Abstract:
The aim of our study was to compare the macrozoobenthos structure as well as water and organic matter content of the bottom sediments of two hydrologically different zones of a strongly fluvial the Włocławek Dam Reservoir. Samples were collected from the Włocławek Dam Resevoir at six sites. Three of them were located in the upper, rheolimnic part (URP) of the reservoir and three others in its lower, limnetic part (LLP). Water transparency, sediment water content and organic matter content in the sediments were higher in the LLP. The higher number of taxa and diversity of macrozoobenthos were found in the URP of the reservoir. The density of bottom fauna was slightly higher in the LLP (90,990 ind. m−2) than in the UPR (73,486 ind. m−2), while the significantly higher biomass of macrozoobenthos was found in the URP of the reservoir (2314 g · m−2) than in the LLP (336 g · m−2). The dominant taxa of Oligochaeta and Chironomidae larvae did not show significant differences in the density between both zones. Some taxa were found only in the URP. In this zone, significantly higher densities of other taxa were observed. The only species with greater abundance in the LLP was Potamothrix hammoniensis (Oligochaeta). Due to the very short water residence time, the differences in the parameters under study between the URP and the LLP of the reservoir were smaller as well as the abundance of the macrozoobenthos was distinctly higher than in other reservoirs.

Abstract:
We cloned cDNAs encoding urotensin II (UII)-related peptide (URP) and UII in Japanese eel, Anguilla japonica , the former being the first such cloning in teleost fishes. Unlike the exclusive expression of UII in the urophysis, the URP gene was expressed most abundantly in the brain (medulla oblongata) followed by the urophysis. Peripheral injections of URP into eels increased blood pressure by 16.1 ± 0.8 mmHg at 0.1 nmol/kg in ventral aortic blood pressure (PVA) and with similar potency and efficacy to that of UII (relative potency of URP to UII = 0.83). URP/UII and ANG II preferentially acted on the branchial and systemic circulations, respectively, and the duration of effect was distinct among the three peptides in the order of UII (60 min) >URP (30 min) >ANG II (14 min) in PVA. Urantide, a mammalian UII receptor antagonist, inhibited the URP effect (−63.6 ± 5.2%) to a greater extent than for UII (−39.9 ± 5.0%). URP and UII constricted isolated eel branchial and systemic arteries, showing their direct actions on the vascular smooth muscle. Central injection of URP increased blood pressure by 12.3 ± 0.8 mmHg at 50 pmol/eel in PVA and with similar efficacy but less potency (relative potency = 0.47) and shorter duration compared with UII. The central actions of URP/UII were more potent on the branchial circulation than on the systemic circulation, again opposite the effects of ANG II. The similar responses to peripheral and central injections suggest that peripheral hormones may act on the brain. Taken together, in eels, URP and UII are potent cardiovascular hormones like ANG II, acting directly on the peripheral vasculature, as well as a central vasomotor site, and their actions are mediated to different degrees by the UII receptor.

Abstract:
Twenty primers of 20 mer referred to universal rice primer (URP) were developed from a repetitive sequence of rice genome. URP-PCR protocol employed stringent PCR with high annealing temperature throughout the thermo-cycling reaction, giving high reproducibility. Under the PCR condition, each single URP primer produced characteristic fingerprints from diverse genomes of bacterial species. The universal application of URP-PCR was demonstrated by applying it to 24 strains from Pectobacterium carotovoum subsp. carotovorum, 41 Agrobacterium vitis strains, 3 Xanthomonas spp. 5 Pseudomonas spp, Rhizobium sp. plant pathogenic bacteria, human and animal pathogenic bacterial strains including 6 Escherichia coli, 4 Salmonella spp., 7 Mycobacterium spp and 3 Blucella abortus strains. In addition, thermophilic bacteria were randomly isolated form high temperature compost and their URP-PCR polymorphisms were characterized with genetic relatedness. PCR approach using URP primers will be useful for studying DNA diversity of diverse prokaryotic genomes, especially at inter- and intra species levels.

