Full textThe lebanese restraining bend is the most prominent geometric irregularity along the Dead Sea transform : The Arabia/Nubia (Sinai) plate boundary. Mont-Lebanon, bounded to the East by the Yammouneh Fault - The most active local branch of the Transform - is the highest relief along. While previous studies had only indirectly addressed the question of the range's build up, our fieldwork reveals the existence of active thrusts along the western and eastern flanks of Mount-Lebanon. The lebanese Flexure corresponds to a major crustal thrust ramp at depth accommodating NW-SE directed shortening resulting from transpression along the restraining bend. This thrust system can be observed onland north of Enfeh as it forms a foreland thrust belt, and also offshore between tripoli and Saida as revealed by the new geophysical data collected during the SHALIMAR marine campaign. The age of the folding onland, and the deformation in Messinian and Plio-Quaternary sediments observed offshore on eismic profiles suggest a Late-Miocene age for the onset of deformation, with the main relief built up happening since 5My. Combined use of biological sea-level indicators with historical, geophysical and geological data suggests that the tsunamogenic 551AD destructive earthquake took place on this major thrust system. Balanced crustal-scale cross-sections suggest a cumulative shortening of 10-15Km corresponding to rates of 1-3mm/yr. On and offshore observations hints for the inversion of the Lebanese passive margin between Saida and Tripoli

Full textThe aim of this study was to provide further knowledge of the effects of anthropogenic disturbance on the conservation status of important shallow habitats (vermetid reefs, Cystoseira forests) in a relatively poorly known region of the Mediterranean Sea: the Lebanese coast. Multiple approaches were applied for the first time along the Levantine Sea, such as the CARLIT (CARtography of rocky-shore LITtoral communities) index, a tool conceived to quantify the Ecological Status (ES) in the framework of the Water Directive (WFD 2000/60/EU). A general description of the Lebanese coastline, summarizing the major environmental features and anthropogenic activities influencing benthic communities, highlighted that Lebanon has been (and is at present) subject to multiple stressors. The application of the CARLIT, based on macroalgal, gives important insights on the ES of Lebanese ecosystems in relation to human impacts and provides a detailed cartography on the distribution and abundance of shallow communities, and in particular Cystoseira and Sargassum forests. The survey of some Lebanese vermetid reefs allowed the evaluation of their current status according to different human pressures and highlighted that they are highly affected by human pressures and only few of them are still alive. In a view of the upcoming oil and gas discovery offshore Lebanon, Lebanese marine ecosystems are under multiple and massive threats; the data provided in the research performed provide a baseline of the distribution and conservation status of key habitats along Lebanese coasts, in order to follow their evolution and to have a management tool in case of catastrophic oil spills

Full textWe present in this thesis some improvements of acoustical pressure prediction by ray-based methods involving curved surfaces. The framework is an adaptative beam tracing algorithm. The first part deals with diffraction by obstacles. First we solve the problem of successive wedges diffractions. Then we extend this problem to creeping waves calculation on curved surfaces using an appropriate and frequency independant tesselation of these surfaces. The reconstruction of continuous and derivable surfaces from meshes to improve reections computation is treated in the second part. These surfaces have to be suitable for ray tracing technics. Starting from Powell-Sabin splines we extend its use to any kind of surfaces. This implies different projection planes that creates holes on the surfaces. We show that these gaps can be filled with polynomial implicit surfaces of degree four ; consequently their intersections with rays can calculated analytically

