Abstract:
The shortage of organs and the growing need for them over recent years have led to the adoption of less stringent donor acceptance criteria, resulting in the approval of marginal organs for transplant, especially from elderly donors. This implies a higher risk of graft dysfunction, a higher frequency of immunological and vascular complications, and shorter graft survival. Several strategies have been implemented in clinical practice to assess graft quality and suitability for transplantation. We have started to test the prospective intraoperative use of thermo-vision cameras during graft reperfusion. Images were acquired using the FLIR One Pro thermo-vision camera for android devices. We hypothesized that thermal images would give a better perspective about the quality of arterial perfusion and graft revascularization of the renal cortex. Thermo-vision cameras provide an easy-to-use, noninvasive, cost-effective tool for the global assessment of kidney graft cortical microcirculation in the immediate post-reperfusion period, providing additional data on the immediate viability and function of a graft.

Abstract:
The article tackles the issues related to a process of heating of three way catalytic converter during the cold start and the heating of the spark ignition engine. The measurements on the test bench were performed, taking into consideration how engine works directly after the start, on the idle speed and under the load, during which the temperature of the exhaust gases in the exhaust system and coolant on the cylinder head were measured. At the same time the track of the heat state of the catalytic converter was monitored using thermo-vision camera. The results of the measurements were presented as charts and selected thermo-grams, qualitatively representing the issue of heating of the catalytic converter.

The present study was investigated on the melting phenomena and the accelerative factors of phase change material (PCM) by acoustic streaming induced ultrasonic vibrations. To investigate the melting phenomena and accelerative factors, the experimental study was measured the liquid temperature and melting time of PCM and was observed the velocity vectors and thermal fluid flow induced acoustic streaming to investigate the heat transfer using particle image velocimetry (PIV) and infrared thermo vision camera, respectively. Also, the numerical study based on a coupled finite element-boundary element method (Coupled FE-BEM) was performed to investigate the analysis of pressure field in the PCM. The results of experimental works revealed that acoustic streaming observed by PIV and infrared thermo vision camera is one of the prime effects accelerating phase change heat transfer. And, the final temperature of PCM is lower and melting speed is 2.6 times faster than that without ultrasonic vibrations when ultrasonic vibrations are applied. The results of numerical work presented that acoustic pressure is higher near the ultrasonic transducer than other points where no ultrasonic transducer was installed and develops more intensive flow such as acoustic streaming, destroying the flow instability. Moreover, the profile of acoustic pressure variation is consistent with that of enhancement of heat transfer.

Contact lenses are widely prescribed for vision correction, and as such they are an attractive platform for drug delivery to the anterior segment of the eye. This manuscript explores a novel strategy to drive the reversible adsorption of peptide-based therapeutics using commercially available contact lenses. To accomplish this, thermo-sensitive elastin-like polypeptides (ELPs) alone or tagged with a candidate ocular therapeutic were characterized. For the first time, this manuscript demonstrates that Proclear CompatiblesTM contact lenses are a suitable platform for ELP adsorption. Two rhodamine-labelled ELPs, V96 (thermo-sensitive) and S96 (thermo-insensitive), were employed to test temperature-dependent association to the contact lenses. During long-term release into solution, ELP coacervation significantly modulated the release profile whereby more than 80% of loaded V96 retained with a terminal half-life of ~4 months, which was only 1–4 days under solubilizing conditions. A selected ocular therapeutic candidate lacritin-V96 fusion (LV96), either free or lens-bound LV96, was successfully transferred to HCE-T cells. These data suggest that ELPs may be useful to control loading or release from certain formulations of contact lenses and present a potential for this platform to deliver a biologically active peptide to the ocular surface via contact lenses.

The paper presents the simulation model of down milling process of titanium alloy (Ti6Al4V) with a tool made of sintered carbides, considering a tool stiffness. A finite element method, applied for numerical computations was experimentally verified with the help of force dynamometer, thermo-vision and high-speed video cameras. Differences between measured and simulated values were less than 22 % for cutting forces and about 8% for maximum cutting zone temperature and about 5% for tool deflection in the active part of the cutting edge.

The paper discusses the problem of thermal state of three-way catalytic converter depending on engine load with spark ignition fueled with gasoline and natural gas. The measurements on the test bench were performed, during which the temperature of the exhaust gases in the exhaust system was measured with the help of thermocouples, and at the same time, the track of the thermal state of the catalytic converter was monitored using thermo-vision camera. The stable work of engine with different rotation speed and values of load was considered together with transient states. The results of the measurements were presented in forms of charts and selected thermograms, qualitatively presenting the issue of thermal state of the catalytic converter.