Abstract:
UII (urotensin II) and its paralogue URP (UII-related peptide) are two vasoactive neuropeptides whose respective central actions are currently unknown. In the present study, we have compared the mechanism of action of URP and UII on cultured astrocytes. Competition experiments performed with [125I]UII showed the presence of very-high- and high-affinity binding sites for UII, and a single high-affinity site for URP. Both UII and URP provoked a membrane depolarization accompanied by a decrease in input resistance, stimulated the release of endozepines, neuropeptides specifically produced by astroglial cells, and generated an increase in [Ca2+]c (cytosolic Ca2+ concentration). The UII/URP-induced [Ca2+]c elevation was PTX (pertussis toxin)-insensitive, and was blocked by the PLC (phospholipase C) inhibitor U73122 or the InsP3 channel blocker 2-APB (2-aminoethoxydiphenylborane). The addition of the Ca2+ chelator EGTA reduced the peak and abolished the plateau phase, whereas the T-type Ca2+ channel blocker mibefradil totally inhibited the Ca2+ response evoked by both peptides. However, URP and UII induced a mono- and bi-phasic dose-dependent increase in [Ca2+]c and provoked short- and long-lasting Ca2+ mobilization respectively. Similar mono- and bi-phasic dose-dependent increases in [3H]inositol incorporation into polyphosphoinositides in astrocytes was obtained, but the effect of UII was significantly reduced by PTX, although BRET (bioluminescence resonance energy transfer) experiments revealed that both UII and URP recruited Gαo-protein. Finally, UII, but not URP, exerted a dose-dependent mitogenic activity on astrocytes. Therefore we described that URP and UII exert not only similar, but also divergent actions on astrocyte activity, with UII exhibiting a broader range of activities at physiological peptide concentrations.

Abstract:
Gut eubiosis is essential for the host’s health. In athletes, the gut microbiota can be altered by several factors, including diets. While eubiotic gut microbiota in elite rugby players has been reported, our survey found that university rugby players suffered from loose stools and frequent urgency to defecate. To establish the causes of the condition, the microbiota and the concentrations of organic acids in fecal samples of university male rugby players (URP) were analyzed and compared with those of age-matching, non-rugby playing males (control). Body mass indices were significantly (p < 0.05) different between groups. Chao1 index was significant (p < 0.05) lower in URP than in control. The relative abundances of phyla Firmicutes and Bacteroidetes were significantly (p < 0.05) higher and lower, respectively, in URP than in control. Potential pathobiont genera Collinsella, Enterobacter, and Haemophilus were significantly (p < 0.05) abundant, whereas beneficial Akkermansia was lower (p < 0.05) in URP than in control. Succinate, a potential causative of gut inflammation, was five-fold higher in URP than in controls. Our findings all but confirmed that the dysbiotic status of gut in URP.

Abstract:
The study was undertaken with major objective to differentiate fungal isolates belonging to various species of Bipolaris and Exserohilum infecting various agricultural crops. After routine isolation and purification of more than 35 attempts, 13 isolates were morphologically characterized into 8 species namely, Bipolaris cynodontis (IQBC-1 and DQBC-2), B. holmii (IQBH-1), B. micropus (IQBM-1), B. oryzae (DQBO-1, DQBO-2), B. sorghicola (DQBS-1, DQBS-2), B. tetramera (DQBT-1, DQBT-2), Exserohilum longirostrata (IQEL-1) and E. rostratum (DQER-1 and DQER-2). Molecular characterization of all the isolates using RAPD, ISSR and URP markers also revealed genetic variation. Out of 24 RAPD primers, only 9 (OPA-2, OPA-4, OPA-9, OPB-4, OPB-8, OPC-1, OPC-2, OPC-5 and OPE-5) could amplify the DNA sequence showing good polymorphism. Out of 20 ISSR primers, only 6 (ISSR 1, ISSR 13, ISSR 15, ISSR 17, ISSR 19 and ISSR 20) could show good polymorphism. Out of 13 URPs, only 5 (URP 4R, URP 6R, URP 9F, URP 13R and URP 38F) could amplify with a greater number of bands. In all the three makers, similarity matrix value using Jaccard’s coefficient ranged from 0.149 to 0.548 revealing genetic co-relation among isolates. Combined clustering of RAPD, ISSR and URP data showed relatively low genetic similarity coefficient ranging from 14.92 to 54.76 among isolates which revealed stronger co-relation among isolates. Cluster analysis based on UPGMA provided a clear resolution of relationship among all the 13 isolates belong to 6 Bipolaris and 2 Exserohilum species and confirmed the morphological characterization of the same. Further, molecular markers especially, RAPD, ISSR and URP have revolutionized the genetic analysis of plant pathogens including fungi and their application can greatly help in characterizing related genera and delineation of inter and intra-species complex of fungi.

