Abstract:
Ubiquinone (UQ) is a polyisoprenoid lipid found in the membranes of bacteria and eukaryotes. UQ has important roles, notably in respiratory metabolisms which sustain cellular bioenergetics. Most steps of UQ biosynthesis take place in the cytosol of E. coli within a multiprotein complex called the Ubi metabolon, that contains five enzymes and two accessory proteins, UbiJ and UbiK. The SCP2 domain of UbiJ was proposed to bind the hydrophobic polyisoprenoid tail of UQ biosynthetic intermediates in the Ubi metabolon. How the newly synthesised UQ might be released in the membrane is currently unknown. In this paper, we focused on better understanding the role of the UbiJ-UbiK2 heterotrimer forming part of the metabolon. Given the difficulties to gain functional insights using biophysical techniques, we applied a multiscale molecular modelling approach to study the UbiJ-UbiK2 heterotrimer. Our data show that UbiJ-UbiK2 interacts closely with the membrane and suggests possible pathways to enable the release of UQ into the membrane. This study highlights the UbiJ-UbiK2 complex as the likely interface between the membrane and the enzymes of the Ubi metabolon and supports that the heterotrimer is key to the biosynthesis of UQ8 and its release into the membrane of E. coli.

Abstract:
Coenzyme Q10 (CoQ10) is a lipid-soluble compound with important physiological functions and is sought after in the food and cosmetic industries owing to its antioxidant properties. In our previous proof of concept, we engineered for CoQ10 biosynthesis the industrially relevant Corynebacterium glutamicum, which does not naturally synthesize any CoQ. Here, liquid chromatography–mass spectrometry (LC–MS) analysis identified two metabolic bottlenecks in the CoQ10 production, i.e., low conversion of the intermediate 10-prenylphenol (10P-Ph) to CoQ10 and the accumulation of isoprenologs with prenyl chain lengths of not only 10, but also 8 to 11 isopentenyl units. To overcome these limitations, the strain was engineered for expression of the Ubi complex accessory factors UbiJ and UbiK from Escherichia coli to increase flux towards CoQ10, and by replacement of the native polyprenyl diphosphate synthase IspB with a decaprenyl diphosphate synthase (DdsA) to select for prenyl chains with 10 isopentenyl units. The best strain UBI6-Rs showed a seven-fold increased CoQ10 content and eight-fold increased CoQ10 titer compared to the initial strain UBI4-Pd, while the abundance of CoQ8, CoQ9, and CoQ11 was significantly reduced. This study demonstrates the application of the recent insight into CoQ biosynthesis to improve metabolic engineering of a heterologous CoQ10 production strain.

Abstract:
Northern analysis demonstrated that levels of ubiquitin transcript increased during the chicken testis maturation process, in agreement with the previously published increase of ubiquitin during this differentiation process. Specific probes for four different ubiquitin genes (two polyubiquitins, UbI and UbII, and two ubiquitin-fusion genes, UbCep52 and UbCep80) allowed us to analyse the expression of each individual gene. UbI polyubiquitin gene was expressed in all the tissues tested, and its transcript was the most abundant ubiquitin RNA in all of them. Unspliced UbI transcript, already detected in stressed chicken-embryo fibroblast, was also present in immature testis and reticulocytes. UbII, a chicken polyubiquitin gene not previously found expressed and not heat-shock-inducible, was specifically stimulated during the testis maturation process. Two minor ubiquitin fusion transcripts of 0.6 and 0.7 kb, corresponding to UbCep52 and UbCep80 respectively, were also found in chicken testis. Although differentially expressed, it was found that UbI and UbII chicken polyubiquitin genes had an HTF (‘HpaII tiny fragments’) island (CpG-rich and constitutively unmethylated region) in their 5′ proximal non-coding region. In addition, we demonstrated the coexistence of 3′ and/or 5′ relatively distal methylated sites together with these 5′ proximal HTF islands in both chicken polyubiquitin genes. 3′ and 5′ distal UbI CCGG sites were specifically hypermethylated in mature testis, whereas a 3′ distal UbII CCGG site was found to be about 50% methylated in all DNAs tested.

