Abstract:
Objective. To determine the in vitro antibacterial effect of four concentrations of the hydroalcoholic extract of Solanum tuberosum “tocosh” (HET) against Streptococcus mutans ATCC 25175TM. Methods. This was a prospective, experimental, comparative study. Fermented tocosh was subjected to hydric stress to obtain a hydroalcoholic extract at four different concentrations: 100%, 50%, 75%, and 25%. S. mutans strains were cultured in brain heart infusion agar using the swab technique. The antibacterial effectiveness of HET was evaluated following the Kirby–Bauer disk diffusion method and compared with 0.12% chlorhexidine (positive control group). Results. The highest mean inhibitory effect was achieved with HET at 100% (33.1 ± 2.2 mm, showing a gradual reduction in the other HET groups at 75%, 50%, and 25% (29.7 ± 1.3 mm, 26.6 ± 2.0, and 20.1 ± 1.8 mm, respectively)). Inferential analysis found statistically significant differences among all the experimental groups (p=0.001). The post hoc analysis also showed significant differences among all the experimental groups evaluated; however, there were no significant differences between HET 50% and chlorhexidine 0.12% (p>0.05). Conclusions. It was found that the highest antibacterial effectiveness was obtained by HET 100%, being even higher than the 0.12% chlorhexidine positive control, and was statistically significant. Post hoc analysis showed that almost all the concentrations showed optimal efficacy against S. mutans.

Abstract:
Potato tocosh is a naturally processed potato for nutritional and curative purposes from traditional Peruvian medicine. The aim of this study was to investigate the acute and sub-acute toxicity of tocosh flour (TF). For sub-acute toxicity, TF was administered orally to rats daily once a day for 28 days at doses of 1000 mg/kg body weight (BW). Animals were observed for general behaviors, mortality, body weight variations, and histological analysis. At the end of treatment, relative organ weights, histopathology, hematological and biochemical parameters were analyzed. For acute toxicity, TF was administered orally to mice at doses of 2000 and 5000 mg/kg BW at a single dose in both sexes. Body weight, mortality, and clinical signs were observed for 14 days after treatment. The results of acute toxicity showed that the median lethal dose (LD50) value of TF is higher than 2000 g/kg BW but less than 5000 mg/Kg BW in mice. Death and toxicological symptoms were not found during the treatment. For sub-acute toxicity, we found that no-observed-adverse-effect levels (NOAEL) of TF in rats up to 1000 g/kg BW. There were statistically significant differences in body weight, and relative organ weight in the stomach and brain. No differences in hematological and biochemical parameters were observed when compared with the control group. For sub-acute toxicity, histopathological studies revealed minor abnormalities in liver and kidney tissues at doses of 5000 mg/Kg. Based on these results, TF is a traditional Peruvian medicine with high safety at up to 1000 mg/kg BW for 28 days in rats.

Abstract:
The consumption of vegetable oil is an important way for the body to obtain tocols. However, the impact of oil types and grades on the tocopherol and tocotrienol contents in vegetable oils is unclear. In this study, nine types of traditional edible oils and ten types of self-produced new types of vegetable oil were used to analyze eight kinds of tocols. The results showed that the oil types exerted a great impact on the tocol content of traditional edible oils. Soybean oils, corn oils, and rapeseed oils all could be well distinguished from sunflower oils. Both sunflower oils and cotton seed oils showed major differences from camellia oils as well as sesame oils. Among them, rice bran oils contained the most abundant types of tocols. New types of oil, especially sacha inchi oil, have provided a new approach to obtaining oils with a high tocol content. Oil refinement leads to the loss of tocols in vegetable oil, and the degree of oil refinement determines the oil grade. However, the oil grade could not imply the final tocol content in oil from market. This study could be beneficial for the oil industry and dietary nutrition.

