In order to cause disease, CD8+ T cells must overwhelm the mechanisms of peripheral tolerance in the skin and if we understand the suppressive mechanisms that are compromised during vitiligo, we can potentially use this

Vitiligo is a pigmentation disorder, which causes the destruction of melanocytes in the skin. The affected areas lose their pigmentation resulting in the appearance of irregular white patches over the surface of the skin. Although the appearance of the skin can change dramatically, vitiligo does not cause any pain or physical discomfort, nor is it in any way physically limiting. Abstract:

Abstract:
Vitiligo is an autoimmune skin disease in which the pigment producing cells of the epidermis, melanocytes, are targeted for destruction by CD8+ T cells specific for melanocyte/melanoma-shared antigens. Previous work has identified IFNg as the central cytokine driving disease pathogenesis in both human patients and in our mouse model of vitiligo. IFNg signaling induces production of the chemokines CXCL9 and CXCL10, which trigger autoreactive T cell migration into the epidermis where effector T cells can target and destroy melanocytes. However, both IFNg and type I IFN signaling through activati

Background: Vitiligo is a complex autoimmune skin condition characterized by the death of melanocytes, the principle pigment producing cells in the skin. Transcriptome analysis of vitiligo skin revealed significantly reduced levels of proteolipid 1 (PLP1) gene, which is known to be expressed by Schwann cells, as well as significantly up-regulation of genes that are associated with natural killer (NK) cell activity. Hypothesis and Objectives: Schwann cells may be adversely affected in vitiligo and NK cells may potentially play a role in the overall disease pathogenesis. Therefore, the purpose o

Abstract:
Vitiligo is an acquired depigmenting disorder characterised by the loss of functional melanocytes and of melanin from the cutaneous epidermis. A role for autoimmunity in vitiligo pathogenesis is supported by the presence of anti-melanocyte autoantibodies and autoreactive T lymphocytes in patients with the disorder. Recently, the melanin-concentrating hormone receptor 1 (MCHRI) has been identified as an autoantigen in vitiligo. The aim of this study was to characterise the properties of MCHR1 autoantibodies. The epitopes on MCHR1 that are recognised by autoantibodies in vitiligo patients were i
