Abstract:
Managing groundwater resources requires computer modeling software, which in turn brings its own costs in education and usage fees. Although many groundwater modeling programs can be obtained at relatively inexpensive rates the hidden costs of software training remain high in general. What has been done is to make both the software and training available online for free, but this method delegates all responsibility to the users for both accessing the software in addition to learning how to use it properly. In this research the accessibility of groundwater models has been improved upon by creating a web app of an open source software called TimML. I divided up the task of building a general purpose groundwater modeling web app into three different apps with increasing sophistication. The first app I developed simulates construction dewatering using the Dupuit assumptions. The second app also simulates construction dewatering and uses the same basic graphical user interface I developed for the first app while implementing TimML as the backend processor. The culminating app, TimML-Cloud, builds on the user interface created in the first two apps and exposes more of the modeling capacity of TimML. By making TimML a cloud app, people with internet access have greater accessibility to an analytic element groundwater model than before. Because TimML is more accessible than before, people can more readily implement TimML as a decision-making tool in water resource management.

Thymol (2-isopropyl-5-methyl-phenol) an isomer of Carvacrol. Present as large colorless crystals or white crystalline powder. It is a monoterpene essential oil extracted from various herbs, and the main component of the essential oil of rosemary-pepper (Lippia sidoides), constituting approximately 48% of its composition. This paper presents the behavioral actions of thymol in animal models of anxiety, depression, seizures and sedation, such as elevated plus maze (EPM), open field, rota rod, forced swim, tail suspension, seizure induced by pentylenetetrazol and pentobarbital-induced sleep time. Thymol was administered orally in male mice at doses of 25mg/kg and 50mg / kg. The results showed that thymol at both doses studied did cause muscula relaxament activits change locomotor activity on the test rota rod nor the number of crossings in the open field test, but reducedThe grooming was reduced with the administration of thymol in the open field test. At both doses studied LCE thymol increased all parameters observed suggesting a possible anxiolytic effect. Flumazenil, an antagonist of GABA / benzodiazepine, reversed this effect. In the test of sleep time induced by pentobarbital thymol not alter the latency but increased sleep duration. Although this test is sensitive to CNS depressant agents but is not specific for compounds which would interfere with the biotransformation of pentobarbital. In the test of pentylenetetrazol-induced seizures, thymol increased latency of seizure at the dose of 50mg/kg, however was not able to change the mortality of animals. Thymol showed antidepressant effect in the forced swim test and the tail suspension. This effect was reversed by pretreatment of animals with PCPA (a serotonin synthesis inhibitor), Prazosin and Yohimbine (adrenoreceptor antagonists), SCH23390 and Sulpiride (dopamine receptor antagonists). These results suggest that thymol shows anxiolytic effects probably associated with the GABAergic system and antidepressant effects associated with the serotonergic, noradrenergic and dopaminergic without causing sedation.
O Timol (2-isopropil-5-metil-fenol) um isômero do Carvacrol. Apresenta-se como cristal incolor grandes ou pó cristalino branco. É um monoterpeno extraído do óleo essencial de diversas plantas aromáticas, sendo o componente principal do óleo essencial do alecrim-pimenta (Lippia sidoides), constituindo aproximadamente 48% da sua composição. Este trabalho avaliou as ações comportamentais do timol em modelos animais de ansiedade, depressão, convulsão e sedação, tais como labirinto em cruz elevado (LCE), campo aberto, rota rod, nado forçado, suspensão da cauda, convulsão induzida por pentilenotetrazol e tempo de sono induzido por pentobarbital. Timol foi administrado por via oral, em camundongos machos, em doses únicas de 25mg/Kg ou 50 mg/Kg. Os resultados mostraram que o timol nas duas doses estudadas, não causou efeito relaxante muscular no teste do rota Rod nem alterou o número de cruzamentos no teste do campo aberto, mas reduziu o grooming. No LCE ambas as doses estudadas do timol aumentaram todos os parâmetros observados sugerindo um possível efeito ansiolítico. O flumazenil, um antagonista dos receptores GABAA/Benzodiazepínico, reverteu este efeito. No teste do tempo de sono induzido por pentobarbital o timol não alterou a latência, mas aumentou a duração do sono. Entretanto este teste embora sendo sensível para agentes depressores do SNC não é específico pois compostos que possam interferir com a biotransformação do pentobarbital. No teste da convulsão induzida por pentilenotetrazol, timol aumentou a latência da convulsão na dose de 50mg/Kg, no entanto não foi capaz de alterar a latência de morte e a mortalidade dos animais. Timol apresentou efeito antidepressivo no teste do nado forçado e da suspensão da cauda. Este efeito foi revertido pelo pré-tratamento dos animais com PCPA (um inibidor da síntese de serotonina), Prazosina e Ioimbina (antagonistas dos receptores adrenérgicos), SCH23390 e Sulpirida (antagonistas dos receptores dopaminérgicos). Estes resultados sugerem que o timol apresenta efeitos ansiolíticos provavelmente relacionados com o sistema GABAérgico e efeitos antidepressivos associados aos sistemas serotonérgico, noradrenérgico e dopaminérgico sem causar sedação.

