Le déclin du système immunitaire naturellement lié à l'âge - appelé immunosénescence - contribue à la moindre efficacité des vaccins chez les personnes âgées. La sénescence cellulaire est caractérisée par une inflammation chronique de bas grade. Les cellules sénescentes s'accumulent avec l'âge et sont résistantes à la mort cellulaire, notamment du fait de l'augmentation de l'expression des membres de la famille Bcl-2. Les cellules sénescentes produisent de nombreux facteurs inflammatoires et des immunomodulateurs, ce qui contribue à l'expression de leurs fonctions délétères. Ces cellules causent de nombreux désordres liés à l'âge et l'élimination contrôlée de ces cellules pourrait représenter une stratégie thérapeutique prometteuse.Le rôle potentiel des cellules sénescentes dans le déclin de l'efficacité vaccinale n'a pas été étudié à notre connaissance. Pour aborder cette question, l'une des possibilités est d'éliminer sélectivement ces cellules à l'aide de drogues sénolytiques. Nous avons utilisé l'ABT-263 (Navitoclax), une drogue qui cible Bcl-2, et avons étudié l'effet du traitement sur la réponse immunitaire induite par vaccination. Pour cela, des souris âgées (22 mois) et des souris adultes jeunes (2 mois) ont été traitées avec le Navitoclax et quelques jours plus tard les souris ont été immunisées avec l'antigène ovalbumine (OVA) en présence d'adjuvant (Quil-A ou CpG ODN). La production des anticorps spécifiques a été étudiée par ELISA et la réponse cellulaire par quantification de l'IFN-gamma suite à une re-stimulation antigénique. Pour étudier l'efficacité de la réponse immunitaire générée par la vaccination, les souris ont été inoculées avec le mélanome B16 exprimant OVA et la croissance tumorale a été mésurée.Dans un premier temps, nous avons montré que le Navitoclax élimine les cellules sénescentes chez les souris âgées, notamment dans la rate : réduction du nombre de cellules exprimant le marqueur de sénescence p16 et Bcl-2 (RTPCR, immunohistochimie) et de l'activité beta-galactosidase, un autre marqueur de sénescence. Cet effet s'accompagne d'une réduction de la production des cytokines inflammatoires dans le sang. Par ailleurs, les splénocytes isolés des souris âgées traitées avec le Navitoclax produisent moins de cytokines inflammatoires en réponse au LPS comparativement aux animaux contrôles. Ayant validé l'efficacité du Navitoclax, nous avons mesuré la réponse immunitaire post-vaccination. La déplétion des cellules sénescentes est associée à une réduction (non-significative) de la production des anticorps (IgM et IgG). De façon contrastée, le traitement par le Navitoclax augmente de la production de l'IFN-gamma par les lymphocytes T suite à la re-stimulation antigénique. Ce dernier effet est également observé chez la souris adulte jeune. De façon inattendue, le Navitoclax abroge l'effet protecteur de la vaccination sur la croissance tumorale chez les souris âgées et dans une moindre mesure chez les souris adultes jeunes. Pour conclure, la déplétion préventive des cellules sénescentes avant la vaccination influence l'efficacité de la réponse immunitaire et a un impact négatif sur la réponse anti-tumorale chez la souris âgée<br>Age-related decline of immunity reduces vaccine efficacy in the elderly. Cellular senescence - a hallmark of aging - is a physiological process characterized by a state of chronic low-grade inflammation. Senescent cells accumulate with age and are resistant to cell death as a result of increased Bcl-2 expression. Senescent cells show an enhanced pro-inflammatory phenotype, as a part of senescence associated secretory phenotype (SASP) which contributes to inflammation and other detrimental effects. Pre-existing senescent cells cause many aging-related disorders and therapeutic strategies aiming at selectively eliminating these cells have recently gained attention.The potential role of pre-existing senescent cells in vaccine efficacy in the aged populations has not yet been reported to our knowledge. This can be achieved through different approaches such as the use of senolytic drugs that selectively target and eliminate these cells. Using the specific Bcl-2 family inhibitor senolytic ABT-263 (Navitoclax), we investigated the effects of senolysis on the immune response induced by vaccination. To this end, aged mice (22-months) and young adult mice (2 months) were treated with Navitoclax before immunization and few days later mice were immunized with the antigen Ovalbumin (OVA) plus adjuvant (Quil-A and CpG ODN). Antibody production was quantified by ELISA and the T cell response was quantified by measuring the production of interferon gamma after antigen re-stimulation. To study the efficacy of the immune response post-vaccination, mice were engrafted with OVA-expressing B16 melanoma cells and melanoma outgrowth was measured.ABT-263 treatment depleted senescent cells in the spleen. This was evidenced by immunohistochemistry using antibodies against p16 (a marker of senescence) and Bcl-2 and by quantifying beta-galactosidase activity, another marker of senescence. Depletion of senescent cells also led to a reduced production of systemic SASP-related factors in blood. In the same line, splenocytes isolated from Navitoclax-treated aged mice produced less inflammatory cytokines in response to LPS compared to controls. Having validated the efficacy of Navitoclax, we then turned to analyze the consequences of senescent cell's removal on the immune, anti-tumor response. Navitoclax treatment slightly reduced antigen-specific antibody production. Both IgM and IgG were affected. In contrast, T cells from Navitoclax-treated aged mice produce more IFN-gamma compared to controls. A similar effect was observed in young adult mice. Strikingly, depletion of pre-existing senescent cells before vaccination abrogated the protective effect of the vaccine on tumor outgrowth in aged mice, and to a lower extent, in young adult mice. We conclude that senolysis influences the quality of the immune responses post-vaccination and strongly affects the anti-tumor response in vaccinated aged mice

L'objet de ce travail a ete l'etude immunochimique des antigenes excretes-secretes par t. Gondii et l'evaluation de leur role dans l'immunite anti-toxoplasmique, dans le cadre de recherches pour la mise au point d'un vaccin contre la toxoplasmose. Apres avoir caracterise les molecules les plus immunogenes en toxoplasmose humaine et experimentale (24, 27, 35, 39, 43, 63, 78 et 98 kda), nous avons produit et caracterise des anticorps monoclonaux diriges contre les antigenes excretes-secretes. Nous avons mis en evidence dans ces antigenes la presence de molecules de surface du parasite (p22, p30, p35 et p43) et de molecules specifiques du complexe apical du parasite (45/52, 58/62, 70, 78 et 98 kda). Nous avons recherche parmi les antigenes excretes-secretes, ceux qui presentaient des epitopes communs aux tachyzoites (forme proliferative presente au cours de l'infection aigue) et aux bradyzoites (forme enkystee caracteristique de l'infection chronique) et identifie six molecules qui repondaient a ces criteres (27, 35, 39, 43, 63 et 78 kda). La comparaison des produits d'excretion-secretion des tachyzoites de la souche rh et des tachyzoites de la souche ts4 (mutant non pathogene pour la souris), a montre que les molecules de 29 et 78 kda etaient specifiquement relarguees par les tachyzoites de la souche rh, suggerant ainsi un role de ces deux molecules dans la penetration ou dans la multiplication du parasite dans la cellule infectee. Les antigenes de 26. 5 et 34/36 kda ont pu etre purifies par chromatographie d'affinite a l'aide des anticorps monoclonaux et les molecules de 27, 45/52 et 63 kda par chromatographie d'echange d'ions. Nous avons ensuite montre l'induction par les antigenes excretes-secretes d'une reponse immune t chez l'homme, et demontre le pouvoir protecteur chez la souris des antigenes excretes-secretes totaux et de deux molecules purifiees, la p35 et la p100

La protéine VAR2CSA de P. Falciparum est le candidat principal pour le développement du vaccin contre le paludisme gestationneL Cependant, le haut poids moléculaire de VAR2CSA (350 kDa) ainsi que le taux élevé du polymorphisme du gène CAR2CSA sont deux éléments limitant dans la faisabilité du vaccin. L'identification d'une région minimale de VAR2CSA incluant des épitopes majeurs de VAR2CSA est nécessaire afin de pouvoir élaborer un vaccin protégeant les femmes enceintes contre le paludisme. Nombreuses études ont été réalisées sur ce sujet, parmi lesquelles une qui a été effectuée dans notre laboratoire et qui a mis en évidence, en accord avec d'autres études récentes, la localisation N-terminale (NTS-DBL1X-ld1-DBL2X) des épitopes protectifs contre le paludisme gestationneL Dans le premier article, nous avons montré que l'usage de la technique de vaccination génétique présentait de nombreux avantages pour réaliser la cartographie antigénique des protéines, en prenant exemple de la protéine VAR2CSA. Dans le deuxième article, nous avons zoomé sur les domaines N-terminal de VAR2CSA afin de minimiser autant que possible la région incluant les épitopes protectifs majeurs. Ainsi, nous avons réalisé une cartographie raffinée sur la partie N-terminale de la protéine VAR2CSA, en utilisant 5 constructions dérivées (NTS-DBL1X, NTS-DBL1X-ld1, Idl, ld1-DBL2X et DBL2X) du fragment NTS-DBL1X-Id1-DBL2X, pour l'immunisation génétique de souris via électroporation. Les anticorps produits en réponse à chaque construction ont été testés en ELISA puis par FACS pour vérifier leur réactivité avec la protéine recombinante et les hématies infectées. Finalement, ces anticorps ont été examinés pour leur capacité à bloquer la liaison des hématies parasitées aux CSA in vitro. Nous avons identifié ldl-DBL2X comme le fragment le plus court de la protéine VAR2CSA, hautement immunogène et capable de déclencher les anticorps qui bloquent l'adhésion des érythrocytes infectés au CSA, avec la même efficacité que les antisera anti-VAR2CSA extracellulaire. Nous avons également montré que les anticorps anti-ld1-DBL2X reconnaissent spécifiquement la surface des érythrocytes infectés provenant des isolats de femmes enceintes du Bénin et que ces anticorps inhibent jusqu'à 95% selon les isolats, l'adhésion de ces érythrocytes parasités au CSA. Enfin, le dernier volet de la thèse est dédié à la caractérisation de la diversité génétique du fragment ld1-DBL2X. Nous avons donc séquence le fragment du gène VAR codant pour la région ld1-DBL2X de la protéine VAR2CSA, sur environ 380 échantillons provenant des isolats naturels VAR2CSA de P. Falciparum collectés des régions géographiques où le paludisme s'est majoritairement développé : Bénin, Sénégal, Tanzanie, Madagascar, Afrique, Cambodge, Asie, Pérou, Amérique Latine, Papouasie Nouvelle Guinée et en Océanie<br>VAR2CSA is considered as the main target of protective immunity against pregnancy-associated malaria. VAR2CSA high molecular weight complicates scaling up production of VAR2CSA recombinant protein for large-scale vaccination programmes. We previously demonstrated that antibodies induced by NTS-DBL1X-ld1-DBL2X efficiently block parasite binding to CSA in a similar manner to antibodies induced by the full-length extracellular part of VAR2CSA. This work aimed first at identifying the shortest fragment of VAR2CSA carrying major protective epitopes able to elicit inhibitory antibodies. To achieve this goal we performed a refined antigenic mapping of NTS-DBLlX-ld1-DBL2X through a DNA vaccination technique. Likewise, five single or double domains constructs encoding NTS-DBL1X, NTS-DBL1X-ld1, Id1, ld1-DBL2X and DBL2X were made and used to immunize mice. The NTS-DBL1X, NTS-DBLIX-ld1, and ldl-DBL2X fragments all raised high titer immune response, as measured by ELISA. The DBL2X fragment raised a weaker antibody titer, and the Id1 construct failed to elicit antibody. Sera from mice immunized with NTS-DBL1X or DBL2X constructs failed to block infected erythrocytes binding to CSA, whereas sera from mice immunized with NTS-DBLIX-ld1 showed partial inhibitory activity, and the ld1-DBL2X fragment elicited antisera that totally abrogated infected erythrocytes adhesion to CSA. IgG purified from ldl-DBL2X antisera showed a similar inhibitory profile than ld1-DBL2X antisera. Anti-FCR3 anti-ld1-DBL2X antibodies also efficiently block the adhesion of erythrocytes infected by the HB3 parasite line to CSA. Ld1-DBL2X antisera recognized the surface of field isolates from pregnant women, and inhibited CSA-binding of all 8 isolates tested, although to a variable level. We raised high-titer antibodies against several parts of the protein, and identified ldl-DBL2X as the minimal VAR2CSA fragment inducing antibodies with CSA-binding inhibitory efficiency in the same range as the full-length extracellular part of VAR2CSA. The second part of the work is dedicated to a global characterization of genetic diversity of id1-DBL2X fragment within eight Worldwide isolates of natural P. Falciparum populations coming from four continents. To perform this study, we have take advantage of Next Generation Sequencing (NGS). A moderate level of genetic differentiation is observed up to the position 569 of the nucleotide sequence, whereas a significant increase is observed in the following part of the fragment. Interestingly, this boundary nearly matches the separation between the Id1 and DBL2X domains. This means that on Id1 fragment the genetic differentiation between the eight populations is moderated while it become much more important on DBL2X level

