Full textFrom brain to bones, stiffness and composition of the extracellular matrix vary greatly and play a role in cell responses. Substrate rigidity also impacts plasma membrane tension, which has a close relationship with membrane trafficking. How substrate rigidity and chemistry sensing may regulate exocytosis, which in turn regulates membrane tension, is still largely unknown. Here, I used pHluorin imaging of single vesicle exocytosis in cells cultured on substrates of controlled rigidity and composition to explore the regulation of VAMP2 and VAMP7-mediated exocytosis. I developed a computer software to automatically identify fusion events thus allowing quick analysis of batch data. I contributed to the study showing that VAMP7 exocytosis is regulated by src kinase which phosphorylates VAMP7 in its Longin domain (LD) (Burgo et al. JBC 2013). I further found that VAMP7 but not VAMP7 lacking LD- or VAMP2-mediated secretion was stimulated by substrate stiffness on laminin. VAMP7 and VAMP7 lacking LD were similarly sensitive to osmotic chock-induced membrane tension changes. Finally, i showed that LRRK1, a regulator of egf receptor transport, is a partner of the LD, and controls the retrograde transport of VAMP7. These approaches allowed me to reveal a new mechanism whereby substrate rigidity, by acting on integrin signalling, enhances VAMP7 exocytosis via LRRK1- and Rab21-dependent regulation of its peripheral readily-releasable pool (Wang et al. submitted). This mechanism may have broad potential relevance for plasma membrane dynamics in normal conditions and diseases, particularly cancer

Full textMembrane trafficking is a universal process that is essential for neuronal function in a wide spectrum of applications. From neuronal growth and morphological development to neurotransmitter release and synaptic plasticity, it supports neuronal activity and gives countless questions that drive today’s neurobiology research. Notably, the trafficking of recycling endosomes (REs) in somatodendritic compartments participates in synaptic transmission and plasticity, such as long-term synaptic potentiation (LTP). However, the fusion machinery mediating RE exocytosis is still unclear. To identify the vesicular SNAREs (v-SNAREs) involved in different forms of postsynaptic RE exocytosis, we first imaged neuronal VAMP proteins fused with pH-sensitive pHluorin in cultured hippocampal neurons, and found that only VAMP2 and VAMP4, but not VAMP7, underwent somatodendritic exocytosis in mature neurons. After identifying these two candidate proteins, we used a combination of different downregulation techniques to chronically or acutely deactivate their function and observe consequences on REs exocytosis, basal synaptic transmission and LTP. Our results suggest that VAMP2 is involved in activity-regulated exocytosis important for LTP, but not constitutive postsynaptic AMPARs exocytosis, supporting basal transmission. VAMP4 is required for constitutive exocytosis of at least a large proportion of REs, but the functional implication of these endosomes still need to be explored, as VAMP4 downregulation did not alter basal synaptic transmission

Full textMembrane trafficking is essential for neuronal function: from growth of neurons and synapse formation to recycling of synaptic vesicles and receptors, questions concerning exocytosis and endocytosis are stimulating neurobiology research. In particular, trafficking of glutamate receptors present in recycling endosomes (REs) is necessary for the expression of long term potentiation (LTP). To investigate the mechanism of exocytosis in dendrites, we have imaged cultured rat hippocampal neurons transfected with transferrin receptor, a classical marker of REs, tagged with phluorin. As for AMPA receptors or β2-adrenegric receptors, single exocytic events has revealed two main behaviors: in most cases, receptors diffuse quickly in the plasma membrane after exocytosis (discharge events), but receptors can also remain clustered (display events). Using fast extracellular pH changes around the recorded cell, we show that for display events exocytosis is transient: after a few seconds (median 2.6 s) receptors are internalized. Moreover, using two color imaging of single exocytosis events with markers of neuronal compartments, we found that Rab11 is enriched at the exocytosis site, confirming the endosomal origin of the vesicles. Overexpression of a dominant negative form of Rab11 known to impair LTP decreases selectively the frequency of discharge events. As SNARE proteins are involved in virtually all membrane fusion processes, we investigated the role of Vamp proteins in somatodendritic exocytosis events. We found that Vamp4, unlike Vamp2 or Vamp7, is enriched in TfR containing compartments and can undergo exocytosis at high frequency and is required for TfR exocytosis

Full textThe hippocampal mossy fiber is characterized by its particular morphology, distinctsynaptic transmission and presynaptic plasticity. Moreover, this synapse has beencalled ―teacher‖ or ―detonator‖ for its proposed functional role in episodic memoryencoding. Nevertheless, the molecular mechanisms underlying its specific functionalproperties remain elusive. This work is composed of two main parts:1) Phenotyping Hippocampal Mossy Fiber Synapses in VAMP7 KO MiceVAMP7 is a vesicle SNARE of the longin family important in neurite growth duringdevelopment. In the adult brain, VAMP7 is enriched in a subset of nerve terminals,particularly at the hippocampal mossy fiber. We analyzed VAMP7 function inneurotransmitter release by characterizing basal and evoked transmission at thissynapse in KO mice and fully tested hypotheses relevant to short-term plasticity.Loss of VAMP7 has been previously reported not to cause major developmental orneurological deficits (Sato et al., 2011; Danglot et al., 2012). Presynapticmechanisms of short-term plasticity at the hippocampal mossy fiber also seemunaffected for potential reasons that will be discussed.2) CA3 Circuits Probed with RABV-Tracing and Paired RecordingsWe developed a technique to establish paired recordings between connected dentategyrus granule cells and CA3 pyramidal cells (GC-CA3) in mouse hippocampalorganotypic slice cultures. To identify direct presynaptic partners to a defined targetCA3 pyramidal cell, we combined single-cell electroporation (SCE) and mono-transsynaptictracing based on a pseudotyped, recombinant rabies virus (EnvApseudotyped RABV ΔG). Using SCE we transfected a single CA3 pyramidal cell perslice with the plasmids encoding: the RABV envelope glycoprotein (RG), afluorescent reporter, and TVA (the EnvA cognate surface receptor, which has nohomologue in mammalian cells). The slices were subsequently infected with EnvApseudotyped RABV ΔG. After 3-4 days, the RABV mono-trans-synaptic tracingrevealed the presynaptic inputs of that single neuron. Then, we were able toestablish paired recordings between connected GC-CA3 cells, as well as to quantifythe presynaptic partners of the starter CA3 pyramidal cell
De mosvezel van de hippocampus kenmerkt zich door een bijzondere morfologie,uitzonderlijke synaptische transmissie en presynaptische plasticiteit. De synapswordt ook wel "leraar" of "detonator" genoemd vanwege zijn waarschijnlijke rol in decodering van het episodisch geheugen. Toch blijven de specifieke moleculairemechanismen van dit synaps onbekend. Dit werk bestaat uit twee delen:1) Fenotypering van mosvezel synapsen van de hippocampus in VAMP7 KO muizenVAMP7 is een vesicle-SNARE van de longin familie van belang bij de groei vanneurieten tijdens de ontwikkeling. Full textThis research analyzes the development of the application called "Vamos App", which allows measuring the capacity of the premises in real time. The idea arises given the current situation in the country, where we are in a constant process of adaptation to current needs, with reduced capacity and mandatory social distancing. In addition, the lifestyle in general is increasingly dynamic and people have less time to wait. Therefore, this application will allow users to know the capacity of the premises and to know which ones have available space according to the location of the person. In addition, in our "Premium" version you can access other benefits such as discounts, the possibility of making reservations, among others. The research has allowed us to confirm the viability of the project, entering our application in the Peruvian market. For this, we have used social networks as our main sales channel, which allowed us to obtain satisfactory results expressed in the sales made. Subsequently, the process of validation of the problem and the business idea was carried out, to then carry out the future plans, with which the financial plan was obtained. The results of our research can be summarized in that our NPV is 53,933.27, with a return in year 2 and our IRR is 57.70%, which shows the viability of the project.