Der norddeutsche Schriftsteller Timm Kröger ist in Haale (Kreis Rendsburg-Eckernförde) geboren, einem Dorf, das in nächster Nähe von Luhnstedt, dem Heimatdorf von F.E. Peters, liegt. Peters und Kröger sind sich nie persönlich begegnet, hatten aber gemeinsame Bekannte. „Timm Kröger – der Dichter unserer Heimat“ stammt aus dem Nachlass von F.E. Peters, der ein Bewunderer der Kunst Timm Krögers war und den „besonderen lyrischen Schimmer“ seiner Erzählkunst geliebt hat. Die Rede ist anlässlich des 110. Geburtstages von Timm Kröger entstanden. Peters nimmt das Jubiläum zum Anlass, anhand von Zitaten aus Krögers Vorwort zu einer Gesamtausgabe seiner Werke die Gattung Heimatdichtung kritisch zu beleuchten und zu unterscheiden zwischen einer ideologisch gefärbten schablonenhaften Heimatdichtung und einer Dichtung, welche die Landschaften der Heimat in den Mittelpunkt stellt, das Provinzielle jedoch überwindet: „Es sei zugegeben, dass von sogenannten Heimatdichtern viel plattes Zeug geliefert wird. Based on the vast literature on the therapeutic properties of the monoterpenes carvacrol and thymol, these compounds were used to obtain derivatives and the pharmacological activities of them were evaluated. Modifications were made to the hydroxyl group of C-1, obtaining eighteen derivatives of carvacrol (1a-1i) and thymol (2a-2i), with satisfactory yields. The structures were confirmed by spectroscopic methods, such as GC/MS, IR, 1H and 13C NMR. The compounds obtained and their precursors were submitted to an inhibition test of the acetylcholinesterase enzyme using the colorimetric method in which derivatives 1b and 2b showed positive results against the enzyme. Their cytotoxic activity in vitro were evaluated against SF295 (glioblastoma), HCT116 (colon - human) and PC3 (prostate) cells by the MTT method. Results showed weak cytoxiticy activity and also larvicidal activity to larvae of Aedes aegypti in the third stage, presenting significant results for derivatives 1a, 1b, 1c, 1h, 2a, 2b, 2c and 2h.
: Baseado na vasta literatura sobre as propriedades terapêuticas dos monoterpenos carvacrol e timol, utilizou-se desses compostos para a obtenção de derivados, bem como a avaliação farmacológica dos mesmos. Foram realizadas modificações no grupo hidroxila do C-1, com a obtenção de dezoito derivados, do carvacrol (1a-1i) e do timol (2a-2i), com rendimentos satisfatórios. A confirmação das estruturas foi feita utilizando métodos espectroscópicos, tais como CG/EM, IV, RMN de 1H e 13C. As substâncias obtidas e seus precursores foram submetidas a ensaio de inibição da enzima acetilcolinesterase, pelo método colorimétrico em que os derivados 1b e 2b apresentaram resultados positivos frente à enzima. Foram submetidas à avaliação da atividade citotóxica in vitro, pelo método do MTT, frente às linhagens de células SF295 (glioblastoma), HCT116 (cólon – humano) e PC3 (próstata), apresentando atividade fraca, e também, atividade larvicida que foi realizada frente às larvas de Aedes aegypti no terceiro estágio, tendo apresentado resultados significativos para os derivados 1a, 1b, 1c, 1h, 2a, 2b, 2c e 2h.

Dengue is an infectious disease that may also happen in a severe form with hemorrhagic events. The etiological agent of the disease is an arbovirus wich is transmitted by the mosquito Aedes aegypti, the primary vector of the disease. The main actions to combat the disease are mosquito control and personal protection that can take place using repellents. Almost all repellents have DEET as active substance, which has use restrictions. In this context, thymol (monoterpene) has become a potential insect repellent against Ae. aegypti, and the use of technologies is essential to the viability of a repellent thymol based product. Given the above, the aim of this study was the preparation and pharmaceutical characterization of thymol nanocapsules and evaluation of skin permeation, in vitro cytotoxicity and mosquito repellence (Ae. aegypti). For this purpose, we developed and validated analytical method for identification and quantification of thymol by HPLC-PDA. Nanocapsules thymol (NCT - 1%) showed an active content of 85%. Continuing the characterization, NCT were analyzed for the mean diameter (D), polydispersity index (PDI), potential zeta (PZ), encapsulation efficiency, pH and product stability . Results shared showed an average diameter of approximately 150 nm, negative PZ (-27,83 ± 2,60), PDI less than 0.2 and an encapsulation efficiency of 98%. NCT and ethanolic solution of thymol were evaluated for release, permeation and retention of thymol through dialysis membrane or the porcine ear skin in a Franz diffusion cell model. NCT showed release control of thymol in relation to the ethanolic solution of thymol, which showed a maximal release of 53.8% of the dose within 24 hours of testing, higher than the amount released by the NCT (17.6%) indicating the decrease in permeability when nanoencapsulated. Cytotoxicity analysis of free and encapsulated thymol (50 to 100 ug / ml) in human keratinocytes by MTT assay showed cytotoxicity of free thymol - 100 ug / mL (% viable cells: 16.1 ± 3.2 ), which was virtually devoid with this encapsulation (% viable cells: 92.7 ± 31). Preliminary assessment of the repellent potential of NCT against Ae. aegypti showed that in human topical administration of the product promoted up to 67% repellency. The results suggest that standardized NCT has morphological and chemical characteristics of interest to a nanossystem, plus the potential repellent against Ae. aegypti mosquito.
