Muscle-derived stem cells (MDSCs) have been successfully transplanted into both skeletal (1) and cardiac muscle (2) of dystrophin-deficient (mdx) mice, and show potential for improving cardiac and skeletal dysfunction in diseases like Duchenne muscular dystrophy (DMD). Our previous study explored the regeneration of dystrophin-expressing myocytes following MDSC transplantation into environments with distinct blood flow and chemical/mechanical stimulation attributes. After MDSC transplantation within left ventricular myocardium and gastrocnemius (GN) muscles of the same mdx mice, significantly more dystrophin-positive fibers were found within the myocardium than in the GN. We hypothesized that the differences in mechanical loading of the two environments influenced the transplantation and explored whether using MDSCs exposed to mechanical stimulation prior to transplantation could improve transplantation. Our study shows increased engraftment into the heart and GN muscle for cells pretreated with mechanical stretch for 24 hours. This increase was significant for transplantation into the heart. These studies have implications in a variety of applications including mechanotransduction, stem cell biology, and Duchenne muscular dystrophy.

Abstract:
Organ transplantation is a form of a specialty treatment aimed at saving, prolonging and improving the quality of life of gravely ill patients. Although the organ transplantation is a field of medicine, this procedure is connected to the need for respecting and protecting of a number of individual rights which makes it also the subject of the regulatory framework. In that context, the goal of this paper is to investigate the principles which lay the grounds for the procedure of human organs transplantation in Serbian law and to answer the question whether the provisions of Serbian Law on human organ transplantation allow the adequate implementation of these principles. In order to reach this goal, the author of this paper compares the national concepts on this matter to those adopted in German and Croatian law, as well as to those foreseen in the European supranational law passed by the Council of Europe. Following a detailed analysis of the meaning and the scope of influence of each of four major Serbian law principles of human organ transplantation on the protection of basic human rights and dignity of both the organ donor and recipient, which constitutes the major part of the paper, in the concluding remarks the author presents her recommendations for the improvement of the existing norms regulating this matter.

Abstract:
In the relatively young field of cardiac tissue engineering, different biomaterials, methods and techniques have been tested for cardiac repair. In this study we examined the validity of a series of new preformed membrane scaffolds, based on collagen type I, for the transplantation of cardiac cells. One type of membrane, cross-linked with 1,4-butanediol diglycidyl ether (BDDGE) and fibronectin-enriched, gave rise to spontaneously beating heart cell constructs 5–9 days after seeding with neonatal rat cardiac cells. This membrane was then grafted, with and without beating cardiac cells, onto the infarcted area of rat models of heart failure. Seriate echocardiography, performed on rats before transplantation and at 4 and 8 weeks after transplantation, showed that rats that received collagen membranes with beating cells showed an improvement in cardiac function after 8 weeks. These results suggest that this new type of collagen membrane can be used as vector for the transplantation of beating heart cells to the injured myocardium, hence representing an important potential tool for cardiac tissue repair technologies.

Organ transplantation, as an extremely demanding, but impоrtant medical intervention, has always provided significant scope for opposing different legal and ethical principles. That scope is being expanded by considering the possibility that a sensitive category of persons, such as minors, may appear in the role of donor. While transplantation of organs from living minors is prohibited for the above reasons, cadaveric transplantation of organs from deceased children is legal in the law of the Republic of Serbia. This paper analyzes precisely such legally regulated organ donation, which implies the possibility of the appearance of a deceased minor as a donor. While, on the one hand, there is a growing need to take organs from children in order to transplant them and preserve the life of another child of appropriate age, such a decision has a significant emotional impact, above all, on the parents of the child. Keeping in mind that parents appear as the only authorized holders of the right to decide on the transplantation of an organ of a deceased minor, it is necessary to analyze the legal nature of the rights they have on the body and organs of the child. Also, it is necessary to point out the importance of more precise legal regulation of the conditions under which such transplantation can be performed, as well as the necessity of ensuring the autonomy of the will of the minor in terms of organ donation, which he could achieve for life.

Muscle-derived stem cell (MDSC) transplantation has shown potential as a therapy for cardiac and skeletal muscle dysfunction in diseases such as Duchenne muscular dystrophy (DMD). In this study we explore mechanical environment and its effects on MDSCs engraftment into cardiac and skeletal muscle in mdx mice and neoangiogenesis within the engraftment area. We first looked at transplantation of the same number of MDSCs into the heart and gastrocnemius (GN) muscle of dystrophic mice and the resulting dystrophin expression. We then explored neoangiogenesis within the engraftments through quantification of CD31 positive microvessels. This study is important to aid in determining the in vivo environmental factors leading to large graft size which may aid in determining optimum transplantation conditions for muscle repair.

Abstract:
Factor XII clotting activity (FXII), Plasma Pre-kallikrein amidolytic activity (PK), 2-Macroglobulin ( α2-M) and cT-Inhibitor (cT-Inh) antigen have been measured in 17 patients immediately before and sequentially up to four months after kidney transplantation. Based on suspected Cyclosporin A (CyA) induced endothelial damage, activation of the contact system with resulting consumption of the contact activation factors was^evaluated. Before transplantation, FXII, PK, α2-M, Cl-Inh levels were 99±27%, 102±21%, 115±55%, and 129±32%, respectively. In the first two weeks after transplantation FXII decreased to 65±27%, PK to 67±20% and α2-M to 88±42%; Cl-Inh rose to a maximum of 201±44% (mean ± S. D.)(2p<0.001). Mean FXII levels correlated positively with PK, α2-M and albumin and negatively with CyA level and dose and serum bilirubin. PK and α2-M correlated positively with each other and with albumin and negatively with creatinine, bilirubin and CyA (p<0.01). The changes of FXII, PK and α2-M after transplantation suggest an influence of CyA on production or consumption of these factors. The behaviour of the cT-Inh may be unspecific and related to its action of an acute phase reactant.
