Private business subjects are characterised by their continuous effort for increasing their market position. Therefore they are permanently monitoring their own sthrengths and weaknesses, and they are also monitoring their competitors´ behaviour and customers´ needs and wishes. Such pieces of information can be received by analysis of business surroundings and from custommers´ opinion. Activities like these are called marketing research. The research should include all aspects that influence the way of perception of the company by any other market subject, or even the atmosphere inside the company. Information received is valuable for managers and their decision-making. Marketing research is a dynamicly developing field where new findings of social sciences are permanently applied. A significant developement in this field was reached mainly in last years. It is a result of the rapid progress of information and communication technologies. Modern forms of marketing research often use the Internet for collecting various information on the market.

This thesis is intent on the introduction to problems of statistical approach to research tasks. It focuses on research assignments, position of research worker and statistician while analyzing, ways of gathering data files and problems connected with them, main types of multivariate statistical methods and possible views of their classification. Moreover, this work includes overview of examples of research assignments, possibilities of their solutions and related data files. First chapter describes statistical approach to the research assignments, and the second one shows concrete examples of these assignments. The enclosed CD includes data files to most of the statistical examples.

The main aim of doctoral thesis is the study on physicochemical properties of humic substances (HS) by modern instrumental techniques. The subject of the study were HS isolated from South Moravian lignite, South Bohemian peat, forest soil Humic Podzol and finally extract from brown sea algae Ascophyllum nodosum. With respect on determination of structure and reactivity of these unique “biocolloids”, standard samples (Leonardite HA, Elliott Soil HS and Pahokee Peat HS) were also studied. These samples were obtained from International Humic Substances Society (IHSS). All mentioned substances were characterized by elemental analysis (EA), molecular absorption spectroscopy in ultraviolet and visible region (UV/Vis), infrared spectroscopy with Fourier transformation (FTIR), nuclear magnetic resonance spectroscopy of carbon isotope 13C (LS 13C NMR), steady-state and time resolved fluorescence spectroscopy. Obtained fluorescence, UV/Vis and 13C NMR spectra were used for calculation of fluorescence and absorption indexes, values of specific absorbance and structural parameters respectively, which were used for fundamental characterization of these “biocolloidal” compounds. Infrared spectroscopy with Fourier transformation was utilized for the identification of functional groups and structural units of HS. Evaluation of infrared spectra is quiet complicated by overlapping of absorption bands especially in fingerprint region. This problem was overcome by Fourier self-deconvolution (FSD). Steady-state fluorescence spectroscopy was used for deeper characterization of HS with respect to origin, structural units, amount of substituents with electron-donor and electron-acceptor effects, content of reactive functional groups, “molecular” heterogeneity, the degree of humification, etc. Parameters of complexation of samples Elliott Soil with heavy metal ions (Cu2+, Pb2+ and Hg2+) were obtained by using modified Stern-Volmer equation. These ions were chosen purposefully, because the interaction of HS with these ions is one of the fundamental criteria for the assessment of the reactivity of HS. Key part of the whole doctoral thesis is time-resolved fluorescence spectroscopy. It is able to determine the origin of emission of HS by method Time-Resolved Area Normalized Emission Spectra (TRANES). The viscosity of micro medium about excited fluorophores of HS was determined by Time-Resolved Emission Spectra (TRES).

In this contemporary world, when the globalization is on the first place, is possible to satisfy one's needs immediately. Road haulage becomes in last few years a significant market's part, which is necessary to the right function in all branches in the Czech Republic. The aim of this thesis is to analyse level of principle operation's research's support in TASW solution for the society, which is directed at road transport. Simultaneously I project a study, which supports a solution for small shippers, who aren't able or willing to acquire an expensive software for haulage entrepreneurship. I characterize issues of transportation's company to achieve my aim and I think about the principle's uses and method of operation's research in process of haulage entrepreneurship. I analyse a situation of operation's research's support with TASW, which is oriented on haulage entrepreneurship. After that I describe a study, where I project a solution for small shippers, which takes in the consideration the components of operation's research in their processes and TASW and this contributes to make the decision of small shippers easier to buy the suitable software for haulage. This thesis is divided into few parts -- at the begging I theoretically follow up haulage entrepreneurship, its division and kind of current freight. I describe the price creation to make really obvious, which knowledge the shipper have to know. I explain particular disciplines of research's operation, which are connected to make more effective the workings of haulage entrepreneurship. I use these results of the theoretical parts to create my own model of haulage entrepreneurship. I analyse the market's system in the Czech market, which is fixed to support process of haulage entrepreneurship, which applies the principles of operation's research. In conclusion I compare the variants of using different types of IS for transport. The biggest benefit of my thesis I see in that also the laymen can understand it. It makes easier to choose a specialized software for haulage entrepreneurship and the process analyse binding on possibly company's analyse and on field operation's research, which enable more effective delivery.