Herein, the solidification cracking susceptibility of the CM247LC superalloy and its welds with ERNiFeCr-2 filler wire was quantitatively evaluated using a novel modified Varestraint testing method, for the successful manufacturing of CM247LC superalloy gas turbine blades. It was found that the solidification brittle temperature range (BTR) of the CM247LC superalloy was 400 K. This measurement was obtained with a high-speed thermo-vision camera. The BTR increased to 486 K for the CM247LC/ERNiFeCr-2 welds (dilution ratio: 74%). Theoretical calculations (i.e., the Scheil equation, performed using Thermo-Calc software) were conducted to determine the temperature range in which both solid and liquid phases coexist, together with the microstructural characterization of the solidification cracking surfaces. The greater increase in BTR for the CM247LC/ERNiFeCr-2 welds than that for CM247LC was attributed to the enlargement of the solid–liquid coexistence temperature range. This correlated with the formation of a low-temperature Laves phase during the terminal stage of solidification, and was affected by the diluted Nb and Fe components in the ERNiFeCr-2 filler metal. Based on the experimental and theoretical results, the proposed modified Varestraint testing method for dissimilar welds is expected to be an effective testing process for solidification cracking behavior in the manufacturing of high-soundness CM247LC superalloy welds.

Abstract:
The Inverse Gas Chromatography (IGC) technique has been employed for the surface thermo-dynamic characterization of the polymer Poly(vinylidene chloride-co-acrylonitrile) (P(VDC-co-AN)) in its pure form. IGC attributes, such as London dispersive surface energy, Gibbs free energy, and Guttman Lewis acid-base parameters were analyzed for the polymer (P(VDC-co-AN)). The London dispersive surface free energy ( γ S L ) was calculated using the Schultz and Dorris–Gray method. The maximum surface energy value of (P(VDC-co-AN )) is found to be 29.93 mJ·m − 2 and 24.15 mJ·m − 2 in both methods respectively. In our analysis, it is observed that the γ S L values decline linearly with an increase in temperature. The Guttman–Lewis acid-base parameter K a , K b values were estimated to be 0.13 and 0.49. Additionally, the surface character S value and the correlation coefficient were estimated to be 3.77 and 0.98 respectively. After the thermo-dynamic surface characterization, the (P(VDC-co-AN)) polymer overall surface character is found to be basic. The substantial results revealed that the (P(VDC-co-AN)) polymer surface contains more basic sites than acidic sites and, hence, can closely associate in acidic media. Additionally, visual traits of the polymer (P(VDC-co-AN)) were investigated by employing Computer Vision and Image Processing (CVIP) techniques on Scanning Electron Microscopy (SEM) images captured at resolutions ×50, ×200 and ×500. Several visual traits, such as intricate patterns, surface morphology, texture/roughness, particle area distribution ( D A ), directionality ( D P ), mean average particle area ( μ a v g ) and mean average particle standard deviation ( σ a v g ), were investigated on the polymer’s purest form. This collective study facilitates the researches to explore the pure form of the polymer Poly(vinylidene chloride-co-acrylonitrile) (P(VDC-co-AN )) in both chemical and visual perspective.

Abstract:
This paper was visually studied the thermal and dynamic characteristics of two phase flow by vertical upward bubbles in the liquid. Air-water model in a ladle was visually studied with a particle image velocimetry (PIV) and a thermo vision camera. Once gas was injected into the liquid, bubbles-liquid plume started to form and then circulations was caused by bubbles' behavior helped to reach temperature equilibrium and remove impurities in the liquid. The experimental results showed that temperature equilibrium between water and gas could be achieved in a short time. High speed CCD camera and argon illumination laser were used for the visualization of test section. Liquid circulation was repeated in the local or whole liquid region and accelerated the mixing rate. Visualization of qualitative descriptions such as velocity vectors, turbulent intensity and kinetic energy in the flow field presents useful information on thermal and dynamic characteristics of two phase flow.

Abstract:
Constructing the power curve of a power generation facility integrated with complex and large-scale industrial processes is a difficult task but can be accomplished using Industry 4.0 data analytics tools. This research attempts to construct the data-driven power curve of the generator installed at a 660 MW power plant by incorporating artificial intelligence (AI)-based modeling tools. The power produced from the generator is modeled by an artificial neural network (ANN)—a reliable data analytical technique of deep learning. Similarly, the R2.ai application, which belongs to the automated machine learning (AutoML) platform, is employed to show the alternative modeling methods in using the AI approach. Comparatively, the ANN performed well in the external validation test and was deployed to construct the generator’s power curve. Monte Carlo experiments comprising the power plant’s thermo-electric operating parameters and the Gaussian noise are simulated with the ANN, and thus the power curve of the generator is constructed with a 95% confidence interval. The performance curves of industrial systems and machinery based on their operational data can be constructed using ANNs, and the decisions driven by these performance curves could contribute to the Industry 4.0 vision of effective operation management.

