Abstract:
The intention of this research was to inspect the amount of social work students addiction in using Facebook, the amount of their study habits such as management of time, competency in attending test, note taking, reading, writing, field work practice, report writing, presentation of class seminar, participation in group discussion in the class and if correlation exists between the two. Researchers used the Bergen Facebook Addiction Scale BFAS , it contains the six important elements of addiction such as salience, mood modification, tolerance, withdrawal, conflict, and relapse and the Study Habits Questionnaire SHQ were used to 123 Social work students from different post graduate colleges affiliated under Tumkur University. SPSS and Excel used to compute data, researchers adopted correlation and simple percentile statistical test to examine impact of facebook addiction. The Social work students studying in different colleges in Tumkur University have largely high degree of addiction in Facebook. On the other hand they are found to exhibit low extent of practicing study related activities. Correlation analysis revealed significant yet positive coefficient values of the six Facebook addiction elements with overall study habits. Findings can be used Social work educators by integrating it in classroom activities. At the same time this study is path way for further studies with bigger scope and inclusion of demographic variables in estimating Facebook addiction, social network or online addiction are recommended. Abstract:
Despite the availability of the HPV vaccine in our country, the mortality secondary to cervical cancer is still high as not many are interested in vaccinating the at-risk population. Hence, a study was conducted to identify the knowledge, attitude, and practice of the Human papilloma virus (HPV) vaccination in pre-university female students.A cross-sectional study was conducted in the Tumkur district of India amongst female students studying in pre-university colleges. A total of 271 female students were included in the study. The mean knowledge score of the study population was 5.6/17 and only 169 (62%) students were aware of the effects of HPV. A total of 144 (53%) study participants were willing to take HPV vaccination if provided but 1/3 of them backed out after knowing the cost of vaccination. The study participants from the urban area had more knowledge about the effects of HPV (118/160 vs. 51/111; p <0.0001) than those from the rural area but there was no difference between them regarding attitude and practice toward HPV infection and its vaccination. The study population was found to have poor knowledge about HPV infection and HPV vaccination. Only 53% of study participants were willing to take HPV vaccination if provided but 1/3 of them backed out after knowing the cost of vaccination. The urban group was more knowledgeable than the rural group but both groups had a similar attitude and practice toward HPV infection and its vaccination.

Arnold-Day, Christel. "Paediatric brain tumours: The University of Cape Town experience from 1996 - 2017." Master's thesis, Faculty of Health Sciences, 2019. https://hdl.handle.net/11427/32084.

Full text
Abstract:
Brain tumours are the second most common malignancy in children(1) (2), and despite some advancements being made over the last 2 decades, patient outcomes in general remain poor when compared with other childhood cancers. Optimal treatment of children with brain tumours is challenging and expertise and resources are not widely available in South Africa. This is important because the outcomes of children with brain tumours depend critically on the expertise and resources of a multidisciplinary team tasked with their treatment. Despite the importance of paediatric brain tumours though, little is known about childhood brain tumours in South Africa as limited data have been published and there have been no funded studies to support research in this area. In addition, we know very little about the resources available across the country to treat these children. In international centres of excellence the best outcomes are achieved by combining good epidemiological data, strong multidisciplinary teams, centralization or regionalization of services, available resources, and a research foundation. To start, we need to know more about the patients presenting to us with brain tumours. PURPOSE The overall aim of this project was to collect epidemiological data for childhood brain tumours at a tertiary paediatric hospital in South Africa with a dedicated multidisciplinary team. METHODS Study design: A retrospective review of records of patients diagnosed with a primary brain tumour and who presented to Red Cross Children’s Hospital (RCCH) system from 1 January 1996 to 31 December 2017. 2 Patient selection & data collection: Patients were identified by combining databases and admission logs from paediatric neurosurgery, oncology, radiotherapy, histopathology and radiology. Data collected included: age at diagnosis, sex, province of referral, tumour site and diagnosis. RESULTS A total of 554 paediatric patients with primary brain tumours were identified over the study period. Tumours were more common among males (55.4%) and were located in the supratentorial compartment in 52%. The median age at diagnosis was 5.92 years. The commonest tumours were astrocytomas (n=114 patients; 20.3%), followed by medulloblastomas (incl. PNETs) (n=107 patients; 19.1%), and craniopharyngiomas (n=55; 9.8%). As expected, most patients referred and seen at RCCH/GSH were from the expected drainage area in the Western Cape (73%), but a significant number of referrals (27%) were from outside the province referrals, especially in the last 10 years. CONCLUSION Our findings were largely consistent with the published literature in terms of histological diagnosis, sex profile and age ranges for children diagnosed with brain tumours with some small differences possibly related to referral bias. More patients than expected were referred from outside of the province, which emphasizes the need for establishing an ongoing tumour database registry and co-ordinating patient care across institutions. A follow-up study to assess patient management and outcomes is of critical importance to assess resource availability and patient outcomes.

