Abstract:
La tossina botulinica è una neurotossina prodotta dal batterio anaerobio Clostridium botulinum. L’iniezione di tossina botulinica è un trattamento sicuro ed efficace quando viene usata per inibire la funzione delle ghiandole salivari in patologie come gli sialoceli o le fistole salivari di origine post-traumatica o iatrogena. Negli scialoceli e fistole salivari, il trattamento con tossina botulinica può essere preso in considerazione anche dopo il fallimento o in aggiunta ai trattamenti conservativi non chirurgici. La terapia con tossina botulinica si sta dimostrando promettente anche nel trattamento della scialoadenite cronica. La tossina botulinica ha un elevato tasso di risoluzione nella Sindrome di Frey ed è considerata il gold standard nel trattamento di questo disturbo neurologico.

Le alterazioni congenite del sistema nervoso centrale e le patologie degenerative possono comportare disturbi a carico dell'attivitŕ muscolare sotto forma di spasticitŕ e/o distonia. Nel grave cerebroleso l'iperattivitŕ muscolare dŕ facilmente luogo ad esiti invalidanti che, sommati ad altre comorbilitŕ, si traducono in quadri molto complessi: a volte l'alterazione motoria non č compatibile con posture che consentano l'interazione con l'ambiente; in alcuni casi la stessa sopravvivenza del paziente viene messa a rischio da complicanze, come le gravi patologie dell'apparato urinario conseguenti a iperattivitŕ vescicale. Chirurgia ortopedica, tossina botulinica, baclofen intratecale, impianto di stimolatori a livello dei nuclei della base sono strumenti che possono migliorare in misura significativa la qualitŕ di vita del paziente se vengono usati tenendo ben presenti i bisogni che prevalgono nel singolo caso.

Il microbiota intestinale è un ecosistema batterico cha ha proprietà difensive per l’ospite ma che in particolari condizioni può produrre metaboliti tossici e dannosi per l’organismo. Metaboliti benefici sono gli acidi grassi a catena corta (SCAF), i metaboliti biliari ed i probiotici. Metaboliti dannosi sono la trimetilamina-N-ossidata (TMAO), i lipopolisaccaridi (LPS) e le tossine uremiche. La permeabilità della mucosa intestinale è la causa principale del passaggio in circolo di metaboliti dannosi. Il microbiota può intervenire in modo difensivo o dannoso in molte patologie cardiovascolari come la cardiopatia ischemica e lo scompenso ed in situazioni cliniche come il diabete, l’obesità, la malattia renale, la colite ulcerosa, il morbo di Chron e le malattie neurodegenerative. La dieta corretta è il cardine per mantenere una una favorevole funzionalità del microbiota.

Abstract:
AbstractDustbathing in fowls (eg chickens, Gallus gallus) consists of tossing the litter onto and between the fluffed feathers and subsequently enclosing it by flattening the feathers. The proximal contact between litters like sand and peat, and the integument is intensified by rubbing the body. This is not the case in wood-shavings which adhere to the distal plumage after tossing; rubbings in wood-shavings are frequently interrupted by reinitiated tossings. Lipids accumulate on the feathers and become stale during dust deprivation. Only baths in sand and peat are effective in removing excessive lipids from the proximal downy feather parts. Fluffiness of the down is highest in hens on peat, intermediate on sand and lowest on wood-shavings, while the reverse is true for the plumage surface temperature of the back Hens monitor the integumental lipid condition which becomes causally connected with bathing in the course of experiencing litter bathing effects. It is shown from the author's experimental study that hens naive of bathing in litter, and hens experienced with wood-shavings initially shifted bathing litters but finally preferred peat or sand (in that order) when peat, sand and wood-shavings were simultaneously presented Most of the hens experienced with bathing in sand did not shift to another substrate when presented with one. Recommendations are given concerning adequate dustbathing litter. A chronic deprivation of adequate litter leads to an uncontrollable condition of the lipids on the integument and an abnormal development of dustbathing. Such a deprivation therefore reduces animal welfare. Moreover, it is suggested to be costly.