Esta dissertação apresenta reflexões e análises de uma experiência musical coletiva, nomeada Roda de Música, realizada no contexto de um Centro de Referência da Assistência Social - CRAS, situado na região metropolitana de Curitiba. A pesquisa teve caráter participativo e comunitário e se configurou como um tipo de pesquisa-intervenção. Os temas aqui desenvolvidos nascem de inquietações em torno dos espaços criados para o fazer musical dentro do contexto da Proteção Social Básica. Por meio de uma relação dialógica entre a Psicologia Sócio-História e a Musicoterapia de base social e comunitária, buscou-se estabelecer construções teóricas que contribuam para a compreensão do fazer musical quando em processos comunitários. De igual modo, a descrição e a análise da experiência da Roda de Música, pretendeu abrir questões em torno do tipo de atuação dos profissionais em contexto socioassistencial. Este trabalho apresenta a música como mediadora de encontros que podem expandir as possibilidades de ser, sentir, agir e pensar dos sujeitos, sobretudo por meio de um tensionamento na configuração sensível. A partir dos encontros da Roda de Música os participantes puderam construir um cancioneiro com o repertório do grupo, dedicaram alguns encontros para a re-criação das canções escolhidas e envolveram-se em um processo criativo de composição. Eles utilizaram suas produções musicais para construção de visibilidades coletivas por meio do fortalecimento dos laços comunitários, bem como com a criação de relações outras com o território quando ocuparam o espaço urbano com suas sonoridades.
Abstract : This research presents some reflections and analysis from a colletive musical experience, named Music groups, held in the context of a Reference Center for Social Welfare - CRAS, located in the metropolitan region of Curitiba. This research realized was parcipatory and have community character, as well is configured as a kind of intervention research. The themes developed were born in concerns around the spaces created for the music making within the context of Basic Social Protection. Through a satisfactory relationship between the Socio-Historical Psychology and Social and Community Music Therapy, it attempted to establish theoretical constructs that contribute to understand what music does in those communitary processes. Similarly, the description and analysis of the experience of music groups, intended to open questions about the type of professional activities in social assistance context. This paper presents music as a mediator of meetings that would expand the possibilities of being, feeling, acting and thinking from the subjects, especially through a tension in the sensitive setting. From the Music Groups meetings participants were able to build a songbook with the repertoire of the group; some meetings were devoted to the re-creation of songs chosen and engaged in a creative process of composition. They used their musical production for construction of collective visibility through the strengthening of community ties and the creation of other relations with the territory when they occupied the urban space with their sounds.

The present work has as its theme: Algebra or Geometry? Let the question! This study discusses mainly the relationship between these two distinct areas of the curriculum perspective, basic education. The goal is to show how tenuous, and at the same fruitful time, the border that separates the basic algebra and plane geometry, and how the teacher should investigate this boundary in the very first year of high school, even before submitting Analytic Geometry. This study is organized in the form of chapters, covering the following topics in order: why the mathematics teaching, the method used in problem solving as a process, the theoretical assumption, highlighting the similarity of triangles, the Pythagorean theorem, the laws of sines and cosines, the Ptolemy's theorem, and the list of proposed issues and an epistemological discussion of the proposed problems. The work was in the light of theoretical proposals Elon Lages Lima, Terence Tao and Paulo Freire, the latter a master of pedagogy. The survey was conducted exploratory and bibliographic way, qualitative character. Finally, the study aims to clarify that the curriculum division of mathematics classrooms in primary school in Algebra and Geometry is only one curriculum division and should not affect the view that all the contents studied are part of a whole perfectly consistent; given that, being on a math problem, students and teachers can make use of both tools of algebra as geometry of the results to solve it.
O presente trabalho tem como tema: Álgebra ou Geometria? Vamos à questão! Esse estudo discute, principalmente, a relação entre essas duas áreas distintas do ponto de vista curricular, no ensino básico. O objetivo é mostrar como é tênue, e ao mesmo tempo frutífera, a fronteira que separa a Álgebra básica e a Geometria Plana e, como o professor deve investigar esta fronteira logo nas primeiras séries do ensino médio, mesmo antes de apresentar a Geometria Analítica. Este estudo está organizado em forma de capítulos, abordando as seguintes temáticas em ordem: o porquê do ensino da Matemática, o método utilizado na resolução de problemas como processo, o pressuposto teórico, dando destaque à semelhança entre triângulos, ao teorema de Pitágoras, às leis dos Senos e Cossenos, ao teorema de Ptolomeu, e à lista de problemas propostos e uma discussão epistemológica dos problemas propostos. O trabalho foi à luz das propostas teóricas de Elon Lages Lima, Terence Tao e de Paulo Freire, este último um mestre da Pedagogia. A pesquisa foi feita de modo exploratório e bibliográfico, de caráter qualitativo. Por fim, o estudo pretende deixar claro que a divisão curricular das aulas de Matemática, no ensino básico, em Álgebra e Geometria é apenas um divisão curricular e não deveria afetar a visão de que todos os conteúdos estudados fazem parte de um todo perfeitamente coerente; tendo em vista que, estando diante de um problema de matemática, estudantes e professores podem lançar mão tanto de ferramentas da álgebra quanto de resultados da geometria para resolvê-lo.

