The millimeter-wave rotational spectra of the ground and excited vibrational states v8= 1 and v8= 2 of the symmetric top molecule CD3CN have been analyzed again. Thel= ± 1 in v8=1,l= 0 andl= ± 2 series in v8= 2 states have been assigned respectively. The assignment and analysis of the measurements with a least – squares procedure have made it possible to obtain the rotational, quartic and sextic centrifugal distortion constants with more reliable and higher accuracy. Analysis of the v8= 2 state gave the following rotational parameters: Aζ = 62218.96 MHz and xll= 87527.70 MHz. Investigation in v8= 2 state indicates thatl-resonance is observed for this molecule aroundk=xℓℓ+(A−B)−2AζAζ−(A−B)≈4.

Abstract:
The objective to evaluate the maize yield components as a function of the top-dressing nitrogen partitioning in maize plants with conventional and transgenic technology. The experiment was carried out in the agricultural crops of 2012/2013 and 2013/2014, in the municipality of Tenente Portela-RS, Brazil. The experiment was set up in a randomized block design in a factorial scheme with two genetic technologies x 11 nitrogen fertilization treatments, arranged in three replications. The treatments were composed of top-dressing applications in the phenological stages V2 - two fully expanded leaves, (V2), V4 - four fully expanded leaves, (V4), V6 - six fully expanded leaves (V6) and V8 - with eight fully expanded leaves (V8) and split applications in V2+V4; V2+V6; V2+V8; V4+V6; V4+V8; V6+V8; and V2+V4+V6+V8. There was interaction between genetic technologies and levels of nitrogen fertilization in the maize crop. The highest grain yield was obtained with conventional technology because it presented plants with greater prolificacy, ear diameter and number of grains per row. Grain yield was superior with nitrogen fertilization in V4 and in nitrogen splitting in the V4 + V6, V4 + V8 and V2 + V4 + V6 + V8 stages.

ABSTRACT For over 3 decades, sexual development in the human fungal pathogen Cryptococcus neoformans and other fungi has been initiated by growing compatible mating partners on V8 juice medium. Although this medium is an efficient inducer of sexual development, the mechanism by which it promotes the process is unknown. To understand how V8 juice medium induces sexual development, we attempted to purify inducing factors from V8 juice, and we carried out a complete compositional analysis of V8 juice. We discovered that no single factor is responsible for the effects of V8 juice medium. Rather, the unique composition of V8 juice medium provides the proper nutrient composition for inducing and sustaining complete sexual development. Utilizing these findings, we developed a defined V8 (DV8) medium that mimics V8 juice medium in sexual development assays. Then, using DV8 as a tool, we explored the roles that specific molecules play in enhancing sexual development. Surprisingly, we discovered that copper is a key factor, leading to an upregulation of the mating pheromone genes MF a and MFα, both required for the initial steps in sexual development. The utilization of DV8 to investigate the effects of copper on sexual development presented here is an example of how defining the conditions that induce sexual development will advance the study of C. neoformans.

Effects of culture media, near-ultraviolet exposure time, and temperature on sporulation of Alternaria alternata were investigated. Strains RL 671-2 and ATCC 36068 were cultivated on Potato Dextrose Agar (PDA), V8 Juice Agar (V8 Agar) and Mycological Agar (MA). The best culture medium for sporulation of strain RL 671-2 was PDA, followed by V8 agar, with only negligible numbers of spores appearing on MA. Near-UV exposure significantly increased sporulation in strain RL 671-2 on PDA and V8 agar. Significantly higher (P<0.01) spore counts were found in PDA cultures of this strain exposed to near-UV at 35 than at 20°C. On V8 agar significantly more spores were observed at 20 than at 35°C. MA was not a satisfactory medium for sporulation of ATCC 36068. Both PDA and V8 agar equally supported sporulation for this strain (ATCC 36068) at all exposure times.

In Brazil, corn planted area increased by 30%. Therefore, there was also an increase in the incidence of pathogens in the crop. The objective of this work was to evaluate the influence of fungicide applications on corn crops in the different growth stages for the control of the diseases and its effect on the occurrence of mycotoxins in the grain. The experiment was carried out in the 2017/2018 summer crop, in the municipality of Cruz Alta, state of Rio Grande do Sul/BR. This study used P 1630 hybrid, in a randomized block design with eight treatments (control, V8, PT, V4 + V8, V4 + V8 + PT, V8 + PT, V8 + PT + 15, PT + 15) and three replicates. The fungicide fluxapiroxade + pyraclostrobin + epoxiconazole at the dose of 1.0 L/ha-1 was used. The diseases that affected the crop were the white spot and helmintosporiosis, therefore, reducing the incidence of diseases. Also the highest yields were obtained where the applications started at the phenological stage V4 and V8. The levels of mycotoxins detected in this work remained within the limits of the legislation, with a significant reduction in the accumulation of fumonisin and aflatoxin with fungicide applications.

