Full textDirect numerical simulation of two-phase ow is used extensively for engineering research and fundamental fluid physics studies [54, 81]. This study is based on the Volume-Of-Fluid (VOF) method, originally created by Hirt and Nicols [30]. This method has gained increased popularity, especially when geometric advection techniques are used coupled with a planar reconstruction of the interface [14, 89]. The focus of the first part of this work is to investigate the hydrodynamics of isothermal cavitation in large bubble clouds, which originated from a larger study of micro-spalling [61], conducted by the French CEA. A method to deal with volume-changing vapour cavities, or pores, was formulated and implemented in an existing code, PARIS . The ow is idealized by assuming an inviscid liquid, negligible thermal effects and vanishing vapour pressure. A novel investigation of bubble cloud interaction in an expanding liquid using direct or detailed numerical simulation is presented. The simulation results reveal a pore competition, which is characterised by the Weber number in the ow. In the second part of the study the governing equations are extended to describe incompressible ow with phase change [79]. The description of the work commences with the derivation of the governing equations. Following this, a novel, geometric based, VOF solution method is proposed. In this method a novel way of advecting the VOF function is invented, which treats both mass and energy conservation in conservative form. New techniques include the advection of the interface in a discontinuous velocity field. The proposed algorithms are consistent and elegant, requiring minimal modifications to the existing code. Numerical experiments demonstrate accuracy, robustness and generality. This is viewed as a significant fundamental development in the use of VOF methods to model phase change

Full textScania CV AB är en ledande koncern inom utveckling och produktion av bussar, lastbilar samt industri- och marinmotorer. Nya lagkrav för miljö och säkerhet ställer ständigt högre krav på de tillverkade produkterna. Ett nära förestående lagkrav för lastbilar, Euro 6, innebär att gasläckage från motorn ska detekteras under körning. Om läckaget förutom att detekteras också kan isoleras, det vill säga särskiljas från andra fel, går reparationen i verkstaden snabbare då man slipper lokalisera läckaget. En snabbare reparation ökar up-time, det vill säga tiden som lastbilen kan användas på åkeriet. I detta exjobb används befintliga metoder för residualgenerering framtagna på Scania för att åstadkomma modelbaserad läckagediagnos. Arbetet tar även fram mätdata för olika givarfel samt för två läckage i motorn. Denna mätdata används för att utvärdera det erhållna diagnossystemets faktiska prestanda. Resultatet, som bygger på residualerna som metoden genererat, visar att läckage går att detektera, och att läckagen går att isolera från fel på tryckgivarsensorer på laddluftssidan och avgassidan. Denna isolering anses vara den svåraste att uppnå av alla sensorfel samt läckage varvid övrig isoleringsprestande verkar lovande. Däremot behövs mer mätdata för att säkert kunna fastställa övrig isoleringsprestanda. Diagnosmetoden lämpar sig troligen för exekvering i realtid ombord på lastbilen.

Full textSubmarine communications are the main usage of the very low frequency (VLF). This frequency band allows to radiate up to a few tens meter of depth and to a very long distance. Antennas for such telecommunications are necessarily small in regard to the wavelength. However, these structures are composed of hundred meters of thin metallic cables and their locations are often chosen for their dielectric characteristics or the structural advantage provided by the relief. To evaluate such antennas, we propose to use a home-made software, based on the TLM method. Such technique can be efficient for studying wide band electromagnetic problems in complex dielectric environment. Then we have improved the TLM Thin Wire model and we have evaluated its performances in realistic environment. In this document, we present our work and its validation by comparing our results to those obtained with the commercial software FEKO, based on the MoM, considered as the most suitable technique for this kind of problem. A solution assuring a good accuracy of the model for an arbitrary orientation of the Thine Wire in the 3D cartesian grid was proposed. We have also specified the limitations of the bent wire and the wires junction. In addition, we have studied the interaction between the wire and inhomogeneous media. This is an ambitious problem for which we brought some elements of answer but which remains a challenge. Finally, we have tested our software on some realistic antenna systems. The simulations of a valley span T antennas system allow to understand the functioning of such radiating structure and to show the interest of our method. The computation times are significantly lower with the TLM method than with FEKO to deal with antennas above such complex ground. However, the simulations of a Trideco antenna with aerial or buried radial ground plane showed the actual limitations of the TLM software which remains handicapped by an insufficient accuracy of the wires junction model and the inhomogeneous media interactions

