Abstract:
Abstract Vegetative incompatibility in fungi has long been known to reduce the transmission of viruses between individuals, but the barrier to transmission is incomplete. In replicated laboratory assays, we showed conclusively that the transmission of viruses between individuals of the chestnut blight fungus Cryphonectria parasitica is controlled primarily by vegetative incompatibility (vic) genes. By replicating vic genotypes in independent fungal isolates, we quantified the effect of heteroallelism at each of six vic loci on virus transmission. Transmission occurs with 100% frequency when donor and recipient isolates have the same vic genotypes, but heteroallelism at one or more vic loci generally reduces virus transmission. Transmission was variable among single heteroallelic loci. At the extremes, heteroallelism at vic4 had no effect on virus transmission, but transmission occurred in only 21% of pairings that were heteroallelic at vic2. Intermediate frequencies of transmission were observed when vic3 and vic6 were heteroallelic (76 and 32%, respectively). When vic1, vic2, and vic7 were heteroallelic, the frequency of transmission depended on which alleles were present in the donor and the recipient. The effect of heteroallelism at two vic loci was mostly additive, although small but statistically significant interactions (epistasis) were observed in four pairs of vic loci. A logistic regression model was developed to predict the probability of virus transmission between vic genotypes. Heteroallelism at vic loci, asymmetry, and epistasis were the dominant factors controlling transmission, but host genetic background also was statistically significant, indicating that vic genes alone cannot explain all the variation in virus transmission. Predictions from the logistic regression model were highly correlated to independent transmission tests with field isolates. Our model can be used to estimate horizontal transmission rates as a function of host genetics in natural populations of C. parasitica.

Abstract:
ABSTRACT Vegetative incompatibility in the chestnut blight fungus,Cryphonectria parasitica, in Europe is controlled by six unlinked vic loci, each with two alleles. Four previously identified vic loci (vic1, vic2,vic3, and vic4) were polymorphic in European vegetative compatibility (vc) types. Two new loci, vic6 andvic7, also were identified among European vc types. In one cross, vic genes segregated independently at five loci, and 194 progeny were assigned to 32 vc types; none of these loci were linked. A total of 64 vc types were identified from all crosses. All 64 genotypes possible from six vic loci, each with two alleles (26 = 64), were identified and assigned to vc types. Based on our model, vc types v-c 5 and v-c 10, which had been used in previous genetic studies, differ by only five vic genes. Future studies of vc types in C. parasitica can use knowledge of vic genotypes for analysis of population genetic structure based on vic allele frequencies and to determine the effect of each vic gene on virus transmission between vc types.

Abstract:
AbstractSpatiotemporal features of anthocyanin accumulation in a model legume Lotus japonicus (Regel) K.Larsen were elucidated to develop criteria for the genetic analysis of flavonoid biosynthesis. Artificial mutants and wild accessions, with lower anthocyanin accumulation in the stem than the standard wild type (B-129 ‘Gifu’), were obtained by ethyl methanesulfonate (EMS) mutagenesis and from a collection of wild-grown variants, respectively. The loci responsible for the green stem of the mutants were named as VIRIDICAULIS (VIC). Genetic and chemical analysis identified two loci, namely, VIC1 and VIC2, required for the production of both anthocyanins and proanthocyanidins (condensed tannins), and two loci, namely, VIC3 and VIC4, required for the steps specific to anthocyanin biosynthesis. A mutation in VIC5 significantly reduced the anthocyanin accumulation. These mutants will serve as a useful system for examining the effects of anthocyanins and proanthocyanidins on the interactions with herbivorous pests, pathogenic microorganisms and nitrogen-fixing symbiotic bacteria, Mesorhizobium loti.

Abstract:
ABSTRACT During the last decade, the hns gene and its product, the H-NS protein, have been extensively studied in Escherichia coli. H-NS-like proteins seem to be widespread in gram-negative bacteria. However, unlike in E. coli and inSalmonella enterica serovar Typhimurium, little is known about their role in the physiology of those organisms. In this report, we describe the isolation of vicH, an hns-like gene in Vibrio cholerae, the etiological agent of cholera. This gene was isolated from a V. cholerae genomic library by complementation of different phenotypes associated with anhns mutation in E. coli. It encodes a 135-amino-acid protein showing approximately 50% identity with both H-NS and StpA in E. coli. Despite a low amino acid conservation in the N-terminal part, VicH is able to cross-react with anti-H-NS antibodies and to form oligomers in vitro. ThevicH gene is expressed as a single gene from two promoters in tandem and is induced by cold shock. A V. choleraewild-type strain expressing a vicHΔ92 gene lacking its 3′ end shows pleiotropic alterations with regard to mucoidy and salicin metabolism. Moreover, this strain is unable to swarm on semisolid medium. Similarly, overexpression of the vicH wild-type gene results in an alteration of swarming behavior. This suggests that VicH could be involved in the virulence process in V. cholerae, in particular by affecting flagellum biosynthesis.