The present thesis intercrosses the visual cultures studies and the gender studies, having as object of analysis six encounters which made up the NarrATIVIDADES extension project. Genders Lab and narratives, which happened in November and December 2015, March and April 2017, in addition to the narratives produced by the people who committed themselves to the registered design from the School of Visual Arts (FAV) at the Pro-rector of Extension and Culture (PROEC) from the Federal University of Goiás (UFG). Masculinities in this study are treated as cultural constructs that are supported by the variety of speeches to operate in societies. For this reason, after reports of several visualities are problematized, these constructs of discipline and control that not only relate to the scope of the affective-sexual, but also to the domain of symbolic production. In this sense, this research craved analyze the possibilities and deconstruction strategies of visualities on masculinities and masculinization of looks, this from hypervisuals talks, works with memories and narratives produced by the group of students and teachers on the main subject of research: objectification and women hypersexualization, especially black women and black men; invisibility of the non heteronormative identities and androcentrism in using audiovisuals and media devices. The methodological approach nicknamed and used in this process corresponds to the hypervisual conversations that consist in an expansion of a narrative research expansion from works with visualities and countervisualities. To better comprehend the proper use of the term ‘hypervisuals’ are threshed, throughout the text, discussions around the concept of visual, visual images, the visualities, the countervisualities, the scopic regimes and many other terms of the studies of visual cultures. Since it’s a study linked to the Line of Image Cultural Research of image and Mediation Processes, this research outlines also the use of hypervisual narratives as possible to develop pedagogical processes with deconstructivist bias.
A presente tese entrecruza os estudos das culturas visuais e os estudos das relações dos gêneros, tendo como objeto de análise seis encontros que compuseram o projeto de extensão NarrATIVIDADES. Laboratório de gêneros e narrativas, os quais aconteceram em novembro e dezembro de 2015, março e abril de 2017, além das narrativas produzidas pelas pessoas que se vincularam ao projeto inscrito desde a Faculdade de Artes Visuais (FAV) ante a Pró-reitora de Extensão e Cultura (PROEC) da Universidade Federal de Goiás (UFG). As masculinidades neste estudo são tratadas enquanto construções culturais que têm como suporte os mais variados discursos para operar nas sociedades. Por este motivo, a partir dos relatos sobre múltiplas visualidades são problematizados esses construtos de disciplinamento e controle que não referem só ao âmbito do afetivo-sexual, mas também ao domínio da produção simbólica. Nesse sentido, esta investigação almejou analisar as possibilidades e estratégias de desconstrução das visualidades sobre as masculinidades e a masculinização dos olhares, isto a partir das conversações hipervisuais, trabalhos com memórias e as narrativas produzidas pelo grupo de estudantes e docentes sobre as temáticas principais da pesquisa: objetualização e hipersexualização das mulheres, principalmente das mulheres negras, e dos homens negros; invisibilização das identidades não heteronormativas e androcentrismo no uso de aparelhos e mídias audiovisuais. A abordagem metodológica alcunhada e usada neste processo corresponde às conversações hipervisuais que consistem em uma expansão da pesquisa narrativa a partir de trabalhos com visualidades e contravisualidades. Para compreender o uso do termo hipervisual são trilhadas, ao longo do texto, discussões em torno do conceito do visual, das imagens visuais, das visualidades, das contravisualidades, dos regimes escópicos e tantos outros termos caros aos estudos das culturas visuais. Por se tratar de um estudo vinculado à Linha de Pesquisa Culturas da Imagem e Processos de Mediação, esta investigação delineia, também, a utilização das narrativas hipervisuais como possível forma de desenvolver processos pedagógicos com viés desconstrutivista.

Programas de mudança de comportamento têm proporcionado melhora a saúde por intermédio da adoção de hábitos de vida saudáveis. Atentar-se para a percepção do sujeito tem um papel importante na compreensão da saúde, bem como os benefícios que a mudança de comportamento traz para as suas vidas. Nesse sentido, o objetivo deste estudo é analisar as percepções dos participantes no Programa Vida Ativa Melhorando a Saúde (VAMOS), quanto à promoção da saúde, barreiras e facilitadores. A pesquisa é caracterizada como qualitativa. Para compreender o objeto, este objetivo derivou de um estudo maior, que é o VAMOS, aplicado nos Centros de Saúde em Florianópolis (SC). A amostra foi constituída por todos os participantes do grupo de mudança do comportamento que concluíram o programa em suas duas versões: a primeira em 2012; e a segunda em 2014; e, que participaram do Grupo Focal (GF). Na primeira versão, 39 participantes iniciaram o Programa, desses, 16 participaram do GF. Na segunda versão, participaram 40 pessoas, desses, 19 do GF. Foi aplicada uma ficha diagnóstica aos participantes para formação do baseline, um GF, de onde foram extraídas as categorias sobre a percepção por meio da análise do conteúdo. Os dados sociodemográficos (sexo, estado civil, idade e renda) foram coletados por meio de entrevista face a face e transcritos e armazenados no Epidata. As variáveis qualitativas foram analisadas com auxílio do programa Nvivo 10 for Windows para criar as categorias de análise do conteúdo, para os dados sociodemográficos foi utilizada a análise descritiva desvio padrão e frequência pelo Programa Statistical Package for the Social Sciences (SPSS - versão 17.0®). O VAMOS possibilitou aos participantes a construção de processos cognitivos tanto individuais quanto coletivos. Estes processos cognitivos são fatores importantes para a tomada de atitudes que contribuem na mudança de comportamento, podendo contribuir na prevenção de problemas de saúde ou manutenção de uma vida saudável. A análise da percepção possibilitou uma avaliação sobre o programa, auxiliando as possíveis modificações de suas versões futuras. Assim, acredita-se que os achados da pesquisa possam contribuir na melhoria do programa, especialmente buscando potencializar os facilitadores e minimizar as barreiras para que mudanças de comportamento sejam alcançadas, que é a característica principal do programa.
Abstract : Behavior change programs have provided improved health through the adoption of healthy lifestyle habits. To be attentive to the perception of the subject is an important role for the understanding of health, as well as the benefits that the behavior change brings to their lives. In this sense, the objective of this study is to analyze the perceptions of participants of the Active Life Program Improving Health (VAMOS), as regards to health promotion, barriers and facilitators. The research is characterized as qualitative to better understand the study object, and was derived from a larger study, VAMOS, that is applied in health centers of Florianópolis (SC). The sample was consisted of all participants of the behavior change group that completed the program in their two versions: first in 2012, and second in 2014; and who participated in the Focus Group (FG). In the first version, 39 participants began the Program, of which these, 16 participated of the GF. In the second version, there were 40 people, of which, 19 participated of the GF. Diagnostic record was applied to the participants to form the baseline, one GF, where the categories of the perception were extracted using the content analysis. The socio-demographic data (gender, marital status, age and income) were collected using face-to-face interview, than were transcribed, and stored in Epidata. Qualitative variables were analyzed with the assistance of Nvivo10 program for Windows to create the content analysis categories; to the demographic data were used the descriptive analyzing, standard deviation and frequency, by the Statistical Package Program for Social Sciences (SPSS - version 17.0®). VAMOS enabled the participants to build both individual and collective cognitive processes. These are important factors for taking actions that contribute to the change behavior, and thus contribute to preventing health problems or to maintenance a healthy life. The analysis of the perception enabled an evaluation about the program, helping the possible changes for the future versions. In consequence, it is believed that the research findings can contribute to the improvement of the program, especially looking for enhance the facilitators and minimize barriers to behavior changes are achieved, the main characteristic of the program.

