A LesÃo Central de CÃlulas Gigantes dos maxilares (LCCG) à intra-Ãssea, nÃo tem predileÃÃo por sexo, classifica-se em agressivas e nÃo-agressivas, histologicamente consistem tecido fi-broso e celularizado fusiforme associado a cÃlulas gigantes multinucleadas (CGM), focos de hemorragia e neovascularizaÃÃo, tendo na cirurgia seu habitual tratamento. Novas abordagens terapÃuticas foram propostas, sendo a principal delas o uso de corticÃides intralesionais. Este trabalho analisa retrospectivamente 21 pacientes portadores de LCCG que foram tratados por hexacetonido de triancinolona intralesional, atravÃs do seguinte protocolo: injeÃÃo de hexace-tonido de triancinolona 20mg/ml diluÃdo na soluÃÃo anestÃsica de lidocaÃna 2%/epinefrina 1:200.000 numa proporÃÃo de 1:1; infiltrando 1ml de soluÃÃo para cada 1cm3 de lesÃo, totali-zando 06 aplicaÃÃes em intervalos quinzenais. Estabeleceu-se 04 critÃrios clÃnicos para classi-ficar a resposta ao tratamento: 1- estabilizaÃÃo ou regressÃo clÃnica da lesÃo 2- ausÃncia de sintomas 3- aumento da densidade nos controles radiogrÃficos 4- aumento da resistÃncia a infiltraÃÃo intralesional da droga, bem como, fez-se uma anÃlise imunohistoquÃmica quanto à expressÃo dos Receptores de corticÃides (GCR) e Calcitonina (CTR), Cox-2, proteÃna p16 e amplificaÃÃo gÃnica da Ciclina D1 por CISH, comparando quanto a agressividade e a resposta terapÃutica a corticoterapia intralesional. Dos 21 pacientes incluÃdos neste estudo, 11 eram homens e 10 mulheres, 09 tinham lesÃo em maxila, 12 em mandÃbula. Dez eram lesÃes agres-sivas e 11 nÃo-agressivas, 15 (71,4%) apresentaram uma boa resposta ao tratamento, 04(19%) moderada e 02(9,1%) negativa. Das 11 nÃo agressivas, 10(90,9%) apresentaram boa resposta e 01 (9,1%) resposta moderada, das 10 agressivas 05(50%), 03(30%) e 02(20%) apresentaram boa, moderada e negativa resposta respectivamente, nenhuma apresentou recidiva apÃs o tra-tamento, com preservaÃÃo que variou entre 04 a 08 anos. Os achados histopatolÃgicos mos-traram uma reduÃÃo da densidade e do tamanho das CG, e um estroma fibro-colagenoso das lesÃes. Dentre os marcadores pesquisados, apenas GCR em CG antes do tratamento mostrou significÃncia estatÃstica (p<0,004) com relaÃÃo a uma boa resposta terapÃutica. O CTR ex-pressou-se em cÃlulas gigantes e mononucleares de forma variada. A p16 apresentou-se ex-pressa em 30% da amostra, COX2 nÃo apresentou expressÃo na lesÃo e 33% da amostra apre-sentou amplificaÃÃo gÃnica da ciclina D1. NÃo mostraram significÃncia estatÃstica nem quanto à agressividade, nem quanto resposta ao tratamento, nenhum dos marcadores, exceto o GCR. O estudo mostrou que a corticoterapia intralesional à efetiva e segura para o tratamento das LCCG, com tendÃncia a melhor resposta nas lesÃes nÃo-agressivas do que nas agressivas. Mostrou ainda que a marcaÃÃo para GCR em CG demonstrou ser um parÃmetro confiÃvel para prever a resposta à terapÃutica com a corticoterapia intralesional e que 33% das LCCG tÃm comportamento neoplÃsico pela amplificaÃÃo gÃnica da ciclina D1.