Abstract:
AbstractVitamin E (tocols) is a key metabolite for efficient scavenging of lipid peroxy radicals that cause membrane breakdown during seed ageing. However, in rice, this hypothesis has been tested for very few lines only and without considering intraspecific variation in genomic structure. Here, we present a correlation study between tocols and seed longevity using a diverse rice panel. Seeds of 20 rice accessions held in the International Rice Genebank at the International Rice Research Institute, representing Aus, Basmati/Sadri, Indica, temperate Japonica and tropical Japonica variety groups, were used for tocols analysis (quantification of α-, β-, γ- and δ-tocopherol/tocotrienol by ultra-performance liquid chromatography) and storage experiments at 45°C and 10.9% seed moisture content. To examine the effects of DNA-haplotype on the phenotype, the 700 K high-density single-nucleotide polymorphism marker dataset was utilized. Both seed longevity (time for viability to fall to 50%; p50) and tocols content varied across variety groups related to the heterogeneity in the genetic architecture. Among eight types of tocol homologues, α-tocopherol and γ-tocotrienol were significantly correlated with p50 (negatively and positively, respectively). While temperate Japonica varieties were most abundant in α-tocopherol, Indica varieties recorded 1.3- to 1.7-fold higher γ-tocotrienol than those of other groups. We conclude that the specific ratio of tocol homologues rather than total tocols content plays an important role in the seed longevity mechanism.

Abstract:
Three Latin American oilseeds obtained from native fruits: nopal (tuna) ( Opuntia ficus-indica), cherimoya (chirimoya) ( Annona cherimola), and papaya, Chilean variety ( Carica pubescens or C. candamarcensis) were studied for their fatty acid composition and bioactive compounds, such as tocols and phytosterols, looking for new sources of special oilseeds for this region. The results indicated that each species represents an interesting possibility. Nopal oilseed is a good source of linoleic acid (62%), with a good balance between SFA and MUFA (1:1.3). Cherimoya oilseed presents quite a different composition, with 24% SFA, 43% MUFA and 33% PUFA. Palmitic and stearic acids (15% and 7.6%, respectively) are the main SFA. A good balance between oleic acid (42.7%) and linoleic acid (31%) was observed. Papaya oilseed is a highly MUFA oil (72% with 71% oleic acid), with a very interesting composition, according to the new nutritional and technological recommendations. With respect to bioactive compounds, the main tocol in these three oilseeds was γ-tocopherol, with 136, 300 and 317 mg/kg for cherimoya, papaya and nopal oilseeds, respectively. According to the total tocol content, papaya oilseed presented the highest value with 384 mg/kg. The total amount and distribution of phytosterols was different, with values of 3092, 3554 and 5474 mg/kg for nopal, cherimoya and papaya oilseeds, respectively, with β-sitosterol forming 47.6%, 65.0 % and 78.7% of the total phytosterol fractions, respectively. From the results obtained, Chilean papaya oilseed can be considered as a very promising new source of special plant oil for different applications, followed by cherimoya and nopal oilseeds.

Abstract:
The separation and simultaneous quantitation of diastereomers of DL-α-tocopherol, eight tocol forms, and retinols (trans and cis) have been conducted by reversed-phase liquid chromatography followed by solid-phase extraction. A chiral silica stationary phase modified with polysaccharide derivative on the monodisperse macroporous silica gel (Unichiral OD-5H column, 150 mm × 4.6 mm, 5 μm, NanoMicro Technology Co., Ltd.) was employed for eluting each target compound. Instead of conventional solvent extract, a green and eco-friendly solid-phase extraction column, packing with nonpolar polystyrene divinylbenzene, was optimized in terms of capacity and solvent used in steps. Validation of the method was examined and confirmed to be satisfactory, with excellent linearity regression (r > 0.9999), acceptable accuracy (74.66%∼112.92%), and precision (0.20%∼10.52%) results. Limit of detection ranged from 0.05 mg·kg−1 (retinols) to 0.4 mg·kg−1 (tocols). The method was checked by infant formula reference material SRM 1849a as well, which illustrated good agreement of mass fraction with certified value and enriched the important isomer data.

Abstract:
This paper reviews the contribution of tocopherols and tocotrienols (tocols) to food quality as well as their bioactivity and health-promoting properties, which have attracted researchers and food technologists. Tocols are lipophilic phenolic antioxidants encompassing tocopherols that are characterized by a saturated side chain and tocotrienols with an unsaturated isoprenoid side chain. Tocols are natural constituents of several foods like dairy, vegetable oils, nuts, and grains. Their presence in foods, namely, as food additives, helps prevent lipid oxidation, which negatively affects the sensorial quality of foods, and even the nutritional value and safety. Supplementation of animals’ diets with tocopherols has proven its effectiveness in preserving fresh color and flavor of the meat. Although alfa-tocopherol displays much higher vitamin E activity than other tocols, health outcomes have been reported for tocotrienols, thus calling for more studies.

