Abstract:
A large proportion of vaginal and vulvar squamous cell carcinomas (SCCs) and intraepithelial neoplasias (VAIN and VIN) are associated with HPV infection, mainly type 16. The purpose of this study was to identify HPV genotypes, as well as E6/E7 mRNA expression of high-risk HPVs (16, 18, 31, 33, and 45) in 56 histology samples of VAIN, VIN, vaginal, and vulvar SCCs. HPV was identified in 56% of VAIN and 50% of vaginal SCCs, 71.4% of VIN and 50% of vulvar SCCs. E6/E7 mRNA expression was found in one-third of VAIN and in all vaginal SCCs, 42.9% of VIN and 83.3% of vulvar SCCs. Our data indicated that HPV 16 was the commonest genotype identified in VAIN and VIN and the only genotype found in SCCs of the vagina and vulva. These findings may suggest, in accordance with other studies, that mRNA assay might be useful in triaging lesions with increased risk of progression to cancer.

Cet article analyse le double processus de la circulation du vin comme marchandise et des significations qui l’entourent dans le contexte de la mondialisation. À la lumière de l’étude de Bestor (2004) concernant la circulation transnationale du thon, j’identifie certaines caractéristiques de la commercialisation transnationale du vin, notamment l’existence de deux systèmes de connaissances (les lois et les normes oenophiles) qui influencent la manière pour les consommateurs et les producteurs de penser et de se représenter le vin, systèmes qui participent au caractère fétiche de cette marchandise. Via une analyse du développement de l’industrie vitivinicole à Cirò Marina, en Calabre, je démontre l’influence conjointe des législations et de l’internationalisation du commerce du vin sur l’évolution la production locale. Sont aussi relevés des décalages au niveau des représentations locales du vin. Plutôt que de circuler, la manière avec laquelle les personnes produisent et interprètent les significations du vin varie davantage en fonction de leur position au sein de la production.

Érasme ne s’est jamais présenté comme un anachorète et croit au contraire profondément que la présence du Christ jaillit lorsque des amis se réunissent autour d’un verre de vin. Habité par les textes des Anciens et par les Évangiles, Érasme sait que le vin a été célébré bien avant lui et il ne cesse de se référer aux paraboles bibliques ou aux adages grecs. Convaincu que le vin est source de vie et de bienfaits, il boit chaque jour sa boisson préférée, issue des terres de Bourgogne, qui embrassent alors aussi les vignobles du Jura et de Besançon. Lorsque la vieillesse le frappe, le vin n’apparaît plus seulement comme un délicieux nectar qui permet d’oublier les tourments du siècle, mais aussi comme un puissant remède. <p style="text-align: justify;">L'action des glycosidases exogènes d'origine fongique (α-arabinosidase, α-rhamnosidase, β-apiosidase, β-glucosidase) a été étudiée en vinification en vin doux naturel (VDN) et en vin sec du moût de Muscat de Frontignan. La teneur en fraction libre de l'arôme a montré une forte augmentation dans le cas du vin sec issu du moût enzymé : d'une part la concentration en certains terpénols (nérol, géraniol, citronellol, α-terpinéol, hydroxylinalols) a sensiblement augmenté, d'autre part des composés nor-isoprénoïdiques (β-damascénone, 3- hydroxy-β-damascone), intéressants sur le plan olfactif, ont apparu. On a démontré que l'augmentation de la concentration de la plupart des composés aromatiques était le résultat de l'action hydrolytique des glycosidases exogènes sur les précurseurs glycosidiques. Le vin sec issu du moût enzymé a été jugé plus aromatique (floral) et plus typé. Dans le cas du VDN issu du moût enzymé, l'hydrolyse des précurseurs glycosidiques d'arôme a été interrompue au stade de monoglucosides, dû à l'inhibition de l'activité β-glucosidase par la présence du glucose. Ainsi la fraction libre de l'arôme n'a pu augmenter comme dans le cas du vin sec enzymé.</p>

