Abstract:
The objective of this study was to validate the shelf-life of marinated and frozen chicken tenderloins. Treatments were randomly assigned to the age of the tenderloins post-harvest, days aged (DA): DA4, DA5, DA6, DA7, and DA8. Microbial analyses were used to analyze the growth of aerobic, psychotropic, and lactobacilli bacteria to assess the shelf-life of bulk-packaged chicken tenderloins. Tenderloins were sampled fresh, then vacuum tumbled in a marinade. After marination, the tenderloins were sampled with the remaining tenderloins packaged and frozen (−25 °C). After freezing the chicken tenderloins were slacked in a refrigerated cooler (2.2 °C) for up to 132 h (h) and sampled at 36 h, then every 24 h following. After marination, each treatment significantly (p < 0.05) decreased in aerobic and psychotropic counts except DA4. During slacking, no treatment crossed the threshold of 106 CFU/mL (Log 6) set for this study. Though none crossed the threshold, treatments DA4, DA5, and DA6 had significant (p < 0.05) increases in aerobic bacteria after 7 days of age. The psychotropic bacteria continuously grew at each sampling period, with DA4 and DA5 surpassing the other treatments (p < 0.05) at 108 h and 132 h reaching 105 CFU/mL. Every treatment remained below the spoilage threshold, suggesting that this method of storage is suitable for chicken tenderloin shelf-life.

Abstract:
Device-assisted therapies (DAT) benefit people with Parkinsons Disease (PwP) but many referrals for DAT are unsuitable or too late, and a screening tool to aid in identifying candidates would be helpful. This study aimed to produce such a screening tool by building a classifier that models specialist identification of suitable DAT candidates. To our knowledge, this is the first objective decision tool for managing DAT referral. Subjects were randomly assigned to either a construction set (n = 112, to train, develop, cross validate, and then evaluate the classifier’s performance) or to a test set (n = 60 to test the fully specified classifier), resulting in a sensitivity and specificity of 89% and 86.6%, respectively. The classifier’s performance was then assessed in PwP who underwent deep brain stimulation (n = 31), were managed in a non-specialist clinic (n = 81) or in PwP in the first five years from diagnosis (n = 22). The classifier identified 87%, 92%, and 100% of the candidates referred for DAT in each of the above clinical settings, respectively. Furthermore, the classifier score changed appropriately when therapeutic intervention resolved troublesome fluctuations or dyskinesia that would otherwise have required DAT. This study suggests that information from objective measurement could improve timely referral for DAT.

SUMMARYIn this study, we assessed the sensitivity, specificity, and diagnostic accuracy of a previously developed direct agglutination test (DAT) using a freeze-dried antigen derived fromLeishmania infantumpromastigotes and composed in a prototype kit for visceral leishmaniasis (VL) diagnosis, named DAT-LPC. To evaluate DAT-LPC reproducibility, the kit was used to analyse 207 serum samples from VL patients and 80 serum samples from patients with other parasitic infections or healthy subjects in four laboratories from different public health institutions in Brazil. DAT-LPC showed sensitivity between 96·2 and 99·5% (P= 0·14), specificity ranging from 96·2 to 97·5% (P= 0·95), and diagnostic accuracy ranging from 96·5 to 99% (P= 0·34). The inter-laboratory reproducibility of qualitative results was classified as excellent (κindex: 0·94–0·97). The reproducibility of the end-titre results in relation to the reference laboratory, ranged from 31 to 85%. These results demonstrate an excellent performance of the DAT-LPC, and validate it for the diagnosis of VL that could replace the immunofluorescent antibody test as the routine diagnostic test in the Brazilian public health system.

The article deals with the quality of data obtained in the quantitative research process. The authors decided to raise the subject, because in the Polish political science – in the opposition to sociology – it’s not well described. The text was made mainly on the basis of a literature review devoted to particular parts of the article and is its synthesis. During the work it was discovered that thanks to relatively simple tools, as well as the use of some programs the quality of the data set can be checked in a simple and precise way.

Iris laevigata Fisch., is restricted geographically in Manipur, north eastern India. An experiment was planned with Factorial Randomized Block Design during 2018 to 2020 (3 seasons) in the study plots of ICAR Research Complex for NEH Region, Manipur Centre, Lamphelpat. The study encompasses on the growth and development of I. laevigata seedlings collected from Ipa Thoukok Complex: influenced by spacing, trimming and correlations amongst the growth parameters, so as to validate the most favourable conservation method for this endangered plant. Among the treatments, T6 = S2T3 (45 cm &times; trimming 40 DAT) was found the best treatment. T9 = S3T3 (60 cm &times; trimming 40 DAT) was second, T3 = S1T3 (30 cm &times; trimming 40 DAT) was the third and lowest was found in case of T1 = S1T1 (30 cm &times; 0 trimming, i.e., without trimming). Strong positive correlation between leaf surface area and plant height on the growth and development was found to be effective among other correlations.

Abstract:
Estudos sobre padrões alimentares dependem da validade dos métodos, como o questionário de frequência alimentar (QFA), para estimar consumo alimentar de longo prazo, que pode ser superestimado e o método precisa ser validado. Questiona-se se estudos de validação aninhados a estudos com desenho amostral complexo (DAC) deveriam ignorar a dependência dos dados. Este estudo objetiva validar o QFA aplicado a adolescentes escolares e estimar os fatores de calibração em um estudo com amostragem complexa. A validade do QFA é testada frente a registros alimentares (RA) aplicados a 85 adolescentes em um estudo com DAC, com estimação dos fatores de calibração (λ) para energia, nutrientes e grupos alimentares (GA). Coeficiente de correlação intraclasse foi calculado; diferenças médias foram comparadas via modelagem, e foram realizadas análises de concordância. As médias entre os métodos diferiram para energia e nutrientes (p<0,05), mas quatro GA foram similares. Limites de concordância não se mostraram aceitáveis para os nutrientes. Coeficientes de correlação de concordância de fibras, zinco e ácidos graxos, ajustados pela energia, variaram de 0,16 a 0,22 e para os GA variaram de 0,07 a 0,36. Os fatores de atenuação corroboraram as diferenças (λ<0,40). O QFA apresentou validade relativa, sobretudo para os GA e λ aproximou as estimativas do QFA ao RA, sendo relevante quando nutrientes são usados em análises epidemiológicas. O ajuste das análises pelo DAC confirmou que a estrutura de dependência dos dados deve ser considerada, mesmo em estudos de validação, devendo-se evitar tratar tais dados como se fossem oriundos de amostra aleatória simples.