La vaccination est l'un des plus grands progrès réalisés en santé publique. Toutefois, malgré de nombreuses connaissances sur le système immunitaire, de nombreux pans d’ombre empêchent la conception de vaccins contre des pathogènes complexes. Pour pallier ce problème, une meilleure compréhension des modes d'action des vaccins est requise. En particulier, la plupart des vaccins nécessitent plusieurs immunisations pour induire une mémoire immunitaire adaptative au long terme, mais l'impact du délai entre primo-vaccination, induisant une mémoire primaire, et rappel(s) la restimulant pour générer une mémoire secondaire, est peu défini. De plus, la réponse immunitaire innée, induite à chaque immunisation et façonnant l'immunité adaptative, reste peu caractérisée dans ce contexte vaccinal. En vaccinant des macaques cynomolgus avec le virus de la vaccine modifiée Ankara, selon un schéma de primo-vaccination suivie d’un rappel homologue à deux mois, et en utilisant la cytométrie de masse couplée à des analyses bio-informatiques, nous avons caractérisé la réponse innée induite par chaque immunisation. Les réponses innées diffèrent entre primo-vaccination et rappel, avec induction par la primo-vaccination d’une modification phénotypique tardive des cellules innées, suggérant une meilleure capacité à répondre au rappel. De surcroît, la réduction à deux semaines du délai entre primo-vaccination et rappel abroge la mobilisation de ces cellules innées phénotypiquement modifiées et altère la qualité de la réponse humorale. En définitive, en plus de la réponse innée précoce, ce projet a mis en évidence l'induction par la primo-vaccination d'un vraisemblable entraînement inné tardif, un concept émergent traduisant la capacité de mémorisation des cellules innées via des modifications épigénétiques. Ce vraisemblable entraînement, non seulement des monocytes et cellules tueuses naturelles, mais aussi des cellules dendritiques et surprenamment des neutrophiles, est corrélé à la qualité de la mémoire immunitaire adaptative, de manière hautement dépendante du délai entre primo-vaccination et rappel. Ces résultats contribuent à ouvrir la voie vers l’optimisation rationnelle des futurs vaccins, via l'optimisation des calendriers vaccinaux et la valorisation de l'entraînement inné<br>Vaccination is one of the best achievements made in public health. However, designing vaccines against complex pathogens is currently challenging. The immune system is indeed uncompletely characterized, despite large amount of accumulated knowledges. A better understanding of vaccine-induced immunity is then required to optimize vaccine design. In particular, while most vaccines require several immunizations to induce a long-lasting adaptive immune memory, little is known on the impact of the delay beween the prime inducing a primary memory and the boost restimulating it to induce a secondary memory. Also, the innate immunity induced by each immunization and shaping the adaptative immunity is poorly characterized in this vaccine context.We studied the innate immune responses in cynomolgus macaques immunized with the modified vaccinia virus Ankara, following an homologous prime-boost vaccination at two months apart. We applied mass cytometry and bioinformatic analyses to characterize the innate response induced by each immunization. We showed that prime and boost vaccination triggered distinct innate responses. Actually, prime induced late phenotypic modifications of innate cells. These phenotypic changes suggest a stronger ability to react to the boost. Moreover, reducing the delay between prime and boost to two weeks impeded the mobilization of these phenotypically modified innate cells, and qualitatively altered humoral response.In conclusion, beyond the early innate responses, these results highlight the late induction by the prime of "likely trained" innate cells. This emerging concept corresponds to the ability of innate cells to display memory features based on epigenetic modifications. This "likely training" occured not only on monocytes and NK cells, but also on dendritic cells and strikingly on neutrophils. It was deeply connected with adaptive immune memory establishment, in a prime-boost delay dependant fashion. These findings contribute to pave the way towards to the rationale design of future vaccines, via vaccine schedule optimization and harnessment of innate training

Le développement de nouveaux vaccins administrables par voie orale est un objectif important. De tels vaccins permettraient l'induction de réponses immunitaires au niveau des muqueuses, qui constituent les principales voies d'entrée des agents pathogènes dans l'organisme, et seraient plus pratiques et plus sûrs pour l'Homme. La voie orale est aussi la voie la plus adaptée à la vaccination de la faune sauvage. À l'heure actuelle peu de vaccins sont administrés par voie orale et la plupart sont des vaccins vivants atténués, qui présentent certains risques. Les vaccins vectorisés non réplicatifs dérivés des adénovirus (VVA) représentent une alternative attractive. Ils sont plus sûrs et sont très efficaces lorsqu'ils sont administrés par voie parentérale. Néanmoins, leur efficacité est diminuée après administration par voie orale. Les objectifs de ce travail se divisent en trois points : 1. Comprendre les différentes étapes de mise en place de la réponse immunitaire après une administration par voie orale d'un VVA. 2. Explorer diérentes stratégies d'optimisation de ces vaccins. 3. Déterminer l'impact des adjuvants sur les étapes précoces de-mise en place de la réponse Immunitaire. Par des techniques utilisant la bioluminescence et la RT-PCRq, nous avons montré qu'après une administration par voie infra-gastrique d'un VVA chez deux souches de souris (C57BL/6 et BALB/c), l'expression du transgène était restreinte à l'intestin et limitée à certaines régions anatomiques. Nous avons également évalué l'action de plusieurs adjuvants sur les réponses immunitaires induites par les VVA par voie orale ainsi que leurs eets sur l'expression du transgène Chez les souris C57BL/6 et BALB/c<br>The development of new vaccines for oral delivery is a desirable objective. Such vaccines would permit induction of immune responses at mucosal surfaces, which constitute the principal portais of entry for pathogens. Moreover, the oral delivery route, as compared with parenteral routes, would be safer and more practical for human usage, and would facilitate the vaccination of wildlife. At present, bowever, few vaccines are administered by die oral route. Of these, most are live attenuated vaccines, which present a certain risk. Non-replicative vectored vaccines derived from adenoviruses (rAd) tepresent an attractive alternative, as they are much safer than live attenuated vaccines and are capable of eliciting robust immune responses when delivered by parenteral routes. Nevertheless, their efficacy is diminished upon oral administration. The objectives of this study were three-fold : 1. To understand the different steps leading to induction of an immune response after oral delivery of rAd, 2. To explore different strategies for optimisation of such vaccines, 3. To determine the impact of adjuvants on the early steps involved in the induction of the immune response. After intragastric delivery of rAd, we measured transgene activity by bioluminescence and RT-qPCR, and showed that transgene expression was confined to the intestine, and restricted to delimited anatomical zones. Moreover, the distribution of bioluminescence and antigen-encoding transcripts was dissimilar in C57B1/6 and BALB/c mice. Finally, after intragastric delivery of rAd, we showed that adjuvants affect transgene expression and the ensuing immune response in both C57BL/6 and BALB/c mice

<p>Tuberculosis (TB) remains one of the world’s most serious infectious diseases. It is estimated that a third of the world’s population is latently infected and 8 million new cases are recorded each year. Although BCG vaccination triggers protective immune responses in the neonates, it confers protection against only certain forms of childhood TB. Protection mediated by BCG, against pulmonary TB, is controversial as reported with variable efficacy ranging from 0-80%. In addition to the problems associated with the BCG vaccine, diagnosis of TB cannot be performed readily with the available tools. At present, an effective control of TB is highly dependent on the development of a new TB-vaccine as well as proper identification and treatment of individuals with active disease. Therefore, we particularly focused on identification of biomarker (s) of infection and the development of better vaccines, with special emphasis on the immune responses in the respiratory tract.</p><p>In the first study, we aimed to identify immune biomarker (s) of infection for better diagnosis of TB. Mice were infected with BCG administered i.n. or i.v., and the bacterial burden in the lungs, spleen and liver was examined. We measured IL-12, IFN-γ, TNF, soluble TNF receptors (sTNFR) and mycobacteria-specific antibodies in the broncho-alveolar lavage (BAL) and in serum in order to find immune correlates of infection. Results showed that sTNFR and mycobacteria-specific antibodies in BAL, but not in serum, might be useful in distinguishing active from latent infection or exposure to mycobacterial antigens.</p><p>In the second study, we investigated whether we could improve the currently used BCG vaccine. For this purpose, we tested a combination of neonatal vaccination protocol using BCG and posterior boosting with the protein heparin-binding hemagglutinin adhesion (HBHA). It has been described that immunization with native (n) HBHA but not recombinant (r) HBHA conferred protection against M. tuberculosis challenge in mice.</p><p>This protection was comparable to that afforded by the BCG vaccine. In order to improve the protective efficacy of the nHBHA vaccine we followed heterologous prime-boost strategy, comprising BCG vaccination at the neonatal age, followed by nHBHA boosting at the infant and adult ages. We also examined whether the rHBHA protein could boost BCG-mediated protective immunity. Cellular immune responses and protection as measured by control of bacterial growth in the lungs of the treated animals were followed. Our results showed an improved effect of BCG-priming on HBHA-immunization. The BCG/HBHA immunization protocol was more effective in induction of HBHA-specific immune responses, as well as in protection than when the animals received only BCG or HBHA alone. Importantly, our study revealed that nHBHA does not require co-administration with adjuvant provided that mice were primed with live BCG before boosting.</p><p>Finally, we hypothesized that in utero sensitization of the fetal immune system with nHBHA may improve nHBHA-specific immune responses after birth. The pregnant mother was immunized with nHBHA 1 week before delivery. After birth, the offspring received two doses (week 1 and week 4) of nHBHA formulated with cholera toxin. We examined HBHA-specific recall responses and protection after challenge with a high dose of BCG. We found that immune responses were improved by priming the pregnant mother, and that this also provided better protection than when the offspring received only BCG or HBHA neonatal vaccinations.</p>

Afin d'étudier la distribution des lymphocytes b-like et t-like dans les organes lymphoïdes rein, rate, thymus et le tissu lymphoïde associe à l'intestin du turbot et de la truite arc-en-ciel, des ac polyclonaux de lapin dirigés spécifiquement contre les immunoglobulines (ig) et les thymocytes de chaque espèce ont été produits. Un manque de spécificité des sérums antithymocytes n'a pas permis la détection des lymphocytes t-like. Seuls les anticorps anti-ig ont donc été utilisés dans cette étude menée sur des poissons, indemnes de toute maladie. Quels que soient les organes considérés, un plus grand nombre de cellules ig + est observé chez la truite. En ce qui concerne l'intestin des différences majeures sont notées entre les deux espèces : les cellules ig + sont présentes à la fois dans la lamina propria et la lamina epithelialis chez la truite alors qu'elles ne sont détectées que dans la lamina propria chez le turbot. Dans un second temps la mise au point de techniques de prélèvement du mucus intestinal, et d'isolement des cellules mononuclées présentes dans l'intestin postérieur du turbot, avec la possibilité d'obtenir séparément les cellules issues de la lamina epithelialis et de la lamina propria, a permis d'appréhender les mécanismes de déclenchement de la réponse immune au niveau intestinal a la suite de vaccinations orales (vo) contre la vibriose à vibrio anguillarum. L’absorption de l'ag et la réponse immune systémique ont également été étudiées. Différentes formulations vaccinales ont été testées : des vaccins constitués de corps bactériens entiers, gastroproteges (vog) ou non (voe), et des vaccins constitués d'ag solubles (vos). La voie intraperitoneale a toujours été testée en parallèle car utilisée comme référence. A la suite des vaccinations par vo, l'ag est detecte dans la lamina propria et dans les organes systémiques, où il est transporte jusqu'aux cellules immunocompetentes. En réponse a cette stimulation antigénique des cellules b-like et/ou des plasmocytes apparaissent dans l'intestin au niveau de la lamina propria, puis dans la rate et le rein. Contrairement aux cellules intestinales issues de la lamina propria et aux cellules rénales qui possèdent une aptitude a la phagocytose et sont capables de sécréter des ac, aucune activité comparable n'est mise en évidence sur les cellules issues de la lamina epithelialis. Les réponses observées dans l'intestin restent modérées. A la suite d'une vaccination par vo, une réponse immunitaire locale et générale existe donc et ceci malgré l'absence d'ac anti-vibrio anguillarum, dans le mucus intestinal et dans le sérum. L’injection parentérale, a montré quant à elle que l'ag atteint les organes systémiques mais parvient également dans la lamina propria, dans laquelle une lymphoprolifération de cellules b-like est observée. De plus des ac sont détectés dans le plasma et le mucus indiquant que le système immunitaire général et intestinal sont conjointement sollicites. Enfin, l'étude d'un transfert passif d'immunité, démontre l'existence d'une réaction immunitaire humorale consécutive aux vaccinations par ip mais aussi par vo avec l'ag soluble, sans que la protection induite soit réellement attribuable aux ig.