<p style="text-align: justify;">Le carbamate d'éthyle est une substance naturellement présente dans tous les milieux fermentés contenant de l'éthanol. Son origine reste encore en partie inexpliquée. Dans les vins, les teneurs habituellement rencontrées sont de l'ordre d'une dizaine de microgrammes par litre.</p><p style="text-align: justify;">Dans les eaux-de-vie de vin, les teneurs varient de quelques dizaines à quelques centaines de microgrammes par litre. Le carbamate d'éthyle fait l'objet d'une réglementation dans certains pays à cause de ses propriétés cancérogènes. Dans les eaux-de-vie de vin, le carbamate d'éthyle ne provient ni de celui initialement présent dans le vin distillé, ni de l'urée. Il pourrait se former à partir d'un précurseur volatil non identifié qui réagirait avec l'éthanol après la distillation.</p><p style="text-align: justify;">Il était intéressant de connaître, d'une part les variations de la teneur en carbamate d'éthyle des eaux-de-vie de vin à partir de la fin de la distillation et d'autre part s'il était possible d'éliminer totalement ou même partiellement ce précurseur avant qu'il ne réagisse avec l'éthanol.</p>

The pronoun ‘vin’ (‘who’) is an ancient lexical item. It has neither a complete paradigm of declension nor a category of number; abundance can be expressed by doubling the stem: vin movida? – vin da vin movidnen? (tamari da nino movidnen) (Who came? – Who and who came? (Tamar and Nino came). From the 19th century, the –‘eb’ form of the pronoun ‘vin’ – ‘vinebi’ (‘whos’) emerges in the mountainous dialects of the Georgian language, as “ an attempt to create a plural form’’ of the ‘vin’ pronoun (A. Shanidze). From the following period, ‘vinebi’ (the ‘whos’) spread to other dialects, in general, to the Georgian language, and here and there even in fiction. Today it is especially noticeable in the speech of young people and children. It seems that the media, social network, and other means of communication also contribute to this process. In our opinion, the basis for the emergence and further spread of the ‘vinebi’ can be: 1. The need for formal agreement (apart from the semantic one) of the pronoun ‘vin’ (‘vints’) (who ([the one] who) with the verb and the corresponding word (vin iq’o stumari -vin iq’o stumrebi>vin iq’vnen stumrebi || vin da vin iq’vnen stumrebi || vinebi iq’vnen stumrebi) (Who was the guest – Who was the guests > Who were the guests || Who and who were the guests || Whos were the guests); 2. The analogy of the production of other pronouns formed with the suffix -eb (ra - ra-n-i, ra da ra || raebi. (‘what’) Cf. vin-vin and vin || vinebi (‘who’); also dialect forms: amaebi, magaebi, magnebi, agenebi, eseebi, isebi/isnebi… (these/those); 3. Preparation andin vitroandin vivoevaluation of vinpocetine (VIN) elementary osmotic pump (EOP) formulations were investigated. A method for the preparation of VIN elementary osmotic pump tablet was obtained by adding organic acid additives to increase VIN solubility. VIN was used as the active pharmaceutical ingredient, lactose and mannitol as osmotic agent. Citric acid was used as increasing API solubility and without resulting in the API degradation. It is found that the VIN release rate was increasing with the citric acid amount at a constant range. Cellulose acetate 398-3 was employed as semipermeable membrane containing polyethylene glycol 6000 and diethyl-o-phthalate as pore-forming agent and plasticizer for controlling membrane permeability. In addition, a clear difference between the pharmacokinetic patterns of VIN immediate release and VIN elementary osmotic pump formulations was revealed. The area under the plasma concentration-time curve after oral administration of elementary osmotic pump formulations was equivalent to VIN immediate release formulation. Furthermore, significant differences found for mean residence time, elimination half-life, and elimination rate constant values corroborated prolonged release of VIN from elementary osmotic pump formulations. These results suggest that the VIN osmotic pump controlled release tablets have marked controlled release characters and the VIN osmotic pump controlled release tablets and the normal tablets were bioequivalent.