Full textThis PhD thesis shows the development of a numerical method for solving two-phase flows with vaporisation. A coupling between Level Set and VOF methods is realised for the interface capturing. In order to solve fluid mechanics equations, a pressure based method is employed and, to decouple velocity and pressure, a projection method is performed. This method allows the implicitation of the acoustic terms and the time step constraint reduction. Liquid and gas are considered as compressible allowing local density variations with equations of state. The vaporisation is computed in two different ways ; a first one where the vaporisation rate is constant and a second one, where this rate is calculated by thermodynamics. Along with this topic, a study on curvature distribution in a liquid injection configuration is realised. This study allows to extend the drop size distribution concept in a spray and to improve available informations on ELSA model. Finally, an other study is performed on thedevelopment of a criterion, based on interface curvatures, which estimates the quality of a simulation

Full textThe study of the plasma spraying process is investigated in this work, in particular the interaction between a liquid phase and a plasma flow. Different physical phenomena occureas the jet fragmentation, the liquid vaporization and the particles fusion. The numerical simulation is used to better understand the physical phenomena and is an alternative to experimental measurements, which could be locally difficult to perform because of the measurement techniques limitations and the particular characteristics of the plasma. A compressible two-phase model is developped to take into account the compressible effects of the plasma flow. Validations are realized on academic test cases and on a argonjet. Good agreements are obtained between simulations and theory or experiments. Then different mixtures of plasmagen gas, classically used in plasma spraying, are studied :argon/hydrogen and argon/helium. An analysis of unsteady and turbulent character isled and allows seeing the importance of unsteady effects in these flows. Numerical simulations of the injection of a liquid phase into the plasma flow are realized. The influence of the plasmagen gas nature and the injection type is anlysed. Structures observed by experiments are also obtained by the simulations

Full textWe study in this work the modelling of two-phase cocurrent downflows in fixed bed reactors, a process widely used in industry. Due to the flow complexity, i.e., the presence of different interface configurations and, therefore, different phase interactions, most models have been developed using empirical approaches, with the assumption of a homogeneous flow in the reactor. However, several studies showed that local heterogeneities, geometric and hydrodynamic, such as the liquid distribution, could have a great influence on the flow at the reactor-scale and, therefore, on the reactor performance. Consider the microscopic phenomena in a macroscopic model require the use of multi-scale approaches. However, due to the flow complexity, the upscaling cannot be done directly and requires the development of modelling tools suitable for an intermediate scale. In a first step, the direct numerical method \ Volume-Of-Fluid" (VOF) is validated in the case of a two-phase flow in a capillary tube with the presence of a thin film. Then, this method is used, at a microscopic level to propose and validate closure laws for a pore-network model which will be used to simulate the flow at the intermediate scale. This upscaling approach is first tested in a two-dimensional case,i.e., an array of cylinders between two walls. This configuration allows the set up of an experimental approach, coupled with 2D VOF simulations at the intermediate scale, in order to validate the pore-network approach. The pore-network approach is then extended to a real fixed bed, i.e. in three dimensions, whose geometry is obtained by micro-tomography. Local laws of the pore-network model are redefined using direct numerical simulations at a microscopic scale. Pore-network simulations are then compared, for a homogenous phase distribution, with 1D models typically used for two-phase flow in fixed beds. Finally, an experimental campaign was set up to observe, by imaging scanner, the spreading of a liquid jet on a fixed bed pilot. A qualitative comparison is then performed between experimental observations and pore-network simulations in the specific case of the spreading of a liquid jet

