In the yeast Saccharomyces cerevisiae, trehalose is synthesized by the trehalose synthase complex in two steps. The Tps1 subunit catalyses the formation of trehalose 6-phosphate (Tre6P), which is dephosphorylated by the Tps2 subunit. Tps1 also controls sugar influx into glycolysis; a tps1 deletion strain is therefore unable to grow on glucose. It is unclear whether this regulatory function of Tps1 is mediated solely by Tre6P or also involves the Tps1 protein. We have developed a novel sensitive and specific assay method for Tre6P. AbstractThe organisation of a test performance study (TPS) involves different steps that are mostly sequential, but some may be conducted simultaneously. This chapter details the following: the steps regarding the selection of the tests to be validated; the selection of the laboratories to participate in the TPS; the preparation of the materials and the dispatch of the samples; and the completion of the TPS (including the collection and analysis of the TPS results). The reader will be able to get the detailed information on how to define and plan timeline of the TPS, the appropriate number of samples (including replicates) and of laboratories that should be included in the TPS to ensure an appropriate statistical analysis, and how to perform basic analyses of the obtained data. In addition, this chapter covers the most important critical points which can endanger successful TPS organization providing the future TPS organisers in the field of plant health (but also in other similar fields) with the possibility to identify them in advance and carry-out successful TPS.

In this book authors have used collective experience gained in the organization of 12 TPS studies, as well as previous extensive experience in the organization of similar studies to prepare a ‘textbook’ for organisers of future TPS not only in the field of plant pest detection, but also in other areas of microbiology. Abstract:
AbstractThere are two types of interlaboratory comparison studies: proficiency tests, which aim at monitoring the proficiency of laboratories, and test performance studies (TPS), which aim at evaluating the performance of (a) specific test(s) and whether it (they) is (are) fit for purpose. This booklet covers only TPS organisation. A TPS is usually organised to monitor the performance of a newly developed test to detect and/or identify pests or to compare the performance of different tests. The results of TPS also provide information on how (a) test(s) perform(s) in different laboratories. This allows a better estimation of the accuracy and reproducibility of tests. Organising a TPS is a complex process that requires considerable effort from the organisers in terms of time, expertise, and finances. Therefore, the selection of e.g. pests and of participants are important steps which are described in this chapter It also discusses the minimum criteria that the TPS organiser should meet.

High-speed aviation brings many challenges, one being the materials used ensure the aircraft and rockets travelling at hypersonic speed arrive at their destination safely. Control surfaces and thermal protection systems for vehicles flying at Mach 5 or above must withstand extremely hot temperatures and intense mechanical vibrations at launch, during cruising and re-entry into the Earth’s atmosphere. UHTCMCs (Ultra-High Temperature Ceramic Matrix Composites) belong to a new subclass of ceramic matrix composites (CMCs) with superior properties in terms of structural and chemical stability at el

Abstract Thermal protection system (TPS) plays a pivotal role in safeguarding space vehicles from extreme aerothermal heating during atmospheric entry. To ensure effective thermal management, Hybrid Thermal Protection seamlessly integrating both active and passive TPS is an innovative solution. The passive TPS consists of an insulative layer on vehicle skin to counteract the aerothermal heat. Although a thicker layer enhances insulation, a trade-off arises between the thickness of the passive layer and the spacecraft’s weight. Beneath the passive TPS layer lies the active TPS functioning as a

Text irregularities pose significant challenges to scene text recognizers. Thin-Plate Spline (TPS)-based rectification is widely regarded as an effective means to deal with them. Currently, the calculation of TPS transformation parameters purely depends on the quality of regressed text borders. It ignores the text content and often leads to unsatisfactory rectified results for severely distorted text. In this work, we introduce TPS++, an attention-enhanced TPS transformation that incorporates the attention mechanism to text rectification for the first time. TPS++ formulates the parameter calcu

Thermoplastic starches (TPS) are important bio-based, biodegradable polymers used in flexible packaging. However, their mechanical properties, processability, and high hydrophilicity limit their applications. This study examines the effects of chemical modifications and mechanical reinforcements on TPS matrices. Combinations of native and acetylated TPS, reinforced with native (CNF) and acetylated cellulose nanofibers (CNFA) at 1%, 2%, 3%, and 10%, were analyzed. TPS films were prepared with CNF using compression molding, followed by structural, morphological, mechanical, and hygroscopic analy

Starting with a review of the manufacturing scenario in the mid- to late 1980s when the Toyota Production System (TPS) gained a global foothold, the P-Insights report From Lean to Smart Manufacturing by Dr. James C. Chen details how the TPS has evolved by incorporating elements of smart manufacturing, technological advances, and the insights of employees. Relationships among the TPS, lean, kaizen, digitalization, Industry 4.0, and smart manufacturing are analyzed in different manufacturing scenarios. Concluding with two brief case studies, the report provides examples for manufacturers to cons