Le paludisme est la maladie parasitaire la plus mortelle pour les populations humaines. L'agent étiologique de l'infection, Plasmodium spp, est inoculé par des anophèles hématophages. Le candidat-vaccin MSP3, constitué d'une portion du domaine C-terminal de la protéine montra récemment qu'il était capable d'induire la protection contre la maladie chez des enfants africains. Récemment l'intérêt vaccinal de la MSP3 fut soulignée par la définition d'une famille multigénique de protéines partageant une même organisation structurale et ciblées par des anticorps cross réagissant avec l'ensemble des membres de la famille, offrant de nouvelles perspectives pour le développement de vaccins multiantigéniques. Dans cette optique, huit poly protéines (PP) de taille et de composition différentes combinant les antigènes des communs entre les membres de la famille furent synthétisées dans Escherichia coll. Les études d'immunogénicité vis-à-vis de la famille MSP3 dans trois souches de souris génétiquement distinctes (C57BL/6 ; BALB/c ; Swiss), montrèrent qu'elles induisaient assez précocement une réponse anticorps de haute amplitude et homogène indépendamment du fond génétique murin ou de l'immunogène. Cette réponse était constituée principalement de sous-classes d'IgG cytophiles murines (IgG2a, IgG2b, IgG2c). Une observation concordant avec celle obtenue en antigénicité, qui montrait que les anticorps naturellement présents et spécifiques. Des PP dans les sérums issus d'individus en zone d'endémie étaient principalement de nature cytophiles (IgGl, IgG3). Par ailleurs, l'analyse de la réactivité des anticorps induits révéla qu'ils étaient capables de reconnaitre les protéines natives du stade sanguin du parasite, et d'exercer en coopération avec des monocytes humains, une activité antiparasitaire in vitro en ADCL Si ces PP dérivées de MSP3 constituent une version améliorée du premier multigène générée à la suite de la découverte de la famille, la réactogénicité hautement similaire de chacune d'elles est un handicap à surmonter à travers d'autres outils de criblage afin de sélection 2 ou 3 à conduire en expérimentation de phase I chez l'Homme<br>MSP3 has been shown to induce protection against malaria in African children. The characterization of a family of Plasmodium falciparum merozoite surface protein 3 (MSP3) antigens sharing a similar structural organization, simultaneously expressed on the merozoite surface and targeted by a cross-reactive network of protective antibodies offers new perspectives for the development of subunit vaccines against malaria. Eight recombinant polyproteins containing carefully selected regions of this family covalently linked in different combinations were ail efficiently produced in Escherichia coll. The polyproteins consisted of one monovalent, one bivalent, one trivalent, two tetravalents, one hexavalent construct, and two tetravalents incorporating coiled-coil repeats regions from LSA3 and p27 vaccine candidates. All eight polyproteins induced a strong and homogeneous antibody response in mice of three distinct genotypes, with a dominance of cytophilic IgG subclasses, lasting up to six months after the last immunization. Vaccine-induced antibodies exerted a strong monocyte-mediated in vitro inhibition of P. Falciparum growth. Naturally acquired antibodies from individuals living in an endemic area of Senegal recognized the polyproteins with a reactivity mainly constituted of cytophilic IgG subclasses. Combination of genetically conserved and antigenically related MSP3 proteins provides promising subunit vaccine constructs, with improved features as compared to the first generation construct employed in clinical trials (MSP3-LSP). These multivalent MSP3 vaccine constructs expand the epitope display of MSP3 family proteins, and lead to the efficient induction of a wider range of antibody subclasses, even in genetically different mice. Le système immunitaire est capable de rejeter une tumeur, même si l'interaction entre système immun et tumeurs est et reste complexe. Contrôler à long terme la croissance de la tumeur est un challenge, probablement suite à divers mécanismes de tolérance centrale et périphérique. Différentes approches d'immunothérapie du cancer ont été et sont toujours développées. Nous avons, au laboratoire, investigué et démontré, dans le cadre de deux différents modèles tumoraux murins, les capacités thérapeutiques d’une nouvelle stratégie de vaccinations combinées associant des injections de cellules dendritiques (DC) et de cellules tumorales sécrétrices de GM-CSF.<p><p>Notre travail de thèse a consisté à poursuivre l'étude de cette stratégie de vaccins combinés en étudiant plus particulièrement le rôle des lymphocytes T régulateurs (Treg) dans l'efficacité thérapeutique des vaccinations. <p>Les Treg regroupent différentes populations cellulaires immunosuppressives dérivées du thymus qui jouent un rôle clé dans le maintien de la tolérance périphérique. Les Treg naturels dont le mécanisme de suppression principal nécessite un contact cellule-cellule, expriment de façon constitutive les molécules de surface CD4, CD25 et CTLA-4 mais le marqueur le plus spécifique est le facteur de transcription Foxp3 qui est indispensable à leur développement et à leur fonction suppressive. Dans un premier temps, nous avons donc caractérisé les Treg de rat pour l'expression de Foxp3 et la nature et spécificité antigénique de leur fonction suppressive. Nos résultats démontrent d’une part, une expression de Foxp3 restreinte aux LT CD25+ natifs et liée à une fonction suppressive s'exerçant par contact cellule-cellule et d’autre part, une spécificité antigénique non-restreinte de ces Treg. <p><p>Ensuite, nous avons utilisé ce modèle de vaccination associant des injections de DC et de cellules tumorales sécrétrices de GM-CSF pour analyser comparativement les réponses immunes induites chez les rats vaccinés guéris ou non-guéris et les rats contrôles non vaccinés et identifier les paramètres cruciaux conduisant à l'éradication de la tumeur. Nos résultats ont montré que la principale différence entre rats vaccinés guéris et non guéris ne réside pas dans l'induction de réponses cytotoxiques systémiques spécifiques de la tumeur. Par contre, la guérison est associée à la persistance d'une réponse systémique LT CD4+ TH1 ainsi qu'au recrutement important en intratumoral de LT CD8+ cytotoxiques lié à une faible proportion relative de Treg. <p><p>Comme dans la majorité des études publiées chez l'animal, les DC utilisées dans ces vaccins combinés ont été générées à partir de rats naïfs, par différenciation de précurseurs de la moëlle osseuse en présence de GM-CSF seul. Par analogie avec les vaccins DC administrés aux patients cancéreux, nous avons aussi dérivé des DC à partir de la moëlle osseuse de rats porteurs d’une tumeur (TUM+) et nous avons constaté que ces mêmes vaccins combinés montraient in vivo une efficacité thérapeutique nettement moins bonne que leurs équivalents naïfs. Nous avons établi que cette différence d’efficacité était liée à la présence de Treg fonctionnels dans les vaccins DC dérivés de rats TUM+. Puis, nous avons établi le lien entre la présence de Treg dans les vaccins DC TUM+ et leur moins bonne efficacité thérapeutique en montrant qu'un traitement in vivo des rats TUM+ au témozolomide (TMZ) avant de générer les vaccins DC résultait in vitro, en une moindre expression de Foxp3 et une fonction suppressive diminuée et in vivo, en une bien meilleure survie des rats vaccinés.<p><p>Enfin, dans le but d’une utilisation future en clinique, nous avons développé une approche simplifiée ‘tout in vivo’ de notre modèle de vaccinations combinées, en utilisant la tumeur localement irradiée in vivo comme source d’antigène pour des DC autologues injectées en péri-tumoral et un apport exogène de GM-CSF. Nous avons utilisé des adénovirus associés recombinants porteurs du gène du GM-CSF (AAV1-GM-CSF) pour transduire la tumeur in vivo. Ces injections intratumorales d’AAV1-GM-CSF ont montré de bons résultats en vaccinations combinées puisque 60% des rats ont pu être guéris d’une tumeur 9L pré-implantée. Nous avons ensuite expérimenté l’enrobage des AAV1-GM-CSF dans un polymère biocompatible et thermosensible, le poloxamère, avant de les injecter en intratumoral, sans observer de meilleur effet thérapeutique. Cependant, nous avons constaté que l'utilisation du poloxamère pour enrober du GM-CSF recombinant permettait d'améliorer nettement la survie des rats vaccinés par comparaison à l'utilisation de GM-CSF recombinant seul.<p><p>La stratégie de vaccinations combinées que nous avons largement explorée et validée chez le rat serait une alternative intéressante à développer en clinique, particulièrement en combinaison avec un traitement permettant d'éliminer les Treg à la fois dans les vaccins et chez les patients vaccinés.<p><br>Doctorat en Sciences biomédicales et pharmaceutiques<br>info:eu-repo/semantics/nonPublished

La réponse immune anti-tumorale naturelle est contrecarrée par différents mécanismes d'échappement de la tumeur au système immunitaire dont le recrutement de lymphocytes T régulateurs (LTreg) inhibant les réponses LT-CD8+ anti-tumorales notamment. La vaccination anti-tumorale cherche donc à induire des réponses LTCD8 tout en bloquant ces LTreg. J'ai analysé deux nouvelles stratégies d'inhibition de cette population par un antagoniste de CCR4 ou un antiangiogénique, le sunitinib. L'antagoniste de CCR4, associé à une vaccination, inhibe l'activité des LTreg et permet l'induction de LT-CD8 contre différents antigènes du soi. Chez des patients atteints de cancers du rein métastasiques, le sunitinib entraine une diminution significative des LTreg corrélée à la survie globale. Les LTreg représentent un biomarqueur potentiel de suivi de la réponse au traitement antiangiogénique. I analysed two novel molecules which inhibit LTreg: a CCR4 antagonist and an antiangiogenic molecule, Sunitinib. The CCR4 antagonist inhibits LTreg activity and is able to induce CD8+ T cells responses against various selfantigens when it was combined with vaccination. Sunitinib induces significannt LTreg diminution which is correlated with overall survival of patients with metastasic renal carcinoma treated by antiangiogenic therapy. The measurement of LTreg may have a predictive value on antiangiogenic clinical response. These two molecules may endogenous antitumor response and synergize with immunotherapy protocols

La pandémie COVID-19 a posé des défis majeurs aux systèmes de santé mondiaux, notamment en raison de la capacité du SARS-CoV-2 à acquérir des mutations. Cela a entraîné l'émergence séquentielle de variants préoccupants (VOCs de l’anglais Variants of concern) tels que Alpha, Beta, Delta, et désormais Omicron qui circule sous forme de sous-variants successifs (BA.1, JN.1, KP.3). Ces VOCs soulèvent des questions sur leur capacité à échapper à la réponse immunitaire induite par une infection et/ou une vaccination. Alors que les campagnes de vaccination se poursuivent, il est crucial d'évaluer comment différents schémas d'immunisation conférés par la vaccination homologue ou hétérologue ainsi que par la combinaison d’une infection et de vaccination (immunité hybride), impactent la réponse immunitaire contre les variants émergents. Grâce à une cohorte prospective de soignants, ce projet de thèse visait à étudier la capacité des VOCs à échapper à la réponse immunitaire, l'efficacité des différents schémas d'immunisation, et la durabilité des réponses immunitaires générées. Nos résultats montrent que les variants Alpha et Beta échappent aux anticorps neutralisants induits par l'immunisation contre la souche ancestrale, indépendamment du schéma d'immunisation. Cette capacité d’échappement immunitaire s'étend au-delà des premiers variants, car Delta et Omicron ont aussi présenté une baisse significative de leur capacité à être neutralisés par les anticorps produits lors d’une immunisation préalable. Ces résultats soulignent la nécessité cruciale de prendre en compte l'évasion immunitaire spécifique aux variants pour définir les seuils de protection et adapter les stratégies de vaccination. En plus de l’échappement immunitaire des VOCs, l'affaiblissement de la réponse immunitaire contribue également à une diminution de la protection contre le SARS-CoV-2 au fil du temps. Nos résultats montrent que le type d'immunisation, infection ou vaccination, impact significativement le pic et la demi-vie des anticorps dirigés contre le domaine de liaison au récepteur (RBD de l’anglais Receptor binding domain). Cela nous a mené à étudier la réponse immunitaire induite par différents schémas de vaccination 6 mois après l'immunisation. Nous avons montré que l'immunité hybride conduit à une réponse immunitaire plus robuste en comparaison à l'immunité induite par l'infection ou la vaccination seule. Cette réponse améliorée est observée à travers divers paramètres tels que la capacité de neutralisation et le pool de cellules B mémoires, et se traduit par une protection significativement améliorée contre le variant Delta. Ainsi, les individus avec une immunité hybride ont un risque d'infection par Delta réduit de 4,5 fois par rapport à ceux ayant reçu une vaccination homologue. Ces résultats soulignent l'importance de prendre en compte ces différences dans les recommandations vaccinales. Néanmoins, des infections dites «breakthrough», c'est-à-dire survenant malgré une vaccination antérieure, sont souvent observées pendant l’ère Omicron chez des individus vaccinés ou même présentant une immunité hybride. Notre étude sur la réponse humorale après une infection «breakthourgh» par BA.1 a révélé que, bien que l'immunité hybride empêche l’augmentation des taux d'IgG4 anti-S et maintient une activité ADCC élevée, elle limite la diversification du pool de cellules B mémoires spécifiques du RBD par rapport à l'immunité induite par la vaccination. Ces résultats indiquent que l’infection «breakthrough» induit des réponses immunitaires distinctes selon les schémas d'immunisation antérieurs, soulignant l’intérêt de considérer l’historique d’immunisation pour personnaliser les recommandations vaccinales. En somme, les résultats obtenus lors de ce travail de thèse soulignent la nécessité d’établir des recommandations vaccinales en fonction des immunisations antérieures pour répondre efficacement aux capacités d’échappement immunitaire des VOCs en circulation<br>The COVID-19 pandemic has presented significant challenges to global healthcare, largely due SARS-CoV-2’s ability to acquire new mutations. This has led to the sequential emergence of variants of concern (VOCs) such as Alpha, Beta, Delta, and now Omicron that exhibited different successive subvariants (notably BA.1, JN.1, and KP.3). These VOCs have raised concerns about their capacity to escape the immune response induced by infection and/or vaccination. As vaccination campaigns continue worldwide, it is crucial to evaluate how different immunization schemes, including homologous and heterologous vaccinations as well as infection combined with vaccination (hybrid immunity), impact the immune response against emerging variants. With a prospective cohort of healthcare workers, this PhD project aimed to investigate i) the capacity of viral variants to escape the immune response, ii) the effectiveness of different immunization schemes, and iii) the durability of the resulting immune responses. Our findings indicated that the Alpha and Beta variants are able to escape neutralizing antibodies induced by immunization against the ancestral strain, regardless of the immunization scheme. This capacity for immune evasion extends beyond these earlier variants, as both the Delta and Omicron variants also demonstrated significant resistance to neutralization by antibodies elicited through prior immunization. Such findings underscore the critical need to consider variant-specific immune escape when establishing protection thresholds and updating vaccination strategies. In addition to viral immune escape the waning of the immune response also contributes to a decreased protection against SARS-CoV-2. Our results show that the type of immunization, i.e. infection or vaccination, significantly influences the peak levels and half-life of antibodies targeting the receptor binding domain (RBD). This led us to investigate the immune response induced by different immunization schemes 6 months post-immunization. In particular, we showed that hybrid immunity leads to a more robust immune response 6 months post-immunization compared to immunity induced by either infection or vaccination alone. This enhanced response is observed across various immunological parameters, such as neutralization capacity and the pool of memory B cells, and translates into significantly improved protection against the Delta variant. Individuals with hybrid immunity experienced a 4.5-fold reduction in the risk of Delta infection compared to those with immunity induced solely by homologous vaccination. These findings highlight the importance of considering these differences when formulating vaccination recommendations. Nevertheless, breakthrough infections, i.e. infections occurring despite previous vaccination, are frequently reported during the Omicron era among individuals fully-vaccinated and those with hybrid immunity. Our investigation into the humoral immune response following BA.1 breakthrough infection revealed that while hybrid immunity prevents an increase in anti-S IgG4 levels and maintains a high antibody-dependent cellular cytotoxicity (ADCC) activity, it limits the diversification of the RBD-specific memory B cell pool compared to vaccination-induced immunity. Hence, our results indicate that BA.1 breakthrough infection elicits distinct immune responses that vary based on prior immunization schemes, which emphasizes the interest to consider immunization history with the aim to personalize vaccination recommendations. Overall, the results obtained throughout this PhD project emphasize the need to incorporate prior immunization history into ongoing adjustments of vaccination strategies and policies to effectively address the evolving immune escape capabilities of VOCs