Forty isolates of Macrophomina phaseolina, a pathogen causing charcoal dry root rot of soybean, cotton, and chickpea, were genetically characterized with universal rice primers (URP; primers derived from DNA repeat sequences in the rice genome) using polymerase chain reaction (URP-PCR). Out of 12 URPs used in this study, 5 primers were effective in producing polymorphic fingerprint patterns from the DNA of M. phaseolina isolates. Three primers (URP-2F, URP-6R, and URP-30F) were quite informative and produced high levels of polymorphism among the isolates of M. phaseolina. Analysis of the entire fingerprint profiles using unweighted pair-group method with arithmetic averages (UPGMA) clearly differentiated M. phaseolina isolates obtained from soybean, cotton, and chickpea hosts into specific groups. In this study, we found for the first time transferability and use of PCR primers derived from plant genomes to generate host-specific fingerprint profiles of M. phaseolina, a broad host range plant pathogenic fungus. These results demonstrate that URPs are sensitive and technically simple to use for assaying genetic variability in M. phaseolina populations.Key words: Macrophomina phaseolina, molecular variability, soybean, cotton, chickpea.

Abstract:
The urotensin II (UII) family is currently known to consist of two paralogous peptides, namely UII and UII-related peptide (URP). In contrast to UII, which has been identified in all vertebrate classes so far, URP has only been characterized in tetrapods. We report here the occurrence of two distinct URP genes in teleosts, which we have named URP1 and URP2. Synteny analysis revealed that teleost URP1 and URP2 genes and tetrapod URP genes represent three distinct paralog genes that, together with the UII gene, probably arose from the two rounds of tetraploidization, which took place early in vertebrate evolution. The absence of URP in fish indicates that the corresponding gene has been lost in the teleost lineage, whereas it is likely that both the URP1 and URP2 genes have been lost in the tetrapod lineage. Quantitative RT-PCR analysis revealed that the URP2 gene is mainly expressed in the spinal cord and the brain in adult zebrafish. In situ hybridization experiments showed that in zebrafish embryos, URP2 mRNA-containing cells are located in the floor plate of the neural tube. In adult, URP2-expressing cells occur in close contact with the ventral side of the ependymal canal along the whole spinal cord, whereas in the brain, they are located below the fourth ventricle. These URP-expressing cells may correspond to cerebrospinal fluid-contacting neurons. In conclusion, our study reveals the occurrence of four distinct UII paralogous systems in vertebrates that may exert distinct functions, both in tetrapods and teleosts.

ABSTRAKPenelitian ini bertujuan untuk mengetahui pengaruh pemangkasan akar (underground root pruning/URP) terhadap fitoremediasi tanah yang tercemar merkuri di Kokap Kulon Progo Yogyakarta. Penelitian ini dilaksanakan menggunakan rancangan acak kelompok lengkap dengan tiga ulangan. Faktor pertama adalah URP, yaitu dengan dan tanpa URP. Faktor kedua ialah takaran Jamur Mikoriza Arbuskula (JMA), meliputi 0; 50; 100, dan 150 g/pot. Hasil penelitian telah membuktikan bahwa pemangkasan akar serta inokulasi JMA ke dalam tanah sisa olahan penambangan emas telah berhasil memacu proses fitoremediasi melalui perluasan rizosfer. Serapan merkuri oleh tanaman Jati yang menjalani pemangkasan akar serta inokulasi JMA sebanyak 100% paling tinggi dibanding perlakuan lain maupun kontrol. Pada perlakuan ini, efisiensi penurunan merkuri di dalam tanah oleh tanaman jati sebesar 88,61%, sedangkan kontrol hanya mencapai 64,11%. ABSTRACTThis study aims to determine the effect of underground root pruning (URP) of the mercury-contaminated soil phytoremediation in Kokap Kulon Progo, Yogyakarta. This study was conducted using complete randomized block design with three replications. The first factor is the URP, that divided as with and without URP (root pruning). The second factor is the dose of JMA, are 0; 50; 100, and 150 g/pot. Results of studies have proven that the root pruning and inoculation of JMA into the residual gold mining soil has successfully accelerated the process of phytoremediation through the expansion of the rhizosphere. Mercury uptake by Tectona grandis, Linn F that is with root pruning and 100% JMA inoculation are highest other treatment and control. In this treatment, the removal efficiency of mercury in the soil by plants amounted to 88.61% identity, meanwhile control only reached 64.11%.

China has 287 million migrant workers and identifying and preventing their unemployment risk holds theoretical and practical significance. This study collected data in 2019 to explore the structure of migrant workers’ perception of unemployment risk. In the first stage of this study, in-depth interviews and grounded theory analysis were conducted and an interpretation framework for migrant workers’ unemployment risk perception (URP) was developed. In the second stage, a URP scale for migrant workers was developed and then tested and verified using a questionnaire survey and factor analysis. The results showed that the URP of migrant workers is composed of many dimensions: mental, financial, relationship, citizenization and re-migration.