Abstract:
The idea for this article came from a problem that was published before the widespread availability of graphing calculators (North Carolina School for Science and Mathematics 1988). In that publication, Geometric Probability, an interesting analytic solution to the problem about comparing the areas of squares was given and is described later in Algebraic-Geometric Solution. We have adapted the problem and have used successfully another, equally interesting method of solution in numerous classes from seventh-grade prealgebra through precalculus, as well as with several groups of teachers. This article presents our solution and, in addition, an example of how a rational function, a type not commonly found in applications of mathematics at this level, can model a solution of a related problem. The problem that we use with our students follows:

Abstract:
The attested association of Helicobacter pylori to gastro-duodenal pathologies such as chronic gastritis, peptic ulcer and gastric carcinoma has always been of great interest. The bacterium produces several virulence factors: the vacuolating cytotoxin (known as VacA) is definitely one of the most interesting ones, for the multiplicity of the effects on mammalian cells. For this reason VacA has been defined a multifunctional toxin [D’Elios et al., 2007; Cover et Blanke, 2005]. VacA is secreted by the bacterium as a mature protein of 95 kDa, and it can furher be cleaved in the N-terminal domain (called p37), and a C-terminal domain (called p58) which can remain non covalently linked. Secreted toxin is a flower-shaped oligomer, formed by 6 or 7 monomers, which can insert into lipid bilayers forming voltage-dependent anion channels [Tombola et al., 1999]. Several properties of VacA are strictly dependent on the formation of these channels: cellular vacuolation [Willhite et al., 2003], a rapid drop of the trans-epithelial resistance in epithelial monolayers [Papini et al., 1998], and finally apoptosis in epithelial cells [Willhite et Blanke, 2004]. In the last few years several efforts have been devoted to the characterization of the latter. It was reported that VacA intoxication is responsible for the decrease in mitochondrial membrane potential [Kimura et al., 1999] and cytochrome c release from the organelle [Galmiche et al., 2000]. Mitochondria have been reported to be the intracellular destination of VacA, as assessed when the toxin or its p37 domain were intracellularly expressed through transient transfection [Galmiche et al., 2000] and also when the toxin was extracellularly applied [Willhite et Blanke, 2004]. Mutant forms of VacA, lacking channel activity (VacA P9A, G14A) are incapable of inducing either the decrease of the mitochondrial trans-membrane potential, or cytochrome c release [McClain et al., 2003; Willhite et Blanke, 2003 and 2004]. In 2006, Yamasaki et al. showed that VacA-induced cell death and cytochrome c release depend on the activation of pro-apoptotic proteins Bax and Bak. However, the molecular mechanisms that regulate VacA-induced apoptosis and by which mechanism VacA is targeted to mitochondria remained two unanswered questions. In order to address these questions we first performed cell viability experiments and verified the involvement of pro-apoptotic proteins BAX and BAK in our cell model (Mouse Embryonic Fibroblasts, MEFs). Considering also that endoplasmic reticulum Ca2+ level is crucial to control the fate of cells in response to certain apoptotic stimuli and following sensitization of mitochondria [Breckenridge et al., 2003], we verified whether VacA-induced cell death could primarily involve the reticulum or mitochondria. As the apoptogenic role of VacA depends on its channel activity, we first performed cell death analyses inducing the intracellular expression of the full length toxin (p95), or its p37 domain where resides the membrane-channel forming ability. We also performed the same experiments with the toxin extracellularly applied: MEFs were then incubated with H. pylori supernatant containing the wt toxin, and with the supernatant containing a mutant form of the toxin, VacA P9A, which was unable to form any channel. It is known that after being endocytosed VacA localizes to late endosomes [Molinari et al., 1997], where vacuoles arise. It is not known the mechanism which allows VacA to further target mitochondria. We analyzed, though immunofluorescence and subcellular fractionation assays, the pathway followed by the toxin inside the cell, from its endocytosis to cell death. Here we report that BAX and BAK proteins are required for VacA-induced cell death; this process involves only mitochondria, and not the endoplasmic reticulum so it is Ca2+ independent. The role of BAX is crucial: it is recruited by VacA on early endosomes and activated by the toxin. Its following translocation to mitochondria, and VacA targeting to the organelle, seem to be the consequence of the cell trafficking alteration induced by the toxin. This process is characterized by the migration of endosomal membranes to the mitochondrial ones, and maybe by their fusion. All the effects so far described depend on the channel activity of VacA: it is not responsible of the release of apoptogenic factors from mitochondria, but also of all the events that precede this process.