Full textThe aim of this study is to propose an adapted model for the simulation of mould filling that must be a compromise solution between computational time and results accuracy. The double difficulty is to take into account the filling phenomenon that is a complex problem due to the presence of free boundaries and to the Glass specificities: viscosity that is highly thermal dependant and high melting temperature that requires taking into account radiation effects. Chapter I is devoted to the melting Glass flow. The numerical libraries Aquilon/Thétis, adapted for solving such type of problems and the thermal coupling between Air/Glass/Walls, has been used. (V.O.F method for front tracking, Augmented Lagrangian/Vector Projection methods for solving Pressure/Velocity coupling). For radiative aspect, different approaches are proposed: equivalent radiative conductivity (Chapter II), direct explicit method for validation (Chapter III) and spherical harmonics method or PN method (Chapter IV). In the Chapter V, the selected PN method is validated through simple cases and is then applied in other cases with convective coupling in complex geometries including semi-transparent inclusions (1D, 2D and 3D, 2D axi-symmetric and non grey medium). A P1 modified version is presented. The results are close to those given by P3 method but with reduced computational time. The main interest of this model is that it can be easily implemented in existing numerical codes: a single stationary second order partial differential equation to solve in 3D

In recent years, as the cost of aircraft design is growing rapidly, and aviation industry is interested in saving time and cost for the design, an accurate design result during the early design stages is particularly important to reduce overall life cycle cost. The purpose of the work to reducing the design cost at the early design stage with design accuracy as high as that of the detailed design. The method of an efficient global optimization (EGO) with variable-fidelity analysis and multidisciplinary design is proposed. Using the variable-fidelity analysis for the function evaluation, high fidelity function evaluations can be replaced by low-fidelity analyses of equivalent accuracy, which leads to considerable cost reduction. As the aircraft system has sub-disciplines coupled by multiple physics, including aerodynamics, structures, and thermodynamics, the accuracy of an individual discipline affects that of all others, and thus the design accuracy during in the early design states. Four distinctive design methods are developed and implemented into the standard Efficient Global Optimization (EGO) framework: 1) the variable-fidelity analysis based on error approximation and calibration of low-fidelity samples, 2) dynamic sampling criteria for both filtering and infilling samples, 3) a dynamic fidelity indicator (DFI) for the selection of analysis fidelity for infilled samples, and 4) Multi-response Kriging model with an iterative Maximum Likelihood estimation (iMLE). The methods are validated with analytic functions, and the improvement in cost efficiency through the overall design process is observed, while maintaining the design accuracy, by a comparison with existing design methods. For the practical applications, the methods are applied to the design optimization of airfoil and complete aircraft configuration, respectively. The design results are compared with those by existing methods, and it is found the method results design results of accuracies equivalent to or higher than high-fidelity analysis-alone design at cost reduced by orders of magnitude.

Full textA 3-D full-wave model, based on the integral representation of the electric field and dedicatedto the analysis of bi-static scattering mechanisms by a forest in the VHF and UHF bands wasefficiently enhanced. In order to overcome the limitation of a previous 3D model to small simulationsscenes and low frequencies, we have developed, during this research work, a new model using basis functionsadapted to the problem of interest, in the context of the Characteristic Basis Function Method(CBFM) and we investigated the suitability of this direct method for computing the electromagneticfields inside and outside three-dimensional dielectric objects representing the tree trunks and branches.The CBFM has shown great performances, when applied to the forest scattering modeling, both interms of CPU time and memory resources needed. Once properly set, the CBFM-E is so efficient thatit is able to treat in few minutes electromagnetic problems totally intractable with the classical MoM.Consequently, we have developed a powerful 3D forest electromagnetic scattering tool which allows ustoday to compute large forest electromagnetic problems in few minutes without worrying about theaccuracy of the solution. On the other hand, we have demonstrated the efficiency and accuracy of theCBFM-E when applied to 3D dielectric objects in the context of the electric volumetric integral equation,and have consolidated thus its leading position in the computational electromagnetics, especiallyagainst the iterative solvers based numerical methods