Abstract:
Sleep is a fundamental physiological process that is essential for sustaining a healthy body and mind. The gold standard for clinical sleep monitoring is polysomnography(PSG), based on which sleep can be categorized into five stages, including wake/rapid eye movement sleep (REM sleep)/Non-REM sleep 1 (N1)/Non-REM sleep 2 (N2)/Non-REM sleep 3 (N3). However, PSG is expensive, burdensome and not suitable for daily use. For long-term sleep monitoring, ubiquitous sensing may be a solution. Most recently, cardiac and movement sensing has become popular in classifying three-stage sleep, since both modalities can be easily acquired from research-grade or consumer-grade devices (e.g., Apple Watch). However, how best to fuse the data for greatest accuracy remains an open question. In this work, we comprehensively studied deep learning (DL)-based advanced fusion techniques consisting of three fusion strategies alongside three fusion methods for three-stage sleep classification based on two publicly available datasets. Experimental results demonstrate important evidences that three-stage sleep can be reliably classified by fusing cardiac/movement sensing modalities, which may potentially become a practical tool to conduct large-scale sleep stage assessment studies or long-term self-tracking on sleep. To accelerate the progression of sleep research in the ubiquitous/wearable computing community, we made this project open source, and the code can be found at: https://github.com/bzhai/Ubi-SleepNet.

Abstract:
Road traffic has many negative socioeconomic impacts on society. A key problem is the risk of deadly accidents. The risk, to a high extend is reduced in developed societies. However, the accidents are still ubiquitous. There are various approaches for reducing accidents such as improving the infrastructure, educating better drivers and incentivizing drivers for driving safe. For the latter, the way is to analyse driving behaviour and this is possible using sensors such as inertial measurement units (IMU) without hindering privacy. Insurance companies approach this issue via Usage Based Insurance (UBI) products, where the premium is dynamically calculated by evaluating the driver based on measuring vehicle dynamics and other contextual data. Due to utilization of devices that use different IMUs, generalization of measured data is an issue for correct evaluation and fairness. The thesis deals with providing tools for filling the evaluation gap of IMUs for this purpose. The study began with a survey involving IMUs in the market. Considering technical and economic aspects, the most suitable ones were selected for evaluation. A modular system called quad-IMU (QIMU) was designed and developed. A selected IMU (BMI160) was incorporated into a QIMU and compared to two widely used IMUs in two scenarios; harsh breaking and static measurement using raw digital linear acceleration measurements. Root mean square errors (RMSE) showed that the BMI160 outperformed the others by approximately one and two orders of magnitude, respectively. The QIMU showed to be a promising framework that needs to be explored further for evaluating IMUs in-house in a rapid, low-cost and reliable manner.<br>Vägtrafiken har många negativa socioekonomiska effekter på samhället. Risken, i stor utsträckning, minskar i utvecklade samhällen.

Marquette, Scarlet Jacquelyn. "Ubi Cogito, Ibi Sum: Paranoid Epistemology in Russian Fiction 1833-1907." Thesis, Harvard University, 2013. http://dissertations.umi.com/gsas.harvard:11046.

Abstract:
This dissertation addresses two questions fundamental to Russian nineteenth-century intellectual history: 1) Why does literature about paranoid psychosis figure so centrally in the nineteenth-century canon? and 2) How did the absence of an epistemological tradition of reflexive self-consciousness influence the development of Russian ideas of subjectivity? I propose that the presence of paranoia in Russian fiction extends beyond the medical or psychoanalytic aspects of character traits or themes. I argue that literary representations of paranoia perform fundamental philosophical gestures and function as "epistemological speech acts." Russian narratives of paranoia (e.g., Pushkin, Gogol, Dostoevsky, Chekhov, Garshin, Sologub) constitute a means of exploring the operations of a self-reflexive consciousness, familiar in the West through the Cartesian Cogito. In other words, the theme of paranoia in nineteenth-century Russian fiction actively responds to the regnant philosophical discourse and functions as a praxis for the exploration of philosophical questions. However, this is done in an alternative discourse to the propositional language generally favored in philosophical texts; as a result, the philosophical function of the fictions of paranoia has gone unrecognized, and the genre has been "exiled" from philosophical discourse. I argue that Russian texts of paranoid psychosis should be reconceived as venues for the play of the transcendental ego outside social or communal axes. Paranoia emerges as the Jakobsonian “dominant” within these texts, in that it is paranoia that engages with other narrative components and transforms them. Further, as prose fiction, these texts had the discursive and social capacity to resonate and divagate in ways impossible to philosophical texts. Ultimately, these narratives of paranoia are meta-epistemologies that interrogate their own discursive function and status. They raise critical questions not only about the ways in which we represent truth but about the ontological status of truth itself.<br>Slavic Languages and Literatures

