Abstract:
The form of the second movement of the String Quartet in A major, Op. 7 is original not only in Dohnányi’s oeuvre but in music history in general, as well. This new musical idea is a fusion of two traditional forms: a variation and a ternary form. The theme of the movement is followed by four variations, but at the end of the second one there is an unexpected break: a contrasting Trio-like section comes in between and the flow of variations continues only after it finishes. This unique structure is analysed in detail for the first time in present study. Relying on analyses of Dohnányi’s compositions, this study traces similar formal characteristics in other works of the composer too. Finally, the article provides with an example for this hybrid form in one of its three closest relatives: the second movement of the Piano Quintet in Eb minor, Op. 26.

Abstract:
Skeletal dysplasias (SDs) comprise a series of severe congenital disorders that have strong clinical heterogeneity and usually attribute to diverse genetic variations. The pathogenesis of more than half of SDs remains unclear. Additionally, the clinical manifestations of fetal SDs are ambiguous, which poses a big challenge for accurate diagnosis. In this study, eight unrelated families with fetal SD were recruited and subjected to sequential tests including chromosomal karyotyping, chromosomal microarray analysis (CMA), and trio whole-exome sequencing (WES). Sanger sequencing and quantitative fluorescence PCR (QF-PCR) were performed as affirmative experiments. In six families, a total of six pathogenic/likely pathogenic variations were identified in four genes including SLC26A2, FGFR3, FLNB, and TMEM38B. These variations caused disorders following autosomal dominant or autosomal recessive inheritance patterns, respectively. The results provided reliable evidence for the subsequent genetic counseling and reproductive options to these families. With its advantage in variation calling and interpreting, trio WES is a promising strategy for the investigation of fetal SDs in cases with normal karyotyping and CMA results. It has considerable prospects to be utilized in prenatal diagnosis.

Abstract:
The variability and phylogeny among TRI5, TRI8 and TRI11 nucleotide and translated protein sequences of isolates from Trinidad belonging to Fusarium incarnatum-equiseti species complex (FIESC) were compared with FIESC reference sequences. Taxa appeared to be more divergent when DNA sequences were analyzed compared to protein sequences. Neutral and non-neutral mutations in TRI protein sequences that may correspond to variability in the function and structure of the selected TRI proteins were identified. TRI5p had the lowest amino acid diversity with zero predicted non-neutral mutations. TRI5p had potentially three protein disorder regions compared to TRI8p with five protein disorder regions. The deduced TRI11p was more conserved than TRI8p of the same strains. Amino acid substitutions that may be non-neutral to protein function were only detected in diacetoxyscirpenol (DAS) and fusarenon-X (FUS-X) producers of the reference sequence subset for TRI8p and TRI11p. The deduced TRI5 and TRI8 amino acid sequences were mapped to known 3D-structure models and indicated that variations in specific protein order/disorder regions exist in these sequences which affect the overall structural conservation of TRI proteins. Assigning single or combination non-neutral mutations to a particular toxicogenic phenotype may be more representative of potential compared to using genotypic data alone, especially in the absence of wet-lab, experimental validation.