Abstract:
In the frame of African artisanal small-scale extraction of palm oil, the effects of post-harvest storage time and freezing of palm fruits on the extraction yield and quality of crude palm oil (CPO) were studied at laboratory scale using a process mimicking artisanal extraction. The extraction yield and free fatty acid (FFA) content of CPO increased with the length of fruit storage time at room temperature and freezing. FFA, total fatty acid and triacylglycerol profiles indicated a lack of specificity of the lipases at work. During post-harvest storage, the carotenoid content of CPO decreased slightly, tocopherols and tocotrienols (tocols) remained steady, while lipid oxidation remained at a very low level. For frozen fruits, carotene and tocol contents decreased sharply as a function of storage time, thought remaining quite high, but decreasing amounts of secondary oxidation products were detected. The FFA content and lipid oxidation level were highly correlated, indicating a pro-oxidant effect of FFAs. To conclude, three days of storage prior to artisanal extraction seemed the best trade-off between extraction yield and CPO quality. Combined storage at room temperature and freezing of palm fruits led to a large range of FFA contents in CPO. Optimization of the FFA contents of artisanal CPOs in line with local consumer’s demand calls for an assessment of their functional properties and sensory perception of foods formulated with CPOs.

Abstract:

Abstract:
Data about the movements of diverse objects, including human beings, animals, and commodities, are collected in growing amounts as location-aware technologies become pervasive. Clustering has become an increasingly important analytical tool for revealing travel patterns from large-scale movement datasets. Most existing methods for origin-destination (OD) flow clustering focus on the geographic properties of an OD flow but ignore the temporal information preserved in the OD flow, which reflects the dynamic changes in the travel patterns over time. In addition, most methods require some predetermined parameters as inputs and are difficult to adjust considering the changes in the users’ demands. To overcome such limitations, we present a novel OD flow clustering method, namely, TOCOFC (Tree-based and Optimum Cut-based Origin-Destination Flow Clustering). A similarity measurement method is proposed to quantify the spatial similarity relationship between OD flows, and it can be extended to measure the spatiotemporal similarity between OD flows. By constructing a maximum spanning tree and splitting it into several unrelated parts, we effectively remove the noise in the flow data. Furthermore, a recursive two-way optimum cut-based method is utilized to partition the graph composed of OD flows into OD flow clusters. Moreover, a criterion called CSSC (Child tree/Child graph Self-Similarity Criterion) is formulated to determine if the clusters meet the output requirements. By modifying the parameters, TOCOFC can obtain clustering results for different time scales and spatial scales, which makes it possible to study movement patterns from a multiscale perspective. However, TOCOFC has the disadvantages of low efficiency and large memory consumption, and it is not conducive to quickly handling large-scale data. Compared with previous works, TOCOFC has a better clustering performance, which is reflected in the fact that TOCOFC can guarantee a balance between clusters and help to fully understand the corresponding patterns. Being able to perform the spatiotemporal clustering of OD flows is also a highlight of TOCOFC, which will help to capture the differences in the patterns at different times for a deeper analysis. Extensive experiments on both artificial spatial datasets and real-world spatiotemporal datasets have demonstrated the effectiveness and flexibility of TOCOFC.

Abstract:
Saponin-rich extracts from edible seeds have gained increasing interest and their hydrolysis to sapogenin-rich extracts may be an effective strategy to enhance their potential bioactivity. However, it remains necessary to study the resulting chemical modifications of the extracts after hydrolysis as well as their impact on the subsequent bioaccessibility of bioactive compounds. The chemical composition of non-hydrolyzed and hydrolyzed extracts from fenugreek (FE, HFE) and quinoa (QE, HQE), and the bioaccessibility of saponins, sapogenins and other bioactive compounds after an in vitro gastrointestinal digestion was assessed. In general, FE mainly contained saponins (31%), amino acids (6%) and glycerides (5.9%), followed by carbohydrates (3.4%), fatty acids (FFA) (2.3%), phytosterols (0.8%), tocols (0.1%) and phenolics (0.05%). HFE consisted of FFA (35%), sapogenins (8%) and partial glycerides (7%), and were richer in phytosterols (1.9%) and tocols (0.3%). QE mainly contained glycerides (33%), FFA (19%), carbohydrates (16%) and saponins (7.9%), and to a lesser extent alkylresorcinols (1.8%), phytosterols (1.5%), amino acids (1.1%), tocols (0.5%) and phenolics (0.5%). HQE mainly consisted of FFA (57%), partial glycerides (23%) and sapogenins (5.4%), were richer in phytosterols (2.4%), phenolics (1.2%) and tocols (0.7%) but poorer in alkylresorcinols (1%). After in vitro digestion, saponins from FE and QE were fully bioaccessible, sapogenins from HFE displayed a good bioaccessibility (76%) and the sapogenin from HQE was moderately bioaccesible (38%). Digestion of saponin and sapogenin standards suggested that other components of the extracts were enhancing the bioaccessibility. Other minor bioactive compounds (phytosterols, alkylresorcinols, tocols and some phenolics) also displayed optimal bioaccessibility values (70–100%).