This paper purpose is to provide recommendations after a thermal energy analysis and inspection of a hotel building and its related facilities, in order to increase energy efficiency saving primary resources, to ensure a healthy indoor climate for users, and last but not least to ensure a reduction of CO2 emissions in order to ensure an external environment with low pollutant emissions. All the carried out analysis has as main objective to consider a symbiosis between all the parameters involved (energy, indoor/outdoor environment, comfort), when a building (envelope and installations) is renovated, especially when excess in order to save energy tend to endanger the indoor climate. Non-invasive methods were used to inspect the building by using thermo-vision both for the building envelope and for inspection of existing installations. The building utilities bills were analyzed for a period of one year to highlight current energy performance. The given recommendations achieve the proposed goal and highlight the active role of building management, continuous monitoring of energy and utilities to assess and improve energy efficiency, and ultimately to minimize specific energy costs. The results of this study can be used successfully on a wide range of hotel buildings in Romania.

Energy prices are constantly rising and that ́s why people are always looking for new ways to reduce energy costs. The main trend is to eliminate heat losses to the maximum extent. The demands for more sophisticated outer insulation for building still envelope (roofs, floors, walls, doors, and windows) are growing. Increased demands are evident also in the standards used in this area. Buildings with nearly zero energy (nZEB) become not only a vision but reality. Several ways for reduction of the energy needs are offered. The application of various colour adjustments of the building facade is one of the options. The impact of colour on the demand for heating and cooling at different thermal insulation capability is analyzed from the thermo - technical point of view. However, the question is whether it is possible to influence the annual costs for heating and cooling through the selection of facade colour (respectively through the absorption of solar radiation). We can say that today's architecture tries to use the structure of the building as some kind of thermal radiation (or rather solar energy) accumulator. In such a way we can characterize the majority of the structural designs for energy low cost buildings, where the main aim of such a solution is to reduce the energy need for heating and cooling.

Car camera lens is widely used for the safety system of all kinds of vehicles. The car lens should have very wide field angle, strong ability to endure vibration and thermo-mechanical instability as well as erosion. According to these requirements, a new car lens is designed based on the aspheric glass lenses. The whole structure of car lens contains six separated glass lenses including two aspheric lenses. To reduce the cost, the aspheric lenses are made from low-Tg glass which is adaptable for mass production by glass molding press. The total track of lens is less than 30mm. For the enhancement of night vision, the F-number is set to be 2.4. The full FOV (field of view) of 160 is wide enough to capture all the dangerous information around the car. The imaging resolution is designed to be 3 Mega pixels. The designed result proves that the new car lens features high imaging quality. Its MTF (modulation transfer function) value is not less than 0.15 at the Nyquist frequency as for the field angle of 160. At the half Nyquist frequency, the MTF values of the most field angles are not less than 0.5. Therefore, the new lens could be used in the front of car to monitoring the dangerous cases around the corner.

The paper presents the results of mechanical properties testing of thin sheet metal of INCONEL 625 and 718 alloys. These studies are a continuation of experience in the preparation of the technological characteristics of metal strips plasticity necessary for carrying out numerical simulations [1]. In order to process sheets now become necessary to design the process using software such as thermo-mechanical simulation e.g. Eta/DYNAFORM. On the road of numerical simulation are sought optimal conditions for processing sheets. It brings reducing the cost of industrial tests. However, becomes strictly necessary characteristics of mechanical and technological properties describing the characteristics of the charges for forming. Here the problem is solved if we forming limit curves (FLCs) designated and technological tests conducted. Using the FLCs is comprehensively defined stamping sheet metal press formability and technological tests allow the mapping of the actual operating conditions selected stamping operations. In the presented study used modern digital analyzer AutoGrid of local deformations and the method of image analysis of deformed mesh subdivision. The use of mesh analyzer and vision systems method significantly speeds up the possibility of producing FLCs. Also measurement accuracy is very high. Selected Inconel alloys are evaluated quantitatively and qualitatively by preparing their properties characterization. The acquired data entered into the database material properties of sheet metal and used in the numerical simulation of stamping process of Inconel 625 cone drawpiece. The legitimacy of the use of modern strain analyzer AutoGrid has been confirmed.

