Fluorogenic substrates [Ala-Pro]2-cresyl violet and Ala-Pro-rhodamine 110 have been tested for microscopic detection of protease activity of dipeptidyl peptidase IV (DPPIV) in living cells. DPPIV activity is one of the many functions of the multifunctional or moonlighting protein CD26/DPPIV. As a model we used Jurkat cells, which are T-cells that lack CD26/DPPIV expression, and CD26/DPPIV-transfected Jurkat cells. Ala-Pro-rhodamine 110 is not fluorescent, but after proteolytic cleavage rhodamine 110 fluoresces. [Ala-Pro]2-cresyl violet is fluorescent by itself but proteolytic cleavage into cre

A detailed map of the vestibular nuclear complex of the guinea pig has been established by Gstoettner and Burian (1987), using cytoarchitectonic (cresyl violet staining) and fiberarchitectonic criteria. However, the exact borders between the different subdivisions are not always evident in Nissl stained sections. In the present study, serial sections of the vestibular nuclei of the guinea pig were stained to visualize acetylcholinesterase (AChE) activity, and compared with corresponding sections stained with cresyl violet. Abstract:
Mild traumatic brain injury (mTBI), often referred to as concussion, has become increasingly recognized as a serious health issue in the general population. The prevalence of mTBI in athletes, particularly repeated injuries in young athletes, is of great concern as injuries to the developing brain can have long-term detrimental effects. In this study we used a novel awake closed-head injury (ACHI) model in rodents to examine repeated mTBI (rmTBI), to determine if repeated injuries produced the neurological and molecular changes evident with human concussion. This entire nerve is typically sampled over the course of a few centimeters; however, in some cases fascicular biopsy is performed. The sural nerve biopsy specimen is studied using hematoxylin and eosin, trichrome (for connective tissue), and silver stains (for axons), as well as with specialized stains as needed (e.g., Congo red for amyloid or Hirsch-Peiffer acidified cresyl violet stain for metachromatic materials). We examine a spectral region centered at the pump wavelength, 618 nm, and ranging from 570 to 670 nm. Differential absorption spectra are taken at intervals of 25 fs following the initial excitation of the dye. A principal problem in obtaining optical pulses with bandwidths approximating that of the visible spectrum and durations approaching the transform limit has been the uncompensated cubic phase distortion of pulses compressed by grating pairs. We obtain pulses of 6-fs duration which can be used on a routine basis for time-resolved spectroscopy. We have used these pulses to study the dynamics of spectral h
