This Thesis examines the formation and evolution of early-type elliptical and S0 galaxies. In order to shed light on the key processes which lead to the formation of elliptical and S0 galaxies a range of techniques have been employed. The kinematics and stellar populations of the major and minor axes of the nearby edge-on S0 galaxy NGC 3115 have been examined through deep Gemini-GMOS longslit spectroscopy. The behaviour of the radial profiles of the line strength indices is found to be well explained by a simple model where NGC 3115 is composed of two components; an older classical spheroidal

In this thesis, we present a new photometric catalogue of the local Coma galaxy cluster in the Johnson U- and V- bands. We cover an area of 3360arcmin(^2) of sky, to a depth of V = 20mag in a 13 arcsec diameter aperture, and produce magnitudes for ~ 1400 extended objects in metric apertures from 8.8 to 26arcsec diameters. The mean internal RMS scatter in the photometry is 0.014mag in V, and 0.026mag in U, for Via(_13) < 17mag. In this thesis I investigate the dynamics and stellar populations of a sample of 28 edge-on early-type (S0--Sb) disk galaxies, 22 of which host a boxy or peanut-shaped bulge. I begin by constructing mass models of the galaxies based on their observed photometry and stellar kinematics. Subject to cosmologically motivated assumptions about the shape of dark haloes, I measure in a purely dynamical way their stellar and dark masses. We have measured central line strengths and line-strength gradients for a complete sample of early-type galaxies in the Fornax cluster, comprising 11 elliptical and 11 lenticular galaxies, more luminous than M(_B) = -17. We find that the centres of Fornax ellipticals follow the locus of galaxies of fixed age in Worthey's models and have metallicities varying roughly from half solar to 2.5 times solar. Chemical abundances as measured from stellar spectral lines are often subject to uncertainties due to lack of accurate data for inelastic collisions, which is needed for non-local thermodynamic equilibrium (non-LTE) modelling. For cool stars, understanding of collision processes with electrons and hydrogen atoms is required to achieve high precision measurements. In this thesis, I have investigated the role of these collisions on the non-LTE formation of Li and Mg spectral lines in late-type stars. The formation of gas hydrates in the oil and gas industry causes numerous problems that require costly solutions and operation downtime. A great deal of hydrate research has focused on their prevention either through kinetic or thermodynamic inhibitors. Recently, antifreeze proteins (AFPs) produced by cold adapted organisms have been found to have a kinetic inhibitory effect on clathrate hydrates.<br>Kinetic experiments were conducted on the methane-water system in the presence of AFPs by measuring the gas uptake during the formation of methane hydrate in a 610 cc high pressure crystallizer.