A dengue é uma doença infecciosa que pode se manifestar de forma grave, com quadros hemorrágicos. O agente etiológico é um arbovirus transmitido pela picada do mosquito Aedes aegypti, principal vetor da doença. As principais medidas de combate são controle do mosquito e proteção individual, que pode dar-se com o uso de repelentes. Quase a totalidade dos repelentes no mercado possui DEET como ativo, que possui restrições de uso. Neste sentido, o timol (monoterpeno) tem-se mostrado promissor como repelente de inseto (Ae. aegypti), sendo a agregação de tecnologias essencial para a viabilidade de um produto repelente à base de timol. Diante do exposto, o objetivo do presente trabalho foi a preparação e caracterização farmacêutica de nanocápsulas de timol, com avaliação da permeação cutânea, citotoxicidade e atividade repelente de inseto (Ae. aegypti). Para tanto, foi desenvolvido e validado método analítico para identificação e quantificação do timol por CLAE-DAD nas nanocápsulas de timol (NCT – 1%), que apresentou um teor de ativo em torno de 85%. Prosseguindo a caracterização das NCT, foram determinados o diâmetro médio, índice de polidispersão (PDI), potencial zeta (PZ), eficiência de encapsulação e pH, além do estudo de estabilidade. Análises das NCT mostraram um diâmetro médio de aproximadamente 150 nm, PZ negativo (-27,83 ± 2,60), PDI abaixo de 0,2 e uma eficiência de encapsulação de 98 %. NCT e solução etanólica de timol foram avaliadas quanto à liberação, permeação e retenção de timol através de membrana de diálise ou da pele de orelha suína, em um modelo de células de difusão de Franz. A NCT mostrou controle da liberação de timol em relação à solução etanólica de timol, que apresentou por sua vez uma liberação máxima de 53,8% da dose em 24 horas de ensaio, superior à quantidade liberada pela NCT (17,6%), evidenciando a diminuição da permeabilidade do timol quando nanoencapsulado. Análise da citotoxicidade do timol livre e encapsulado (50 e 100 g/mL) em queratinócitos humano através do teste de MTT, mostrou a citotoxicidade do timol livre - 100 g/mL (% células viáveis: 16,1 ± 3,2), que foi praticamente destituída com a encapsulação deste (% células viáveis: 92,7 ± 31). Avaliação preliminar do potencial repelente das NCT ao mosquito Ae. aegypti em humano mostrou que a administração tópica desse produto promoveu até 67 % de repelência. Os resultados obtidos sugerem que as NCT padronizadas possuem características morfológicas e químicas de interesse para um nanossistema, além do potencial repelente ao mosquito Ae. aegypti.

Objetivou-se com esse trabalho avaliar o efeito in vitro de metabólitos secundários de plantas no volume de metano (VCH4), degradabilidade in vitro da matéria seca (DIVMS), volume de metano por grama de matéria seca degradada (VCH4/DIVMS), concentração de nitrogênio amoniacal (N-NH3) e ácidos graxos voláteis (AGV). Foram realizados três experimentos, conduzidos no Laboratório Química Analítica de Plantas (LQAP) da Embrapa Cerrados. O meio de cultura foi preparado seguindo-se as recomendações de Theodorou et al. (1994). A mensuração dos gases produzidos foi realizada às 6 e às 24 horas após o início dos experimentos, pelo deslocamento de água medido pelo aparato de vasos comunicantes. DIVMS foi determinada às 24 h. Amostras de Brachiaria brizantha cv. Marandu e concentrado foram pesadas na relação 80:20 em sacos F57 da Ankom ® e foram usadas como tratamento controle (TC) para os experimentos 1 e 2. Para o experimento 3, somente amostras de B. brizantha cv. Marandu foram usadas como controle (TC). O delineamento foi inteiramente casualizado com cinco tratamentos e três repetições e as médias foram comparadas pelo teste de Tukey a 5% de probabilidade. No experimento 1 foi avaliado duas doses (0,3 e 0,6 mL/L de solução) de óleo essencial de alho (A3, A6), canela (C3, C6) e orégano (O3, O6) e duas doses (300 e 600 mg/L de solução) de timol (T3, T6) e resveratrol (R3, R6) em pó. Para VCH4 todos os tratamentos diferiram (P<0,05) de TC, reduzindo o volume produzido. R6 não diferiu (P>0,05) de TC para DIVMS, sendo que todos os outros tratamentos aumentaram (P<0,05) DIVMS. A relação VCH4/DIVMS foi reduzida (P<0,05) com a adição dos metabólitos. A concentração de N-NH3 não foi alterada por nenhum dos metabólitos avaliados. As concentrações dos ácidos acético, propiônico e soma AGV não foram alteradas pelo tratamento R3, tendo todos os outros reduzido (P<0,05) a concentração de ambos os ácidos, o que evidencia o