Abstract:
Most renal transplants ultimately fail secondary to chronic allograft nephropathy (CAN). Vimentin (vim) is a member of the intermediate filament family of proteins and has been shown to be important in the development of CAN. One of the pathways leading to chronic renal fibrosis after transplant is thought to be epithelial to mesenchymal transition (EMT). Even though vim expression is one of the main steps of EMT, it is unknown whether vim expression is required for EMT leading to renal fibrosis and allograft loss. To this end, the role of vim in renal fibrosis was determined via unilateral ureteral obstruction (UUO) in vim knockout mice (129 svs6 vim −/−). Following UUO, kidneys were recovered and analyzed via Western blotting, immunofluorescence, and transcriptomics. Cultured human proximal renal tubular (HK-2) cells were subjected to lentiviral-driven inhibition of vim expression and then treated with transforming growth factor (TGF)-β to undergo EMT. Immunoblotting as well as wound healing assays were used to determine development of EMT. Western blotting analyses of mice undergoing UUO reveal increased levels of vim soon after UUO. As expected, interstitial collagen deposition increased in control mice following UUO but decreased in vim −/− kidneys. Immunofluorescence analyses also revealed altered localization of β-catenin in vim −/− mice undergoing UUO without significant changes in mRNA levels. However, RNA sequencing revealed a decrease in β-catenin-dependent genes in vim −/− kidneys. Finally, vim-silenced HK-2 cell lines undergoing EMT were shown to have decreased cellular migration during wound healing. We conclude that vim inhibition decreases fibrosis following UUO by possibly altering β-catenin localization and downstream signaling.

ABSTRACT A transferable plasmid from Klebsiella pneumoniae carried a class 1 integron containing bla VIM-12, a novel bla VIM-type gene, flanked by two copies of aacA7. bla VIM-12 was clustered between bla VIM-1 and bla VIM-2 and differed from bla VIM-1 by 18 nucleotides that were all located at the 3′ end and matched the corresponding nucleotides in bla VIM-2. The bla VIM-12-associated 59-base element was identical to that described in bla VIM-2 alleles.

Abstract:
In this paper, we present a comparative study between Sinc–Galerkin method and a modified version of the variational iteration method (VIM) to solve non-linear Sturm–Liouville eigenvalue problem. In the Sinc method, the problem under consideration was converted from a non-linear differential equation to a non-linear system of equations, that we were able to solve it via the use of some iterative techniques, like Newton’s method. The other method under consideration is the VIM, where the VIM has been modified through the use of the Laplace transform, and another effective modification has also been made to the VIM by replacing the non-linear term in the integral equation resulting from the use of the well-known VIM with the Adomian’s polynomials. In order to explain the advantages of each method over the other, several issues have been studied, including one that has an application in the field of spectral theory. The results in solutions to these problems, which were included in tables, showed that the improved VIM is better than the Sinc method, while the Sinc method addresses some advantages over the VIM when dealing with singular problems.

Abstract:
ABSTRACTThe metallo-β-lactamase VIM-4, mainly found inPseudomonas aeruginosaorAcinetobacter baumannii, was produced inEscherichia coliand characterized by biochemical and X-ray techniques. A detailed kinetic study performed in the presence of Zn2+at concentrations ranging from 0.4 to 100 μM showed that VIM-4 exhibits a kinetic profile similar to the profiles of VIM-2 and VIM-1. However, VIM-4 is more active than VIM-1 against benzylpenicillin, cephalothin, nitrocefin, and imipenem and is less active than VIM-2 against ampicillin and meropenem. The crystal structure of the dizinc form of VIM-4 was solved at 1.9 Å. The sole difference between VIM-4 and VIM-1 is found at residue 228, which is Ser in VIM-1 and Arg in VIM-4. This substitution has a major impact on the VIM-4 catalytic efficiency compared to that of VIM-1. In contrast, the differences between VIM-2 and VIM-4 seem to be due to a different position of the flapping loop and two substitutions in loop 2. Study of the thermal stability and the activity of the holo- and apo-VIM-4 enzymes revealed that Zn2+ions have a pronounced stabilizing effect on the enzyme and are necessary for preserving the structure.