ABSTRACT Background: per il trattamento focale della spasticità dopo l’evento di stroke lo strumento più utilizzato a livello farmacologico è la tossina botulinica, che porta alla riduzione della spasticità e di conseguenza ad effetti benefici per quanto riguarda il dolore e la funzione motoria. Un aspetto molto importante è l’approccio riabilitativo dopo l’iniezione di tossina botulinica: non è ancora stato stabilito un protocollo di riabilitazione ben definito. Obiettivo: riportare le principali evidenze disponibili in letteratura relative alle metodiche di trattamento riabilitativo del paziente con spasticità correlata ad evento di stroke in seguito all’iniezione di tossina botulinica. Metodi: la seguente scoping review ha incluso studi delle banche dati PubMed e PEDro. Gli articoli presenti trattano metodiche riabilitative su soggetti adulti con anamnesi di stroke e trattati con iniezione di tossina botulinica. Risultati: dei 573 records ottenuti dalla ricerca bibliografica, 34 sono stati inclusi nella seguente scoping review. Esistono vari approcci riabilitativi dopo l’iniezione di tossina botulinica; in particolare vengono valutati prevalentemente i trattamenti con esercizio riabilitativo, terapia fisica e tape. Tra questi, l’esercizio terapeutico risulta essere la tipologia di trattamento più considerato e con i migliori risultati. Conclusioni: non esiste una tipologia di trattamento riabilitativo standard e riconosciuto in modo univoco. L’eterogeneità degli studi, delle loro caratteristiche e dei loro risultati non permette di giungere ad una conclusione sull’efficacia di un determinato trattamento o su quale trattamento sia maggiormente indicato.

Abstract:
Introduzione: la paralisi cerebrale infantile (PCI) è una patologia che colpisce circa il 2-3,5 per mille dei bambini nati vivi nei Paesi Occidentali. La diplegia spastica è un tipo di PCI che coinvolge gli arti inferiori e, in maniera meno severa, gli arti superiori. La spasticità è l’impairment motorio più comune e l’utilizzo della tossina botulinica di tipo A (BONT-A) risulta essere un metodo efficace per il trattamento della spasticità focale. Tuttavia, risulta poco chiaro quali siano le migliori strategie riabilitative aggiuntive da adottare dopo l’inoculo di BONT-A. Obiettivo: lo scopo della presente revisione è quello di ricercare le evidenze scientifiche riportate in letteratura per il trattamento riabilitativo dei bambini con paralisi cerebrale infantile, in particolare quelli con diplegia spastica, in seguito all’utilizzo di tossina botulinica di tipo A, per quanto riguarda gli effetti su outcome come spasticità e cammino. Materiali e metodi: la ricerca è stata condotta tra il 1° marzo e il 30 settembre 2020. I database consultati per la ricerca degli studi sono stati: The Cochrane Library; Pubmed; PEDro. Le parole chiave utilizzate e combinate attraverso l’operatore boleano “AND” sono state: “Diplegic children”; “Diplegia”, “Botulinum”, “Spasticity”. Sono stati poi indicati i criteri di inclusione ed esclusione relativi al tipo di studio, di popolazione e di outcome per individuare gli studi pertinenti al quesito di ricerca. Risultati: l’analisi della letteratura e la selezione degli studi attraverso i criteri di inclusione e di esclusione hanno condotto all’individuazione di cinque studi randomizzati controllati. Conclusioni: l’analisi degli studi ha dimostrato che l’utilizzo del cast seriale alternato, del cast inibitorio e della stimolazione elettrica reciproca sono trattamenti efficaci, in concomitanza al trattamento fisioterapico e in seguito all’inoculo di BONT-A, per migliorare outcome come spasticità e cammino nei bambini con diplegia spastica.

