Abstract:
Pruritus vulvae, a common complaint, affect approximately 10% of patients attending the gynecologic clinic. The most common cause of this condition is infections of the vulva caused by Candida albicans or Trichomonas vaginalis. Diabetes is among the potent risk factors that causes Candidiasis. Present report provides details of management of this condition with Ayurveda in a 40-year-old woman with severe itching and burning in the vulvar region for a month while concurrently managing her diabetes with allopathic medication. Clinical examination revealed significant inflammation and discoloration of the vulva and vaginitis. As per Ayurveda the case can be diagnosed as Acharna Yonivyapada as the main symptom given is itching vulvae. The treatment included internal medicines like Ashwagandhadi Choorna, Mamejak Ghanavati and Avipattikar Choorna along with local therapies comprised of Avagahana using Panchavalkala Kwatha and Lepana with Gandhak Malam, chosen for their properties to alleviate itching, combat infections, and restore healing and balance of Dosha. The patient was treated with Sthanik chikitsa for a period of 45 days, except menstrual days. Over the period of two months, the patient was recovered with complete cessation of pruritus vulvae and normalization of skin color in the affected areas. These encouraging results highlight the potential efficacy of Ayurvedic interventions in addressing pruritus vulvae, especially in patients with diabetes who are prone to such conditions due to factors like elevated sugar levels in urine.

Abstract The Caenorhabditis elegans locus lin-15 negatively regulates an intercellular signaling process that induces formation of the hermaphrodite vulva. The lin-15 locus controls two separate genetic activities. Mutants that lack both activities have multiple, ectopic pseudo-vulvae resulting from the overproduction of vulval cells, whereas mutants defective in only one lin-15 activity appear wild-type. lin-15 acts non-cell-autonomously to prevent the activation of a receptor tyrosine kinase/ras signaling pathway. We report here the molecular characterization of the lin-15 locus. The two lin-15 activities are encoded by contiguous genomic regions and by two distinct, non-overlapping transcripts that may be processed from a single mRNA precursor by trans-splicing. Based on the DNA sequence, the 719- and 1,440-amino acid lin-15 proteins are not similar to each other or to known proteins. lin-15 multivulva mutants, which are defective in both lin-15 activities, contain deletions and insertions that affect the lin-15 genomic region.

Abstract:
Functional morphology of the male gonopods, female holding systems and vulvae of Libinia spinosa are described using scanning electron microscopy and histological sections, establishing differences between immature and mature individuals. The first gonopod of mature males, larger than 35.1 mm of carapace width (CW), is long, slender, and ending in two wing-like processes; abdominal, characterized by the presence of a sub-terminal seminal canal, and sternal, closed and where the cuticle holds setae. In immature males, smaller than 29.9 mm CW, both processes are shorter, and the tip of the abdominal process closed. Six different types of setae are described for both gonopods. Internally, the gonopods have rosette glands in immature and mature individuals. The presence of secretions, which may be involved in the moulting process, was observed in immature males towards the external cuticle corresponding to the rosette glands. The rosette glands may be related to reproduction in mature males. Females presented a press-button holding system and a biconcave vulva. In immature females, smaller than 36.1 mm CW, the vulva aperture was closed by integument bulging to the outside. In mature females, larger than 42.6 mm CW, the locking system was almost fused to the cuticle, and the vulvae showed an open biconcave form with a more inner flexible membrane. Morphological characteristics shown by the gonopods of mature males develop synchronously with the acquisition of physiological maturity, not with morphometrical maturity. The mature holding systems and vulvae of females are developed synchronously with physiological and morphometrical maturity.

Males and swollen mature females of Achlysiella trilineata and Achlysiella magniglans were discovered for the first time in Australia. These life stages confirmed that both species show well-marked sexual dimorphism in the anterior region of the body, with the males having reduced cephalic frameworks, stylets, and oesophagi. Males of A. magniglans displayed an extreme reduction in the style such that only the cone could be seen, and the rest of the esophagus was barely visible. In both species, the swollen females had postmedian vulvae, and the anterior branch of the ovary was longer than the posterior branch. In A. magniglans, the disparity in length was more pronounced, and the vulva was more posterior. The finding of swollen females confirmed the classification of both species in Achlysiella rather than Radopholus. Vermiform females of both species have large dorsal oesophageal glands and short ovaries. Justifiably, other species of Radopholus with these latter features (R. brevicaudatus, R. capitatus, and R. vacuus) should also be moved to Achlysiella.

