squamous cell carcinoma (OSCC) and bone cells are poorly understood. We hypothesized that tumor cells can directly stimulate osteoclastogenesis. Methods: OSCC cell lines, bone-invasive BHY and metastatic but not bone-invasive HN were cultured and conditioned medium (CM) was collected. Osteoclast formation from RAW 264.7 mouse monocytic cell-line was assessed. Results: When RAW 264.7 were primed with receptor activator of nuclear factor κB ligand (RANKL) and then treated with BHY-CM, marked 2-6 fold induction of osteoclastogenesis was observed. In contrast, HN-CM did not significantly affect osteoclastogenesis. In addition, BHY-CM, but not HN-CM promoted survival of mature osteoclasts. Using pharmacological inhibitors, we found that Protein kinase C (PKC)/Extracellular signal-regulated kinase (ERK)1/2/Mitogen activated protein kinase (MAPK) p38 as well as Phosphatidyl-inositol 3-kinases (PI3K)/Serine/threonine protein kinase Akt/Mammalian target of rapamycin (mTOR) pathways mediate BHY-CM induced osteoclastogenesis. Conclusion: OSCC-cells produce soluble factors that stimulate osteoclastogenesis from RANKL-primed precursors. Tumor-derived factors act by stimulating ERK1/2 and p38 MAPK pathways in osteoclast precursors.<br>Objectif: L'invasion du tissu osseux est un problème majeur dans le traitement des cancers de la tête et du cou, cependant les mécanismes d'interactions entre le carcinome de cellules de squamous oral (OSCC) et les cellules du tissu osseux sont mal compris. Nous avons posé comme hypothèse que les cellules tumorales peuvent stimuler directement le phénomène d'ostéoclastogenèse. Méthodes: Deux différentes populations cellulaires de la lignée OSCC furent utilisées: les cellules BHY ayant un potentiel de colonisation du tissu osseux et les cellules HN ayant un potentiel métastatique mais non colonisant. Ces deux lignées cellulaires ont été cultivées et le milieu de culture conditionné (CM) a été collecté. La formation de cellules ostéoclastiques à partir de cellules de la lignée monocytaire de souris RAW 264.7 a été évaluée. Résultats: Une augmentation significative du phénomène d'ostéoclastogenèse d'un facteur 2 à 6 fut observée lors d'une activation des cellules RAW 264.7 avec RANKL suivit d'un traitement avec BHY-CM. De plus, la survie des cellules ostéoclastiques matures était favorisée en présence de BHY-CM uniquement. L'utilisation d'inhibiteurs pharmacologiques nous a permis de mettre en évidence que la stimulation du phénomène d'ostéoclastogenèse induite par BHY-CM est médiée par les voies de signalisations PKC/ERK/p38 et PI3K/AKT/mTOR. Conclusion : Les cellules OSCC produisent des facteurs solubles stimulant la formation d'ostéoclastes à partir de précurseurs activées par RANKL. Les facteurs dérivés de tumeurs agissent en stimulant les voies de signalisation ERK1/2 et p38 dans les précurseurs ostéoclastiques.

Umbilical cord blood (UCB) is a source of hematopoietic stem and progenitor cells (HSPC) used for allogeneic transplantation. Ex vivo expansion of HSPC can improve the slow platelet and neutrophil engraftment associated with UCB transplants. HSPCs reside in niches, some of which are near the endosteal bone surface, where they can associate with immature osteoblasts. Interestingly, osteoblasts can enhance the growth of HSPC in culture and their platelet engraftment activity. Using a proteomics approach, I identified 47 differentially expressed proteins between mesenchymal stem cells and immature osteoblasts. Several of these were previously implicated in HSPC maintenance such as IGF2, IGFBP2, DCN, GAS6 and VCAM1. Moreover, several other proteins belong to the alternative and classical complement pathways. Finally, I discovered that microvesicles found in osteoblast conditioned medium may also modulate the growth of HSPC, at least in ex vivo cultures.

