Full textThe human 5-HT7 receptor is the latest 5-HT receptor subtype identified. Its implication in important functional roles such as learning and memory processes has been established. It is also involved in psychiatric and neurological disorders, such as epilepsy and schizophrenia. In order to increase the knowledge about the different roles of 5-HT7R, the radioligand development for this receptor has a potential interest. Taking into account the scientific literature and preliminary works performed in our laboratory, we have synthetized radioligands for Single Photon Emission Computed Tomography (SPECT) studies of 5-HT7R in the indazole series. To reach this goal, two methodologies of selective functionalisation of the indazole ring have been developed. The first method is a selective metallation of iodoindazoles at C-3 position using TMPMgCl. LiCl as base allowing an efficient insertion of thioalkyl chains. The second method is a selective iodination of 1-phenyl-1H-indazoles at ortho position via a C-H activation reaction. With these two new methods, a synthetic route has been established to obtain the desired compounds. A total of 17 compounds have been obtained. Two of them have an affinity for the 5-HT7R around 10. 0 nM. These two molecules are currently under investigation as 5HT7R SPECT radiotracers

Full textWe study the possibilities of tomographic measurements of regional Cerebral Blood Flow with a double headed gamma-camera and by the inhalation method of Xenon 133. The results showed that a speed of rotation of at least one revolution per minute and véry efficient Collimators are necessary, which the parameters are optimized. Also, we study the method of intravenous injection of 99m Tc-HM-PAO which allows non quantitative tomographic measurements of rCBF and we proposed, in this case, a « Simple » method of quantification of 99m Tc-HM-PAO slices from a previous non tomographic acquisition by the inhalation method of Xenon-133. Finally, we realized pre and post-reconstruction filtering programmes in the frequency domain by using the linear filters of Wiener and Metz in order to improve the contrast and the spatial resolution of 99m Tc-HM-PAO images obtained with the system: Rota-Camera-SIEMENS

Full textThe question of how acetylcholine (ACh) and dopamine (DA) could be involved together in the pathophysiology of some neurodegenerative disorders is essential, particularly in dementia with Lewy bodies (DLB). The present study aims at assessing an in vivo molecular imaging method of both ACh and DA brain systems using single photon emission computed tomography. In the first part of the present study, a method based on pharmacokinetic analysis making it possible to quantify ACh neurons in vivo, using [123I]-IBVM, a specific radioligand of vesicular acetylcholine transporter, was developed and validated in healthy subjects and Alzheimer’s disease patients. Then, we showed the ability of our method to demonstrate a differential alteration of ACh pathways in Progressive Supranuclear Palsy and Multiple System Atrophy patients. In the last part of this study, we imaged for the first time both ACh and DA systems in DLB patients, using not only [123I]-IBVM, but also [123I]-FP-CIT, a specific radioligand of dopamine transporter. Concomitantly, a behavioral exploration of hallucinations, fluctuating cognition and disturbances of circadian rhythms was achieved in these patients, as well as a neuropsychological examination. This study is currently in progress. The first results show consistent links between imaging and clinical data