The systematics of two problematic potamid species, Sinopotamon koatenense (Rathbun, 1904) and Sinopotamon wuyiensis Li, Lin, Cheng &amp; Tang, 1985, both originally described from the Wuyi Mountains are resolved in this study. Sinopotamon koatenense is transferred to the genus Huananpotamon Dai &amp; Ng, 1994, as the new combination Huananpotamon koatenensecomb. nov. The new combination differs from its congeners in the form of the carapace, male pleon, male first gonopod, and vulvae. Phylogenetic analyses based on mitochondrial 16S rDNA sequences support the identification of Huananpotamon koatenensecomb. nov. and a redescription is also provided. In addition, S. wuyiensis is confirmed as a junior synonym of Sinopotamon fukienense Dai &amp; Chen, 1979 based on morphological similarities and phylogenetic lineages.

Abstract:
The enormous burning of fossil fuel and deforestation have caused an increase in the atmospheric CO₂ concentration ([CO₂]) during the last century. This will invariably have profound direct and indirect effects on plant carbon metabolism. The majority of research on the effects of CO₂ enrichment on plants are short-term and are done on other crops, but very little have been done on barley. This project aimed to determine the effects of long-term CO₂ enrichment on photosynthesis, growth and grain yield on barley. Hordeum vulgare L. cvs Stirling and Schooner plants were grown from seeds in controlled environment chambers at ambient (350) and elevated (600) μmol molˉ¹ [CO₂]. Measurements of net assimilation rate (NAR), photosynthetic pigments content and growth parameters were started 7 days after germination (DAG) and continued until senescence. The anatomy of matured fully developed leaves was also monitored. Elevated [CO₂l resulted in an increase in NAR in the two cultivars from days 7 until 14, after which the stimulation of NAR of CO₂-enriched plants started to decrease. At the onset of senescence, NAR was almost equal in plants grown under both ambient and elevated [CO₂]. The response of assimilation as a function of internal [CO₂l (C₁) at the end of the experimental period showed a significant decrease in both the initial slope of the A/C₁ curves and the CO₂-saturated photosynthetic rates in the two cultivars. Stirling showed no significant changes in the content of chlorophyll α,chlorophyll б or in total carotenoids. However, Schooner showed a stimulation in chlorophyll α content at day 7, but decreased at day 28. Chlorophyll б and total carotenoids content were not affected by CO₂ enrichment. While total above-ground biomass was not affected by elevated [CO₂] in the two cultivars, total plant height decreased significantly after 14 days in Stirling whereas no significant change occurred in Schooner throughout the experimental period. Leaf area was not significantly affected by CO₂ enrichment in the two cultivars although the leaves in CO₂ enriched plants were slightly shorter. Anatomical studies reveal that leaf thickness was significantly increased by CO₂ enrichment in Stirling, but the increase was not significant in Schooner. Both cultivars did not show any significant effect on chloroplast morphology and ultrastructure as a consequence of elevated CO₂ exposure. No signs of starch accumulation were evident in variety Schooner, but Stirling showed some form of starch accumulation, under increased atmospheric [CO₂]. Elevated CO₂ resulted in a significant reduction by more than 50 % in the number of grain yield per plant in both Stirling and Schooner. Results from this study therefore indicate that CO₂ enrichment will not be beneficial in terms of growth and yield in this important crop.