Este estudo aborda numa perspectiva antropológica, a trajetória política e intelectual de Oliveira Silveira considerando um intelectual contemporâneo com uma significativa contribuição para a atualização do pensamento social brasileiro, por meio da elaboração de contranarrativas sobre a presença negra no país. As múltiplas frentes de atuação de Oliveira Silveira impulsionam a refletir sobre os atravessamentos identitários por meio dos quais este se constitui na pessoa-personagem narrada pelos interlocutores dessa pesquisa. Tomando como aporte as discussões que procuram atualizar o campo antropológico em termos teórico-metodológico, este trabalho se inspira na perspectiva de Sahlins (1990), Turner (2005) e Gonçalves (2010), autores que pensam a relação entre ação e estrutura, bem como a relação entre esquemas culturais prévios e as apropriações que os sujeitos fazem deles. Incorpora-se a perspectiva de Said (1993) que discute o papel do intelectual na sociedade contemporânea. Busca-se nos estudos pós-coloniais elementos convergentes para situar Oliveira Silveira como intelectual diaspórico e como portador de uma identidade híbrida representada pela articulação conjunta de sua identidade regional e de sua identidade negra. O estudo procura descrever os deslocamentos vivenciados por Oliveira Silveira tanto no sentido geográfico como sentido simbólico. Este foi o proponente do Vinte de Novembro como data de maior significado para a história negra do país, sendo também atuante em organizações negras, tais como: clubes sociais negros, escolas de samba, congadas. Além disso, organizou vários grupos de ativismo político na capital, o primeiro deles, o Grupo Palmares e depois o Grupo Semba, Razão Negra, Revista Tição e Associação Negra de Cultura. Todas as organizações lideradas por Oliveira Silveira se destinavam, simultaneamente, à luta política e à promoção da cultura negra. Essa dissertação se propõe a apresentar o percurso etnográfico percorrido para apreender como diferentes pessoas, que conviveram com Oliveira Silveira em diferentes momentos compreenderam, significaram e valorizaram a trajetória deste intelectual e ativista. A partir da proposta de etnografia multisituada de George Marcus (1994), a pesquisa procurou identificar a forma como cada interlocutor(a) elaborou sua percepção e interpretação a respeito da trajetória de Oliveira Silveira, convertendo-as em narrativas.
The presuppositions of this study address the political and intellectual trajectory of Oliveira Silveira based on an anthropological perspective. This study takes into consideration that he is a contemporary intellectual that provides a meaningful contribution to the update of the Brazilian social thought, through the presentation of counter-narratives related to the black presence in the country. The many places that Oliveira Silveira occupies stimulated the reflection about the identity crossings from which he is constituted in the person-character, narrated by his interlocutors.Taking as a guide to the discussions that focus on updating the anthropological field in theoretical-methodological terms, it was adopted in this study the perspectives of Sahlins (1990), Tunner (2005) e Gonçalves (2010), authors that think the relation between action and structure, as well as the place of the individual in the cultural life. In this study, it was incorporated the perspective of Said (1993) who discusses the role of the contemporary intellectual. Studies on convergent elements of the post-colonial period were also consulted in order to place Oliveira Silveira as a diasporic intellectual with a hybrid identity represented though the simultaneous articulation of his regional and black identity. In this study, it is intended to describe this displacement lived by Oliveira Silveira in the physical as well as in the symbolic way. This was the proponent of November twentieth as the date of greatest meaning to the black history of the country, being also active in black organizations, such as: clubs, samba schools and congadas. Besides, he has organized many groups in the capital, the first of them, the group Palmares and after the group Semba, Razão Negra, Revista Tição and Associação Negra de Cultura. All these organizations were lead by Oliveira Silveira and aimed at the political fight and to the promotion of the black culture. These dissertations aim at showing how different people, at different moments, understood, made sense and valued the intellectual trajectory of this activist. The choice of the topic of this study brings, a priori in an implicit way, the observation of the meaningful role played by Oliveira Silveira. This research aimed at identifying how each one chose to narrate their perception and comprehension related to the trajectory of Oliveira Silveira.

Resumo: O cenario de producao em campos de petroleo tem apresentado nos anos recentes condicoes cada vez mais severas e complexas. Muitas descobertas em regioes de maior profundidade e com formacoes geologicas mais diversas tem demandado solucoes tecnológicas que beiram a fronteira do conhecimento, de forma que a producao tem andado lado a lado com o desenvolvimento de novas tecnologias. Assim, a necessidade do aumento do fator de recuperação para justificar os investimentos nestas condicoes sao desafios presentes nos novos projetos de pocos, de elevacao artificial e de recuperacao secundaria, o que vem levando ao desenvolvimento de solucoes submarinas e integradas, como o caso do separador submarino VASPS. No entanto, o controle de processos continuos, como a vazao de producao, a temperatura e a pressao, ainda e tratado da mesma forma que era feito quando do comeco da producao offshore em campos de oleo leve. Este trabalho visa permitir a melhoria de desempenho operacional do sistema VASPS, mensurada pelos custos totais gerados pelo sistema e pelos proventos gerados na producao de oleo e gas, bem como propor novas metodologias de sistemas de controle para sistemas similares, utilizando tecnologias recentes de controle inteligente. Vale ressaltar que o problema de controle e atualmente tratado em campo com solucoes de automacao, sem o estudo devido, causando danos a equipamentos que exigem intervencoes custosas. Os controladores propostos neste trabalho visam minimizar os esforcos aos quais estes atuadores estao expostos, provendo uma maior vida util as tecnologias de elevacao artificial de alta vazão e podendo ser aplicado de forma modular a outros processos relacionados.
Abstract: The oil production scenario has been becoming more and more complex and severe in the recent years. Plenty discoveries in deeper and more diversified formations have been requiring cutting edge technological solutions., such that the production has been deployed side by side with the technological development. Thus, the need for the enhancement of the recuperation factor in order to justify the investment in these new conditions are actual challenges within the recent design of wells, artificial lift systems and secondary recuperation projects, which has been leading to the development towards subsea integrated solutions, such as the VASPS subsea separator. However, the control of continuous processes, i.e. flow, temperature and pressure, is still dealt the same way it was done when the offshore production in light oil fields began. This work envisions the enhancement of the operational performance of the VASPS system, measured as a function of the total costs implied by the technology, altogether with the receipts generated in the gas and oil production, as well as the delivering of new methodologies for the design of controllers using late technologies of intelligent control. Also, it is worth recalling that nowadays the control task is dealt with automation solutions, without the proper handling, causing failures in equipments that require expensive rig intervention for workover. The controllers proposed in this work are designed to minimize the control effort, to which these actuators are exposed, providing extended lifetime to the high level production artificial lift technologies, being also able to be applied to other related processes in a modular form.