Little is known of the precise relationship between the expression of disease-related metabolic patterns and nigrostriatal dopaminergic dysfunction in parkinsonism. We studied 51 subjects with Parkinson's disease (PD) (18 non-demented, 24 demented, and 9 dementia with Lewy bodies) and 127 with atypical parkinsonian syndromes (47 multiple system atrophy (MSA), 38 progressive supranuclear palsy (PSP), and 42 corticobasal syndrome (CBS)) with 18F-fluorodeoxyglucose PET to quantify the expression of previously validated disease-related patterns for PD, MSA, PSP, and CBS and 18F-fluoropropyl-β-CIT PET to quantify caudate and putamen dopamine transporter (DAT) binding. The patients in each group exhibited significant elevations in the expression of the corresponding disease-related pattern ( p < 0.001), relative to 16 healthy subjects. With the exception of cerebellar MSA (MSA-C), all groups displayed significant reductions in putamen DAT binding relative to healthy subjects ( p < 0.05). Correlations between the dopaminergic and metabolic measures were significant in PD and CBS but not in MSA and PSP. In all patient groups with the exception of MSA-C and CBS, pattern expression values and DAT binding correlated with disease duration and severity measures. The findings suggest that in these parkinsonian disorders, metabolic network expression and DAT binding provide complementary information regarding the underlying disease process.

Abstract:
Dolphin-Assisted Therapies (DAT) are alternative therapies aimed to reduce anxiety levels, stress relief and physical benefits. This paper is focused on measuring and analyzing dolphins brain activity when DAT is taking place in order to identify if there is any differences in female dolphin’s neuronal signal when it is interacting with control or intervention subjects, performing our research in Delfiniti, Ixtapa, Mexico facilities. We designed a wireless and portable electroencephalographic single-channel signal capture sensor to acquire and monitor the brain activity of a female bottle-nose dolphin. This EEG sensor was able to show that dolphin activity at rest is characterized by high spectral power at slow-frequencies bands. When the dolphin participated in DAT, a 23.53% increment in the 12–30 Hz frequency band was observed, but this only occurred for patients with some disease or disorder, given that 0.5–4 Hz band keeps it at 17.91% when there is a control patient. Regarding the fractal or Self-Affine Analysis, we found for all samples studied that at the beginning the dolphin’s brain activity behaved as a self-affine fractal described by a power-law until the fluctuations of voltage reached the crossovers, and after the crossovers these fluctuations left this scaling behavior. Hence, our findings validate the hypothesis that the participation in a DAT of a Patient with a certain disease or disorder modifies the usual behavior of a female bottle-nose dolphin.

Abstract:
Chamomile is a medicinal plant, which presents several biological effects, especially the anti-inflammatory effect. One of the compounds related to this effect is apigenin, a flavonoid that is mostly found in its glycosylated form, apigenin-7-glucoside (APG), in natural sources. However, the affectivity and safety of this glycoside have not been well explored for topical application. In this context, the aim of this work was to develop and validate a reversed-phase high-performance liquid chromatography (RP-HPLC-DAD) method to quantify APG in chamomile preparations. Additionally, the safety and the anti-inflammatory potential of this flavonoid were verified. The RP-HPLC-DAD method was developed and validated with linearity at 24.0–36.0 μg/mL range (r=0.9994). Intra- and interday precision (RSD) were 0.27–2.66% and accuracy was 98.27–101.21%. The validated method was applied in the analysis of chamomile flower heads, glycolic extract, and Kamillen cream, supporting the method application in the quality control of chamomile preparations. Furthermore, the APG safety was assessed by MTT cytotoxicity assay and mutagenic protocols and the anti-inflammatory activity was confirmed by a diminished TNF-αproduction showed by mice macrophages treated with APG following LPS treatment.

ABSTRAK Indonesia dikenal dengan negara yang memiliki beragam suku, budaya, dan bahasa. Pada tahun 2010, data jumlah penduduk Indonesia yang paling terakhir menyatakan jumlah penduduk Indonesia sebesar 237,56 juta jiwa dengan 85.1% nya beragama Islam dan 288.405 penduduknya berprofesi sebagai seorang perawat. Pemahaman perawat tentang keperawatan transkultural saat ini sangatlah dibutuhkan. Agama, salah satu dari tujuh faktor yang menurut Leininger memiliki andil besar dalam mempengaruhi perilaku dan sikap para perawat maupun para klien. Simanjuntak (2011) berdasarkan studi kualitatif mengutarakan 13 karakteristik yang perlu di validasi sebagai karakteristik dari perawat Islam saat melakukan perawatan kepada klien. Penelitian ini dilakukan untuk memvalidasi butir-butir gambaran identitas profesional perawat Islam Indonesia melalui rancang bangun dari lembar kaji. Desain dalam penelitian ini adalah validasi kuesioner terhadap faktor rancang bangun, dimensi dan construct validity. Penelitian ini dilakukan dengan melakukan pilot study terlebih dahulu di Rumah Sakit Cibabat dan Rumah Sakit Imanuel dengan kemudian meminta responden untuk mengisi kuesioner serta memberikan komentar terhadap setiap butir-butir pernyataan, Data aktual berasal dari Rumah Sakit Santosa Bandung, Rumah Sakit Meilia Cibubur, dan Rumah Sakit Umum Daerah Koja Jakarta dengan cara meminta responden untuk mengisi kuesioner. Sampel dipilih dengan menggunakan metode snowballing kepada perawat Islam Indonesia yang bekerja di Bandung dan Jakarta. Data dianalisa dengan Cronbach α, didapatkan hasil pada uji validitas terhadap 13 karakteristik identitas profesional perawat Islam Indonesia masing-masing memiliki nilai Cronbach α >0,70. Hasil penelitian ini menyarankan agar perawat Islam Indonesia dapat menjadi perawat yang profesional dalam mengkaji diri agar dapat menuju keperawatan lintas budaya yang berkompetensi.  Kata Kunci: Validasi, Identitas Profesional, Perawat Islam Indonesia.  ABSTRACT Indonesia is known as a country with diverse in ethnic, cultural, and dialect. Religion is one of the seven factors that according Leininger determined the behavior and attitudes of nurses and clients. Therefore, understanding of transcultural nursing is imperatively necessary. In a qualitative study, Simanjuntak (2011) states 13 characteristics of Indonesian Islam nurse professional identity need to be validated. This study conducted to validate the grains picture of Indonesian Islam nurses professional identity through the design of identity assessment tool. This study validates the identity assessment tool, its design, dimensions and the construct validity. Pilot study was done in the Cibabat Hospital and Immanuel Hospital, and the actual data were collected from Santosa Bandung Hospital, Meilia Cibubur Hospital, and the Koja General Hospital in Jakarta. Samples were selected using snow-ball method among Islam nurses who work in Bandung and Jakarta. The result obtained on the validity of the 13 characteristics of professional identity of Indonesian Islam professional nurses with the value of Cronbach α> 0.70. The results of this study suggest that Indonesian Islam nurses can become a professional nurse in assessing themselves in order to lead a cross-cultural nursing competence. Multiplex immunofluorescence (mIF) and digital image analysis (DIA) have transformed the ability to analyse multiple biomarkers. We aimed to validate a clinical workflow for quantifying PD-L1 in non-small cell lung cancer (NSCLC). NSCLC samples were stained with a validated mIF panel. Immunohistochemistry (IHC) was conducted and mIF slides were scanned on an Akoya Vectra Polaris. Scans underwent DIA using QuPath. Single channel immunofluorescence was concordant with single-plex IHC. DIA facilitated quantification of cell types expressing single or multiple phenotypic markers. Considerations for analysis included classifier accuracy, macrophage infiltration, spurious staining, threshold sensitivity by DIA, sensitivity of cell identification in the mIF. Alternative sequential detection of biomarkers by DIA potentially impacted final score. Strong concordance was observed between 3,3’-Diaminobenzidine (DAB) IHC slides and mIF slides (R2 = 0.7323). Comparatively, DIA on DAB IHC was seen to overestimate the PD-L1 score more frequently than on mIF slides. Overall, concordance between DIA on DAB IHC slides and mIF slides was 95%. DIA of mIF slides is rapid, highly comparable to DIA on DAB IHC slides, and enables comprehensive extraction of phenotypic data and specific microenvironmental detail intrinsic to the sample. Exploration of the clinical relevance of mIF in the context of immunotherapy treated cases is warranted.

