ENGLISH ABSTRACT: The challenge facing managers and employees in the 21st century is to cope with the changing environment in which they find themselves. This study project aims to give managers guidelines to optimise human capital through emotional intelligence and, in so doing, assist them with a practical tool to challenge the modern business environment. Here the key question is: What does it mean to optimise human capital through emotional intelligence in practice? In an attempt to answer this question, the primary aim of this study project is to develop a practical and accessible tool which the modern manager can use in understanding and optimising the concepts of human capital and emotional intelligence, both in his personal and professional life. Gone are the days when managers and employees were told to leave their emotions at home. This study project also wishes to highlight the importance of emotions at work, how the management of emotions can significantly affect an individual's potential for development and promotion at work and, lastly, how different the workplace might be if everyone was helped to recognise and manage their emotional intelligence. The methodology used involves a thorough exploration of international and local literature, including books, articles and relevant electronic resources. Existing definitions, theories, text and themes regarding human capital and emotional intelligence are broken down into practical advice that is accessible to and can be understood by the modern manager. Thirty specific coaching guidelines are given that can be used as a management tool. The author attempts to anchor the study with reference to a South African case study. This case study involved testing the EQ of fourteen individuals from Media24 FamilyMagazines, a division of Media24, by means of the BarOn EQ-I test. It is concluded that if managers and employees develop their emotional intelligence, both parties will benefit. The key lies in engaging people's heads and hearts and in optimising human capital through emotional. intelligence.
AFRIKAANSE OPSOMMING: Die uitdaging wat bestuurders en werknemers in die 21ste eeu in die gesig staar is om optimaal te funksioneer in die veranderende omgewing waarin hul hulself bevind. Hierdie studieprojek het ten doel om moderne bestuurders riglyne te gee om menslike kapitaal deur middel van emosionele intelligensie te optimaliseer en hulle sodoende te voorsien van 'n praktiese hulpmiddel waarmee hulle die uitdagings van die 21ste eeuse sake-omgewing die hoof kan bied. Die sleutelvraag is: Wat beteken dit in die praktyk om menslike kapitaal deur emosionele intelligensie te optimaliseer? In 'n poging om hierdie vraag te beantwoord is die primêre doel van hierdie studie om 'n praktiese en toeganklike hulpmiddel te ontwikkel wat die moderne bestuurder kan gebruik om die konsepte menslike kapitaal en emosionele intelligensie in sy persoonlike en professionele lewe te verstaan en te optimaliseer Die dae toe bestuurders en werknemers gevra is om hul emosies tuis te laat, is verby. Hierdie studieprojek het ook ten doel om die belangrikheid van emosies by die werk uit te lig, hoe die bestuur van emosies 'n beduidende uitwerking op 'n individu se potensiaal vir ontwikkeling en vordering by die werk kan hê, en laastens, hoe anders die werkplek kon wees as almal gehelp word om hul emosionele intelligensie te herken en te bestuur. Bestaande definisies, teorieê, tekste en temas oor menslike kapitaal en emosionele intelligensie word verwerk tot praktiese raad wat toeganklik is vir en deur die moderne bestuurder verstaan kan word. Dertig spesifieke opleidingsriglyne word gegee wat as 'n bestuurshulpmiddel gebruik kan word. Die skrywer poog om die studieprojek met verwysing na 'n Suid-Afrikaanse gevallestudie te anker. Abstract:
The speed and complexity of globalisation and reduction of natural resources on the one hand, and interests of large multinational corporations on the other, necessitates proper management of mineral resources and consumption. The need for scientific research and application of new methodologies and approaches to maximise Net Present Value (NPV) within mining operations is essential. In some cases, drill core logging in the field may result in an inadequate level of information and subsequent poor diagnosis of geological phenomenon which may undermine the delineation or separation of mineralised zones. This is because the interpretation of individual loggers is subjective. However, modelling based on logging data is absolutely essential to determine the architecture of an orebody including ore distribution and geomechanical features. For instance, ore grades, density and RQD values are not included in conventional geological models whilst variations in a mineral deposit are an obvious and salient feature. Given the problems mentioned above, a series of new mathematical methods have been developed, based on fractal modelling, which provide a more objective approach. These have been established and tested in a case study of the Kahang Cu-Mo porphyry deposit, central Iran. Recognition of different types of mineralised zone in an ore deposit is important for mine planning. As a result, it is felt that the most important outcome of this thesis is the development of an innovative approach to the delineation of major mineralised (supergene and hypogene) zones from ‘barren’ host rock. This is based on subsurface data and the utilisation of the Concentration-Volume (C-V) fractal model, proposed by Afzal et al. (2011), to optimise a Cu-Mo block model for better determination of an ultimate pit limit. Drawing on this, new approaches, referred to Density–Volume (D–V) and RQD-Volume (RQD-V) fractal modelling, have been developed and used to delineate rock characteristics in terms of density and RQD within the Kahang deposit (Yasrebi et al., 2013b; Yasrebi et al., 2014). From the results of this modelling, the density and RQD populations of rock types from the studied deposit showed a relationship between density and rock quality based on RQD values, which can be used to predict final pit slope. Finally, the study introduces a Present Value-Volume (PV-V) fractal model in order to identify an accurate excavation orientation with respect to economic principals and ore grades of all determined voxels within the obtained ultimate pit limit in order to achieve an earlier pay-back period.