To prove the post-natal depression model, the antidepressant sertraline, was assessed in rat mothers (n=14) divided into Prenatally Stressed (PS) and Non-Stressed (NS) groups. The data failed to support the hypothesis that ‘the progeny of 10mg of sertraline-treated PS mothers displayed less anxiety than the progeny of vehicle-treated PS mothers’. The forced swim test (FST) was used to examine depressive-like behaviour in mice. Barley successfully increased mobility in mice exposed to the FST. Barley was antidepressant at low doses (0.8g/kg and upwards) if used subchronic; and at high doses(6.4g/kg and 12.8g/kg) if used acutely;(n=113,56acute,57 subchronic- treated). Barley (6.4g/kg) was also able to alleviate the depressive-behaviour in mice induced by the Reserpine Test (n=114, 58 reserpinised, 56 non-reserpinised) and Social ‘Defeat’ Test (n=24, 8 vehicle undefeated, 8 barley defeated, 8 vehicle defeated mice). To confirm that the anti-depressant effects of barley(6.4g/kg) were not simply due to increased locomotor activity in the FST, an Open Field Test(OFT) was undertaken (n=14,7 vehicle, 7 barley). Barley had no effect on locomotor activity and also caused no significant changes in weight (n=16, 8vehicle, 8 barley). In mice,Barley(6.4g/kg) significantly delayed the tremorogenic effects of Physostigmine (n=18, 6 control,6 Physostigmine, 6 Physostigmine with barley); reduced bradykinesia induced by reserpine (n=18,6 control, 6 vehicle, 6 barley treated);and was analgesic in nociception tests (n =20, 5 control, 5 barley, 5 pain, 5 pain with barley). Abstract:
The gas exchange responses of two cultivars of Hordeum vulgare L., to light, temperature, CO₂ and water stress were investigated in the laboratory. The optimum temperature for net CO₂ assimilation was found to be 25°C and 22.5°C for cv. Clipper and cv. Dayan respectively. Net CO₂ assimilation was reduced at 30°C in cv. Dayan. At low light intensity the highest quantum yield efficiency was 0.051 mol.mol⁻¹ at 30°C for cv. Clipper, and 0.066 mol.mol⁻¹ at 20°C for cv. Dayan. At the same temperature, cv. Clipper had a higher water use efficiency than cv. Dayan, but stomatal conductance for cv. Dayan was higher than cv. Clipper. Stomatal limitation to CO₂ was lowest at the optimum temperature for CO₂ assimilation in both cultivars. Stomata limited CO₂ assimilation in cv. Clipper to a larger degree than in cv. Dayan. Relative stomatal limitation for cv. Clipper at 25°C was 0.280 ± 0.010, and for cv. Dayan at 22.5°C was 0.028 ± 0.011. Short-term exposure to elevated CO₂ concentrations increased CO₂ assimilation in both cultivars, but more so for cv. Clipper. Transpiration rate at elevated CO₂ partial pressures were higher in cv. Dayan than in cv. Clipper. At very high CO₂ (860 μmol.m⁻²s⁻¹) partial pressure water use efficiency in cv. Clipper was higher than cv. Dayan, but at low CO₂ partial pressures water use efficiency in cv. Dayan was higher than cv. Clipper. Water stress reduced the relative leaf water content and net CO₂ assimilation in both cultivars. Cultivar Dayan was more tolerant to water stress, and CO₂ assimilation in this cultivar was less affected by water stress. In both cultivars water stress increased the concentration of chlorophyll a, chlorophyll b, and chlorophyll a+b. The chlorophyll a:b ratio remained relatively constant throughout the stress period. No correlation between relative leaf water content and total carotenoid concentration was observed.

Abstract:
Editing Middle High German texts today is led much more by the ventilation of practical necessities than by theoretical consideration. Especially lacking discussions of a basic concept of ›text‹ raises the risk of developing blind spots. The article will try to identify these blind spots as places where the traditional strategy of editing texts on the basis of the faulty-replication model persists. This will be illustrated by new editions of the ›Wunderer‹ and the ›Rosengarten‹. Furthermore, the article evolves a semiotic model of the philological praxis of editing Middle High German heroic epics.<br>Die Redaktion mittelhochdeutscher Texte wird heute viel mehr von der Lüftung praktischer Notwendigkeiten als von theoretischen Überlegungen geleitet. Gerade die fehlende Diskussion über einen grundlegenden Begriff von 'Text' birgt die Gefahr, dass sich blinde Flecken entwickeln. Der Artikel versucht, diese blinden Flecken als Orte zu identifizieren, an denen die traditionelle Strategie der Textredaktion auf der Grundlage des Fehlerreproduktionsmodells fortbesteht. Dies wird durch Neuauflagen des 'Wunderers' und des 'Rosengartens' illustriert. Darüber hinaus entwickelt der Artikel ein semiotisches Modell der philologischen Praxis der Edition mittelhochdeutscher Heldenepen.