e17635 Background: Vulvar intraepithelial neoplasia (VIN) is a precursor lesion of vulvar cancer. Human papillomavirus (HPV) is causative for part of VIN lesions. This retrospective study aimed to investigate HPV distribution in patients with vulvar intraepithelial neoplasia (VIN) and the factor associated with the recurrence of high-grade vulvar intraepithelial neoplasia (VIN2+), high-grade vaginal intraepithelial neoplasia (VAIN) , cervical intraepithelial neoplasia(CIN) or progression to vulvar cancer. Methods: We retrospectively analyzed patients who diagnosed as vulvar squamous intraepithelial lesions between 1990 and 2014 in our institution. All the diagnoses and classifications were confirmed by two pathologists in 2015 ISSVD terminology. HPV DNA was extracted from formalin-fixed and paraffin-embedded samples. SPF1/GP6+ polymerase chain reaction (PCR) followed by HPV Blot and E6 type-specific PCR were performed. Results: Ninety-eight VIN patients were included in our analysis. Ninety-three percent were positive for HPV and HPV16 (43.9%), HPV6 (11.38%), HPV11 (8.94%), and HPV58 (4.88%) were the most prevalent types. Patients with positive HPV were diagnosed younger than those with negative results (43.0 years vs. 61.1 years, P = 0.049). In 15 patients who with recurrent VIN (n = 11) or progression to vulvar cancer (n = 4), had positive HPV. All 4 patients who had progression to cancer were positive for HPV16 in their initial VINs. High risk type HPV infection, positive HPV16, and positive alpha-9 types are significant risk factors for recurrent VIN and progression to cancer. Conclusions: Positive HPV was detected in 92.8% of VINs and was related to younger age at diagnosis. HPV16 was the most prevalent type and was significantly related to recurrence/ progression. Other significant risk factors for recurrent VIN or progression to cancer were high-risk HPV and alpha-9 types. Closely monitor of those patients with risk factors was indicated for early detection and treatment of recurrent VIN and vulvar cancer.

Cells lacking vinculin are highly metastatic and motile. The reasons for this finding have remained unclear. Both enhanced survival and motility are critical to metastasis. Here, we show that vinculin null (vin−/−) cells and cells expressing a vinculin Y822F mutant have increased survival due to up-regulated activity of extracellular signal–regulated kinase (ERK). This increase is shown to result from vinculin's modulation of paxillin–FAK interactions. A vinculin fragment (amino acids 811–1066) containing the paxillin binding site restored apoptosis and suppressed ERK activity in vin−/− cells. Both vinY822F and vin−/− cells exhibit increased interaction between paxillin and focal adhesion kinase (FAK) and increased paxillin and FAK phosphorylation. Transfection with paxillin Y31FY118F dominant-negative mutant in these cells inhibits ERK activation and restores apoptosis. The enhanced motility of vin−/− and vinY822F cells is also shown to be due to a similar mechanism. Thus, vinculin regulates survival and motility via ERK by controlling the accessibility of paxillin for FAK interaction.

Abstract BACKGROUND Vincristine is a commonly used chemotherapeutic agent that results in debilitating untreatable peripheral neuropathy. OBJECTIVE To determine the effects of pulsed high-intensity focused ultrasound (HIFU) on sensory thresholds in a validated vincristine-induced neuropathy (VIN) rodent model. METHODS VIN was induced and mechanical allodynia was confirmed by nociceptive testing. von Frey fibers and Randall-Sellito test were used as measures of innocuous and noxious mechanical thresholds, respectively, and the hot plate test for thermal thresholds. Tests were performed before VIN, after 2 wk of vincristine, at 24, 48, 72, and 120 h after HIFU applied to the left L5 dorsal root ganglia at 3 Watts for 3 min. Comparisons were made between a VIN cohort who underwent HIFU, a VIN cohort who underwent sham HIFU, and naïve rodents who underwent HIFU. RESULTS VIN HIFU rats had significantly increased mechanical thresholds at 24 h (P &lt; .001), 48 h (P = .008), 72 h (P = .003), and 120 h (P = .03) after treatment, when compared to pre-HIFU thresholds. Furthermore, at 24 and 48 h following treatment, VIN HIFU rats had significantly higher innocuous and noxious mechanical thresholds and thermal thresholds than VIN sham HIFU rats (P &lt; .001). Thresholds were not altered in naïve rodents who underwent HIFU. Histological data of L5 dorsal root ganglia of VIN HIFU rats suggest that transient cellular edema resolves by 48 h. CONCLUSION Our data suggest that HIFU increases mechanical and thermal thresholds in VIN rodents. Whether HIFU can preclude the development of reduced thresholds in the VIN model warrants further study.