Abstract:
Les cellules contiennent de nombreux ARNs non-codants qui jouent un rôle essentiel dans toutes les étapes de l'expression génique. Nos travaux concernent la caractérisation de possibles nouvelles fonctions associés à deux classes d'ARNs non-codants : l'ARN U1 et les scaARNs. Le petit ARN nucléaire U1 forme une ribonucléoparticule (snRNP U1) qui joue un rôle clé dans l'épissage des ARN pré-messagers. De plus, l'ARN U1 semble intervenir dans d'autres étapes de la synthèse des ARNm fonctionnels. Nos travaux ont démontré qu'une fraction de l'ARN U1 interagit spécifiquement avec la protéine TAF15, dans un complexe qui diffère dans sa composition protéique de la particule d'épissage canonique Sm snRNP U1. Nous avons constaté qu'à la différence de la particule d'épissage, la snRNP U1-TAF15 est fortement attachée à la chromatine. De plus, suite à l'arrêt de la transcription par l'ARN polymérase II, la particule U1-TAF15 se relocalise dans la coiffe périnucléolaire, à l'opposé de la particule Sm snRNP U1 qui s'accumule alors dans les "speckles". Une étude protéomique de la particule U1-TAF15 nous a permis d'établir des hypothèses quant à sa fonction qui sont actuellement à l'étude dans le laboratoire. Récemment, une protéine essentielle à l'adressage des scaARNs (ARNs spécifiques des Corps de Cajal) dans les Corps de Cajal a été identifiée (Tycowski et al. , 2009; Venteicher et al. , 2009). Une analyse par séquençage à haut-débit de l'ensemble des petits ARNs associés à WDR79 a permis d'identifier de nombreux nouveaux scaARNs putatifs. Parmi eux, l'analyse de deux ARNs à boite C/D a défini des structures essentielles pour leur localisation dans les corps de Cajal. Des études plus approfondies devraient nous permettre de déterminer des éléments spécifiques nécessaires à l'adressage des scaARNs à boite C/D vers les corps de Cajal. Par ailleurs, nous avons commencé la caractérisation d'un nouveau scaARN qui ciblerait la modification d'un ARN de transfert. Alors que les ARN C/D connus chez les Eucaryotes participent à la biogenèse des ARN ribosomiques et des petits ARN nucléaires, nous avons identifié pour la première fois chez les Eucaryotes un ARN C/D qui ciblerait un ARN de transfert
Noncoding regulatory RNAs (ncRNAs) are in the focus of current research, since they participate in nearly all cellular processes. To get further insights into the functional and structural complexity of ncRNAs, we studied human ncRNAs belonging to two classes of ncRNAs, the nucleoplasmic spliceosomal snRNAs and the nucleolar and Cajal body-specific box C/D 2'-O-methylation guide RNAs. The U1 snRNP is an evolutionarily conserved, abundant nucleoplasmic snRNP that plays a central role in pre-mRNA splicing. According to a recently emerging view, besides its constitutive role in splicing, the U1 snRNP has important regulatory functions in different steps of pre-mRNA production. We demonstrated that a fraction of the human U1 snRNA specifically associates with the nuclear RNA-binding protein TAF15 that is known to interact with a subpopulation of TFIID and RNA polymerase II complexes. The U1-TAF15 snRNP is structurally and functionally distinct from the well-characterized U1 spliceosomal snRNP and it tightly associates with chromatin. The function of U1-TAF15 snRNP remains unknown; it might contribute to the coupling of transcription and splicing. WDR79 (also called WRAP53) has been recently identified as an essential factor for targeting a subclass of box C/D and H/ACA modification guide RNAs, as well as telomerase H/ACA RNA, into the Cajal bodies. Accumulation of box C/D and H/ACA RNPs in Cajal bodies is essential for the biogenesis of functional spliceosomal snRNPs and telomere synthesis. Co-immunopurification of WDR79-associated human RNAs, followed by cDNA synthesis and deep sequencing identified a large number of novel Cajal body-specific RNAs. We are currently dissecting the cis-acting RNA element responsible for WDR79-binding and for targeting box C/D 2'-O-methylation guide RNPs into Cajal bodies. We have also identified a novel Cajal body-specific 2'-O-methylation guide RNA that is predicted to direct methylation of cytidine 34 at the Wobble position of tRNA-Met-CAT elongator. Interestingly, tRNA modification is a novel function for vertebrate box C/D scaRNPs