This contribution examines the co-existence in French of two impersonal constructions with a light verb and a weather noun, as in il y a / il fait du vent ‘it’s windy’ (lit. there is / it makes wind). Our starting point stems from Bauer’s (2000) observations on Vulgar Latin postulating that facere ‘do, make’ with weather nouns appears to compensate for the diminishing use of intransitive weather verbs. Based on a diachronic and synchronic corpus study of weather sentences extracted from Frantext, we argue that Bauer’s hypothesis can apply at best to Middle French. For Modern and Contemporary French, our analysis reveals that both constructions are existential with il fait+N specializing in expressing ambient states and the il y a-construction in expressing atmospheric events or substances.

Apologies have been a rewarding target for research in the pragmatics tradition; however, few studies connect apologies to hypocrisy. This chapter is an attempt to do so. I approach the pragmalinguistic aspects of hypocrisy by examining two notorious apologies: Clinton’s 1998 apology for his Monica Lewinsky affair and Trump’s 2016 apology for his vulgar comments about women. I argue that hypocritical apologies can be identified via an analysis of felicity conditions (including the speakers’ motivations and intentions) and move structures. Clinton’s and Trump’s apologies share most features with non-hypocritical apologies, but they also include self-promotional and accusatory moves toward the end of the speech event which are absent from genuine apologies. These extra moves turn their apologies into hypocritical ones.

Vaginal intraepithelial neoplasia is a rare, premalignant condition of the vagina, which is caused by persistent infection with oncogenic strains of the human papillomavirus (HPV). It occurs either concurrently with intraepithelial neoplasia of other parts of the anogenital tract, or can develop after treatment of cervical lesions or pelvic irradiation. It can be difficult to diagnose and treat, due to the proximity of surrounding structures such as the bladder and rectum, and the need to preserve sexual function. Squamous carcinoma of the vulva is the most common vulval malignancy. It may arise from two distinct types of vulval intraepithelial neoplasia (VIN). Vulval extramammary Paget’s disease is a rare intraepithelial adenocarcinoma which accounts for less than 2% of primary vulval tumours. VIN and Paget’s disease are treated in order to relieve symptoms, such as severe pruritus, to exclude invasive disease, and to decrease the risk of developing cancer. Specialist follow-up in multidisciplinary clinics, with access to conservative surgery and reconstruction, as well as psychosexual support, are important in the management of women with vulval premalignant disease. Vulvar and vaginal cancer are rare gynaecological malignancies that occur predominantly in elderly women. Where the cornerstone of vulvar cancer treatment is surgery, radiotherapy is the most common treatment given in vaginal cancer. Vulvar cancer treatment has undergone significant modifications during the last decades, all with the aim to reduce treatment-related morbidity without compromising survival rates. The introduction of the sentinel node procedure has been a major advantage in the treatment of this disease.