Hyaluronová kyselina je v současné době velmi významná biomolekula používaná v medicíně a kosmetice, a její výzkum je důležitý pro další budoucí použití. Zaměření této práce je studování povrchových vlastností hyaluronové kyseliny, jejích hydrofobních derivátů a roztoků SDS pro srovnání, za použití tenziometrie. Tyto sloučeniny byly zkoumány ve vodě a v roztocích ve formě sodné soli. Sledované vzorky byly měřeny dvěmi metodami v různých koncentračních rozmezích při laboratorní teplotě pomocí nového BPA-800P bublinového tenziometeru. Byly navrženy takové experimenty, aby se zjistily využitelné možnosti tohoto přístroje pro další výzkum. Výsledky práce ukazují rozdílnosti jednotlivých povrchově aktivních látek v závislosti na jejich koncentraci a použitém prostředí. Hyaluronová kyselina nevykazuje povrchovou aktivitu, zatímco její deriváty a SDS ano.

The aim of this diploma thesis is a selection of the best current account. This thesis describes how the bank system functions in the Czech Republic. The characteristics of the banking institutions which provide current accounts and the characteristics of the compared current accounts are presented in this thesis. The Monte Carlo method and the methods multi-criteria assessment of variants are described theoretically. The selection is based on the methods of the operational research. The first part of the practical chapter focuses on the Monte Carlo method which selects the best account by charges for different types of clients. The methods of multi-criteria assessment of variants, specifically methods TOPSIS and PROMETHEE, are also applied. The calculations are carried out by add-in SANNA for MS Excel. In conclusion, both approaches to solution are compared and the current account according to the preferences of each client type is recommended.

This diploma thesis deals with the ethics of marketing research conducted with underage respondents. Child security is a very hot topic these days; therefore the field of market research should use only methods tolerated by parents. Global as well as local market research associations regularly publish codes of conduct and guidelines dealing with the ethics in market research. However, they define only basic rules and regulations and do not reflect the specificity of each method and technique. The goal of this diploma thesis is to identify methods used in market research that are not perceived as ethical by parents. Results should motivate research agencies to eliminate using such methods or at least modify them to decrease the level of controversy among general public. In order to identify unethically perceived market research methods, the author of the thesis conducted a quantitative research among the needed target group: parents of school kids. The survey found 5 methods that are considered ethical and thus do not pose any significant threat to the industry of market research: taste test, observation, product test, eye-tracking and communication test. Two methods were labeled as unethical by the respondents: field experiment and online discussion groups. Those methods can be used without any major changes provided that agencies communicate openly with parents all details regarding the research. They have to make sure that parents understand the methodology and are sure to let their children participate in such a project. Finally, three methods were recognized as highly unethical or even illegal: neuromarketing, netnography and geolocation (geomarketing). These methods carry a high probability of damaging the positive image of market research industry among general public. Less than one quarter of respondents consider these methods as suitable for use with kids and at the same time more than 25 % of parents think that those methods are illegal. In case of neuromarketing, it is recommended to firstly explain the used methodology in detail and, if possible, demonstrate its 100 % safety in presence of a doctor. Netnography was evaluated as highly unethical. On the other hand, it is a type of observation; therefore, its use in reality is unlikely to be registered by public. As a result, there does not have to be any severe steps taken to regulate this method. Geolocation scored as the most unethical market research method in the survey. This method is therefore not recommended for use as it carries an enormous risk of damaging the whole market research industry.

The presented dissertation is focused on research in the field of dowels and design of concrete pavements using the finite element method. The theory part describes the history and the currently used techniques in the world and in the Czech Republic. The practice part of the dissertation contains a description of design methods, diagnostics of concrete pavements by using FWD and a description of laboratory tests on beams with inserted dowels. The results of measurements and modelling are verified on real operating conditions on testing fields. Based on the dissertation results, the amendment of the Czech standard ČSN 73 6123-1 regarding the position of dowels were proposed and approved. In addition, methodological guidelines of the Ministry of Transport regarding the measurements of the dowels and tie bars position and diagnostic by FWD was approved. The conclusion of the work summarizes and emphasizes the results of the dissertation.

The output of this work is device that is able to implement and compare quality of codification methods mainly for scanning of problematic surfaces. The functionality of equipment was verified by experimental measurement.