Abstract:
A new disease of tomatoes (Lycopersicon esculentum) in South Africa has caused severe yield losses. The causative virus, Tomato curly stunt virus (ToCSV) is distantly related to Tomato yellow leaf curl virus (TYLCV). Thirty-two tomato accessions resistant to TYLCV and 8 local cultivars were assessed for their responses to ToCSV infection. Natural infection in a field trial and a screen-house trial with controlled ToCSV-inoculation were conducted. Symptoms were rated using a severity index (SI). Yield parameters were determined in the screenhouse trial to support SI evaluations. Average SI's obtained demonstrate that TYLCV-tolerant tomato accessions, with a few exceptions, have milder symptoms of ToCSV infection than TYLCV-susceptible, sensitive accessions. None of the accessions were immune, with some virus detected in all plants. Yield losses due to ToCSV infection of TYLCV-tolerant tomato accessions were not significant (P > 0.10) with some exceptions. However, TYLCV-sensitive accessions and 8 local cultivars had significant (P < 0.10) yield losses of 49 to 100%. TYLCV-tolerant cultivars may be useful for control of ToCSV, but require evaluation prior to release or use in breeding programs in South Africa.

Abstract:

Abstract:

Abstract:
The dynamics of the world’s larger permanently open estuaries have been well documented, but much less is known about the complex dynamics of smaller temporarily open/closed estuaries (TOCEs). Seventy per cent of South Africa’s 250 estuaries are classified as TOCEs, and many are less than 50 ha. A conceptual model of the water-quality dynamics of TOCEs has been proposed, primarily considering three hydrodynamic states (closed, semi-closed and open). This paper uses data from the East Kleinemonde Estuary, a small TOCE in the warm-temperate region of South Africa, to verify the model, focusing on physico-chemical parameters (salinity, temperature, pH, dissolved oxygen and turbidity) and inorganic macronutrients (dissolved inorganic nitrogen and dissolved inorganic phosphate) for the open and closed states. The results generally fitted the model in terms of the behaviour of the physico-chemical parameters and macronutrients in the water column. This knowledge provides a first approach to predict the water-quality dynamics in small TOCEs, thereby ensuring that the often limited resources are targeted at resolving uncertainties at appropriate temporal and spatial scales. TOCEs are not unique to South Africa and also occur along other coasts (Mediterranean and south-western Australia) where this type of model could also be applied.

Abstract:
The potentially life-threatening effects of total body ionizing radiation exposure have been known for more than a century. Despite considerable advances in our understanding of the effects of radiation over the past six decades, efforts to identify effective radiation countermeasures for use in case of a radiological/nuclear emergency have been largely unsuccessful. Vitamin E is known to have antioxidant properties capable of scavenging free radicals, which have critical roles in radiation injuries. Tocopherols and tocotrienols, vitamin E analogs together known as tocols, have shown promise as radioprotectors. Although the pivotal mechanisms of action of tocols have long been thought to be their antioxidant properties and free radical scavenging activities, other alternative mechanisms have been proposed to drive their activity as radioprotectors. Here we provide a brief overview of the effects of ionizing radiation, the mechanistic mediators of radiation-induced damage, and the need for radiation countermeasures. We further outline the role for, efficacy of, and mechanisms of action of tocols as radioprotectors, and we compare and contrast their efficacy and mode of action with that of another well-studied chemical radioprotector, amifostine.