Dans un milieu désordonné, une onde peut être localisée exponentiellement par des effets d'interférence. Ce phénomène de localisation d'Anderson conduit notamment à une annulation de la conductance d'un fluide quantique unidimensionnel. Des travaux théoriques ont cependant montré que l'application d'un champ électrique pouvait réduire, voire supprimer, cette localisation. Nous étudions ici l'effet d'une force sur la localisation d'une onde quantique de matière dans un système unidimensionnel. En lien direct avec les expériences d'atomes ultrafroids, qui permettent d'observer la localisation d'Anderson d'un paquet d'onde en étalement, ou bien l'effet du désordre sur le transport entre deux réservoirs, nous nous intéressons à deux systèmes : la diffusion et la transmission d'une particule. Afin d'étudier la transmission à travers un guide, nous étendons un formalisme de matrices de transfert à la présence d'une force, éventuellement inhomogène. Deux approches analytiques complémentaires nous permettent d'étendre les résultats au cas d'un désordre de tavelures tel que celui utilisé dans les expériences d'atomes ultrafroids. Nous montrons que la force peut être entièrement prise en compte à l'aide d'une renormalisation de la longueur du guide par un libre parcours moyen local de la particule. Pour un désordre blanc, la force conduit alors une localisation plus faible, algébrique, tandis qu'une délocalisation apparaît pour un désordre corrélé. Nous nous intéressons ensuite à la diffusion d'une particule, à l'aide d'une approche numérique. Nous mettons en évidence une délocalisation de la position à grande force sous la forme d'une croissance temporelle algébrique, dont l'exposant augmente avec la force. Nous montrons de plus que la localisation est systématiquement détruite dans un désordre corrélé
A wave can be exponentially localized in a disordered medium, due to interference effects. This Anderson localization phenomenon leads to a cancellation of the conductance of a quantum fluid in 1D. However, theoretical works pointed out that an electric field may reduce or cancel this localization. We study here the effect of a force on the localization of a 1D quantum matter wave. Since both Anderson localization of an expanding wave packet and the effect of disorder on the transport between two reservoirs have been studied in ultracold atom experiments, we focus on two systems, namely the diffusion, or the transmission, of a particle.In order to calculate the transmission, we generalize a transfer matrix formalism to the presence of a, possibly inhomogeneous, force. The case of a speckle disorder as used in ultracold atom experiments is dealt with using two other analytical approaches. Our main is result is that the force can be entirely taken into account by renormalising the length with a local mean free path of the particle. For white-noise disorder, the force leads to a weaker, algebraic localization, whereas full delocalization appears for a correlated disorder. We then focus on the diffusion of a particle, using a numerical approach. A transition of delocalization of the particle for strong forces is shed into light through a power law increase of its position, whose exponent increases with the force. Moreover, we show that localization is systematically destroyed in a correlated disorder

Johann Sebastian Bach's Chaconne from his Partita in D Minor is considered a crown jewel of the violin repertoire. Since its composition, this piece has taken many forms, undergoing transformations allowing its performance by other instruments and ensemble formations. These include transformations for piano, orchestra, flute, clarinet, and trumpet amongst others. This projects serves as another transformation, bringing a new versions of the Chaconne to the repertoire of the alto saxophone. Additionally, the project discusses in detail the necessary changes made to the music, as well as offering comprehensive technical suggestions for the inherent issues encountered within this arrangement.