Abstract:
Despite surgery and/or chemotherapy more than 80% of patients affected by pancreatic cancer do not survive more than 5 years after diagnosis. Gene therapy with bacterial toxins might be an effective approach for pancreatic cancer treatment. The aims of the present study were: 1. to create expression vectors, encoding highly toxic compounds, which can be activated and modulated by heat; 2. to verify, in vitro, their possible application in pancreatic cancer gene therapy. First objective The catalytic domain of diphtheria toxin (DTA) and of its variants CRM176 and CRM197 have been employed in this study as potent inhibitors of protein synthesis. To confine the expression of these cytotoxic agents to neoplastic tissue we focused our attention on heat inducible promoters, which allow a selective spatial and time control of gene induction by external heat. We chose the promoter of HSPA6 (encoding Hsp70B' protein) because it is strictly inducible, its basal expression levels being barely detectable in most tissues. We engineered three expression vectors with different heat-inducible HSPA6-derived promoter sequences driving the reporter gene eGFP: V1 containing a 473bp commercial sequence with a 91% homology to HSPA6; V2 with a 104bp sequence designed by us containing five Heat Shock Elements in tandem and the minimal promoter of HSPA6; V3 containing the 104bp sequence located downstream the 473bp promoter. In order to define the optimal heat shock temperature and exposure time we used four pancreatic cancer cell lines stably transfected with V1: the highest levels of eGFP expression (measured by Q-RT-PCR and FACS analysis) were obtained with a heat shock of 42,5°C for 1,5 hours. After setting the experimental conditions we compared the eGFP protein and mRNA levels in V1, V2 and V3 transfected pancreatic cancer cell lines. At 37°C there were minimal eGFP expression levels in cells transfected with all three vectors. After heat shock, however, V1, V2 and V3 transfected cells behaved differently: V1 and V2 transfected lines showed low and similar levels of expression (fold increase in mRNA about 8) while V3 transfected cells were highly induced by heat reaching up to 30 times the basal levels. V3, therefore, was shown to offer the best combination of high transciption efficiency and low background levels and was selected to drive the expression of DTA and its less toxic variants. Second objective The growth of all cell lines, transfected with DTA or its twenty fold less active variant CRM176, was significantly delayed even at 37°C. In other words the basal transcription levels of these toxins are sufficient to cause cell death; therefore DTA and CRM176 cannot be considered suitable candidates for gene therapy protocols using V3 promoter. At 37°C the supposedly inactive toxin CRM197 caused mild distress in transfected cells. After heat shock this phenomenon was amplified: cell growth was reduced in all CRM197 transfected cell lines. These findings indicate that the lethal effects of CRM197 are probably dose correlated. A spatial and time controlled expression of this toxin variant might offer the opportunity of combining its cytotoxic effects with its immunogenic properties, which may help antitumor immune system reaction. Conclusions The expression vector with the V3 heat-inducible promoter driving the CRM197 variant can be considered a promising starting point for future in vivo applications of pancreatic cancer gene therapy.

Abstract:
Nello studio clinico DENDR1, attivo presso la Fondazione IRCCS Istituto Neurologico Carlo Besta, 24 pazienti affetti da glioblastoma primario sono stati sottoposti a sette vaccinazioni con cellule dendritiche (DC) in combinazione al chemioterapico temozolomide (TMZ) come adiuvante. 