Background: Recently, the immunohistochemical markers cytokeratin 17 (CK17) and SRY-box2 (SOX2) have been evaluated as adjuncts for the diagnosis of high-grade vulvar intraepithelial neoplasia (VIN). In the present study, the aim was to assess CK17 and SOX2 expression in VIN by studying 150 vulvar lesions, originally reported as high-grade VIN and to assess the diagnostic accuracy. Methods: All slides (H&E, p16INK4a, p53, Ki-67, CK17, and SOX2 stains) were independently assessed by six pathologists and the final diagnosis was reached in consensus meetings, as follows: 46 human papillomavirus (HPV)-independent VIN (including 30 p53 mutant and 16 p53 wild-type lesions), 58 high-grade squamous intraepithelial lesions (HSILs), 4 low-grade SILs (LSILs), 37 non-dysplastic lesions, and 5 lesions where the histology was inconclusive. Results: CK17 positivity was observed in 100% p53 wild-type HPV-independent VIN, compared to 73% p53 mutant HPV-independent VIN, 14% HSILs, 0% LSILs, and 24% non-dysplastic lesions. SOX2 positivity was observed in 13% p53 wild-type HPV-independent VIN, 43% p53 mutant HPV-independent VIN, 2% HSILs, 0% LSILs, and 3% non-dysplastic lesions. The highest diagnostic accuracy (89%) for HPV-independent VIN was obtained when combining p53 and CK17 immunohistochemistry. The addition of SOX2 did not further increase diagnostic accuracy. Conclusion: To conclude, aside from p53, both CK17 and SOX2 can be of value for reaching an accurate diagnosis of HPV-independent VIN.

AC Vin-Pro is a large-seeded soybean [Glycine max (L.) Merr.] cultivar with elevated seed protein levels. AC Vin-Pro has yellow hila and acceptable tofu processing quality for foreign and domestic markets. Its brighter seed colour should make it attractive to the Japanese miso market. Abstract:
Purpose: Doxorubicin (DOX) is a widely used chemotherapeutic agent complicated with cardiotoxic adverse effects. Up till now, there are no researches discussing the role of vinpocetine (VIN) in DOX cardiotoxicity. Thus, the aim of our work was to study this effect and explore the different involved mechanisms. Methods: 50 male Wistar albino rats were subjected to DOX toxicity via administration of single i.p. Dose (15 mg/kg) on the 4th day with or without co-administration of VIN (10, 20, 30 mg/kg/day) orally for 5 days. Results: Our data revealed that VIN succeeded in protecting the heart against DOX induced damage as manifested by significant decrease of cardiac enzymes, hypoxia inducible factor alpha (HIF-1α), vascular endothelial growth factor-A (VEGF-A), tissue malondialdehyde (MDA), tumor necrosis factor alpha (TNF-α) and caspase3 levels. Furthermore, VIN given group showed marked improvement of the histopathological changes of cardiac injury, total antioxidant capacity (TAC), elevation of reduced glutathione (GSH), cyclic guanosine monophosphate (cGMP), cyclic adenosine monophosphate (cAMP) and sirtuin-1 (SIRT-1). Conclusion: We concluded that VIN could ameliorate DOX induced cardiac damage and this effect may be attributed to modulation of HIF/VEGF signaling pathway, up-regulation of cGMP/cAMP/SIRT pathway, inhibition of phosphodiesterase enzyme, besides its anti-apoptotic, anti-inflammatory, and anti-oxidant properties.

