Abstract:

This study aims to analyze the home visite services of people with mental disorders in Kalawa Atei General Hospital in Central Kalimantan Province. Given the implementation of the program for people with mental disorders to access health services whose handling is different from other health services. This study uses a qualitative approach that aims to describe the complexity and dynamics of handling people with mental disorders. The primary data in this study were sourced from several informants, namely Home Visite Officers from Kalawa Atei General Hospital, Social Service and Povinsi / Regency Health Service, local Puskesmas, and Family of Ganguan Mental Panderita. Meanwhile, Secondary Sources in this study include the Implementation of Home Visite Regulation, Home Visite Service Profile, Home Visite Patient Ratio and other related documents. The results of the study show the conclusions of the study that the implementation of home visite services organized by the Central Kalimantan Atei Central Hospital along with other cross sectors has been running quite well. This is seen in terms of the facilities and infrastructure that have been provided in providing home visite services for ODGJs that have been prepared and are maximally sought. Home visite services are in accordance with SOPs, competencies and disciplines of each service officer. Home visite services provide guarantees to each patient with a special schedule by officers to come to the regions to provide these services. In this case, home visite services in terms of empathy by service personnel are good enough. Considering that home viste activities are shown to severely mentally ill patients, who cannot access Kalawa Atei General Hospital for various reasons, such as financial incapacity, no family support, family ignorance, and the existence of savings. However, there are several inhibiting factors in the implementation of the Home Visite Service at Atawa Kalawa Hospital, namely: 1) equipment completeness, 2) field conditions, 3) communication.

Hvilke opplevelser og læring hadde foreldrene av å diskutere barneoppdragelse i en fokusgruppe? Fokusgruppeintervjuer ble gjennomført med fire foreldre, fra Norge og to ikke-vestlige land. Det ble gjennomført ett oppfølgingsintervju med faren fra ett av de ikke-vestlige landene. Samtlige deltakere hadde barn i åpen barnehage. Dataene fra fokusgruppeintervjuene viste at foreldrene fortalte hverandre hverdagslige historier om <span style="color: black; mso-themecolor: text1;">blant annet</span> søvnrutiner, ros og ris i oppdragelsen. Dataene viste også at foreldrene var fornøyde med møtene. De opplevde at samtalene var lærerrike, nyttige og fortrolige. Dessuten uttrykte de at det var beroligende å oppleve at andre foreldre hadde samme utfordringer. Faren som deltok i oppfølgingsintervjuet, fortalte at fokusgruppa fungerte som hjelp i oppdragelsen av eget barn. Diskusjonen tar opp betydningen av at foreldre med ulik kulturell bakgrunn snakker sammen om hverdagsdilemmaer vedrørende oppdragelsen av egne barn. Videre viser diskusjonen at det er av betydning<span style="mso-spacerun: yes;">  </span>å arrangere formelle møter mellom etnisk norske- og minoritetsforeldre fordi det er med på å fremme akkulturasjonsprosessen. </span></span></span></em></p>

Studentenes ukentlige refleksjonsmøter i løpet av deres medisinske praksisperiode er blitt til i løpet av et pedagogisk aksjonsforskningsprosjekt (2006 – 2009), kalt ”Når praksisfelt og skole flyter sammen”. Studentevalueringer og loggdata viste at refleksjonsgruppene var viktige for studentenes læring, men sa lite om på hvilken måte. Dette ble utforsket gjennom et fokusgruppeintervju med 8 studenter. Det utkrystalliserte seg fem </em><a name="OLE_LINK2"></a><a name="OLE_LINK1"><span style="mso-bookmark: OLE_LINK2;"><em>tema: Å utvikle kunnskap, Erfaringslæring, Anerkjennelse av egen sårbarhet, Å oppøve faglig skjønn og </em></span></a><em>Å være i bevegelse mot profesjonell yrkesutøvelse. I en sammenfattende tyding fremkom Dannelse som et overordnet tema. Refleksjonsrommet drøftes som en dannelsesarena, en ”refleksjonsoase”, hvor studentene erfarer at de får et personlig og reflektert forhold til kunnskapene de erverver seg. Kunnskaper og dannelse vil da sammen, og i vekselvirkning med hverandre, hjelpe dem mot en bedre yrkesutøvelse. </em></span>

Abstract:

The PDZ domain (PSD95-Discs large-ZO1) is a widespread modular domain present in the living organisms. A prevalent function in the PDZ family is to serve as scaffolding and adaptor proteins connecting multiple partners in signaling pathways. An explanation of the flexible functionality in this domain family, based just on a static perspective of the structure–activity relationship, might fall short. More dynamic and conformational aspects in the protein fold can be the reasons for such functionality. Folding studies indeed showed an ample and malleable folding landscape for PDZ domains where multiple intermediate states were experimentally detected. Allosteric phenomena that resemble energetic coupling between residues have also been found in PDZ domains. Additionally, several PDZ domains are modulated by post-translational modifications, which introduce conformational switches that affect binding. Altogether, the ability to connect diverse partners might arise from the intrinsic plasticity of the PDZ fold.