9 pazienti hanno beneficiato del trattamento e l’aumento della sopravvivenza è correlato ad una persistente risposta mediata da cellule natural killer, ma non da linfociti CD8. Nello specifico, il trattamento con TMZ ha avuto un impatto negativo sull’attivazione di una risposta mediata da linfociti CD8 compromettendo la formazione di una memoria immunologica, un requisito fondamentale per una risposta immunitaria anti-tumorale prolungata nel tempo. La resistenza alla TMZ è risultata essere legata all’espressione di ABCC3, una proteina coinvolta nella resistenza ai farmaci, nelle cellule natural killer ma non nei linfociti CD8. Nello studio clinico DENDR2, 12 pazienti con GBM recidivo sono stati trattati con cinque vaccinazioni con cellule dendritiche in combinazione con TMZ. In questi pazienti non abbiamo trovato un’attivazione significativa dei una risposta immunitaria anti-tumorale o un beneficio in termini di sopravvivenza. Dati recenti indicano che l’iniezione intradermica dell’anatossina tetanica/difterica nel sito di somministrazione delle cellule dendritiche determina l’insorgenza di un processo infiammatorio con aumento di fattori, quali la CCL3 che sembra favorisce la migrazione delle DC ai linfonodi e di conseguenza l’efficacia della vaccinazione. Questo tipo trattamento che viene definito preconditioning. In uno studio pilota chiamato variant –(V) DENDR2, che include otto pazienti con GBM recidivo, le DC sono state somministrate a livello intradermico nella stessa area dopo 24 ore è stata iniettata la tossina tetanica al fine di predisporre un ambiente favorevole alla loro attivazione e migrazione. I pazienti arruolati in questo studio non hanno ricevuto TMZ come trattamento adiuvante. 5 pazienti, che hanno raggiunto una sopravvivenza di 9 mesi e sono stati pertanto definiti lungo-sopravviventi, hanno mostrato una forte attivazione di linfociti CD8 con formazione di memoria. Inoltre abbiamo osservato a un aumento di linfociti CD4 attivati dalla tossina tetanica parallelamente a un’attivazione di linfociti CD4 più propriamente associati all’azione delle DCs. Inoltre, il sito di vaccinazione è risultato essere molto infiltrato dal linfociti CD4 esprimenti CCL3, a supporto di una possibile maggior migrazione di cellule dendritiche ai linfonodi. Complessivamente questi dati indicano che il protocollo di somministrazione della TMZ dovrebbe essere rivalutato in modo da potenziare la risposta immunitaria con il contributo dei linfociti CD8. Inoltre, il preconditiong del sito di vaccinazione e l’assenza di TMZ permetterebbero l’attivazione di linfociti CD8 e la formazione di una memoria immunologica, supportata da un aumento di linfociti CD4 che sostengono la persistenza di una risposta anti-tumorale durante il trattamento.
In the DENDR1 clinical study, active at Fondazione IRCCS Istituto Neurologico Carlo Besta, 24 patients with first diagnosis of glioblastoma (GBM) were treated with seven vaccinations with dendritic cell (DC) combined to temozolomide (TMZ) as an adjuvant. 9 patients took advantage of this approach and the gain in survival was associated with a specific and long-lasting NK cell, and not CD8+ T cell, response. Specifically, TMZ administration limited the T cell response impairing on CD8+ T cell activation and memory formation, that is a critical requirement for a long-lasting anti-tumor immune-response. Resistance to TMZ was associated with the expression of the multidrug resistance protein ABCC3 in NK but not CD8+ T cells. In the DENDR2 clinical study, 12 patients with recurrent GBM were treated with five DC vaccinations combined with a dose-dense TMZ. These patients did not experience any significant immune response activation and survival advantage. Recent data suggested that pre-conditioning the vaccine site with the recall antigen tetanus/diphtheria toxoid induces a local inflammation with the involvement of CCL3, which improves lymph nodes homing of DC and consequently, the efficacy of DC vaccination. In a pilot study named variant (V)-DENDR2, including 8 patients with recurrent GBM, the vaccine site was pre-conditioned with tetanus toxoid (TT) 24 hours before each DC vaccination. TMZ was avoided. 5 patients, who reached OS9 and were indicated as long-term survivors, showed a strong CD8+ T cell activation and effector to memory transition. Moreover, a vaccine-specific CD4+ T cell activation was detected, accompanied by a TT- specific CD4+ memory T cell response. Moreover, the injection site was strongly infiltrated by CD4+ T cells expressing the chemokine CCL3, supporting a potential increase of DC - homing. Taken together these results suggest that the schedule of TMZ administration should be carefully reconsidered in order to improve and complete the immune response with the contribution of CD8+ T cells. Furthermore, pre-conditioning of the vaccine site and TMZ absence allowed CD8+ T cell activation and memory formation, supported by an increased frequency of CD4+ T helper cells sustaining the persistence of the anti-tumor response during the treatment.

