ABSTRACTVancomycin-resistant enterococci (VRE) are notorious clinical pathogens restricting the use of glycopeptide antibiotics in the clinic setting. Routine surveillance to detect VRE isolated from patients relies on PCR bioassays and chromogenic agar-based test methods. In recent years, we and others have reported the emergence of enterococcal strains harboring a “silent” copy of vancomycin resistance genes that confer a vancomycin-susceptible phenotype (vancomycin-susceptible enterococci [VSE]) and thus escape detection using drug sensitivity screening tests. Alarmingly, these strains are able to convert to a resistance phenotype (VSE→VRE) during antibiotic treatment, severely compromising the success of therapy. Such strains have been termed vancomycin-variable enterococci (VVE). We have investigated the molecular mechanisms leading to the restoration of resistance in VVE isolates through the whole-genome sequencing of resistant isolates, measurement of resistance gene expression, and quantification of the accumulation of drug-resistant peptidoglycan precursors. The results demonstrate that VVE strains can revert to a VRE phenotype through the constitutive expression of the vancomycin resistance cassette. This is accomplished through a variety of changes in the DNA region upstream of the resistance genes that includes both a deletion of a likely transcription inhibitory secondary structure and the introduction of a new unregulated promoter. The VSE→VRE transition of VVE can occur in patients during the course of antibiotic therapy, resulting in treatment failure. These VVE strains therefore pose a new challenge to the current regimen of diagnostic tests used for VRE detection in the clinic setting.

Abstract Background Vancomycin-variable enterococci (VVE) are a potential risk factor for vancomycin resistance gene dissemination and clinical treatment failure. vanM has emerged as a new prevalent resistance determinant among clinical enterococci in China. A total of 54 vancomycin-susceptible enterococci (VSE) isolates carrying incomplete vanM gene clusters were isolated in our previous study. Objectives To determine the potential of vanM-carrying VSE to develop vancomycin resistance and investigate the mechanism of alteration of the resistance phenotype. Methods Fifty-four vanM-positive VSE strains were induced in vitro by culturing in increasing concentrations of vancomycin. Genetic changes between three parent VVE strains and their resistant variants were analysed using Illumina and long-read sequencing technologies, quantitative PCR and Southern blot hybridization. Changes in expression level were determined by quantitative RT–PCR. Results Twenty-five of the 54 VSE strains carrying vanM became resistant upon vancomycin exposure. A significant increase in vanM copy number was observed ranging from 5.28 to 127.64 copies per cell in induced resistant VVE strains. The vanM transposon was identified as tandem repeats with IS1216E between them, and occurred in either the plasmid or the chromosome of resistant VVE cells. In addition, an increase in vanM expression was observed after resistance conversion in VVE. Conclusions This study identified tandem amplification of the vanM gene cluster as a new mechanism for vancomycin resistance in VVE strains, offering a competitive advantage for VVE under antibiotic pressure.

Growth of susceptible VVE occurred after 24 to 72 h of exposure to vancomycin due to excision of the ISL3-family element. Venous insufficiency results from several factors responsible for the progression of inflammation and oxidative damage of veins. Recently, natural extracts have been proposed for the treatment of venous insufficiency, but their efficacies have not been fully elucidated. In the present study, we evaluate the combinatorial effects on anti-inflammatory and anti-vascular leakage potential of mixed compositions containing different proportions of Centella asiatica extract (CE) and Vitis vinifera L. leaf extract (VVE) using an inflammation model of lipopolysaccharide- (LPS-) stimulated RAW264.7 cells and various vascular permeability models in mice (acetic-acid-induced peritoneal vascular model, mustard-oil-stimulated ear vascular model, and carrageenan-induced paw edema model). Pretreatment of CE and VVE in a 1 : 3 combination dose dependently inhibited the production of nitric oxide (NO) and prostaglandin E2 (PGE2) and mRNA expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) through downregulation of the nuclear factor-κB (NF-κB) pathway in LPS-stimulated RAW264.7 macrophages. In vascular permeability-related mouse models, pretreatment with the CE-VVE 1 : 3 combination significantly reduced the permeability of peritoneal or ear veins caused by acetic acid and mustard oil, respectively. Furthermore, pretreatment of the CE-VVE 1 : 3 combination ameliorated inflammation and edema of the hind paw caused by carrageenan injection. Thus, the combination of CE and VVE showed significant anti-inflammatory qualities and anti-vascular leakage effects. These findings indicate that an optimal combination of CE and VVE may have a more synergistic effect than that of CE or VVE alone as a putative agent against vascular incompetence.