Central Giant Cells Lesion (CGCL) of the jaws is an intra-bone lesion with no predilection for sex and clinically divided into aggressive and non-aggressive subtypes. Histological, it shows as fibrous tissue with fusiform cells, as well as multinucleated giant cells (GC) clusters, he-morrhagic foci and neovascularization. Surgery is the regular treatment option. As new the-rapeutic approaches have been proposed, intralesional glucocorticoid injection is the main option. This paper assesses retrospectively 21 patients presenting CGCL, treated with intrale-sional triamcinolone hexacetonide by using the following protocol: intralesional injection of triamcinolone hexacetonide 20mg/mL, diluted in a solution of lidocain 2% plus epinephrine 1:200000, at a 1:1 proportion; 1mL of this final solution for each 1cm3 of lesion volume was the injected, with a total of 06 injections, one in every 15 days. Four clinical criteria were sta-bilished to evaluate treatment outcome: 1- Clinical regression or stabilization of the lesion; 2- Absence of symptoms; 3- Raising in density on radiographic controls; 4-Increased resistence when injecting the drug intralesionally. It was also performed immunohistochemical assess-ment for glucocorticoid receptor (GCR) expression, calcitonin receptor (CTR) expression, COX-2 expression, p16 expression and Ciclin D1 gene amplification by CISH, making com-parisons related to aggressivity and to therapeutic outcome. Eleven out of 21 patients of this study were women, and 10 were men. Nine of the patients had lesion located in the maxilla, 12 in the mandible. Ten patients showed aggressive lesions and 11 non-aggressive lesions. Fifteen patients showed good treatment outcome, four patients showed moderate outcome, and two patients showed negative answer to the treatment. Among the 11 patients with non-aggressive lesions, ten showed good outcome and the other, moderate outcome. Among the ten aggressive lesions, five patients showed good outcome, three patients showed moderate outcome and the remaining two patients showed negative answer to the treatment. None of them showed reicidive in a four to eight years follow-up period. Morphologic analysis found positive correlation between volume density of GC/mm2 and lesion aggressiveness, as well as significant reduction in number of GC/mm2 after treatment. Among the markers, only GCR in GC showed statistical relevance associated to the treatment. CTR was espresse in GC and in mononuclear cells in a varying way; p16 was expressed in 30% of the sample; COX-2 was not expressed at all in lesion samples and 33% of the sample showed gene amplification in Ciclin D1. None of the markers showed any statistical significant difference related to aggres-siveness nor to treatment outcome, except for GCR. The study showed the feasibility of the adopted treatment, with tendency to better outcomes in non-aggressive lesion, if compared to the aggressive ones. It also showed evidence pointing to GCR expression in GC as a reliable parameter to predict therapeutic responsiveness to glucocorticoids; and it showed that 33% of CGCL have neoplastic behaviour by Ciclin D1 gene amplification.