The Cordel Literature, also known as string literature , constitutes one of the traditional poetical verses of Northeastern Brazil, characterized by the written production and specific typographic records. The communicational processes occupy, hence, a privileged position. In the last two decades, the cordelists have started performing through poetic contests by means of e-mail or chat communities on social networks, publishing what is called virtual battles , battles based on the cordel s imaginary battles, which can already be seen in titles of the 20th century. Thus, these written contests tradition harks back to the poetic challenges between two improvisers who would perform before an enthralled audience. The contests of cordelists and improvisers safe guard several codes and simultaneously interweave poetical elements which have been updated for centuries. Combining fixed forms, rhythm, themes, verbal duels, be they improvised or not, are recurrent in the poetry of cordelists, guitar players, coquistas, cirandeiros, maracatu masters, carnival ox, caboclinhos, rural carnival parades, samba of the countryside man. Face to face challenges, printed challenges, web-mediated challenges pave the way for the articulation of these expressions into a great poetical, oral text going through a continuous process in updating the virtual matrices. The research corpus harbors battle booklets (virtual and fictional); face-to-face challenges; poetry performing events; recorded virtual battles that took place on the web, cds and dvds by improvisers and cordelists. Participants observation, interviews and qualitative analysis of large texts that have been utilized in virtual contests (mediated by twitter, msn, facebook, orkut, sites, portals, email, and so forth) are in the methodology covering the fictional battles of cordelists and improvisers. Pursuant to the interdisciplinary concepts of communication by Jesús Martín-Barbero; tradition, performance and movement by Paul Zumthor; great oral text and virtual matrices by Jerusa Pires Ferreira; cyber-culture by Francisco Rüdiger, I propose to carry out a comparative study on the production of improvisers and cordelists spanning mainly the decades from 1990/2000. The purpose of the study is to investigate the existence of memories confluence and combination in the articulation of this large communication text in an ever adapting process, and also to speculate to what extent one can define such poetry as a recognizable cultural series
A literatura de cordel se constitui numa das poéticas tradicionais do Nordeste do Brasil, caracterizada por produção escrita e registro tipográfico específicos. Os processos comunicacionais ocupam, aí, posição privilegiada. Nas duas últimas décadas, os cordelistas passam a realizar disputa poética via e-mail ou bate-papo nas redes sociais, publicando o que denominam pelejas virtuais, calcadas nas pelejas imaginárias de cordel, que aparecem, já, em títulos do início do século XX. Por sua vez, a tradição dessas pelejas escritas remete ao desafio poético entre dois improvisadores, diante de uma platéia atenta. As pelejas de cordelistas e repentistas guardam variados códigos e simultaneamente entrelaçam elementos poéticos que se atualizam há séculos. Combinando formas fixas, ritmo, temas, os duelos verbais, de improviso ou não, são recorrentes na poesia de cordelistas, violeiros, coquistas, cirandeiros, mestres de maracatu, boi de carnaval, caboclinhos, blocos rurais de carnaval, samba de matuto. Em desafios ao vivo, desafios impressos, desafios mediados pela web, é possível articular essas expressões poéticas como um grande texto oral em contínuo processo de atualização de matrizes virtuais. No corpus da pesquisa, folhetos de pelejas (virtuais e fictícias); desafios ao vivo; eventos com performance de poesia; registro de pelejas virtuais ocorridas na web; cds e dvds de cordelistas e repentistas. Na metodologia, a observação participante, entrevistas e análise qualitativa do grande texto que se realiza nas pelejas virtuais (mediadas por twitter, msn, facebook, orkut, sites, portais, correio eletrônico etc.), nas fictícias pelejas de cordelistas e nas de improvisadores. Respaldada nos conceitos interdisciplinares de comunicação, de Jesús Martín-Barbero; tradição, performance e movência, de Paul Zumthor; grande texto oral e matrizes virtuais, de Jerusa Pires Ferreira; cibercultura, de Francisco Rüdiger, proponho-me realizar um estudo comparativo da produção de pelejas de cordelistas e repentistas, abrangendo sobretudo as décadas 1990/2000. Full textO maracatu cearense é uma manifestação cultural que representa um cortejo em homenagem aos Reis Negros. Esta dissertação pretende compreender como um bem cultural que exalta os negros se inseriu num Estado que disseminou uma ideologia de embranquecimento, considerando os negros ausentes da sua formação étnica / cultural. Pretende, ainda, apresentar o universo de dois grupos: o maracatu Rei de Paus e o Nação Baobab, ao detalhar as formas como seus brincantes dialogam com essa ideologia e qual a sua influência para a manifestação estudada. A dissertação reforça discussões sobre tradição, memória, identidade e cultura popular, através das interpretações dos sujeitos. Ela demonstra que é possível problematizar questões como políticas públicas e religião através de um bem cultural . Propõe a particularização dos maracatus, pois, a maioria dos estudos e pesquisas realizadas têm homogeneizado essa manifestação popular. Cada maracatu é um universo complexo precisando ser analisado no contexto em que está inserido. Nesse sentido, o maracatu do Ceará incorpora e ao mesmo tempo se contrapõe à ideologia do embranquecimento. Existe um jogo de identidades: a elite reforça a ausência dos negros, enquanto o cortejo coroa uma rainha negra, mesmo tendo o rosto pintado de preto, ela representa a realeza africana. O cortejo em seu conjunto resgata a história dos negros, suas alas representam personagens como: o casal de preto velho, os africanos, dentre outros

Full textIn the face of the existence of an endocannabinoid system in humans, possibly dysfunctional in anxiety disorders and the growing interest in the therapeutic use of cannabidiol (CBD), justified a study comparing plasma concentrations of CBD, administered orally in powder form and dissolved in oil, in ordre to define the optimal mode of administration of the CBD and their therapeutic efficacy at a dose of 150mg, based on dynamic and kinetic studies (PK/PD). In this contexto, the study researched 39 healthy volunteers, allocated randomly in 3 groups, receiving 150 mg of CBD, capsules in powder form (Group 1), dissolved in corn oil (Group 2) and corn starch (placebo group). The plasmatic concentration of CBD was determined by gas chromatography coupled to mass spectrometry at the times: 0, 0.5, 1.0, 1.5, 2.0, 2.5, 3.0, 4.0 h, after administration of the drug. Assessment instruments of psychological measures were: analogical scale mood (VAMS) and scale identification of trait anxiety state (IDATE) and physiological measures were monitored in arterial pressure (PA) and heart rate (HR). The anxiogenic test applied was Simulated Public Speaking (TSFP). In recognition of facial expressions (REF) was performed the task touch, that consists in indicate the emotion shown the facial expression of the individual. The results of subjective measures (VAMS and IDATE) was not found the anxiolytic effect; there was increase of systolic and diastolic pressure and heart frequency; the response intensity in the REF was 93% in group 1 (powder) and 81% in Group 2 (oil). The peak mean plasma was 20.70 ± 1.68 ng / mL (CBD powder) in 2.5 h and 82.59 ± 9.52 ng / mL (CBD in oil) in 2h. Thus, this study PK / PD demonstrated that the ideal pharmaceutical form for administration of the CBD has been in oil and study of dose / response, 150mg has no therapeutic efficacy in relation to anxiety.

Através de uma abordagem feita por viés histórico, estético, identitário e linguístico, esta dissertação apresenta uma reflexão sobre a proposição antropofágica elaborada por Oswald de Andrade, na sua relação com os discursos históricos, e com os discursos nacionalistas dos anos 30 no Brasil. A mesma demonstra como Oswald de Andrade constrói uma narrativa sobre a nacionalidade na qual a relação entre a identidade étnica e a nacional estão constante conflito. Partindo desse fato, a dissertação localiza duas maneiras pelas quais a antropofagia oswaldiana vem sendo narrada: uma que a territorializa no campo dos discursos nacionalistas, outras que tentam lhe retirar desse espaço, dando-lhe novos significados e potencialidades de transformação estética e sociocultural. Full textThe immune response against a broad spectrum of pathogens or against tumor cells requires the CD4 + T lymphocytes activation. The triggering of T Cell Receptor (TCR) by peptide-MHC through antigen-presenting cells (APC) leads to numerous T-cell remodeling and the establishment of a specialized structure at the interface between the T lymphocyte and the APC: the immunological synapse. The synapse is the site of intense signaling events where various signaling molecules are recruited in order to amplify and diversify the signal initiated by the TCR. These signaling events are regulated by the transmembrane adapter LAT ("Linker for activation of T cells") which is present at the plasma membrane as well as in intracellular compartments. The intracellular fraction of LAT is recruited at the immune synapse and it is proposed that these vesicular compartments participate in T lymphocyte signaling. The objective of this thesis work was to determine the intracellular trafficking pathways required for the recruitment of the intracellular pool of LAT to the immunological synapse and understand the role of this transport in T cell activation and response. By silencing the expression of different intracellular transport molecules in Jurkat T cells or primary human CD4 + T lymphocytes, or by using Knock-Out (KO) mice, we have highlighted several trafficking pathways involved in the transport of LAT to the immune synapse. We have demonstrated that the recruitment of LAT to TCR activation sites requires a secretion pathway dependent on the vesicular SNARE protein VAMP7. Further analysis of the transport of LAT showed that LAT is present in recycling compartments whereas VAMP7 is mainly located in the Golgi apparatus. The study of the small GTPase Rab6 and the t-SNARE syntaxin-16 protein that are involved in the retrograde transport pathways between the recycling endosomes and the Golgi apparatus, demonstrated that this route of transport is required for the recruitment of LAT to the immunological synapse and for T lymphocyte response in vitro, ex vivo and in vivo as well. Finally, the role of intraflagellar transport protein IFT20, which has already been implicated in the transport of TCR, was analyzed in mice and also showed defects in LAT recruitment to synapse and T lymphocyte activation ex vivo and in vivo. Our results thus show that the regulation of intracellular transport plays a crucial role in T lymphocyte activation. We thus propose a model in which LAT is constitutively internalized from the plasma membrane and pursues a Golgi-dependent recycling pathway that contains the secretion machinery associated with VAMP7. This intracellular transport pathway would thus allow the polarized LAT re-secretion to the immunological synapse under activation conditions and a robust T lymphocyte response