Nematoda Lagochilascaris minor is the etiologic agent of lagochilascaríase in human beings. Until today, no one knows the natural host of this helminth and Brazil registers about 90% of the cases of lagochilascaríase world-wide human being. The life cycle of L. minor was described using an experimental model in mice and domestic cat, behaving as an intermediate host and definitive host respectively. The objective of this study was to evaluate the histology of the adult males of this helminth worms, using light microscopy. These specimens were removed after necropsy of cats kept in their cages cataloged in male and female worms, and the adult male specimens were fixed in formalin solution 10 % hot, AFA and 4% paraformaldehyde in sodium cacodylate buffer 0 1 M, pH 7.2. After the different fixations, the samples were dehydrated in a gradual series of ethanol in different concentrations of alcohol levels, and included in historesin cut with 3μm –thick microtome, with horizontal and transverse serial sections. The sections were stained with Hematoxylin & Eosin (HE), Carmin, Giemsa and Toluidine Blue. The best fixation was achieved with the use of AFA and 4% paraformaldehyde in 0.1 M sodium cacodylate buffer pH 7.2 and the best staining was obtained from the use of the dye HE, which showed histological structures of adult male worms L. minor loop as testicular, seminal vesicle, ejaculatory duct, sperm and spicule. It was also observed structures of the digestive system, muscular system, cuticle, wing side and lateral line. The sections were photomicrographed using photomicroscope with the help of the program Motic Images Plus 2.0, selected and analyzed.
O nematoda Lagochilascaris minor é o agente etiológico da lagochilascaríase em humanos. Até hoje, não se conhece o hospedeiro natural deste helminto e o Brasil registra cerca de 90% dos casos da lagochilascaríase humana mundial. O ciclo evolutivo de L. minor foi descrito utilizando um modelo experimental com camundongo e gato doméstico, comportando-se como hospedeiro intermediário e hospedeiro definitivo, respectivamente. O objetivo deste trabalho foi avaliar a histologia dos vermes adultos machos deste helminto, utilizando microscopia de luz. Estes exemplares foram retirados após necropsia de gatos mantidos em biotério, catalogados em vermes machos e fêmeas, sendo que os exemplares machos adultos foram fixados em solução de Formalina a 10% a quente, AFA e Paraformaldeído a 4% em tampão cacodilato de sódio 0,1 M pH 7,2. Após as diferentes fixações, as amostras foram desidratadas em uma série gradual de etanol em diferentes concentrações de teores alcoólicos, incluídas em historesina e cortadas com 3μm de espessura em micrótomo, com secções seriadas horizontais e transversais. Os cortes foram corados com Hematoxilina & Eosina (HE), Carmim, Giemsa e Azul de Toluidina. A melhores fixações foram alcançadas com o uso de AFA e Paraformaldeído a 4% em tampão cacodilato sódio 0,1 M pH 7,2 e a melhor coloração obteve-se a partir da utilização do corante HE, que evidenciou estruturas histológicas dos vermes adultos machos de L. minor como alça testicular, vesícula seminal, ducto ejaculatório, espermatozoides e espículo. Observou-se também estruturas do sistema digestório, sistema muscular, cutícula, asa lateral e linha lateral. As imagens dos cortes foram realizadas utilizando um fotomicroscópio com o auxílio do programa Motic Images Plus 2.0, selecionadas e analisadas.