The aim of the diploma thesis is to design the extension of tram line no. 12 from the current terminal station Technologicky park to the city district Brno-Medlanky. The tram track is situated on the separate road bed outside the built up urban area. Directional and elevational solution of the track is designed in several variants, after the evaluation the best variant is selected and elaborated. The diploma thesis contains the design of the tram stop and the terminal station with baloon loop, including access paths. Adjustment of the crossed road and the drainage system solution is part of the thesis as well.

News coverage of high profile criminal matters has increased in South Africa. Such matters are of public concern, as every citizen has a right to receive and impart information and to debate openly and frankly matters which are of public concern, including matters before the courts. The legitimacy of the courts is dependent on robust media reportage and public scrutiny of judicial matters which such reportage stimulates. However, criminal trials of high profile accused persons such as Oscar Pistorius, Shrien Dewani and J Arthur Brown, turn easily into a show with strong entertainment value, giving the media strong profitmaking reasons to cover it. In their pursuit of profit and in seeking to satisfy the curiosity of their readers, listeners or viewers, the media regularly resort to trial by media or adverse pre-trial publicity. Trial by media is nothing more than commercially motivated expression which does not warrant constitutional protection. At the receiving end of such coverage are accused persons. Public censure of crime and of accused persons which follows trial by media should not be imposed on the innocent. The right to a fair trial requires that an accused be treated fairly from the inception of the criminal process, from which point the person suspected of committing the crime in question is considered innocent. Any pre-trial process which implies that the accused is guilty, including any such process influenced by media reports surrounding criminal offences, violates the presumption of innocence. Despite the availability of remedies, the media in South Africa usually are not held to account for their actions and persist with adverse, biased and irresponsible pre-trial reporting. Courts have shown a tendency to protect the media in these cases, despite the effect of such reporting on the judicial process, the administration of justice and the fair trial rights of accused persons. The reason for this is usually the hesitation on the part of judges to recognise their susceptibility to extraneous matters. Judges should not be placed in a position where their independence and impartiality are questioned as a result of media sensationalism. Where the media create mistrust in the integrity of the judiciary, the rule of law is in peril.

Four trap types were used to trap boll weevils at 1 m heights, with and without grandlure, and 2 trap types were compared at 0.5 m height, with and without grandlure. The results indicate trap height may be an important factor in boll weevil catches at certain times during the cotton season. Also trap color, design and other factors may influence trap catches since traps not baited with grandlure caught high numbers of boll weevils during certain times during the cotton growing season.

Abstract:
This research investigates the influence of layout and design on the severity of trail degradation. Previous trail studies have been restricted by relatively small study areas which provide a limited range of environmental conditions and therefore produce findings with limited applicability; this research improves on this limitation by analyzing a representative sample of the Appalachian Trail with significant ecological diversity. Most trail science studies have also focused on a singular form of trail degradation, whereas this study investigates trail soil loss, widening and muddiness, providing a more cohesive analysis and revealing interrelationships between trail degradation processes. ANOVA testing of the mean values of three trail impact indicators for trail transects within several trail layout frameworks confirms the broad relevance of core trail design principles, specifically the sustainability advantages of trails with low grades and side-hill alignments. Findings also reveal the importance of landform grade in determining the susceptibility of trails to degradation and the influence of routing decisions; these relationships have received relatively little attention in the literature. The results also reveal several methodological considerations for trail alignment metrics and trail impact indicators.

Natural surfaced trails are an essential infrastructure component in parks and protected natural areas. They provide transportation routes through otherwise undeveloped areas and outdoor recreation opportunities for hikers, mountain bikers, and equestrians. Over time, recreational use and natural processes such as rainfall can lead to negative ecological impacts that damage trail treads in ways that impair their utility for visitors and require costly repairs. Environmental factors like unstable soils or extreme precipitation can make trails more susceptible to degradation. However, sustainable trail layouts and effective maintenance can reduce the rate and severity of degradation. This research investigates the influence of trail layout on three chief forms of trail degradation: trail soil loss, muddiness, and widening. Many trail science studies have occurred in small protected natural areas where the limited range of represented environmental conditions reduces the applicability of their findings in dissimilar settings. This study investigates a dataset from a large and ecologically diverse representative sample of the entire Appalachian Trail from Georgia to Maine which significantly broadens the relevance of its findings. Furthermore, many previous trail studies have focused on single forms of trail degradation whereas this study which investigates three, which provides a more cohesive analysis and reveals interrelationships between impacts. Findings confirm the broad pertinence of core sustainable trail design principles, specifically the benefits of low trail grades and side-hill alignments, and suggests that landform grade is an important factor which has received little attention in the literature. The study also revealed several methodological improvements and considerations which may be useful to trail scientists and practitioners.

Abstract:
Basic economic theory suggests that the decision to go to college should be based only on the expected costs and benefits of college. The income of the family the student comes from should have no effect. Yet, it does. The two common explanations for this discrepancy, inadequate primary school funding and liquidity constraints are at odds with the facts. I offer a third explanation, economically disadvantaged students attend college at lower rates because they have biased information. This analysis connects to the existing literature in at least three ways. It provides a rational basis for the neighborhood effect., extends work on human capital development indicating that educational paths are set at or before age 16 and helps provide an explanation for the both the increase in the return to education and the slowdown in college graduation growth among young men in the United States.