Phospholipase C-gamma 1 (PLC-gamma 1) was treated with Staphylococcus aureus V8 protease (V8) and the digestion products were analysed with site-specific antibodies. V8 treatment generated three immunodetectable PLC-gamma 1 fragments of 120, 97, and 39 kDa. The 39 kDa fragment is derived from the C-terminus of PLC-gamma 1 and includes the conserved Y domain present in all PLC isoenzymes. The 120 and 97 kDa fragments are derived from the N-terminus of PLC-gamma 1, possess the conserved X domain common to all PLC isoenzymes, and the src-homology domains unique to PLC-gamma 1 and -gamma 2. It is likely that the 97 kDa fragment is a V8 product of the 120 kDa fragment. As the C-terminal 39 kDa fragment, and either of the N-terminal 120 or 97 kDa fragments, were precipitable with antibody specific to a sequence present in only the 39 kDa fragment, the data indicate co-precipitation of separate polypeptide chains that remain associated after V8 proteolysis. Importantly, V8 treatment increased the activity of PLC-gamma 1 and did not alter the calcium requirement. The influence of other modulators of PLC-gamma 1 activity, however, was lost following V8 treatment. These results suggest the stable association of the X and Y domains within PLC-gamma 1, and demonstrate that proteolysis in the region of PLC-gamma 1 that is subject to tyrosine phosphorylation can enhance catalytic activity.

Percutaneous balloon pulmonary valvuloplasty is the preferred therapy for pulmonary valve stenosis. However, the designs of the cylindrical balloons historically used for valvuloplasty have limitations, especially in patients who have large pulmonary annular diameters. The hourglass-shaped V8 Aortic Valvuloplasty Balloon may prove to be an effective alternative. The balloon has 2 large bulbous segments that are separated by a narrowed waist. The geometric shape is maintained throughout inflation, improving fixation and enabling broader leaflet opening. We present our first experience with the V8 balloon in 3 adults who had severe, symptomatic pulmonary valve stenosis. In addition to describing their cases, we detail our sizing technique for pulmonary valvuloplasty with the V8 balloon. Our successful results suggest that the V8 balloon is efficient and safe for balloon pulmonary valvuloplasty in adults with severe pulmonary valve stenosis.

Summary Background: Capillary electrophoresis (CE) is used for the screening of protein abnormalities in serum and other biological fluids. Since the Helena V8 CE system has been recently commercialized, the aim of this study was to assess its analytical performance under routine laboratory conditions. Methods: The evaluation of Helena V8 (Medical Systems S.p.A., Genova, Italy) for protein fractions (i.e., albumin, alpha-1, alpha-2, beta-1, beta-2, gamma, albumin/globulin ratio [A/G]) and monoclonal component (MC) quantification included imprecision studies, linearity, and comparison with Sebia Capillarys (Sebia, Evry Cedex, France), which was considered the reference analyzer. Results: The imprecision of Helena V8 on all parameters was comprised between 0.8% and 15.2%; the linearity was excellent up to an MC value of 14.5% (r=1.00). The comparison studies showed excellent Spearman’s correlations with Sebia Capillarys, with coefficients comprised between 0.84 and 0.99 (all p<0.001). The mean bias of Helena V8 versus Sebia Capillarys was -0.02% for G/A, -0.2% for albumin, 2.1% for alpha-1, 0.1% for alpha-2, 0.3% for beta-1, -0.2% for beta-2, -2.2% for gamma and 1 g/L for MC. Conclusions: The results of this evaluation attest that Helena V8 is a suitable alternative for routine separation and quantification of protein fractions and monoclonal component.

AbstractArea-wide sterile insect technique (SIT) programs assume that offspring reduction of the target population correlates with the mating success of the sterile males released. However, there is a lack of monitoring tools to prove the success of these programs in real-time. Field-cage tests were conducted under the environmental conditions of the Mediterranean coast of Spain to estimate: (a) the mating success of sterile Vienna-8 (V8) Ceratitis capitata males using molecular markers and (b) their efficacy to reduce C. capitata populations under six release ratios of wild females to wild males to V8 males (1:0:0, 1:1:0, 1:1:1, 1:1:5, 1:1:10, and 1:1:20). Statistical models were developed to predict: (a) the number of females captured in traps, (b) sperm ID (sterile or not) in spermathecae of the trapped females, and (c) the viable offspring produced, using release ratio and temperature as predictors. The number of females captured was affected by relative humidity. However, its influence in the model was low. Female captures were significantly higher in ratios 1:0:0 compared to ratios where V8 males were released. The proportion of V8 sperm in spermathecae increased with temperature and with the number of V8 males released, but leveled off between ratios 1:1:10 and 1:1:20. In all seasons, except winter (no offspring), viable offspring increased with temperature and was lowest for ratio 1:1:20. For the first time, a strong negative relationship between proportion of V8 sperm detected by molecular tools and C. capitata offspring was established. The models obtained should contribute to enhance the efficacy of SIT programs against this pest.