Abstract:
We previously found that, among human parainfluenza virus type 3 (HPIV3) proteins, the interaction of nucleoprotein (N) and phosphoprotein (P) provides the minimal requirement for the formation of cytoplasmic inclusion bodies (IBs), which are sites of RNA synthesis, and that acetylated α-tubulin enhances IB fusion and viral replication. In this study, using immunoprecipitation and mass spectrometry assays, we determined that vimentin (VIM) specifically interacted with the N–P complex of HPIV3, and that the head domain of VIM was responsible for this interaction, contributing to the inhibition of IB fusion and viral replication. Furthermore, we found that VIM promoted the degradation of α-tubulin acetyltransferase 1 (α-TAT1), through its head region, thereby inhibiting the acetylation of α-tubulin, IB fusion, and viral replication. In addition, we identified a 20-amino-acid peptide derived from the head region of VIM that participated in the interaction with the N–P complex and inhibited viral replication. Our findings suggest that VIM inhibits the formation of HPIV3 IBs by downregulating α-tubulin acetylation via enhancing the degradation of α-TAT1. Our work sheds light on a new mechanism by which VIM suppresses HPIV3 replication.

Abstract:
ABSTRACT During a survey conducted to evaluate the incidence of class B carbapenemase (metallo-β-lactamase [MBL])-producing Pseudomonas aeruginosa strains from hospitals in Majorca, Spain, five clinical isolates showed a positive Etest MBL screening test result. In one of them, strain PA-SL2, the presence of a new bla VIM derivative (bla VIM-13) was detected by PCR amplification with bla VIM-1-specific primers followed by sequencing. The bla VIM-13-producing isolate showed resistance to all β-lactams (except aztreonam), gentamicin, tobramycin, and ciprofloxacin. VIM-13 exhibited 93% and 88% amino acid sequence identities with VIM-1 and VIM-2, respectively. bla VIM-13 was cloned in parallel with bla VIM-1, and the resistance profile conferred was analyzed both in Escherichia coli and in P. aeruginosa backgrounds. Compared to VIM-1, VIM-13 conferred slightly higher levels of resistance to piperacillin and lower levels of resistance to ceftazidime and cefepime. VIM-13 and VIM-1 were purified in parallel as well, and their kinetic parameters were compared. The k cat/K m ratios for the antibiotics mentioned above were in good agreement with the MIC data. Furthermore, EDTA inhibited the activity of VIM-13 approximately 25 times less than it inhibited the activity of VIM-1. VIM-13 was harbored in a class 1 integron, along with a new variant (Ala108Thr) of the aminoglycoside-modifying enzyme encoding gene aacA4, which confers resistance to gentamicin and tobramycin. Finally, the VIM-13 integron was apparently located in the chromosome, since transformation and conjugation experiments consistently yielded negative results and the bla VIM-13 probe hybridized only with the genomic DNA.

Abstract:
The vortex induced vibration VIV is a main subject studied in the offshore engineering, having its name adapted to VIM by the occurrence in Spar platforms in the Gulf of México. As the first monocolumn unit, the Sevan Piranema, was in Northeast Brazil in 2007, VIM occurrence in cylindrical units like this was questioned, having the mooring lines design dimensioned without this load. Firstly a detailed study about the works developed in this field was performed, and not being found enough publications about VIM in monocolumns, its characterization was aimed in terms of oscillation amplitude and period through model tests, due to the difficulty to obtain reliable results by means of analytical or numerical approaches. Later, with the objective of verifying the impact of VIM in extreme loads and, mainly, in fatigue damage, analyses with two different methods found in the literature were performed. The first consider the application of VIM as a motion in the numerical model, based on the experimental results, and the second applies the observed forces. For these studies the mooring system for a typical scenario was analyzed, under the wave, wind and current environmental force. At last, due to the importance observed from the results, there were discussed the relation of VIM with some external factors, like span wise relation, presence of waves and viscous damping, being presented for this last a comparison of the numerical model results with the experimental is presented.

Abstract:

Abstract:
The Random Forest model is commonly used as a predictor function and the model have been proven useful in a variety of applications. Their popularity stems from the combination of providing high prediction accuracy, their ability to model high dimensional complex data, and their applicability under predictor correlations. This report investigates the random forest variable importance measure (VIM) as a means to find a ranking of important variables. The robustness of the VIM under imputation of categorical noise, and the capability to differentiate informative predictors from non-informative variables is investigated. The selection of variables may improve robustness of the predictor, improve the prediction accuracy, reduce computational time, and may serve as a exploratory data analysis tool. In addition the partial dependency plot obtained from the random forest model is examined as a means to find underlying relations in a non-linear simulation study.