Results are relevant to a wide range of natural and man-made applications, including the transport of oil droplets created accidentally during a deep-sea oil spill and sediment released purposefully during deep-sea mining operations. The subsequent transport and fate of small oil droplets with and without treatment using chemical dispersants is addressed experimentally and analytically. Next, we characterize the dependence of plume trapping behavior with crossflow velocity, bridging a gap in the current plume parametrization scheme. We also present a novel laboratory study aimed at understanding and quantitatively predicting secondary intrusion formation. In addition, a sensitivity analysis is conducted on plume trapping behavior to variations in key factors observed in the 2010 Deepwater Horizon oil spill field data. Another important environmental application of the study is deep-sea mining. Part of the work includes a field investigation to elucidate the trapping and dilution of mid-water plume consisted of fine sediment residuals from the mining operations, with the goal of assessing the potential environmental impact of this type of tailing disposal technique. Analytical and numerical model results are validated against field observations of the plume dynamics. Many practitioners commonly deal with implementing a change that is imposed by an organization. Some imposed changes require practitioners to alter more than what they do in practice but also to change their identity. Many researchers have studied identity change through the lens of sociocultural theory, specifically utilizing communities of practice theory (CoP). However, the majority of these studies used CoP theory as a vehicle to implement the imposed change. Yet some studies have found that after the trial period ends many practitioners revert back to the way they performed in practice prior to the study. One reason for this problem could be that the nature of the change experience that practitioners must undergo is not understood. The purpose of this phenomenological study was to reveal the nature of the change experience of six released-time seminary teachers in response to the adoption of the teaching and learning emphasis (TLE) within the Seminaries and Institutes of Religion (S&I) for The Church of Jesus Christ of Latter-day Saints. Specifically, the research question was "What is the nature and meaning of the change experience of a sample of released-time seminary teachers who are considered to be effective at learning to understand and implement TLE"? Researching S&I teachers' change experience is important because it may relate not only to the needs of the S&I organization, but also in a broader sense to the nature of the experience of practitioners who undergo an imposed change by the organization for which they work that alters their professional identity. Three central themes were found that represented the nature of the change experience for the sample group. The themes were represented as transformational, sociocultural, and self-reflective change. Each participant's experience varied in the particular details of his individual change experience. Nevertheless, all participants experienced some degree or kind of transformational change within their particular configuration of knowledge, character, and professional practice. Furthermore, all study participants engaged in sociocultural learning practices to facilitate their change. Finally, study participants experienced self-reflective changes.

Pollution spills are a serious threat to rivers causing fish kills, and lasting damage to the biological systems of rivers. At present financial penalties are used to deter such incidents but no mitigation system exists. A multiple method approach involving a hydrological and water chemistry based field study, a flume tank experiment, a computer fluid dynamics model and a 1D flow model was under taken to examine the feasibility of using a wave of water released from a reservoir to dilute pollution spills. Nine waves were released from reservoirs on three different catchments. Water quality was measured downstream of sewage treatment works and the progress of the wave was tracked down the river with gauges. Additionally in one experiment a slug of rhodamine dye was released into the river ahead of a wave. To understand the impact of a wave on mixing processes within the water column, in particular longitudinal dispersion, a series of flume tank and computer fluid dynamics experiments were ran. In both experiment sets a wave was released from one end of a tank and a slug of either rhodamine dye, or tracked particles was released mid tank, the interaction between the wave and the dye was then captured as footage and analysed. Across the three sets of experiments waves were found to move significantly faster than the baseflow with mean velocities ranging between 0.86ms-1 1.63ms-1. In the Holme River a dye slug would be caught within 3 hours and 46 minutes. Catch up times and response times were both demonstrated with a dye test and estimated with a 1D model providing management focused results previously unreported in the literature. Dilution of water quality parameters including NH4 and conductivity was recorded during wave passage at the sewage treat works outflows in the majority of experiments. Peak dilutions of 59% for NH4 and 58% for conductivity were recorded. An increase in longitudinal dispersion with wave magnitude was observed in the computer fluid dynamics model but unclear within the flume tank.