Abstract: The objective of this work was to validate a fast method with low-solvent use, for the analysis of trans-resveratrol and quercetin in red wines. The wines were prepared, using a classical method, from the grape (Vitis vinifera) cultivars Cabernet Franc, Cabernet Sauvignon, Malbec, Merlot, Petit Verdot, Pinot Noir, Ruby Cabernet, Syrah, Tannat, and Tempranillo. Samples were filtered and analyzed by high-pressure liquid chromatography (HPLC) with a diode array detector (DAD), at 306 and 371 nm, for trans-resveratrol and quercetin, respectively. An octylsilane column was used, and the mobile phase was composed by a gradient of methanol, water, and formic acid. The method was validated according to the following figures of merit: specificity, linearity, limit of detection, limit of quantification, precision, accuracy, and robustness. The proposed HPLC-DAD method may be established for the analysis of trans-resveratrol and quercetin in red wines.

The dopamine transporter (DAT) is an important imaging target as changes in DAT have been implicated in a variety of neurologic and psychiatric disorders and can result from certain classes of medications. [11C] N-(3-iodoprop-2 E-enyl)-2β-carbomethoxy-3β-(4-methylphenyl)nortropane ([11C]PE2I), a radioligand with high specificity for DAT, has been shown to exhibit favorable kinetics and to produce high contrast positron emission tomography (PET) images. To better characterize this ligand and to assess its measurement reliability, PET images of seven subjects were acquired in a test–retest paradigm. For optimal model performance, each subject was scanned for 120 mins, ensuring that high binding regions could reach equilibrium, a validated coregistration method was performed for accurate anatomic delineations and an exhaustive search for a reference region having one-tissue compartment kinetics was undertaken. Eleven modeling methods were tested and six metrics were used for method evaluation. A noniterative two-tissue compartment method with 100 mins of scanning time was found to be optimal for characterizing [11C]PE2I.

É necessário encontrar meios para quantificar de maneira eficiente e econômica o nível de atividade física da população. Os questionários são formas viáveis e econômicas, embora seja discutível a fidedignidade dessas medidas. Este estudo objetivou determinar a reprodutibilidade e a validade do Questionário Internacional de Atividade Física (IPAQ) em homens idosos. A amostra foi composta por 29 homens idosos acima de 60 anos, com média de idade de 66,6 anos (DP = 4,3), participantes do programa de extensão da Universidade Federal de Santa Catarina. Os instrumentos utilizados foram: a) IPAQ, forma longa; b) pedômetro; e c) Diário de Atividade Física de Bouchard (DAF). Para a reprodutibilidade foram realizadas duas aplicações do IPAQ, com intervalo de 21 dias (r s = 0,95). A análise estatística adotada foi a correlação de Spearman (r s), o percentual de concordância (%C), o índice kappa (k) e a plotagem de Bland e Altman. A amostra foi dividida, utilizando-se como critério a mediana. A reprodutibilidade apresentou correlação de r s = 0,95. A associação entre o IPAQ e o DAF foi de: r s = 0,38; %C = 69 e k = 0,04 e a associação entre o IPAQ e o pedômetro de: r s = 0,24; %C = 62 e k = 0,19. Concluiu-se que a validade variou de moderada a baixa, enquanto a reprodutibilidade foi adequada.

In this study, the HPLC and Total Organic Carbon (TOC) analysis methods have been developed and validated for the determination of the amount of Mometasone furoate and CIP100 residues on the surface of production equipment and to confirm the efficiency of the cleaning procedure. Mometasone furoate and CIP100 have been chosen based on the worst case assessment approach. The limit of mometasone furoate contamination that has been determined is 100 ug/swab and the specified limit of CIP100 contamination is 100 ug/swab which should not be exceeded during sequential cleaning of the equipment. Mometasone furoate shows good linearity in the range 0.1-1.0 ppm for the HPLC method and CIP100 2.0-10.0 ppm for TOC Analysis. The percentage of recovery from stainless steel plates using swab sampling techniques was found in the limits of 95.12% and 99.93% respectively in the HPLC and TOC methods. Both methods are simple, inexpensive, short analysis time and high sensitivity for quantitative determination of Mometasone furoate and CIP100 on the surface of manufacturing equipment well below the contamination limit. The validated method meets the requirements for demonstration of the validation of residual cleaning of mometasone furoate and CIP100 on the surface of production equipment

Objective: This study aims to develop the extraction of the marker kaempferol in the fluid extract (FE) and validate an analytical method that monitors the quality of extracts of P. pyramidalis. Methods: The P. pyramidalis leaves were collected and then were dried to milling process. The extracts were drawn up at 20% weight: Volume (w/v) by maceration, and the extraction system used was hydroethanol solution ratio at 50:50 volume: Volume (v: v). From the hydroalcoholic extract, a method of extracting the kaempferol biomarker was developed and validated by high-performance liquid chromatography coupled with diode array detector. To validate a method, the following parameters were evaluated: Specificity, selectivity, linearity, limit of quantification (LOQ) and detection (LOD), precision, accuracy, robustness, and stability. Results: The method developed proved to be efficient, as it allowed the analysis of the interferents free marker, with recovery above 90%, linear over the range 1.4–26.6 μg/mL, correlation coefficient R2=0.999, and LOD and LOQ 0.07 and 0.22 μg/mL, respectively, specificity, precision, accuracy, and robustness. Conclusion: The extraction methodology of the kaempferol marker was successfully developed interferents free and the validated method by HPLC-DAD represents a useful tool in the quality control of P. pyramidalis herbal medicines.