Abstract:
Background: Hibernomas are rare benign soft-tissue tumours arising from brown fat tissue. Although imaging characteristics are not specific certain imaging features, common locations and patient demographics may suggest hibernoma as a differential diagnosis. Case presentation: We report on two 48-year-old male patients with hibernoma. The tumour presented with local swelling of the inguinal region in the first patient and was an incidental imaging finding in the second patient. Imaging included magnetic resonance imaging in both patients and computed tomography as well as 18 F-fluorodeoxyglucose positron emission tomography-computed tomography in the second patient. In both cases histological diagnosis was initially based on excisional and needle core biopsy, respectively. Complete surgical resection confirmed the diagnosis of hibernoma thereafter. Conclusion: In soft tissue tumours with fatty components hibernoma may be included into the differential diagnosis. Because of the risk of sampling errors in hibernoma-like tissue components of myxoid and well-differentiated liposarcoma, complete resection is mandatory. This article also reviews the current imaging literature of hibernomas.

Abstract:
AbstractThe WHO Classification of Tumours (WCT) is a series of authoritative and concise reference books for the histological and molecular classification of tumors that underpins treatment and care of cancer patients, as well as research into cancer epidemiology, prevention, diagnosis, and treatment, and is essential for cancer diagnosis worldwide. This classification relies traditionally on consensus of pathological expert opinions as basis for cancer classification, but the understanding of cancer at a molecular level advances in prognosis, and other related fields have moved the WCT to find ways of translating diagnostic research into evidence synthesis that can effectively inform decisions relevant to the classification. Systematic reviews represent the top of the hierarchy of scientific evidence and allow to summarize evidence from many publications to inform decisions. This evidence-based approach is the cornerstone of evidence-based medicine and well established in many medical specialties. However, uptake of these principles within pathology has been slow, and such high-quality reviews of available evidence are not easily available for authors contributing to the WCT. In an effort to overcome reluctance in the field to adopt these methods, a collaborative project between the Advanced Oncology program of the University of Ulm in Germany, the Cochrane Netherlands, the Universidad de Campinas in Brazil, and the WCT has been started. This project called Evi-Pat (Evidence-Based Pathology Training Initiative) aims to develop and evaluate an online training for oncologists and pathologists to train them in the application of evidence-based practice to pathology and related specialties, thereby addressing and overcoming challenges in this, and to pathology, novel approach. Scientific and didactic evaluation of such an effective training tool for online-educated pathologists and oncologists will improve the evidence base in pathology and hence provide a solid foundation for diagnostic and prognostic decision-making. We believe that an evidence-based approach to informing key decisions that feed into tumor classification will allow the WCT editorial board to mitigate the potential inclusion of biased decisions into the classification and also benefit authors by providing structured, transparent, and reliable methods for the synthesis of available evidence for each tumor type, as the same time training to apply these methodologies.