efeito antimicrobiano dos óleos essenciais sobre os micro-organismos ruminais. No experimento 2 foram avaliadas as combinações entre 300 e 600 mg/L de resveratrol com 0,6 mL/L dos óleos essenciais de alho (R3A6, R6A6), canela (R3C6, R6C6) e orégano (R3O6, R6O6), e 600 mg/L de timol (R3T6, R6T6). Também foi avaliada a dose de 0,6 mL/L de todos os metabólitos sem associação com resveratrol (A6, C6, O6, T6, R6) e a menor dose de resveratrol (R3). O VCH4 e a DIVMS reduziram (P<0,05) em relação à TC para todos os tratamentos combinados ou não com o resveratrol, mas não houve efeito (P>0,05) para os tratamentos R3 e R6. O VCH4/DIVMS também foi reduzido (P<0,05) em relação à TC para todos os tratamentos. A concentração de N-NH3 foi reduzida (P<0,05) para os tratamentos R6, R6A6 e R6T6. Os tratamentos A6, C6, O6 e R3C6 aumentaram (P<0,05) a concentração de N-NH3. Não houve efeito, em relação à TC, na concentração de acetato, butirato e soma de AGV para os tratamentos combinados com o resveratrol, à exceção de R3C6, indicando que tais aditivos podem ser usados para melhorar a eficiência do uso da dieta de ruminantes com efeito tóxico sobre os micro-organismos metanogênicos. O experimento 3 avaliou o efeito de extratos de plantas do cerrado, barbatimão e sucupira, nos mesmo parâmetros citados acima. Amostras de Brachiaria brizantha foram usadas como controle (Br). As doses avaliadas foram 100, 200, 300, 400 e 600 mg/L de solução para barbatimão (B100, B200, B300, B400 e B600) e para sucupira (S100, S200, S300, S400 e S600). Para VCH4, B300, B600, S300 e S400 mg/L, diferiram (P<0,05) de Br. Nenhum tratamento teve influência na DIVMS. A relação VCH4/DIVMS aumentou (P<0,05) em 33,8% para S300. A concentração de N-NH3 aumentou (P<0,05) para S400 e S600 Em relação a concentração do AGV, B100 e S300 mg/L reduziram (P<0,05) a concentração de ácido acético, porém S400 aumentou (P<0,05) em 21% a concentração desse ácido. Os tratamentos não diferiram estatisticamente de Br para o ácido propiônico. Os tratamentos reduziram (P<0,05) a concentração de ácido butírico entre 37,6 e 76,4%. A soma de AGV também reduziu significativamente (P<0,05) para todos os tratamentos com extrato de barbatimão e sucupira, a exceção do tratamento S400. Os tratamentos com extrato de barbatimão provocaram queda na produção de CH4, porém reduziram a SAGV, o que mostra que tal extrato, nas doses avaliadas, não é recomendado para melhorar a eficiência de uso da dieta por bovinos. Os tratamentos com extrato de sucupira apresentaram variação nos resultados dos parâmetros avaliados nesse experimento. Mais estudos devem ser realizados para elucidar os efeitos desse extrato sobre os parâmetros de fermentação ruminal. ________________________________________________________________________________ ABSTRACT
The objective of this study was to evaluate in vitro effect of plant secondary metabolites on the volume of methane (VCH4), in vitro dry matter degradability (IVDMD), volume of methane per gram of degraded dry matter (VCH4/IVDMD), ammonia concentration (N-NH3) and volatile fatty acids (VFA). The incubation medium was prepared following the recommendations of Theodorou et al. (1994). In vitro gas production was performed by the use of a simple apparatus for measuring gas production through communicating vessels and methane concentration was determined by gas chromatography. IVDMD were determined at 24 h. Samples of Brachiaria brizantha and concentrate were weighed in the ratio 80:20 into Ankom® filter bags (F57) and were used as the control treatment (TC) for experiments 1 and 2. For experiment 3 only B. brizantha was used as control (Br). In Experiment 1 it was evaluated two doses (0.3 and 0.6 mL/L solution) of cinnamon (C3, C6), garlic (A3, A6) and oregano (O3, O6) essential oils and two doses (300 and 600 mg/L solution) of, thymol (T3, T6) and resveratrol (R3, R6) powder. All treatments differed (P<0.05) from TC for VCH4, reducing the volume produced. R6 did not differ (P>0.05) for IVDMD compared to TC, but all the others treatments increased (P<0.05) IVDMD. The VCH4/IVDMD ratio was reduced (P<0.05) with the addition of the secondary metabolites. N-NH3 concentration was not altered by any of the metabolites evaluated. The concentrations of acetic, propionic and total VFA acids were not altered by treatment R3, while all others reduced (P<0.05) the concentration of both acids, which shows the antimicrobial effect of essential oils on rumen microorganisms. In experiment 2, the combinations between 300 and 600 mg/L of resveratrol with 0.6 mL/L of cinnamon (R3C6, R6C6), garlic (R3A6, R6A6) and oregano (R3O6, R6O6) essential oils and with 600 mg/L thymol (R3T6, R6T6) were evaluated. 