Les contrôleurs du VIH (HICs) sont de rares patients qui contrôlent spontanément la réplication du VIH en absence de thérapie antirétrovirale. Ces patients sont caractérisés par un taux normal de cellules T CD4+, le maintien d’une charge virale indétectable (&lt;50 copies d’ARN viral/mL de plasma), et présentent un très faible risque de progression vers le SIDA. De nombreux travaux suggèrent que le contrôle de la réplication virale chez ces patients est dû à une réponse cellulaire antivirale particulièrement efficace. Notre équipe a récemment montré que les HICs inclus dans la Cohorte ANRS CO21 CODEX maintiennent des réponses T CD4+ très sensibles, associées à l'expression de TCRs ayant une forte avidité pour certains peptides Gag du VIH. Nous avons montré en particulier que les cellules T CD4+ de HICs répondent à de très faibles concentrations de l’épitope immunodominant Gag293. Dans une première étude, nous avons pu mettre en évidence que la réponse de haute avidité observée pour les cellules T CD4+ des HICs semble s'expliquer par une propriété intrinsèque de leur TCR. Pour identifier les déterminants moléculaires qui sous-tendent cette réponse de haute avidité, nous avons caractérisé le répertoire TCR des cellules T CD4+ spécifiques de l’épitope immunodominant Gag293. Les HICs présentent un répertoire TCR hautement biaisé, caractérisé par une expression préférentielle des chaînes variables TRAV24 et TRBV2, la présence de motifs conservés au sein des régions CDR3 de ces deux chaînes, et une prévalence élevée de clonotypes publics. Le clonotype public le plus représenté est capable de conférer une réponse spécifique à Gag293 très sensible et polyfonctionnelle aux cellules T CD4+ primaires, ainsi qu'une reconnaissance de multiples allèles HLA-DR. Nous cherchons à présent à déterminer si les candidats vaccins peuvent induire des réponses T CD4+ similaires à celles observées chez des patients HIC. Pour cela, nous avons analysé les réponses spécifiques de Gag293 chez des volontaires sains ayant reçu le candidat vaccin ADN ADVAX administré par soit par électroporation (EP) ou soit par injection intramusculaire (IM). La comparaison des réponses spécifiques de Gag293 dans des lignées T CD4+ primaires par test ELISpot IFNγ a montré que la sensibilité à l'antigène chez les volontaires vaccinés était proche de celle des patients contrôleurs et significativement plus élevée que celle mesurée chez les patients traités (p&lt;0.05). Par contre, les volontaires du groupe IM n'ont pas montré de réponse détectable contre Gag293. L'analyse du répertoire TCR des cellules spécifiques de Gag293 chez les volontaires vaccinés a révélé une amplification préférentielle de la chaîne variable TRBV2, chaîne qui prédomine également chez les patients HIC. Cependant, l'utilisation des chaînes variables TRAV est apparue plus diverse chez les sujets vaccinés, avec une amplification préférentielle des chaînes TRAV29 et TRAV24. L'analyse des clonotypes TCR amplifiés chez les vaccinés a mis en évidence un recoupement inattendu avec le répertoire des HIC, avec une fréquence élevée de motifs partagés (30%). Les clonotypes publiques TRAV24 et TRBV2 les plus fréquemment retrouvés dans le répertoire des Contrôleurs ont également été induits par la vaccination ADN EP, indiquant le potentiel pour une réponse de forte avidité. Par contre, le clonotype TRAV29 le plus exprimé, chez les vaccinés a induit des réponses de plus faible avidité, même si ces réponses étaient restreintes par de multiples allèles HLA-DR. L'ensemble de ces résultats suggère que les clonotypes TRAV24, plutôt que les clonotypes TRAV29, sont responsables de la réponse de forte avidité contre Gag293. En conclusion, un candidat vaccin ADN administré par EP possède la capacité d'induire des clonotypes TCR associés au contrôle du VIH. Cette étude suggère que les clonotypes TCR publics pourraient servir de nouveaux biomarqueurs permettant d'évaluer la qualité des réponses vaccinales induites contre le VIH<br>HIV controllers are rare patients who spontaneously control HIV replication to levels below 50 copies viral RNA/mL in the absence of antiretroviral therapy. Converging evidence indicate that HIV controllers mount a cellular antiviral response that is highly efficient at detecting and eliminating infected cells. In particular, we previously reported that HIV controllers from the ANRS CO21 CODEX cohort harbour a pool of memory CD4+ T cells able to respond to minimal amounts of viral antigens, due to the expression of TCRs with high avidity for immunodominant Gag epitopes. In a first study we show that the high functional avidity observed in Controller CD4+ T cells could be explained by the expression of high avidity T cell receptors (TCRs) which efficiently bound Gag peptide-loaded MHC class II tetramers. Repertoire analysis of CD4+ T cells specific for the immunodominant Gag293 epitope showed a highly skewed repertoire in HIV controllers, with a predominance of the TRAV24 and TRBV2 variable gene families, the presence of conserved motifs in both CDR3 regions, and a high prevalence of public clonotypes. When transferred to primary CD4+ T cells, the most frequent public TCR clonotypes could confer properties associated with an efficient T cell response, such as polyfunctional cytokine secretion, high affinity antigen recognition, and broad HLA-DR crossrestriction. To determine whether candidate HIV vaccines could induce the high sensitivity responses seen in controllers, we analysed Gag293-specific responses in healthy volunteers who received the ADVAX DNA vaccine administered by electroporation (EP) or intramuscular injection (IM). Comparison of Gag293-specific responses in primary CD4+ T cell lines via IFNγ ELISpot revealed that the median antigen sensitivity in vaccinees was close to that observed for controllers but significantly higher than that in treated patients (p &lt;0.05). In contrast, volunteers from the IM group did not show a detectable Gag293-specific response. TCR repertoire analysis of Gag293-specific CD4+ T cells from vaccinees revealed a preferential amplification of TCRβ family chain TRBV2, which also predominates in controllers. However, TRAV family gene usage appeared more diverse in vaccinees compared to controllers, with the preferential amplification of TRAV29 and TRAV24. Sequence analysis revealed an unexpected degree of overlap between the specific repertoires of vaccinees and HIV controllers, as evidenced by a high frequency of shared TCR motifs (&gt;30%). The most frequent TRAV24 and TRBV2 public clonotypes found in the controller TCR repertoire were also induced by EP DNA vaccination, suggesting the potential for high avidity responses. In contrast, an abundant TRAV29 clonotype from vaccinees induced Gag293-specific responses of lower avidity, even though these responses were broadly HLA-DR cross-restricted. Thus, our results suggest that TRAV24 rather than TRAV29 clonotypes may be responsible for the high avidity Gag293-response. In conclusion, DNA vaccination by electroporation has the potential to inducer TCR clonotypes associated with HIV control. Monitoring the amplification of public TCR clonotypes could provide a novel approach to evaluate the quality of HIV vaccine responses

Vγ9Vδ2 T cells are a subset of human T lymphocytes activated by phosphoantigens in a T cell receptor-dependent manner to fight microbial invaders or kill transformed cells. Phosphoantigens are low molecular weight nonpeptidic pyrophosphate containing metabolites produced both endogenously (upregulated in transformed cells) and by microbes. Vγ9Vδ2 T cells are the first T cells generated in the foetus and have programmed functions before encountering the post-partum environment.In this PhD thesis, the aim was to assess the origin of Vγ9Vδ2 T cells in early versus adult life and to evaluate their T cell receptor repertoire and effector potential in the neonatal and infant period. First, human Vγ9Vδ2 T cells were characterised coming from foetal blood and generated by the foetal thymus and then similarities and differences with adult blood Vγ9Vδ2 T cells were identified. The data showed that there is a post-natal thymic output of Vγ9Vδ2 T cells which are different from their foetal counterparts. This finding could help guide the development of cancer immunotherapy strategies aiming to improve the resistance and tenacity of Vγ9Vδ2 T cells which enter an exhaustion state after long encounter with the antigen.Furthermore, human Vγ9Vδ2 T cells were studied early after birth regarding their T cell receptor repertoire and function. At 10 weeks after birth, Vγ9Vδ2 T cells had expanded, and a big part of the Vγ9Vδ2 T cell repertoire was foetal-derived. Additionally, Vγ9Vδ2 T cells had undergone significant functional polarisation toward potent killer effector cells. The expansion and shift in effector functions were not influenced by neonatal BCG vaccination, highlighting the role of environmental exposure upon birth. The data gathered here highlight the unique properties of this innate-like lymphocyte population which can act as a first wave of protection in early life while conventional αβ T cells are not yet optimal. Depuis peu, l’immunothérapie a émergé comme une nouvelle stratégie chez les patients atteint de Cancer Bronchique Non à Petites Cellules (CBNPC), confirmant ainsi le rôle du système immunitaire dans cette maladie. Malgré ces nouveaux traitements (thérapies ciblées, immunothérapie), les taux de réponses restent faibles avec un impact modeste sur la survie globale. Des biomarqueurs sont donc nécessaire pour définir les populations cibles de ces traitements. Une des pistes explorées est le statut immunitaire des patients, en effet celui-ci a un impact pronostic et pourrait influencer la réponse aux traitements standards tels que la chimiothérapie, les thérapies ciblées et même l’immunothérapie. Parmi les cellules du système immunitaire, les cellules Natural Killer (NK) auraient un rôle effecteur dans le CBNPC. Il est maintenant clairement établit que les cellules NK favorisent la mise en place d’une immunité adaptative fonctionnelle et efficace. Ainsi une altération des fonctions NK pourrait être un mécanisme associé à l’échappement à l’immunité adaptative de la tumeur. Dans notre première étude, nous avons mis en évidence que les exosomes de cellules dendritiques stimulaient les cellules NK via NKp30, cette activité étant associée à un gain de survie sans progression chez des malades inopérables porteurs d’un CBNPC avancé. Notre second projet a révélé, pour la première fois, un rôle pronostic des transcrits de NCR3 (gène de NKp30) chez des patients naïfs de tout traitement. L’activation des cellules NK via NKp30 pourrait être une stratégie efficace d’immunomodulation chez les patients atteints de CBNPC avancé. Ces travaux confirment le rôle important des cellules NK dans le CBNPC avancé<br>Recently, immunotherapy has emerged as a new strategy in Non-Small Cell Lung Cancer (NSCLC) patients, confirming the key role of the immune system in this disease. Despite these new treatments (targeted therapies, immunotherapy), response rates remain low with a modest impact on overall survival. Biomarkers are needed to define the target population of these treatments. One of the options explored is the immune status; indeed the immune status of cancer patients has a prognosis impact and may influence the response to standard treatments such as chemotherapy, targeted therapies and even immunotherapy. Among the immune cells, Natural Killer cells (NK) have an effector role in NSCLC. It is now established that NK cells can promote a functional and efficient adaptive immunity. Therefore, an impaired NK functions could be a mechanism associated with the escape from adaptive immunity of the tumor. In our first study, we demonstrated that exosomes from dendritic cells stimulated NK cells through NKp30, this activity is being associated with improved survival in advanced NSCLC. Our second project has revealed, for the first time, a independent prognostic role of NCR3 transcript (NKp30 gene) for naïve advanced NSCLC. Activation of NK cells via NKp30 could be an effective strategy for immunomodulation in advanced NSCLC patients. These studies confirm a major role of NK cells in advanced NSCLC

Un des défis majeurs de l’immunologie antitumorale repose sur l’induction efficace et prolongée de laphase effectrice de la réponse immune antitumorale. Une meilleure compréhension des mécanismesimpliqués dans la potentialisation de l’activité antitumorale des effecteurs immunitaires, en particulier lesCTL infiltrant la tumeur représente donc un enjeu considérable dans le développement de nouvellesapproches d’immunothérapie visant à induire une réponse immunitaire spécifique efficace contre latumeur. Dans ce contexte, nous nous sommes particulièrement intéressés à étudier le rôle de CD5 dans lecontrôle de la progression tumorale, en particulier dans la potentialisation de l’activité T cytotoxique desCTL infiltrant le mélanome B16 in vivo. Nos résultats ont montré un ralentissement significatif de lacroissance tumorale chez des souris déficientes pour l’expression de CD5 comparées à leurs équivalentessauvages. Le contrôle de la progression de la tumeur chez les souris CD5-/- ne semble pas corréler avec unrecrutement plus important de lymphocytes T, mais avec une efficacité accrue de leur réactivité vis-à-visdu mélanome B16. Nous avons montré aussi que cette réponse est transitoire et qu’un échappementtumoral au système immunitaire ait lieu à un stade plus tardif de la progression de la tumeur. Cetéchappement tumoral semble être associé à une augmentation de la mort par AICD des TIL CD8+/CD5-par rapport aux TIL CD8+/CD5+ et ce par induction de FasL à la surface des TIL. La modulation de la voieFas/FasL in vivo avec un adénovirus Fas-Fc protège les lymphocytes T CD8+/CD5- infiltrant la tumeur dela mort par apoptose et empêche ainsi l’échappement tumoral. De plus, l’analyse du répertoirelymphocytaire T spécifique du mélanome B16 dans les souris CD5-/- suggère que le contrôle de laprogression de la tumeur serait lié à une meilleure prolifération de lymphocytes T CD5-/- spécifiques insitu. A better understanding of the mechanisms involved in potentiatingthe antitumor activity of immune effectors, particularly the CTL infiltrating the tumor represents aconsiderable challenge in developing new approaches to immunotherapy. In this context we areparticularly interested in studying the role of CD5 in the control of tumor progression, particularly inpotentiating the cytotoxic activity of CTLs infiltrating B16 melanoma. Our results showed a significantdelay in tumor growth in CD5-/- mice as compared with wild type mice. The control of tumor growth inCD5-/- mice does not seem to correlate with a higher recruitment of T cells, but with increased cytotoxicityof infiltrating T cells against B16 cells. We have also shown that this response is transient and that thetumor escape from immune system takes place at a later stage of tumor progression. This tumor escapeappears to be associated with increased death by AICD of TIL CD8+ from CD5-/- mice as compared withTIL CD8+ from wild type mice wich correlated with the induction of FasL on the surface of TIL. In vivomodulation of Fas/FasL with an adenovirus AdFas-Fc protects tumor infiltrating T CD8+/CD5- fromapoptosis and thereby prevents the tumor escape. In addition, analysis of specific T lymphocyte repertoirein CD5-/- mice suggests that the control of tumor progression could be linked to a greater in situproliferation of specific CD5- T lymphocytes. Our results also show that immunization of CD5-/- miceusing different melanoma associated antigens contributes to the potentiation of antitumor immuneresponse. All these results highlight a particular interest in targeting of CD5 to improve currentapproaches to tumor immunotherapy