Full textPulmonary infections in young foals by Rhodococcus equi result in severe pneumonia, leading to death a large number of animals every year. There is no globally accepted vaccine for the prevention of equine rhodococosis so far. To date, the only acceptable treatment is based on antibiotics, but the clinical protocols are long lasting, expensive, having side effects and favoring the emergence of drug resistant strains. Several strategies for developing a safe and effective vaccine against rhodococosis have been proposed, however, none has induced a significant protective effect. The main virulence factor of R. equi described and extensively studied is the protein vapA, although other proteins encoded by genes in pathogenicity islands are detected in samples of virulent R. equi, isolated from infected animals. Recent works have demonstrated that the vapG gene is present in all virulent strains, and that this gene is highly expressed when bacteria reside within macrophages, making it a potential vaccine target. Our group has already been working with an attenuated Salmonella enterica Typhimurium strain expressing the VapA protein. Even though our results were very promising, we decided to construct a second vaccine strain expressing the VapG protein. Based on the positive results, it was constructed an attenuated strain of S. enterica Typhimurium 3987 expressing the vapG protein. Interestingly, the VapG-expressing strain induced protection against virulent R. equi in mouse model of infection. We could observe colonization and persistence of Salmonella vaccine cells in target organs, with reduction of bacterial loads of R. equi and induction of a protective immune profile similar to that seen in resistant adult animals. We could also observe increased production of IL-12p70 and IFN- in addition to the presence of increased levels of IL-4 and reduced levels of TNF-. Moreover, we detected an increase in the subpopulation of T helper cells (CD4), with a profile of memory, as well as in the population of B lymphocytes, when compared to control groups. This set of results shows that immunization with Salmonella enterica Typhimurium expressing the VapG protein raises an efficient cellular immune response, making this strain a potential candidate for vaccine vector. Furthermore, this work suggests that other R. equi antigens may be taken into account for vaccine construction, besides the VapA protein utilized in the majority of studies.

Full textShared economy is an economic system based on the share of underutilized goods free of charge or for a fee. With the advent of the internet, the digital platforms based on this concept have been developed, emerging the need of understanding its relationship with two-side markets. This expansion has reached various sectors of the economy, as well as created new demand, such as the Uber platform, which ,however, has generated disturbances in the taxi sector, traditionally regulated with its worldwide recognised inefficiencies. Thus, the goal of this dissertation is to understand what the digital platforms are and to analyse the specific case of Uber company, showing the main factors which allowed this type of business to arise; where we are, defining what Uber effectively is and the services provided by it, addressing the welfare gains provided (consumer surplus) and regulatory problems faced; and where we go, prospecting a path for the shared economy regarding the digital platforms in two-side markets.

Este trabalho objetiva, através de uma análise de interface entre gramática e discurso, verificar como se operam as construções apositivas, cuja unidade base seja constituída do elemento linguístico o seguinte, em interações argumentativas, mais especificamente, em audiências de conciliação do PROCON/JF. Tais construções se apresentaram com notável frequência em nossos dados, revelandose uma importante estrutura de interação interpessoal modelada por características particulares. No plano interacional argumentativo, o falante se utiliza deste tipo de construção apositiva para: (i) introduzir seu ponto de vista; (ii) sustentar o ponto de vista; (iii) apresentar um acordo; e (iv) sugerir um acordo para as partes envolvidas na audiência. Já o elemento linguístico o seguinte, presente na unidade base da construção apositiva, apresenta comportamento semelhante a um marcador discursivo, desempenhando duas funções na organização do discurso: a textual (direcionada para a estrutura do texto, para o seu “amarramento”) e a interacional (relacionada aos interlocutores, aos participantes da interação). O seguinte comporta-se sintaticamente como: (i) argumento de um verbo; (ii) conjunção integrante; e (iii) conjunção explicativa. Cabe destacar que as ocorrências deste tipo de construção apositiva são todas relevantes nessa interação investigada, principalmente pela peculiaridade do elemento híbrido o seguinte que permite ao falante sinalizar ao seu interlocutor a relevância do que será dito a seguir.
This paper aims, through an analysis of the interface between grammar and discourse, see how to operate the apposition buildings, whose basic unit consists of the linguistic element o seguinte in argumentative interactions, more specifically, in conciliation hearings of PROCON / JF. Such constructions are presented with remarkable frequency in our data, revealing an important structure of interpersonal interaction modeled by particular characteristics. On the interaction of argument, the speaker uses this type of construction for patching: (i) introduce their point of view, (ii) support the view, (iii) make an agreement, and (iv) to suggest an agreement for the parties at the hearing. However the linguistic element o seguinte, that is present in the base unit of apposition buildings, behaves like a discourse marker, playing two roles in the organization of discourse: the text (directed to the structure of the text, for its "tethering") and the interactional (related to partners, the participants of the interaction). O seguinte behaves syntactically as: (i) argument of a verb, (ii) integral conjunction, and (iii) explanatory conjunction. It should be noted that occurrences of apposition constructions are all relevant in this interaction investigated, mainly by the peculiarity of the hybrid element o seguinte that allows the speaker to signal the interlocutor the relevance of what will be said below.

The structures fictionists and the forms of the wonderful story, presents in the interactive narratives, propitiate a rescue and actualization of the contemporary production in the perspective of the collective creation of the plot and in the characters\' elaboration in the flow of fictions. It was used as a corpus of discussions in the forums of the gender, players\' registrations on happy cases and unhappy results in the use of the rules of the grammar of the fiction. The practice that turns the reader a careful manipulator of the fiction, subject and creator of narratives, it can be observed in the experience with authorship and education, accomplished at a public school. Finally, the interactive narratives are an indicative of the creation need that the human being possesses where, when telling their stories, they not just denounce the literary structures and fictionists in game, but a skilled reader\'s presence in the manipulation of the cultural universe that surrounds it, whose elementary rules are the one of the authorship.

