Abstract:
The TyrA protein family includes prephenate dehydrogenases, cyclohexadienyl dehydrogenases and TyrAas (arogenate dehydrogenases). tyrAa from Synechocystis sp. PCC 6803, encoding a 30 kDa TyrAa protein, was cloned into an overexpression vector in Escherichia coli. TyrAa was then purified to apparent homogeneity and characterized. This protein is a model structure for a catalytic core domain in the TyrA superfamily, uncomplicated by allosteric or fused domains. Competitive inhibitors acting at the catalytic core of TyrA proteins are analogues of any accepted cyclohexadienyl substrate. The homodimeric enzyme was specific for L-arogenate (Km=331 μM) and NADP+ (Km=38 μM), being unable to substitute prephenate or NAD+ respectively. L-Tyrosine was a potent inhibitor of the enzyme (Ki=70 μM). NADPH had no detectable ability to inhibit the reaction. Although the mechanism is probably steady-state random order, properties of 2′,5′-ADP as an inhibitor suggest a high preference for L-arogenate binding first. Comparative enzymology established that both of the arogenate-pathway enzymes, prephenate aminotransferase and TyrAa, were present in many diverse cyanobacteria and in a variety of eukaryotic red and green algae.

ABSTRACT In order to get insights into the feedback regulation by tyrosine of the Escherichia coli chorismate mutase/prephenate dehydrogenase (CM/PDH), which is encoded by the tyrA gene, feedback-inhibition-resistant (fbr) mutants were generated by error-prone PCR. The tyrA fbr mutants were selected by virtue of their resistance toward m-fluoro-d,l-tyrosine, and seven representatives were characterized on the biochemical as well as on the molecular level. The PDH activities of the purified His6-tagged TyrA proteins exhibited up to 35% of the enzyme activity of TyrAWT, but tyrosine did not inhibit the mutant PDH activities. On the other hand, CM activities of the TyrAfbr mutants were similar to those of the TyrAWT protein. Analyses of the DNA sequences of the tyrA genes revealed that tyrA fbr contained amino acid substitutions either at Tyr263 or at residues 354 to 357, indicating that these two sites are involved in the feedback inhibition by tyrosine.

The rapid number of social media users and changes in consumer behavior today requires marketers to adapt quickly. The use of celebrity endorser "Titan Tyra" in the #JajanAsikDiXXI campaign on Instagram is one of XXI's digital promotional activities. This study measures the effect of celebrity endorser characteristics "Titan Tyra" in the #JajanAsikdiXXI campaign on the attitudes of Communication Science students at Bakrie University toward the XXI brand. This research uses a quantitative approach with an explanatory case study method. The data was collected using questionnaires, observation, tracing professional documents, and digital documentation. The distribution of questionnaires was carried out to 86 active students of Communication Science at Bakrie University using cluster random sampling where the clusters used were classes from each course that took place in the even semester of 2019/2020. The results showed that all the characteristics of celebrity endorsers attached to Titan Tyra had a significant and simultaneous influence on the attitudes of active students of Communication Science at Bakrie University towards the XXI brand. This research is worthwhile to provide insight to marketers about promotional activities using celebrity endorsers to increase positive brand attitudes.

Abstract:
This descriptive qualitative research essentially aimed to find out the types of code mixing on Brown Sugar Battle video. Titan Tyra published the video on June 17th, 2019. The researchers chose the utterances of Titan Tyra that contained code mixing as the data source of this research. In collecting the data, the researchers conducted observational method and non-participatory technique. Additionally, in analyzing the data, the researchers applied bilingual-differentiating identification method and differentiating comparative link technique. Furthermore, the researcher applied the theory of code mixing proposed by Muysken (2000). The result revealed that the speaker frequently inserted English words and phrases in the utterance of Indonesian language. The speaker mixed language to do the review of brown sugar drink. From 53 data, there were 31 data of insertion and 22 data of congruent lexicalization. The type of insertion became the most frequently uttered type. Also, the speaker did not utter English clause in utterances.