Eleven head lettuce varieties and four leaf lettuce varieties were grown in a replicated small plot trial on the Safford Agricultural Center in 1998. Desertgreen variety of head lettuce from Harris Moran produced a crop with the best head weight, size and firmness rating. Premiere variety followed closely behind. Of the leaf lettuce varieties tested, Saguaro Romaine produced the best quality and quantity product. Per acre yields are calculated for each variety in the study.

Willing to overstep literary conventions in order to ensure that meaning and purpose reign over structure, cross-genre writing works to push boundaries of genre and tear down the walls of limitation. This cross-genre thesis aims to test literary restrictions of structure and style and, as literary endeavors often do, to rattle our existence. In this thesis, nonfiction and fiction work together to drive meaning to the surface of the page, meaning that is universal in the individual stories as well as in the human experience. Although some characters are fictional and some real, they often intersect, their journeys and discoveries merging into one. The many voices of this thesis, while diverse, speak to similar themes and meaning. The main character of "Silhouettes," a homosexual male who yearns to find his identity away from the place he once called home, experiences feelings of abandonment and loss. The narrator of "A Crimson Trail" longs to uncovers truths about her uncle's suicide and endures similar feelings of loss. "Abandoned Laurels" explores a complex mother-daughter relationship and wades through themes of mourning, regret, and shame. The remaining stories explore similar themes, including those of longing, death, and familial relationships. Shorter pieces are scattered amongst longer works and supplement themes developed in the thesis. Each section contributes to the characters' longing for identity, recovery, and understanding of the past. These related characters and their stories--both real and fictional--merge in a collective endeavor to sift through loss, explore the past, and, most importantly, find identity and hope in the future amidst the rubble of the present.

Abstract:
Obesity is one of the largest health problems our nation currently faces. There are many contributing factors to the development of obesity, including genetics, lifestyle, diet, social and economic status, and lack of education or resources. In the rural East Tennessee community of Johnson County, lack of exercise has been identified as a major contributor to obesity. In addition, community members have expressed concern of a lack of knowledge of public places to exercise, such as walking paths and hiking trails. Previous research has shown that access to educational materials, such as informational brochures and maps, can increase knowledge of exercise locations. Our hypothesis, based on the Theory of Planned Behavior, is that creating and placing brochures in various locations throughout the community will normalize and encourage the use of these locations to increase exercise and ultimately improve the health of the community. An initial survey was sent to community members via email to determine what content should be included in the brochure. In conjunction with the Johnson County Health Council, a list of all walking paths and trails in the county was compiled. Each location was then visited to take note of the amenities available, the difficulty and the length of each path, and the hours available to the public. This information was then formatted into easy-to-read bullet points and grouped by geographic region in the county and a cartoon map developed. The completed brochure was then evaluated for effectiveness at increasing awareness of local paths and trails. Participants at a local doctor’s office were asked to complete a four question pre-survey assessing their current knowledge of walking paths and trails. The brochure was then given to the participants and sufficient time allowed to look through it. Participants completed an eight question post-survey which assessed any change in knowledge which occurred as a result of the brochure. The study was reviewed and approved by the ETSU Institutional Review Board. Preliminary results (n=6) show that the brochure is effective at increasing awareness and participants are more likely to visit one of the listed trails in the next month as a result. Further data collection is currently underway. By using a community based participatory approach, the specific needs of the community could be addressed. One drawback of this study is that it does not assess actual changes in trail usage or physical activity. However, it is hoped that the creation of this brochure and its use in the Johnson County community will lead to a more active community and will inspire future health initiatives.