Abstract:
I performed my doctorate research activity studying three important human pathogens that are A-B toxins: Diphtheria Toxin (DT), Tetanus Neurotoxin (TeNT) and Botulinum neurotoxins (BoNTs), the etiologic agents of diphtheria, tetanus and botulism respectively. In terms of structural organization these toxins consist of three domains, which are termed L chain (the N-terminal catalytic domain), HN (the transmembrane domain), and HC (the C-terminal binding domain). These domains are closely related to the common four step mechanism of action: membrane binding mediated by HC, endocytosis, membrane translocation mediated by HN and L-chain mediated substrate modification. I studied the conformational change of diphtheria toxin at acidic pH. DT includes a T domain which is known to mediate the pH-dependent membrane translocation, by forming a channel through which the catalytic domain crosses the endocytic vesicle membrane. To date no structural data are available about the pore/channel formed by the T domain, nor is known if it is monomeric or oligomeric. I have performed biochemical and structural studies to characterize the T domain of DT. The T domain is also considered a prospective anti-cancer agent for the targeted delivery of cytotoxic therapy to cancer cells. I obtained the crystal structure of DT in the presence of lipid bicelles (which simulate the endocytic vesicle membrane) and grown at pH 5.5, pH that mimics the acidic environment where translocation takes place. The reported structure throws lights on the initial event of this process, the destabilization of the three α-helices present at the bottom of the toxin (Leka et al., 2014). I then worked on a project which aimed to unravel the three dimensional structure of tetanus neurotoxin by crystallization studies. Because TeNT is considered “uncrystallizable” I focused on the use of antibody fragments (Fabs) as crystallization chaperons to aid the structural determination. Native gel analysis and size exclusion chromatography showed the formation of a stable complex in vitro between TeNT and the relative Fabs. Several crystallization experiments were carried out by high throughput crystallization screens. Further, I performed functional analysis on the trafficking of botulinum neurotoxin at the neuromuscular junction (NMJ). I expressed the binding domains of different BoNT serotypes, which are both necessary and sufficient for binding to the neuronal surface and internalization. The two step purifications, chromatography and gel filtration, were sufficient to yield purifications of each binding domain to >90% purity. Using cerebellum granular neurons (CGNs), I tested their functionality and specificity. I performed also in vivo assays in order to analyze their distribution along the NMJ. The data from fluorescence analysis show high specificity of these binding domains at the NMJ, and a different staining between different BoNT serotypes, reflecting their different time of intoxication, and perhaps a different pathway of vesicular trafficking.
Ho effettuato la mia attività di ricerca studiando tre importanti patogeni umani, che sono tossine di tipo A-B: la tossina difterica (DT), la neurotossina tetanica (TeNT) e le neurotossine botuliniche (BoNTs), gli agenti eziologici di difterite, tetano e botulismo, rispettivamente. In termini di organizzazione strutturale queste tossine sono costituite da tre domini: il dominio catalitico (LH), il dominio di translocazione (HN) e il dominio di legame (HC). Questa organizzazione dei domini è strettamente correlata al loro comune meccanismo d’azione che comprende: il legame alla membrane cellulare mediato dal HC, la traslocazione del dominio catalitico nel citoplasma mediata dal canale di permeazione formato dal HN. Ho studiato il cambiamento conformazionale della tossina difterica a pH acido. DT include un dominio di translocazione (dominio T), che forma il canale attraverso il quale il dominio catalitico attraversa la membrana della vescicola endosomica. Fino ad oggi non ci sono dati strutturali che riguardano il canale formato dal dominio T, non si sa neanche se è un monomero o oligomero. Ho eseguito studi biochimici e strutturali per caratterizzare il dominio T di DT. Il dominio T è anche considerato un agente anti-cancro nelle terapie mirate contro le cellule tumorali. Ho ottenuto la struttura tridimensionale della tossina difterica in presenza di doppi strati lipidici (che simulano la membrana della vescicola endosomica) ed in condizioni di pH 5,5 (pH corrispondente all'ambiente acido in cui avviene la il processo di traslocazione). La struttura riportata getta luci sull'evento iniziale di questo processo, la destabilizzazione di tre alfa-eliche presenti nella parte inferiore della tossina (Leka et al., 2014). Ho poi lavorato su un progetto che mirava a caratterizzare la struttura tridimensionale della tosssina tetanica. Poiché la cristallizzazione di questa tossina risulta d’essere molto difficile, mi sono concentrata sull'utilizzo di frammenti di anticorpi (Fab) come tools per aiutare la determinazione strutturale. Analisi da gel nativo e da cromatografia ad esclusione mostrano la formazione di un complesso stabile in vitro tra la tossina ed i relativi Fab. Diversi esperimenti di cristallizzazione sono stati eseguiti, e per il momento non abbiamo ancora informazioni strutturali sulla tossina. Inoltre, ho studiato anche la localizzazione ed il processo di internalizzazione delle tossine botuliniche a livello della giunzione neuromuscolare (NMJ). Ho espresso i domini di legame di diversi sierotipi di tossine botuliniche, domini che sono necessari e sufficienti per il legame alla superficie dei neuroni. I domini di legame sono stati purificati utilizzando cromatografia di affinità e per esclusione, ottendo alla fine una purezza > 90% . Utilizzando i neuroni granulari di cervelletto (CGN), ho testato la loro funzionalità e specificità. Questi domini sono stati iniettati in vivo al fine di analizzare la loro localizzazione a livello della giunzione neuromuscolare. I dati ottenuti con analisi di microscopia confocale ed a fluorescenza mostrano che questi domini si localizzano proprio a livello della giunzione muscolare. Nelle marcature si osserva anche una colorazione diversa tra i diversi sierotipi BoNT, e questo risultato riflette il diverso tempo di intossicazione tra i vari serotipi di tossine botuliniche, e forse anche una diversa localizzazione in diverse vescicole endosomiche.