Abstract:
RNAs undergo different posttranscriptional chemical modifications, which affect their structural stability and functional diversity. RNA methylation is a very common type of post-transcriptional modification and is present in all domains of life: Archaea, Eukaryotes and Bacteria. Some of these methylations are catalyzed either by a RNA-protein complex or by stand-alone enzymes. The RNA-protein complex (Ribonucleoprotein complex) is comprised of a small RNA known as the guide RNA (Box C/D RNA) and core proteins (L7Ae, Nop5, and Fibrillarin). Box C/D RNAs contain conserved regions, called box C and box D near their 5’ and 3’ termini, respectively, and their imperfect copies called box C’ and box D’, internally. A short stretch of sequence between these Boxes are known as the guide/spacer regions, as the guide region helps in recruiting and positioning a specific target RNA for modification. Both in Archaea and Eukarya, box C and box D, as well as box C’ and box D’ together can form a structure called a Kink-turn (K-turn) that is characterized by a canonical Watson-Crick base-paired stem on one side, and a non-canonical stem on the other, separated by a 3-nucleotide loop. In Archaea box C’ and D’ can also form a K-loop, where the canonical stem of K-turn is replaced by a loop. Archaeal L7Ae binds first to the K-turn or K-loop and allows the recruitment of other proteins to form the complex. The presence of a unique box C/D RNA of Haloferax volcanii, called sR-tMet has been reported previously to guide the 2’-O-methylation of C34 in elongator pre-tRNAMet. Here we tried to characterize the structure-function relationship of this guide RNA under in vivo conditions. This RNA lacks a conventional K-turn or K-loop at its C’/D’ motif. We have created an H. volcanii strain that has a genomic deletion of sR-tMet. The sR-tMet gene is not essential for H. volcanii but this sR-tMet deleted strain lacks the 2’-O-methylation of C34 of its elongator tRNAMet. Unlike the close sR-tMet homologs (sR8 from Methanocaldococcus jannaschii and sR49 from Pyrococcus abyssi), the Box C’/D’ motif of sR-tMet is neither a K-turn nor a K-loop. The introduction of proper K-loop in the Box C’/D’ motif (sR-tMet with K-loop) abolished its Cm34 modification function in ΔsR-tMet strain. Direct interaction between L7Ae and the K-loop is not an absolute requirement for its function. However, disruption of the G/A and A/G pairing in Box C/D motif and Box D’ suggests the importance of these non-Watson crick base pairings in respect to sR-tMet’s function. Several other mutational studies have revealed that peculiar sR-tMet guide RNA from H. volcanii, behaves more like a Eukaryotic Box C/D RNA (where the K-loop is not required and presence of longer spacer length) than regular Archaeal one. Pseudouridine synthase 10 (Pus10) is the most recently identified Ψ synthase, found only in higher eukaryotes and Archaea. Archaeal Pus10 produces either tRNA Ψ55 or both tRNA Ψ54 and Ψ55 modifications. In Human, its Ψ synthase activity is not yet confirmed and interestingly it has been implicated in apoptosis. Herein for the first time we revealed that this putative RNA Ψ synthase protein, Human Pus10 (HuP10), translocates from the nucleus to the cytoplasm in TRAIL induced apoptosis. This nucleo-cytoplasmic movement of HuP10 occurs through the CRM1 mediated nuclear export pathway and Caspase 3 influences this movement. HuP10 also mediates crosstalk between the extrinsic and intrinsic pathways during TRAIL-induced apoptosis. Other than its involvement in apoptosis, we have also uncovered that HuP10 is involved in regulation of cell proliferation. Depletion (knockdown) of this protein in different cancer cell lines, promotes cell migration and anchorage-independent cell growth in the absence of any apoptotic stimulation.