L'hormonothérapie permet de contrôler le cancer de la prostate métastatique en induisant la régression des cellules cancéreuses en provoquant leur apoptose. Malheureusement la pathologie face à ce type de traitement évolue alors vers une hormono-indépendance qui sera fatale à plus ou moins court terme au patient. L'étude des gènes impliqués dans l'apoptose et sensibles aux androgènes permettrait une meilleure compréhension de cet échappement hormonal actuellement inéluctable. Dans cette thèse nous avons étudié la relation existante entre le ligand TRAIL et ses récepteurs et les androgènes au sein de la prostate. Notre premier travail a consisté, grâce à l'utilisation d'un modèle animal pertinent, de mettre en évidence la présence de TRAIL et des ses récepteurs dans la prostate ventrale de rat et d'étudier la relation système TRAIL/androgène dans ce même organe. Il apparaît intéressant de noter que le récepteur leurre DcR2 est sensible aux androgènes, une suppression de cette hormone induisant alors une diminution d'expression corrélée avec une augmenetation d'apoptose. Ce premier travail nous permet d'émettre l'hypothèse qu'une augmentation d'apopotose induite par le ligand TRAIL peut danc certains cas être due à la diminution d'expression d'un récepteur leurre, DcR2 dans notre cas, plutôt qu'à la surexpression de récepteurs de mort. Le changement du ratio récepteur leurre-récepteur de mort devient alors plus favorable à l'entrée des cellules en apoptose. Dans un second travail nous avons étudié l'androgénodépendance du système TRAIL dans un modèle de cancer prostatique androgénosensible humain, la lignée cellulaire LNCaP. Il apparaît alors que DR5 et DcR2 subissent une augmentation de leur expression pour de fortes concentrations d'androgènes, corrélée avec une absence d'apoptose. L'apoptose générée par le ligand TRAIL sur ces cellules dépend de la concentration d'androgène présente. Par utilisation du siRNA nos travaux montrent que le niveau d'expression des récepteurs DR5 et DcR2 influence l'apoptose induite par le ligand TRAIL et plus particulièrement la possibilité d'induire ou d'augmenter l'apoptose lorsque le récepteur DcR2 est sous exprimé. Ce travail de thèse permet de mieux comprendre la relation qui existe entre l'apoptose induite par l'homonothérapie et le système TRAIL. Cela pourrait contribuer à l'étude de nouvelles stragégies thérapeutiques

While many people enjoy hiking as a weekend activity, to many outdoor enthusiasts there is a hiking culture with which they feel affiliated. However, the way that these cultures interact with each other is still unclear. Exploring these different cultures and understanding how they relate to each other can help in engaging stakeholders of the trail. This is an important step toward finding ways to encourage environmentally friendly outdoor recreation practices and developing hiker-approved (and environmentally conscious) technologies to use on the trail. We explored these cultures by analyzing an extensive collection of tweets (over 1.5 million). We used topic modeling to identify the topics described by the communities of Triple Crown trails. We labeled training data for a classifier that identifies tweets relating to depreciative behaviors on the trail. Then, we compared the distribution of tweets across various depreciative trail behaviors to those of corresponding blog posts in order to see how tweets reflected cultures in comparison with blog posts. To harness metadata beyond the text of the tweets, we experimented with visualization techniques. We combined those efforts with ethnographic studies of hikers and conservancy organizations to produce this exploration of trail cultures. In this thesis, we show that through the use of natural language processing, we can identify cultural differences between trail communities. We identify the most significantly discussed forms of trail depreciation, which is helpful to conservation organizations so that they can more appropriately share which Leave No Trace practices hikers should place extra effort into practicing.

This diploma thesis deals with the possibilities of building a tram track between the Prague districts of Suchdol and Bohnice. It summarizes basic facts about public transport in general and in Prague, mentions the organization of the Prague Integrated Transport system and its financing. The practical part deals with the specific project of the tram track connecting Suchdol and Bohnice using the already designed tracks Kobylisy - Bohnice and Podbaba - Suchdol. Moreover, the nearby Troja area is added to the project. Based on current bus traffic data, new tram lines serving these areas are suggested. At the final stage, the project is complemented by the new P + R parking lots and the possibilities of their use.