La vaccination est l’une des interventions humaines ayant le plus contribué à la diminution de la mortalité reliée aux maladies infectieuses. Les succès enregistrés par les vaccins traditionnels, composés de pathogènes atténués ou inactivés, ont été obtenus principalement grâce à la stimulation d’anticorps neutralisants. Or, ce corrélat de protection immunologique ne convient plus aux nouveaux pathogènes émergeants tels que le virus de l’hépatite C ou le virus de l’immunodéficience humaine, qui requiert également la stimulation d’une réponse de type cellulaire forte. Une solution intéressante à ce problème est l'ajout d'un adjuvant au vaccin, une méthode reconnue pour augmenter l'ampleur et la diversité de la réponse immunitaire contre le vaccin. Cependant, peu d’adjuvants sont reconnus pour leur capacité à générer des réponses cellulaires et aucun n’est disponible en vaccination humaine en Amérique du nord. A cet effet, les pseudo-particules virales (PPVs) du virus de la mosaïque de la papaye (PapMV) ont précédemment démontré un certain potentiel adjuvant au niveau de la réponse cellulaire. L’objectif de ma thèse de doctorat a donc été d’étudier le potentiel adjuvant des PPVs du PapMV sur des vaccins commerciaux, ainsi que sur des cibles protéiques complètes. Dans un premier temps, nous avons évalué l’effet adjuvant des PPVs de PapMV sur le vaccin inactivé utilisé pour lutter contre le virus influenza. Ensuite, nous avons évalué l’effet de cet adjuvant, ainsi que d’une version de haute avidité, sur la nucléoprotéine (NP) du virus influenza, une cible intéressante dans le développement d’un vaccin universel contre cet important pathogène. Finalement, nous avons évalué la possibilité de développer un vaccin candidat contre le virus de l’hépatite C (VHC) basé uniquement en tout ou en partie sur la protéine de coque du VHC, protéine la plus conservée du virus, en combinaison avec notre adjuvant. Globalement, l’adjuvant s’est avéré efficace à augmenter l’immunogénicité de l’ensemble des cibles vaccinales utilisées et a même contribué à augmenter l’effet protecteur généré par le vaccin inactivé et la protéine NP contre le virus de l’influenza. Son utilité pour le développement d’un vaccin protecteur contre l’hépatite C, comprenant uniquement la protéine de coque, reste à déterminer.<br>Vaccination is one of the human interventions that having the most contributed to the decrease of mortality attributed to infectious diseases. The successes recorded by the traditional vaccines, composed of attenuated or inactivated pathogens, were principally obtained by the induction of neutralizing antibodies. Now, this correlate of immunologic protection is no longer suitable for new emergent pathogens such as the hepatitis C virus or the human immunodeficiency virus that equally require the stimulation of a strong T-cell response. An interesting solution to this problem is the addition of adjuvants to vaccines, a method known to increase the breadth and diversity of the immune response against the vaccine. However, few adjuvants are known for their ability to generate cellular responses and no adjuvant of this type is available for human vaccination in North America. To this end, the virus-like particles (VLP) of Papaya mosaic virus (PapMV) have previously demonstrated potential adjuvant effect towards the cellular responses. The objective of my Ph.D. thesis was to study the adjuvant potential of the PapMV VLPs on commercial vaccines and on complete protein targets. First, we evaluated the effect of PapMV VLPs adjuvant on the inactivated vaccine used to fight the influenza virus. Secondly, we evaluated the adjuvant effect of this adjuvant, and the high avidity version, on the nucleoprotein (NP) of influenza virus, a promising target to develop a universal vaccine against this important pathogen. Finally, we evaluated the possibility of developing a candidate vaccine against the hepatitis C virus (HCV) based in whole or in part on the core protein of HCV, the most conserved protein of the virus, in combination with our adjuvant. Overall, the adjuvant was effective of increasing the immunogenicity of all vaccine targets used and even helped to increase the protective effect generated by the inactivated influenza vaccine and the NP protein. Its usefulness for the development of a protective vaccine against hepatitis C based solely on the core protein, remains to be determined.

La vaccination muqueuse, notamment celle administrée par voie nasale, est considérée comme la méthode optimale permettant de protéger les sites muqueux de l'infection par des pathogènes. Cependant, le manque d'adjuvants muqueux spécifiques ainsi qu'une prise en compte insuffisante du système immun nasal limite le développement des vaccins muqueux. P1, un domaine conservé de gp41, la glycoprotéine d'enveloppe du VIH-1, a été récemment utilisé par le laboratoire d'acceuil pour développer un vaccin prophylactique muqueux après immunisation combinée par voie nasale et intra-musculaire. Ce vaccin s'est montré efficace lors études pré-cliniques et clinique de Phase I chez l'Homme, grâce à sa capacité d'induire des IgA muqueux contre P1 bloquant la pénétration muqueuse du VIH-1 par transcytose et de stimuler la production d'IgG spécifiques de gp41 induisant la lyse des cellules infectées par le VIH-1 par cytotoxicité cellulaire médiée par anticorps (ADCC). Dans le travail présenté ici, nous avons caractérisé les mécanismes immuns induits par ce vaccin basé sur P1 au niveau de la muqueuse nasale humaine. Tout d'abord, nous avons démontré que P1 initie une réponse immune en induisant la sécrétion de la cytokine TH2, la Thymic Stromal LymphoPoietin (TSLP), par les cellules épithéliales nasales. TSLP est connyu pour ses propriétés d'adjuvant muqueux puissant et son récepteur, le TSLP-R, joue un rôle déterminant dans la réponse muqueuse à IgA. Nous avons montré que P1 induit l'expression de TSLP via l'interaction de P1 avec le galactosyl céramide, un récepteur du VIH-1 permettant l'entrée muqueuse du virus. De plus, nous avons identifié la voie de signalisation Calcineurin/NFATet le microRNA- comme partenaire décisifs dans la régulation de la production de TSLP induite par P1. Dans un second temps, nous avons montré que le peptide P1 module la réponse innée en activant les cellules dendritiques (DCs). Cette stimulation par P1 induit l'augmentation de l'expression des molécules de co-stimulation par les DCs. La sécrétion de l'IL-6 et de l'IL-10 par les DCs est aussi augmentée par P1 alors que celle de l'interféron gamma, IFN-', est réduite, démontrant ainsi que les DCs activée par P1 se polarisent en une phénotype facilitant une réponse Th2 et IgA. De plus, l'IL-8 et les chimiokines CCL20 et CCL22 sont secrétées indiquant que les DCs ont acquis la capacité de recruter des cellules immunes dans la muqueuse pour initier une réponse muqueuse adaptative. La métalloprotéinase MMP-9 permettant la dégradation de la matrice extracellulaire et facilitant la migration des cellules hors de la muqueuse, est aussi produite. Une boucle positive autocrine de TSLP est observée, puisque P1 induit la sécrétion par les DCs de TSLP en conséquence l'augmentation de l'expression du TSLP-R par les DCs induite par P1. Finalement, P1 active la prolifération des lymphocytes TCD4+ médiée par les DCs. En conclusion, nous avons démontré que P1 est un peptide multifonctionnel avec un grand potentiel dans le développement de vaccin, non seulement comme antigène vaccinal candidat mais aussi comme potentiel adjuvant qui pourrait être combinés à d'autres vaccins muqueux<br>Mucosal vaccination, especially intranasal administrated ones, has been considered to be ideal for protection from pathogens invading through mucosal sites. However, the lack of specific adjuvant and insufficient acknowledgement of nasal immune system limits the development of vaccine. P1, a conserved region of gp41 envelope glycoprotein, was recently developed into a prophylactic HIV-1 vaccine immunized via both the intramuscular and intranasal routes. It showed high efficiency in pre-clinical and phase I clinical trial due to induction of P1 specific mucosal IgA with transcytosis blocking activity and IgG inducing antibody dependent cell cytotoxicity. In this study, we characterized the immunological mechanism underneath P1-vaccine in nasal mucosa. Firstly, we demonstrated that P1 initiate immune responses by inducing nasal epithelial cells to secret the Th2 cytokine Thymic Stromal LymphoPoietin (TSLP). TSLP has been reported to be a strong mucosal adjuvant, and its receptor TSLP-R plays a critical role in IgA response. We showed that P1 induce TSLP expression via the interaction with galactosyl ceramide, the receptor of HIV-1 mucosal entry. Furthermore, we identified Calcineurin/NFAT signaling pathway and microRNA-4485 as important players in the regulation of TSLP production induced by P1. Secondly, we showed that P1 modulates innate immune responses by activate dendritic cells (DCs). P1 stimulation results in enhanced expression of costimulatory molecules on DCs. Furthermore, the secretion of IL-6, IL-10 were increased, while IFN-γ was reduced, indicating that P1 activated DCs polarize into a Th2 and IgA prone phenotype. In addition, IL-8, CCL20, CCL22 were produced indicating a capacity at recruiting immune cells to mucosal surface for initiation of an adaptive immune response. MMP-9 was also produced allowing degradation of the extracellular matrix and facilitating the migration of immune cells out of the mucosa. Stricingly, a TSLP autocrine loop was observed as P1 induced DCs to secret TSLP and meanwhile, enhanced DC expression of TSLP-R. Finally, P1 activated DCs enhanced the proliferation of CD4+ T cells. In conclusion, we demonstrated that P1 is a multi-functional protein with a great interest for vaccine development, not only as an antigen for vaccine candidate, but also as a potential adjuvant that can be combined to other mucosal vaccines

Le cancer est la seconde cause de mortalité dans le monde et les cancers de localisation muqueuse (poumon, estomac, colorectal, du col de l’utérus, …) représentent la première cause de mortalité due au cancer dans le monde. La majorité des vaccins contre les cancers muqueux n’ont à ce jour, pas montré de résultats cliniques significatifs. Au cours de ce travail, nous avons développé une immunothérapie efficace basée sur la sous-unité B non toxique de la toxine de Shiga et montré pour la première fois dans le domaine de la cancérologie que la localisation de l’immunisation était cruciale pour induire des réponses immunitaires anti-tumorales. En effet, dans un modèle préclinique, une immunisation systémique intramusculaire n’a pas permis d’induire de protection thérapeutique efficace contre le développement de tumeurs muqueuses de la langue, alors que la voie d’immunisation intranasale a induit une réponse clinique complète. Nous avons identifié les lymphocytes T CD8+ comme les cellules nécessaires à cette protection et plus précisément la population de lymphocytes T résidents mémoires (Trm). Ces Trm présentent le phénotype classique CD103+ mais expriment également l’intégrine CD49a qui joue un rôle dans la migration/rétention au sein des tumeurs mais également dans la survie à long terme des Trm. Par ailleurs nous avons montré que les cellules dendritiques muqueuses pulmonaires permettaient d’induire ce phénotype CD49a sur les lymphocytes T CD8+ alors que les cellules dendritiques de la rate non. Notre travail montre que l’aspect quantitatif de ces Trm joue un rôle dans la protection anti-tumorale, en effet nous avons pu pour la première fois moduler in vivo le nombre de Trm en traitant les souris par un anticorps anti-TGF-β. La diminution du nombre des Trm est corrélée à la diminution de la protection anti-tumorale. Les patients atteints de cancers des voies aérodigestives supérieures sont majoritairement traités par radiothérapie. Dans l’optique d’essais cliniques à court terme, nous avons montré que la radiothérapie localisée associée à notre immunothérapie permet une protection plus efficace que le traitement seul de l’un ou de l’autre notamment en provoquant un remodelage du microenvironnement tumoral associé à une normalisation vasculaire. Nos résultats ouvrent de nouvelles perspectives dans le développement d’immunothérapies thérapeutiques efficaces contre les cancers muqueux et pourront mener rapidement à des essais cliniques<br>Cancer is the second mortality cause worldwide while mucosal cancers (lung, stomac, …) is the first mortality cause from. The majority of cancer vaccines against mucosal tumors have not given rise yet to significant clinical results. In this work we developed a strong immunotherapy based on the nontoxic subunit B from shiga toxin and showed for the first time that the localization of the immunization is crucial to induce potent and effective anti-tumoral responses. In a preclinical model a systemic immunization failed to induce a therapeutical protection against mucosal tumor challenge while intranasal immunization completely succeed. We identified a CD8 T lymphocyte population as a required cells in this protection and more precisely the T resident memory (Trm) cells. This Trm showed the classical CD103 phenotype as well as the CD49a which can play a specific role in the retention or the migration of this cells in the tumor tissue and might play a role in the survival. We also demonstrate that dendritic cells from the mucosal parenchyma was required to induce the CD49a expression on CD8 T cells while dendritic cells from the spleen was not. Our work shows that the Trm number as an impact in the anti-tumoral protection. We were able to reduce the Trm number in vivo using an anti-TGF-β antibody. This number diminution was correlated with a less efficient anti-tumoral protection. Patients with head and neck cancers are treated with radiotherapy. In this situation we showed that the combination of radiotherapy and our immunotherapy was associated with a better protection than radiotherapy alone or immunotherapy alone thanks to a vascular normalization. These results might rapidly lead to clinical trials and might open new ways to work with immunotherapies