Les réseaux de capteurs sans fil sont maintenant un moyen crédible de collecte de données sur les cultures. L'installation d'une structure de communication fixe pour relayer les données surveillées depuis la tête de grappe jusqu'à sa destination finale peut être soit impraticable en raison de la topologie du terrain, soit prohibitive en raison du coût initial élevé. Une solution plausible consiste à utiliser des véhicules aériens sans pilote (UAV) comme moyen alternatif de collecte de données et de contrôle de supervision limité de l'état des détecteurs. Dans cet article, nous considérons le cas des parcelles agricoles disjointes comprenant chacune des grappes de capteurs, organisées de manière prédéterminée en fonction des objectifs d'élevage. Cette recherche vise à trouver une solution optimale pour la recherche de UAV et la collecte de données à partir de tous les capteurs installés dans un champ de culture. En outre, le protocole de routage des capteurs tiendra compte d'un compromis entre la gestion de l'énergie et les frais généraux de diffusion des données. Le système proposé est évalué en utilisant un modèle simulé et il devrait trouver une classe parmi toutes les sous-considérations
Wireless sensor networks are now a credible means for crop data collection. The installation of a fixed communication structure to relay the monitored data from the cluster head to its final destination can either be impractical because of land topology or prohibitive due to high initial cost. A plausible solution is to use Unmanned Aerial Vehicles (UAV) as an alternative means for both data collection and limited supervisory control of sensors status. In this paper, we consider the case of disjoint farming parcels each including clusters of sensors, organized in a predetermined way according to farming objectives. This research focuses to drive an optimal solution for UAV search and data gathering from all sensors installed in a crop field. Furthermore, the sensor routing protocol will take into account a tradeoff between energy management and data dissemination overhead.The proposed system is evaluated by using a simulated model and it should find out a class among all under consideration

Abstract:
Mancha marrom ou Helmintosporiose é uma das principais doenças do trigo, causada pelo fitopatógeno Bipolaris sorokiniana, sendo responsável por grandes perdas econômicas no cultivo do trigo no mundo inteiro. Este fungo apresenta uma grande diversidade morfológica, fisiológica e genética. Com objetivo de caracterizar a diversidade molecular de amostras monospóricas de B. sorokiniana isolados de sementes do Brasil e outros países, foram utilizados 60 isolados monospóricos tendo o DNA genômico desses isolados sido submetidos à amplificação por PCR. Foram utilizados 12 oligonucleotídeos iniciadores universais construídos a partir de sequências repetidas do genoma do arroz (URP). Os perfis gerados foram analisados pelo método de médias aritméticas não ponderadas (UPGMA). O método PCR – URP permitiu obter informações importantes sobre o perfil genético das culturas monospóricas mostrando distinções entre os três conídios isolados oriundos da mesma cepa polispórica. Os oligonucleotídeos URP-30F, URP-6R, URP-17R e URP-38Famplificaram com um elevado índice de isolados. Em contra partida, os oligonucleotideos URP-13R, URP- 25F e URP-32F apresentaram um perfil de amplificação menor entre os isolados do fitopatógeno. O número total de fragmentos gerados com cada um dos oligonucleotídeos variou entre 41 e 77 fragmentos. Os oligonucleotídeos URP-17R, URP-30F, URP-32F e URP-6R apresentaram maior índice de fragmentos polimórficos. Os isolados apresentaram uma grande diversidade intra-especifíca entre os oligonucleotideos iniciadores URP e entre os conídios monospóricos. A análise forneceu informações relevantes sobre a variabilidade genética e a relação entre os isolados de B. sorokiniana.
Leaf spot disease or helmintosporiosis is one of the most important diseases of wheat cultures caused by the phytopathogem Bipolaris sorokiniana. It is responsible for great economic loss of wheat cultivation worldwide. This fungus presents a great morphologic, physiologic and genetic diversity. The aim of this study was to characterize the molecular diversity of B. sorokiniana monosporic isolates isolated from seeds from Brazil and other countries. For this purpose 60 monosporic isolates were used. The genomic DNA of this isolates were submitted to PCR amplification using 12 universal primers built from repeated sequences of rice genome (URP). The analysis of the amplification profile were calculated by Unweighted Pair Group Arithmetic Mean (UPMGA) Method. The PCR-URP method provided important information about the genetic profile to the monosporic culture, showing distinctions between the three spore isolates of the same polysporic strain. The primers URP-30F, URP-6R, URP-17R and URP-38F were able to amplify a higher percentage of the isolates been the more efficient in the study of this phytopathogen. However, the primers URP-13R and URP-32F have shown a lower amplification profile between the B. sorokiniana isolates. The number of fragments that resulted from the amplification of each primer had varied between 41 and 77 fragments. The primers URP-17R, URP-30F, URP-32F and URP-6R generated a higher number of polymorphic fragments with the isolates. The analysis provided relevant information about the variability and genetic relationship between B. sorokiniana isolates.