0.6 mL/L of all metabolites without association with resveratrol (A6, C6, O6, T6, R6) and the lowest dose of resveratrol (R3) were also evaluated. The VCH4 and IVDMD decreased (P<0.05) compared to TC for all treatments combined or not with resveratrol, but there was no effect (P>0.05) for R3 and R6 treatments The VCH4/IVDMD was also reduced (P<0.05) compared to TC for all treatments. The N-NH3 concentration was reduced (P<0.05) for R6, R6A6 and R6T6 treatments. The C6, A6, O6 and R3C6 treatments increased (P<0.05) the concentration of N-NH3. Compared to TC, there was no effect in acetate, butyrate and the sum of VFA concentrations combined with resveratrol, with the exception of R3C6 treatments, indicating that such additives can be used to improve efficiency of feed utilization by ruminants, with toxic effect on the methanogenic microorganisms. Experiment 3 tested doses were 100, 200, 300, 400 and 600 mg/L of solution for barbatimão (B100, B200, B300, B400 and B600) and sucupira (S100, S200, S300, S400 and S600). Treatments B300, B600, S300 and S400 mg/L differed (P<0.05) from Br to VCH4. IVDMD was not affected. The VCH4/DIVMS ratio increased (P<0.05) in 33.8% for S300. The N-NH3 concentration increased (P<0.05) to S400 and S600. S300 and B100 reduced (P<0.05) acetic concentration, however S400 increased (P<0.05) in 21% the concentration of this acid. The treatments did not differ statistically from Br to propionic acid. The treatments reduced (P<0.05) butyric acid concentration from 37.6 to 76.4%. The sum of VFA also reduced (P<0.05) for both barbatimão and sucupira treatments, except to S400 treatment. Treatments with barbatimão extract caused decreased in CH4 production, but reduced SVFA, which shows that this extract in the doses evaluated, it is not recommended to improve the efficiency of use of the diet for cattle. Treatment with extract sucupira showed variation in the results of the parameters evaluated in this experiment. More studies should be conducted to elucidate the effects of this extract on ruminal fermentation.

O timol, composto natural encontrado no óleo essencial do orégano e do tomilho, apresenta efeitos benéficos como atividade antioxidante e antimicrobiana e é muito utilizado na indústria de alimentos. No entanto, seu uso pode ser limitado pela sua alta instabilidade a fatores ambientais como a temperatura, oxigênio, luz e umidade. A degradação do timol pode ser retardada pela microencapsulação, técnica em que o composto ativo é envolvido por uma matriz que protege e possibilita a liberação controlada. Dentro desse contexto, o objetivo desse trabalho foi produzir e caracterizar micropartículas de zeína carregadas com timol. As micropartículas foram obtidas pelo processo de atomização (spray-drying). A temperatura de secagem e a proporção de timol na amostra foram definidas através da metodologia de superficíe de resposta, onde as melhores condições selecionadas foram, 3,85% (g de timol/100g de zeína) e 130 °C de temperatura de secagem. Nessa condição uma eficiência de encapsulação de 49,5% foi atingida e as micropartículas obtiveram diâmetro médio de 14,3 µm de acordo com a análise por difração de feixe de laser. Em uma segunda etapa as micropartículas foram reticuladas utilizando os compostos 1-etil-3-(3-dimetilaminopropil) carbodiimida (EDC) e (1R,2R,6S)-2-Hidroxi-9-(hidroximetil)-3-oxabiciclo[4.3.0]nona-4,8 dieno-5-carboxilato de metila (genipin) e sua morfologia foi alterada, uma vez que seu formato esférico não foi encontrado após esse processo. A ligação promovida pela reticulação entre as cadeias da proteína diminuiu os sítios de ligação disponiveis, diminuindo a absorção de água em relação às não reticuladas. Isso foi evidenciado pela isoterma de sorção de água. As análises térmicas mostraram que a microencapsulação proporcionou proteção ao timol até uma temperatura acima da temperatura de degradação do composto puro. Os testes de liberação realizados mostraram que as micropartículas não reticuladas apresentaram uma liberação mais acentuada do timol chegando a 90% de liberação no primeiro dia. As micropartículas reticuladas mostraram liberação mais lenta classificada como não-fickiana, com maior resistência na taxa de relaxamento das cadeias proteicas reduzindo a liberação do timol.