This article examines Vin Diesel’s use of his public Facebook Page to mourn the loss of his friend and co-actor Paul Walker in the period from 2013-2015. It discusses how Vin Diesel performed his grief and how his mourning process was communally reflected and repeated by both Vin Diesel and Walker fans, who used Vin Diesel’s page to share and verbalise their own feelings of loss in a both public and safe space. An analysis of Vin Diesel’s own status updates and 1800 comments reacting to three popular status updates related to the death of Paul Walk posted over the course of more than a year show that commentary was used to make condolences to both Vin Diesel and Walker’s familes and to affectively express the users’ immediate feelings, both verbally and through the use of emojis. However, over time, both the form and intensity of expression of both Vin Diesel and his followers changed, pointing to the need to further study celebrity mourning processes on social media over extended periods of time.

Вульварная интраэпителиальная неоплазия (vulvar intraepithelial neoplasia, VIN) – неинвазивное предраковое поражение вульвы, которое встречается у 50–70% пациенток, страдающих плоскоклеточным раком вульвы. До недавнего времени радикальные хирургические методы были стандартом лечения VIN, однако, учитывая психофизиологические осложнения, связанные с удалением обширных зон поражения и невозможностью вести нормальный образ жизни, с конца ХХ века осуществлялся поиск менее агрессивных методов лечения. Фотодинамическая терапия (ФДТ) – эффективный и безопасный вид лечения предрака вульвы с благоприятными результатами, в том числе косметическими у женщин репродуктивного возраста. Методы. В статье представлены данные о классификации, этиологии и патогенезе, методах диагностики и видах лечения VIN, результаты исследований, посвященных применению ФДТ при лечении VIN, и собственные клинические наблюдения. Результаты. Описаны 2 клинических случая пациенток с VIN различной степени тяжести, которым до ФДТ были проведены разнообразные виды терапии. После применения ФДТ наступило значительное улучшение. Фотодинамическое лечение не вызывает выраженного дискомфорта при применении, хорошо переносится и является практичным. Полученные результаты свидетельствуют о безопасности и удобстве применения данного метода лечения. Заключение. Преимущество ФДТ при VIN заключается в ее способности воздействовать на многоочаговое заболевание с минимальным разрушением тканей, сохранением анатомии вульвы и отличными результатами купирования симптомов. Благодаря этим свойствам ФДТ является одним из перспективных вариантов лечения VIN. Vulvar intraepithelial neoplasia (VIN) is a non-invasive precancerous lesion of the vulva, which occurs in 50–70% of patients suffering from squamous cell carcinoma of the vulva. Until recently, radical surgical methods were the standard method of treating VIN, however, given the psycho-physical complications associated with the removal of extensive lesion areas and the inability to lead a normal lifestyle, since the end of the 20th century, less aggressive treatment methods have been sought. Photodynamic therapy (PDT) is an effective and safe type of treatment for vulvar cancer with favorable cosmetic results. Methods. The article presents data on the classification, etiology and pathogenesis, diagnostic methods and types of treatment of VIN, the results of studies on the use of PDT in the treatment of VIN, and our own clinical observations. Results. Two clinical cases of patients with VIN of varying severity who underwent various types of therapy before PDT were described. After the application of PDT, there was a significant improvement. Photodynamic treatment does not cause pronounced discomfort when used, is well tolerated and is practical. The results obtained indicate the safety and convenience of using this treatment method. Conclusions. The advantage of PDT for VIN lies in its ability to affect a multi-focal disease with minimal tissue destruction, preservation of the anatomy of the vulva and excellent results in relieving symptoms. Due to these properties, PDT is one of the promising treatment options for VIN.

Vulvar intraepithelial neoplasia (VIN) is the proliferation of atypical basal cells in the vulvar epithelium. The global VIN incidence has recently doubled; its incidence among white women under 35 years of age has almost tripled with a tendency for further growth. Such an increase in the number of usual-type VIN cases in young women is primarily attributed to infection with highly oncogenic human papillomavirus. The second type of dysplasia, namely differentiated VIN, is usually found in older women and is associated with chronic dystrophic diseases of the vulva, most frequently with lichen sclerosus of the vulva. VIN diagnosis is quite challenging; no screening programs for this disorder have been developed so far. Patients with VIN practice self-treatment for a long time, which aggravates their condition and might trigger the development of vulvar cancer. Several treatment options are currently available; however, their efficacy worldwide is not high.