Abstract:

From its creation as a military housing development to its ultimate transformation into private housing, Linda Vista, in San Diego, ran the ideological spectrum—ranging from a foil for alleged communism, to a repository for proto Right Wing conservatism—simultaneously revealing burgeoning sunbelt politics and the conflict between the housing needs of military families and the anti-public housing ethos of the city's political class. Though the Navy required such projects to house its service personnel and their dependents, the city and many residents sought to eliminate public housing. Linda Vista also demonstrates the intersection of military housing, race, and local politics. For the left, it served as a fortress of political support in the 1940s, but by the 1950s, Linda Vista came to be a Republican stronghold. Ultimately, Linda Vista's shift previewed the New Right conservatism that Sunbelt metropolises would promote in the latter half of the twentieth century.

Abstract:

The immunoglobulin (Ig) superfamily consists of many critical immune regulators, including the B7 family ligands and receptors. In this study, we identify a novel and structurally distinct Ig superfamily inhibitory ligand, whose extracellular domain bears homology to the B7 family ligand PD-L1. This molecule is designated V-domain Ig suppressor of T cell activation (VISTA). VISTA is primarily expressed on hematopoietic cells, and VISTA expression is highly regulated on myeloid antigen-presenting cells (APCs) and T cells. A soluble VISTA-Ig fusion protein or VISTA expression on APCs inhibits T cell proliferation and cytokine production in vitro. A VISTA-specific monoclonal antibody interferes with VISTA-induced suppression of T cell responses by VISTA-expressing APCs in vitro. Furthermore, anti-VISTA treatment exacerbates the development of the T cell–mediated autoimmune disease experimental autoimmune encephalomyelitis in mice. Finally, VISTA overexpression on tumor cells interferes with protective antitumor immunity in vivo in mice. These findings show that VISTA, a novel immunoregulatory molecule, has functional activities that are nonredundant with other Ig superfamily members and may play a role in the development of autoimmunity and immune surveillance in cancer.

El estudio expone una comparativa sobre el tratamiento procesal que puede recibir un asunto de Derecho de Familia, desde el punto de vista del profesional de la abogacía, bien como profesional humano, bien como inteligencia artificial. El supuesto concreto ha sido extraído de la praxis forense real de un despacho de abogados. El planteamiento se enfoca a la resolución de la conducción del proceso civil desde sus inicios hasta su finalización en la vía judicial. El supuesto de hecho aborda la problemática de la decisión de las medidas a acordar en unas relaciones paterno-filiales especialmente conflictivas y con varios factores problemáticos. El análisis trata de confrontar las respuestas dadas por el jurista, desde un marco subjetivo inherente a su cualidad de persona, frente al de una inteligencia artificial, desde su contexto puramente objetivo. El Código Civil y la Ley de Enjuiciamiento Civil constituyen la regulación legal que sirve de base para las distintas interpretaciones.

Verificou- se a presença de duas cavidades: a)cavidade temporária e b) cavidade permanente. O índice de incapacitação foi aferido baseado na forma e profundidade da cavidade temporária que cada calibre produzia na argila. A cavidade temporária foi medida em volume e expressa em ml.</p></span></span>

Abstract:

Programmed death-1 homolog (PD-1H), a CD28/B7 family molecule, coinhibits T cell activation and is an attractive immunotherapeutic target for cancer and inflammatory diseases. The molecular basis of its function, however, is unknown. Bioinformatic analyses indicated that PD-1H has a very long Ig variable region (IgV)-like domain and extraordinarily high histidine content, suggesting that unique structural features may contribute to coinhibitory mechanisms. Here we present the 1.9-Å crystal structure of the human PD-1H extracellular domain. It reveals an elongated CC′ loop and a striking concentration of histidine residues, located in the complementarity-determining region-like proximal half of the molecule. We show that surface-exposed histidine clusters are essential for robust inhibition of T cell activation. PD-1H exhibits a noncanonical IgV-like topology including an extra “H” β-strand and “clamping” disulfide, absent in known IgV-like structures, that likely restricts its orientation on the cell surface differently from other IgV-like domains. These results provide important insight into a molecular basis of T cell coinhibition by PD-1H.