Abstract:
In this thesis we applied proteomic techniques to basic biological topics as well as to clinically relevant issues both in diagnostic and in investigative perspectives. The procedure we employed consisted of electrophoretic separation of the relevant proteins, in-gel-proteolytic fragmentation and finally identification either of precise sites of labelling in irreversible protein PTM by Tgase 2 or of the protein itself in clinically oriented oncologic studies. In protein PTM we have analysed three substrate proteins, troponin, tubulin and mutant dyphtheria toxin. We obtained evidence that TnT was the subunit modified in native troponin and that glutamine 13 is the site of Q-labelling, while the protein is not a K substrate. Data on tubulin and DT are not yet so advanced. Both proteins are however substrates of Tgase 2. Tubulin is better substrate when in the aggregated form and is labelled in the C-terminal region. Moderate crosslinking also occurred. The protein when we focused on is the DT mutant CRM197, which instead undergoes massive crosslinkage forming protein aggregates whose accumulatioon is competed by incorporation of external amines, confirming the occurrence of g-glutamyl-e-lysine isopeptide bonds. In the oncologic studies we used the bi-dimensional electrophoresis and mass spectrometry approach to isolate specific proteins and we identified from breast cancer tissues to detect candidate disease biomarkers for diagnosis and prognosis and proteins involved in Epithelial Mesenchymal Transition in cholangiocarcinoma cell cells. In our four investigations we took particularly of the technical aspects to optimize resolution of the experimental data in both perspectives.

Abstract:
l numero di persone affette da grave insufficienza renale è in forte aumento ogni anno e con esso coloro che devono sottoporsi ad emodialisi. Tale trattamento consente la rimozione di tossine uremiche accumulatesi nel sangue per scambio di materia con una soluzione salina, il dialisato. Per singolo trattamento dialitico si consumano fino a 150 L di acqua per paziente e ciò rende tale terapia costosa nonché poco accessibile in zone con limitate risorse idriche. Il progetto di tesi propone lo sviluppo di membrane a matrice mista per la rimozione delle tossine uremiche dal dialisato esausto consentendone così un ricircolo seppure parziale e la conseguente riduzione di liquido necessario per paziente. Il primo passo è stata l’identificazione dei materiali da utilizzare come fase dispersa con migliori proprietà adsorbenti. Carbone attivo e zeoliti ZSM-5 e ZUF trattate con HCl sono risultati degli ottimi candidati. Sono poi state prodotte membrane tramite elettrofilatura di soluzioni di acido poliacrilico (PAA) con particelle di materiale adsorbente disperse. Tale tecnica consente di produrre fibre nanometriche continue e da esse membrane porose caratterizzate da un’elevata capacità filtrante e proprietà meccaniche superiori. Le immagini acquisite al microscopio elettronico mostrano fibre ben tessute e particelle di materiale adsorbente ben inglobate al loro interno. Le condizioni utilizzate sono quindi adeguate, se non ottimali, per il sistema acido poliacrilico-fase dispersa. Tuttavia occorre ancora risolvere problemi legati alla cattiva dispersione delle particelle solide all'interno del polimero e problemi di insolubilità della membrana.