Abstract : Thymol is a natural compound found in the essential oil of oregano and thyme, with antioxidant and antimicrobial activities. It is highly used in the food industry. Its use may be, however, limited due to high unstableness to environmental factors such as temperature, oxygen, light and humidity. Thymol degradation may be delayed by microencapsulation by which the active compound is wrapped in a matrix that protects and makes possible controlled release. Current research produces and characterizes microparticles of thymol-bearing zein. Microparticles were obtained by spray-drying and drying temperature and thymol percentage in the sample were defined by response surface methodology in which the best conditions were 3.85% (g thymol/100g zein) and drying temperature at 130 °C, obtaining 49.5% encapsulation efficiency. Microparticles had an average diameter 14.3 µm following laser diffraction analysis. Microparticles were then recticulated by compounds 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) and (1R,2R,6S)-2-Hydroxy-9-(hydroxymethyl)-3-oxabicycle[4.3.0]nona-4,8 dieno-5-carboxylate of methyl (genipin), with changed morphology since sphere shape was not possible after the process. Bond provided by reticulation between protein chains decreased available sites and reduced water absorption when compared with non-reticulated ones. Water sorption isotherm is evidence of the above. Thermal analyses showed that microencapsulation protected thymol up to a temperature above the degradation of the pure compound. Release tests also demonstrated that non-reticulated microcapsules had a greater thymol release, reaching 90% during the first day. Reticulated microparticles revealed a slower release, classified as non-Fickian release, with a greater resistance in relax rate of the protein chains, with a decrease in thymol release.

Computer Assisted Surgery (CAS) summarizes techniques which assist a surgeon in the preparation and conduction of surgical interventions. Today still predominantly based on radiology images, these techniques include the preoperative determination of an optimal surgical strategy and intraoperative systems which aim at increasing the accuracy of surgical manipulations. CAS is a relatively young field of computer science. One of the unsolved "teething troubles" of CAS is the absence of technical standards for the interconnectivity of CAS system. Current CAS systems are usually "islands of information" with no connection to other devices within the operating room or hospital-wide information systems. Several workshop reports and individual publications point out that this situation leads to ergonomic, logistic, and economic limitations in hospital work. Perioperative processes are prolonged by the manual installation and configuration of an increasing amount of technical devices. Intraoperatively, a large amount of the surgeons' attention is absorbed by the requirement to monitor and operate systems. The need for open infrastructures which enable the integration of CAS devices from different vendors in order to exchange information as well as commands among these devices through a network has been identified by numerous experts with backgrounds in medicine as well as engineering. This thesis contains two approaches to the integration of CAS systems: - For perioperative data exchange, the specification of new data structures as an amendment to the existing DICOM standard for radiology image management is presented. The extension of DICOM towards surgical application allows for the seamless integration of surgical planning and reporting systems into DICOM-based Picture Archiving and Communication Systems (PACS) as they are installed in most hospitals for the exchange and long-term archival of patient images and image-related patient data. - For the integration of intraoperatively used CAS devices, such as, e.g., navigation systems, video image sources, or biosensors, the concept of a surgical middleware is presented. A c++ class library, the TiCoLi, is presented which facilitates the configuration of ad-hoc networks among the modules of a distributed CAS system as well as the exchange of data streams, singular data objects, and commands between these modules. The TiCoLi is the first software library for a surgical field of application to implement all of these services. To demonstrate the suitability of the presented specifications and their implementation, two modular CAS applications are presented which utilize the proposed DICOM extensions for perioperative exchange of surgical planning data as well as the TiCoLi for establishing an intraoperative network of autonomous, yet not independent, CAS modules
Die moderne Hochleistungschirurgie des 21. Jahrhunderts ist auf vielerlei Weise abhängig von Computern oder, im weiteren Sinne, der digitalen Datenverarbeitung. Administrative Abläufe, wie die Erstellung von Nutzungsplänen für die verfügbaren technischen, räumlichen und personellen Ressourcen, die Rechnungsstellung und - in zunehmendem Maße - die Verwaltung und Archivierung von Patientendaten werden mit Hilfe von digitalen Informationssystemen rationell und effizient durchgeführt. Innerhalb der Krankenhausinformationssysteme (KIS, oder englisch HIS) stehen für die speziellen Bedürfnisse der einzelnen Fachabteilungen oft spezifische Informationssysteme zur Verfügung. Chirurgieinformationssysteme (CIS, oder englisch SIS) decken hierbei vor allen Dingen die Bereiche Operationsplanung sowie Materialwirtschaft für spezifisch chirurgische Verbrauchsmaterialien ab. Während die genannten HIS und SIS