Full textIn 1994, the vote of the French "bioethics' laws" offers to innovating biomedical technologies a specific regulatory status, which was what law people and scientists had been expecting. These laws were meant to forbide practices which could be discriminatory or eugenic and at the same time to allow others practices necessary to medical innovation, even if some of those are still discussed. A reform of the bioethics' laws was scheduled five years later, in 1999. But it is only in 2002 that the French Parliament has decided to modify the initial texts. This revision, which has became necessary due to a very quick evolution of biotechnological innovation, is as difficult to put in place as it was the case for the vote of the initial laws. Today, gene therapy and cell therapy are expected to allow new fields of scientific research in hope to fing alternative treatments to incurable diseases. .

Full textAcute stimulation of β-adrenergic receptors (β-ARs), for example during physical activity, leads to the synthesis of the second messenger cAMP in cardiomyocytes, which triggers a cascade of events leading to the increase of cardiac function. While acute β-AR stimulation is beneficial to the heart, chronic β-AR activation is detrimental because it promotes cardiac remodeling and ultimately leads to heart failure (HF). HF is defined by the heart's inability to overcome hemodynamic needs of the body. While the majority of patients die of worsening heart function, a significant proportion dies suddenly of cardiac arrhythmias.Phosphodiesterases (PDEs) are crucial enzymes since they allow not only to terminate cAMP signals by degrading this second messenger into inactive 5’AMP but permit their spatial organization in subcellular compartments. HF is accompanied by profound rearrangements of the β-AR pathway and the expression of PDEs is modified under pathological conditions, thus disrupting cAMP intracellular compartmentation. The expression of one of these enzymes, PDE4B, is decreased in cardiac hypertrophy and the invalidation of the gene encoding PDE4B promotes ventricular arrhythmias under β-AR stimulation in mice. Conversely, the expression of another enzyme, PDE2A, is up-regulated in human and animal models of HF which may constitute an important defense mechanism during cardiac stress since its overexpression attenuates hypertrophy induced by norepinephrine or phenylephrine and limits cardiac arrhythmias.The purpose of my work was to test the hypothesis that an increase of PDE activity could constitute an alternative to conventional HF treatments to limit cardiac remodeling, HF progression and associated arrhythmias. To do so, I performed a cardiac gene therapy in mouse models of HF using serotype 9 adeno-associated viruses (AAV9) encoding for PDE4B or PDE2A. My results suggest that this approach may be a promising new therapeutic strategy during HF by limiting cardiac dysfunction, left ventricular hypertrophy, and could protect ventricular arrhythmias only when PDE2A is overexpressed

Full textThe detailed characterization of large area is a challenging task because time and resources are frequently limited. This Master’s thesis is part of the ParaChute research project. The aim of this project is to develop a rockfall susceptibility rating system along linear infrastructures. A partially automated method has been developed to facilitate field works planning while optimizing time and resources. It is mainly based on 3D rockfall simulations carried out systematically and efficiently on every rock slopes located nearby the infrastructure to help identify potential hazardous natural cliffs. Automation tools were developed to allow the realization of simulations over large area. The proposed method also uses the software Rockyfor3D and only requires surface elevation model obtained from airborne LiDAR survey as input data. However, other data, such as orthophotos, were used for calibration. The method was applied along the ArcelorMittal Infrastructures Canada railway. The covered zone starts near Port-Cartier (Québec) and extends 260 km north along the ArcelorMittal Infrastructures Canada railway up to the Groulx Mountains. In this Master’s thesis, a partially automated method that helps to choose on which areas to focus field work by telling if there is a possibility for a block to reach the linear infrastructure is detailed.

Full textThis work concerns the study of the reinforcement of stone structures with composite materials. Two reinforcing techniques are used. One is reinforcement by cement matrix reinforced with textile TRC of glass fiber. The other is by reinforcing FRP fiberglass rods according to NSM technique (near-surface mounted). The first phase consists of materials characterization and analysis of mechanical behavior in compression shear and tensile of small masonry specimens made of stone and mortar. The characteristic values of the mechanical behavior of the masonry are compared with the different models in the literature which calculate the characteristic parameters and give a good approximation with the experimental results. In addition, the behavior of reinforced materials is tested in tension and the behavior of the adhesion between the reinforcement and the masonry is studied. The second phase focuses on strengthening masonry walls subjected to two types of loading. One is in-plane shear of walls made with the diagonal compression test. Represents the second bending loads to 3 points out of plane walls accompanied by axial compression forces which cause a compressive stress of 0.2 MPa. The shear test was performed on square walls of five rows of stones with mortar. Strengthening by two vertical bars NSM FRP significantly improves the ductility and shear modulus of the walls and the reinforcement with two orthogonal layers TRC improves shear strength. Finally the application of analytical models used to compare the experimental results with the results given by the models. In addition, the bending test with 3-point is carried out on walls composed of twelve rows of stones with mortar. The reinforcement with two vertical bars and two crossbars increased more elastic stiffness. However, the reinforcement with TRC composed of a single layer of textile accompanied with a vertical bar FRP does not increase the resistance of the wall relative to the resistance of the reinforced wall with a single layer of textile or with a simple vertical bar

Full textRetinitis pigmentosa (RPs) or inherited retinal degenerations are a group of inherited degenerative blinding diseases affecting more than 1, 5 million people. Many genetic mutations causing RPs being now identified gene therapy holds promises for these untreatable diseases. The chimeraplasty/genoplasty is a recent non-viral gene therapy approach based on the use of oligonucleotides (ODNs). The delivered specific wild-type ODNs, in contact with the corresponding gene that contains a point mutation, would activate the endogenous gene repair systems in the targeted cells and stably correct the targeted genomic DNA. The aim of this thesis was to apply the chimeraplasty/genoplasty strategy in the wildest studied human RP animal model, resulting from a point mutation : the rd1 mouse. First, we have optimized ODNs delivery into the targeted retinal photoreceptor nuclei using a treatment combining iontophoresis and intravitreal injection.