Apoptosis is a physiological form of cell death. It is a highly evolutionarily conserved process that is non-inflammatory or anti-inflammatory in nature. This anti-inflammatory nature of apoptosis is evident by the fact that neutrophils are histologically absent from sites where homeostatic apoptosis rates are high. The rapid phagocytosis of apoptotic cells as a means to prevent the release of noxious inflammatory compounds also accounts for the anti-inflammatory environment of such sites. However, the mechanisms that enable mononuclear phagocytes to migrate to sites where homeostatic apoptosis rates are high, and not granulocytes, the professional phagocytes that accumulate at sites of inflammation, have not been determined yet. Using Burkitt’s lymphoma (BL) as a model of apoptosis, the aim of this thesis was to identify the regulatory mechanisms or factors underlying the non-phlogistic features of sites where homeostatic apoptosis rates are high and in particular, those preventing the recruitment of neutrophils - a major granulocyte subclass to these sites. BL is a highly aggressive B cell lymphoma that is mainly characterised by a high rate of apoptosis. By carrying out a series of in vitro chemotaxis assays and biochemical approaches, it was found in this thesis that BL cells actively inhibit neutrophil migration by releasing factors that were identified to be lactoferrin, a 80 kDa iron-binding glycoprotein with anti-bacterial and anti-inflammatory properties. It was further demonstrated that lactoferrin selectively inhibited migration of granulocytes (both neutrophils and eosinophils) but not mononuclear phagocytes and this effect was irrespective of its iron saturation status and the chemoattractant used. Also, lactoferrin potently inhibited neutrophil migration, as assessed by thioglycollate-induced in vivo model of mouse peritonitis. This anti-inflammatory function of lactoferrin was attributed to its effect on granulocyte signalling pathways that regulate cell adhesion and motility. Finally, it was demonstrated that in cell types of diverse lineages, induction of apoptosis results in de novo synthesis and secretion of lactoferrin. In subsequent proliferation assays determining the in vitro growth of a number of BL cell types, it was demonstrated that lactoferrin is an essential component of BL cells and promotes their proliferation, as its antibody-mediated neutralisation or shRNA-mediated expression knockdown, reduced BL cell growth. Together, the results of this thesis identified lactoferrin as one of the few characterised antiinflammatory components of the apoptosis milieu that negatively regulate granulocyte migration. This effect may provide opportunities for broad therapeutic interventions concerning the use of lactoferrin in chronic inflammatory conditions characterised by aberrant neutrophil influx as well as atopic allergic disorders, such as asthma. Moreover, based on the tumour-supporting role of lactoferrin described in this study, targeting its expression in tumours could lead to tumour regression and thus, be a promising therapeutic molecule in tumour immunotherapy.

Forests have as a of late become increasingly plagued with bark beetle infestations as a result of climate change. The damage caused by tree killing bark beetles has within recent years seen a substantial increase. Detecting and removing infested trees at an early stage is an essential part of mitigating the spread of and the damage caused by the beetle. Today, the most common way of early detection is visual detection by forestry personnel. However, this is time consuming with highly variable results. In this thesis a novel approach to tracing the European spruce bark beetle through pheromone detection is investigated. With this approach, the antennae of the beetle were paired with an epitaxial graphene chip in order to create a bioinspired smell sensor. Tests were conducted on the sensor in order to investigate how the resistance changed over the chip as a result of the sensor being exposed to the pheromone 2-methyl-3-buten-2-ol. As a result of the tests, a corelation between exposing the sensor to pheromone and an increase of the resistance over the graphene chip was noted. However, more tests need to be conducted in order to draw any definite conclusions about the efficacy of the sensor in its current form. Additionally there are opportunities to investigate further optimization alternatives regarding the design of the sensor.

