Abstract:
The conserved fungal velvet family regulatory proteins link development and secondary metabolite production. The velvet domain for DNA binding and dimerization is similar to the structure of the Rel homology domain of the mammalian NF-κB transcription factor. A comprehensive study addressed the functions of all four homologs of velvet domain encoding genes in the fungal life cycle of the soil-borne plant pathogenic fungus Verticillium dahliae. Genetic, cell biological, proteomic and metabolomic analyses of Vel1, Vel2, Vel3 and Vos1 were combined with plant pathogenicity experiments. Different phases of fungal growth, development and pathogenicity require V. dahliae velvet proteins, including Vel1-Vel2, Vel2-Vos1 and Vel3-Vos1 heterodimers, which are already present during vegetative hyphal growth. The major novel finding of this study is that Vel1 is necessary for initial plant root colonization and together with Vel3 for propagation in planta by conidiation. Vel1 is needed for disease symptom induction in tomato. Vel1, Vel2, and Vel3 control the formation of microsclerotia in senescent plants. Vel1 is the most important among all four V. dahliae velvet proteins with a wide variety of functions during all phases of the fungal life cycle in as well as ex planta.

Abstract:
Abstract The large collections of ChIP-seq data rapidly accumulating in public data warehouses provide genome-wide binding site maps for hundreds of transcription factors (TFs). However, the extent of the regulatory occupancy space in the human genome has not yet been fully apprehended by integrating public ChIP-seq data sets and combining it with ENCODE TFs map. To enable genome-wide identification of regulatory elements we have collected, analysed and retained 395 available ChIP-seq data sets merged with ENCODE peaks covering a total of 237 TFs. This enhanced repertoire complements and refines current genome-wide occupancy maps by increasing the human genome regulatory search space by 14% compared to ENCODE alone, and also increases the complexity of the regulatory dictionary. As a direct application we used this unified binding repertoire to annotate variant enhancer loci (VELs) from H3K4me1 mark in two cancer cell lines (MCF-7, CRC) and observed enrichments of specific TFs involved in biological key functions to cancer development and proliferation. Those enrichments of TFs within VELs provide a direct annotation of non-coding regions detected in cancer genomes. Finally, full access to this catalogue is available online together with the TFs enrichment analysis tool (http://tagc.univ-mrs.fr/remap/).

Abstract:
In this article, we propose new algorithms for multi-objective coordination graphs (MO-CoGs). Key to the efficiency of these algorithms is that they compute a convex coverage set (CCS) instead of a Pareto coverage set (PCS). Not only is a CCS a sufficient solution set for a large class of problems, it also has important characteristics that facilitate more efficient solutions. We propose two main algorithms for computing a CCS in MO-CoGs. Convex multi-objective variable elimination (CMOVE) computes a CCS by performing a series of agent eliminations, which can be seen as solving a series of local multi-objective subproblems. Variable elimination linear support (VELS) iteratively identifies the single weight vector, w, that can lead to the maximal possible improvement on a partial CCS and calls variable elimination to solve a scalarized instance of the problem for w. VELS is faster than CMOVE for small and medium numbers of objectives and can compute an ε-approximate CCS in a fraction of the runtime. In addition, we propose variants of these methods that employ AND/OR tree search instead of variable elimination to achieve memory efficiency. We analyze the runtime and space complexities of these methods, prove their correctness, and compare them empirically against a naive baseline and an existing PCS method, both in terms of memory-usage and runtime. Our results show that, by focusing on the CCS, these methods achieve much better scalability in the number of agents than the current state of the art.