Abstract:
The object of research is the works of V. Bibik written at the beginning of his mature period. The aim of the research is to reveal the main features of Bibik’s style. Methods of research include technical analysis of the works in the context of the innovative tendencies in the Ukrainian music of 1960–70s, as well as comparative research. Research results. Outstanding Ukrainian composer Valentyn Bibik (1940–2003) wrote over 150 works. Mostly they are large-scale symphonic, choral, vocal, and chamber pieces. Among them are 11 symphonies, over 20 concerti for various instruments with orchestra, vocal and choral cycles, chamber compositions (the last group includes 5 string quartets, 3 piano trios, sonatas for string instruments both solo and with piano), 10 piano sonatas, piano solo works (two sets of preludes and fugues – 24 and 34 total, Dies Irae – 39 variations). The composer was born in Kharkiv. In 1966 he completed studies at Kharkiv Conservatory, where he attended the composition class of D. Klebanov. Since 1994, he had been living in St.-Petersburg, and since 1998, in Israel where he died in 2003. Bibik’s formative period coincided with a substantive modernization of Ukrainian culture in the 1960s. During those years, members of the “Kyiv avantgarde” group (L. Hrabovsky, V. Sylvestrov, V. Godziatsky, et al.) sought to utilize modernistic idioms and techniques, such as free atonality, dodecaphony, sonoristic and aleatoric textures, cluster harmony, etc. Unlike the others, Bibik started with a more conservative style, which bore the influences of Shostakovich, Prokofiev, and Bartók. Bibik’s mature period started several years later in the early 1970s with Piano Trio No. 1 (1972) and the composition Watercolors for soprano and piano (1973). Together with his next piano work 34 Preludes and Fugues, these compositions show extremely individual features of Bibik’s style, such as: 1. Special treatment of the sound, which is considered not just a material for building certain musical structures but a self-valuable substance (Bibik has an original manner of organizing sound). Hence, timbral and textural aspects draw special attention to the composer. 2. The pitch and rhythmic structure of the themes is quite simple. A combination of several simple motives becomes the starting point of long and sophisticated development. These motives are derived from folk music, however, due to rhythmic transformation, they have lost their direct connection with the folk source. 3. Rhythmic structures areal so very simple. They often include sequences of equal rhythmic values (usually crotchets or eights). However, the composer avoids monotony dueto due to variable time signatures and permanent rubato, as well as significant flexibility in phrasing. 4. The development relies mostly on melodic and polyphonic elaboration of initial simple motives. The composer utilizes various kinds of polyphony, such as canonic imitations, various combinations of the main and supportive voices, heterophony, hyper-polyphony. In fugues he employs both traditional and new methods of thematic and tonal distribution. 5. The harmony in Bibik’s works is mostly modal, as well as a combination of modality with free atonality and extended tonality. The structure of the dense chords is close to clusters, while more transparent chords include mostly seconds and fourths (as well as their inversions). He almost never used traditional tonal harmony and chords built up from thirds, and was interested in their color aspect rather than their tonal functionalism. 6. The sonoristic texture is very important. It does not diminish the importance of the melody but gets into special collaboration with it (“singing sonority”). A special “mist” around a clear melodic line is one of Bibik’s most typical devices. Due to special “pedal” orchestration, both the line and the “surrounding” sounds become equally important. 7. Elements of limited aleatoric music may be found in his rhythm and agogics, and sometimes inpitch structures (passages and figurations with free choice of the pitches). His favorite technique is a superposition of two rhythmically and temporally independent textural layers (for instance, a combination of the viola solo and the sonoristic orchestral background in the third movement of the Fourth Symphony). 8. Sonata for mand the fugue were significantly reinterpreted within free atonality and modal harmony. These provisions are the scientific novelty of the study.

Abstract:
Trichothecene mycotoxins are a class of secondary metabolites produced by multiple genera of fungi, including certain plant pathogenic Fusarium species. Functional variation in the TRI1 gene produces a novel Type A trichothecene called NX-2 in strains of F. graminearum. Using a bioinformatics approach, a systematic analysis of 52 translated TRI1 sequences of Fusarium species, including five F. graminearum NX-2 producers and four F. graminearum non-NX-2 producers, was conducted to explain the functional difference of TRI1p of FGNX-2. An assessment of several signature motifs of fungal P450s revealed amino acid substitutions in addition to the post-translational N-X-S/T sequons motif, which is indicative of N-linked glycosylation of this TRI1-encoded protein characteristic of NX-2 producers. There was evidence of selection bias, where TRI1 gene sequences were found to be under positive selection and, therefore, under functional constraints. The cumulative amino acid changes in the TRI1p sequences were reflected in the phylogenetic analyses which revealed species-specific clustering with a distinct separation of FGNX-2 from FG-non-NX-2 producers with high bootstrap support. Together, our findings provide insight into the amino acid sequence features responsible for the functional diversification of this TRI1p.