BackgroundCraniopharyngioma (CP) is a common refractory tumor of the central nervous system. However, little is known about the expression and clinical significance of B7 family ligands/receptors in CP patients. Thus, we conducted the present study to address this issue in a cohort of 132 CP cases.MethodsWe mapped and quantified the expression of B7 family ligands/receptors molecules programmed cell death ligand 1 (PD-L1), B7-H3, B7-H4 and V-domain Ig-containing suppressor of T cell activation (VISTA) in 89 adamantinomatous-type CP and 43 papillary-type CP samples using immunohistochemistry and immunofluorescence. Associations between the marker levels, clinicopathological variables and survival were evaluated.ResultsThe positive rates of PD-L1, B7-H3, B7-H4 and VISTA in the cohort of 132 CP cases were 76.5%, 100%, 40.2% and 80.3%, respectively. The cut-off values of PD-L1, B7-H3, B7-H4 and PD-L1 expression were determined by survival receiver operating characteristic (ROC) package, which was 70, 182, 0 and 20, respectively. Elevated expressions of PD-L1, B7-H3, B7-H4 and VISTA were significantly associated with some clinicopathological characteristics. Kaplan-Meier analysis indicated that higher VISTA expressions correlated with better overall survival (OS) and progression-free survival (PFS) (p=0.0053 and p=0.0066, respectively). Multivariate Cox regression analysis indicated that VISTA was an independent prognostic factor for OS (p=0.018) but not for PFS (p=0.898).ConclusionsWe found variable expression of PD-L1, B7-H3, B7-H4 and VISTA proteins in CPs. The results suggest that the expression level of VISTA may be used as an important indicator to predict the OS and PFS of CPs. B7 family ligands/receptors could be potential immunotherapeutic targets when treating CPs.

Background:V-domain Ig suppressor of T cell activation (VISTA) is an immune checkpoint receptor with an established role as an inhibitory receptor on T cells. However, its role in myeloid cell biology remains unclear. Owing to the much higher expression of VISTA on monocytes and macrophages, we investigated its role in regulating myeloid biology. Methods: Mouse bone marrow-derived macrophages (BMDMs) were differentiated and treated with agonistic anti-VISTA mAb under steady-state and conditions of M1 inflammatory polarization (LPS + IFN-𝛾). Human CD14+ monocytes were isolated from peripheral blood and treated with anti-VISTA in vitro. Phosphoarrays were used to assess signaling downstream of VISTA engagement and Luminex analysis was used to evaluate cytokine production. Time-course total RNA-seq was performed on BMDMs (at 1, 2, 4, 8 and 16 hours of anti-VISTA treatment). To evaluate the effect of anti-VISTA in vivo, single-cell RNA-seq (scRNA-seq) was performed on FACS sorted CD11b+ myeloid populations 12 hours after mice were treated with intravenous anti-VISTA. Results: Anti-VISTA treatment of BMDMs and human monocytes in vitro induced rapid phosphorylation events including robust phosphorylation of Raf-1 kinase at the suppressive serine residue 43.Next, we generated a comprehensive timestamp of VISTA activation in BMDMs in vitro by time-course deep RNA-seq analysis of agonistic anti-VISTA treated cells under rigorous M1 inflammatory polarization. This revealed a unique activation state marked by the upregulation of inhibitory cytokines IL-10 and IL1RA as well as ablation of IL-12 family cytokines, suggestive of an M2-like signature. The presence of an M2-like immunoregulatory signature defined by upregulation of the IL-10 was also seen in macrophages identified post clustering of in vivo scRNA-seq data on myeloid cells. Conclusion: Using novel agonistic anti-VISTA antibodies, we reveal that VISTA triggering on myeloid cells can induce an alternative anti-inflammatory cell-state, even under conditions of strong inflammatory polarization. Desde o ano de 1950 este festejo é realizado pelo grupo de Congado local, como um modo de prestar homenagens à sua padroeira. As festas executados pelo Congado apresentam grande notoriedade no cenário das manifestações religiosas de cunho popular do estado de Minas Gerais, preservando as tradições que remetem a um passado escravocrata do Brasil Colonial. O objetivo desse trabalho é o de analisar a identidade congadeira a partir dos símbolos culturais que compõem a rememoração da coração dos Rei e Rainha congos a partir dos conceitos de hibridismo, tradução cultural e terceiro espaço. A metodologia adotada foi a observação participante, e a análise dos dados foi realizada a partir dos pressupostos teóricos dos Estudos Culturais. Este estudo revelou que a construção da identidade cultural congadeira foi capaz de superar as posições binárias e hierarquizadas das religiosidades católica e de matriz africana, criando uma terceira narrativa que contribuiu de forma positiva para o reconhecimento dessas identidades diaspóricas.</span></span></p>