Abstract:
The notion of being connected all the time and everywhere has been a primary goal of the deployment of ubiquitous computing technologies such as wireless networks in urban areas. This has endangered traditional third places and disrupted conventional human relationships, leading to social scientists referring to modern technologically mediated cities as “lonely places”. While it may be impossible to recreate the traditional third places, it is feasible to utilize ICT as a mediator for social interaction. This thesis introduces the UBI-Collage system for mediating co-located social image sharing for the purpose of establishing a third place with a community of users. The design of the system was based on a detailed study of contemporary trends in image sharing practices, in particular among younger generations. Two incremental prototypes of the system were implemented that coupled a large situated display and personal mobile devices into a user interface providing public and private views into the photo gallery. The prototypes were evaluated in form of controlled user tests conducted in a laboratory environment with 21 and 19 participants, respectively. They used the system for 30–45 minutes and then provided feedback on questionnaire statements focusing on three themes: entertainment value, the facilitation of socialising, and contribution towards an effective third place. The results of the user tests indicated that the participants generally accepted image sharing as an entertaining social activity. However, the evidence on the facilitation of socializing and contribution towards an effective third place was much less conclusive. This is at least partly due to the controlled experiment conducted in a laboratory environment that is hardly an ideal setting for establishing a third place. The user tests also revealed that user experience depended heavily on the interaction design of the system<br>Aina ja kaikkialla saavutettavissa oleminen on yksi urbaaneille alueilla käytettäville jokapaikan tietotekniikan teknonologioille kuten langattomille verkoille asetetuista tavoitteista.

The study concerning the Slavic district Neletici on the Mulde River follows up on the research done on the Slavic settlement of the Leipzig Land and also of the pagus Quezici (around and to the west of Eilenburg) and is based on the same methodical principles. Taking into account relevant research done by Horst Naumann and Ernst Eichler 96 Slavic place names are again analysed in regard to etymology, formation und meaning. Accordingly, the use of two multi-colored maps aid in illustrating the area settled by the Slavs who immigrated from Bohemia. The core area of the regio Neletici is not the territory directly around Wurzen but lies to the north between the Mulde an the Lossabach. The borders of Neletici are distinctly established by the Slavic districts Chutici and Quezici in the west and northwest and by the Slavic settlements on the Elbe River. The border to the south could be determined even more precisely by analogical research.

Abstract:
To assess pain we have tools that are one-dimensional, as the name implies, consider only a single dimension and multidimensional instruments, which provide a more comprehensive, better representing the reality of pain experience. To measure pain intensity scales are validated at the international level, including the visual analog scale (considered as a standard numeric scale for purposes of statistical analysis). A new device (UBI Algigraph) enables a new way to assess pain intensity, allowing a non-verbal communication by the patient towards health professionals. Preliminary studies suggest that Algigraph, correlates satisfactorily with the visual analog scale. The Algigraph was initially developed for instant pain situations where there is no possibility of verbalizing the experience of pain / discomfort. Therefore it could be used in procedures such as endoscopy of the upper digestive tract which, although not really produce what is commonly known as pain, can cause severe discomfort. Objective: Evaluate the UBI Algigraph for evaluate and record the discomfort caused by upper endoscopy of the digestive tract, considering the visual analog scale as the standard by which the new method is compared. Methods: We design our study with features of a preliminary study, to be generating information enabling a basis for the necessary estimates for the design of further studies on the same subject. 28 adult patients were enrolled, but 7 were subsequently excluded from analysis for technical reasons which led to incomplete records. Therefore, we analyzed data from 21 adult patients, 8 females and 13 males, 62.0 + 16.1 years old, undergone to endoscopy. A questionnaire developed for this purpose, there is personal information, the reason for endoscopy, co-morbidities, antidepressants, tranquilizers or other and surgical history. After teaching the patient about the use of the apparatus during the procedure it was observed correct use of Algigraph. At the end of the intervention, patients were asked about what was the maximum amount of discomfort, using for that VAS. Statistical analysis used the Pearson correlation. Data are presented as mean and standard-deviation. Results and Discussion: During endoscopy, we found with Algígrafo the maximum value 3,9 + 2,5, as the average 1,1 + 0,8 and number of events 8,6 + 8,5. At the end of the intervention Visual Analog Scale correlates with Algigraph, with r values of 0.53 for it maximum value (p = 0.013), of 0.16 for the average value p = 0.49) and 0.42 for the number of events (p = 0.056). Conclusion: This study found that, statistically, the data from Algigraph correlated with that recorded with VAS, with values of r that we considered acceptable. The limitations inherent in the method that we used – a comparison against a standard - although mandatory for these situations it allows us to assess the relative value of both methods. Being a pilot study, we believe that our data give an indication towards the usefulness of Algigraph during endoscopy, but new studies including more patients and evaluating other dimensions are now needed.