Full textImpact craters are formed when a large meteorite or comethits the Earth. At the impact a shock wave is released causingabundant fracturing in the surrounding bedrock. This type offracturing is intense and occurs throughout a very large volume(>100 km3) of the bedrock. Fractures of this type have beenobserved in deep drilling, to 5 km depth, in thePuchezh-Katunki Impact Crater. At theses depths the ambienttemperature is high. Thus impact structures are candidates forpotential heat-exchange sources for extraction of geothermalenergy.
There is a relation between fracture intensity and electricresistivity in bedrock predominated by impact-generatedfractures. In crystalline bedrock changes in electricresistivity is mainly due to fracturing which is the mainsource of porosity in these rocks. Electric resistivity methodsare highly sensitivity to porosity. Furthermore highfracture-intensities have generally been associated with lowelectric resistivity. Electro-magnetic methods like Very LowFrequency Resistivity (VLF-R) and Magnetotellurics (MT) canindirectly measure electric resistivity to relatively largedepths in the bedrock.
This study will quantify the relationship between fractureintensity and electric resistivity which can be used as aprospecting tool for geothermal energy resources at largedepth.
To meet that end, a method for fracture mapping on outcropsin Swedish terrain and a method to calculate thethree-dimensional fracture frequency from two-dimensionalfracture data has been developed. The fracture traces measuredin two dimensions on outcrops are assumed to represent avertical surface and must be converted to a three-dimensionalmeasure of the fracture frequency per unit volume. Spacing, dipand trace length of fractures have been accounted for. Thebiases associated with the mapping method have also beenaccounted for (II).
The correlation between impact-induced fracturing andelectric resistivity in crystalline rocks in the Lockne Areashows that the extent of impact fracturing in crystalline rockscan be measured with electro-magnetic or electric techniques.In addition the electric resistivity of crystalline basementand impact generated Tandsby Breccia from the Lockne Craterwere determined (I).
The relation between fracture frequency and electricresistivity in fresh water conditions using the VLF-R method isestablished from data collected from both two drill holes andfrom numerous outcrops in the Björkö region. Apreliminary quantification of the fracture frequency has beenmade. The MT resistivity models, related to the two drillholes, show that porosity and mineral-conductivity variationsof the bedrock affect this relation more than the salinityvariations in the bore-hole fluid. Further research is neededto establish a firm relation between fracture frequency,salinity of rock fluid, conductivity and porosity in order tovalidate the MT resistivity models (III).

Full textA 2D numerical modeling of flows in the swash zone is proposed using a Volume-Of- Fluid code. The detailed flow structure of a bore-driven swash event over an impermeable beach is first studied. The numerical model is then developed to account for porous media flow within the beach. The unique VARANS momentum equation and 1-fluid method used allow to solve simultaneously both surface and subsurface flows of air and water phases in the swash zone. This model is validated against a series of analytical tests cases and confronted to recent experimental measurements. The numerical results highlight the ability of the VOF-VARANS model to reproduce swash flows over and within a permeable beach