An enhanced elastic-plastic constitutive equation taking into account strain induced transformation is developed. The variation of the Young’s modulus with the martensitic transformation during stamping process is also introduced to improve the accuracy of springback simulation for TRIP steel. The accuracy of the proposed method is verified by comparing with the conventional FE model and experimental results. The enhanced material model is testified to be efficient in accurate prediction on springback for TRIP steels, which will put forward the application of TRIP steel sheet in auto body manufacturing.

Abstract Background The Simultaneous Multiplex Real Time PCR (SIMUL-qPCR) Top 7 STEC Assay Collection is a quick, reliable method for detecting top seven Shiga toxin-producing Escherichia coli (STEC) in raw beef trim, raw ground beef, and beef carcass sampling sheets. Each assay multiplexes several targets in one run to identify E. coli O157: H7, O26, O45, O103, O111, O121, O145, Shiga toxin, and intimin genes. This collection uses specifically optimized proprietary media for single-step recovery and enrichment of enterohemorrhagic E. coli. Objective This report details the method validation study to validate raw beef trim, raw ground beef, and beef carcass sampling sheets. Method Matrix studies for raw beef trim, raw ground beef, and beef carcass sampling sheets, inclusivity/exclusivity, product consistency/stability, and robustness testing were conducted to assess the method’s performance. Results Fifty top seven STEC isolates were analyzed with the SIMUL-qPCR assay. Thirty-two isolates, including closely related non-E. coli species and E. coli non-STEC strains, were also tested. Inclusivity showed the collection detected the Shiga toxin (stx) gene, intimin (eae) gene, and top seven serogroups. None of the 32 exclusivity strains were detected. The candidate and reference methods’ results had no statistically significant differences during matrix studies. Small variations in critical test parameters (enrichment time, extraction reagent volume, and extracted sample volume) did not adversely affect the assay’s performance, and stability testing indicated consistent results for at least one year. Conclusions The data presented demonstrate that the SIMUL-qPCR Top 7 STEC Assay is a reliable method for detecting the top seven STEC. Highlights The Applied Food Diagnostics, Inc. Simultaneous Multiplex Real Time PCR (SIMUL-qPCR) Top 7 STEC Assay is capable of detecting the top seven Shiga toxin-producing Escherichia coli in beef trim, ground beef, and beef carcass sampling sheets in as little as 10 hours of enrichment.

Abstract:
Max Reger was a prolific composer on the threshold of modernism. The style of his extensive musical output was polarizing among his contemporaries. A criticism of Reger's music is its complex and dense musical structure. Despite writing tonal music, Reger often pushes the boundaries of tonality so far that all sense of formal organization is seemingly imperceptible. In this dissertation, I offer what I observed to be a new way of discerning Reger's motivic relationships and formal structures within and between movements. There are three primary tools and methods I incorporated to make these observations: Schoenberg's developing variation; melodic contour analysis as discussed by Elizabeth West-Marvin and Diana Deutsch; and Janet Schmalfeldt's motivic cyclicism stemming from internal themes. In this dissertation I examine five different musical works by Reger: D minor Piano Quartet, Clarinet Quintet, Piano Concerto, String Quartet, op. 121 and E minor Piano Trio, op. 102. My analysis shows how Reger relies on melodic contours of his motives to connect musical moments across entire movements and entire works with multiple movements. These motives are developed and often mark structurally significant moments providing the organization often perceived as missing in Reger's music.