<p><strong>Objective: </strong>The ethanolic extract of <em>Vitis vinifera</em> leaves (VVE) (500 mg/kg body weight), ranitidine (50 mg/kg body weight) and both of them were tested for their gastroprotective and curative effects against the incidence of peptic ulcer. <strong></strong></p><p><strong>Methods</strong>:<strong> </strong>The antiulcer effects of VVE were investigated using a combination of indomethacin and cold-stress for 2h. To ascertain the mechanism of action of VVE, its protective and curative effects were studied on gastric volume, gastric juice acidity, ulcer index and malondialdehyde (MDA), glutathione (GSH), catalase (CAT), glutathione–S-transferase (GST), superoxide dismutase (SOD), nitric oxide (NO) activities of both stomach and duodenum of rats. Moreover, histopathological effects on stomach and duodenum were determined.</p><p><strong>Results: </strong>The antioxidant activity of VVE was demonstrated using 2, 2-diphenyl-1-picrylhydrazyl (DPPH) test. VVE was found to reduce the ulcer index, gastric volume and the level of gastric juice acidity. Also, VVE showed gastroprotective and curative activities mainly through improvement of antioxidant status and decreasing lipid peroxidation accompanied with amelioration of both stomach and duodenum architectures.</p><p><strong>Conclusion: </strong>The prophylactic and curative effects of VVE proved to be effective in preventing gastric and duodenal ulceration which may be probably due to its antioxidant and anti-acid secretory effects.</p>

De grampositiva bakterierna enterokocker tillhör normalfloran i tarmen hos människor samt djur. I detta arbete studeras vancomycin-variabla enterokocker (VVE) som är vanA positiva enterokocker, känsliga för vancomycin och kan utveckla resistens in vivo. Vancomycin-resistenta enterokocker (VRE) består av två arter bakterier Entrococcus faecalis och Enterococcus faecium. VVE består av Enterococcus faecium och är svåra att detektera med odlingsbaserad diagnostik och kan därför leda till att bakterien är underdiagnostiserad, samt att den tyst kan sprida sig inom sjukhusmiljön. VVE har en förmåga att kunna övergå från vancomycin-variabla till att vara vancomycin-resistenta fenotyper. Därför undersöks i denna studie vid vilken vancomycinkoncentration (5, 4, 3, 2 mg/L) som VVE kan anrikas i buljongerna inför amplifiering med hjälp av real-time polymerase chain reaction (real-time PCR). Resultatet visade att vancomycinkoncentrationen skulle behövas sänkas till 2 mg/L, dock kan bakgrundsfloran vara för hög vid denna koncentration av antibiotika. The Gram-positive bacteria Enterococci, belong to the normal intestinal flora in humans and in animals. In the following study, vancomycin-variable enterococci (VVE) are investigated. VVE is a vanA-positive bacteria, meaning that they are sensitive to vancomycin and can develop a resistance in vivo. Vancomycin-resistant enterococci (VRE) consists of two different species of bacteria, Enterococcus faecalis and Enterococcus faecium. Furthermore, VVE consists of E. faecium and can be difficult to detect with a culture-based diagnostic andcan therefore lead to the bacterium being underdiagnosed. This in turn means that the bacteria can be quietly spread within the hospital. VVE has a unique ability, it can transition from vancomycin-variables to having vancomycin-resistant phenotypes. Therefore, this study investigates at what vancomycin concentration the VVE broths can be enriched in the broths before the qPCR. The results show that the vancomycin concentration is optimal at 2 mg/L. However, for the background flora this might be a too high concentration.