The purpose of this study is to evaluate the complex effects of the hot and cold procedures, practiced in modern Spa & Wellness, on skin elasticity, as that indicator is an integral factor defining the aesthetic vision of the skin. Material and Methods: The present study tracked 60 women aged 35 to 65 years. The methods included an aesthetic skin rejuvenation program that contained a complex of 12 Spa and Wellness rituals conducted in a period of one month, three times per week. Each treatment was а temperature contrast procedure through a Parafango body thermo-application and a face cryo-gel application. The facial skin condition was assessed by an A-ONE apparatus - an automatic skin diagnostic analyzer. Results: A significant improvement in the skin elasticity in the three studied age groups was observed after the procedures. This indicator increased 3.75 units (9.93%) in the 35-44 age group, 3.5 units (11.36%) in the 45-54 age-group and 1.80 units (8.22 %) in the observed individuals from 55-65 age-group. At the end of a treatment, the increase in all three age groups was statistically significant (p<0.05). Skin elasticity assessment was significantly lower for all three age-groups before the procedures, compared to the established standards. After the applied procedures, the skin elasticity was approaching the established standard (the first age group exceeded the standard, the second approached the standard and the third was slightly below the standard). Conclusion: The study reveals that the complex effects of hot and cold procedures result in improved skin elasticity.

Dual-Phase (DP) steels are composed of martensite islands dispersed in a ductile ferrite matrix, which provides a good balance between strength and ductility. Current processing conditions (continuous casting followed by hot and cold rolling) generate 'banded structures' i.e., irregular, parallel and alternating bands of ferrite and martensite, which are detrimental to mechanical properties and especially for in-use properties. We present an original and simple method to quantify the intensity and wavelength of these bands. This method, based on the analysis of covariance function of binary images, is firstly tested on model images. It is compared with ASTM E-1268 standard and appears to be more robust. Then it is applied on real DP steel microstructures and proves to be sufficiently sensitive to discriminate samples resulting from different thermo-mechanical routes.

To conduct a comparative analysis of the expression of the following markers: Ki‑67, cyclin D1, E‑cadherin, CD44, MMP‑9, VEGF, P53, vimentin in renal tumor tissue in clear cell kidney cancer depending on the prevalence of the tumor process.Materials and methods. The material for the study was the operating material of 100 patients with light cell kidney cancer who were treated at the National Medical Reseaгch Centгe fог Oncology of the Russian Federation Health Ministry from 2015 to 2018. 50 patients were diagnosed with local cancer (T1–3an0m0), 50 – generalized cancer (T1–4N0M1). For the immunohistochemical (IHC) study, the material was fixed in 10% neutral formalin for 24 hours and encased in paraffin. Dewaxing and restoration of antigenic activity of the material was carried out in the RT module (Thermo Fisher Scientific) using Tris buffer pH=9, for 20 minutes at 98 °C. the Formulation of the IGC reaction was carried out in the immunohistostainer «Autostainer 480S» (Thermo Fisher Scientific). Used system detection UltraVision Quanto Detection System (Thermo Fisher Scientific), and the Chromogen DAB. Antibodies used: E‑cadherin (EP700Y) Cell Marque, 1: 100; CD 44 (EPR1013Y) Cell Marque 1:150; Ki‑67 (SP6) Spring Bio, 1:200; P53 (DO‑7) Cell Marque, 1:200; cyclin D1 (EP12) Dako, 1:200; VEGF Termo Fisher, 1:100; Vimentin (V9) Dako, 1:150; MMP‑9 (EP100902) Epitomics, 1:100. The results of the reactions with markers were evaluated by counting the number of colored cells in each 3rd field of view of the entire drug at magnification of the X200 lens in the AXIO Scope microscope. A1 (Carl Zeiss). The results were expressed as a percentage-the proportion of stained cells in relation to all tumor cells in the field of vision. Parametric methods of statistics were used for statistical processing of the results. The reliability of the difference between the two averages was determined by the student's t‑test for unrelated populations.Results. In clear cell kidney cancer, a low level of proliferative activity was observed in General, but in generalized, compared with local, it was significantly higher (P<0.05) (8±0.5% and 5±0.6%, respectively), and in generalized cancer, there was an overexpression of Cyclin D1–70±3.9%, compared to 14.4±2.3% in local stages, P<0.05. In generalized kidney cancer, epithelial-mesenchymal transformation processes are more pronounced in comparison with local cancer (a significant increase in Vimentin expression by 28% and CD44 by 16.6% (P<0.05), a decrease in E‑cadherin expression by 24% (P<0.05), and activation of neoangiogenesis processes (a significant increase in VEGF expression by 32%, P<0.05). The P53 protein was absent in local kidney cancer cells and was extremely low when generalized – 3.8±0.7%. One of the main markers of extracellular matrix degradation MMP‑9 was approximately at the same level at both stages: at local‑50±6% and 49.6±7.2% at generalized, the difference in indicators is not reliable (P<0.05).Conclusion. Progression of clear cell kidney cancer from local to generalized forms is accompanied by hyperexpression of cyclin D1, a decreased e‑cadherin expression while increasing vimentin expression (increasing signs of epithelial-mesenchymal transformation), an increase in CD44 and VEGF expression.