This thesis aimed to the identification of immune biomarkers of mycobacterial infection for better diagnosis of tuberculosis (TB) and also focused on new vaccination strategies with a particular emphasis on the immune responses in the respiratory tract using murine models. Since the lung is the natural habitat for the M. tuberculosis, we reasoned that immune responses detected locally in the lungs would be good correlates of infection (Paper I). Likewise, immune responses induced in the respiratory tract following immunization would be more effective against mycobacterial infection. We showed that cytokines (IL-12, TNF, and IFN-γ) and cytokine receptors (sTNFR1 and sTNFR2) together with specific antibodies in the respiratory tract correlated better with the bacterial burden in the organs. In Paper II, we investigated the role of the BCG vaccination as a priming vaccine in a heterologous prime-boost immunization protocol. The results showed that the neonatal BCG vaccination primed the immune system for a relevant antigen and showed a generalized adjuvant effect. Using this immunization protocol, protective immune responses in the lungs were generated independently of the route used for the booster immunization. In Paper III, We showed that exposure to mycobacterial antigens during the gestational period led to antigen transportation from the mother to the fetus and this resulted in an early priming of the fetal immune system. Immunization with the same antigen during the postnatal life increased antigen-specific recall IFN-γ responses and protection against infection. We examined the role of innate immunity for the induction of acquired immune responses upon immunization with mycobacterial antigens using TLR2 deficient mice (Paper IV). Our data indicated that suboptimal innate immune responses in the TLR2-/- mice might compromise the induction of acquired immune responses. Overall, the current findings suggested that a better understanding of the mucosal immunity would be useful for the improvement of diagnostic procedures and the development of efficient vaccines against TB.<br>At the time of the doctoral defense, the following paper was unpublished and had a status as follows: Paper 4: Manuscript

La sous unité B de la toxine de Shiga (STxB) est un vecteur vaccinal ciblant les cellules dendritiques. Des réponses lymphocytaires T-CD4 et CD8 ainsi que la production d'anticorps ont été observées après vaccination de souris avec des protéines chimériques associant STxB couplée à différents antigènes. STxB ne favorisant pas la maturation des cellules dendritiques, nous avons évalué l'efficacité vaccinale de STxB mélangée à différents adjuvants. Ainsi, STxB couplée à différents antigènes et associée à l'aGalCer, glycolipide activant les cellules NKT, permet d'augmenter la fréquence de LT-CD8 et de réduire de façon drastique les doses d'antigènes. Cette combinaison vaccinale a également permis de rompre une tolérance à des antigènes du soi et de protéger contre le développement d'une infection virale. Par ailleurs, nous avons montré que la voie d'immunisation influait sur le type de réponses immunitaires (humorale muqueuse, cellulaire systémique) observées après l'utilisation de STxB<br>The Shiga toxin subunit B (STxB) is a vaccinal vector targeting dendritic cells. CD4+ and CD8+ T cells responses as well as antibody production were observed after vaccination of mice with chimeric proteins composed of STxB coupled with different antigens. STxB doesn't favour maturation of dendritic cells, thus we assessed the STxB efficiency as vector in combination with different adjuvants. STxB coupled with different antigens and mixed with aGalCer, a glycolipide activating NKT cells, resulted in an increase CD8+ T cells frequency and it also allowed the dramaticaly reduction of antigen doses. This vaccine also permitted to break tolerance to self-antigens and to protect against the development of viral infection. In addition, we have showed that the route of immunization had an influence on the type of immune response (mucosal humoral response jind cellular^ systemic response) observed after the use of STxB

Cette thèse avait pour objectif de fournir des données sur l’efficacité et la tolérance des vaccins dans un contexte d’immunodépression chronique, dans diverses situations cliniques et pour différents antigènes vaccinaux. Un premier essai randomisé chez 95 patients infectés par le VIH a ainsi montré que 15% des patients avaient perdu leurs anticorps protecteurs quatre ans après vaccination contre l��hépatite A, quel que soit le nombre de doses de vaccin reçues dans le schéma de primo-vaccination (2 ou 3 doses). Puis une revue bibliographique systématique et une méta-analyse étendues aux autres antigènes du calendrier vaccinal et à d’autres catégories d’immunodépression (hémodialysés et transplantés d’organe solide) ont confirmé que la durée de persistance des anticorps après vaccination était plus courte chez ces sujets qu’en population générale. La question de la tolérance vaccinale a été abordée au travers de deux études observationnelles : la première n’a pas mis en évidence d’impact significatif des vaccinations sur l’incidence des poussées chez les patients atteint de vascularite systémique, et la seconde a montré que les patients sous corticoïdes rapportaient plus fréquemment des réactions locales modérées ou sévères après vaccination contre la fièvre jaune que les témoins (RR = 8,0 (IC 95% : 1,4 ; 45,9)). Ces différents travaux montrent que la tolérance et l’immunogénicité des vaccins présente des particularités chez les personnes immunodéprimées qui nécessitent d’être prises en compte par la rédaction de recommandations spécifiques en termes de schémas et de fréquence des rappels vaccinaux<br>Safety, immunogenicity and durability of responses after vaccination are not well documented in the context of immune suppression. We first present the results of a clinical trial in 95 patients living with HIV comparing the immunogenicity of a reinforced 3-dose vaccine scheme against hepatitis A to the standard 2-dose regimen. Maintenance of seroprotection was not improved in the 3-dose arm since 15% of responders had lost protective antibodies by year 4 after immunization in both arms (p=1). We next extended our work to other categories of immune suppression by performing a systematic review of published works providing original long term (i. E. More than 6 months) immunogenicity data with licensed vaccines in three common conditions increasingly met in clinical practice: HIV infection, end-stage renal disease and solid organ transplantation. A meta-analysis of the estimations confirmed that duration of seroprotection provided by most licensed vaccines was shorter in immunocompromised patients than in otherwise healthy persons. Safety issues have been addressed through two observational studies: the first focused on the impact of immunizations on the course of systemic vasculitis and the second showed that patients on corticosteroids reported moderate/severe local reactions more frequently than controls (Relative Risk (95%CI) = 8. 0 (1. 4 to 45. 9)) to the live-attenuated yellow fever vaccine. Safety and immunogenicity of vaccines in persons with impaired immunity are not comparable to that observed in healthy individuals, calling for specific recommendations in these patients

En vue de l'exploration de l'immunité muqueuse chez des sujets sains vaccinés par un vaccin SIDA, une étude portant sur une population de sujets infectés par le VIH-1 et au stade CDC II de la maladie a été menée. Les IgG totales, les IgA totales et les IgG et IgA anti-gp 120 MN et anti-p24 LAI ont été testés dans les salives totale, parotidienne et sous-linguale, les sécrétions cervico-vaginales (CVS) le liquide séminal et les prélèvements rectaux. L'albumine sérique était mesurée afin de calculer les coefficients relatifs d'excrétion (RCE). 44 volontaires infectés par le VIH-1 et 52 volontaires sains ont été inclus dans cette étude. Les concentrations en IgG totales dans tous les fluides sont plus élevées chez les sujets séropositifs que dans le groupe contrôlé. Le calcul des RCE a permis de mettre en évidence 3 catégories de prélèvements muqueux : des sécrétions sans production locale d'immunoglobulines comme le sperme, des sécrétions produisant des IgA et dont les IgG ont une origine transudative comme les fluides salivaires et rectaux et enfin les CVS pour lesquelles des IgA et des IgG sont produites localement. Pour tous les fluides la réponse anti-VIH-1 était portée principalement par l'isotype IgG, les titres les plus élevés étant observés dans les CVS. La réponse des IgA anti-VIH-1 est plus faible dans les sécrétions par rapport au sérum. Dans un deuxième temps, la nature de la réponse IgG a été explorée plus avant et les sous-classes d' IgG ont été testées. Le calcul des RCE indique une origine transudative des sous-classes, excepté dans les CVS pour lesquelles on détecte des IgG 1, IgG 2 et IgG 4 produites localement. Seules les CVS montrent une production locale en IgG 1 anti-gp120 MN. Les techniques Elisa développées ont été utilisées pour évaluer la réponse muqueuse anti-VIH dans deux essais vaccinaux des sujets vaccinés par voie parentérale et les niveaux de réponse ont pu être comparés à ceux obtenus chez les sujets séropositifs.

Bovine tuberculosis (TB), caused by Mycobacterium bovis, is increasing in incidence in the United Kingdom and detailed knowledge of host-pathogen interactions in the natural host is essential to facilitate disease control. Vaccination of neonatal calves with Bacille Calmette Guerin (BCG) induces a significant level of protection from infection with M. bovis. Since neonatal vaccination of humans with BCG induces activation of natural killer (NK) cells, and neonatal calves have high circulating numbers of these cells, it is proposed that NK cells are important in the response to BCG. Furthermore, NK cells play an important role in shaping adaptive immune responses through interactions with dendritic cells (DCs). The overall hypothesis of this project was that the enhanced efficacy of BCG in neonates is due to the increased number of NK cells, which through interactions with DCs can polarise Th1-type CD4+ and CD8+ T cell responses, both of which are involved in protection against M. bovis infection. Initially, the frequency and phenotype of NK cells across the blood, afferent lymph and the lymph nodes in steady-state conditions were compared. CD2- NK cells were the principal subset of NK cells migrating from the skin to the draining lymph node and were highly activated in afferent lymph and lymph nodes, compared with peripheral blood. It was also demonstrated that CD2- NK cells were the main subset of NK cells egressing from the lymph node via the efferent lymphatic vessel to return to circulation. Since many vaccines including BCG are delivered subcutaneously, NK cell responses in the blood and the skin draining afferent lymphatic vessel, lymph nodes and efferent lymphatic vessel were determined after BCG vaccination. Alterations in the frequency and receptor repertoire were evident following vaccination, supporting a role for NK cells during BCG vaccination of neonatal calves. To investigate the interactions of NK cells and BCG-infected DCs, in vitro co-cultures were established. CD2- NK cells were preferentially activated following culture with BCG-infected DCs and secreted high levels of IFN-γ. Overall, this thesis provides novel evidence that NK cells may re-circulate in steady-state conditions, play a role in BCG vaccination of neonatal calves, and that through interactions with BCG-infected DCs, may be involved in driving protective Th1-type adaptive immune responses.