This work focuses onthe build-up of a representative volume of the coat considering a great range of phenomenonappearing in APS process such as gas flow properties, kinetic (multiphase flow,wettability) and thermal (heat transfers, thermal contact resistance, solidification) duringthe impact and steaking of particles. An original numerical method, named VOF-SM(Volume Of Fluid - Sub Mesh) is developped. The simulation of the impact of an unsteadyand turbulent ArH2 plasma flow is carried out in order to define the gas flow closeto the wall and heat transferred to the substrate by the plasma. Specific phenomena of theimpact of particles were incorporated into the CFD code (Thétis) and validated independentlyby caparison with analytical solutions, then together combined by the comparisonto a millimeter size impact experimental data. Full textImmunoassays are based on the use of antibodies that selectively bind the analyte. The trueness of the results obtained depends to a great extent on the selectivity of the antibodies and can be affected by interferences. In this study, a method was developed in which the sample is separated into fractions by using high-performance liquid chromatography (LC) and these fractions are measured using an enzyme-linked immunosorbent assay (ELISA). This method was referred to as LC-ELISA. The profile obtained from the measured analyte concentration by ELISA as a function of the elution time was referred to as LC-ELISAgram and offers the possibility to detect interferences which otherwise remain undetected during the ELISA. Sulfamethoxazole (SMX), carbamazepine (CBZ) and estrone (E1) were used as model analytes for the ELISA and LC-ELISA measurements. Various environmental matrices such as surface water and wastewater were examined for their interference in the respective ELISA. The good quantification properties of the validated LC-ELISA have been used to demonstrate an approach to distinguish between specific and non-specific interferences from environmental samples. By this approach and application of acidic hydrolysis of the sample, it was possible to detect a previously unknown metabolite of SMX and estimate its cross-reactivity to probably 460 ± 150%. Furthermore, a new tracer was developed in a linear 13-step synthesis, which resulted in the novel conjugation of the peroxidase at the N1-position of SMX. The new hapten was also used for the synthesis of a novel immunogen.

Full textThe focus of the thesis was the characterization of a monoclonal antibody, which binds specifically to cisplatin-treated DNA and is already used for diagnostic purposes in cancer therapy. The structure and composition of the antibody, as well, as the antigen specificity were investigated by mass spectrometry. Using nano-ESI-MS it was possible to analyse the antibody in its native state. Tandem-MS experiments revealed not only specific antibody fragments but also gave information on the primary structure of the antibody. The Fc-bound glycans that are responsible for antibody structure and functionality, were cleaved enzymatically and studied with MALDI-MS. For a detailed determination of the primary structure, the antibody was proteolysed with different enzymes and analyzed via high resolution Tandem-MS. The data was evaluated by comparism with results from sequence analysis on DNA level and known antibody sequences from protein databases. Simultaneously many different posttranslational modifications were identified. Furthermore, the antibody was successfully labelled with a metal complex at its sugar structures without loss of its structure and function. For investigation of the antibody specificity antigens were generated by incubation of well-defined single and double stranded DNA oligomers with cisplatin. Antibody-antigen-complexes were synthesized and analyzed by mass spectrometry under native conditions. The stability of the complex with double stranded DNA was also investigated by MS/MS-experiments. Thus, it was also possible to determine the specific fragmentation behavior for the complex just as for the free antibody.

Full textHepcidin-25, a key iron-regulatory peptide hormone discovered in 2000, has revolutionized the understanding of iron-related pathology. This study applied LC-MS/MS, using the triple quadrupole mass spectrometer, in a rapid and robust analytical strategy for the quantification of hepcidin-25 in human serum, to be implemented in routine laboratories. For this purpose, two sample preparation strategies and two complementary LC conditions were investigated, where the use of acidic mobile phases (0.1% TFA) was compared with a novel approach involving solvents at high pH (0.1% NH3). The application of these LC-MS/MS methods to human samples in an intra-laboratory comparison, using the same hepcidin-25 calibrators, yielded a very good correlation of the results. The LC-MS/MS employing trifluoroacetic acid-based mobile phases was selected as a highly sensitive (LOQ of 0.5 µg/L) and precise (CV<15%) method and was recommended as a reference method candidate for hepcidin-25 quantification in real samples. One of the novel aspects of the methodology was the use of amino- and fluoro-silanized autosampler vials to reduce the interaction of the 25-residue peptide to laboratory glassware surfaces. Moreover, this LC-MS/MS method was used for an international round robin study, applying a secondary reference material as a calibrator and its performance was found to be in the optimal range as defined by the International Consortium for Harmonization of Clinical Laboratory Results (ICHCLR). In this work, the formation of hepcidin-25 complexes with copper(II) was investigated. The first reversed-phase chromatographic separation of hepcidin-25/Cu2+ and hepcidin-25 (copper “free”) was achieved by applying mobile phases containing 0.1% NH3. LC-MS/MS and FTICR-MS were applied for the characterization of the formed hepcidin-25-Cu(II) species at pH values of 11 and 7.4 respectively. A new species corresponding to hepcidin-25 complexed with two copper ions was identified at high pH.