Schistosoma mansoni, é um dos mais importantes parasitas que infectam seres humanos. No nordeste do Brasil, a esquistossomose é historicamente endêmica e considerada como um problema de saúde pública. No estado de Pernambuco, particularmente no litoral e zona da Mata, predomina a cepa São Lourenço da Mata (SLM). Estudos ultraestruturais desta cepa até então ainda não foram realizados, embora análises morfométricas e morfológicas de outras cepas existentes no Brasil já tenham sido publicadas na literatura. Os vermes são bem adaptados ao hospedeiro, tendo a sua longevidade vista como uma conseqüência do eficaz escape do sistema imune. Na circulação sanguínea os vermes devem obter lipídios a partir do hospedeiro, já que eles não sintetizam tais compostos, sendo as lipoproteínas plasmáticas a possível fonte de tais lipídios, além de poderem atuar mascarando o reconhecimento do verme pelo sistema imune. Neste trabalho, microscopia eletrônica de varredura (SEM) foi utilizada para a análise morfológica e morfométrica de cercárias e vermes adultos de S. mansoni cepa SLM, bem como para estudar a interação da lipoproteína de baixa densidade (LDL) com o tegumento do parasito. Também foram usadas diferentes técnicas para a localização e identificação das proteínas envolvidas no processo de absorção e/ou transporte de lipoproteínas no verme adulto de S. mansoni. As cercárias foram obtidas de caramujos Biomphalaria glabrata e os vermes adultos de camundongos machos Mus musculus e Swiss, ambos infectados pela cepa SLM de S. mansoni. Os resultados da SEM mostraram que os corpos da cercárias são cobertos por espinhos, com uma ventosa ventral, uma ventosa oral com receptores sensoriais, e um par de glândulas de penetração na cabeça. Seu comprimento total variou de 174 a 290μm. O comprimento dos vermes adultos machos foi de 4mm e das fêmeas 5mm. O comprimento da região anterior do macho foi de 470μm e 271μm para o sexo feminino. Todos os parâmetros foram realizados em dez amostras. Os valores encontrados na morfometria da cepa SLM foram menores que em outras cepas de S. mansoni já descritas. Através da microscopia eletrônica de transmissão (TEM), os experimentos de imunomarcação mostraram partículas de ouro localizadas dentro do tegumento, na região muscular e espículas dos machos e em torno das células vitelínicas das fêmeas. A utilização de técnicas tais como imunoblotting e “liquid chromatography–mass spectrometry” (LC-MS) proporcionaram a identificação no S. mansoni de duas formas diferentes da chaperona Hsp 70, proteínas envolvidas na interação com a apo-B e o seu transporte para o retículo endoplasmático, sendo responsável também pela ruptura das interações clatrina-clatrina na endocitose humana. Com a descoberta destas proteínas, novos estudos serão necessários para esclarecer o papel funcional das mesmas no S. mansoni.

So as to apply this perspective to a concrete historical situation, the theory of the aesthetic imagination will be used in an attempt to integrate vermeer's concept of painting, huygens's concept of optics, and spinoza's concept of philosophy. In each of these men's work, euclid's geometry is a fundamental presence. Please note that vermeer, spinoza and huygens not only shared a common geographical origin but also a common period of time. On the philosophical "borders" of this work two fundamental figures arise: ignace of loyola and descartes. They are briefly analyzed so as to establish connections between this present piece of research and conceptions which either preceded it