DNA vaccination is based on in vivo delivery of plasmids encoding an antigen, leading to antigen synthesis and specific immunity. This technology is still in its infancy but has advantages over conventional vaccination, such as the stimulation of all arms of the immune response, i.e., humoral immunity, T-helper (17h) cells and cytotoxic T lymphocytes (CTLs). We studied DNA vaccination against human carcinoembryonic antigen (CEA) in mice. Our hypothesis was that codelivery of cytokine- and CEA-encoding plasmids could regulate responses, and allow polarization of Th responses to either type 1 (Th1) or type 2 (Th2). We found that intramuscular injection of the CEA plasmid alone induced antibodies, Th1 cells and CTLs reactive to CEA. These mice had increased immunity against transplanted syngeneic CEA+ stably transfected tumor cell lines, but always developed lethal tumors. Coinjection of the CEA plasmid with a vector encoding either IL-12 or interferon gamma (IFNgamma) markedly enhanced IgG2a production (IFNgamma-dependent), IFNgamma secretion by spleen cells (a Th1 cytokine) and CTL-mediated tumor cell lysis, in a CEA-specific way. Moreover, resistance to a tumor challenge was greatly improved, such that up to 80% of mice survived tumor free. In contrast, coinjection of CEA and IL-4 genes increased CEA-specific IgG1 levels (IL-4-dependent) and IL-4 secretion by lymphocytes (a Th2 cytokine), but decreased both CTL activity and tumor resistance. Thus, we could readily enhance or polarize immunity. The IL-12 cDNA had the strongest adjuvant effect, which was only observed when it was injected at the same site as the CEA gene. To analyze effector components, we studied IL-12-plasmid-enhanced DNA vaccination in gene knockout mice, lacking either CD3, CD4, CD8. IFNgamma, perforin or Fas ligand (FasL). Les lymphocytes T CD8+ spécifiques de l'antigène sont impliqués dans la réponse immunitaire adaptative et jouent un rôle essentiel dans la protection de l'hôte contre l'infection par des pathogènes intracellulaires. Cette protection de longue durée dépend de la génération de réponses lymphocytaires T CD8+ mémoires, hautement fonctionnelles en termes de fréquence et de fonctionnalité, après réinfection.Après présentation de l'antigène, une cellule T CD8 naïve subit une forte expansion clonale, générant une population hétérogène de cellules activées qui est dominée, au sommet de l'expansion, par des effecteurs CD8 de courte durée (SLEC). Cette expansion est suivie d'une phase de contraction massive par apoptose. Quelques cellules survivent à cette phase de contraction et finissent par se différencier en cellules mémoire hautement compétentes. Les processus par lesquels et le moment où se différencient les précurseurs de mémoire (MPECs) restent largement inconnus, tout comme les étapes ultérieures de leur maturation en cellules mémoire pleinement fonctionnelles. Ces précurseurs précoces, qui ont un phénotype effecteur, se développent et migrent temporairement à la jonction des zones T et B, où ils interagissent avec les Tfh puis perdent leur expression CXCR5.Cette interaction avec les Tfh, considérés jusqu'à présent comme des auxiliaires exclusifs des cellules B, est nécessaire pour que les MPECs CD8+ deviennent des cellules mémoire compétentes sensibles à l'IL-21, capables de générer des réponses effectrices secondaires efficaces.Cette étude dévoile les premières étapes cruciales dans la génération de la mémoire CD8+, identifie CXCR5 comme le premier marqueur connu des MPECs CD8+, révèle l’implication fondamentale des Tfh dans le CD4 help et indique une coordination possible, via les Tfh, entre les voies de différentiation mémoire CD8+ et B. Ces résultats peuvent avoir des implications pour la conception du vaccin et de l'immunothérapie<br>Antigen-specific CD8 T cells are involved in the adaptive immune response and play a critical role in protecting the host from infection by intracellular pathogens. This long-lasting protection depends on the generation of memory CD8+ T cell responses, which are highly functional in terms of frequency and functionality, after secondary infection.Following antigen activation, a naive CD8 T cell undergoes strong clonal expansion, generating a heterogeneous population of activated cells that is dominated, at the peak of expansion, by short-lived CD8 effectors (SLECs). This expansion is followed by a phase of drastic contraction through massive apoptosis. A few cells survive this contraction phase and eventually become highly competent memory cells. Precisely when and how these memory precursors (MPECs) are generated is largely unknown, and so are the subsequent steps of their maturation into fully functional memory cells. Help signals from CD4+ T cells are clearly required throughout the MPEC maturation process.Our team has previously shown that FoxP3+ regulatory CD4+ T cells (Tregs) favor MPECs maturation by limiting exposure to IL-2 and by providing inhibitory signals, but this is probably only one facet of the complex and multifaceted help provided by CD4+ T cells to MPEC, and Tregs act on pre-existing MPECs.B-cell memory and CD8+ T cell memory share some common features, such as the expression of the transcription factor Bcl-6. Tfh are major producers of the cytokine IL-21. The mechanisms by which Tfh induces Bcl-6 in B-cells need to be clarified, they might include IL-21 and CD40-CD40L.In this PhD project, we have investigated the potential role of Tfh on the initiation of CD8 memory differentiation, in transgenic mice models, allowing transient and selective depletion of Tfh cells, infected by recombinant Listeria monocytogenes-OVA.We have shown that as early as 2 days after infection, very early memory precursors can be identified by their expression of the chemokine receptor CXCR5. These early precursors, which have an effector phenotype, expand and temporarily migrate to the junction of T-cell and B-cell zones, where they interact with follicular CD4 T cells (Tfh) then lose their CXCR5 expression.Remarkably, this interaction with Tfh, hitherto considered as exclusive B-cell helpers, is required for memory precursors to become competent memory cells responsive to IL-21 and capable of mounting efficient cytotoxic secondary effector responses.This study thus unveils critical early steps in the generation of CD8 memory, identifies CXCR5 as the earliest known marker of CD8 memory precursors, reveals a major helper role for Tfh, and points to possible coordination, through Tfh, between the pathways of CD8 and B-cell memory generation. These findings may have implications for vaccine and immunotherapy design

Human subjects maintain long-term immunological memory against infective organisms but the mechanism is unclear. CD4+ T helper memory cells (Thmem) are pivotal in controlling humoral and cellular responses, therefore their longevity and response to vaccination are critical for maintenance of protective immunity. To probe the dynamics of the Thmem response to antigenic challenge, we investigated subjects following a booster injection with tetanus toxoid (TT). Expansion of TT-specific Thmem cells, and cytokine production, showed complex kinetics. Strikingly, parallel expansion and cytokine production occurred in pre-existing Thmem cells specific for two other common antigens, Purified Protein Derivative of tuberculin (PPD), and Candida albicans (C.alb). Bystander expansion occurred in Thmem but not in Thnaive cells. Antibody production against TT peaked ~2 weeks post-vaccination and gradually declined. However, pre-existing antibody against the other antigens did not change. It appears that, although all Thmem cells are readily stimulated to expand, antibody responses are controlled by antigen availability. These human findings which relate to the maintenance of memory and have consequences for assessments of specific T-cell responses to vaccination, have been further investigated in a mouse model. A transgenic model (OT-II) where CD4+ T cells express a TCR specific for an ovalbumin peptide (peptide 323-339, OVAp) was used first to ask the question as to whether naïve or antigen-activated T cells were influenced in a bystander manner during a secondary immune response directed against a protein antigen that was unrelated to their cognate one. For this, carboxyfluorescein diacetate succunimidyl ester (CFSE)-labeled OT-II cells were adoptively transferred, either as naïve or 4 following in vitro activation with OVAp, into C57/BL6 wild type recipient mice which were immune to TT. Recipient mice were then challenged with TT antigen and susceptibility of OT-II cells to bystander activation and proliferation was tested. Naïve T cells were found not to be influenced, but antigen-activated cells were responsive and underwent further activation and bystander proliferation, with accompanying phenotypic changes. Interestingly bystander proliferation appeared to be proportional to the strength of TT-specific cellular immune response. The second question was whether the bystander influence on activated T cells was also evident during a primary immune response to TT. To address this question, antigen-activated OT-II cells and control naïve cells were adoptively transferred into wild type naïve recipient mice and their activation and proliferation was assessed after challenge with TT. In this case no bystander activation or proliferation of OT-II cells was observed. These results underline the susceptibility to bystander activation and proliferation as a unique feature of antigen-activated OT-II cells as opposed to naïve OT-II cells. They mirror those obtained in our study on human subjects and add formal proof of bystander proliferation occurring in vivo. Furthermore this well defined mouse model paves the way for further investigations aimed at addressing the mechanisms responsible for the observed phenomenon.

<p>Vaccination with DNA encoding the encephalitogenic autoantigen myelin oligodendrocyte glycoprotein (MOG), pMOG91-108, induce a protective immunity against experimental autoimmune encephalomyelitis (EAE), an animal model of human multiple sclerosis. By injection of a DNA vaccine that contains a DNA region encoding short interfering RNA specific for IFNβ (pMOG-IFNβ) the protective effect of the DNA vaccination is totally inhibited. This demonstrates that IFN-β is directly involved in the protective mechanism against EAE.</p><p>The objective of this project was to study how molecules involved in the inflammatory process in EAE are regulated by suppressive DNA vaccination. mRNA expression of IL-1β, TGF β, IL-23p40 and Axl receptor tyrosine kinas did not show any significant differences between the groups vaccinated with these DNA vaccines. IL-6 and IFNγ mRNA expression after MOG stimulation in rats treated with pCI, a control vaccine was significantly higher compared to the group vaccinated with vaccine containing pMOG-IFNβ. IL-17 m RNA expression after MOG stimulation in pCl-treated rats was significantly higher compared to the group vaccinated with vaccine containing pMOG-91-108. Of these results the mRNA expression of IL-17 and IL-6 were of interest for the project.</p><p>The immune system normally protects the body against infections and T-cells have an important role in this defence system. In MS and EAE, the immune system attacks the myelin and this process is caused by a dysregulation of the T-cells. IL-17-producing Th17 cells mediate EAE. Naïve CD4 T-cells in the presence of IL-6 and TGFβ are differentiated to Th17 cells instead of differentiating into T-helper or regulatory T-cells. These IL-17-producing T-cells are highly pathogenic and essential for the development of EAE. The results showed that pMOG IFNβ vaccine had an effect at the immune response, which resulted in an inhibition of the IL-6 production and that vaccination with pMOG91-108 impairs differentiation of IL-17-producing T-cells.</p>

Because of the well established epidemiologic and biologic interactions between human immunodeficiency virus (HIV) transmission and genital ulcer disease, measures to control chancroid could have a significant impact on the epidemic of HIV. I developed an IgG and IgM antibody serologic enzyme linked immunosorbent assay (ELISA) using pooled sera from clinically and microbiologically proven cases of chancroid as positive controls, and pooled sera from normal individuals without a history of sexually transmitted diseases as negative controls. Cross reactivity was minimized by absorption of serum samples with a sorbent prepared from three other Haemophilus species. Performance of the ELISA was enhanced in the period of early convalescence from acute primary chancroid and was not diminished in the presence of HIV coinfection, reflecting the tempo of the natural serologic response to and acute infection. I also developed an inhibition ELISA to determine antigenicity of potential vaccine candidates in human H. ducreyi infection, using lipooligosaccharide (LOS) as the test antigen. In a panel of 10 sera from cases of primary natural H. ducreyi infection reactive to H. ducreyi 35000 soluble antigen, only 4 samples were identified with reaction to H. ducreyi 35000 LOS. Inhibition ELISA confirmed that pre-adsorption with LOS substantially diminished reactivity of these sera to LOS but not to soluble antigen. Using the ELISA to detect immunogenicity by measuring the serologic response to vaccine candidates in rabbits, we further tested the feasibility of three bacterial antigen preparations to induce protective immunity against infection and disease. LOS carbohydrate and a pilus preparation were purified from H. ducreyi 35000 and used in a booster immunization procedure. Virulence was assayed by intra-epithelial challenge and measurement of disease for homologous strain 35000 or the virulent clinical isolate RO-34. LOS and the pilus preparation both induced humoral responses to the corresponding antigen, but the carbohydrate preparation did not. Immunization with LOS or carbohydrate did not modify virulence of infection with H. ducreyi 35000. Immunization with the strain 35000 pilus preparation significantly reduced the severity of disease, and the duration of infection and disease compared with controls. I conducted passive immunization experiments, and characterized the inflammatory infiltrate of chancroidal lesions. Naive rabbits were passively immunized with 24 or 48 mg of purified polyclonal IgG intravenously and 24 hours after infusion, challenged with the homologous strain 35000. No significant difference in disease resulting from infection with the homologous strain was observed. I then comparatively evaluated the serial immunohistology of lesions produced by infectious challenge with the homologous strain in sham-immunized or rabbits immunized with the pilus preparation. Flow cytometric analysis of rabbit peripheral blood leucocytes with CD5 and CD4 rabbit lymphocyte markers prior to infectious challenge revealed two T lymphocyte populations: CD5+CD4+ and CD5+CD4$-$. Pilus preparation vaccinee lesions showed significant quantitative acceleration and increase in T lymphocyte infiltration, and similar early increased recruitment of the CD4+ T lymphocyte subset preceding early lesion sterilization and early healing without ulceration. Master of Science<br>Department of Clinical Science<br>Elizabeth Davis<br>Background: While there is a commercially-available vaccine for Streptococcus equi subsp. equi licensed for the intranasal route of administration, some equine practitioners are administering this vaccine orally despite a lack of evidence for its efficacy by this route of administration. Objectives: To compare systemic and local immune responses following intranasal or oral administration of the USDA-approved, live-attenuated Streptococcus equi subspecies equi vaccine (Pinnacle IN®, Zoetis, Florham Park, New Jersey). Study Design: Experimental, randomized clinical trial Methods: Eight healthy horses with low Streptococcus equi M protein (SeM) titers (<1:1600) were randomly assigned to an intranasal or oral two-vaccine series. SeM-specific serum immunoglobulins G (IgG) and A (IgA) and nasal secretion IgA were assessed using a commercially-available ELISA (Equine Diagnostic Solutions, LLC, Lexington, Kentucky) and a novel magnetic microsphere assay utilizing fluorescence. A general linear mixed models approach was used for statistical data analysis. Results: As expected, intranasal vaccinates showed substantial increases in both serum SeM-specific IgG and IgA levels post-vaccination (P=0.0006 and P=0.007, respectively). Oral vaccinates showed an increase in serum SeM-specific IgG post-vaccination (P=0.0150), though only one-third the magnitude of intranasal vaccinates. Oral vaccinates showed no evidence of change in SeM-specific IgA post-vaccination (P=0.15). Main Limitations: Changes in mucosal antibody responses were not identified in this study which may be related to small change in antibody response, timing of sample collection, or method of nasal secretion collection. Conclusions: Results indicate that intranasal or oral vaccine administration resulted in increased serum SeM-specific IgG, though the magnitude of response differed between routes.

Tick-borne encephalitis virus is the cause of much morbidity and sometimes a fatal infection. A vaccine based on formaldehyde inactivated virus is currently the only available way of preventing disease. This vaccine gives a high rate of seroconversion but there are reports of vaccination breakthrough, even in people who have demonstrated a neutralizing antibody response. The T cell response to inactivated TBE vaccine is largely unknown, but could be of importance for the effect of the vaccine. This study characterizes aspects of the T cell response by investigating the expression of two transcription factors, T-bet and GATA-3 with RT-PCR. T-bet is expressed in CD4+ T cells of the Th1 type, while GATA-3 is expressed in CD4+ T cells of the Th2 type. Our data show that vaccination with inactivated TBE vaccine leads to increase in expression of the T-bet gene when cells of vaccinated subjects are cultured with TBE virus. In contrast, the expression of GATA-3 remains unaffected by vaccination. Thus, this study suggests that the inactivated TBE vaccine leads to a Th1-like immune response in humans.