Abstract:
O presente ensaio foi realizado com o objetivo de avaliar a produção de biomassa micelial bem como a esporulação de Cercospora piaropi, nos meios líquidos V8, ETD (Extrato de Tomate Diluído) e BD (Batata - Dextrose), em períodos de cultivo de 96, 120, 144 e 168 h, sob agitação constante. Adicionalmente foi avaliado o efeito de períodos de desidratação da biomassa micelial (24, 48, 72, 96 e 120 h) sobre a esporulação. Os inóculos obtidos foram avaliados quanto à severidade da doença em plantas de aguapé (Eichhornia crassipes). De acordo com os resultados, o meio ETD proporcionou maior crescimento micelial em relação aos meios BD e V8, destacando-se o período de 144 h de agitação. Entretanto, o meio V8 induziu esporulação superior do patógeno, quando cultivado por 120 h. Os inóculos obtidos nos meios V8 e ETD causaram maiores valores de severidade da doença. O período de desidratação da biomassa micelial a partir de 72 h favoreceu maior produção de conídios. Não houve efeito do período de desidratação sobre a severidade da doença.

O objetivo do trabalho foi quantificar a resposta ao número de aplicações de fungicida, na intensidade de doenças foliares, produtividade, massa de mil grãos e grãos manchados em arroz irrigado. Os experimentos foram conduzidos com o cultivar SCS 116 Satoru nas safras 2011/12 e 2012/13, no município de Rio do Oeste, Alto Vale do Itajaí, estado Santa Catarina. O delineamento foi de blocos casualizados, com quatro repetições e seis tratamentos constituídos de aplicações de mistura de fungicidas triazol (difenoconazole) e estrobilurina (azoxistrobina), sendo uma aplicação em final de perfilhamento (V8), duas aplicações (V8 + R0, iniciação da panícula), três aplicações (V8 + R0 + R2, emborrachamento), quatro aplicações (V8 + R0 + R2 + R4, floração), cinco aplicações (V8 + R0 + R2 + R4 + R6, grão leitoso) e um tratamento (testemunha) sem aplicação. Antes de cada aplicação foi determinada a incidência e a severidade das doenças foliares. Nas duas safras as doenças foliares predominantes foram brusone, mancha parda e escaldadura. O aumento de uma até cinco aplicações de fungicida apresenta resposta significativa na redução da intensidade das doenças foliares, com incremento médio de 19,5%, 30%, 70,3%, 86,6% e 100,8% na produtividade, 0,2%, 13,7%, 28,5%, 41,3% e 54% na massa de mil grãos e redução de 22%, 31,8%, 44,1%, 75,9% e 168,5% de grãos manchados, em relação à testemunha.

Utilizando a equação com melhor ajuste nas duas safras e substituindo o valor de severidade de 1% obtém-se incidência de 30,80; 35,86; 25,91; 30,26; 32,90 respectivamente nos estádios fenológicos de perfilhamento, iniciação da panícula, emborrachamento, floração e grão leitoso. A severidade estimada com base na incidência e vice-versa possibilita seu uso pela assistência técnica no monitoramento das manchas foliares na cultura do arroz irrigado.

Abstract:
O objetivo com esse trabalho foi verificar os resultados do parcelamento da adubação nitrogenada com ureia na cultura do milho grão no estágio V4 e V8. O experimento foi realizado na fazenda Ponte Alta, no município de Silvânia-GO. O delineamento experimental adotado foi o de blocos ao acaso, com quatro tratamentos e quatro repetições sendo: T1 – Testemunha; T2 – Ureia 100% V4; T3 – Ureia 50% V4 e 50% V8; T4 – Ureia 100%V8. A demanda nutricional da cultura foi estabelecido de acordo com a análise do solo sendo de 261 kg ha-1 de N. Foram avaliados a altura da planta, diâmetro de colmo, massa verde e a altura da inserção da espiga até o florescimento pleno e a produtividade no final do experimento. Os resultados foram submetidos à análise de variância, e quando ocorreram diferenças significativas, identificadas pelo teste F (P<0,05), se aplicou o teste de médias de Tukey, utilizando-se programa estatístico Sisvar, versão 5.6. O milho FS 2B610 PW respondeu positivamente à adubação nitrogenada de cobertura, em todas as características estudadas. Nas condições de solo e clima da área analisada, a adubação de cobertura nitrogenada para o desenvolvimento do milho é mais eficiente quanto a aplicação do N em cobertura é realizada no estádio vegetativo V4, se assemelhando produtivamente à dose parcelada em V4 e V8. A realização da adubação nitrogenada total de forma tardia, em V8, não gera efeito positivo no desenvolvimento da planta e na produtividade.

