Conjugation is a fundamentally important mechanism of horizontal DNA transfer between bacteria, bacteria x archea, and bacteria x eukaryotes. This work has concentrated on conjugation between bacteria x eukaryotes, specifically Escherichia coli x Saccharomyces cerevisiae. Teil werden die Untersuchungen des ACDS-Komplexes aus A. fulgidus mittels Transmissionselektronenmikroskopie (Negativkontrastierung und der Kryo-Einbettung) geschildert. Die 3D-Rekonstruktion mit einer Auflösung von 29 Å wird de novo ermittelt und drei mögliche Positionen für die CODH-Untereinheit vorgeschlagen. Im 2. Teil wird die Röntgenkristallstrukturanalyse der CODH-Untereinheit des ACDS Komplexes aus A. fulgidus geschildert.

The aim of this thesis is to investigate the methodology of three dimensional chemical imaging of nanoparticles through the use of scanning transmission electron microscope (STEM) – energy dispersive X-ray (EDX) spectroscopy.
