Elkin, Naomi D. "Evaluation of biomarkers for testicular toxicity." Thesis, University of Edinburgh, 2010. http://hdl.handle.net/1842/4412.

Abstract:
Non-clinical safety assessment is essential during the drug development process in the pharmaceutical industry, and involves numerous, detailed in vitro and in vivo toxicology tests (general, reproductive and genetic), and safety pharmacology studies. The testis is a common organ for adverse drug effects leading to attrition of potential compounds. It would, therefore, be useful to detect testicular toxicity as early as possible in the drug development process. Histopathology is the standard method for assessing testis toxicity, but a biomarker for ‘early warning’ detection of testicular toxic

Ecotoxicological values were collected from several databases, and screened according to quality criteria. A hierarchical QSAR approach was applied for the prediction of acute aquatic toxicity. Organophosphorus (OP) pesticide toxicity is a global health problem. Respiratory failure due to neuromuscular transmission dysfunction accounts for about 300,000 deaths annually in rural Asia. However, the clinical manifestation is complex, and described in terms of acute, intermediate, and chronic syndromes. The underlying mechanism of toxicity is still unclear. OP pesticides contain inhibitors of acetylcholinesterase (AChE), for example dimethoate, emulsified in an organic solvent, typically cyclohexanone. A hypothesized mechanism is initial excitotoxicity through inhibition of acetylcholine

Abstract:
With a view to establishing a model system for examining toxicity in skin, primary lingual keratinocyte cultures , a keratinocyte cell line and freshly isolated keratinocytes all derived from rat sub-lingual epithelium, were partially characterized both morphologically and enzymically and the toxicity and metabolism of the mycotoxin T-2, studied therein. A number of techniques for obtaining pure suspensions of sublingual keratinocytes (NCK) were examined and dispase is recommended for completely separating the epithelium from its dermis prior to trypsinization. Existing methods for the culture

Abstract:
Diclofenac was responsible for the death of millions of Gyps vultures (G. bengalensis, G. indicus and G. tenuirostris) in the Indian sub-region with the safety of the other members of nonsteroidal anti-inflammatory drugs (NSAIDs) being questionable. This has resulted in calls to test all the available NSAIDs for their vulture toxicity potential especially as studies have shown meloxicam to be safe; and ketoprofen, carprofen, flunixin and phenylbutazone as toxic. Unfortunately, due to the cost of testing, the time taken to establish toxicity reliably and the questionable ethics of repeat toxici

Abstract:
Leptinotarsa decemlineata (Say) is the most important defoliating pest of potato Solanum tuberosum L., in North America and Europe.  Management of this pest relies heavily on chemical control and insecticide resistance is a persistent problem.  This phenomenon has increased the need for developing novel insecticides, resistance evaluation, and the development of alternative control strategies regarding this insect pest.  From 2010 to 2013, field and lab experiments were conducted to evaluate the efficacy of a novel insecticide tolfenpyrad on L. decemlineata.  In leaf-dip assays, tolfenpyrad wa