Abstract:
Many real-world domains are subject to a structured non-stationarity which affects the agent's goals and the environmental dynamics. Meta-reinforcement learning (RL) has been shown successful for training agents that quickly adapt to related tasks. However, most of the existing meta-RL algorithms for non-stationary domains either make strong assumptions on the task generation process or require sampling from it at training time. In this paper, we propose a novel algorithm (TRIO) that optimizes for the future by explicitly tracking the task evolution through time. At training time, TRIO learns a variational module to quickly identify latent parameters from experience samples. This module is learned jointly with an optimal exploration policy that takes task uncertainty into account. At test time, TRIO tracks the evolution of the latent parameters online, hence reducing the uncertainty over future tasks and obtaining fast adaptation through the meta-learned policy. Unlike most existing methods, TRIO does not assume Markovian task-evolution processes, it does not require information about the non-stationarity at training time, and it captures complex changes undergoing in the environment. We evaluate our algorithm on different simulated problems and show it outperforms competitive baselines.

Abstract:
In this investigation an intensive experimental study was conducted to study the flow quality in a test section of a low speed, V∞ = 10–100, closed return wind tunnel. An interesting result was obtained when a trip strip, a guitar wire with D = 1.14mm, was installed near the end of the contraction region. Addition of this trip wire resulted in a turbulent reduction almost at all speed ranges along the entire test section. Further, it smoothness the variation of turbulent intensity along the test section too. Velocity profiles along the tunnel wall at various speeds using a rake were measured with and without the trip wire. In addition, pressure contours at different location of the test section was measured too.

Abstract Elbow fittings are manufactured using quenching and tempering heat treatment processes. Such fittings can occasionally exhibit localized regions with lower yield strength than the design target, potentially due to non-uniform heat treatment. This paper presents an analytical methodology to examine the influence of these localized lower yield zones on the load capacity of the affected pipe fitting. In parallel, full-scale testing has been performed to quantify the actual response of the elbows under a combination of different loading conditions. The experimental data is used to validate the analytical approach. Details of the analytical method include a two-fold criterion: a global failure based on elastic–plastic stress analysis and a local failure based on the tri-axial strain limit per ASME Boiler and Pressure Vessel Code Section VIII, Division 2. This paper presents the details of the finite element model development, assessment procedure, validation and parametric analysis of the size and location of the low yield zones in the elbow fittings. The fittings are analyzed for three possible operating scenarios: internal pressure, internal pressure with opening moment and internal pressure with closing moment. To characterize the influence of the low yield zone on the strength of the pipe, a parameter termed as “effective yield strength” is introduced. This approach is further demonstrated and found suitable for predicting burst pressures of components with lower yield zones of various diameters and thicknesses. This assessment method can be further extended to assess other pipeline components that exhibit similar behavior.

Abstract Energy storage systems provide a variety of benefits, including taking better advantage of renewable electricity when available and smoothing demand by shifting demand peaks to times when electricity prices and demand are lower. When low electricity demand occurs during the nighttime, system wide advantages also occur. These lower nighttime ambient temperatures lead to efficiency improvements throughout the grid, including power generators, transmission and distribution systems, chillers, etc. An analysis of ice thermal energy storage carried out by T. Deetjen et al. in 2018 analyzed fuel consumption of the power generation fleet for meeting cooling demand in buildings as a function of ambient temperature, relative humidity, transmission and distribution current, and baseline power plant efficiency. Their results showed that the effective round trip efficiency for ice thermal energy storage could exceed 100% due to the efficiency gains of nighttime operation. However, their analysis was performed on a case study in Dallas, where relatively high humidities lead to a relatively small diurnal temperature variation during the cooling season. In order to expand on this limitation, our study extends this analysis to a mountain west climate, using northern Arizona as a case study. The climate of the mountain west has several key differences from that of the Dallas case study in the previous work, including lower relative humidity, higher diurnal temperature variation, and near- and below-freezing nighttime temperatures during shoulder seasons that also exhibit cooling demand in buildings. To address these differences, this paper updates the models of Deetjen et al. to consider generator fleet efficiency and chiller/icemaking COP for local weather characteristics relevant to the mountain west, as well as considering the differences between fuel mixes of the generator fleet in nighttime and daytime. Compared to Dallas, the larger temperature variation of northern Arizona leads to higher round trip efficiencies (RTE) over the course of the year in most days of the year (e.g. 313 days of the year in northern Arizona in comparison with 182 days in Dallas), demonstrating frequent achievement of over 100% effective round trip efficiency. The presence of a mature commercial market and the possibility of gaining over 100% effective round trip efficiency create a strong case for cooling thermal energy storage as an energy storage approach. Future work will investigate emissions impacts as well as extend the analysis to additional western climates, including the hot dry and marine climates.