This article analizes the labors of memory in Vanessa Núñez Handal’s novel Dios tenía miedo. By looking into both the familial and the national past in El Salvador through an autofictional exploration of the ways in which her own family was involved in the war she herself experienced as a child, Vanessa Núñez Handal experiments with the explanatory potential of both the creative imagination and the use of symbolic language, in a process where writing seems to be as much a method as a goal in order to remember and, above all, understand the percepticide that took place during the war. The ambiguity of the autofictional pact rules over the novel, giving her the liberty to combine fictional, autobiographical, and testimonial techniques, with the aim of articulating her interpretation of the traumatic past.

Background. B7 family members and ligands have been identified as critical checkpoints in orchestrating the immune response during sepsis. V-domain Ig suppressor of T cell activation (VISTA) is a new inhibitory immune checkpoint involved in restraining T cell response. Previous studies demonstrated that VISTA engagement on T cells and myeloid cells could transmit inhibitory signals, resulting in reduced activation and function. The current study was designed to determine the potential therapeutic effects of a high-affinity anti-VISTA antibody (clone MH5A) in a murine model of sepsis. Methods. Polymicrobial sepsis was induced in male C57BL/6 mice via cecal ligation and puncture. Expression profiles of VISTA on T lymphocytes and macrophage were examined at 24 and 72 h postsurgery. The effects of anti-VISTA mAb on the 7-day survival, lymphocyte apoptosis, cytokine expression, bacterial burden, and vital organ damage were determined. Furthermore, the effects of anti-VISTA mAb on CD3+ T cell apoptosis and macrophage activation were determined in vitro. Results. VISTA was substantially expressed on T cells and macrophages in sham-operated mice; septic peritonitis did not induce significant changes in the expression profiles. Treatment with MH5A improved the survival of septic mice, accompanied by reduced lymphocyte apoptosis, decreased cytokine expression, and enhanced bacterial clearance. Engagement of VISTA receptor with MH5A mitigated CD3+ T cell apoptosis cultured from CLP mice and suppressed LPS-induced cytokine production by macrophage in vitro. Conclusion. The present study identified VISTA as a novel immune checkpoint in the regulation of T cell and macrophage response during sepsis. Modulation of the VISTA pathway might offer a promising opportunity in the immunotherapy for sepsis.

Members of B7-CD28 family ligands and receptors play the important role in the co-stimulation and inhibition of the T-cell mediated immune response. Blocking of the B7 family molecules inhibiting the immune response (also called immunological checkpoints), CTLA4 and PD-1 / PD-L1 promoted to important therapeutic achievements in cancer immunotherapy and improved survival of cancer patient. The recent discovery of new B7 family molecules (such as B7-H3 [CD276], B7-H4 [B7S1 / B7x / Vtcn1], B7-H5 [VISTA / PD-1H / GI24], B7-H6 and B7-H7 [HHLA2]) has expanded therapeutic possibilities for the management of cancer patients. In this article, we discuss the clinical application of known blockers of immunological checkpoints and the prospects for the use of newly discovered molecules.

Es importante señalar que al tratarse de un acercamiento novedoso al análisis de investigación histórica, las propuestas que se desarrollan aquí están en proceso de trabajo y pretenden ampliar las discusiones respecto al uso de la imagen en la investigación. A partir de un breve análisis de las fotografías, en tanto forman parte de las disputas por la definición de significados, se realizará una descripción de la diversidad de trabajadores participantes en huelgas y manifestaciones. Se observan ámbitos de lucha, la importancia de la exposición pública de los reclamos, el rol de las mujeres y niños y la existencia de códigos gestuales particulares. Finalmente se propone una periodización que divide un momento de afinidad y otro de aversión de la revista “Caras y Caretas” hacia los sectores obreros.</span>

O Programa de Aquisição de Alimentos (PAA) tem se convertido em uma vitrine em termos de políticas públicas dirigidas a agricultura familiar no Brasil. Dois de seus principais objetivos são, de um lado, o fortalecimento econômico e social dos produtores familiares através da compra da produção por parte dos organismos públicos e, por outro lado, a luta contra a fome e a desnutrição mediante a distribuição dos alimentos para a população em situação de insuficiência alimentar. Neste artigo, analisa-se a execução do PAA no município de Boa Vista (estado de Roraima) e, mediante a utilização do enfoque de capital social, se descreve a rede social gerada em torno do Programa e o tipo de relações que se produz entre os atores que formam parte dela. O estudo está centrado em analisar os efeitos do PAA no âmbito dos produtores que participam do Programa, e os resultados mostram uma rede social débil, que se sustenta no protagonismo de alguns atores, a exemplo da CONAB, e também onde são nítidas a carência de um estoque de capital social suficientemente equilibrado em suas distintas dimensões. Esse quadro conjuntural levanta inquietações sobre a eficiência de uma estratégia de desenvovimento como o PAA em territórios de baixa institucionalidade e escasso nível de coesão social, que por sua vez nos traz incertezas sobre os atuais efeitos positivos entre os beneficiários possam ser sustentáveis no tempo.