Abstract The aim of this study was to evaluate the relationship of hyposmia in Parkinson’s disease (PD) with other motor and non-motor symptoms and with the degree of nigrostriatal dopaminergic cell loss. A total of 295 patients with a diagnosis of PD were included. Olfactory function was measured using the University of Pennsylvania Smell Identification Test (UPSIT). Motor symptoms were rated using the Unified Parkinson’s Disease Rating Scale motor subscale (UPDRS III). To evaluate other non-motor symptoms, we used the Mini-Mental State Examination (MMSE) as a measure of global cognitive function and validated questionnaires to assess sleep disturbances, psychiatric symptoms, and autonomic dysfunction. A linear regression model was used to calculate correlation coefficients between UPSIT score and motor and non-motor variables [for psychiatric symptoms a Poisson regression was performed]. In a subgroup of patients (n = 155) with a dopamine transporter (DaT) SPECT scan, a similar statistical analysis was performed, now including striatal DaT binding. In the regression models with correction for age, sex, disease duration, and multiple testing, all motor and non-motor symptoms were associated with UPSIT scores. In the subgroup of patients with a DaT-SPECT scan, there was a strong association between olfactory test scores and DaT binding in both putamen and caudate nucleus. Hyposmia in PD is associated with various motor and non-motor symptoms, like cognition, depression, anxiety, autonomic dysfunction and sleep disturbances, and with the degree of nigrostriatal dopaminergic cell loss. This finding adds further confirmation that hyposmia holds significant promise as a marker of disease progression.

Sibling conflict has some influences on children’s development, positively and negatively. It is not only about the frequency, but also about the strategies used in managing conflict. The main purpose of this study was to adapt and to validate Sibling Conflict Strategy Scale developed by Recchia (2009), so it can be used by parents in Indonesia to identify their children’s strategies. The process of adaptation was guided by the International Testing Commission (ITC) Guidelines model for test adaptation, which include forward – backward translation processes, followed by expert reviews to test language and conceptual equivalence. Then, we conducted the validation process by testing the Content Validity Index (CVI) and Confirmatory Factor Analysis (CFA). Two hundred and six parents in Surabaya who have children in the age range of 52-87 months (M = 71.21) were participated in the pilot study. The result showed that each indicator, dimension, and construct met all the Goodness of Fit criteria (RMSEA < .08; CFI > .9). In addition, the value of Construct Reliability (CR) > .70 and Average Variance Extracted (AVE) > .50 were also obtained. Thus, the Sibling Conflict Strategy Scale can be used as a valid and reliable measurement to measure children’s sibling conflict strategies in Indonesia.

ObjectiveTo validate the role of the dopamine transporter (DAT) imaging as a biomarker in multiple system atrophy (MSA), we analyzed the association between spatial patterns of [18F]fluoro-propyl-carbomethoxy-iodophenyl-tropane ([18F]FP-CIT) PET and the clinical characteristics of MSA.MethodsSixty-five patients with MSA who underwent [18F]FP-CIT PET between 2009 and 2018 were retrospectively enrolled. To identify spatial patterns of [18F]FP-CIT PET, principal component (PC) analysis was used and correlated with the clinical presentation.ResultsOf the 65 patients, 42 presented with parkinsonian subtype of MSA, and 23 presented with cerebellar subtype of MSA (mean age 63.7 ± 9.3 years; disease duration, 1.8 ± 1.8 years). Each PC represents a specific pattern of degeneration: PC1 and PC2 were associated with the DAT binding of the entire putamen and the posterior putamen, respectively. PC3 was associated with increased [18F]FP-CIT uptake of the caudate and decreased uptake of the dorsal pons. PC2 was significantly correlated with the presence of parkinsonism (p = 5.34 × 10−5) and a positive levodopa response (p = 0.044), with age as a cofactor. PC3 was correlated with the presence of urinary incontinence (p = 0.036). Onset age was significantly correlated with both PC2 (R = 0.48, p = 5.0 × 10−5) and PC3 (R = −0.39, p = 0.0013).ConclusionsThe spatial pattern of DAT binding can reflect distinct clinical features of MSA and provides insight into the underlying pathophysiology of a broad spectrum of clinical features in MSA.

Global staging measures for dementia of the Alzheimer type (DAT) assess the influence of cognitive loss on the ability to conduct everyday activities and represent the “ultimate test” of efficacy for antidementia drug trials. They provide information about clinically meaningful function and behavior and are less affected by the “floor” and “ceiling” effects commonly associated with psychometric test. The Washington University Clinical Dementia Rating (CDR) is a global scale developed to clinically denote the presence of DAT and stage its severity. The clinical protocol incorporates semistructured interviews with the patient and informant to obtain information necessary to rate the subject's cognitive performance in six domains: memory, orientation, judgment and problem solving, community affairs, home and hobbies, and personal care. The CDR has been standardized for multicenter use, including the Consortium to Establish a Registry for Alzheimer's Disease (CERAD) and the Alzheimer's Disease Cooperative Study, and interrater reliability has been established. Criterion validity for both the global CDR and scores on individual domains has been demonstrated, and the CDR also has been validated neuropathologically, particularly for the presence or absence of dementia. Standardized training protocols are available. Although not well suited as a brief screening tool for population surveys of dementia because the protocol depends on sufficient time to conduct interviews, the CDR has become widely accepted in the clinical setting as a reliable and valid global assessment measure for DAT.