Redmond, Paul, Seamus McGuinness, and Klavs Ciprikis. A universal basic income for Ireland: Lessons from the international literature. ESRI, 2022. http://dx.doi.org/10.26504/rs146.

Abstract:
A universal basic income (UBI) is defined as a universal, unconditional cash payment that is made regularly, is sufficient to live on, is not means tested, carries no work requirements and is paid on an individual basis. This study examines the international evidence on universal basic incomes and identifies key issues for consideration in the design of any UBI pilot for Ireland. Despite the mainstream interest in UBI as a potential policy tool, relatively little is known about the associated consequences of such policies. Even the definition of a UBI appears to be poorly understood and is often misused in the public discourse. Several pilot studies have been recently implemented across different countries. However, some pilot studies depart from the accepted definition of UBI. For example, some are not universal, in that they only target a specific subgroup of the population and/or have eligibility restrictions based on earnings. Others provide a relatively low level of payment, which may fall short of what an individual could reasonably be expected to live on. There are a number of potentially positive impacts associated with a UBI. A universal, unconditional payment could eliminate the stigma associated with welfare receipt. If replacing existing welfare payments, a UBI would also involve lower transaction costs, both on the recipient (in terms of the application procedure) and on Government (in terms of administering the payment). Universal, unconditional payments would also avoid situations where people choose not to work in order to retain means-tested benefits. UBI could give individuals the freedom to turn down or leave insecure, exploitative or low-paid work in pursuit of better or improved work opportunities. In addition, it would mean that persons in informal and often unpaid work, such as childcare and eldercare, which is mostly done by women, receive some compensation for their labour. Empirical results from several pilot studies have found evidence of positive health impacts following the implementation of a UBI. In terms of potential disadvantages, a UBI, by definition, may not target those that are most in need, as a large percentage of recipients will be high-earning individuals. Furthermore, the cost of a UBI is likely to be very expensive, even if other existing benefits (such as unemployment benefits) are no longer required. The net impacts of a UBI on labour supply are unclear, with both positive and negative influences on labour market participation potentially arising as a consequence of a UBI. In this study, we undertake some basic calculations relating to four possible UBI approaches, all of which would involve an unconditional payment to every individual aged over 18 in Ireland.

Abstract:
Previous research has demonstrated the use of single-channel porcine-derived urinary bladder matrix (UBM) conduits in segmental-loss, peripheral nerve repairs as comparable to criterion-standard nerve autografts. This study aimed to replicate and expand upon this research with additional novel UBM conduits and coupled therapies. Fifty-four Wistar Albino rats were divided into 6 groups, and each underwent a surgical neurectomy to remove a 7-millimeter section of the sciatic nerve. Bridging of this nerve gap and treatment for each group was as follows: i) reverse autograft—the segmented nerve was reversed 180 degrees and used to reconnect the proximal and distal nerve stumps; ii) the nerve gap was bridged via a silicone conduit; iii) a single-channel UBM conduit; iv) a multi-channel UBM conduit; v) a single-channel UBM conduit identical to group 3 coupled with fortnightly transcutaneous electrical nerve stimulation (TENS); vi) or, a multi-channel UBM conduit identical to group 4 coupled with fortnightly TENS. The extent of nerve recovery was assessed by behavioural parameters: foot fault asymmetry scoring measured weekly for six weeks; electrophysiological parameters: compound muscle action potential (CMAP) amplitudes, measured at weeks 0 and 6; and morphological parameters: total fascicle areas, myelinated fiber counts, fiber densities, and fiber sizes measured at week 6. All the above parameters demonstrated recovery of the test groups (3-6) as being either comparable or less than that of reverse autograft, but none were shown to outperform reverse autograft. As such, UBM conduits may yet prove to be an effective treatment to repair relatively short segmental peripheral nerve injuries, but further research is required to demonstrate greater efficacy over nerve autografts.