Les entreprises dont le cœur de métier est orienté vers la conception et la production de systèmes réputés complexes cherchent en permanence à améliorer leur position sur des marchés généralement mouvants face à la concurrence internationale. Une des solutions possibles consiste à déployer, améliorer et gérer des processus d'Ingénierie Système. Entre autres avantages, les entreprises peuvent par ce biais espérer gagner à la fois en termes de qualité de leurs prestations et de réactivité avec laquelle elles les fournissent.Ce déploiement nécessite cependant de porter une attention particulière aux besoins, capacités et motivations des ressources et organisations, tant internes qu'externes à l'entreprise, devant être impliqués dans le déploiement ou l'exécution de ces processus. De fait, la maîtrise de leur interopérabilité doit être considérée comme un facteur clef de la réussite du déploiement. Par ailleurs, comme le déploiement de processus appelle à structurer le travail, à modifier l'organisation et le fonctionnement même des entreprises, ces dernières doivent être en mesure de mettre en œuvre des efforts importants de compréhension, d'anticipation, d'adaptation voire de gestion du changement. Elles se trouvent alors souvent dépourvues face au peu de solutions pragmatiques disponibles et applicables réellement pour déployer des processus en milieu industriel.Pour répondre à ces besoins, les travaux de recherche développés au cours de cette thèse ont pour objectif de fournir un cadre méthodologique et technique permettant de lever certains des verrous empêchant ou freinant la réussite du déploiement de processus. Ils sont appliqués ici au déploiement de processus d'Ingénierie Système dans un contexte aéronautique. Ils concernent plus particulièrement les processus d'ingénierie de produits de type hélicoptère et des services associés.La contribution globale de ces travaux consiste donc à définir un guide méthodologique outillé pour assister le déploiement de processus dans une entreprise multi-sites et pluridisciplinaire. Ce guide inclut :• Une analyse comparative détaillée des standards de l'Ingénierie Système applicable en aéronautique,• Un méta-modèle permettant d'établir une vision/compréhension partagée des concepts nécessaires au déploiement,• Un modèle de maturité permettant de connaitre le degré de préparation de l'entreprise pour le déploiement,• Une méthodologie de déploiement formalisée (BPMN 2.0 et modèle formel sous-jacent) facilement automatisable et non ambiguë,• Une méthodologie d'évaluation de l'interopérabilité des ressources et des unités d'organisations à impliquer dans un processus à déployer, formalisée mathématiquement et outillée entre autres par des questionnaires types facilitant son application immédiate,• Des modèles de processus et en particulier celui de définition des exigences réalisé pour valider la méthodologie de déploiement,• Un ensemble cohérent et interopérable de solutions informatiques comprenant un moteur de workflow, des outils de modélisation, des outils d'évaluation de la maturité et de l'interopérabilité, une application récupérant la connaissance des modèles et un générateur de documents
Companies whose primary business is the design and the supply of complex systems try to constantly improve their performances to be ahead of their international competitors. For this, a solution is to deploy, improve and manage Systems Engineering processes. Indeed, among other things, this may enable companies improving the quality of their services but also the reactivity with which they supply them.Nonetheless, this deployment requires paying a special attention to needs, abilities and motivations of resources and organisations, should they be internal or external to the company, likely to be involved in the deployment or execution of these processes. Thus, mastering their interoperability should be considered as an important need and should be considered as a key factor in the deployment.Moreover, since a process deployment implies to structure the work share, to modify the organisation and the functioning of enterprises, the latter must be able to carry out considerable effort to understand, anticipate, adapt and lead the change. However, they often experience difficulties considering the few pragmatic and applicable solutions available to directly deploy processes in industry.To meet these needs, this thesis research aims to supply a methodological and technical guide that enables getting over obstacles preventing or impeding the success of the deployment. It is here applied for the deployment of Systems Engineering processes in an aeronautics context. More precisely, this work focus on processes in charge of the design of helicopters along with services related to them.Consequently, the global contribution of this thesis consists in the definition of an equipped methodological guided to support the deployment of processes in a company that is multi-sites and multi-cultural. This guide includes:• A detailed comparative analysis of international Systems Engineering norms applicable in a aeronautics,• A meta-model that enables sharing a common vision/understanding of concepts required for the deployment,• A maturity model that enables appraising the readiness of a company for the deployment,• A formalised deployment methodology (BPMN 2.0 and an underlying formal model) that is unambiguous and that can be easily automated,• A methodology enabling the assessment of the interoperability of resources and organisation units to involve in a process to deploy, mathematically formalised and equipped with typical questionnaires to facilitate its immediate application,• Processes models and more particularly the one in charge of the stakeholders requirements definition to validate the deployment methodology,• A consistent and interoperable set of computer applications including a workflow engine, modelling tools, applications for the assessment of maturity and interoperability, an application to collect knowledge in models and a document generator

Solos for euphonium with electronic media present the unique challenge of incorporating an active, physical involvement in the live accompaniment through sound-altering technology such as guitar pedals or digital processors. Instructions for this solo genre are often vague and demand a general knowledge of how to use non-traditional devices. Due to the lack of information available on newly-composed pieces for this medium, students and professionals easily overlook the artistic merit of electroacoustic music. This dissertation provides a comprehensive performance guide that aids in the set-up and operation of advanced technology and presents a methodical approach to performing common musical and technical challenges found in modern euphonium repertoire with electronic media. Included in this dissertation are tables of common audio vocabulary and images of connectors, safety precautions, equipment recommendations with performance settings, a list of required connectors, adapters, cables, speakers, and amplifiers, performance set up diagrams, background information, and analyses of both the technical and musical aspects of each piece. In the appendices are signal flow charts, visual illustrations of polar recording patterns, and an updated catalog of published and unpublished original, adapted, and arranged euphonium solos with live electronics and electronic media accompaniment between 1970 and 2017.