Full textThe motion of a nitrogen Taylor bubble in glycerol-water mixed solutions rising through different types of expansions and contractions is investigated by a numerical approach. The CFD procedure is based on an open-source solver Basilisk, which adopts the volume-of-fluid (VOF) method to capture the gas-liquid interface. The results of sudden expansions/contractions are compared with experimental results. The results show that the simulations are in good agreement with experiments. The bubble velocity increases in sudden expansions and decreases in sudden contractions. The bubble break-up pattern is observed in sudden expansions with large expansion ratios, and a bubble blocking pattern is found in sudden contractions with small contraction ratios. In addition, the wall shear stress, the liquid film thickness, and pressure in the simulations are studied to understand the hydrodynamics of the Taylor bubble rising through expansions/contractions. The transient process of the Taylor bubble passing through sudden expansion/contraction is further analyzed for three different singularities: gradual, parabolic convex and parabolic concave. A unique feature in parabolic concave contraction is that the Taylor bubble passes through the contraction even for small contraction ratios. Moreover, a phase change model is developed in the Basilisk solver. In order to use the existed geometric VOF method in Basilisk, a general two-step geometric VOF method is implemented. Mass flux is calculated not in the interfacial cells but transferred to the neighboring cells around the interface. The saturated temperature boundary condition is imposed at the interface by a ghost cell method. The phase change model is validated by droplet evaporation with a constant mass transfer rate, the one-dimensional Stefan problem, the sucking interface problem, and a planar film boiling case. The results show good agreement with analytical solutions or correlations

Full textThe explosive Pentaerythritol tetranitrate (PETN) is extremely difficult to detect. An improved antibody against PETN was developed by using the bioisosteric concept. These polyclonal antibodies are highly selective and sensitive. The limit of detection (LOD) of the ELISA was determined to be 0.15 µg/L. The dynamic range of the assay was found to be between 1 and 1000 µg/L. The antibodies are sufficiently pH-stable and resistant to solvent additives. An HPLC-compatible TNT-affinity column with porous glass as support material was prepared for the environmental analysis. In order to isolate the anti-TNT antibodies of the TNT sera a separation was carried out on a dinitrophenyl-affinity column. To optimize the immobilization method, orange-coloured dabsyl proteins were synthesized and bound to the surface. The colour intensity was found to be an indicator for the immobilization rate. In consequence of the high affinity constants of the anti-TNT antibodies, TNT can''t elute by a typical acidic elution step. Therefore, a novel separation approach, the thermal online-elution was developed. The maximum capacity of an affinity column was 650 ng TNT or 10 µg/mL of column volume. To quantify the immobilization rate of proteins, a new method has been developed, because the usual protein determination methods were unsuitable. Therefore an HPLC separation method of Tyr and Phe was developed without prior derivatization. Two internal standard compounds, HTyr and FPhe, were used for calibration. The LOD was estimated to be 0.05 µM (~ 10 µg/L) for Tyr and Phe at 215 nm. The protein hydrolysis time was reduced from 22 h to 30 min using microwave technique. This procedure, that was termed aromatic amino acid analysis (AAAA), has been validated for protein determination of homogeneous samples with NIST-BSA. The LOD for proteins was calculated to be below 16 mg/L (~ 300 ng BSA absolute). The relative standard deviation, including the hydrolysis step, is 5%.