In light of the growing strain on the energy grid and the increased awareness of the significant role buildings play within the energy ecosystem, the need for building operational strategies which minimize energy consumption has never been greater. One of the major hurdles impeding this realization primarily lies not in the lack of decision strategies, but in their inherent lack of adaptability. With most operational strategies partly dictated by a dynamic trio of social, economic and environmental factors which include occupant preference, energy price and weather conditions, it is important to realize and capitalize on this dynamism to open up new avenues for energy savings. This paper extends this idea by developing a dynamic optimization mechanism for Net-zero building clusters. A bi-level operation framework is presented to study the energy tradeoffs resulting from the adaptive measures adopted in response to hourly variations in energy price, energy consumption and indoor occupant comfort preferences. The experimental results verify the need for adaptive decision frameworks and demonstrate, through Pareto analysis, that the approach is capable of exploiting the energy saving opportunities made available through fluctuations in energy price and occupant comfort preferences.

The automatization of the drilling process opens the opportunity to faster reactions in case of unexpected drilling conditions, therefore reducing the risk that a drilling incident escalates to a serious situation. It also allows to push the drilling performance to be as close as possible to the limits of drillability as a function of the varying drilling conditions. But to achieve high level of drilling process automation, it is necessary to have access to accurate mathematical models of the complex physical system that is composed of the drilling rig, the drill-string, the drilling fluid and the borehole itself. As the development of accurate heat transfer, mechanical and hydraulic models and their utilization in full scale drilling applications is a huge and complex task, it is tempting to recreate drilling automatization problems in a laboratory scale setup. Because of sudden variations of the downhole drilling conditions, like when transitioning from soft to hard rock or when the bit is subject to large torque variations induced by interbedded rock layers, the boundary conditions at the bit change suddenly and require quick response from the automatic top-drive and hoisting system controllers. At a small laboratory scale, the necessary reaction times are of the order of milliseconds and therefore exclude any manual intervention. It is therefore crucial that the automatic control methods utilize precise mathematical models of the physical system to accurately estimate the limits by which the drilling process can be managed under safe conditions. For that reason, a general purpose mathematical model of small-scale drilling rigs has been developed. First, the Rayleigh-Ritz method is used to determine the deflection of the drill-string and to estimate the side forces at the contact points along the drill-string and BHA (Bottom Hole Assembly). Then the dynamic response of the power transmission system is modelled for both variable frequency drive controlled tri-phase motors and for stepper motors, including friction effects at the contact points. Friction is modelled using Stribeck theory rather than the classical Coulomb laws of friction. Finally, the expected response of 3D accelerometers, that could be placed on the outside of a BHA component, is modelled to retrieve possible inclination variations and potential vibration modes such as torsional oscillations, forward or backward whirl. The generality of the model is such that it can be used for many small-scale drilling rig designs.