Abstract:
The International vocabulary of metrology ― Basic and general concepts and associated terms (VIM), 3rd edition, also known as the VIM3, was published by the Joint Committee for Guides in Metrology (JCGM) in 2008, and was subsequently published with corrections in 2012. Since its publication, various questions have arisen about some of the changes between the VIM3 and the earlier editions of the VIM. The committee responsible for the development and maintenance of the VIM, Working Group 2 of the JCGM, has undertaken to develop a set of Frequently Asked Questions (FAQs) about the VIM3, which can be used to provide answers to these questions, and rationale for some of the new direction taken in the VIM3. Examples include “Why is the title of the VIM3 different from the titles of the VIM1 and VIM2?”, “What is the difference between a term, a concept and a definition?”, and “Why are some of the definitions in the VIM3 so complicated?” This paper will address some of the more frequently asked questions about the VIM3 and provide readers with information about how to access (online) the more complete set of VIM3 FAQs. The talk is intended to provide attendees with an opportunity to ask their own questions and suggest additional VIM3 FAQs.

Abstract:
Abstract This paper presents results from a case study of vortex-induced motions (VIM) of a floating, spar type offshore wind turbine. VIM response can lead to increased motions of the wind turbine tower as well as contribute to fatigue of moorings and cables. Reliable predictions of VIM response are therefore needed for safe and cost-effective design. One option is to extend the use of analysis methods developed for vortex-induced vibrations (VIV) of slender structures. Three different analysis methods are compared in the present work: • Linear frequency domain analysis with coefficient-based VIV loads. • Nonlinear time domain simulation with a harmonic load found using a VIM design curve. • Nonlinear time domain simulation with the recently developed time domain VIV load model. The nonlinear time domain simulations can include the dynamic wave and wind loads together with the VIM loads. Nonlinear interaction effects can then be included, and fatigue damage can be calculated from the total response instead of combining damages calculated separately. Only the analysis with time domain VIV loads gives in-line VIM response. The results show that if in-line motions occur, they may give significant contributions to the mooring line tensions and fatigue. The effects of the higher Reynolds number and the low length to diameter ratio of floating spars must be investigated. Comparisons with full-scale measurements and adjustment of load coefficients are necessary before a time domain VIM load model is established.

Abstract:
New discoveries of petroleum reservoirs in ultra deep-water depths, like Pre-salt fields in Santos Basin, are demanding new riser systems concepts. In this scenario, the Free-Standing Hybrid Riser (FSHR) system is a viable choice. A submersible buoy connected by rigid and flexible risers constitutes this riser system. The sea current can cause the Vortex-Induced Motion (VIM) of the buoy, which can increase significantly the riser fatigue damage. Although the VIM phenomenon is similar to Vortex-Induced Vibration (VIV), it generally occurs in rigid bodies with low aspect ratio, where end effects causes tridimensional flow behavior. Therefore, the vortex wake characteristics and the hydrodynamics coefficients found for VIV is no longer valid for VIM. In this context, wake oscillator models used for VIV prediction in actual form is not adequate for the VIM prediction of the buoys. In this paper, a VIV wake oscillator model is calibrated for VIM, through hydrodynamic coefficients found in the technical literature. In order to verify accuracy, the VIM calibrated wake oscillator model is used to reproduce some FSHR reduced model tests. The results of amplitude and frequency of oscillation against the reduced velocity obtained from the numerical simulation are compared with the experimental results. The numerical results presented the same trend with some differences in amplitude. The amplitude deviation could be related to the hydrodynamics coefficients used in the calibration of the wake oscillator model.

Abstract:
Vortex-Induced Motion (VIM) is another way to denominate the Vortex-Induced Vibration (VIV) in floating units. The main characteristics of VIM in such structures are the low aspect ratio (L/D < 4.0) and the unity mass ratio (m* = 1.0, i.e. structural mass equal water displacement). The VIM can occur in MPSO (Monocolumn Production, Storage and Offloading System) and spar platforms. These platforms can experience motion amplitudes of around their characteristic diameters. In such cases, the fatigue life of mooring and riser systems can be greatly reduced. Typically, the VIM model testing campaigns are carried out in the Reynolds range between 200,000 and 400,000. VIV model tests with low aspect ratio cylinders (L/D = 1.0, 1.7 and 2.0) and unity mass ratio (m* = 1.0) have been carried out at the Circulating Water Channel facility available at NDF/EPUSP. The Reynolds number range covered in the experiments was between 10,000 and 50,000. The characteristic motions (in the transverse and in-line direction) were obtained using the Hilbert-Huang Transform method (HHT) and then compared with results obtained in experiments found in the literature. The aim of this investigation is to definitely establish the similarity between the VIM and VIV phenomena, making possible to increase the understanding of both and, at same time, allowing some analytical models developed for VIV to be applied to the VIM scenario on spar and monocolumn platforms, logically under some adaption.