Abstract:
Understanding how the human brain functions on a molecular and cellular level is nearly impossible with current technology and ethical considerations. Utilizing the small nematode, Caenorhabditis elegans, and its innate behavioral responses to olfactory social cues, we can begin to unravel the mechanisms underlying social behavior. This is made possible given that innate behaviors are crucial for survival, and therefore hardwired into the genome of organisms. This allows for genetic-level analysis of neural circuitries driving behavior. Studying the neuronal mechanisms underlying C. elegans’ behavioral responses to social cues will not only assist in our overall understanding of how the brain perceives stimuli to enact a behavioral response at the cellular and molecular level, but also our understanding as to how the nervous system properly integrates information to enact social behavioral responses: mis-integration and social abnormalities are commonalities seen in many neuropsychiatric disorders, and these studies will provide fruitful insights into the defects observed in these disorders. Lastly, by comparing the perception of several different types of social chemicals, we can further our understanding of neural coding strategies for the various behaviors crucial for survival. Chapter One of this thesis orients the reader to social, innate behavior, and the usefulness of C. elegans as a tool for understanding behavioral coding. Chapter Two explores and establishes the required components of a socially aversive pheromone, providing insight into signaling evolution and co-option of biological machineries. Chapter Three examines how multiple, competing stimuli are integrated to modulate behavioral output, furthering our understanding of molecular and cellular integration and decision making within the nervous system. Chapter Four highlights the importance of predator pressure, and provides insights into circuit strategies of redundant and promiscuous networks of threat detection. Lastly, Chapter Five considers the implications of these findings as a whole, in the perspective of evolutionary strategies leading to neuronal coding of different behavioral outputs. Taken together, this dissertation aimed to fill the void in our understanding of social behavior neural circuitries, and how integration governed at the molecular and cellular level of the nervous system affects those behaviors.

Abstract:
This thesis describes and analyzes the symbolist artist Tyra Kleen’s life as a student and professional artist in late 1900th- and early 2000th-century, using biography as methodology, in combination with Pierre Bourdieu’s theories on Cultural Fields, and Feministic theory. A few of her Symbolistic art works will be analyzed using Ragnar Josephson’s theories on the Making of Art. The years in between 1892 – 1908 are in focus of the thesis, when the artist mainly lived, studied and worked abroad; in Germany, Paris and Rome.  Tyra Kleen (1874-1951) was born in Sweden, but lived for many years in Europe. She saw herself more as a European or a Cosmopolitan, than a Swedish person. She went to several art schools in Europe; in Dresden, Karlsruhe, Munich, Paris and Rome. She lived in Paris during the years of ”the fine de siècle”, and then moved to Rome where she lived and worked for almost ten years. During her years abroad, with influences from artists as Böcklin and Puvis de Chavannes, authors as Charles Boudelaire and Edgar Allan Poe, and from Theosophy, she picked up a continental Symbolistic style, that is unique, and is not to be found in any other Swedish artists’ styles. The thesis gives a new piece to the puzzle regarding Swedish late 1900th-early 2000th-century Art History.

ABSTRACT This dissertation examines how Tyra Banks, Shonda Rhimes, and Tyler Perry negotiate blackness in terms of racial representation both in their interactions with the press and public as well as in their final product. Banks, Rhimes, and Perry are among the few prominent African American executive producers working in an industry of inequality. Each is the creative figure behind a prominent prime-time television show. This project contributes to the discussion of race and representation in the field of television studies. I argue there is a connection between how Banks, Rhimes, and Perry publicly discuss race and how these perspectives are encoded in America’s Next Top Model (Banks), Grey’s Anatomy (Rhimes), and House of Payne (Perry) from 2005-2010. These three are vital case studies because their shows offer a range of African American representations and extra-textual discourses about representations. Chapter Two historically contextualizes the industrial shifts in mainstream broadcast networks and basic cable channels as it relates to blackness onscreen and diversity behind the scenes. ABC, The CW, and TBS are the focus of this chapter because they are the outlets for Banks, Rhimes, and Perry’s shows. I also position ABC, The CW, and TBS in relation to the rest of the industry as it has moved from the multi-channel transition to the post- network era. Chapter Three examines how Banks, Perry and Rhimes promote and publicize themselves as the key creative figure of their shows. This analysis places each individual in different sites of the burden of representation, which each handles differently. Chapter Four explores the connection between the role blackness plays in their image as public creative figure and how it is represented in their texts through a representational analysis of America’s Next Top Model, Grey’s Anatomy and House of Payne. This dissertation advocates a neo-racial framework to examine blackness on television and behind the scenes. A neo-racial framework acknowledges that racial inequities continue to exist and the context surrounding these inequities needs to be examined. I conclude that the Banks, Rhimes, and Perry cases show that we are not in a post-racial society or in the post-network era.