Abstract:

Abstract:
The supercritical carbon dioxide (SC-CO2) extraction technique has emerged as one of the best possible alternatives to organic solvent (hexane) extraction. However, very limited information is available on process optimization for this extraction technique and the lack of available engineering data is causing the slow growth of this technique. In the present investigation, SC-CO2 extraction was carried out to extract the oil from wheat germ under various operating conditions and the oil samples were characterized for properties such as phosphorous and tocol contents (vitamin E). A three-level Box Behnken design from response surface methodology was applied to optimize the SC-CO2 extraction parameters such as pressure, temperature and CO2 flow rate with an objective to obtain high oil yield, rich tocol contents and low phosphorous content. The process parameters were maintained between 30 to 50 MPa, 40 to 60 °C and a flow rate of 10 to 30 g·min-1 in a Box Behnken design matrix. Three different second order polynomial models were obtained for oil yield, phosphorous content and tocol contents with high R2 values. The optimum conditions were found to be 50 M Pa, 60 °C and 30 g·min-1 where the predicted oil yield, phosphorous content and tocol contents were found to be 8.87%, 31.86 mg·Kg-1 and 2059.92 mg·Kg-1 respectively. Under the optimum conditions, the experimental oil yield, phosphorous content and tocol contents obtained were found to be very close to the values predicted by the model.

Abstract:
Hybrid nanobiocomposite films are prepared using a solution casting by incorporating TEMPO cellulose nanofibrils (TOCNs) and carbon nanotubes (CNTs) into an aqueous solution of pullulan (PULL). The presence of CNT is confirmed by XRD characterization, and the prepared film shows an increased degree of crystallinity after the addition of TOCNs and CNT. The maximum degree of crystallinity value is obtained for CNT 0.5 % (59.64%). According to the Fourier-transform infrared spectroscopy, the shifts of the characteristic -OH peak of PULL occurred after the addition of TOCNs and aqueous CNT (3306.39 to 3246.90 cm−1), confirming interaction between the TOCNs, CNTs, and PULL matrix. The prepared films show enhanced material properties including higher tensile strength (65.41 MPa at low CNT content (0.5%)), water barrier properties, and reduced moisture susceptibility (5 wt.% CNT shows the lowest value (11.28%)) compared with the neat PULL film. Additionally, the prepared films are almost biodegradable within 64 days and show excellent electrical conductivity (0.001 to 0.015 S/mm for 0.5–5% CNT), which suggests a new approach to transform natural polymers into novel advanced materials for use in the fields of biosensing and electronics.

The activity of vitamin E, total content of tocols and the content of individual isomers: α-tocopherols, β-tocopherols, γ-tocopherols and δ-tocopherols was monitored in samples of hop cones of the world-important Saaz variety. Hop cone samples originated from hop-breeding area Tršice, Czech Republic. The method used for the determination of vitamin E in barley was modified and used for this quantitative analysis. The results indicate that monitored characteristics are influenced by the year of harvest (2010 or 2011) but also by the age of hop-gardens (hop bucks). High values of vitamin E activity (up to 67.79 mg.kg−1) and total content of tocols (up to 76.31 mg.kg−1) in hop cones are worth further attention from the viewpoint of alternative use of hops.

Abstract:

Abstract:
Thermococcus onnurineus NA1 is an anaerobic archaeon usually found in a deep-sea hydrothermal vent area, which can use elemental sulfur (S0) as a terminal electron acceptor for energy. Sulfur, essential to many biomolecules such as sulfur-containing amino acids and cofactors including iron-sulfur cluster, is usually mobilized from cysteine by the pyridoxal 5′-phosphate- (PLP-) dependent enzyme of cysteine desulfurase (CDS). We determined the crystal structures of CDS from Thermococcus onnurineus NA1 (ToCDS), which include native internal aldimine (NAT), gem-diamine (GD) with alanine, internal aldimine structure with existing alanine (IAA), and internal aldimine with persulfide-bound Cys356 (PSF) structures. The catalytic intermediate structures showed the dihedral angle rotation of Schiff-base linkage relative to the PLP pyridine ring. The ToCDS structures were compared with bacterial CDS structures, which will help us to understand the role and catalytic mechanism of ToCDS in the archaeon Thermococcus onnurineus NA1.