BackgroundV-domain Immunoglobulin Suppressor of T cell Activation (VISTA/PD-1H) is a B7 family ligand expressed on circulating and intratumoural myeloid cells as well as Treg and NK cells. It has been shown to inhibit T cell responses in vitro and in preclinical models. In patients, VISTA is also a potential mediator of resistance to anti-CTLA-4 and anti-PD1 therapies and therefore is a valuable new target for cancer immunotherapy.MethodsKineta has analyzed 107 fully human ScFv antibodies directed against VISTA.ResultsOur lead candidates exhibit high potencies in the subnanomolar range and are also characterized by a long kDis. They specifically target human and cynomolgus monkey VISTA on a singular unique epitope. In a Staphylococcus Enterotoxin B T-cell activation assay, Kineta’s anti-VISTA antibodies efficiently induce IFNg secretion. They also promote strong maturation of Antigen Presenting Cells with an increase of CD80 and HLA-DR surface expression as well as CXCL10 secretion. The mechanism of action is mediated in part by NK cells. We demonstrated that myeloid cells acquire a high level of VISTA expression during MDSC or M2 differentiation in vitro and that Kineta’s anti-VISTA antibodies prevent the differentiation of MDSC as well as their immunosuppressive activity against T cells. Anti-VISTA antibodies mediate single-agent antitumor effects in syngeneic tumor models in wild-type mice and show enhanced activity in combination with anti-PD1 and anti-CTLA-4 treatment. Candidate anti-VISTA antibodies have also been evaluated in exploratory tolerability and PK studies in cynomolgus monkey. Abstract:

BackgroundV-domain immunoglobulin suppressor of T cell activation (VISTA) is a B7 family inhibitory immune checkpoint protein and is highly expressed on myeloid cells and T cells.1 VISTA acts as both an inhibitory ligand when expressed on antigen-presenting cells and a receptor when expressed on T cells. Our recent study has shown that VISTA is a myeloid cell-specific immune checkpoint and that blocking VISTA can reprogram suppressive myeloid cells and promote a T cell-stimulatory tumor microenvironment.2 In this study, we further demonstrate that VISTA blockade directly alters the differentiation and the suppressive function of myeloid-derived suppressor cells (MDSC).MethodsFlow cytometry was performed to examine VISTA expression on MDSCs in multiple murine tumor models including the B16BL6 melanoma model, MC38 colon cancer model, and the KPC pancreatic cancer models. To examine the role of VISTA in controlling the differentiation and suppressive function of MDSCs, we cultured wild type (WT) and VISTA.KO bone marrow progenitor cells with GM-CSF and IL-6 to induce BM -derived MDSCs.ResultsOur preliminary results show that VISTA is highly expressed on M-MDSCs in B16BL6, MC38 and KPC tumors. In BM-derived MDSCs, VISTA deletion significantly altered the signaling pathways and the differentiation of MDSCs. Multiple inflammatory signaling pathways were downregulated in VISTA KO MDSCs, resulting in decreased production of cytokines such as IL1 and chemokines such as CCL2/4/9, as well as significantly impaired their ability to suppress the activation of CD8+ T cells. The loss of suppressive function in VISTA KO MDSCs is correlated with significantly reduced expression of iNOS. To validate the results from BM-MDSCs, we sorted CD11b+CD11c-Ly6C+Ly6G- M-MDSCs and CD11b+CD11c-Ly6G+ G-MDSCs from B16BL6 tumor tissues and tested the ability of a VISTA-blocking mAb to reverse the suppressive effects of tumor-derived MDSCs. Our results show that blocking VISTA impaired the suppressive function of tumor-derived M-MDSC but not G-MDSCs.ConclusionsTaken together, these results demonstrate a crucial role of VISTA in regulating the differentiation and function of MDSCs, and that blocking VISTA abolishes MDSC-mediated T cell suppression, thereby boosting.Ethics ApprovalAll in vivo studies were reviewed and approved by Institutional Animal Care and Use Committee (Approval number 2019-2142).ReferencesXu W, Hire T, Malarkannan, S. et al.