Abstract:
El objetivo principal de esta investigación fue analizar el resultado de tres técnicas muy utilizadas en la estrategia del scaffolding. Para llevarla a cabo, se trabajó durante un semestre con el curso de Inglés 4 del Centro de Idiomas de la URP, nivel intermedio. Para lograr esto, se desarrollaron lecciones y actividades planificadas a través de la enseñanza de las técnicas del skimming, scanning y subrayado, cuyo propósito fue el de mejorar las habilidades de comprensión lectora de los estudiantes de dicho curso. El enfoque de la investigación fue cuantitativo, de nivel explicativo y de tipo aplicada, seleccionándose el diseño experimental de clase cuasiexperimental. Se utilizó como técnica de recojo de información la encuesta y como instrumento se aplicó una prueba al grupo de control, antes y después de la aplicación de las técnicas. Luego se analizaron las puntuaciones para ver los logros de la aplicación. Los resultados confirmaron la hipótesis general al encontrarse diferencias significativas en el grupo de control debido a la aplicación de las técnicas; es decir, se logró desarrollar una mejor comprensión de los textos leídos.

Se ha probado que la metodología aplicada en el curso de Investigación de Mercados, con la simulación de estrategias de marketing del programa Markstrat, contribuye a mejorar significativamente el rendimiento académico de los alumnos de la especialidad de Administración y Gerencia. La investigación fue realizada a los alumnos del pre-grado matriculados en el curso de Investigación de Mercados durante el semestre académico 2014-I, en dos secciones, para lo cual se diseñó una investigación cuasi experimental, habiéndose considerado un grupo de control y un grupo experimental. Al iniciar la investigación se aplicó una prueba de entrada a ambos grupos, obteniéndose como resultado estadístico que entre las calificaciones de ambos no existe diferencia significativa. Seguidamente se desarrolló en curso de Investigación de Mercados, diferenciando para el grupo experimental, la aplicación de la simulación con el programa Markstrat, y para el grupo control se mantuvo el contenido del curso, tal como se venía realizando. Al final del curso, se aplicó a ambos grupos una prueba de salida, y del análisis estadístico de las calificaciones se obtuvo como resultado que entre ellas si existe diferencia significativa, determinándose el efecto positivo de la metodología con el uso del programa Markstrat en el rendimiento académico de los alumnos de Investigación de Mercados de la carrera de Administración y Gerencia en la FACEE-URP.

Abstract:
Objetivos: Determinar la prevalencia de Escherichia coli BLEE en urocultivos del Hospital Central FAP en el periodo Enero-Junio 2016. Material y métodos: Se realizó un estudio observacional, descriptivo, analítico, retrospectivo y transversal. Se revisaron 1772 aislamientos bacterianos en orina (urocultivos) generados desde el laboratorio de microbiología del Hospital en el periodo comprendido entre Enero a Junio del 2016. Resultados: Encontramos 1175 urocultivos positivos de los cuales el 26.5% resultaron ser urocultivos E coli BLEE (+).El 80 % de los urocultivos E coli BLEE (+) fueron de género femenino y el 20% masculino. La media de la edad de los pacientes con E coli BLEE (+) fue de 64+/-22,2 años, siendo la mínima de 1 año y la máxima de 102 años. En la mujer, la prevalencia de Escherichia coli BLEE (+), fue más frecuente en el grupo etáreo de 35 a 64 años (34,7%), y en el hombre fue más frecuente en el grupo etáreo mayor de 80 años (42,6%). Observamos que la media de edad de los BLEE (+) fue mayor en relación a los BLEE negativos (64 vs 60.9 años). Encontramos una mayor frecuencia de sensibilidad antimicrobiana de los aislados de Escherichia coli BLEE (+) con amikacina (91,7%), e imipenem (91,5%); y la frecuencia de mayor resistencia fue encontrada con el ácido Nalidíxico (94%), y Cefalotina (89,8%). En los BLEE negativos hubo mayor sensibilidad con la nitrofurantoína (95,5%), Cefalotina (85,1%) y gentamicina (80%), asimismo en el mismo grupo se evidenció mayor resistencia al ácido nalidíxico (64.3%). Conclusiones: La prevalencia de Escherichia coli BLEE fue del 26.5%, afectando principalmente al sexo femenino y el grupo etáreo de 35 a 64 años. En los urocultivos E. coli BLEE (+), la media de edad fue de 64 años, y en los urocultivos E. coli BLEE (-) fue 60 años. Hubo una mayor frecuencia de sensibilidad antimicrobiana de los aislados de Escherichia coli BLEE positivos con amikacina, e imipenem; y de mayor resistencia con el ácido Nalidíxico, y Cefalotina; y de los BLEE negativos hubo mayor sensibilidad con la gentamicina, Cefalotina, nitrofurantoína y mayor resistencia a ácido nalidíxico.