In the course of our work, we have understood that the esthetic causes represent the faithful reflection of the social and political reality experienced by the athenians. Moreover, these socio-political values are solidly anchored in the official religion consisting of myths coming from a distant past. Nous faisons d'abord un tour d'horizon de l'histoire et de la théorie du point de vue tracées par Rabatel. Nous étudions ensuite le point de vue comme un élément fondamental dans la construction de certains composants essentiels de l'énonciation narrative de "La Vie mode d'emploi" : personnages, narrateur et narrataire. Dans cette thèse, nous insistons sur deux enjeux majeurs de notre recherche qui résultent d'une certaine réciprocité de la théorie et de l'oeuvre étudiée : le premier enjeu met en avant la polyphonie comme une particularité de l'écriture perecquienne dans "La Vie mode d'emploi" : nous voyons comment la polyphonie s'installe à partir des relations que nous avons établies entre certaines notions comme l'ambiguïté, l'argumentation indirecte et les citations implicites. La mise en évidence de la polyphonie à la lumière du point de vue démontre la spécularité et la complexité de l'écriture perecquienne dans laquelle toutes les voix, celles des personnages, celle du narrateur ainsi que celle du narrataire sont représentées. Le second enjeu est l'apport de notre lecture de "La Vie mode d'emploi" à la théorie du point de vue. Notre parcours lectoral nous permet de modifier l'appareil théorique pour le rendre plus souple et répondre aux particularités textuelles du grand roman de Perec, qualifié de Tour de Babel de la littérature contemporaine.

Remi évêque de Reims (ca. 461-535) appartient à la petite noblesse sénatoriale de Laon. Influencé par Lérins, il accompagne la mise en place du pouvoir franc (baptême de Clovis). L'identification de sa famille éclaire le développement de son culte : au VIe s. , c'est un culte civique rémois, relayé au VIIe s. Par un culte aristocratique et familial (Agilolfides, Étichonides). Vers 800, saint Remi est célébré comme celui qui a converti les Francs. L'évolution culmine avec la Vie de saint Remi par Hincmar de Reims (845-882). En germe dès 852, c'est le fruit d'au moins deux rédactions, d'où son caractère hétérogène. Après Hincmar, les archevêques de Reims font de saint Remi l'image de leur autorité et l'incarnation de leurs revendications politiques. Après le sacre de Philippe Ier (1059), il devient patron de la monarchie sacrée. L'enquête s'appuie surtout sur la prosopographie (Antiquité et Moyen Age), sur l'hagiographie rémoise (datation) et sur l'édition d'un texte de Rotgar (ca. 883) Remi, bishop of Reims (ca. 461-535), was born in a senatorial family of Laon, deeply touched by the lerinian monasticism. He took his place in the bilding of the Frankish monarchy (christening of Clovis). Searching his relative, we can partly explain the expansion of his cult: during the 6th century, it is a civic cult for people of Reims, changing to an aristocratic cult during the 7th century. With the beginning of the 9th century, we observe the first celebration of saint Remi's role in converting the Franks as a nation. The theme is emphasized by Hincmar's Vita Remigii, written continually from 852 to 882. After Hincmar (845-882), the archbishops of Reims use Remi as an image of their authority and an incarnation of their political ambitions. After the coronation of Philip Ith (1059), saint Remi becomes the patron of the anointed kings. Our inquiry mainly leans on prosopography, hagiography of Reims province and the first edition of a text attributed to the monk Rotgar (ca. 883)