Dual antiplatelet therapy (DAT) is recommended for all patients undergoing percutaneous coronary intervention (PCI), as it significantly reduces the ischemic risk at the cost of increasing the incidence of bleeding events. Several clinical predictive models were developed to better stratify the bleeding risk associated with DAT. This systematic review aims to perform a literature survey of both standard and emerging bleeding risk scores and report their performance on predicting hemorrhagic events, especially in the era of second-generation drug-eluting stents and more potent P2Y12 inhibitors. We searched PubMed, ScienceDirect, and Cochrane databases for full-text studies that developed or validated bleeding risk scores in adult patients undergoing PCI with subsequent DAT. The risk of bias for each study was assessed using the prediction model risk of bias assessment tool (PROBAST). Eighteen studies were included in the present systematic review. Bleeding risk scores showed a modest to good discriminatory power with c-statistic ranging from 0.49 (95% CI, 0.45–0.53) to 0.82 (95% CI, 0.80–0.85). Clinical models that predict in-hospital bleeding events had a relatively good predictive performance, with c-statistic ranging from 0.70 (95% CI, 0.67–0.72) to 0.80 (95% CI, 0.73–0.87), depending on the risk scores and major hemorrhagic event definition used. The knowledge and utilization of the current bleeding risk scores in appropriate clinical contexts could improve the prediction of bleeding events.

Methods: Five activities of daily living were incorporated into a newly developed and validated Dupuytren’s assessment tool (DAT). Sixty healthy participants were assessed with the DAT wearing a range of 12 dorsal blocking splints. Half wore them on their right little finger, the other half on their ring finger. These induced flexion deformities mimicking DC of the MCPJ, PIPJ and a combination of the two. The angles of flexion deformity at which important hand disability occurred were calculated using receiver operating characteristic curves. Results: Clinically important hand disability occurred for the little finger MCPJ, PIPJ and combined MCPJ and PIPJ angulation at 52.5, 67.5, and 75 degrees respectively. For the ring finger joint, the angulations were 52.5, 67.5 and 75 degrees respectively. Conclusions: This information will provide information for clinicians and patients as to when clinically important disability tends to occur in DC.

AbstractAlthough dicamba-resistant crops can provide an effective weed management option, risk of dicamba off-site movement to sensitive crops is a concern. Previous research with indeterminate soybean identified 14 injury criteria associated with dicamba applied at V3/V4 or R1/R2 at 0.6 to 280 g ae ha−1. Injury criteria rated on a 0 to 5 scale (none to severe), along with percent visible injury and plant height reduction, and canopy height collected 7 and 15 d after treatment (DAT) were analyzed using multiple regression with a forward-selection procedure to develop yield prediction models. Variables included in the 15 DAT models (in order of selection) for V3/V4 were lower stem base lesions/cracking, plant height reduction, terminal leaf epinasty, leaf petiole droop, leaf petiole base swelling, and stem epinasty, whereas for R1/R2 variables were lower stem base lesions/cracking, terminal leaf chlorosis, leaf petiole base swelling, stem epinasty, terminal leaf necrosis, and terminal leaf cupping. To validate the models, experiments including the same dicamba rates and application timings used in previous research were conducted at two locations. For the variables specific to each model, data collected for the dicamba rates were used to predict yield. For the V3/V4 15 DAT model, predicted yield reduction (compared with the nontreated control for dicamba at 0.6 to 4.4 g ha−1) underestimated or overestimated observed yield reduction by an average of 1 and 3 percentage points. For 8.8 g ha−1, predicted yield reduction overestimated observed yield reduction by 8 points and for 17.5 g ha−1 by 20 points. For the R1/R2 15 DAT model, predicted yield reduction for 0.6 to 4.4 g ha−1 overestimated observed yield reduction by an average of 3 to 5 percentage points. For dicamba at 8.8 g ha−1, predicted yield reduction underestimated observed yield reduction by 8 points and for 17.5 g ha−1 overestimated by 6 points.

Abstract Clove (Eugenia caryophyllata) is a well-known medicinal plant used for diarrhea, digestive disorders, or in antiseptics in Korea. Eugenol is the main active ingredient of clove and has been chosen as a marker compound for the chemical evaluation or QC of clove. This paper reports the development and validation of an HPLC-diode array detection (DAD) method for the determination of eugenol in clove. HPLC separation was accomplished on an XTerra RP18 column (250 4.6 mm id, 5m) with an isocratic mobile phase of 60 methanol and DAD at 280 nm. Calibration graphs were linear with very good correlation coefficients (r2 &gt; 0.9999) from 12.5 to 1000 ng/mL. The LOD was 0.81 and the LOQ was 2.47 ng/mL. The method showed good intraday precision (RSD 0.080.27) and interday precision (RSD 0.321.19). The method was applied to the analysis of eugenol from clove cultivated in various countries (Indonesia, Singapore, and China). Quantitative analysis of the 15 clove samples showed that the content of eugenol varied significantly, ranging from 163 to 1049 ppb. The method of determination of eugenol by HPLC is accurate to evaluate the quality and safety assurance of clove, based on the results of this study.

Abstract Introduction/Objective The Direct Antiglobulin Test (DAT) is a useful screening test to determine whether a patient’s red blood cells have been sensitized to immunoglobulin or complement. Like the majority of screening tests, the DAT compromises specificity for sensitivity in order to quickly assess for hemolysis and its dangerous sequelae. Because of the high sensitivity of the DAT, improper ordering of this test can confuse the clinical picture at best and result in misdiagnosis at worst. Pre-tests for ruling-out disease have proven effective in limiting test ordering, such as the Pulmonary Embolism Rule-out Criteria (“PERC rule”) for limiting the use of D-dimers. A similar pre-test could prove useful in DATs. We use “in silico testing” – testing via computer simulation – to predict the likelihood of any DAT being positive, given common patient attributes and laboratory values. Methods A three-layer deep-learning artificial neural network (ANN) was created using Python and the machine learning framework Keras. The ANN was compiled to maximize specificity while retaining 100% sensitivity. Input variables to the model were patient sex and age, along with most recent lab values for hemoglobin, hematocrit, white blood cell count, platelet count, total bilirubin, direct bilirubin, and haptoglobin, where available. The output of the ANN was a binary variable: “ruled-out” versus “further testing necessary”. The ANN was trained on all positive (n=30) and negative (n=63) DATs performed on November 2019 through March 2020, with a final total of ninety-three patients. A 10-fold cross-validation of the entire dataset was used to measure performance. Results The ANN was able to maximize specificity to 83% while retaining 100% sensitivity. A ROC curve of the model shows performance well above that of the no-discrimination line. Conclusion “In-silico testing” can accurately screen the likelihood of a positive result for a labor and time-intensive test, such as the DAT, before the actual test is performed. This has the potential to reduce unnecessary testing if validated in clinical practice. Analogous to their clinical counterpart “the PERC rule,” computer models that maximize specificity while retaining 100% sensitivity could achieve more effective test utilization and more informative results.