O filo Actinobacteria é um importante grupo de bactérias Gram positivas amplamente distribuídas nos ambientes aquáticos e terrestres, são grandes produtores de compostos biologicamente ativos e, portanto, de grande interesse biotecnológico. O gênero Streptomyces destaca-se como maior produtor destes compostos, sendo responsável por cerca de 70% dos antibióticos que hoje utilizamos. A identificação dos organismos deste gênero ainda é um desafio. Durante muitos anos a identificação foi realizada somente com base em características morfológicas e fisiológicas. Atualmente, com o avanço das técnicas moleculares, há um grande número de espécies relatadas em bancos de dados genômicos. Este trabalho tem por objetivo identificar isolados de actinobactérias presentes no Laboratório de Microbiologia Ambiental ICBS/UFRGS com auxílio das técnicas de BOX-PCR, amplamente utilizado em estudos de diversidade dentro deste filo, e URP-PCR. E, além disso, realizar purificação parcial de um composto antimicrobiano efetivo contra bactérias produzido pelo isolado Streptomyces 8S. Os primers URP e BOX1AR produziram distintos padrões de amplificação nos isolados estudados, porém não foi possível investigar as relações de similaridade entre eles. Ainda o sequenciamento da região 16S rDNA não foi eficiente para identificar as espécies. Para a purificação do composto antimicrobiano foi realizada extração líquido-líquido com o solvente acetato de etila, posteriormente cromatografia de gel-filtração (Sephadex G-75) e troca iônica (SP-Sepharose e DEAE-celulose). A atividade antimicrobiana do composto foi recuperada após cada etapa. O composto manteve-se ativo após os ensaios de estabilidade frente à adição de EDTA, enzimas proteolíticas e altas temperaturas. Isto sugere que o composto antimicrobiano não é de origem protéica. Este trabalho sugere novos estudos a partir dos resultados preliminares obtidos, como a amplificação de todo o fragmento 16S rDNA dos isolados de actinobactérias e a investigação do composto antimicrobiano através de cromatografias de alta resolução.
The Actinobacteria phylum is an important group of Gram positive bacteria widely distributed in terrestrial and aquatic environments. They are major producers of biologically active compounds and therefore of great biotechnological interest. The genus Streptomyces stands out as the largest producer of these compounds, accounting for about 70% of the antibiotics that we use today. The identification of these organisms is still a challenge. For many years the identification was carried out only based on morphological and physiology characteristics. Today, with the advance of molecular techniques, there are a large number of species reported in genomics database. This work aims to identify isolates of actinobacterias present in the Laboratório de Microbiologia Ambiental ICBS / UFRGS with the help of BOX-PCR techniques, widely used in diversity studies within this phylum, and URP-PCR. And besides that, performing partial purification of an antimicrobial compound effective against bacteria produced by Streptomyces 8S isolated. The URP and BOX1AR primers produced different amplification patterns in the isolates, but it was not possible to investigate the relationship of similarity between them. Also the sequencing of 16S rDNA was not efficient to identify the species. To purify the antimicrobial compound was carried out liquid-liquid extraction with the solvent ethyl acetate, subsequently chromatography: gel-filtration (Sephadex G-75) and ion exchange (SP-Sepharose and DEAE-cellulose). The antimicrobial in question did not adhere to the SP-Sepharose column, showing that it has negative charge. The antimicrobial activity of the compound was recovered after each step. The compound remained active after the stability tests like the addition of EDTA, proteolytic enzymes and high temperatures. This suggests that the antimicrobial compound is not a protein. This work suggests further studies based on the obtained preliminary results such as the amplification of the entire 16S rDNA of actinobacterias isolated fragment and the investigation of the antimicrobial compound using high resolution chromatography.