OBJECTIVE: Our goal was to clarify the clinical significance of vibration-induced nystagmus (VIN). METHODS: One hundred patients with unilateral vestibulocochlear disorders were enrolled into this study. However, patients with spontaneous nystagmus were excluded. Vibratory stimuli (approximately 100 Hz) were presented to the mastoids and the forehead. Patients also underwent caloric testing and vestibular evoked myogenic potential testing. RESULTS: Of the 100 patients, 60 (60%) showed VIN. The nystagmus was mainly horizontal. VIN was more frequently evoked on the mastoids than the forehead. In the majority of patients, the direction of VIN was toward the healthy side, whereas some patients, especially patients with Meniere's disease, showed nystagmus toward the affected side. VIN was frequently evoked in patients with severe unilateral vestibular damages (canal paresis >50%) (39 of 43, or 90%). CONCLUSION: VIN testing is a simple and sensitive clinical test that indicates unilateral vestibular dysfunction.

In this study, we present the findings of a regional retrospective audit of all cases of vulval intraepithelial neoplasia (VIN) diagnosed in Northern Ireland (NI) over an 11-year period (1989–1999). During the period of the study, there were 97 cases of VIN. Cases of VIN were treated by many different clinicians at 14 hospitals. The commonest symptom at presentation was pruritus vulvae. The most common histopathologic diagnosis was VIN III (73%). In 52% of the cases, there was multifocal VIN, and in 43%, there was involvement of other sites such as the cervix, vagina, or anal region. The most common initial treatment was surgical excision, but a multitude of different treatments were performed initially. During the study period, 18 of 90 patients (20%) for whom follow-up was available developed invasive vulval squamous carcinoma. Most of the vulval cancers were superficially invasive, but three patients died of vulval cancer during the study period. This study illustrates that in NI, VIN is treated at many institutions by a multitude of clinicians. Management seems largely dependent on personal clinician preference and has been haphazard with little central coordination and organized strategy. VIN should be managed by clinicians with expertise in this field and who are treating significant numbers of patients according to evidence-based protocols.

L’objet de cet article est d’analyser la construction d’un nouveau discours médical sur le vin dont l’émergence et la cohérence paraissent attribuables à la récupération par les médias et la filière vitivinicole d’un certain nombre de données sur les bienfaits du vin sur la santé. Ce faisant, ces derniers contribuent largement à la mise en forme et à la diffusion d’une nouvelle représentation du vin, si bien que progressivement les populations tendent à percevoir celui-ci à travers ce nouveau discours ou cette nouvelle symbolique. Néanmoins, le point de vue des institutions régissant la santé publique demeure inchangé et aucun État n’a modifié le statut légal du vin. Cette transformation de sens nous renseigne sur la production, l’utilisation et la diffusion d’images et d’informations dans notre société et sur les liens étroits que ces phénomènes peuvent entretenir avec la mondialisation des marchés.

Vulvar intraepithelial neoplasia (VIN) is a dysplastic condition of the squamous epithelium of the vulva. There are two types of VIN: high-grade squamous intraepithelial lesion of the vulva and VIN differentiated type (dDVIN). Management includes excision, laser ablation, and topical therapy. An excisional procedure used in VIN is simple local excision and partial or total skinning vulvectomy. Despite treatment, its recurrence is high. A G5P5 (5004) woman in her 60s presented with vulvar pruritus and vulvar pain of 2 years. She was treated for cervical adenocarcinoma Stage IB1 with surgery and complete radiotherapy 27 years prior. She was diagnosed twice with vulvar dysplasia 12 and 21 years after the diagnosis of cervical malignancy, both times presenting as vulvar pruritus. She was subsequently managed with vulvectomy with bilateral groin node dissection and with wide local excision, respectively. A 3 cm × 2 cm well-circumscribed, irregular erythematous plaque at the introitus’s 11–1 o’clock region was noted on physical examination. She was managed as a case of recurrent VIN III and underwent wide local excision and distal urethrectomy with split-thickness skin graft. The final histological examination of the submitted specimen showed human papillomavirus-associated classic VIN II.