Introdução: O líquen escleroso (LE) é uma dermatose inflamatória crônica que tem preferência pela região genital, especialmente a feminina, podendo acometer todas as idades, embora tenha distribuição bimodal, com um pico pré-puberal e outro na pós-menopausa. A verdadeira prevalência é desconhecida. Representa 18% das patologias vulvares em crianças, sendo precedido apenas pela dermatite atópica e irritativa. Pode ser assintomático, porém frequentemente se apresenta com prurido e algumas vezes com queimação, sangramento, disúria e dor. Em crianças, o envolvimento da região perianal pode levar a casos de constipação e dor ao evacuar. As lesões típicas são manchas marfim ou pápulas avermelhadas, em alguns casos com hiperceratose, que podem coalescer em placas geralmente simétricas. Outras lesões podem incluir fissuras, petéquias, púrpuras, erosões, ulcerações, edema, hiperpigmentação. O diagnóstico geralmente é clínico. Está associado, em alguns casos, a doenças autoimunes. O presente relato consiste na descrição de um caso de LE infantil. Relato de caso: Paciente do sexo feminino, nove anos, branca, sem comorbidades nem uso de medicamento regular. Apresentou queixa de prurido vulvar, que levou ao uso de Trok® (cetoconazol 20 mg/g + dipropionato de betametasona 0,50 mg/g) por anos, sem resolução do quadro. Foi realizada investigação para infecções sexualmente transmissíveis, fungos e doenças autoimunes (FAN, TSH, T4 livre, anti-TPO, anti-tireoglobulina, anti-DNA, ultrassonografia com Doppler da tireoide), com todos os resultados negativos. Após a suspeita do diagnóstico de LE, foi utilizado Psorex® creme (propionato de clobetasol 0,5 mg/g) com melhora da sintomatologia. Além disso, foram orientados cuidados de higiene com sabonete neutro, calcinha de algodão e uso de Dersani® loção oleosa após o banho. Conclusão: A fisiopatologia ainda não é bem esclarecida. Existem teorias que envolvem alterações hormonais, por ser o LE mais visto em extremos de idade, questionando-se se ele ocorre pelo hipoestrogenismo. O diagnóstico é eminentemente clínico, evitando-se biópsias, como no caso acima. A chance de o LE evoluir para carcinoma de células escamosas de vulva é de 4%, que é encontrado normalmente após a menopausa. O LE não tem cura mas pode ser bem controlado clinicamente, como observado no caso descrito. O tratamento consiste em aliviar os sintomas e evitar a progressão da doença. Na fase de estabilização da enfermidade, essas pacientes devem ser examinadas duas vezes por ano. O LE é uma doença de difícil diagnóstico, principalmente quando assintomático, mas deve ser lembrado mesmo na infância.

Introduction: Postablative reconstruction of vulvar defects is a difficult challenge. Local flaps carry a high incidence of delayed wound healing as local flaps may redistribute but not eliminate local wound tension. Repair of defect with distant pedicled flaps may avert local complications by minimising tension to the skin and increasing the initial biomechanical strength of wound. The aim of this study was to determine the clinical outcome of distant musculocutaneous &amp; fasciocutaneous flaps used for postablative reconstruction of large vulvar defects. Methods: Between January 2015 to December 2015 total three patients underwent vulvectomy and immediate reconstruction with distant pedicled flaps for vulvar carcinoma. Postoperative complications were recorded and clinical outcomes were evaluated. Results: Two of the three flaps healed primarily. One flap was complicated by minor wound dehiscence, which healed with conservative treatment. Hospital stay and clinical course was shorter in comparison to local flaps and split skin graft. Conclusions: Distant musculocutaneous and fasciocutaneous flaps provide excellent design flexibility and can be tailored to reconstruct postablative vulvar defects with good outcomes and minimal morbidity.

Effective management of regional lymph nodes is the most important factor in the curative management of early vulvar cancer. Despite careful dissection and maintaining vascularity of skin, surgical morbidity is seen in 50% cases. Video – endoscopic inguinofemoral lymphadenectomy was developed by Bishoff in 2003 by dissecting two cadaveric models and in one patient with stage T3N1M0 penile carcinoma. VEIL is an alternative to reduce the morbidity without compromising the oncologic outcomes. VEIL has continued to evolve into single site and robotic variants. R-VEIL is a minimally invasive procedure duplicating the standard open procedure with less morbidity. Aims and Objectives: A video presentation to describe the technique of R-VEIL in vulvar cancer and discuss the advantages and outcome. Conclusions: R–VEIL is an attractive minimally invasive technique to do inguinal block dissection in a single sitting in patients with vulvar carcinoma as the surgeon does not get tired as happens in VEIL technique. R-VEIL allows the removal of inguinal lymph nodes within the same limits as in open procedure and potentially reduces surgical morbidity. It is better accepted cosmetically and reduces hospital stay. Long term oncological results are not available. Randomized multi-institutional studies are required to prove its efficacy over open counterpart.