L’urotensine II (UII) et l’UII-related peptide (URP) sont des peptides cycliques vasoconstricteurs, ligands endogènes du GPR14. Dans ce travail, nous avons montré que l’immunoréactivité de type UII localisée dans les motoneurones spinaux humains correspond à un peptide mature de 11 aa, H-Glu-Thr-Pro-Asp-Cys-Phe-Trp-Lys-Tyr-Cys-Val-OH. Le blocage de l’extrémité N-terminale et l’amidation C-terminale n’altèrent pas l’activité contractile de l’UIIh sur des anneaux d’aorte de rat. Les Alascan et D-scan de l’UIIh-NH2 ainsi que la synthèse de peptides tronqués révèlent que l’octapeptide C-terminal UIIh(4-11) est le fragment minimal isoactif. La présence d’une chaîne latérale fonctionnalisée sur le résidu N-terminal de l’UIIh(4-11) n’est pas requise pour l’activité du peptide. En revanche, l’intégrité et l’orientation du pont disulfure sont des paramètres essentiels pour l’activité biologique de l’UIIh. La iodation de la Tyr6 de l’UIIh(4-11) potentialise l’effet vasoconstricteur du peptide. Des modifications en position 3 et 5, [Cha3]UIIh(4-11) et [4-amino-Phe5]UIIh(4-11), conduisent à des molécules aux propriétés antagonistes. Les résidus intracycliques de l’URP, qui présentent une conformation unique caractérisée par un coude -inversé, sont directement impliqués dans la liaison et l’activation du récepteur. Enfin, les analogues [D-Trp4]URP, [D-Tyr6]URP et [Orn5]URP se comportent comme des antagonistes du GPR14. L’ensemble de ces travaux démontre que 1) l’UIIh(4-11) est la séquence minimale active de l’UIIh sur le GPR14, 2) la iodation du résidu Tyr6 de l’UIIh(4-11) conduit à un agoniste 5 fois plus puissant que le peptide natif, 3) le remplacement des résidus Phe3 et Lys5 respectivement par un résidu cyclohexyl-Ala ou 4-amino-Phe, de même que l’inversion de configuration du C du Trp4 ou de la Tyr6 de l’URP constituent des voies d’accès à des antagonistes du GPR14 qui pourraient contribuer à la conception de nouvelles molécules anti-hypertensives à visée thérapeutique
Urotensin II (UII) and UII-related peptide are the endogenous ligands of a G-protein coupled receptor named GPR14. In this work, we have shown, in human, that UII-immunoreactivity is located in human spinal motoneurons and that the UII precursor is processed to generate a mature peptide of 11 residues, H-Glu-Thr-Pro-Asp-Cys-Phe-Trp-Lys-Tyr-Cys-Val-OH. N-blocked and C-terminal amidated analogues exhibited the same vasocontractile activity as hUII. Alascan or D-scan of hUII-NH2 and the synthesis of truncated fragments indicated that the C-terminal octapeptide retains full biological activity. A functionalized side-chain is not required on the first residue of hUII(4-11) to maintain full activity. Conversely, the disulphide bridge and its orientation play a crucial role in the biological activity of hUII. Monoiodination of the Tyr6 residue of hUII(4-11) enhances the contractile potency of the peptide. Modification at position 3 and 5 generated compounds, [Cha3]hUII(4-11) and [4-amino-Phe5]hUII(4-11), able to inhibit the hUII-evoked contraction on rat aortic rings. Intracyclic residues of URP, which exhibit a single conformation characterized by an inverse -turn, play a crucial role in the biological activity of the peptide. Finally, [D-Trp4]URP, [Orn5]URP and [D-Tyr6]URP behave as antagonists of GPR14. In conclusion, structure-activity relationship studies have shown that 1) the C-terminal octapeptide hUII(4-11) is the minimal core sequence, 2) monoiodination of the Tyr6 residue of hUII(4-11) yielded to an agonist that was 5 times more potent than the native peptide, and 3) substitution of the Phe3 or the Lys5 residues with 4-amino-Phe or cyclohexyl-Ala, respectively, as well as substitution of the Trp4 and the Tyr6 residues of URP by their D-isomer open new ways for the synthesis of GPR14 antagonists which may contribute to the design of novel ligands of GPR14 with therapeutic value as anti-hypertensive drugs

Hasta el 2012, con la digitalización de las señales de televisión por satélite, por cable y últimamente también de la televisión terrestre, se vienen presentando en el país requerimientos técnicos similares a los de países denominados desarrollados, con un dinamismo cada vez mayor que ha obligado la creación de empresas locales altamente especializadas en la implementación de Up Links para DTH, transmisiones deportivas con tecnología digital de última generación, inserción de nuevos canales en las grandes cabeceras CATV, la centralización de canales en las mismas, etc. El día a día de éste tipo de empresas es la configuración en planta y la instalación de complejos sistemas que incluyen encoders, multiplexers, amplificadores, decoders, módems, routers, antenas parabólicas con diámetros que van desde 1.2m hasta 9m. Los cálculos de enlace (link budget) son realizados ahora localmente sin depender ya de los cálculos de los operadores satelitales, para lo cual se emplean programas de computador avanzados y precisos.

