Full textLa piscirickettsiosis es una enfermedad causada por un agente bacteriano denominado Piscirickettsia salmonis que afecta a los salmónidos cultivados en el sur de Chile principalmente en etapas de cultivo en el mar, cuyo diagnóstico se basa comúnmente en el análisis de laboratorio de muestras de riñón, hígado y cerebro según programas de vigilancia de la autoridad sanitaria acuícola de Chile. La presente memoria de título tuvo por objetivo determinar, mediante un índice de concordancia (Kappa), la asociación estadística entre la presencia de P. salmonis en muestras de heces y la presencia del agente en muestras de riñón e hígado provenientes de 24 truchas arcoíris (Onchorynchus mykiss) adultas con signología clínica de la enfermedad y que se encuentran mantenidas en condiciones de cultivo en el mar, utilizando una técnica molecular para la detección de la bacteria en cada una de las muestras. Se determinó la presencia de P. salmonis en un 70,8%, 83,3% y 75% de las muestras de heces, riñón e hígado respectivamente, y el índice kappa calculado para evaluar la asociación entre la presencia de la bacteria en heces y riñón fue negativo indicando nula concordancia mientras que el valor de kappa para la asociación entre la presencia del agente en heces e hígado fue de 0,276, indicando una débil concordancia. Este estudio es el primero en detectar P. salmonis en muestras de heces de truchas arcoíris mantenidas en condiciones naturales de cultivo en mar pero se requieren mayores estudios para poder determinar la viabilidad de la bacteria en el contenido fecal y así establecer su mecanismo de diseminación

Full text--- This study aimed to standardize and validate the real-time RT-PCR technique for diagnosis of Infectious Pancreatic Necrosis Virus (IPNV) in rainbow trout (Oncorhynchus mykiss) in Peru. Kidney and spleen samples of rainbow trout (n=121) from two fish farms of Junin were taken, 61 animals showed signs of disease and 60 were apparently healthy animals. All samples were assessed by indirect immunofluorescence technique (IIF) to determine presence of virus. Real-time RT-PCR was carried out using a two-step commercial kit. We used WB1 and WB2 primers for amplification of a specific genomic segment of the VP2 structural protein. IPNV Sp strain and 10 samples inoculated with IPNV were used as positive controls; whereas related viruses, such as rotavirus and infectious bursal disease virus from chickens, as well as unrelated viruses were all used as negative controls. Also, we inoculated decreasing dilutions of virus into tissue samples to determine the sensitivity degree of the test. Results were assesssed according to the values of the threshold cycle (Ct) and melting temperature (Tm) of the amplified products. The test was able to detect the virus in concentrations up to 102 PFU/ml. All 121 field samples were negative for IPNV; sick animals were apparently infected by bacterial agents. Real Time RT-PCR’s specificity and sensitivity were around 100%; moreover, the positive predictive value and negative predictive value was also set in 100%. Real time RT- PCR technique standardized in our laboratory is a valuable diagnostic tool for IPNV detection in field samples. Full text--- The purpose of this study was to determine frequency the presentation of anothomohistopathologycal lesions compatibles with metabolic disorders related to energy and proteins in the diet of juvenile Rainbow Trout (Oncorhynchus mykiss ). Forty fish were collected randomly from the one trout hatchery, located in Huancayo-Junín, mainly taking samples of liver, kidney, gill, stomach and spleen. Between the macroscopic findings compatible with metabolic alterations observed with major frequency it was the fatty liver characterized by a brown pale coloration in 67.5% (27/40). As soon as, to the lesions histopatológicas compatible the metabolic alterations were found fat degeneration 97.5% (39/40), predominantly in grade IV and hyaline degeneration in renal tubules, 62.5% (25/40) and only 7.5% in glands gastric stomach. In gills, changes in growth such as hyperplasia (30/40), fusion (29/40) and lamellar atrophy (29/40) by 75% and 72.5% respectively, representing the highest frequency of lesions. Based on the findings, concluded that the lesions compatible the metabolic disorders were observed in a high frequency, principally related to excessive protein as hyaline degeneration in the kidney and stomach, and fatty degeneration in liver for balancing energy. Finally, the lesions on gills suggest an association between metabolic disorders and their polluting effect on the aquatic environment.