Sixteen sweet corn varieties were grown in replicated small plot trials with two dates of planting at the Safford Agricultural Center in 1999. Candy Corner ranked number one and two in the early and late dates of planting, respectively, and FMX 413 ranked number one in the later date of planting study. Both varieties produced over 2,500 dozen ears per acre. Sugar readings were made and tabulated for all varieties using a portable refractometer.

Richard Feynman first proposed the idea of quantum computers thirty years ago. Since then, efforts have been undertaken to realize large-scale, fault-tolerant quantum computers that can factor large numbers much more quickly than classical computers, which would have significant implications for computer security. While there is no universally agreed upon technology for experimentally realizing quantum computers, many researchers look to ion traps as a promising technology. This thesis focuses on ion traps, how they fulfill the Divincenzo criteria, what obstacles must be overcome, and recent achievements in this field. We examine the physical principles of a linear Paul trap, including the confining potential and its quantum dynamics. In addition, we built a mechanical analogue of an ion trap for pedagogical purposes, and we provide an analysis of its trapping potential and compare it to a real ion trap, the Paul trap. Furthermore, we provide guidance for building a course module on ion trap based quantum computing; our guidance is based on course materials from several institutions.

Abstract:
The distillation point efficiencies for the alcohol-water binary, ternary and quaternary systems were measured using a modified Oldershaw column. This column is expanded above the tray to separate the newly formed bubbles from the column wall, thus eliminating the surface tension induced wall effects for positive systems and discouraging wetted wall effects. The excessive and recirculating foam and froth found in the conventional Oldershaw column is due to these wall effects and does not represent conditions in large scale distillation. The point efficiencies measured using this column for the system methanol/water were lower than the point efficiencies deduced from the composition profiles across a large and narrow rectangular distillation column using an eddy diffusion model. The narrow rectangular column had a liquid flow path length of about one meter, thus avoiding stagnant zones and flow non-uniformities. The lower efficiencies were due to the shorter contact time between the gas and the liquid. This contact time was increased markedly by fitting an outlet weir to the modified Oldershaw column, thus increasing the tray liquid hold-up and the point efficiencies. These point efficiencies were about 10 per cent lower than those on the large tray at a similar value of the F. Factor. The eddy diffusion model predicted rectangular tray efficiencies about 10 to 20 per cent lower than those measured, when using the improved modified column point efficiencies. Using a suitable model, the improved point efficiencies were scaled-up to the conditions existing on the rectangular tray. This resulted in the large tray values of 2 to 4 per cent lower tray efficiencies than those measured. The surface tension effect on the point efficiencies of the binary systems MeOH/n.PrOH using the original modified Oldershaw column in the absence of wall effects using the concept of the Marangoni stabilising index. The surface tension of these systems were measured using a glass thermometer. The system MeOH/H20 had the highest Marangoni index and showed the highest point efficiencies throughout the composition range, with the EtOH/H2O/n.PrOH, with low values of the Marangoni index, showed comparable point efficiencies throughout the composition range. These systems demonstrate all the possible types of surface tension behaviour. The effects of the outlet weir height and hole size on the point efficiencies in the rectangular column operating under similar hydrodynamic conditions were also investigated using the system MeOH/H2O. There was an increase in point and tray efficiencies on increasing the outlet weir height from 2 mm to 12.7 mm. There was also small increase in point and tray efficiencies on decreasing the hole size from 6.4 mm to 1 mm at the expense of higher pressure drops. The point efficiencies of these trays under different hydraulic conditions were in the range 85 to 95 per cent, with subsequent high tray efficiencies. This provides further evidence of the high tray efficiencies available to the design engineer if the detrimental effects of stagnant zones and flow non-uniformities were eliminated. Two highly non-ideal ternary systems and quaternary system were also studied using the original modified Oldershaw and the rectangular columns. Considerable differences between the individual component point efficiencies were observed. These differences are probably caused by the interactive nature of the mass transfer in these systems. These systems also exhibited equal component point efficiencies in parts of the composition range, which illustrates the composition dependency of these systems. The individual component tray efficiencies for these systems were noticeably different, even with equal component point efficiencies operating across the tray. These differences were simulated using the eddy diffusion model, highlighting the effects of limited liquid back mixing on the tray. The composition profile for the system MeOH/EtOH/H20 were predicted and compared with the measurements across the rectangular column using three methods derived from the original Maxwell and Stephan mass transfer equations. These predictions were in good agreement with the measurements. However, as the comparison is only based on a one meter flow path length, the actual design of distillation column using these methods would be conservative. The prediction of the composition profiles using the point efficiencies from the original version of the modified 0ldershaw column yielded a similar observation for both the ternaries and the quaternary system. An expanded aluminium tray (Expamet 607A) was also subject to preliminary efficiency tests in the rectangular column. This material has corrugated angled holes, thus encouraging the liquid flow across the tray by using the vapour momentum. This material showed much lower pressure drops, due to its high open area compared with conventional sieve trays, and discourages weeping and entrainment.