Los caminos rurales de nuestro país son fundamentales de recorrer debido a que ellos son el único medio de acceso de las poblaciones rurales; tanto en la sierra, costa y selva; por lo que en mi condición de haber vivido en la sierra me tocó recorrer parte de ellas en su mayoría caminos de tierra y afirmados, generalmente en mal estado; me llamó la atención y me hago la pregunta: ¿cómo podría mejorar esta situación? , ya que en algunos casos los caminos rurales son peligrosos de recorrerlos por su deterioro, falta de mantenimiento y desconocimiento de las causas de tipos de fallas que se presentan. Las redes de carreteras sin pavimentar son vitales para el desarrollo de cualquier país, y particularmente para la mayoría de los países en vías de desarrollo como el nuestro que presentan un 81.8% de caminos sin pavimentar, según la Dirección de Caminos y Ferrocarriles, PROVIAS descentralizado y oficina de estadística- MTC; cómo podemos observar en el Cuadro 1.1, de 84,244.87 Km. de carreteras en el Perú, tenemos 68,931.88 Km. sin pavimentar que representa el 81.8%.

Abstract:
Abstract Kettering University (formerly GMI Engineering & Management Institute) has had a strong link with industry due to its co-op approach to engineering education. This paper presents an extension of this link by collaborating with industry under two different programs: The Ford University Research Program (URP) and the Sloan Faculty Co-op Program. This paper describes the two programs and the various reasons for interaction, documents the nature of the collaborations, outlines potential benefits, and describes the manner by which the relationships were initiated. The paper focuses on the outcomes, makes recommendations towards successful and mutually beneficial relationships, and exhibits conditions for a continuing partnership. It concludes with an outline for a successful collaboration, its implications, and proposes ideas to other institutions for developing similar programs.

Abstract:
OBJETIVO Describir la prevalencia de TUS en una población de pacientes con TMG ingresados en una Unidad de Rehabilitación de Media y Larga Estancia (URP-MyLE) y comparar las características clínicas de estos pacientes. MÉTODOS Estudio retrospectivo transversal con todos los pacientes que ingresaron durante 2020 en una URP-MyLE. Se obtuvieron los datos demográficos, clínicos y tratamiento psicofarmacológico a través de registros médicos electrónicos. Se llevó a cabo un análisis estadístico con IBM SPSS Stastistics (Chicago INC). Se dividió la muestra en dos grupos en función del diagnóstico de TUS. Se usó Chi-cuadrado para comparar variables nominales y t-student o U de Mann Whitney para variables numéricas. RESULTADOS La muestra tenía 60 pacientes (43 hombres – 71,7%; edad media: 45,77 años ± 10,88). El tiempo medio de ingreso fue de 23.7 meses (±25.6). El diagnóstico más frecuente fue Esquizofrenia (56,7%) seguido de Trastorno Esquizoafectivo (26,7%) y Trastorno Bipolar (6,7%). Un 28,3% presentaba un TUS (n=17). Concretamente, un 23% para alcohol, 11,7% para cocaína; 18.3% para cannabis; 1.7% para benzodiacepinas y 1,7% para opiáceos. Los pacientes con TUS presentó una media de tiempo de ingreso menor (18,2 meses [±13.6] vs 25,9 meses [±25.6] en pacientes sin TUS), sin significación estadística (t2=1.047; p=0,299). Los pacientes con PD no presentaron mayor proporción de DEPOT (X2=0,008; p=0,928), ni clozapina (X2=0,202; p=0,653), ni ansiolíticos (X2=0,093; p=0,761), ni antidepresivos. Tampoco presentaron mayor número de factores de riesgo cardiovascular (t2=-0,089; p=0,929); ni comorbilidades somáticas (t2=1,537; p=0,130); ni en IMC (t2=0,775; p=0,442). CONCLUSIONES 1/3 de los pacientes ingresados presentan Patología Dual (PD). Los pacientes con PD no presentaron diferencias en cuanto al perfil de tratamiento farmacológico o comorbilidad somática. Los pacientes con PD tienden a ingresar menos tiempo. Podría guardar relación con fugas o altas voluntarias motivadas por el consumo. Se necesitan programas dirigidos al TUS en estas unidades.