Abstract:
Background. The article discusses psychology of teenagers and elders in detective novels which is considered to be more complicated. Although the society and the social environment change and renew the way of thinking, the changes in the world of childhood and adolescence, like the laws of nature, are constantly changing. Adolescence is a period that is complicated by the transition of a person to the stage of childhood and maturity. Methods. It is important that every teenager at this age pays more attention to the heroes of books and movies, learns from them. Consequently, the task of fiction for teenagers is also very responsible and multifaceted. Among the works of world literature such as J. Verne's "Children of Captain Grant", "Five Weeks in a Balloon", "Mysterious Island", D. Defoe's "Robinson Crusoe", J. Swift's "Gulliver's Travels", Uzbek children's fiction and detective prose, for example The works of H. Shaykhov, T. Malik, O. Mukhtor, H. Tukhtaboyev, as well as the didactic stories of T. Malik, E. El presente artículo de investigación da cuenta de la importancia que tiene la grafología en los procesos de selección de personal a partir del estudio del perfil criminal.

Abstract:
Lin-7 PDZ proteins, also called MALS or Velis, have been shown to coordinate basolateral membrane expression of various target proteins in renal epithelial cell models. Three different Lin-7/MALS/Veli isoforms, encoded by separate genes, have been identified. Here, we show that each Lin-7/MALS/Veli isoform is expressed in the kidney. Using MALS isoform-specific antibodies in combination with cell-specific marker antibodies, we found the products of the three mammalian Lin-7/MALS/Veli genes are differentially expressed along the length of the nephron. MALS/Veli 1 is predominately expressed in the glomerulus, thick ascending limb of Henle’s loop (TAL), and the distal convoluted tubule (DCT). MALS/Veli 2 is exclusively expressed in the vasa recta. MALS/Veli 3 is largely located in the DCT and collecting duct. The subcellular localization of MALS/Veli proteins can vary, depending on the isoform and the cell type. In contrast to the predominate basolateral location of MALS/Veli 1 in the TAL and DCT and MALS/Veli 3 in the DCT, MALS/Veli 1 is found diffusely throughout the cytosol of intercalated cells. In the collecting duct, MALS/Veli 3 is chiefly located on the basal membrane. Collectively, these results suggest that different MALS/Veli isoforms may carry out cell type-specific functions. The TAL and distal segments appear to have the most significant capacity for a basolateral membrane-targeting mechanism involving different MALS/Veli isoforms.

Abstract:
Abstract To survive that is to eat and not to be eaten. Whatever its spatial and temporal level of organisation, every living system owns 7 invariant capacities (the gauge invariance paradigm). Emerging by embedments and juxtapositions of previous systems, every living system-of-systems is both dependent and independent from its new global level of organisation and past and present local situations of emergence. Mass growth governs growth phase duration. Local actors, modules of past, present and new modules of modules become mutually integrated (percolation process) into a new Whole through their merging into an ARMSADA (Association for the Reciprocal and Mutual Sharing of Advantages and DisAdvantages). Reversely (systemic constructal law) the global Whole is integrating the local parceners. Whatever the organisation level, the living systems obey the same principle of evolution and emergence, the volume and time of generation of the adult system obey a correlation: The mass controlled duration of acquisition of the reproductive capacity and the volume at its acquisition are linked by a 3/2 exponent power law. This dynamic fractal law is invariant, between and within le- vels of organisation. Brownian motion is the basic phenomenon of growth control: Matter and energy are exchanged at a constant flow rate. A Pareto-like relationship governs limits and limitations: Mutual survival depends on reciprocal limitations, through interactive feedbacks, for the best and for the worst. To survive that is to transform disadvantages into advantages and to avoid advantages turn to disadvantages.