Background: Patients with renal transplant have a higher incidence of various malignancies. Delivery of adequate radiation dose to the pelvic target in such patients sparing the transplanted kidney is a dosimetric ordeal. Due to lack of sufficient data in the literature regarding the dose constraint of the transplanted pelvic kidney, plan evaluation becomes extremely challenging in this situation. Here we present comparative dosimetric plan evaluation data of treating a patient with carcinoma of the vulva with transplanted kidney. Methods: We compared 3D conformal radiotherapy (3DCRT) and Intensity Modulated Radiotherapy (IMRT) plans for a patient diagnosed to have carcinoma of the vulva with a transplanted kidney. Total dose of radiotherapy (63 Gy) was delivered in two phases (45 Gy in 25 fractions and 18 Gy in 10 fractions respectively). We compared dose to planning target volume (PTV), and dose to organs at risk including the transplanted kidney in these two techniques. The volumes encompassed by different isodoses (50%, 20%, 10%, 5%) were also compared. Weekly renal function test was monitored. Results: The dose received by 95% of the planning target volume in 3DCRT was 43.3 Gy (phase 1), 17.7 Gy (phase 2) and in IMRT was 43.74 Gy (phase 1), 17.3 Gy (phase 2). The mean doses received by kidney in Phase 1 3DCRT, Phase 1 IMRT, phase 2 3DCRT and phase 2 IMRT were 0.98 Gy, 3.05 Gy, 0.74 Gy, 0.13 Gy respectively. The volumes covered by 50%, 20%, 10%, 5% were higher with IMRT plan when compared with 3DCRT plans. The creatinine values remained stable through the treatment. Conclusion: Radiotherapy in renal transplanted patients can be done with high precision radiotherapy techniques with strict dosimetric and image guided set up verification.

Condition being studied: Traditional clinical recommendations for treating diseases of the female lower reproductive tract include topical therapy with drugs, surgery, intravaginal radiation, carbon dioxide (CO2) laser, etc. Although medication is easy to administer, it has a high recurrence rate and adverse effects such as burning sensation, pain, and dyspareunia. The other traditional treatment method is usually invasive, repeated operation of vaginal perforation, scar, easy recurrence, fertility decline, and other shortcomings. At present, the treatment strategy for cervical squamous intraepithelial lesion, vaginal squamous intraepithelial lesion, condyloma acuminatum, and vulvar lichen sclerosis are to protect the normal organ structure and function as much as possible, reduce recurrence, prevent disease progression and carcinogenesis, and preserve female reproductive function.

The goal of this bulletin is to provide timely information and/or updates on issues of intentional and accidental adulteration and mislabeling of oregano herb and essential oil (EO).* It provides information on issues of adulteration and mislabeling of oregano (Origanum vulgare subsp. hirtum, O. onites) herb used as a spice and herbal remedy, in particular with winter savory (Satureja montana, Lamiaceae) herb, sweet marjoram (Origanum majorana, Lamiaceae) herb, Cistus spp. (Cistaceae) leaf, olive (Olea europaea, Oleaceae) leaf, thyme (Thymus spp., Lamiaceae) herb, summer savory (Satureja hortensis, Lamiaceae) herb, strawberry (Fragaria spp., Rosaceae) leaf, sumac (Rhus spp., Anacardiaceae) leaf, hazelnut (Corylus avellana, Betulaceae) leaf, myrtle (Myrtus communis, Myrtaceae) leaf, and colored wheat (Triticum aestivum, Poaceae) bran. Additional species in the family Lamiaceae from the genera Origanum, Thymus, Poliomintha, Plectranthus, Monarda, and Hyptis or in the family Verbenaceae from the genera Lippia, Aloysia, and Lantana are known to have “oregano” as part of their common name, and may thus be mistaken for oregano by purchasing personnel who lack proper training. The bulletin also addresses adulteration of oregano (Origanum vulgare subsp. hirtum) EO, in particular with Origanum and Thymus species, Spanish thyme blend, and synthetic carvacrol and thymol. This bulletin may serve as guidance for quality control personnel, the international herbal products industry and the extended natural products community in general. It is also intended to present a summary of the scientific data and methods on the occurrence of species substitution, adulteration, the market situation, and economic and safety consequences for the consumer and the industry.