vornehmlich der Optimierung administrativer Aufgaben dienen, stehen die Systeme der Computerassistierten Chirugie (CAS) wesentlich direkter im Dienste der eigentlichen chirugischen Behandlungsplanung und Therapie. Die CAS verwendet Methoden der Robotik, digitalen Bild- und Signalverarbeitung, künstlichen Intelligenz, numerischen Simulation, um nur einige zu nennen, zur patientenspezifischen Behandlungsplanung und zur intraoperativen Unterstützung des OP-Teams, allen voran des Chirurgen. Vor allen Dingen Fortschritte in der räumlichen Verfolgung von Werkzeugen und Patienten ("Tracking"), die Verfügbarkeit dreidimensionaler radiologischer Aufnahmen (CT, MRT, ...) und der Einsatz verschiedener Robotersysteme haben in den vergangenen Jahrzehnten den Einzug des Computers in den Operationssaal - medienwirksam - ermöglicht. Weniger prominent, jedoch keinesfalls von untergeordnetem praktischen Nutzen, sind Beispiele zur automatisierten Überwachung klinischer Messwerte, wie etwa Blutdruck oder Sauerstoffsättigung. Im Gegensatz zu den meist hochgradig verteilten und gut miteinander verwobenen Informationssystemen für die Krankenhausadministration und Patientendatenverwaltung, sind die Systeme der CAS heutzutage meist wenig oder überhaupt nicht miteinander und mit Hintergrundsdatenspeichern vernetzt. Eine Reihe wissenschaftlicher Publikationen und interdisziplinärer Workshops hat sich in den vergangen ein bis zwei Jahrzehnten mit den Problemen des Alltagseinsatzes von CAS Systemen befasst. Mit steigender Intensität wurde hierbei auf den Mangel an infrastrukturiellen Grundlagen für die Vernetzung intraoperativ eingesetzter CAS Systeme miteinander und mit den perioperativ eingesetzten Planungs-, Dokumentations- und Archivierungssystemen hingewiesen. Die sich daraus ergebenden negativen Einflüsse auf die Effizienz perioperativer Abläufe - jedes Gerät muss manuell in Betrieb genommen und mit den spezifischen Daten des nächsten Patienten gefüttert werden - sowie die zunehmende Aufmerksamkeit, welche der Operateur und sein Team auf die Überwachung und dem Betrieb der einzelnen Geräte verwenden muss, werden als eine der "Kinderkrankheiten" dieser relativ jungen Technologie betrachtet und stehen einer Verbreitung über die Grenzen einer engagierten technophilen Nutzergruppe hinaus im Wege. Die vorliegende Arbeit zeigt zwei parallel von einander (jedoch, im Sinne der Schnittstellenkompatibilität, nicht gänzlich unabhängig voneinander) zu betreibende Ansätze zur Integration von CAS Systemen. - Für den perioperativen Datenaustausch wird die Spezifikation zusätzlicher Datenstrukturen zum Transfer chirurgischer Planungsdaten im Rahmen des in radiologischen Bildverarbeitungssystemen weit verbreiteten DICOM Standards vorgeschlagen und an zwei Beispielen vorgeführt. Die Erweiterung des DICOM Standards für den perioperativen Einsatz ermöglicht hierbei die nahtlose Integration chirurgischer Planungssysteme in existierende "Picture Archiving and Communication Systems" (PACS), welche in den meisten Fällen auf dem DICOM Standard basieren oder zumindest damit kompatibel sind. Dadurch ist einerseits der Tatsache Rechnung getragen, dass die patientenspezifische OP-Planung in hohem Masse auf radiologischen Bildern basiert und andererseits sicher gestellt, dass die Planungsergebnisse entsprechend der geltenden Bestimmungen langfristig archiviert und gegen unbefugten Zugriff geschützt sind - PACS Server liefern hier bereits wohlerprobte Lösungen. - Für die integration intraoperativer CAS Systeme, wie etwa Navigationssysteme, Videobildquellen oder Sensoren zur Überwachung der Vitalparameter, wird das Konzept einer "chirurgischen Middleware" vorgestellt. Unter dem Namen TiCoLi wurde eine c++ Klassenbibliothek entwickelt, auf deren Grundlage die Konfiguration von ad-hoc Netzwerken während der OP-Vorbereitung mittels plug-and-play Mechanismen erleichtert wird. Nach erfolgter Konfiguration ermöglicht die TiCoLi den Austausch kontinuierlicher Datenströme sowie einzelner Datenpakete und Kommandos zwischen den Modulen einer verteilten CAS Anwendung durch ein Ethernet-basiertes Netzwerk. Die TiCoLi ist die erste frei verfügbare Klassenbibliothek welche diese Funktionalitäten dediziert für einen Einsatz im chirurgischen Umfeld vereinigt. Zum Nachweis der Tauglichkeit der gezeigten Spezifikationen und deren Implementierungen, werden zwei modulare CAS Anwendungen präsentiert, welche die vorgeschlagenen DICOM Erweiterungen zum perioperativen Austausch von Planungsergebnissen sowie die TiCoLi zum intraoperativen Datenaustausch von Messdaten unter echzeitnahen Anforderungen verwenden

Abstract:

Undersökningens syfte är att finna svar på vilka svårigheter elever i matematik kurs B har inom algebra samt undersöka vad svårigheterna beror på. Vi vill även undersöka hur elever går tillväga när de löser algebrauppgifter och särskilt belysa vilka fel eleverna gör. För att tydligt knyta an till målen i läroplanerna konstruerade vi ett algebratest med uppgifter från de nationella proven, vilket genomfördes i en teknikklass med 25 elever. Detta test tillsammans med semistrukturerade intervjuer med tre av eleverna och två lärare kring elevernas test utgjorde vår metod för datainsamling i denna kvalitativa studie. Resultatet visar att eleverna har stora svårigheter med flera olika delar inom algebra i matematik B, exempelvis ekvationssystem, kvadratkomplettering och förenkling av uttryck. Vårt resultat visar också att svårigheterna bland annat beror på att eleverna har brister i sina algebrakunskaper från grundskolan, de använder inte de metoder som lärarna visar och de övar inte tillräckligt.