Abstract:
According to contemporary views, hereditary connective tissue disorders divided classified Marfan syndrome, Loeys-Dietz’s, Ehlers-Danlos syndrome, the primary mitral valve prolapse. It is known that the fibrillinopaty, which include the Marfan syndrome and Loeys-Dietz’s is characterized by activation of TGF-β signaling pathway. With high le vels of TGF-β attributed most of these clinical manifestations these diseases – aneurysm of the aorta, arahnodaktylya, duralectasy. Assessment of the activity of TGF-β in persons with marfanoid habitus has not previously been studied. Materials and methods. As part of this work, surveyed 70 people: 61 patients young age (median age of 20.1 ± 2.1 years), among which 36 boys and 25 girls and 9 men with verified diagnosis Marfan syndrome (median age 27.9 ± 9.3 years). All survey performed Echocardiography with a targeted search of small anomalies of heart. Results. Correlation analysis showed a direct and reliable connection between arahnodaktylya and concentration of TGF-β1 in serum (r = 0.4, p = 0.05). For young people with signs of marfanoid habitus are characterized by reliably a higher concentration in the serum of both isoforms of TGF-β. Excess of threshold levels of TGF-β1 revealed at 20% of the core group and not found at all in the control (p < 0.05). Among persons with exceedances of threshold values for at least one faction of the TGF-β patients with signs of marfanoid habitus met almost three times more often than in the group with normal values of TGF-β (p = 0.01, χ2 = 5.58). In the group of persons with marfanoid habitus and increases TGF-β are detected more frequently such as atrial septal aneurysm, false chord left ventricle papillary muscles, incremental, deflection of shutters of the mitral valve in 1-2 mm, asymmetry tricuspid aortic valve.

Abstract:
Palavras-chave: lã, velo, merino Abstract Wool production has been for many years, in Portugal and in several regions of the world, one of the main reasons for sheep production. This situation changed with the appearance of synthetic fibres. The production of meat or milk has thus become the primary economic aptitude of these animals. In any case, since it is necessary to shear the sheep annually, wool can provide extra yield, especially in sheep with a fleece of superior quality. Portugal presents 16 autochthonous sheep breeds that fall into three large ethnic groups, Merino, Bordaleiro and Churro, according to their external morphological characteristics and aptitudes. Each of these ethnic groups has different types of veils with different textile characteristics. It is assumed that breeds of Churra ethnic group provide wool considered of poor quality, characterised by being smooth and rough touch. In contrast, the breeds of the Merino ethnic group are considered to be producers of quality wool, characterised by being soft to the touch and presenting very undulating fibres. The physical and textile characteristics that sheep present relate not only to genetic factors but also to the conditions of the environment in which they live. This work intends to contribute to the knowledge of the origin of wool and to understand the differentiation of native sheep breeds in the three ethnic groups that exist in Portugal. Abstract:
To test the hypothesis that soft palate muscles are abnormal in cleft palate, we compared soft palate morphogenesis in fetuses with cleft palate (n=4) to age-matched (n=3) and nonmatched (n=1) control specimens. The morphologic status of all soft palate and masticatory structures were classified into one of six stages based on the level of histogenesis. At 54 mm crown-rump length (CRL), the levator vell palatini (L), palatopharyngeus (PP), and palatoglossus (PG) in cleft subjects demonstrated mesenchymal condensation into myoblastic fields, lagging behind the control specimens (97 mm CRL), which displayed definitive fields of myoblasts and myotube formation. In the 175 mm and 225 mm cleft and the 170 mm and 192 mm control specimens, muscular morphology was similar and had reached its postnatal appearance for the tensor veli palatini (175 m only) and L, PP, PG (225 mm only). Muscle fiber directions were, however, disoriented and disorganized, especially close to the medial epithelial edge of the cleft. The levator veli palatini, could not be distinguished as a discrete muscle in the cleft specimens, and what we believed to be the PP and PG seemed “normal” at the level of light microscopy, but malpositioned in a superior direction. This preliminary study demonstrates for the first time that early myogenesis in cleft palates differs from normal.