Abstract:
Our overall, long-term goal is to exploit the genetic diversity present in cereal wild relatives for the development of cultivars with durable disease resistance. Our specific objectives for this proposal were to: 1) Utilize Association Genetics Resistance Gene Enrichment Sequencing (AgRenSeq) to identify and clone powdery mildew and leaf rust resistance gene complements in wild barley and 2) Conduct comparative sequence analyses of the cloned resistance genes to elucidate the basis of their specificity and evolution. The deployment of resistant cultivars is the most effective, economically efficient, and environmentally sound means of controlling plant diseases, especially in small grain cereals. The systems selected for study in this proposal are barley (Hordeum vulgare ssp. vulgare, Hvv), its wild progenitor (Hordeum vulgare ssp. spontaneum, Hvs) and the powdery mildew (Blumeria graminis f. sp. hordei, Bgs) and leaf rust (Puccinia hordei, Ph) pathogens. We compiled a diverse panel of Hvs accessions (the Wild Barley Diversity Collection or WBDC; N = 314) from across its native range and evaluated it to 40 isolates of Bgs and 12 isolates of Ph. We obtained genomic DNA sequences enriched for Nucleotide Binding Site-Leucine Rich Repeat (NLR) type resistance genes for 203 WBDC accessions, plus cultivar Morex for which the first reference genome sequence of barley was based. We assembled the 250 bp Illumina sequencing reads into contigs using CLC assembly cell. From this effort, we successfully assembled the sequences of 201 WBDC accessions plus Morex and used NLR Parser to identify contigs containing NLR genes. AgRenSeq was then used to identify k-mers (short oligonucleotide sequences of length k) that were associated with resistance to each isolate of the two pathogens. This analysis was performed individually for all WBDC accessions and each individual pathogen race (9,898 host accession x pathogen race combinations). We visualized the results from these analyses in Manhattan plots and identified 311 and 144 peaks for powdery mildew and leaf rust resistance, respectively. The next step in the analysis was to identify the contigs associated with the peaks in the Manhattan plots. BLAST (Basic Local Alignment Search Tool) searches were employed to identify closely related contigs in other WBDC accessions or in Morex. We identified two candidate R genes that were only present in resistant WBDC accessions. One of these was present in seven WBDC lines and was associated with resistance to four leaf rust isolates. BLAST analysis of this gene revealed that it was Rph15, one of the most widely effective leaf rust resistance genes reported in Hordeum. This gene was cloned and functionally validated in association with our Australian colleagues (Cheng et al., 2021). We are currently in the process of cloning six of other resistance genes: four for powdery mildew and two for leaf rust. As the contigs do not contain much of the promoter sequences, we have employed a genome walking approach to identify 2,500 bp of promoter sequence. To speed up and simplify the cloning of resistance genes from the WBDC, the PI established the International Wild Barley Sequencing Consortium (IWBSC; https://iwbsc.umn.edu/) comprised of over 60 researchers from 14 different countries and raised over $150,000 through crowdfunding to pay for 10X depth sequence coverage. Genome-wide association study (GWAS) of whole genome sequencing (WGS) data identified extremely strong and clear signals of association for several resistance genes which will facilitate gene cloning in concert with a wild barley pan-genome currently under construction. The cloning of multiple resistance gene can facilitate the development of durably resistant cultivars by inserting, through transgenesis, cassettes of multiple resistance genes.

Methods:This was a prospective study, conducted among 200 patients complaining of Vulvovaginal dischargeSpeciation was done as per standard microbiological methods. Non albicans Candida species were identied. Antifungal resistance was determined by disc diffusion method for uconazole, Voriconazole and by microbrothdilution for uconazole. Results: A total of 200 samples were collected from patients complaining of vaginal discharge . Out of them 69 were identied as Candida species , 31[44%] were C.albicans and 38 [56%] were non albicans Candida among them C.glabrata 22 (57%) , C.tropicalis 12 (31%), and 4 (10%) C.krusei. In the Interpretation and conclusion: present study, all the isolates of C. krusei 4 [10% ] tested showed resistance to uconazole by both disc diffusion and microbroth dilution methods . All isolates of C.glabrata 22 [ 57%] and C. tropicalis 12[31%] , tested were sensitive to uconazole by both disc diffusion and microbroth dilution . For voriconazole there was no resistance among all isolates tested by disc diffusion method. It is essential to perform susceptibility testing for all the Candida isolates for providing crucial information about the resistance pattern and help in choosing the appropriate antifungal drug for therapy. Disc diffusion method which is easy to perform can be utilized for day to day practice