Este estudo narrativo investigou a experiência de construção de um planejamento de aulas de inglês, delineado a partir das vozes de alunos do primeiro ano do ensino médio de uma escola da rede pública estadual de Minas Gerais. Por um período de dois meses, a professora participante e eu construímos um planejamento de aulas de inglês que buscou se aproximar de uma perspectiva interdisciplinar de ensino e escrevemos sobre esta experiência em diários, relatos e notas de campo. A partir do estudo deste material documentário, sob a perspectiva teórico-metodológica da pesquisa narrativa, segundo Connelly e Clandinin (2000), foi possível recontar nossas histórias com planejamentos e destacar alguns aspectos emergentes de nossa experiência. Ainda, compreender como estes aspectos figuraram na imagem do planejamento elaborado e reconfiguraram alguns paradigmas de planejamento de aulas de inglês da professora participante e meus. Algumas vertentes teóricas, ligadas a conceitos e práticas curriculares e interdisciplinares, são apresentadas e discutidas de modo a problematizar o foco em questão neste estudo. Perspectivas curriculares, segundo autores como Schawb (1962), Dewey (1938; 1976), Connelly e Clandinin (1988; 1995), Mello (2005) e, ainda, perspectivas interdisciplinares, segundo Japiassú (1976), Fazenda (1978; 1997) e Freire (1987; 1996) são destaques neste trabalho. Dentre os aspectos emergentes da experiência, que deram forma à imagem do planejamento das aulas de inglês, destacaram-se: o entusiasmo, as surpresas, insatisfações, a esperança, os aspectos responsorial e relacional do planejamento, a insegurança de aproximar língua inglesa e interdisciplinaridade e os desafios de se planejar aulas. Este estudo se encontra na área de ensino e formação de professores de línguas estrangeiras da Linguística Aplicada, poderá colaborar com discussões neste campo e, possivelmente, com a prática de professores de inglês como língua estrangeira.

Full textNão sei dizer se este trabalho é a história que vou contar, ou se esta história é o trabalho que vou apresentar. As duas coisas estão muito imbricadas para sofrerem esta distinção. Tenho como objetivo narrar e analisar a tentativa de uma experiência de aprendizagem em prática de Tandem via MSN Messenger em uma escola pública da região do Triângulo Mineiro, com base nos pressupostos da Pesquisa Narrativa, de acordo com Connelly; Clandinin, 2000, 2004, 2005) e Ely (2007). Relaciono, ao longo deste relato, principalmente, estudos sobre, e experiências com, prática de tandem, de acordo com Migliorança (2007), Brammerts e Calvert (2003), Telles; Vassallo (2005) e Telles (2006); ensino e aprendizagem de línguas e novas tecnologias, segundo Warschauer (1996, 1997), Moita Lopes (2005) e Paiva (2006); e linguagem e poder, com base em Fairclough (1989, 2009). As histórias vividas durante a pesquisa foram analisadas pela composição de sentidos, com base nos estudos realizados por Ely; Vinz; Downing; Anzul (2001). Pude perceber que as relações de poder presentes no campo de pesquisa, incluindo pressuposições que fiz sobre o uso das novas tecnologias, tiveram influência no encaminhamento da pesquisa, resultando no não acontecimento da prática de tandem conforme inicialmente planejado e na mudança de perspectiva de análise após a qualificação. Apesar dos (im)previstos que a Pesquisa Narrativa sempre prevê -, a tecnologia exerceu um papel significativo para os participantes na descoberta de recursos para aprendizagem de língua Inglesa e no início de um possível processo de mudança na prática da professora participante. Além disso, a tecnologia possibilitou aos alunos participantes o contato com alunos de uma escola estrangeira e momentos de tandem cultural, o que pareceu ter um possível impacto na aprendizagem dos alunos em relação à motivação, à autonomia e à compreensão de aspectos culturais.

Full textThe hormonal secretion plays a key role in the maintenance of homeostasis. For example, the maintenance of normoglycaemia requires insulin exocytosis from the pancreatic beta cells. The SNARE membrane family protein has been described as the core machinery of fusion between the vesicle containing hormones and the plasma membrane. This family consists of 3 different membrane proteins that are essential during exocytosis. VAMP2 is localized on the vesicle and Syntaxin 1A - on the plasma membrane. They both are transmembrane protein whereas SNAP25 is linked to the plasma membrane by palmitoylation. The SNAREs appear to be essential as they form the cytosolic SNARE complex to dock the vesicle to the plasma membrane. Even though the role of this cytosolic domain has been studied in depth, much less is known on the role of their transmembrane domain during the fusion. Their study remains necessary to establish a complete model of membrane fusion mediated by the SNARE proteins.Here, we have studied the behavior and the role of the SNARE transmembrane domain during exocytosis. In a multidisciplinary project, we have combined a structural approach with a biological study to evaluate the role of this domain. Using mutagenesis in the transmembrane domain of VAMP2 and a cellular system with a clean background, we have assessed the effect of mutations on the secretion and exocytosis in two different cell lines (INS1E and PC12). The biological system is based on the silencing of endogenous VAMP2 and reconstitution of the expression of VAMP2 wt or mutated in the transmembrane domain. Using biochemistry assay and TIRF microscopy we have shown that mutations in this domain can lead to a missorting of the Golgi apparatus or a reduction of the stimulated secretion and exocytosis. This effect can be correlated to a modification of the structural dynamics of this domain.The obtained results clearly demonstrate the role of the transmembrane domain of VAMP2 during exocytosis probably sustained by its unique structural dynamics observed by physico-chemistry

Resumo: O sistema de separação submarina VASPS (Vertical Annular Separation and Pumping System), que se utiliza tanto do efeito gravitacional quanto do centrífugo é composta por Câmara de Expansão, Helicóide de Separação e Piscina. Com o objetivo de se estudar um separador compacto e de menor custo, foi construído um protótipo da Câmara de Expansão em acrílico. Foram realizados testes de eficiência de separação do protótipo utilizando a mistura ar-água e a mistura ar-glicerina com diferentes viscosidades. Para realizar os testes de eficiência foi construído um circuito fechado todo equipado e instrumentado. Nestes testes de eficiência variou-se o diâmetro de bocal de entrada do separador e o nível de líquido na parte inferior do separador. Foi observado durante os testes experimentais a formação de um vórtice, principalmente para níveis mais baixos de líquido, diminuindo a eficiência do separador. Para solucionar esse problema foi desenvolvido e fabricado um dispositivo chamado Redutor de Vórtice, melhorando a eficiência do protótipo para certas condições de operação. Os resultados mostraram que a eficiência diminui para fluidos mais viscosos e também observou-se que o padrão de escoamento no bocal de entrada do separador teve influencia na eficiência. Resultados do ângulo de inclinação e altura de elevação do filme de líquido na parede do separador também foram obtidos
Abstract: The VASPS (Vertical Annular Separation and Pumping System) is a subsea separation system, which utilizes both the gravitational effect as the centrifugal and is composed by Expansion Chamber, Helicoide Separation and Pool. In order, to study a compact separator and with lower cost, we built a prototype of Expansion Chamber in acrylic. Separation efficiency of the prototype using the air-water mixture and air-glycerin mixture with different viscosities tests were performed. To perform efficiency tests was built a closed circuit all equipped and instrumented. In these tests of efficiency ranged the diameter of the separator inlet nozzle and the liquid level at the bottom of the separator. The formation of a vortex, especially for lower levels of liquid was observed during the experimental tests, decreasing the efficiency of the separator. To solve this problem was developed and manufactured a device called Vortex Reducer, improving the efficiency of the prototype for certain operating conditions. The results showed that the efficiency decreases for more viscous fluids and also observed that the flow pattern at the nozzle inlet separator had an influence in the efficiency. Results of tilt angle and lift height of the liquid film on the wall of the separator were also obtained