Туберкульоми на сьогодні залишаються досить поширеною формою туберкульозу легень (ТЛ). Прогресування захворювання може свідчити про неналежний контроль туберкульозної інфекції в ураженому органі на локальному рівні за можливої відсутності будь-яких системних змін імунного статусу. Ось чому прин­ципове значення має вивчення місцевих імунних реакцій у ділянці ураження, зокрема в легеневій тканині. Мета – встановити локальні патерни локалізації та відносну кількість антигенпрезентуючих клітин, клітин з антигенами M. tuberculosis та величину локального імунорегуляторного індексу в легеневій тканині при активних туберкульомах. Матеріал і методи. Проведено імуногістохімічне дослідження серійних зрізів ділянок легеневої тканини на матеріалі 19 резектатів легень хворих із туберкульомою та морфологічними ознаками активності специфічного туберкульозного процесу. Досліджували ділянки легеневої тканини з мікобактеріями та/або їх антигенами, а саме – грануляційний шар капсули туберкульоми, туберкульозні вогнища та гранульоми. Визначали відносну кількість CD4+ та CD8+ лімфоцитів, CD68+ клітин та клітин з наявністю антигенів М. Tuberculosis. Імуногістохімічне (ІГХ) дослідження проводили на AUTOSTAINER 360-2D виробництва компанії Thermo Fisher Scientific (США), для візуалізації клітин застосовано систему Ultra Vision Quanto HRP DAB. У роботі використано такі моноклональні антитіла фірми Thermo Fisher Scientific: мишаче CD4 Clon 4В12, кроляче CD8 Clon SP16, мишаче CD68 Clon КР1 та поліклональне кроляче Mycobacterium tuberculosis antibody PA1-7231 (Pierce Biotechnology, США). Статистичну обробку даних здійснювали з використанням ліцензійних програмних продуктів, які входять в пакет Microsoft Office Professional 2000 (Excel). Результати. У легеневій тканині, яку досліджували, капсула туберкульоми була представлена в усіх випадках, гранульоми поза туберкульомами визначені в 13 (68,4 %) випадках, а туберкульозні вогнища – у 10 (52,6 %) випадках. CD68+ клітини та макрофаги з наявністю антигенів МБТ, тобто інфіковані, виявлено у 100 % випадків, але кількість цих клітин у різних ділянках тканини була різною. CD8+ лімфоцити були визначені у 100,0 % випадків у грануляційному шарі туберкульоми і туберкульозних вогнищах, та в 90,9 % у гранульомах. CD4+ клітини виявлено у 100,0 % випадків у туберкульозних вогнищах, у 89,5 % випадків – у грануляційному шарі туберкульоми та у 72,7 % – у гранульомах. Відносна кількість цих клітин значно коливалася як в окремих гістологічних структурах, так і в окремих спостереженнях. Обчислені значення локального імунорегуляторного індексу при активних туберкульомах легень були в межах їх фізіологічних коливань (1,1–1,61). Висновки. У ділянках легеневої тканини з туберкульомою, де існує безпосередній контакт антигенпрезентуючих клітин з мікобактеріями туберкульозу, найбільша кількість макрофагів із різними морфофункціональними властивостями локалізується в грануляційному шарі туберкульоми, при цьому CD4+ та CD8+ клітини мають значну неоднорідність кількісного розподілу незалежно від кількості інфікованих макрофагів. Поза туберкульомами у легеневій паренхімі визначаються гранульоми різного клітинного типу – переважають епітеліоїдно-лімфоїдні, також є макрофагальні та гігантоклітинні. У більшості з них наявні багатофункціональні макрофаги, а кількість CD4+ та CD8+ лімфоцитів значно варіює, що зумовлює зміни імунорегуляторного індексу. Найменша відносна кількість CD68+ клітин, які водночас інфіковані мікобактеріями туберкульозу (МБТ), спостерігається в туберкульозних вогнищах, при цьому кількість CD4+ та CD8+ клітин варіює. Імунорегуляторний індекс у цих ділянках тканини вказує на переважання цитотоксичної імунної відповіді.