The fungi have become, over the last two decades, a major cause of illness in humans. Fungi of the genus Penicillium can be found in various substrates and affect immunocompromised people, hospitalized patients, many animals and plants, as well as compromise the quality of air indoors. The indiscriminate use of antibiotics and the resulting framework of resistance of microorganisms to conventional antimicrobial therapy has been stimulating researchers to seek alternative sources of these compounds, among them products derived by medicinal plants. Tendency to get phytochemicals from extracts, fractions, fixed or essential oils obtained from plant species is currently observed. In this context, the present study evaluated the in vitro antifungal activity of seven phytochemicals: carvacrol, citral, geraniol, linalool, pcymene, terpinolene and thymol, against twelve strains of Penicillium. Firstly, screening was carried out to find the phytochemical with the best activity by determining the Minimum Inhibitory Concentration (MIC) by the broth microdilution technique. Following, the tests were proceeded with thymol, which were: determination of Minimum Fungicidal Concentration (MFC), effect of the phytochemical on mycelial growth kinetics, interference of thymol on germination of conidia and evaluation of the influence of thymol on the fungal morphology. To this end, we selected two strains, one that was more resistant and another with a more sensitive profile. The value of thymol MIC50 and MIC90 was 128μg/ml, while the CFM value ranged from 128μg/ml to 1024μg/ml. It was observed total radial mycelial growth inhibition at the three thymol concentrations used (128μg/ml, 256μg/ml and 512μg/ml) over 14 days of exposure. In the study of the interference of thymol on the conidia germination was observed inhibitory effect. At the concentration of 512μg/mL was found greater than 80% inhibition, at 256 μg/mL inhibition was higher than 75%, while in 128μg/mL inhibition was higher than 60%, all this for both strains tested, revealing concentration-dependent inhibitory effect. In the presence of thymol, morphological changes were observed in mycelial structure, such as decrease in the amount of conidia, reduction in the formation of reproductive structures with the appearance of rudimentary reproductive structures, besides the abnormal development of hyphae. Given the above, it is concluded that thymol has antifungal activity against strains of Penicillium and therefore represents a new possibility in the arsenal of products for the treatment of infections caused by this fungus.
Os fungos tornaram-se, ao longo das últimas duas décadas, uma das principais causas de doença em humanos. Fungos do gênero Penicillium podem ser encontrados nos mais variados substratos e afetam indivíduos imunocomprometidos, pacientes hospitalizados, animais e plantas diversas, além de comprometerem a qualidade do ar de ambientes internos. O uso indiscriminado de antibióticos e o decorrente quadro de resistência dos microrganismos à terapêutica antimicrobiana convencional vem impulsionando os pesquisadores a buscarem fontes alternativas desses fármacos, dentre elas, produtos de plantas medicinais. Observa-se, atualmente, uma tendência à obtenção de fitoconstituintes a partir de extratos, frações, óleos fixos ou essenciais, obtidos de espécies vegetais. Nesse contexto, o presente estudo avaliou a atividade antifúngica in vitro de sete fitoconstituintes: carvacrol, citral, geraniol, linalol, pcimeno, terpinoleno e timol, sobre doze cepas de Penicillium. Primeiramente, realizou-se uma triagem para encontrar o fitoconstituinte com melhor atividade, através da determinação da Concentração Inibitória Mínima (CIM), pela técnica de microdiluição. A seguir, prosseguiram-se os testes com o timol, quais foram: determinação da Concentração Fungicida Mínima (CFM), efeito do fitoconstituinte sobre a cinética de crescimento micelial do fungo, interferência do timol sobre a germinação dos conídios e avaliação da influência do timol sobre a micromorfologia fúngica. Para tanto, foram selecionadas duas cepas, uma que se mostrou mais resistente e outra com perfil mais sensível. O valor da CIM50 do timol, bem como da CIM90, foi 128μg/ml, já o valor da CFM variou de 128μg/ml a 1024μg/ml. Observou-se total inibição do crescimento micelial radial, nas três concentrações de timol utilizadas (128μg/ml, 256μg/ml e 512μg/ml), ao longo de 14 dias de exposição. No estudo da interferência do timol sobre a germinação dos conídios, observou-se efeito inibitório. Na concentração de 512μg/mL foi encontrada uma inibição superior a 80%, em 256 μg/mL a inibição foi superior a 75%, enquanto que em 128μg/mL a inibição foi superior a 60%, para as duas cepas testadas, revelando um efeito inibitório dependente da concentração. Na presença do timol, foram observadas alterações morfológicas na estrutura micelial, tais como diminuição do número de conídios, redução da formação de estruturas de reprodução com surgimento de estruturas reprodutivas rudimentares, além de desenvolvimento anormal das hifas. Diante do exposto, conclui-se que o timol apresenta atividade antifúngica contra cepas de Penicillium e, conseqüentemente, representa uma nova possibilidade no arsenal de produtos para o tratamento de infecções por este fungo.