Instrumen dikembangkan dengan mengikuti langkah-langkah yang meliputi (1) menyusun spesifikasi tes, (2) menulis soal tes, (3) menelaah soal tes, (4) melakukan uji coba tes, (5) menganalisis butir tes, (6) memperbaiki tes, dan (7) merakit tes. Uji coba instrumen dilakukan di jurusan PGSD UPP Denpasar pada semester II dengan jumlah mahasiswa sebanyak 90 mahasiswa. Jumlah butir soal yang di cobakan sebanyak 30 butir soal pilihan ganda. Hasil penelitian menunjukkan bahwa instrumen yang dikembangkan valid dan reliabel berdasarkan uji ahli dan uji validitas dengan data empiris dan dengan koefisien koefisien reliabilitas Kuder Richadson (KR-20) sebesar 0,84.

The growth of the hotel business is increasing, along with the development of tourism. This condition encourages hoteliers to compete to advance their business with a variety of service innovation processes. Therefore, it is essential to know and identify the innovative behavior of hospitality employees. This study aims to develop innovative employee behavior measurement tools. We used a quantitative study involving 143 sales marketing employees in hospitality as respondents. The sample selection uses a random sampling technique. The development of innovative behavioral questionnaires was revealed through three dimensions, namely the idea generation dimension, idea championing, and idea implementation. Data analysis used confirmatory factor analysis (CFA) with the help of Amos 22 software. The results of the study revealed that the innovative behavior measurement model met the goodness of fit criteria. These findings indicate that innovative behavior can be measured by the dimensions of idea generation, idea championing, and idea implementation. The results of this study also have implications for several hospitality practitioners to evaluate the innovative behavior of hospitality employees. The instrument development consists of three stages: 1) Focus group discussion (FGD) to find out the description of perceived parents’ supportive communication (35 respondents from three universities). The result of this stage is the items of the instrument; 2) Validity and Reliability testing (326 respondents of high school and university students). The result of this stage is the valid and reliable instrument; 3) Factor Analysis from 600 respondents. The result of this stage is the dimentions of the instrument; dan 4) Confirmatory Factor Analysis (CFA) from 1065 respondents. The result of this stage is the same as final result of the whole study, that is perceived parents’ supportive communication instrument, which consists of two dimentions (empathy and confidentiality).

To date there is no validated, 18F-labeled dopamine transporter (DAT) radiotracer with a rapid kinetic profile suitable for preclinical small-animal positron emission tomography (PET) studies in rodent models of human basal ganglia disease. Herein we report radiosynthesis and validation of the phenyltropane 18F-FP-CMT. Dynamic PET recordings were obtained for 18F-FP-CMT in six untreated rats, and six rats pretreated with the high-affinity DAT ligand GBR 12909; mean parametric maps of binding potential ( BPND) relative to the cerebellum reference region, and maps of total distribution volume ( VT) relative to the metabolite-corrected arterial input were produced. 18F-FP-CMT BPND maps showed peak values of ˜4 in the striatum, versus ˜0.4 in the vicinity of the substantia nigra. Successive truncation of the PET recordings indicated that stable BPND estimates could be obtained with recordings lasting only 45 minutes, reflecting rapid kinetics of 18F-FP-CMT. Pretreatment with GBR 12909 reduced the striatal binding by 72% to 76%. High-performance liquid chromatography analysis revealed rapid metabolism of 18F-FP-CMT to a single, non-brain penetrant hydrophilic metabolite. Total distribution of volume calculated relative to the metabolite-corrected arterial input was 4.4 mL/g in the cerebellum. The pharmacological selectivity of 18F-FP-CMT, rapid kinetic profile, and lack of problematic metabolites constitute optimal properties for quantitation of DAT in rat, and may also predict applicability in human PET studies.

Abstract Accurate and precise TLC-densitometric and HPLC–diode-array detector (DAD) methods have been developed and validated to resolve two binary mixtures containing pyridoxine hydrochloride (PYH) with either cyclizine hydrochloride (CYH) or meclizine hydrochloride (MEH). In the developed TLC-densitometric method, chromatographic separation of the three studied drugs was carried out on silica gel 60 F254 plates using a developing system containing methylene chloride + acetone + methanol (7 + 1 + 0.5, v/v/v) scanning separated bands at 220 nm. Beer–Lambert law was obeyed in the ranges of 0.2–5, 0.2–4, and 0.2–4 µg/band for PYH, CYH, and MEH, respectively. On the other hand, the developed HPLC–DAD method depended on chromatographic separation on a Zorbax Eclipse C18 column using methanol–KH2PO4 (0.05 M; 90 + 10, v/v; pH 5, with H3PO4 and KOH) as the mobile phase, a flow rate of 1 mL/min, and UV scanning at 220 nm. A linear relationship was obtained between the integrated peak area and the concentration in the ranges of 10–50, 10–50, and 7–50 µg/mL for PYH, CYH, and MEH, respectively. The proposed methods were successfully applied for the determination of thecited drugs in their pharmaceutical formulations. Statistical comparison with the reported methods using Student's t- and F-tests found there were no significant differences between the proposed and reported methods for accuracy and precision.

10045 Background: Survivors of childhood cancer have functional limitations and health-related morbidity consistent with an accelerated aging phenotype. We characterized aging using a Deficit Accumulation Index (DAI) which examines the accumulation of multiple aging-related deficits readily available from medical records and self-report. DAI’s are used as surrogates of biologic aging and are validated to predict mortality in adult cancer patients. Methods: We included childhood cancer survivors (N = 3,758, mean age 30 [SD 8], 22 [9] years post diagnosis, 52% male) and community controls (N = 575, mean age 34 [10] 44% male) who completed clinical assessments and questionnaires and who were followed for mortality through December 31st, 2018 (mean follow-up 6.1 [3.1] years). Using the initial SJLIFE clinical assessment, a DAI score was generated as the proportion of deficits out of 44 items related to aging, including chronic conditions (e.g. hearing loss, hypertension), psychosocial and physical function, and activities of daily living. The total score ranged 0 to 1; scores > 0.20 are robust, while moderate and large clinically meaningful differences are 0.02 and 0.06, respectively. Linear regression compared the DAI in survivors and controls with an age*survivor/control interaction and examined treatment associations in survivors. Cox-proportional hazards models estimated risk of death associated with DAI. All models were adjusted for age, sex, and race. Results: Mean [SD] of DAI was 0.17 [0.11] for survivors and 0.10 [0.08] for controls. 32% of survivors had a DAI above the 90th percentile of the control distribution (p < 0.001). After adjustment for covariates, survivors had a statistically and clinically meaningfully higher DAI score than controls (β = 0.072 95%CI 0.062, 0.081; p < 0.001). When plotted against age, the adjusted DAI at the average age of survivors (30 years) was 0.166 (95% CI 0.160,0.171), which corresponded to 60 years of age in controls, suggesting premature aging of 30 years. The mean difference in DAI between survivors and controls increased with age from 0.06 (95% CI 0.04, 0.07) at age 20 to 0.11 (95% CI 0.08, 0.13) at age 60, consistent with an accelerated aging phenotype (p = 0.014). Cranial radiation, abdominal radiation, cyclophosphamide, platinum agents, neurosurgery, and amputation were each associated with a higher DAI (all p≤0.001). Among survivors, a 0.06 increase in DAI was associated with a 41% increased risk of all-cause mortality (HR 1.41 95%CI 1.32, 1.50; p < 0.001). Conclusions: Survivors of childhood cancer experience significant age acceleration that is associated with an increased risk of mortality; longitudinal analyses are underway to validate these findings. Given the ease of estimating a DAI, this may be a feasible method to quickly identify survivors for novel and tailored interventions that can improve health and prevent premature mortality.