Full text--- The aim of this study was to determine the increase in rainfall on the prevalence of Renibacterium salmoninarum detected from tissue samples (kidney and spleen) of rainbow trout in a fish farm located in the Mantaro Valley basin. The study was conducted at a fish farm in the department of Junín, in the province of Huancayo, in the months of August, September, November and February, during the rain cycle 2011-2012. The water source for the fish farm is Chiapuquio river belongs to the basin of the Mantaro Valley. The trout were randomly collected and eggs were from National and American. 60 animals were collected per sample, 240 total trout fish farm, considering a prevalence of 5%. Fish were collected in relation to the cycle of rainfall in the Mantaro Valley. August (dry season), September, November (rainy season start), and February (rainy season). DNA was extracted from kidney and spleen by phenol-chloroform technique and performed the real time PCR, yielding no Renibacterium salmoninarum genome in any of the samples, none of the rainy season cycle. We conclude that the prevalence of Renibacterium salmoninarum in fish farm study is 0%, so could not be established association between the rain cycle and prevalence of Renibacterium salmoninarum. Full textThe aquaculture is a field major priority than other for the development in our country; however, there exist many aspects in which it has not been investigated and others in which is beginning, as is the case of the use of immunoestimulants for to achieve better results in the production of the many species from economic importance. One of the stimulants investigated in other species of fish is the chitin that administered as dietary supplement enhances the immune response, providing them of the assault of pathogenic agents as Flavobacterium psychrophilum. The aim of the investigation was to evaluate immunostimulant activity of chitin administered by oral route to rainbow trout’s youths (Oncorhynchus mykiss) in immunocompetent and immunosupressed conditions e infected with a test strain, Flavobacterium psychrophilum 1947T, causing of the “cold water disease”. To demonstrate the immunostimulating with chitin there were evaluated parameters of the cellular (phagocytary activity and production of nitric oxide) and humoral (complement’s activity by alternative route and lysozyme serum’s) innate immunity. In addition it decided the necessary time of treatment with the immunostimulant to achieve the suitable one immunostimulation. Trouts were fed by pienso with and without chitin (n=20 in each case) for 2 and 4 weeks. The chitin was added to the dose of 100g/Kg of food and was supplied a proportion of 1% of biomass. Immediately of treatment both groups were immunosupressed with ciclophosphamide and infected by intramuscular route. I concluded that there exists a significant improvement of the production of nitric oxide and lysozyme serum’s activity in the immunosupressed fishes and treated with chitin in comparison with the immunocompetents fishes and not treated. The complement by alternative route and in vitro phagocytary activity did not show significant variations for both groups from two weeks of treatment.

Full textLa salmonicultura es una actividad que hoy reporta al país importantes beneficios tanto económicos como sociales; convirtiendo a Chile en el segundo productor mundial de salmones. De ahí la importancia de mantener la sanidad de los peces en sus distintos estados de desarrollo y así reducir al mínimo las pérdidas por enfermedades. La piscirickettsiosis cuyo agente causal corresponde a Piscirickettsia salmonis es una de las principales enfermedades que afectan la salmonicultura nacional. La presente memoria de título tuvo por objetivo caracterizar la interacción de P. salmonis con ovas de trucha arco iris (Oncorhynchus mykiss) sometidas a tratamientos con proteasas y cuya técnica de detección fue “dot blot”. La metodología “dot blot” no permitió comparar la eventual diferencia en la adhesión y/o penetración de la bacteria a la ova entre tripsina y proteinasa K. Por lo cual, esta técnica sólo se pudo utilizar como una técnica cualitativa para la detección de P. salmonis. Mediante la técnica de “dot blot” se estableció que las ovas de trucha arco iris son sensibles a la infección con la cepa SLGO-95 de P. salmonis. Esta memoria también permite establecer que la estructura proteica del corion es afectada por las enzimas proteolíticas tripsina y proteinasa K. Además, dados los resultados obtenidos post-tratamientos enzimáticos, se considera que la unión entre ovas y P. salmonis, disminuye si se degradan estructuras proteicas, por lo tanto en esta unión implicaría partes proteicas o polipéptidos. Finalmente estos resultados sugieren, pero no en forma concluyente en considerar la acción de proteínas o polipéptidos en la unión y posterior internalización de P. salmonis en el interior de la ova

Full text--- This study aimed to determinate the presence of Piscirickettsia salmonis agent in culture rainbow trout (Oncorhynchus mykiss) in the Mantaro river basin in the region of Junin by Indirect Inmunofluorescence test. Kidney and spleen samples at random of 180 culture rainbow trout from three fish farms of the basin (60 animals in each) were taken. The most outstanding clinical signs were sores on the skin, melanosis, irregular swimming in the water surface, swelling of the abdomen and exophthalmoses. The most outstanding lesions were petechial hemorrhage in liver, pyloric caeca and spleen, in addition to splenomegaly and pale liver. 100% of rainbow trout samples were negative for Piscirickettsia salmonis. The result indicates that this agent is not present in fish farms evaluated or the prevalence was very low and could not be detected. Full textManifiesta que el objetivo es detectar la presencia de Renibacterium salmoninarum (R. salmoninarum) en ovas fertilizadas importadas para el cultivo de trucha arco iris (Oncorhynchus mykiss) y la evaluación de la técnica de PCR a Tiempo Real como técnica diagnóstica. Se colectaron 60 ovas embrionadas de trucha arcoíris de Origen Norteamericano y se dividieron en 12 pooles de 5 ovas cada uno. Las muestras se procesaron con el método de extracción de fenolcloroformo seguido de la técnica de PCR a tiempo real. Los resultados obtenidos mostraron que, 2 pooles dieron positivo a R. salmoninarum de los 12 evaluados. En ambas muestras positivas se generó un producto específico con una Temperatura de Disociación (Tm) de 84.2°C. Estos resultados indican que existen ovas portadoras de R. salmoninarum y que la prueba PCR tiempo real puede ser usada para detectar la presencia del patógeno en muestras de ovas embrionadas.