Full textEl estudio y modelado de flujos bifásicos, incluso los más simples como el bubbly flow, sigue siendo un reto que conlleva aproximarse a los fenómenos físicos que lo rigen desde diferentes niveles de resolución espacial y temporal. El uso de códigos CFD (Computational Fluid Dynamics) como herramienta de modelado está muy extendida y resulta prometedora, pero hoy por hoy, no existe una única aproximación o técnica de resolución que permita predecir la dinámica de estos sistemas en los diferentes niveles de resolución, y que ofrezca suficiente precisión en sus resultados. La dificultad intrínseca de los fenómenos que allí ocurren, sobre todo los ligados a la interfase entre ambas fases, hace que los códigos de bajo o medio nivel de resolución, como pueden ser los códigos de sistema (RELAP, TRACE, etc.) o los basados en aproximaciones 3D TFM (Two-Fluid Model) tengan serios problemas para ofrecer resultados aceptables, a no ser que se trate de escenarios muy conocidos y se busquen resultados globales. En cambio, códigos basados en alto nivel de resolución, como los que utilizan VOF (Volume Of Fluid), requirieren de un esfuerzo computacional tan elevado que no pueden ser aplicados a sistemas complejos. En esta tesis, mediante el uso de la librería OpenFOAM se ha creado un marco de simulación de código abierto para analizar los escenarios desde niveles de resolución de microescala a macroescala, analizando las diferentes aproximaciones, así como la información que es necesaria aportar en cada una de ellas, para el estudio del régimen de bubbly flow. En la primera parte se estudia la dinámica de burbujas individuales a un alto nivel de resolución mediante el uso del método VOF (Volume Of Fluid). Esta técnica ha permitido obtener resultados precisos como la formación de la burbuja, velocidad terminal, camino recorrido, estela producida por la burbuja e inestabilidades que produce en su camino. Pero esta aproximación resulta inviable para entornos reales con la participación de más de unas pocas decenas de burbujas. Como alternativa, se propone el uso de técnicas CFD-DEM (Discrete Element Methods) en la que se representa a las burbujas como partículas discretas. En esta tesis se ha desarrollado un nuevo solver para bubbly flow en el que se han añadido un gran número de nuevos modelos, como los necesarios para contemplar los choques entre burbujas o con las paredes, la turbulencia, la velocidad vista por las burbujas, la distribución del intercambio de momento y masas con el fluido en las diferentes celdas por cada una de las burbujas o la expansión de la fase gaseosa entre otros. Pero también se han tenido que incluir nuevos algoritmos como el necesario para inyectar de forma adecuada la fase gaseosa en el sistema. Este nuevo solver ofrece resultados con un nivel de resolución superior a los desarrollados hasta la fecha. Siguiendo con la reducción del nivel de resolución, y por tanto los recursos computacionales necesarios, se efectúa el desarrollo de un solver tridimensional de TFM en el que se ha implementado el método QMOM (Quadrature Method Of Moments) para resolver la ecuación de balance poblacional. El solver se desarrolla con los mismos modelos de cierre que el CFD-DEM para analizar los efectos relacionados con la pérdida de información debido al promediado de las ecuaciones instantáneas de Navier-Stokes. El análisis de resultados de CFD-DEM permite determinar las discrepancias encontradas por considerar los valores promediados y el flujo homogéneo de los modelos clásicos de TFM. Por último, como aproximación de nivel de resolución más bajo, se investiga el uso uso de códigos de sistema, utilizando el código RELAP5/MOD3 para analizar el modelado del flujo en condiciones de bubbly flow. El código es modificado para reproducir correctamente el flujo bifásico en tuberías verticales, comparando el comportamiento de aproximaciones para el cálculo del término d