This essay examines the possible meaning of light as a symbol according to Tyra Kleen. The illustrated prose poetic tale En psykesaga (1902) is packed with mythological and symbolist references, and it becomes clear from the first page of the saga that Kleen was truly a symbolist of her time. The story is a paraphrase of the myth of Cupid and the Psyche, but with a new independent agent. To investigate the meaning of light, the thesis studies contextual studies in relation to the symbolism of Facos and Lundberg, in order to get close to the meta poetry of En psykesaga. In addition, a dream theoretical perspective according to Freud's psychoanalysis has been applied for a further dimension of the possible meaning of light.

Abstract:
These works belong to the research thematic related to the identification of specific molecular markers for each French cattle breeds. They consist in the study of the tyrosinase family genes and of the RAB38 gene. The cloning and the polymorphism study of the coding region of TYR, TYRP1, DCT, RAB38 genes show a very good conservation at the genomic and proteic levels with other species. No racial polymorphism correlated to a phenotype has been detected. However, the transcriptional study shows that highly variable expression levels could explain the coat color differences. Notably, a lack in TYRP1 expression seems to be characteristic of the e/e animals at the EXTENSION locus. These works also confirm the epistatic property of the SILVER allele specific to the Charolaise breed

Abstract:
Studying rubber wear is important because it can save money, minimize health and environmental issues related to the particles generated from tyre wear and reduce fuel consumption. The wear of rubber is considered to be the result of energy dissipation due to friction. There are many models that describe the dynamical behaviour of vehicles and tyre, but less e ort has been dedicated to consider the tyre wear in these models. The purpose of the thesis was to create an easy to understand and trend-accurate tyre wear model for implementation in a complete car model. The tyre wear in the thesis is determined to be the amount of rubber volume loss due to sliding per unit length that the tyre travels. A literature study was performed with the objective of gaining knowledge of tyre models and the a ecting parameters of tyre wear. The most important parameters in determining tyre wear were identied as the forward velocity, side-slip angle, longitudinal slip, vertical load, and tyre ination pressure. The wear was chosen to be calculated with Archards wear law for these parameters both separately and combined in pairs in order to obtain a deeper understanding of the wear. The results show that wear is increasing exponentially for the forward velocity. Tyre wear decreases linearly as tyre ination pressure (vertical bristle sti ness) increases. The vertical load, longitudinal slip and side-slip angle yielded exponentially increasing wear. The most inuential parameters a ecting the tyre wear were the longitudinal slip and side-slip angle, these yielded wear rates up to 107 higher compared with the reference case. The developed tyre wear model is a good base for future work. More measurement data are needed in order to validate the model. For future work it is also recommended to implement camber angle and temperature dependency in order to study these two important parameters inuence on tyre wear.

Due to the dimensions of the tyre-road contact area, transients in a tyre last approximately 0.1s. Thus, in the case of abrupt maneuvers such as ABS braking, the use of a steady-state tyre model to predict the vehicle’s behavior would lead to significant errors. Available dynamic tyre models, such as Pacejka’s MF-Tyre model, are based on steady-state formulations and the transient behavior of the tyre is included by introducing a first order differential equation of relevant quantities such as the slip angle and the slippage. In these differential equations the most significant parameter used to describe the transient behavior is the so-called relaxation length, i.e. the distance traveled by the tyre to settle to a new steady–state condition once perturbated. Usually this parameter is assumed to be constant.

Abstract:
The driver subjective perception of a vehicle is strongly determined by its Noise, Vibration and Harshness (NVH) behavior. Consequently, the NVH performance has become an important design and marketing criterion for vehicle manufacturers. The low-frequency (0–500Hz) noise and vibration perceived by a passenger are mainly determined by the dynamic behavior of rolling tyres. Besides the increasing awareness for the problems associated with road traffic noise has lead to the demand for more quiet tyres and road surfaces. Computer aided engineering tools play an important role for improving the vehicle vibration behavior at the earliest step of the design. In order to carry out precise modeling of the vehicle and its subsystems, an accurate tyre model is a key element in predicting the performance of the vehicle system with respect to ride comfort, NVH, durability, safety (braking manoeuvre) and tyre/road interaction. To obtain the best possible performance from a tyre model, a number of different measurements are required to support the tyre model parameter identification process. Although tyre vibration and noise behavior has been studied for several decades, there are still some missing links in the process of accurately predicting and controlling the overall tyre/road noise and vibration linked to the difficulty of performing modal testing on a tyre when rotating. This paper deals with the main test benches, experimental activities and modal analysis techniques available nowadays for characterizing the dynamic behavior of a static (unloaded tyre on a fixed hub and loaded tyre on a fixed hub) and a rotating tyre under different boundary conditions. The main results arising from modal analysis of a static and dynamic test on a tyre are highlighted and compared. In particular the effect of rolling speed, inflation pressure, preload, temperature and excitation amplitude on the dynamic response of a rolling tyre will be discussed.