Full textAbdominal angiostrongyliasis is caused by Angiostrongylus costaricensis, an intra-arterial nematode, that lives in the ileocecal region in wild rodents. This parasite has been detected from southern United States to northern Argentina. Man is an accidental host and is infected ingesting third stage larvae (L3) that are eliminated with mucous secretions by the intermediate host (veronicelid slugs). Another species, that also may infect man is Angiostrongylus cantonensis, a rat pulmonary worm, responsabile for eosinophilic meningitis, in Asia and Pacific islands. Parasites of parasites are currently being studied not only to open new therapeutics possibilities, but also in order to improve diagnostic techniques. The interest for Wolbachia, a gram-negative endosimbiont bacterium, increased when the mutualistic character of its association with filarias was described. These considerations led to new ideas for treatment of these parasitosis through the use of antibacterial drugs. The main objective of this work is to verify the Wolbachia sp presence in A. costaricensis and A. cantonensis, and study its contribution for the humoral immune response of the vertebrate host. The first step was to look for evidences in favor of the presence of Wolbachia sp. Nucleic acids, through the PCR technique. In some experiments amplification products were obtained, what could be an indication of the presence of the bacterium, but these data must be confirmed by electronic microscopy and immunohistology. Because of the difficulties to get Wolbachia sp. antigen, alternatively serum samples from individuals with abdominal angiostrongyliasis were tested against Escherichia coli antigen, because it is a common bacterium species in vertebrates’ microbiota that could eventually colonize the worm. Through a western-blot analysis it was clearly demonstrated a reactivity not exclusively associated to A. costaricensis’ infected individuals. Moreover, fragments of worms were introduced in bacterial culture medium in order to study the microbiota of the Angiostrongylus adult worm. The fact of being found only one species of a grampositive bacillum in this experiment seems to confirm the hypothesis that in intravascular environment, with a low tolerance for bacteria, the worm’s microbiota is reduced in number and diversity. From the experiments we were not able to identify the presence of Wolbachia sp neither in A. costaricensis nor in A. cantonensis. It remains open to further investigations the contribution of other bacteria or simbionts of helminthes, for antigenic recognition by the vertebrate host, with potential implications for diagnosis, pathogenesis and treatment
A angiostrongilíase abdominal é causada pelo Angiostrongylus costaricensis, um nematódeo intra-arterial, que vive na região íleo-cecal de roedores silvestres. Esta parasitose tem sido registrada desde o sul dos Estados Unidos até o norte da Argentina. O homem é hospedeiro acidental e se infecta ingerindo as larvas de terceiro estágio (L3) presentes no muco do hospedeiro intermediário (veronicelídeos). Outra espécie, que também é parasita do homem, é Angiostrongylus cantonensis, um verme pulmonar de ratos, causador da meningite eosinofílica, que ocorre na Ásia e ilhas do Pacífico. Parasitas de parasitas são atualmente alvo de estudos não somente para abrir novas possibilidades terapêuticas, bem como para aprimorar técnicas diagnósticas. O interesse pela Wolbachia sp, uma bactéria gram-negativa endosimbionte, aumentou no momento em que descobriram sua característica mutualística em relação à filária. Estas considerações levaram a novas idéias para o tratamento destas parasitoses através da utilização de drogas antibacterianas. O objetivo principal deste trabalho é verificar a presença de Wolbachia em A. costaricensis e em A. cantonensis, e estudar a sua contribuição para a resposta imune humoral do hospedeiro vertebrado. O primeiro passo foi buscar evidências da presença de ácidos nucléicos de Wolbachia, através da técnica de PCR. Em alguns experimentos foram obtidos produtos de amplificação, o que poderia ser um indício da presença da bactéria, mas esses dados devem ser confirmados por microscopia eletrônica e por imunohistologia. Diante das dificuldades para se obter antígeno de Wolbachia sp alternativamente amostras de soro de indivíduos com angiostrongilíase foram testados contra antígenos de Escherichia coli por ser uma bactéria comum na microbiota de vertebrados e que eventualmente poderia colonizar o verme. Através da análise por imunoeletrotransferência ficou claramente demonstrada uma reatividade não relacionada exclusivamente aos indivíduos infectados por A. costaricensis. Além disso, fragmentos de vermes foram semeados em meio de cultura a fim de estudar a microbiota do verme adulto de Angiostrongylus. O fato de só ser encontrada um bacilo gram positivo nesse experimento parece confirmar a hipótese de que, por ser o ambiente intravascular pouco tolerante à presença de bactérias, a microbiota do verme deve ser pouco numerosa e diversa. Permanece aberta para futuras investigações a contribuição de outras bactérias ou outros simbiontes em helmintos, para reconhecimento antigênico pelo hospedeiro vertebrado, com possíveis implicações para diagnóstico, patogenia e tratamento.

Full textSchistosomiasis is a neglected tropical disease that affects approximately 200 million people around the world, occurring in America, Africa, the Antilles, Middle East and Near East, besides Southeast Asia. The species found in Brazil is Schistosoma mansoni, the typical treatment being administration of either Praziquantel or Oxamniquine. Although, the infection characteristics of this disease is associated with poor sanitation and hardened socio-economic conditions, resulting in high reinfection rates, which motivates the search for antihelmintic drugs and vaccines that overcome this situation. In this study proteomics techniques are used in the search of proteins potencially involved in the differentiation of individuals of both sexes, in the interactions between them and between the worms and the host. Samples of worm syncytium and adult whole worms of both male and female are studied by two-dimentional electrophoresis, while low molecular weight syncytium proteins from male and female adult worms were investigated by gel-LC. The differential protein expression in the syncytium investigated by gel-LC was analyzed statistically, being detected 5 proteins most abundant in males, and 2 in females, while 6 were identified solely on males and 21 on females. The information concerning protein differential expression allows the investigation of survival strategies developed evolutionarily by these parasites.