A comprehensive investigation of the effect of various tip sealing geometries is presented on the blade tip leakage flow and associated heat transfer of a scaled up HPT turbine blade in a low-speed wind tunnel facility. The linear cascade is made of four blades with the two corner blades acting as guides. The tip section of a HPT first stage rotor blade is used to fabricate the 2-D blade. The wind tunnel accommodates an 116° turn for the blade cascade. The mainstream Reynolds number based on the axial chord length at cascade exit is 4.83 × 105. The upstream wake effect is simulated with a spoked wheel wake generator placed upstream of the cascade. A turbulence grid placed even farther upstream generates the required free-stream turbulence of 4.8%. The center blade has a tip clearance gap of 1.5625% with respect to the blade span. Static pressure measurements are obtained on the blade surface and the shroud. The effect of crosswise trip strips to reduce leakage flow and associated heat transfer is investigated with strips placed along the leakage flow direction, against the leakage flow and along the chord. Cylindrical pin fins and pitch variation of strips over the tip surface are also investigated. Detailed heat transfer measurements are obtained using a steady state HSI-based liquid crystal technique. The effect of periodic unsteady wake effect is also investigated by varying the wake Strouhal number from 0. to 0.2, and to 0.4. Results show that the trip strips placed against the leakage flow produce the lowest heat transfer on the tips compared to all the other cases with a reduction between 10–15% compared to the plain tip. Results also show that the pitch of the strips has a small effect on the overall reduction. Cylindrical pins fins and strips along the leakage flow direction do not decrease the heat transfer coefficients and in some cases enhance the heat transfer coefficients by as much as 20%.

This work presents an experimental study on the aeroelastic wind/sails/rig interaction at full scale in real navigation conditions with the aim to give an experimental validation of unsteady Fluid Structure Interaction (FSI) models applied to yacht sails. An onboard instrumentation system has been developed on a J80 yacht to measure simultaneously and dynamically the navigation parameters, yacht motion, and sails flying shape and loads in the standing and running rigging. The first results recorded while sailing upwind in head waves are shown. Variations of the measured parameters are characterized and related to the yacht motion (trim mainly). Coherence between the different parameters is examined. In the system’s response to the dynamic forcing (pitching motion), we try to distinguish between the aerodynamic effect of varying apparent wind induced by the motion and the structural effect of varying stresses and strains due to the motion and inertia. The simulation results from the FSI model compare very well with the experimental data for steady sailing conditions. For the unsteady conditions obtained in head waves, the first results show a good agreement between measurements and simulation.

Abstract With the energy sector in crisis the worldover, oil and gas operators continue to seek more effective and efficient methods to reach potential prospects. With sharply declining oil prices, it is imperative that operators minimize the non-productive time in the drilling of all wells. Many operators are actively seeking riskier exploration to establish a strong foothold in this volatile market. One such area of interest to operators is HPHT and beyond wells. An HPHT prospect carries a high-risk high-reward potential, therefore newer and advanced methods are being deployed to successfully drill and complete HPHT wells. The Makran Coastal belt in south western Pakistan is one such area containing a potential Ultra-HPHT prospect. Many operators had attempted to drill about 9 wells in the locality but never managed to reach target depth due to drilling operations being plagued with a large number of problems. The drilling problems included high pressure influxes, stuck pipe while controlling influxes, circulation losses with high mud weights and ECD’s, differential sticking against permeable formations, inefficient bottom hole pressure control due to mud weight reduction with high temperatures and swabbing from the formation due to having an insufficient trip margin. The operator was facing an extremely narrow drilling window in the target section. The maximum formation pressure was estimated to be around 2.29 SG while the maximum fracture pressure of the formation was estimated to be around 2.35 SG in EMW. It was abundantly clear that drilling with a conventional mud system would be impossible and impractical on all forthcoming wells. As it was of paramount importance to precisely manage the wellbore pressure profile, the operator decided to apply managed pressure drilling on a candidate well. By applying managed pressure drilling techniques the operator expected to drill the section with an underbalanced mud weight and maneuver the bottom hole pressure just above the pore pressure line and thereby avoid circulation losses, detect influxes early on and control influxes without the need of ever shutting in the well, account for mud density variations with temperatures by executing an advanced thermal hydraulics model in real time, mitigate swabbing from the formation again by maintaining a constant bottom hole pressure while tripping, and finally ascertain the downhole pressure environment by conducting dynamic formation pressure tests. The successful application of MPD enabled the operator to reach target depth for the first time in the history of the area. The paper studies the planning, design, and execution of MPD on the subject well.