AbstractThe research presents in the first part of the article an original experiment recorded with a Thermo-Vision camera. It reveals the possibility of the human being to direct energy inside the own body but also to the exterior, to heal other persons. The subject, the first author of this article, sent continuously energy to his palms about five minutes. During this time the temperature of his hands grew with more than two degrees Celsius. The subject did not move at all. The energy can be thus directed then further to other persons in need, to help them recover their health. The second part of the article reveals how our inventions rewarded with a lot of gold medals and not only, the Bio-Chips Doctor Tech, can be used to improve the human general state. A very significant experiment is presented, where with the help of these devices the first author of the paper reached for example a general balance of 99.98%. The third set of measurements presented in the article was made by means of three original types of Keshe devices. The first author of the article changed the energy at the level of the own third eye by means of the devices and could significantly improve his own state. These results are also presented in the last part of this article. For the second and the third sets of measurements, the general state of the person was evaluated by means of a Bio Well device. All the three previous types of the measurements are very original and suggestive for how we can help the others. The general purpose of the article is to show that we can do more in different ways for the others, about their health, their level of energy and their level of performance.

Abstract The research shows in the first part that it is possible to maintain a very good state of the human being for a long time, almost continuously. This thing is highlighted using the values of the human being general balance, while the other values are in the optimal range. The measurements were performed by means of a Bio Well device. The tested subject was the first author of this article. Different techniques were used continuously to reach this important goal. Of course, this attitude can only lead to excellence in everyone’s activity domain and also, in general, in everybody’s life. The second part of the research refers to the human being’s possibilities to send energy to the energy cover of the body to improve the human being functioning. This thing is proved by the measurements made with the help of a thermo-vision camera. A short film was made to show the temperature increase at the level of the head skin during five minutes due to the energy arrived at that zone. The third part refers of the possibility of the human beings to repair themselves. The analysis of the thyroid energy level of the measured subject by means of the Bio Well device is presented. Because the thyroid was used a little too much to improve the optimization of the human being functioning, as presented in the first part of this article, the device shows a possible future danger. Immediate measures had to be taken. Due to successive measures, the subject succeeds in bringing his thyroid energy level to a normal one, without the help of medicines. All the three types of the measurements are very original, presented maybe for the first time in the world. The general purpose of the article is to show that we can do more than we believe about our health and our level of performance.

CAMs, such as cadherin P, catenins alpha and beta are much less known..We analysed their distribution in tumors of different patients and in respect to its architecture. Methods: 100 FFPE blocks of superficial ( pTa/ pTis and pT1) and myoinvasive (>pT1) carcinomas G1-3 were analysed by semiquantitative IHC- . Antibodies: E-Cadherin, P-Cadherin and Catenin Beta: Thermo Scientific, Freemont CA,USA . N-Cadherin Lab Vision, UK, Ltd, Catenin Alpha and H-CAM (CD44) Novocastra Laboratories Ltd. Results: E-Cadherin: Normal and reactive, inflammatory urothelium and dysplasia express high membranous E-cadherin-concentrations. High grade (HG) in-situ-carcinomas:discontinously ,intensively reacting cellmembranes; myoinvasive HG carcinomas lose their E-cadherin membranous component but show low diffuse intracytoplasmic reaction. N-Cadherin: in basal cells but not in normal transitional cells and LG dysplasia. High expression in HG dysplasia and deep invasive carcinomas, HG. P-cadherin:normal urothelium, as well as papillary LG and HG and CIS show an extensive membranous expression. Catenin alpha: high expression in pTa/pT1 LG tumors; low expression in myoinvasive carcinomas. Catenin beta: high membranous expression in normal and dysplastic urothelium as well as in in pTa LG tumors; low membranous cytoplasmic and intranuclear expression in myoinvasive carcinomas and HG tumors. Transition of beta-catenin from cellmembrane to nucleus in neoplasms was not visible Conclusions: E-Cadherin to N-Cadherin shift in the EMT destabilize tumorcells for CAMs, enabling by this the connection to fibrous tissues and augmenting the invasive potential. Catenins alpha and beta: strong prognosticators of progression and recidives; as they are integral parts of E-cadherin, they disappear together with E-cadherin in invasive tumor cell complexes.