Abstract:
Real-time ray tracing is an important tool in computational research. Among other things, it is used to model sensors for autonomous vehicle simulation, efficiently simulate radiative energy propagation, and create effective data visualizations. However, raytracing libraries currently offered for GPU platforms have a high level of complexity to facilitate the detailed configuration needed by gaming engines and high-fidelity renderers. A researcher wishing to take advantage of the performance gains offered by the GPU for simple ray casting routines would need to learn how to use these ray tracing libraries. Additionally, they would have to adapt this code to each GPU platform they run on. Therefore, a C++ API has been developed that exposes simple ray casting endpoints that are implemented in GPU-specific code for several contemporary device platforms. This API currently supports the NVIDIA OptiX ray tracing library, Vulkan, AMD Radeon Rays, and even Intel Embree. Benchmarking tests using this API provide insight to help users determine the optimal backend library to select for their ray tracing needs. HPC research will be well-served by the ability to perform general purpose raytracing on the increasing amount of graphics and machine learning nodes offered by the DoD High Performance Computing Modernization Program.

Abstract:
Invasive exotic plant (IEP) species are a significant threat to natural ecosystem integrity and biodiversity, and controlling them is a high priority for the National Park Service. The Northern Colorado Plateau Network (NCPN) selected the early detection of IEPs as one of 11 monitoring protocols to be implemented as part of its long-term monitoring program. This report represents work completed at Colorado National Monument during 2021. During monitoring conducted June 9–15, a total of 15 priority IEP species in 395 patches were detected on 53.6 kilometers (33.3 mi) of monitoring routes. An additional four species were detected in transects. Yellow salsify (Tragopogon dubius), ripgut brome (Anisantha diandra), jointed goatgrass (Cylindropyrum cylindricum), and tamarisk (Tamarix sp.) were the most commonly detected priority IEPs along monitoring routes, representing 74% of all priority patches. Except for ripgut brome and broad-leaf pepperwort (Cardaria latifolia), most patches of priority IEPs were than 40 m2. Ninety-five percent of tree patches were classified as seedlings or saplings, which require less effort to control than mature trees. Cheatgrass (Anisantha tectorum) was the most common IEP in transects, found in &gt;63% of transects along every route surveyed. When segments of monitoring routes consistently surveyed from 2005 to 2021 were analyzed, Red Canyon, Wedding Canyon, and Fruita Canyon all had the highest number of IEPs per 100 meters during the entire monitoring period. Increases were driven by increases in jointed goatgrass on all three routes and yellow salsify (Tragopogon dubius) in Wedding Canyon and tamarisk in Red Canyon. Jointed goatgrass appears to be rapidly expanding, increasing from 8 patches in 2019 to 42 patches in 2021 (though the routes surveyed were not all the same). In addition, three species not seen in recent years—Canada thistle (Breea arvensis), bull thistle (Cirsium vulgare), and broad-leaf pepperwort—were detected, but in fewer than 10 patches each. These species should be prioritized for control. The No Thoroughfare, Ute Canyon, and Monument Canyon routes all have stayed relatively stable over time, but at least one patch of jointed goatgrass was found on each. Network staff plans to return to re-sample monitoring routes in 2023.

According to data from 2017 to 2021, an estimated 47,984 new cases of human papillomavirus (HPV)-associated cancers were reported in the United States each year, including 26,280 among females and 21,704 among males. Cervical cancer is the most common HPV-associated cancer among females, and oropharyngeal cancers (cancers of the back of the throat, including the base of the tongue and tonsils) are the most common among males. HPV is a recognized cause of cancer. Although most HPV infections are asymptomatic and clear spontaneously, persistent infections can progress to precancer or cancer. HPV causes most cervical cancers, as well as some cancers of the vagina, vulva, penis, anus, and oropharynx. Cancer registries do not routinely collect information about HPV status, so in this report, HPV-associated cancers are defined as those that occur in parts of the body where HPV is often found.