Malignant pleural mesothelioma (MPM) is a devastating disease for which limited effective therapies are currently available. Tumour necrosis factor-related apoptosis-inducing ligand (TRAIL) and other death receptor (DR) agonists are pro-apoptotic agents that trigger the extrinsic apoptotic pathway selectively in cancer cells. Previous work has shown that supports that loss of function of the nuclear deubiquitinase BRCA associated protein-1 (BAP1) augments sensitivity to recombinant TRAIL (rTRAIL) in MPM cells. This study shows that BAP1 is a candidate biomarker for rTRAIL sensitivity in cell line, early passage culture and tumour explant models of MPM. In addition, BAP1 is a potential biomarker for sensitivity to other DR agonists and for additional cancer types with BAP1 mutations. I have also identified other novel candidate biomarkers for DR agonist sensitivity, notably ASXL1/2, through exploration of the mechanism underlying the BAP1-rTRAIL association. I present data supporting the clinical relevance and utility of the BAP1-TRAIL association. I have shown that loss of BAP1 function occurs in a significant proportion of MPM tumours in the UK and that loss of BAP1 function augments sensitivity to TRAIL in primary tumour tissue. I describe in vitro data supporting the hypothesis that loss of BAP1 function augments sensitivity to rTRAIL in MPM. I have shown that loss of BAP1 function augments sensitivity to other DR agonists and that BAP1 can act as a biomarker for DR agonist sensitivity in other cancers with BAP1 mutations. Finally, I explore the mechanism underlying the BAP1-TRAIL association. I present data supporting the notion that BAP1 binds to the ASXL1/2 proteins to form the polycomb repressor deubiquitinase complex which underlies BAP1-induced TRAIL resistance. Loss of this function results in a change in the expression of proteins of the extrinsic apoptotic pathway, which may favour apoptosis upon DR agonist binding.

When planning for road building and infrastructure, simulations of tra-c  ow are useful tools for further investigation. Two models are analyzed in this report, both supported by the assumption that tra-c  ow behaves as a continous  uid on a macroscopic scale. This means that the vehicle density will be considered instead of observing the vehicles as discrete objects. Conservational laws along with piecewise constant initial data are called Riemann problems. Tra-c  ow in one dimension at a tra-c light can be modeled as such a problem with the continuity equation and a scaled Heaviside function as initial data. Two special cases of the general problem are solved, in the rst model the velocity of the cars is a linear function of the density while in the second model the velocty is a function of the denisty and its derivative. The rst case is solved analytically with a similarity solution. The second case, where diusion is included, is solved numerically and the method utilized is proved to be stable. Simulations are presented in order to see how the density of vehicles develop in front of and after the tra-c light. The results are used to determine a time for the tra-c light to be green, given a xed time for red, that optimzes the  ow of vehicles.