Since white-collar workers spend quite a long time sitting in a chair, they tend to have low back pain, ischium pain, muscle and skeleton injury. Although there are some pressure-release chairs on the market that can solve the problems, those chairs are not really user-friendly and still leave room for improvement. To create a more user-friendly pressure-release chair, the study would start by reviewing the disadvantages and inadequacies of the current products on the market based on the literature review and patent analysis. Previously our laboratory has successfully developed a manual pressure-release chair. Therefore, this study will focus on the development of integrated mechanism of eight to two. After integrating the mechanism with the pressure-release chair, the study would then attempt to compare the integrated electronic pressure-release chair with the manual pressure-release one in terms of stress evaluation. Regarding the result, the pressure-release effect of the electronic chair in this study is the same as that of the manual chair. Both of the chairs can effectively distribute the stress on ischium to thighs in stress evaluation, and allow the lumbar spinal curvature to show lordosis. Additionally, we also apply the integrated mechanism of eight to two to the pressure-release chair. Therefore, users can control the chair by motor, which is more comfortable and convenient operationally.

The polymers are used as materials in the two systems mostly, such as citosan、xanthan gum and hydroxypropylmethylcellulose (HPMC) etc. In reservoir system,using a fluid-bed spray granulator machine to sprinkle polymers on surface of tablet and forming a membrane. The membrane make drug's effect release slowly. In matrix system, using traditional method to form the tablets with the mixture by mixing the medicine with hydrogel or hydrophobic polymers. That is a cheaper and easier method than others. Many traditional drugs are made by dump system.In order to consider both safety and effect,development of oral slow-release tablets is a good decoration.There are many advantages in slow-release tablets, such as it can maintain the concentration of drugs in the circulation of blood,decrease side effect,decrease the frequency of using drugs and the squander of drugs,and further,it is a more acceptable mode for patients.

We document abnormally high foreign institutional trading volume beginning two days before recommendations are publicly released. Abnormal trading is related to recommendation characteristics that would require knowledge of the content of the report – such as whether the recommendation is upgrade or downgrade. Our results are consistent with institutional traders receiving tips regarding the contents of forthcoming analysts’ reports.

Abstract:
The issue of homelessness or inappropriate housing of people with complex needs is a serious problem for individuals who have found themselves frequently in the criminal justice system. Complex needs and challenging behaviours exhibited by those with mental illness, intellectual disability and or acquired brain injury combined with for example, family dysfunction, homelessness, poverty, and drug and alcohol misuse of a cocktail of issues that have been difficult to treat let alone support. Incarceration has been the alternative for the judiciary with the intention of protecting the individual offender as well as the community. However it is argued that proper support in the community may avert the need to seek incarceration as the only option for 'released prisoners' with complex needs. This thesis seeks to contribute to the understanding of what it means to be a person with complex needs and the effect of a short term sentence on the ability to seek stable accommodation once released from prison. It draws on the extensive literature in the field for example, with regard to people with disabilities in the criminal justice system and the housing experiences of ex-prisoners in general and those with complex needs. Through a number of in depth discussions with stakeholders from both state government and non-government representative agencies, a critical analysis was undertaken of the available accommodation options for this cohort in Tasmania. From these discussions it is evident that, current services either must embrace the needs of this cohort or a new dedicated service should be developed. However, the new Whole of Government and Community Sector housing initiative 'Housing Connect', commencing 1 July 2013, may offer the required support for this group of people with complex needs.

We made the herbs to the powder and be dripped into the cooling liquid and made the immediate-release form pellet. Later, with the advances in medical technology, we wanted pellet not only be used at immediate-release pellet form. For our research, we wanted to make extended-release form pellets enhancing the dissolution of poorly soluble drugs and release but keep preservative still staying in the pellet. At first, we referred to the past research and let our poorly soluble preservative and drug dissolve in different amount of agar to develop the optimize formulation of pellets and maintain pellets be sphere shape and smooth surface after freeze-drying, and increase the encapsulation efficiency of model drug. The pellet with agar may have influences in physical properties and dissolution of active constituent; for the prescription of pellet, the different methods to prepare agar gel may also have a difference in pellet’s properties. In our research, we found that if we let agar be gelled in refrigerator, the cooling rate could be faster, so it made agar covered methylparaben and agar covered indomethacin have harder quality. Then we could control agar size after homogenizing, and made these pellets have different release in the same time.