Abstract The traditional approach to the relation between parents and their prenatal child presents the child as a fetus, a mainly passive recipient of the mother’s vital biological resources. Contemporary prenatal psychology and pedagogy recognizes this relationship in a quite different perspective: the prenatal child is a member of the family and may be seen as an active member of the wider family as a community, extended to grandparents and other relatives. Between parents and their child in the womb exists a reciprocal relationship at a physiological (hormonal), psychological and spiritual level. The prenatal child communicates with the parents in different ways and reacts to their stimulation (acoustic, tactile, loco-motoric, chemo-receptive, thermo-receptive, and emotional). This dialogue of the parents and their prenatal child enriches each member of the family community. In this sense, the prenatal child is a gift and a challenge for the parents to develop their personality, social competences and spiritual life. The reflections presented in this paper fit the conception of the paradigm of unity applied into the area of prenatal education and prenatal pedagogy as a new pedagogical subdisciline. The concept of paradigm in the philosophy of science was introduced by T. Kuhn (1962). In his understanding, paradigm means a mental breakthrough in the way of ujmowaniu/describing/approaching the subject of study, and/or in the research methods used, but above all in the theoretical and systemic vision of reality, which is the basis for formulating research questions and hypotheses, operationalising the research procedure, analyzing the obtained research results and a theoretical interpretation of these results. To Kuhn, a paradigm may be understood in such way only in the natural sciences, when new visions of the studied natural reality appear. This is accompanied by a mental change in the cognitive schema itself of understanding reality not only for the creator of this vision, but also in its/his/her broader scientific environment. Kuhn considered the most eminent example of such appearing paradigm in the history of science to be the Copernican revolution, thanks to which the view on the world of nature surrounding the human being has completely changed and there was a mental breakthrough in the 15th and 16th century not only in the area of science, but also in the worldview of people of those times, for whom the world has become more open and broad. Kuhn, however, did not consider a paradigm to be a methodological concept in the humanities or social sciences. He was of the opinion that they must first mature more and make their methodological status more precise if one wishes to talk about a paradigm in these sciences. A scientific event that gave an occasion to reflect on the possibility of introducing the concept of paradigm into social sciences was the occasion of granting the title of doctor honoris causa in the field of social sciences by the Catholic University of Lublin to Chiara Lubich. The Focolare movement, also called The Movement of Unity, founded and led by her, introduced, a new approach to the area of human relations at every level (interpersonal, family, local society, economic relations, intra- and inter-denominational dialogue). The arguments for granting the title included the fact that the practical activity of Ch. Lubich in various areas of social integration may be described as a Copernican revolution in the cultural-religious relations, and even in social-economical ones, according to the new paradigm, understood practically, which may be called a paradigm of unity. It was stressed that these activities may play a serious/significant inspiring role in the methodological reflections on the paradigm of unity in the social sciences (Biela, 1996). Such reflections were actually brought up first generally in the social sciences (Biela, 2006), and then within the methodology of psychology (Biela, 2009) and education (Kozubek, 2009). At present, one may consider introducing this paradigm into prenatal education and pedagogy.

Abstract:
To provide a better picture of the active geodynamics along the Variscan suture zones during the late collisional stage (particularly regarding the evolution of the orogenic system towards HT conditions), we focused here on vaugnerites, which consist of mafic ultra-potassic magmatic rocks, intrusive into the granite-gneiss sequences of the Variscan Vosges crystalline massif. Those rocks, though subordinate in volume, are frequently associated with late-collisional granites. In the Central-Southern Vosges, they appear either as (1) pluton margin of the Southern Vosges Ballons granite complex or (2) composite dykes intrusive into migmatite and metamorphic sequences classically referred to as granite-gneiss unit (Central Vosges). Both types correspond to melanocratic rocks with prominent, Mg-rich, biotite and hornblende (20–40% vol., 64 < mg# < 78), two-feldspar and quartz. Those Vosges vaugnerites display geochemical signatures characteristic of ultra-potassic mafic to intermediate, metaluminous to slightly peraluminous rocks. Zircon U-Pb ages were obtained by Laser Ablation Inductively Coupled Plasma Mass Spectrometry. Zircon grains were extracted from a sillimanite-bearing gneiss from the granite-gneiss unit hosting the Central Vosges vaugnerites. They yielded an age at 451 ± 9 Ma, indicating a pre-Variscan Upper Ordovician protolith for the host sequence. Zircon from the four vaugnerite intrusives display U-Pb ages (± 2σ) of 340 ± 2.5 Ma (Ballons), 340 ± 25 Ma, 340 ± 7 Ma and 336 ± 10 Ma (Central Vosges). Synchronous within uncertainty, vaugnerite age data suggest a relatively early emplacement during the Late Variscan collisional history (i.e. Middle Visean times). These results are in line with previously published ages from the Southern Vosges volcano-sedimentary sequences (Oderen-Markstein) and the nearby ultra-potassic granite complexes from the Central and Southern Vosges (Ballons, Crêtes) thereby arguing for a magmatic event of regional significance. Recent petrological studies on vaugnerites suggest that they derive from partial melting of a metasomatized mantle contaminated to some different degrees by elements of continental crust. We propose here that the major ultra-potassic magmatic pulse at 340–335 Ma is a consequence of a significant change into the dynamics of the Rhenohercynian subduction system below the Central-Southern Vosges. In the light of recent thermo-mechanical modelling experiments on mature continental collision, magmatism could result from a syn-collisional lithospheric delamination mechanism involving (1) first, continental subduction evolving towards (2) the underthrusting of the Avalonian continental margin lower crust and (3) the initiation of lithospheric delamination within the supra-subduction retro-wedge (Saxothuringian-Moldanubian continental block). This delamination would drive the emplacement of an asthenospheric upwelling, initially localized along the Variscan suture zones, and gradually propagating towards the southern front of the belt during the Late Carboniferous, as the delamination front migrated at the base of the crust.