A metodologia escolhida fundamenta-se na metanálise de dados do Censo de 2010, referentes ao perfil religioso dos/das adolescentes brasileiros e na pesquisa etnográfica virtual, através do monitoramento de Páginas Comunitárias do Facebook ligadas ao incentivo e extinção do comportamento autolesivo e suicidário entre adolescentes brasileiros (as) de 13-17 anos de idade. A estrutura foi dividida em quatro capítulos distintos. O primeiro apresenta o indivíduo, na fase da adolescência, a partir do modelo bioecológico de Bronfenbrenner e como este se insere nos estágios da fé e o Deus Google. O segundo verifica a percepção de morte e morrer do/da adolescente virtualizado (a), bem como sua interação com as Páginas Comunitárias do Facebook associadas ao incentivo e extinção do comportamento autolesivo. O terceiro analisa a importância do Ensino Religioso como componente curricular compatível ao desenvolvimento do tema da morte e do morrer no espaço escolar através da Pedagogia Cemiterial. Por fim, o último capítulo propõe um modelo de ação para a aplicação da Pedagogia Cemiterial com adolescentes no espaço escolar.
The goal of this dissertation is to present the Cemeterial Pedagogy as a proposal for action to be developed in primary education. The methodology chosen is founded on the meta-analysis of data from the 2010 Census, regarding the religious profile of Brazilian adolescents and on the virtual ethnographic research, through the monitoring of Community Facebook pages connected to the encouragement and extinction of self-injurious and suicidal behavior among Brazilian adolescents ages 13 to 17. The structure was divided into four distinct chapters. The first presents the individual, in the adolescent phase, based on the bio-ecological model of Bronfenbrenner and how this adolescent inserts him/herself in the faith stages and the Google God. The second verifies the perception of death and dying of the virtualized adolescent as well as his or her interaction with Community Facebook pages associated to the encouragement and extinction of self-injurious behavior. The third analyzes the importance of Religious Education as a curricular component that is compatible to the development of the theme of death and dying in the school space through the Cemeterial Pedagogy. Finally, the last chapter proposes a model of action for the application of the Cemeterial Pedagogy with adolescents in the school space.

Although advances have been noticed, because it is a process which needs conceptual, legal, and practice changes, brings itself some doubts, because of existent contradictions between the discourses that proclaim the inclusion and the Brazilian educational reality. Under the vision of the teachers, in the context of the inclusive school, there are several difficulties that the teachers are facing, in practice, with these students. Considering this, despite of the existing production in this field of research, it is necessary to go deeper into the research of this theme from the point of view of the teacher. So, this research investigates the voice and the optics of the teachers of regular classes that have students in situation of inclusion, more specifically the disable ones and deals with different aspects of the inclusion in the school quotidian and the support to the teacher as one of the necessary actions for the inclusive pedagogic practice highlighting the theorists that discourse the inclusive process. The study, of a qualitative character, was realized in four municipal Elementary schools of São Bernardo do Campo with the participation of nine teachers, four principals and four families of disabled children. Considering the analyze of the observed aspects, we can say that the teachers are searching for a re-signification of their practices, however, they need a constant support to deal with the diversity of these students.(AU)
A inclusão de alunos com deficiência em ensino regular no Brasil é um movimento que vem denotando constante crescimento nos últimos anos. Embora muitos avanços sejam observados, por ser um processo que suscita mudanças conceituais, legais e de prática, traz consigo dúvidas, decorrentes das contradições existentes entre os discursos que proclamam a inclusão e a realidade educacional brasileira. Sob o prisma da docência, no contexto da escola inclusiva, são inúmeras as dificuldades que os professores vêm enfrentando na prática com esse alunado. Considerando essa problemática, apesar da produção já existente, faz-se necessário o aprofundamento de estudos deste tema do ponto de vista do professor. Portanto, esta pesquisa investiga a voz e a ótica dos professores das classes regulares que possuem alunos em situação de inclusão, mais especificamente os que têm deficiência, e aborda os diferentes aspectos da inclusão no cotidiano escolar e o apoio ao professor como uma das ações necessárias para a prática pedagógica inclusiva à luz de teóricos que discutem o processo inclusivo. O estudo, de caráter qualitativo, foi realizado em quatro escolas do Ensino Fundamental (séries iniciais) da rede pública municipal de São Bernardo do Campo, e contou com a participação de nove professores, quatro diretores e quatro famílias de alunos com deficiência. Considerando a análise dos aspectos observados, pode-se inferir que os professores têm buscado a ressignificação de suas práticas, mas desvelam a necessidade de um apoio constante, que lhes dê sustentação para o atendimento à diversidade dos seus alunos.(AU)

Full textO presente relatório final de estágio pretende apresentar o projeto de intervenção-investigação desenvolvido numa perspetiva da educação para a sustentabilidade. O projeto de intervenção-investigação pretendia trabalhar a temática do peixe e os benefícios do consumo do mesmo para as crianças, constituindo-se por uma sequência didática que envolveu o trabalho sobre questões relacionadas com diferentes espécies de peixe que existem. O projeto de intervenção-investigação implementado teve como principais objetivos: i) avaliar o impacte da sequência didática nas aprendizagens das crianças ao nível dos conhecimentos, capacidades e atitudes e valores e ii) avaliar o impacte da sequência didática no desenvolvimento profissional da professora investigadora que a concebeu, implementou e avaliou. O projeto de intervenção-investigação teve como suporte os seguintes pilares teóricos: i)Educação para o Desenvolvimento Sustentável; ii) Literacia Científica; iii) Educação em Ciências nos primeiros anos; iv) Orientação Ciência, Tecnologia e Sociedade e v) IBSE/EPP. O mesmo projeto teve caraterísticas de investigação-ação, em que se utilizaram como técnicas de recolha de dados a observação, o inquérito por entrevista e a compilação documental. Na análise dos dados recorreu-se à análise de conteúdo com suporte ao software WebQDA. O projeto contribuiu para o desenvolvimento de aprendizagens das crianças ao nível dos conhecimentos, como “Conhece algumas espécies de peixe”; capacidades como “Observa”; e atitudes e valores como “Revela gosto pelo consumo de peixe”; tal como para a promoção do desenvolvimento profissional da professora investigadora, o que se evidencia nos resultados.
This final stage of report aims to present the project of intervention-research developed a perspective of education for sustainability. The project intervention-research intended to work the theme of fish and the benefits of its consumption for children, being by a didactic sequence that involved working on issues related to different species of fish that exist. The implemented research-intervention project was primarily to: i) assess the impact of the didactic sequence in the learning of children in terms of knowledge, skills and attitudes and values and ii) assess the impact of the didactic sequence in the professional development of researcher teacher who She conceived, implemented and evaluated. The action-research project was supported by the following theoretical pillars: i) Education for Sustainable Development; ii) Scientific Literacy; iii) Science Education in the early years; iv) Guidance Science, Technology and Society v) IBSE / EPP. The same project was research-action features, which were used as techniques for data collection observation, interview survey and documentary compilation. In analyzing the data resorted to content analysis with support WebQDA software. The project contributed to the development of learning of children at the level of knowledge as "Know some species of fish"; capabilities as "Notes"; and attitudes and values as "reveals taste for fish consumption"; as to promote the professional development of teacher researcher, as evidenced in the results. The results show that the project was constituted as a stimulus to science education from the early years, revealing the possibility and similar oriented project development relevance of different themes for the formation of more responsible and conscious citizens in promoting development sustainable.