En aquesta tesi s'ha desenvolupat un nou solver per a bubbly flow en el qual s'han afegit un gran nombre de nous models, com els necessaris per a contemplar els xocs entre bambolles o amb les parets, la turbulència, la velocitat vista per les bambolles, la distribució de l'intercanvi de moment i masses amb el fluid en les diferents cel·les per cadascuna de les bambolles o els models d'expansió de la fase gasosa entre uns altres. Però també s'ha hagut d'incloure nous algoritmes com el necessari per a injectar de forma adequada la fase gasosa en el sistema. Aquest nou solver ofereix resultats amb un nivell de resolució superior als desenvolupat fins la data. Seguint amb la reducció del nivell de resolució, i per tant els recursos computacionals necessaris, s'efectua el desenvolupament d'un solver tridimensional de TFM en el qual s'ha implementat el mètode QMOM (Quadrature Method Of Moments) per a resoldre l'equació de balanç poblacional. El solver es desenvolupa amb els mateixos models de tancament que el CFD-DEM per a analitzar els efectes relacionats amb la pèrdua d'informació a causa del promitjat de les equacions instantànies de Navier-Stokes. L'anàlisi de resultats de CFD-DEM permet determinar les discrepàncies ocasionades per considerar els valors promitjats i el flux homogeni dels models clàssics de TFM. Finalment, com a aproximació de nivell de resolució més baix, s'analitza l'ús de codis de sistema, utilitzant el codi RELAP5/MOD3 per a analitzar el modelatge del fluxos en règim de bubbly flow. Full textCovalent posttranslational modifications (PTMs) influence the structure and function of proteins. Protein phosphorylations belong to the most important PTMs. Rarely characterized labile phosphorylations, for instance phosphorylations of cysteine and lysine and pyrophosphorylations of serine and threonine residues got into the focus of science. However, the analysis of those delicate modifications via tandem mass spectrometry remains a challenge. In the present work, it is shown that electron-transfer dissociation (ETD) combined with HCD supplemental activation (EThcD) is able to fragment peptides with labile phosphorylations at the side chains without losing the modification. In several bottom-up proteomic approaches, EThcD allowed the reliable identification of a naturally occurring cysteine phosphorylation. Moreover, methods for identification of lysine phosphorylations were developed. For the proteome wide analysis of lysine phosphorylations, considering the lability, a bottom-up phosphoproteomic approach with a highly selective mass spectrometry method was established. The MS-method relies on the generation of diagnostic phospholysine immonium ions during HCD, which trigger in a second step an additional EThcD spectrum of the same precursor ion. This strategy ensures the confident identification of lysine phosphorylated peptides. Furthermore, the present work shows that isobaric pyro- and diphosphorylated peptides differ in their neutral loss pattern during CID. This behavior was a key step in a specific neutral loss triggered EThcD method, which enabled the reliable identification of pyrophosphorylations. This method allowed the identification of the first protein pyrophosphorylations, a new endogenous PTM, in yeast and human embryonic kidney cells.

Full textThe present thesis focuses on an investigation of the sloshing phenomenon in a partially filled fuel tank submitted to a harmonic excitation motion. In the first part, the confrontation of numerical results between a linear approach - taking into account viscosity - and a nonlinear approach based on a commercial code leads to define a parameter of linearity. This parameter allows determining cases of sloshing who require non-linear resolution and those who need a linear theory to predict the phenomenon. An experimental study of fluid sloshing in a rectangular tank submitted to an automotive braking is conducted. Tests leaded allow measuring global forces engendered by the motion of the fluid, pressure of fluid impact and velocity field by PIV. This chapter provides an important data base and helps to investigate on the physical phenomenon. This study is completed by CFD results. To conclude, a numerical model for fluid-structure interactions is presented. Limits of this segregated partitioned coupling in case of sloshing in tank flexibly attached are highlighted, depending mostly on the mass ratio between fluid and tank structure. An added-mass term is integrated to the corrected staggered scheme ensuring systematically the convergence of the coupled solution and reducing significantly the iterations required