Abstract:
Objective: to analyze the correlation between the expression levels of microRNA-101, microRNA-142, microRNA-27, microRNA-339, and microRNA-424 and the plasma concentrations of biomarkers that are potentially significant for the diagnosis of post-transplant complications in heart recipients. Materials and methods. The study enrolled 72 heart recipients, among whom were 56 men (77.8%). The average age of recipients was 48.6 ± 10.9 (16 to 70) years. There were 38 patients with severe chronic heart failure, among whom were 29 men (76.3%). Patients’ mean age was 48.8 ± 9.9 (26 to 70) years. The control group consisted of 12 healthy individuals who did not differ significantly by sex and age. microRNA expression levels in blood plasma were measured via quantitative polymerase chain reaction. Plasma concentrations of VEGF-A, PLGF, MCP-1, and sCD40L were determined using a multiplex method. ST2 and Galectin-3 concentrations were measured via enzyme-linked immunosorbent assay. Results. Patients with end-stage chronic heart failure were found to have significantly higher expression levels of microRNA-27, microRNA-339 and microRNA-424 in blood plasma compared with the healthy individuals. In potential heart recipients, the expression le vels of microRNA-339 and microRNA-424 correlated with serum galectin-3 concentrations, microRNA-101 expression levels correlated with PLGF-1 concentrations, while microRNA-27 expression levels correlated with plasma MCP-1 concentrations. In the early post-transplant period, the expression levels of microRNA-101, microRNA-339, and microRNA-424 in heart recipients were significantly lower than in patients with severe chronic heart failure. In the early post-transplant period (one year or more after transplantation), microRNA-101 and microRNA-27 expression levels were significantly higher than in heart recipients. A year or more after transplantation, the following correlations were found in heart recipients: microRNA-142 expression level correlated with serum levels of galectin-3 (p = 0.05), microRNA-27 and microRNA-424 expression levels correlated with ST2 concentrations (p = 0.02), microRNA-27 expression level correlated with PLGF-1 concentrations (p = 0.02), while microRNA-101 expression level correlated with serum levels of PAPP-A (p = 0.05). Conclusion. In heart recipients, the expression levels of microRNA-142, microRNA-27, microRNA-424, and microRNA-101 correlate with the concentration levels of biomarkers of fibrosis (Galectin-3), rejection (ST2), neoangiogenesis (PLGF), and tissue destruction (PAPP-A). A comprehensive analysis of pre- and post-translational markers may open up new perspectives in diagnosis, assessment of the risks of post-transplant complications, and in understanding the processes leading to their development.

Abstract:

Abstract:
Gothograptid retiolitines were distinctive in being one of the very few graptolite groups to thrive through the late Wenlock extinction event that killed off most graptoloid species, and their distinctive construction may have been a factor in this success amid environmental adversity. New and rich material from two localities in Poland and five localities in Lithuania contains Gothograptus nassa, Gothograptus obtectus and four new species. The detailed morphology and reconstruction of the tubarium shows its specific features, different from other lundgreni Biozone retiolitines. The tubarium is narrow, elongated, finite, massive and compact. The thecal orifices are hidden by simple hoods or by extraordinary reticulated veils, which may cover the ventral walls. The colonies of Gothograptus species from the lundgreni Biozone are short, usually having eight pairs of thecae, whereas species from the post-lundgreni interval may reach more than 20 pairs of thecae. A common feature of Gothograptus species is the presence of singular genicular structures tightly covering thecal orifices. Reticulated hoods and veils are characteristic of species from the lundgreni Biozone. The most common such structures are nassa hoods, located on the proximal thecae in some species from the lundgreni Biozone, and on every theca of post-lundgreni species. Only Gothograptus domeyki n. sp. has no genicular processes on most thecae. Within a total of four species from the lundgreni Biozone and three species from the nassa and praedeubeli Biozones, we describe here the new species Gothograptus domeyki and Gothograptus velo from the lundgreni Biozone, Gothograptus diminutus from the parvus/nassa Biozone, and Gothograptus auriculatus from the dubius/nassa and praedeubeli biozones; G. auriculatus n. sp. is the youngest form of the genus Gothograptus. Variation within the genus Gothograptus during the post-lundgreni interval period is shown for the first time.