Abstract:
Gastrin is a gastrointestinal hormone, produced predominantly by the G cells of the gastric antrum and duodenum, although small amounts of gastrin have been isolated in the pituitary and some vagal nerve fibres. The biologically active forms of gastrin include carboxy-amidated gastrin-17 and carboxy-amidated gastrin-34, which bind mainly to the cholecystokinin (CCK)-2 receptor. The main role of amidated gastrin is the stimulation of gastric acid secretion by regulation of histamine release from the gastric enterochromaffin-like (ECL) cells, while it may also have a trophic effect on gastric mucosa. There is evidence that the precursor forms of gastrin, such as progastrin and glycine-extended gastrin, are also of biological importance, binding to a separate CCK-C receptor. These precursor may induce cellular and tumour growth and they are implicated in several cancers, such as colon and pancreatic adenocarcinomas. Gastrinomas represent a group of functional pancreatic neuroendocrine tumours, characterized by autonomous release of gastrin by the tumour cells, which results in symptoms not only due to the tumour growth per se, but also to gastric acid hypersecretion. In 1955, at the annual meeting of American Surgical Association, Robert M. Zollinger and Edwin H. Ellison presented a study entitled Primary Peptic Ulcerations of the Jejunum Associated with Islet Cell Tumour of the Pancreas. They proposed a new clinical syndrome of: (1) ulceration in unusual locations in the upper gastrointestinal tract or recurrent ulcerations; (2) gastric acid hyperseretion; and (3) non-β‎ islet cell tumours of the pancreas. However, the potent gastric secretagogue for the Zollinger–Ellison syndrome was not identified until 1960, when Rodney Gregory and Hilda Tracy of the University of Liverpool discovered that the extract from the pancreas of a patient with Zollinger–Ellison syndrome was the hormone gastrin. Thus, these pancreatic tumours were termed ‘gastrinomas’.

Abstract:
Brain tumour segmentation is a hard and time-consuming task to be conducted in the process of radiotherapy planning. Deep Learning (DL) applications have a significant improvement in image segmentation tasks. In this work, we apply DL models such as 2D and 2.5D U-NET to the segmentation task of a brain tumour on the BraTS 2021 dataset and our local dataset. The 2.5D network is a modified version of 2D U-NET by using three slices as an input for each magnetic resonance imaging (MRI) sequence. We achieve the best segmentation results with 2.5D U-NET on BraTS with Dice scores of 86.97%, 91.27% and 94.42% for enhancing tumour, tumour core and whole tumour respectively. On the other hand, our best segmentation result of the GTV delineation on the local dataset is a Dice score of 78.51% for 2D U-NET. Although the result of GTV contours is not improved by 2.5D for the local dataset due to non-fixed voxel size, the Dice scores of ET, TC and WT are improved by the proposed 2.5D U-NET for the BraTS dataset.

"Impact of Heparan Sulphate Binding Domain of Chemokine CCL21 to Migration of Breast Cancer Cells." In Qatar University Annual Research Forum & Exhibition. Qatar University Press, 2020. http://dx.doi.org/10.29117/quarfe.2020.0132.