AbstractThe aim of this paper is to optimize and validate a high performance liquid chromatography (HPLC) method for separation and quantification of five isoflavones. A statistical central composite design was used to separate all peaks. These multivariate procedures were efficient in determining the optimal separation condition using resolution, capacity factor, asymmetry and number of theoretical plates. The effective separation of the examined compounds was applied on a Develosil RP Aqueous AR 5 RP-30 column with a gradient mobile phase system and a DAD detector. The isolation and preconcentration of the isoflavones from urine and plasma samples were conducted by means of the solid-phase extraction (SPE). For optimize SPE conditions various sorbents were tested. Furthermore, high recoveries and good relative standard deviations were obtained when the samples were passed through the Oasis HLB column. The developed method was validated and successfully applied for determination of isoflavones in urine and plasma.

There are students in the class who can arrange themselves to rarely connect with the opposite sex. not adjacent to friends of the opposite sex, students are reluctant to keep their distance and there are also students who always play with many students in the class. The above behavior, can be referred to as students who have the characteristics of high sexual deviation. This study was conducted by developing new instruments to measure whether sexual deviation in students can be minimized or lower. this scale is called the decline of sexual deviation. The sample of the study consisted of 8th grade junior high school students as many as 186 people from 8 eight classes that the researcher took 5 classes in SMP Negeri 3 Singaraja. Manually filled instrument format (paper-and-pencil). Analytical analyzes use the Rasch model through the Winsteps software. An interesting finding found that 8th grade school students showed different responses amongst students with other students; while the aitem analysis shows all the items are appropriate and the instrument meets the requirements of unidimensionality and some of the detected items contain biases.

Salah satu cara untuk mengetahui prognosis pasien perdarahan subaraknoid ialah dengan menggunakan sistem skoring. Skor dapat membantu para klinisi untuk menilai kondisi pasien, prognosis, serta menentukan penatalaksanaan yang terbaik bagi pasien. Tujuan Penelitian: Mengukur faktor prediktor yang mempengaruhi mortalitas pada pasien perdarahan subaraknoid dan meramalkan prognosis pasien perdarahan subaraknoid menggunakan skor prediktor mortalitas. Metode: Pelaksanaan penelitian ini menggunakan metode kohort retrospektif. Data penelitian ini diperoleh dari data sekunder register stroke dan rekam medik pasien perdarahan subaraknoid di RS Bethesda Yogyakarta. Uji validitas menggunakan kurva receiver-operating characteristic (ROC) dan untuk cut off point menggunakan area under the curve (AUC). Hasil: Hasil analisis multivariate menunjukkan bahwa ada hubungan yang signifikan antara mortalitas pasien perdarahan subaraknoid dengan kesadaran (RO: 19.722, 95% IK: 1.788- 217.521, p: 0.015), tekanan darah sistolik (RO: 157.4, 95%IK: 2.068- 11990.9, p: 0.022), gula darah sewaktu (RO:12.457, 95%IK: 2.305- 67.322, p: 0.03) dan komplikasi (RO: 30.539, 95%IK: 2.685- 347.377, p: 0.006). Masing-masing variabel memiliki skor untuk menentukan prognosis pasien perdarahan subaraknoid. Skor memiliki kemampuan yang baik dalam memperediksi mortalitas pasien perdarahan subaraknoid (AUC) ROC 0.946 (95% IK: 0.896- 0.995; p: <0.001). This article reports development of new scale to measure entrepreneurial characters. Entrepreneurial Characters Scale (SK-WIRA) measures personality traits indicating a person’s capacity to engage in entrepreneurial activity and can be used to study tendency and suitability to work as entrepreneur. In preliminary study, entrepreneurial character is operationalized based on concepts found in literatures: achievement motivation, innovativeness, risk taking, and autonomy. Four SK-WIRA subscales were constructed according to those dimensions. In phase 1 item selection, exploratory factor analysis and reliability analysis were applied. Initial set of SK-WIRA constituted of 40-48 items per subscale was administered to university students (N = 130). It is found that items were not clustered based on the theory, but item types (favorable-unfavorable). Therefore, it is predicted that entrepreneurial characters and non-entrepreneurial characters are probably two different constructs and need to be measured using different tools. Final compilation of SK-WIRA consists only 22 items (all is favorable items) with good internal consistency (Cronbach’s α = .863). Phase 2 construct validity test using confirmatory factor analysis (CFA) confirms that SK-WIRA consists of four correlated factors. The examination of scale internal structure shows good model fit (c2/df = 244.077/205; RMSEA = .038; CFI = .943; TLI = .936; SRMR = .069).

The aim of this study was to develop and validate growth and photosynthetic models of Kimchi cabbages under extreme temperature conditions at different growth stages. Kimchi cabbage plants were subjected to low and high air temperatures 7–10 days after transplanting (DAT) and 40–43 DAT using extreme weather simulators. Except during these periods, the air temperature, relative humidity, solar radiation, and precipitation were set according to previous meteorological data. The experiments were performed over two years: in the first year, data were used to develop the models; the second-year experimental data were used for validation. The growth parameters and relative growth rate of Kimchi cabbage decreased due to low and high air temperature treatments. Photosynthetic CO2 response curves, which were measured using a portable gas exchange system, were used to calculate three biochemical parameters from measured data: photochemical efficiency, carboxylation conductance, and dark respiration. These parameters were used to develop the photosynthetic models (modified Thornley’s models) representing predictions of net photosynthetic rate by CO2 concentration and growth stage. The simulated photosynthetic rate with extreme high temperature treatment (35/31 °C) was 19.7 μmol m−2 s−1 which was evaluated approximately 3% deduction compared with control. Results of this study indicate that the growth and photosynthetic models developed here could be applied to evaluate retarded growth and net photosynthetic rate under extreme temperature conditions.