Që prej vitit 2012, 1070 shtetas nga vendet e Ballkanit Perëndimor kanë udhëtuar për të jetuar dhe luftuar në territoret e kontrolluara nga organizatat terroriste në Siri dhe Irak. Rreth 200 syresh kanë qenë të mitur në kohën e nisjes. Me kalimin e viteve, në arenën e konfliktit në Siri dhe Irak, kanë lindur një numër edhe më i madh fëmijësh të shtetasve të Ballkanit Perëndimor. Në fund të vitit 2019, rreth 485 individë ishin kthyer në shtëpi, duke e bërë Ballkanin Perëndimor rajonin me përqendrimin më të lartë të të kthyerve nga Siria dhe Iraku në Evropë. Qindra të tjerë, kryesisht të mitur, mbeten të bllokuar në Siri në kushte të rrezikshme, me perspektiva të pasigurta riatdhesimi. Shumë shtete po hasin vështirësi në vendimmarrjen e tyre lidhur me trajtimin e shtetasve që kanë udhëtuar për të jetuar dhe luftuar në të ashtuquajturin "kalifat" të Shtetit Islamik (IS). Megjithatë, përvoja dhe të kuptuarit e faktorëve që lehtësojnë angazhimin efektiv me të kthyerit nga Siria, e veçanërisht me fëmijët e kthyer, është ende në fazat e hershme. Kjo Notë Politike ofron rekomandime mbi qasjet alternative ndaj rehabilitimit dhe riintegrimit (R&R) të fëmijëve të kthyer në Ballkanin Perëndimor, duke ndërthurur praktikat e mira të kohëve të fundit me parimet orientuese të konventave, traktateve dhe protokolleve që fokusohen te të drejtat e fëmijëve.

Mes kërcënimit në rritje të ekstremizmit të dhunshëm (EDH) në të gjithë botën, vendet e Ballkanit Perëndimor përballen me sfida të mëdha në kohezionin dhe stabilitetin social. Si kudo tjetër, narrativat e militantizmin fetar, të ekstremit të djathtë dhe nacionalist përkojnë me popullatat e cenueshme të të rinjve në vendet e Ballkanit Perëndimor, ku një histori konfliktesh etnike, fetare dhe civile ka krijuar një situatë cenueshmërie ndaj rekrutimit nga terroristët brenda dhe jashtë vendit. Personat që kanë shkuar për të luftuar përkrah organizatave ekstremiste të dhunshme jashtë vendit po kthehen në vendet e tyre pas humbjeve territoriale të grupeve ekstremiste në Siri dhe Irak. Në të njëjtën kohë, ekstremizmi etnonacionalist vazhdon të fitojë terren dhe të zgjerohet në të gjithë rajonin. Ndërkohë që disa prej këtyre temave kanë marrë vëmendje të shtuar në tërësinë e literaturës aktuale, të tjera tema mbeten të nënhulumtuara. Çështjet ekzistuese të hulumtimit kërkojnë gjithashtu më shumë studime në terren dhe baza më të thella konceptuale. Boshllëqet që rezultojnë në të kuptuarit tonë kolektiv janë tregues të nevojës për hulumtime të mëtejshme mbi zhvillimin e dinamikave sociale dhe të EDH në Ballkanin Perëndimor. Një hulumtim më rigoroz dhe i bazuar në këtë drejtim mund të ndihmojë në informimin dhe përmirësimin e përpjekjeve për të parandaluar dhe luftuar ekstremizmin e dhunshëm në rajon (prevent and counter violent extremism, P/CVE). Në vitin 2019, rrjeti “RESOLVE” mblodhi ekspertë vendas dhe ndërkombëtarë për të diskutuar boshllëqet kërkimore dhe për të hartuar një listë paraprake të prioriteteve kërkimore për të çuar përpara parandalimin dhe luftën kundër ekstremizmit të dhunshëm në Ballkanin Perëndimor.3Çështjet e identifikuara më poshtë pasqyrojnë ekspertizën e tyre të përbashkët, kuptimin e thellë dhe angazhimin për analizë të vazhdueshme të tendencave dhe dinamikave të EDH në rajon.