Abstract:
Recent work toward predicting spar vortex induced motion (VIM) with computational fluid dynamics (CFD) suggests that such simulations can anticipate many aspects of spar response and thus supplement tow tank experiments and other design methods. However, the results also highlight a number of challenges as well. The spar VIM problem is characterized by very high Reynolds numbers, geometric complexity including the presence of numerous external appendages and the presence of very rough surfaces. In this paper, we first review recent work on spar VIM where CFD was used to simulate tow tank experiments. This work suggests that CFD methods give good results in most cases but also points to some exceptions. In particular, in simulations of small scale vortex induced motion tests of spars, good agreement between analysis and experiments is usually obtained when the flow separates from the spar hull at the strakes. The CFD simulations are sometimes less successful at predicted VIM when flow separation occurs at the spar hull. We then examine our own recent practice in simulating tow tank experiments with CFD with the objective of finding possible modeling deficiencies. The focus is on the resolution of the large eddies in the wake which most influence the fluctuating loads on the spar, but we are also concerned with the use of wall functions to model the boundary layer. All of the calculations use detached eddy simulation (DES). In order to test the method, we make use of wind tunnel experiments at on a fixed truncated cylinder without strakes. The wind tunnel experiments are performed at Reynolds numbers (Re) that are about the same as those used in scale model spar VIM experiments. Wake particle image velocimetry (PIV) and other data from wind tunnel experiments published in the open literature are used for comparison. The comparisons are used to examine requirements for grid resolution in the wake. Finally, it is suggested that specific wind tunnel experiments might be used to gather needed data on the effects of rough walls and appendages at very high Reynolds numbers.

Abstract:
Nowadays, discoveries of petroleum reservoirs are located in ultra deep-water depths. In this scenario, risers systems generally demand submersible buoys to support the riser, in order to reduce weight in floating platform and riser tensioners. Usually, these buoys are installed below 200 meters depth to avoid the wave forces. However, in this condition the sea current cause the Vortex-Induced Motion (VIM) of the buoys, which can increase significantly the riser fatigue damage. It makes complex the prediction of VIM in buoys. In this paper, a semi-empirical model using hydrodynamic coefficients found for low aspect ratio cylinders are presented. In order to verify accuracy of numerical simulations, results are compared with experimental data presented in the literature and a good agreement is found.

Abstract:
Vortex Induced Motion (VIM) of a Spar platform is an important consideration in the design of Spar moorings and risers. During the design phase, the VIM characteristics of a Spar configuration are evaluated by hydrodynamic model testing. The basis for hydrodynamic model testing is that geometric and dynamic similitude between prototype and model parameters is preserved. Reynolds number and Froude number scaling are the two relevant scaling parameters for Spar VIM model testing. However, simultaneously satisfying Reynolds and Froude scaling for the model and prototype conditions is practically impossible. This leads to compromises in model test Reynolds number scaling and inherent uncertainties in extrapolating the model test VIM predictions to full-scale design conditions. Hence measurement of full-scale Spar VIM responses provides valuable data for validation of design assumptions. BP’s integrity management efforts for its Gulf of Mexico deep water production facilities include monitoring of the environmental conditions, and response of the hull, mooring and riser systems. Field measured data for VIM of BP’s Horn Mountain Truss Spar, presented in this paper, shows that the Horn Mountain Truss Spar VIM response is within original design assumptions. The maximum VIM measured in the field (A/D ≈ 0.26) is well below the acceptable design value of A/D = 0.5. The field data is analyzed and results presented in the form of Spar VIM response time traces, Spar VIM response versus Reduced Velocity and variation of period of VIM oscillation with Spar offsets. The field data shows VIM response characteristics consistent with model test observations.

Abstract:
Despite designer’s best efforts, Vortex-Induced Motion (VIM) of spars remains a significant problem. While strakes have proven highly effective at suppressing riser VIM, all three straked classical spars in the Gulf of Mexico continue to experience occasional VIM events. Since VIM can cause riser damage, mooring line fatigue, and even work stoppage, its suppression is a major priority. But designers will only successfully eliminate VIM once reliable predictions tools become available. VIM is both roughness and Reynolds number dependent, and spars lie outside the validated range of current computational tools in both these areas. This paper presents the development of Reynolds-Averaged Navier-Stokes (RANS) methods to predict real-world spar VIM behavior. It includes the ability to address rough surfaces and high super-critical Reynolds numbers. Validation is provided against existing experimental data where available. The resulting algorithms were used to assess effectiveness of two alternative control strategies for suppressing spar VIM. The first strategy consists of actively injecting fluid in a direction tangential to the spar. The second strategy is passive, and relies on adding an external sleeve to the spar. Active injection was found highly effective, and even capable of totally eliminating VIM. The passive alternative was also found to reduce VIM, but is not as effective as active injection.