Abstract:
Positron emission tomography (PET) coupled to 6-[18F]Fluoro-L-Dopa (18F-Dopa) remains the gold standard for assessing dysfunctionality concerning the dopaminergic nigrostriatal pathway in Parkinson's disease and related disorders. The use of ligands of the dopamine transporters (DAT) is an attractive alternative target; consequently, the current aim was to validate one of them, 11C-PE2I, using a multiinjection modeling approach allowing accurate quantitation of DAT densities in the striatum. Experiments were performed in three controls, three MPTP-treated (parkinsonian) baboons, and one reserpine-treated baboon. 11C-PE2I B′max values obtained with this approach were compared with 18F-Dopa input rate constant values (Ki), in vitro Bmax binding of 125I-PE2I, and the number of dopaminergic neurons in the substantia nigra estimated postmortem by stereology. In the caudate nucleus and putamen, control values for 11C-PE2I B'max were 673 and 658 pmol/mL, respectively, whereas it was strongly reduced in the MPTP-treated (B′max = 26 and 36 pmol/mL) and reserpine-treated animals (B′max = 338 and 483 pmol/mL). In vivo11C-PE2I B′max values correlated with 18F-Dopa Ki values and in vitro125I-PE2I Bmax values in the striatum and with the number of nigral dopaminergic neurons. Altogether, these data support the use of 11C-PE2I for monitoring striatal dopaminergic disorders and the effect of potential neuroprotective strategies.

Context Hormone receptors HER2/neu and Ki-67 are markers of residual risk in early breast cancer. An algorithm (IHC4) combining these markers may provide additional information on residual risk of recurrence in patients treated with hormone therapy. Objective To independently validate the IHC4 algorithm in the multinational Tamoxifen Versus Exemestane Adjuvant Multicenter Trial (TEAM) cohort, originally developed on the trans-ATAC (Arimidex, Tamoxifen, Alone or in Combination Trial) cohort, by comparing 2 methodologies. Design The IHC4 biomarker expression was quantified on TEAM cohort samples (n = 2919) by using 2 independent methodologies (conventional 3,3′-diaminobezidine [DAB] immunohistochemistry with image analysis and standardized quantitative immunofluorescence [QIF] by AQUA technology). The IHC4 scores were calculated by using the same previously established coefficients and then compared with recurrence-free and distant recurrence-free survival, using multivariate Cox proportional hazards modeling. Results The QIF model was highly significant for prediction of residual risk (P &lt; .001), with continuous model scores showing a hazard ratio (HR) of 1.012 (95% confidence interval [95% CI]: 1.010–1.014), which was significantly higher than that for the DAB model (HR: 1.008, 95% CI: 1.006–1.009); P &lt; .001). Each model added significant prognostic value in addition to recognized clinical prognostic factors, including nodal status, in multivariate analyses. Quantitative immunofluorescence, however, showed more accuracy with respect to overall residual risk assessment than the DAB model. Conclusions The use of the IHC4 algorithm was validated on the TEAM trial for predicting residual risk in patients with breast cancer. These data support the use of the IHC4 algorithm clinically, but quantitative and standardized approaches need to be used.

ABSTRAK Anemia pada kehamilan merupakan salah satu masalah dalam kehamilan. Data WHO pada tahun 2001 menunjukkan bahwa lebih dari 50 % wanita hamil mengalami anemia. Anemia pada kehamilan merupakan suatu kondisi dimana kadar hemoglobin kurang dari 11 mg/dL. Prevalensi anemia di Indoensia cukup tinggi yaitu sebesar 50-63%, sedangkan prevalensi di DI Yogyakarta adalah 14,32%. Penelitian mengenai pengetahuan ibu hamil terkait kondisi kehamilannya di Selangor diperoleh bahwa pengetahuan ibu hamil pada kategori baik, namun sebagian besar dari subyek memiliki penegtahuan yang rendah terkait suplemen zat besi. Instrumen penelitian mengenai pengetahuan dan suplemen zat besi yang tervalidasi di Indonesia masih belum ada, sehingga data pengetahuan ibu hamil mengenai kehamilan dan suplemen zat besi masih sangat kurang. Metode dalam penelitian ini adalah dengan melakukan pengembangan instrumen penelitian (kuesioner) yang dilakukan validasi oleh tiga orang ahli di bidangnya masing-masing (validasi konten). Langkah selanjutnya adalah dengan melakukan pilot test untuk mengetahui masukan maupun umpan balik dari subyek terkait kesalahan dan item pertanyaan yang membingungkan. Pilot test juga digunakan untuk mengukur waktu pengisian kuesioner. Langkah terakhir adalah melakukan validasi construct yang dilakukan pada 49 subyek penelitian. Analisis statistik menggunakan pearson correlation dan cronbach alpha digunakan untuk mengukur validitas dan reliabilitas kuesioner. Subyek penelitian dalam validasi ini adalah sebanyak 49 pasien hamil yang mendapatkan suplemen zat besi dan atau asam folat di RS PKU Muhammadiyah Gamping pada periode Agustus sampai dengan September 2019. Berdasarkan hasil Uji validitas terhadap 15 item pertanyaan diperoleh 2 item pertanyaan yang tidak valid, selanjutnya item pertanyaan dihapus dan dilakukan validasi ulang dan diperoleh hasil valid dengan nilai koefisien pearson correlation ditasa nilai r tabel. Hasil uji reliabilitas menggunakan cronbach alpa diperoleh semua item pertanyaan memiliki nilai cronbach alpa diatas 0,6 sehingga dinyatakan reliabel. The method in this study was to develop a research instrument (questionnaire) which was validated by three experts in their respective fields (content validation). The next step is to conduct a pilot test to find out input and feedback from subjects related to errors and confusing question items. Pilot tests are also used to measure the time to fill out the questionnaire. The final step is to construct validation conducted on 49 research subjects. Statistical analysis using Pearson correlation and Cronbach alpha was used to measure the validity and reliability of the questionnaire. Research subjects in this validation were 49 pregnant patients who received iron supplements and / or folic acid at PKU Muhammadiyah Gamping Hospital in the period of August to September 2019. Based on the results of the validity test of 15 question items, there were 2 items that were invalid, further items questions were deleted and re-validated and valid results obtained with the Pearson correlation coefficient ditasa r table values. The reliability test results using Cronbach Alpha are obtained all questions items have a Cronbach Alpha value above 0.6 so that it is declared reliable.