The concept of combination of thermo reversibility and mucoadhesion has achieved a much valuable interest in various fields of pharmaceutics. Mucoadhesive drug delivery systems interact with the mucus layer covering the mucosal epithelial surface, and mucin molecules and increase the residence time of the dosage form at the site of absorption. Thermoreversible gels are the hydrogels that are liquids at room temperature but undergo gelation when in contact with body fluids or change in pH. A major problem faced by conventional formulation in various routes of administration viz. nasal, ocular, vaginal, rectal, etc is drainage of instilled formulation, poor patient compliance as in the case of vaginal and rectal delivery system. Also, blurred vision and irritation to mucosa is observed in the case of ocular and nasal delivery respectively. The drainage of the formulation results in inaccurate dose delivery thus decreasing the bioavailability of the drug which demands an increase in the frequency of the administration. To overcome these limitations, there is need of novel delivery system such as thermoreversible mucoadhesive insitu gel which provides an ease of administration and improved patient compliance as these smart delivery system is free flowing liquid at ambient temperature and gels at physiological temperature which is higher than the LCST (low critical solution temperature) of thermoreversible polymer. The combined effect of mucoadhesion and thermoreversible property will enhance prolongation of residence time of dosage form and the accuracy of the dose in body cavity respectively, leading to increased bioavailability of the drug. This review focuses on benefits of mucoadhesive thermoreversible system of drug delivery through rectum for the treatment of internal hemorrhoids.

Abstract Background Postural orthostatic tachycardia syndrome (POTS) is characterised by an excessive heart rate increase upon standing and orthostatic intolerance. Previous studies indicate autoimmune involvement mediated via G-protein coupled receptors (GPCRs) as a possible aetiology. Purpose To test if serum from POTS patients and controls activates selected GPCRs and if the activation associates with the severity of symptoms. Methods A total of 34 POTS patients (age 28.9 [9.9] years; 29 women) and a population-based control group of 25 healthy subjects (age 30.7 [8.6] years; 21 women) were included. The subjects performed an active standing test and completed the orthostatic hypotension questionnaire (OHQ) including ten items related to orthostatic intolerance. The OHQ composite score was calculated (range 0–10). Sera were analysed by a FRET-based reporter system (Tango GeneBLAzer, Thermo Fischer) based on a beta-arrestin-linked transcription factor driving transgenic betalactamase transcription. HEK293M-cells over-expressing one of the GPCRs, adrenergic alpha-1 (ADRA1), adrenergic beta-2 (ADRB2), muscarinic type-2 (CHRM2) and opioid-receptor-like 1 (OPRL1) receptor were plated in 96-well optical plates and allowed to re-attach during 48 hrs. The cells were treated with 10% sera diluted in RPMI for 5 hours, followed by addition of the FRET-substrate, incubation for 60 min and quantification analysis in a CLARIOStar multi-purpose plate reader. Quantification of the activation of the GPCRs was log-transformed and related to the OHQ composite and individual scores in age-adjusted linear regression models. OHQ scores were compared according to the median of sera activation, using independent samples t-test. Results Serum ADRA1 activation associated with the OHQ composite score (beta 0.77 OHQ points per SD of activity; p=0.009), whereas there were no significant associations among controls (p=0.953). The association between ADRA1 and total OHQ was significant also after adjusting for heart rate and systolic blood pressure at 3 min (p=0.024). The OHQ composite score was higher in with above median serum ADRA1 activation (Figure 1). ADRA1 activation associated with a higher score for vision problems (p<0.001) and symptoms during prolonged standing (p=0.037) and walking for short (p=0.042) or long periods (p=0.001). The activity of ARB2, CHRM2 and OPRL1 did not associate with OHQ composite score in neither POTS patients nor controls. Figure 1. OHQ according to ADRA1 activation Conclusions Activating serum proteins for the adrenergic alpha-1 receptor are associated with the severity of orthostatic symptoms in POTS patients, partly independent of the orthostatic hemodynamic response. Serum activity against alpha1 receptors is related particularly to symptoms of disturbed vision and symptoms during walking or prolonged standing. Serum activity against the adrenergic alpha 1 receptor may be a one factor underlying the orthostatic symptoms in POTS.