Els virus de la influença tipus A (VIA) són patògens zoonòtics que poden infectar un ampli nombre d’hostes incloent-hi les aus, els porcs i els homes, entre altres. Anualment es documenten milions d’infeccions en humans causades per virus de la influença estacionals. Les pandemies causades pel virus influença també tenen una elevada repercussió pel que fa a la sanitat i l’economia. Tot i que determinats subtipus de VIA s’adapten millor en espècies d’aus que en humans, hi ha hagut casos d’infeccions en humans per virus de la influença de tipus aviars. La susceptibilitat dels porcs per infectar-se amb virus de la influença tant d’origen aviar com humà és també important pel que fa a la salut pública. El genoma del virus influença és segmentat in consta de vuit molècules de ARN de cadena senzilla i sentit negatiu que codifiquen per 11 o 12 proteïnes. Per tant, si una cèl·lula s’infecta simultàniament per dos VIA diferents, pot succeir un reagrupament amb la conseqüent generació d’una nova soca de virus. A més, mutacions a les glicoproteïnes de superfícies (sobretot a l’hemaglutinina, HA) són les responsables de l’elevada variabilitat de VIA. Tot i que les vacunes front a les epidèmies estacionals són eficaces, no produeixen resposta immunològica front una amplia varietat de VIA. És a dir, les vacunes estacionals només protegeixen front a les soques virals circulants durant una determinada estació. Aquest fet, junt amb el risc de possibles pandèmies, han fet encara més important i urgent el desenvolupament d’una vacuna universal capaç de produir immunitat front a múltiples subtipus virals. En la present tesis s’ha estudiat la resposta immunitària front a la infecció i vacunació del VIA en el context de VIA d’alta patogenicitat (vIAAP) A/H5N1 i A/H7N1 i el virus pandèmic A/H1N1 (pH1N1). El treball s’ha dividit en tres parts i cada part s’ha subdividit en capítols. Part I (capítols 1 i 2), conté la introducció general i els objectius de la tesi doctoral. L’objectiu d’aquesta primera part és donar una visió global i introduir informació per entendre (i) la infecció pel virus de la influença, (ii) la resposta immunològica provocada després de la infecció per VIA i (iii) un breu resum de les vacunes actuals front a influença. A continuació, s’exposen els objectius a aconseguir. Part II, és el cos de la tesis i conté els quatre treballs (del capítol 3 al 6) duts a terme durant els quatre anys que ha durat el programa de doctorat. Tots els capítols presentats han estat publicats o sotmesos a publicació en revistes indexades internacionals. Per tant, cada estudi manté l’estructura estàndard de: resum, introducció específica, materials i mètodes, resultats i breu discussió. Estudiar el paper dels determinants virals i caracteritzar la infecció pel VIA en diversos hostes pot ser de gran interès a l’hora de dissenyar vacunes òptimes. S’ha descrit la proteïna NS1 com a un dels principals determinants de virulència en mamífers, però no s’ha estudiat gaire el paper d’aquesta en aus. En el capítol 3 es va avaluar la implicació de la proteïna NS1 en la patogenicitat viral en pollets. Es van infectar pollets amb vIAAP H7N1 que contenien el segment NS de vIAAP H5N1. Les manifestacions patològiques i la resposta immunològica conseqüència de la infecció amb cada un dels virus van ser avaluades. També és molt important el paper de la immunitat prèvia durant un brot perquè pot ser determinant de la mort o supervivència de l’animal. En el capítol 4 es van exposar pollets a un virus H7N2 de baixa patogenicitat (vIABP) i a continuació es van infectar amb un vIAAP H7N1. Posteriorment es van infectar amb un vIAAP H5N1. Els animals que havien estat infectats prèviament amb vIABP quedaven protegits a la posterior infecció letal amb el vIAAP H7N1. No obstant, la resposta immunitària produïda no era suficient per a protegir els pollets front a la infecció amb un virus heterosubtípic (vIAAP H5N1). La presència o absència d’anticossos inhibitoris front a H7- i H5- correlacionaven amb la presència o absència de protecció, respectivament. Conèixer els programes de vacunació actuals i la seva eficàcia és útil per a planificar i dissenyar futures estratègies de vacunació. Les aus aquàtiques són el reservori dels VIA; per tant, són extremadament importants pel que fa a l’ecologia del virus. Aprofitant els programes de vacunació es va testar el sèrum de diverses espècies d’aus de zoològics i centres de recuperació d’Espanya (capítol 5). Els sèrums es van utilitzar per a l’avaluació de la resposta humoral deguda a la vacuna. El principal objectiu del treball era determinar l’eficàcia de vacunes disponibles (inactivades en suspensió oliosa) en diverses espècies d’aus i comparar la variabilitat inter- i intra-espècie. Finalment, i tenint en compte el potencial risc del VIA, els esforços es van focalitzar en desenvolupar una vacuna capaç de protegir a un ampli nombre de subtipus de VIA. La pandèmia de 2009 amb el virus H1N1 (pH1N1) és un clar exemple que els porcs poden actuar com a “coctelera” i generar nous virus. En el capítol 6 es van immunitzar porcs amb pèptids derivats de l’HA i a continuació es van infectar amb el virus pH1N1. Tot i que els pèptids-HA produïen una molt bona resposta humoral i cel·lular, no es va detectar activitat neutralitzant i només es va obtenir un efecte parcial en l’eliminació del virus. Part III (Capítols 7 i 8), és la secció on es discuteixen les implicacions dels resultats obtinguts en els diferents estudis i on s’enumeren les conclusions principals. En una secció a part, s’han inclòs totes les referències bibliogràfiques utilitzades per a l’elaboració de la tesi. S’ha inclòs també un apèndix per afegir informació addicional.<br>Influenza A viruses (IAV) are zoonotic pathogens that can replicate in a wide range of hosts, including birds, pigs and humans, among others. Millions of human infections caused by seasonal influenza virus are reported annually. Influenza pandemics have also a significant health and economic repercussions. Although certain subtypes of IAV are better selected in avian species than in humans, there are reports that evidence cases of human infections with avian influenza viruses (AIV). The susceptibility of pigs to infection with influenza viruses of both avian and human origins is also important for public health. The genome of influenza virus is segmented and consists of eight single-stranded negative-sense ribonucleic acid (RNA) molecules encoding 11 or 12 proteins. Thus, if a single cell is simultaneously infected by two distinct influenza viruses, a reassortment can occur resulting in the generation of a novel virus strain. Moreover, mutations in the surface glycoproteins (mainly in the hemagglutinin, HA) are the responsible of the high variability of IAV. Influenza vaccines against seasonal epidemics, although have good efficacy do not elicit immune response against a wide variety of IAV. Thus, seasonal vaccines only confer protection against the circulating viral strains. This, together with the risk of potential pandemics, has highlighted the importance of developing a universal vaccine able to elicit heterosubtypic immunity against multiple viral subtypes. In this thesis the immune response to IAV infection and vaccination was evaluated in the light of the risk of highly pathogenic AIV (HPAIV) A/H5N1 and A/H7N1, and the pandemic IAV A/H1N1. The work is divided into three parts and each one is further divided into chapters. Part I (chapters 1 and 2) contains the general introduction and the objectives of the thesis. The aim of this first part is to give a global overview and to introduce information to understand (i) the influenza infection, (ii) the immune responses elicited after IAV infection and (iii) a brief summary of current vaccines against influenza. Afterwards, the initial objectives to be achieved are exposed. Part II is the body of the thesis and it contains four studies (from chapter 3 to 6) developed during the four-year period comprising the PhD program. All the chapters are published or submitted to publish in international peer-reviewed journals. Thus, each study contains an abstract, a specific introduction, the materials and methods section, the obtained results and a discussion. To study the role of IAV determinants and to characterize the influenza infection in different hosts could be of great importance to direct the efforts to the formulation of more efficient vaccines. The non structural 1 (NS1) protein is known to be a major determinant of virulence in mammals but little is known about its role in avian species. In chapter 3, the involvement of NS1 in viral pathogenicity was evaluated in chickens. Birds were challenged with two reassortant AIV carrying the NS-segment of H5N1 HPAIV in the genetic background of an H7N1 HPAIV. The pathological manifestations, together with the immunological outcome were evaluated. The role of pre-existing immunity during an outbreak is also important and can determine whether the animals succumbed to infection or not. In chapter 4, chickens pre-exposed to H7N2 low pathogenic AIV (LPAIV) were challenged with H7N1 HPAIV and subsequently infected with H5N1 HPAIV. Pre-exposed animals were protected against the lethal H7N1-challenge whereas naïve animals succumbed. However, pre-existing immunity did not provide protection against HA-heterosubtypic virus (H5N1 HPAIV). The presence or absence of H7- and H5-inhibitory antibodies correlate with the protection (or lack of it) afforded. The control of current vaccination programs and their efficacy is useful to plan and design better vaccines. It is well known that wildfowl are the reservoirs of IAV; thus they are extremely important concerning the ecology of the virus. Sera from several avian species from Spanish zoos and wildlife centers were collected during two successive vaccination programs and were tested to evaluate the vaccine-elicited humoral response (chapter 5). The main objective of this work was to determine the efficacy of current vaccines (inactivated water-in-oil) in several avian species and to compare the differences inter- and intra-specie. Finally, and taking into account the potential risk that IAV represent to our society, the efforts were focused on developing a broadly protective influenza vaccine. The 2009 human H1N1 pandemic (pH1N1) is a clear example that pigs can act as a vehicle for mixing and generating new assortments of viruses. In chapter 6 pigs were immunized with HA-derived peptides and subsequently infected with pH1N1 virus. Although the HA-peptides induced broad humoral and cellular responses no neutralization activity was detected and only a partial effect on virus clearance was observed. Part III (chapters 7 and 8) is where the implications of all the findings from the studies are discussed and the major conclusions are listed. A list of all the references used to develop the thesis is listed after the three parts, in an independent section. An appendix section is also included to give further information.

Infiltrins, or pathogen-secreted host nucleus infiltrating proteins, are potential new targets for the development of more efficient vaccines against helminthic parasites. The archetypal infiltrin is IL-4 inducing principle from S. mansoni eggs (IPSE/alpha-1, also known as SmIPSE), a glycoprotein secreted by Schistosoma mansoni eggs, and characterised by the simultaneous presence of a classical secretory signal and a nuclear localisation signal. Within minutes following uptake by mammalian host cells, IP SE/alpha-1 translocates to their nucleus and binds to Deoxyribonucleic acid. This suggests that infiltrins, by acting as transcription factors, might play a central role in controlling the host-parasite relationship at the molecular level. Together with their secretory status, this role makes infiltrins interesting targets for vaccination. In this study, similar properties were demonstrated for nuclear localisation signal of ShIPSE03 (125-SKRRRKY-131) located between amino acid position 125 and 131, that plays a necessary and sufficient role in the process of transferring ShIPSE03 and heterologous GFP proteins into the nuclei of host cells. Similarly, a combination of online bioinformatics tools was used to predict the putative nuclear localisation signal motif of Smk5 (256-ELKRRVE-262) from S. mansoni eggs, and FhGST-si (202-LKKRAKT-208) and FhH2A (35-IHRHLKT-41) from Fasciola hepatica. To verify the predicted NLSs, putative infiltrins were used to generate a series of truncated constructs fused with Aequorea coerulescens green fluorescent protein-1 (AcGFP1), which were transfected into mammalian cells. Nuclear localisation of fluorescence confirmed the existence of a single, signal at the C-terminal in ShIPSE03, FhGST-si, and Smk5, and at the N-terminal in FhH2A. The predicted NLS motifs in ShIPSE03 (125-SKRRRKY-131), Smk5 (256-ELKRRVE-262), FhGST-si (202-LKKRAKT-208) and FhH2A (35-IHRHLKT-41) inserted into Tetra-enhanced green fluorescent protein (EGFP), but not corresponding alanine NLS mutants, redirected the encoded ~100 kDa protein entirely to the nucleus. Use of an IPSE-specific monoclonal antibody or an anti-His antibody showed that wild-type recombinant ShIPSE03, Smk5, FhGST-si and FhH2A added exogenously to HTB-9 or Huh7 cells, fully translocated to the nucleus, whereas the alanine NLS mutant remained in the cytoplasm. Overall, the existence of infiltrins in S. haematobium and F. hepatica suggests that infiltrins may represent a more general regulatory principle operating within parasitic trematodes. In terms of the function of IPSE/alpha-1, quantitative real-time polymerase chain reaction data indicated that an increase in Alanine-Transaminase (ALT) activity measured after 72 hours could be a result of increased gene expression after IPSE/alpha-1 nuclear translocation, rather than a true reflection of hepatotoxicity. According to Transepithelial Electrical Resistance (TEER) measurements in electrically tight Caco-2 cells grown in transwell inserts, wild-type IPSE/alpha-1 may play an important role in the down- regulation of intestinal epithelial cell tight junction integrity, might encourage apoptotic mediators and inflammatory responses in intestinal epithelial cells, and impair the intestinal tight junction barrier by causing it to dysfunction. In addition, wild-type IPSE/alpha-1 was able to activate humanised basophil reporter cell lines, such as the RS-ATL8 and NFAT DsRed cell lines.

Influenza is a preventable infectious disease, against which vaccination is the primary means of protection. Health care workers (HCW) are among the most vulnerable to the illness and are likely to be sources of infection transmission while caring for patients. Circumstantial evidence suggests higher rates of vaccination coverage by HCW will coincide with a lower incidence of influenza transmission, yet a gap remains in the literature regarding governing health agencies' (i.e., licensing boards, medical and nursing associations) influence on the influenza vaccination practices of their constituents. Moreover, discrepancies exist between governing health agencies' and the National Vaccine Advisory Committee's recommendations on mandatory influenza vaccination for HCW. The main purpose of this quantitative cross-sectional study was to explore the relationship between influenza vaccination uptake by HCW and guidance from governing health agencies to vaccinate. The health belief model and social cognitive theory were used to identify the most influential determinant for HCW to vaccinate against influenza. The sample consisted of 388 HCW who provided direct patient care at the same hospital. Data were analyzed using Fisher's exact test. Study findings suggest that a workplace mandate for influenza vaccination has an influence on HCW uptake of the vaccine and that governing agencies' lack of uniformity on the matter has minimal impact on their constituents' beliefs and behavior. It is recommended that a universal policy be adopted for health agencies' implementation of an influenza vaccine mandate, which could lead to positive social change by supporting preventive self-care practices, minimizing spread of the disease to workers and patients, and maintaining workplace productivity.