Abstract:
The aim of this work was to compare, in terms of energy efficiency, the degradation of three organic compounds (4-chlorophenol, 4-nitrophenol and succinic acid) by several advanced oxidation processes or AOPs (electric discharge, UV-irradiation, ozonation, UV/H2O2, O3/H2O2, catalytic ozonation and photocatalysis) in aqueous solution, at pH 6. For all studied AOPs, 4-chlorophenol was well removed. Succinic acid was degraded only by the processes that generate OH radical, as well 4-nitrophenol which was also oxidized by ozone alone. A simplified pseudo-first-order kinetic law was applied correctly throughout the most part of the reaction. The energy applied per mole of compound removed (specific energy applied or Eas) was evaluated. The value of Eas was found to be independent of the initial concentration of the target compound. This allows us to compare experiments performed at different concentrations. The UV/H2O2 coupling was found to be the best AOP in terms of energy consumption. The ozonation alone was found to be also the best system to remove phenols with relatively low energy consumption. The degradation of saturated acids, such as succinic acid, can be reached by catalytic ozonation and photocatalysis but with low energetic yields

Vico s Scienza nuova introduces a proposal to science the Vico Variable which is clearly different from the prevailing variable, that of Modern Science as we understand it today, based on Newton s Mechanicism and in Descartes Rationalism, anchored in the Enlightenment of the 18th century and established, once and for all, in Comte s Positivism. In Vico, metaphysics no longer busies itself with the contemplation of and speculation about God, to bring its focus down to earth and seek, in History, that which had, until then, sought in the Divine Mind. To him, the knowledge of human affairs is that which can answer the question characteristic of all times concerning what Man is. The civil world of the Scienza nuova develops in the metaphysical realm, to whose study the new art of criticism is developed: a synergy between philosophical criticism and philological investigation, leading to Vico s hermeneutics, which takes advantage both of Reason and of the faculties of the spirit. In this hermeneutics, microcosm and macrocosm, man and society, particular and universal, are synthesized in constant analogies; the theory of the verum ipso factum is originally applied and the common sense of humanity becomes the supreme objective principle of historic evolution (a manifestation of Divine Providence) and as a subjective ground of a historic understanding. So as to situate nations in the development of the eternal ideal history , to evaluate its degrees of civility and the risks it is subject to, the Scienza nuova proposes, also, a diagnostic art, from which one may criticize the way which knowledge (Science and Education) is considered by the moderns in this time. Such criticism is not restricted to observations and considerations, but it sets the groundwork for an action it is, therefore, a Political Science through the proposal of a focus that is able to overcome the empire of the Mathesis Universalis, and thus, to contemplate, equally, the possibility of knowledge of the Ethical, Political and Human universes, interacting in a selfsame science. The Vico Variable is not only a way that has been forsaken on the road of the development of science; something which might have been. It is a philosophical orientation which privileges the practical life, the understanding of human behavior and civil life, which can be extended to thinkers who came after Vico, whose ideas allow the identification with outstanding aspects of his thought; a philosophical orientation which, identified with the emphasis Pico della Mirandola gave upon the Dignity of Man, can represent the emancipation of humanity

After situating Vico in the Naples of his time and in relation to the questions of modern rationality, as well as carrying out an exercise of philosophical reflection seeking the understanding of the worldview of Vico, this piece of work aims at making explicit the ethic that underlies his work as an ethic of teleological character which privileges the practical life, evidencing a concern for what man can and should do of himself, notwithstanding the guardianship of a divine providence in a cyclical history. However, within the advent of the "age of men," it is verified a process of unfoldment of the human nature in which the development of the individual starts to dictate the development of the community; at the same time, the predominance of abstract rationality dissociated from other human faculties – such as imagination – leads to a preponderant literality in the interpretation of mythology, which pervades the doctrines of religions, moving man away from piety, with the consequent loss of both social cohesion and the human sense of the world. Hence, man decays in the age of the gods, submitting himself to heteronomy in the face of the eternal ideal history. It is identified with the present thesis, therefore, the "master key" indicated by the Napolitan himself with the imagination that underlies poetic wisdom, the application of this interpretative key of the method of his work – the new critical art – is extended to the solution of the pretended paradox between the autonomy and the aimed happiness by a teleological ethic and the cyclical repetition of the eternal ideal history

Abstract: Vico has criticized the rationality of his time and he proposed another one as alternative: the key for this question is the concept of wit. Into his pedagogical program, Vico wished a synthesis among science, prudence, and eloquence. He suggested that it would be possible if one sharpen the young men's wit. Vico is not na irrationalist. His notion about the reason has humanist roots. He intended to overcome the Cartesian reason, in reaffirming the science of topical, the inventio, and the phantasy to enthrone the wit with a right faculty status of knowing. That allows him to make a profound study of the humanities. Vico has condemned the monachal and solitarian feature of the philosophies from the turn of the XVII century to XVIII century, and he prevented us against the evil of the "rational selfish" that society from his time was taken with. Vico is not a solitary thinker. Before seeing him as a forerunner or conservator, it is rather situate him in his own time, answering the last news of the republic letters since that time. The interest of the contribution that Vico emphasized in his discourses was put in practice into the context of the famous Quarrel between the Ancients and the Moderns, more specifically about the question of the Homer's status - a problem that demanded from Vico his wit. The four aspects of the wit, the acuity, the inventiveness, the citizeness prudence, and the historical presence, bring under light the rationality that was thought by Vico

Abstract:

Abstract:
Mechanical stimuli, including the elasticity of the extracellular matrix (ECM), can have profound effects on the function of cells and their responsiveness to other microenvironmental cues, thereby regulating homeostasis and disease development. For example, the response of aortic valve interstitial cells (VICs) to growth factors [1] and VIC differentiation to pathological phenotypes [2] depend on ECM elasticity. The ability of cells to sense and respond to mechanical stimuli depends on several factors, including their inherent cellular-level mechanical properties. The mechanical properties of suspended VICs [3, 4] and VICs grown on stiff glass/polystyrene [5] have been reported. However, neither of these test conditions is physiological, as VICs adhere to ECM that is orders of magnitude more compliant than glass. Some other cell types adapt their stiffness to that of their substrate [6]; we hypothesized that adherent VICs would similarly change their elasticity in response to the elastic properties of their ECM.