La recherche se propose d’introduire en sciences de gestion le concept de cherté de la vie du point de vue des consommateurs. Pour ce faire, la recherche propose, dans un premier temps, de définir le concept de cherté de la vie tel qu’évalué par les consommateurs et d’en identifier son contenu. Dans un second temps, la recherche pose la question des antécédents de cette évaluation et de ses conséquences sur le comportement d’achat des consommateurs.La démarche empirique s’appuie sur une Mixed Methods Research. Il s’agit là d’une méthode de recherche qui implique de combiner à la fois les éléments d’une approche quantitative et d’une approche qualitative à des fins de compréhension et de corroboration. Dans cette optique, deux principales études empiriques ont été réalisées : (1) une étude exploratoire qualitative composée de plusieurs méthodes de collecte de données (entretiens semi directifs, netnographie, sondage en sortie de supermarché) et (2) une étude quantitative confirmatoire menée auprès de 700 résidents de l’île de La Réunion.L’étude qualitative permet (1) de déterminer les causes de cette cherté de la vie du point de vue des consommateurs, (2) de définir l’évaluation de la cherté de la vie et de structurer son contenu et (3) d’analyser les conséquences de cette évaluation sur le comportement d’achat des consommateurs. A la lumière de ces premiers résultats et de la revue de la littérature, une série d’hypothèses ainsi que le modèle de la recherche qui en découle sont alors formulés. Sono stati esaminati vantaggi ed eventuali problematiche, ed esposti esempi pratici di diagrammi di distribuzione nel caso di applicazione ad un motore tradizionale ad accensione comandata a ciclo Otto, ad un motore ad accensione comandata a ciclo Miller e, infine, ad un motore a ciclo Otto funzionante tramite accensione per compressione di una carica omogenea (HCCI).
In tutti e tre i casi si sono ottenuti benefici derivanti dall’applicazione dell’attuazione variabile. Gli unici svantaggi rispetto alla fasatura fissa risiedono nella maggior complicazione costruttiva e nella necessità di una calibrazione adeguata per lo sfruttamento del sistema.
Delle tre tecnologie esaminate, l’applicazione a motori Otto ad accensione comandata risulta essere la più matura, con un livello di sviluppo altissimo e larga diffusione nella produzione commerciale; l’applicazione dei cicli Miller occupa una fetta di mercato molto ridotta, ma diversi costruttori ne stanno portando avanti lo sviluppo da tempo, arrivando ormai ad alti livelli di ottimizzazione, per cui il suo utilizzo sui veicoli commerciali, in particolare quelli ibridi, è destinato a crescere. La combustione HCCI è la più acerba delle tre tecnologie, ma l’interesse dei costruttori sull’argomento è molto vivo e diversi produttori hanno pubblicato risultati di ricerca in questo ambito.

Quand en 1940 la France devient territoire occupé, les conditions de vie se dégradent à tel point que les Français vivent en grande difficulté. Pourtant, aussi épouvantables que les conditions matérielles de vie soient-elles, rien n’est comparable à la dégradation de la vie de l’Esprit, au manque du spirituel. Les Français sont affamés, les Français ont froid, mais pire encore, les Français meurent dans l’esprit. Les Français perdent peu à peu leur identité. « C’est l’âme aujourd’hui qui est tellement déserte. On meurt de soif », s’exclamait Antoine de Saint-Exupéry dans une lettre à sa mère en 1940. Et pourtant on ne peut pas s’abreuver parce qu’il n’existe que « ce marécage ». La douleur est trop forte parce que c’est le patrimoine spirituel français qui est en péril et de nombreux écrivains en sont très conscients. L’essence de la vie, l’eau claire, l’eau limpide, l’eau qui donne la vie, s’est transformée en eau de mort, « une eau qui doit s’assombrir, une eau qui va absorber la noire souffrance. Toute eau vive est une eau dont le destin est de s’alentir, de s’alourdir. Toute eau vivante est une eau qui est sur le point de mourir » disait Bachelard dans son livre L’eau et les rêves. La vie est étouffée et il n’y a que des eaux stagnantes, des « eaux mortes ». Dans cette communication je voudrais montrer comment les écrivains ressentent et manifestent cette crise, cette perte d’identité, cette mort ; et comment l’écriture est proposée comme remède, comme voie de salut grâce à la démarche autobiographique d’une manière spéciale. This paper presents the simulation model of an oil lubrication gerotor pump for the internal combustion engine. The model was constructed by using a mono-dimensional commercial code taking into account all the phenomena that occur during the revolution of the pump shaft. First of all, several geometric considerations and theoretical observation are presented. An experimental campaign was also performed to validate the simulation model. In these experimental tests particular attention was regarded to the behavior of the pressure oscillations during the pump shaft revolutions. The final aim of this activity is to obtain an instrument that allows in-depth analysis of pump and lubrication circuit functioning and that can be coupled with other models (e.g. VVA, VVT, etc.) in order to take into account different problems of the hydraulic components of engines.