Abstract:
Lymph node metastasis constitutes a key event in breast cancer progression. Chemokines are small proteins, which can promote metastatic spread by inducing cancer cell migration and invasion. Chemokine function is dependant upon their binding to both cell surface heparan sulphate (HS) molecules and to their specific receptor. Our group has demonstrated a significant increase in chemokine receptor CCR7 expression in cancerous breast epithelia compared to healthy controls. This study is designed to test the hypothesis that a non-HS binding forms of chemokine CCL21 can disrupt the normal response to CCL21, therefore reducing the metastasis of CCR7-expressing cancer cells. Truncated CCL21 chemokine (Δ98- 134 c-terminal basic extension), was synthesised to investigate a possible linkage between chemokine binding capacity and cell activation. Wild type (WT) and mutant-CCL21 were tested for their ability to stimulate a dose-dependent increase in intracellular-free calcium in peripheral blood mononuclear cell (PBMC) and breast cancer epithelial cells MDA-MB-231. Mutant-CCL21 at concentrations 5 and 10nM showed potential to mobilise Ca2+ at levels similar to that produced by WT-CCl21. A series of experiments was performed to determine how deletion of the HS-binding site altered the ability of CCL21 to stimulate chemotaxis within a concentration gradient generated by free solute diffusion. PBMC stimulated to migrate by wild-type CCL21 was not significantly different from that stimulated by mutant (P> 0.05). Similar results were observed in assays using MDA-MB-231 cells. A further series of experiments was performed to compare the potential of WT and mutant-CCL21 to stimulate the migration of cells across endothelium. In contrast to results for trans-filter migration, it was found that the non HSbinding mutant stimulated no increased in transendothelial cell migration above the background at each of the tested concentrations, 10, 30 and 50 nM respectively (P>0.05). However, WT-CCL21 stimulated significant increased PBMC migration at each of the tested concentration (all P <0.001). Furthermore, the effect of heparin on chemotactic properties of WT and mutant- CCL21 was examined. Interestingly, heparin (250 µg/ml) completely inhibit the chemotaxis mediated by WT-CCL21 (5nM) (P < 0.001), whereas it did not inhibit the chemotaxis at concentrations 100, 250 & 500 µg/ml in response to mutant CCL21 (5nM) (P > 0.05). Similar assay will be performed using MDA-MB-231 cells. Work is ongoing to characterise the biophysical properties of mutant-CCL21 and determine its potential role for a therapeutic blockade of the migration of breast cancer cells in-vivo. Our primarily data showed that mutant CCL21 in xenograft brain tumor models showed substantial inhibition of tumour growth. Our results indicate that truncated CCL21 chemokine might be a potential preventive biofactor for human breast cancer metastasis by targeting chemokine receptor genes.

Abstract:
Clear cell renal cell carcinoma (ccRCC) is the most common subtype of renal cell carcinoma (RCC). If diagnosed in later stages, ccRCC is associated with high renal cancer related morbidity and poor prognosis. Recently, microRNAs (miRNAs) have attracted interest as potential diagnostic and prognostic biomarkers due to their important role in cancer development and progression. Availability of big omics data in the cancer genome atlas (TCGA) coupled with data mining and machine learning have revolutionized the identification of robust diagnostic and prognostic signatures in different types of cancers. In this study, we have utilized the miRNA sequencing data of 516 ccRCC patients from TCGA to identify a diagnostic and prognostic signature by using a combined approach of differential expression analysis, survival analysis and machine learning. Differential expression analysis identified 30 downregulated and 20 upregulated miRNAs in the primary tumor as compared to solid tissue normal samples. Out of these 50 differentially expressed miRNAs, higher expression of 7 and lower expression of 6 miRNAs were found to be significantly associated with poor survival when analyzed using the Kaplan-Maier survival method. Pathway enrichment analyses related to the differentially expressed miRNAs revealed that fatty acid biosynthesis was the most significantly enriched KEGG pathway while proteoglycans in cancer pathway was enriched by the highest number of survival-associated miRNAs target genes. Differential expression and association with poor survival was used as a prefilter for training a support vector machine model capable of classifying tumor samples from solid tissue normal samples with an accuracy and precision of 99.23% and 98.50%, respectively. We have identified here a nine-miRNA signature in ccRCC patients that is capable of segregating tumor from normal tissue samples with high accuracy and precision. The future validation of this classification model in in a clinical cohort will support translation of these findings into clinical practice for early detection and follow-up of ccRCC.

Abstract:
Sanguinarine (SNG), a natural compound with an array of pharmacological activities, has promising therapeutic potential against a number of pathological conditions, including malignancies. This research aimed to investigate the antiproliferative and anti-cancer potential of SNG against two well characterized papillary thyroid cancer (PTC) cell lines, BCPAP and TPC-1 .In both cell lines , SNG was able to inhibit cell proliferation in time and dose dependent manner. Western blot analysis revealed increased expression of apoptosis and autophagy markers , caspase-3,cleaved caspase-3 , P62, and LC3. SNG modulate its anticancer effect through ROS production, because NAC was able to reverse SNG effect. Interestingly, co-treatment of PTC with SNG and cisplatin amplified anticancer activity. Finally, SNG treatment of PTC spheroid suppressed its growth with downregulation of stemness markers including ALDH2 and SOX2 markers. In conclusion, SNG enhanced the anti cancer activity against PTC cells and the effect is amplified when cisplatin is added.
