Amyotrophic Lateral Sclerosis (ALS) is a fatal, late-onset, progressive neurodegenerative disease. A familial form of ALS, autosomal dominant ALS8, is characterized by a mutation in an ER membrane protein, VAPB. To characterize the role of VAPB in motor neurons, two C. elegans models were generated: one expressing human VAPB-P56S and another with the knockdown of C. elegans VAPB ortholog, VPR-1. Liver disease is the fifth largest killer in the United Kingdom and with the numbers of diagnoses increasing each year there is an urgent need for novel therapeutic interventions. This thesis examines one potential target, Vascular Adhesion Protein (VAP)-1: an amine oxidase enzyme with reported adhesin functionality. They are conserved from flies to mammals, where they consist of three family members: Mena, VASP and Evl. Valosin containing protein (VCP)/p97 is a hexameric ATPase of the AAA family, which regulates a wide array of essential cellular processes. Dominant mutations in the N-domain of the VCP give rise to the complex disease syndrome known as Inclusion body myopathy with Paget disease of the bone and frontotemporal dementia (IBMPFD). VCP plays a key role in the ubiquitin-proteasome dependent protein degradation although mutations in VCP seem to result in a late stage autophagy defect. A mutation in ER membrane protein VAPB is responsible for causing a familial form of ALS (ALS8). The VAPB-P56S mutation causes protein aggregation and a nuclear envelope defect, where retrograde transport is disrupted. Over-expression of a FFAT peptide from OSBP1 reduces the size of VAPB-P56S aggregates and restores retrograde transport.