O milho nos tempos contemporâneos tornou-se o grão mais produzido no mundo. Portanto esse trabalho objetivou avaliar o efeito da absorção de fósforo nas características biométricas e produtivas da cultura do milho nas fases V6, V8, florescimento e colheita. Os atributos avaliados foram: teor de fósforo, massa fresca de folha no florescimento, massa fresca de caule no florescimento, massa seca de folha no florescimento, massa seca de caule no florescimento, massa seca do caule, massa fresca do caule, massa fresca da folha na colheita, massa seca da folha, massa fresca de folha em V6, massa seca de folha em V6, massa fresca de caule em V6, massa seca de caule em V6, massa seca de caule em V8, massa fresca de caule em V8, massa fresca de folha em V8, massa seca de folha em V8. Através da análise de trilha foi permitido estabelecer correlações entres estas variáveis que são indicativos de produtividade e qualidade do milho. Resultados mostram pela correlação de Pierson que massa fresca de caule e massa seca de caule nas diferentes fases estudadas, apresentam altos níveis de correlação positiva indicando que suas massas frescas e secas tendem, após testes e análises, ter proximidade de relações. Como conclusões, compreende-se que de maneira direta e positiva o fósforo obteve maior correlação com o peso médio de grãos. Efeito direto e negativo apenas massa fresca de folha no florescimento obteve resultados consideráveis em relação ao teor de fósforo.

Avaliou-se a esporulação de conídios de Mycosphaerella fijiensis (isolado LPM472) em sete meios de cultura (batata dextrose ágar, V8 ágar, V8 CaCO3 ágar, água de coco ágar, batata cenoura ágar, folha de banana ágar e micophil) sob quatro regimes de luminosidade (escuro contínuo, fotoperíodo de 12 h, luz contínua e seqüencial - dez dias no escuro e cinco dias sob luz contínua). O ensaio foi conduzido em delineamento inteiramente casualizado, com cinco repetições. A esporulação foi determinada em erlenmeyer de 125 ml, contendo 20 ml de seu respectivo meio de cultura e 0,5 ml de suspensão com 5 x 10(4) conídios de M. fijiensis/ml, mantidos a 25 ºC + 2 ºC, durante 15 dias. Não ocorreu esporulação em todos os meios de cultura sob regime de escuro contínuo, assim como no meio de folha de banana ágar, sob todos os regimes de luminosidade. No regime de fotoperíodo, ocorreu esporulação apenas nos meios V8 CaCO3 ágar e michophil, e no regime de luz contínua, apenas no micophil. No regime seqüencial, os meios de batata dextrose ágar e V8 CaCO3 ágar propiciaram as maiores produções de conídios.

ABSTRACT In most Staphylococcus aureus strains, inactivation of sarA increases hla transcription, indicating that sarA is a repressor. However, in S. aureus NCTC 8325 and its derivatives, used for most studies of hla regulation, inactivation of sarA resulted in decreased hla transcription. The disparate phenotype of strain NCTC 8325 seems to be associated with its rsbU mutation, which leads to σB deficiency. This has now been verified by the demonstration that sarA repressed hla transcription in an rsbU + derivative of strain 8325-4 (SH1000). That sarA could act as a repressor of hla in an 8325-4 background was confirmed by the observation that inactivation of sarA in an agr sarS rot triple mutant dramatically increased hla transcription to wild-type levels. However, the apparent role of sarA as an activator of hla in 8325-4 was not a result of the rsbU mutation alone, as inactivation of sarA in another rsbU mutant, strain V8, led to increased hla transcription. Northern blot analysis revealed much higher levels of sarS mRNA in strain V8 than in 8325-4, which was likely due to the mutation in the sarS activator, tcaR, in 8325-4, which was not found in strain V8. On the other hand, the relative increase in sarS transcription upon the inactivation of sarA was 15-fold higher in 8325-4 than in strain V8. Because of this, inactivation of sarA in 8325-4 means a net increase in repressor activity, whereas in strain V8, inactivation of sarA means a net decrease in repressor activity and, therefore, enhanced hla transcription.