Master thesis discusses a reflection of a project To have a body, to be a body which took place in the years 2009 ? 2012. Its topic was a human body and the way how we approach our corporeity and how we relate to it in present days. The project included the experience of people with various types of health or social handicap and also a people whose relationship to own bodies was influenced by their profession. The first part which was the research part included the series of six workshops with following groups: people with mental handicap, boys from juvenile detention center, elderly people, people with physical handicap, IT experts and physicians. The teachers of the workshops were utilizing various movement concepts, approaches and methods (e.g. Feldenkrais method, modern dance, rhythmics, dance and movement therapy, dramatherapy, physiotherapy, neutral mask?) with a common goal to bring the participants to self-expression and self-recognition through their own bodies and authentic movement. In the second part which was the performance part, the authorial performance was created (publicly performed in the years 2010 ? 2012), in which the participants of former workshops took part as authors and performers. The project has had inclusive and integrative aspects. The thesis provides an insight into the project by means of thematic profiles based on its major topics ? it includes chapters Lecturers, teachers and their work in the groups, Group dynamics, Assistance and its changes during the project, Integration, inclusion, re-socialization and project etc.). Among others, one of the resources is wide documentary material gathered continuously during the project. It includes quotes from diary records, interviews with teachers, assistants, participants and curators of workshop groups and from questionnaires for participants and authors of the project. There is rich video and photo documentation added to the thesis.

The aim of this Master's Thesis is to create an employee shift work schedule by using selected methods of operations research. The first part is theoretical and deals with the introduction into operational research, specifically in the field of mathematical programming and simulation models, including the possibility of computer processing. This first part highlights important subjects that will be used in the second part, which is practical and is solving a specific problem. Firstly, Cinema City Flora, which provided data for the analysis, is presented. Then there is a solution of the problem in the add-in Solver in MS Excel, and then in the program LINGO. After, a discrete simulation is created in the program SIMUL8. In conclusion, there is a summarization of the main findings and recommendations, comparison of selected methods and their applicability in practice and possibilities of further research.

The master thesis "Research of a megatrend and its commercial potential" uses the qualitative method Delfi and MML-TGI data. The research has showed that one future megatrend is the creation of a new platform which will replace the current internet platform. Furthermore, the research has helped to define the marketing potential of the future platform. The thesis firstly contains a theoretical introduction into the world of megatrends, defines what a megatrend is together with a possible prediction of future events, and looks at the overall characteristics of what makes-up a megatrend. It also looks at current megatrends, ways how to deal with them, and analyses reactions of different industries towards them. Secondly, the thesis follows through with a theoretical analysis of the research methods used, such as the Delfi method. This part contains a detailed methodology of the research with the use of Delfi and MML-TGI. The focus and objective of the thesis lies in researching the future megatrend and its marketing potential. A prerequisite to fulfilling the main objective is to acquire relevant data which will allow for the research to be completed. The thesis deals with the problem in the form of the possibility to predict a future megatrend and its marketing potential, especially in the relation of availability and supply of relevant, trustworthy information.

Next, the relationship between national and organizational culture is described and recent intercultural researches are assessed. On this theoretical framework, the Swedish culture research is based. The main benefit of the research is the subjective comparison method that describe in detail position of Swedish culture compared with another. At the end, the Swedish national culture results are gently transferred to organizational culture. The goal of this paper is contemporary assessment of Swedish culture and topics suggested for next research.

In the first part of thesis the functioning of the electricity market in the Czech Republic is explained and the theoretical assumptions, which are necessary for understanding the issue is described. In the practical part of the thesis some methods of multicriteria decision making and of data envelopment analysis models are presented. The results of each method are explained in detail and they are compared with results of other methods.

The theoretical part introduces Systemic Approach problem solving, as well as the methodology of Six Sigma and Lean Management and its analytical tools, and finally with Logistics as the section of Operations Research. The practical part contains a brief introduction to the company profile, the use of Systemic Approach and methods of Six Sigma and Lean. Procedures of these methods are compared afterwards. Within the System Approach is also done the analysis using Operational Research Method - the Queuing System. Based on the analysis is determined a model that expresses the best use of the system.

This thesis deals with modern statistical approaches and their application aimed at robust methods and neural network modelling. Selected methods are analyzed and applied on frequent practical problems in czech industry and technology. Topics and methods are to be benificial in real applications compared to currently used classical methods. Applicability and effectivity of the algorithms is verified and demonstrated on real studies and problems in czech industrial and research bodies. The great and unexploited potential of modern theoretical and computational capacity and the potential of new approaces to statistical modelling and methods. A significant result of this thesis is also an environment for software application development for data analysis with own programming language DARWin (Data Analysis Robot for Windows) for implemenation of effective numerical algorithms for extaction information from data. The thesis should be an incentive for boarder use of robust and computationally intensive methods as neural networks for modelling processes, quality control and generally better understanding of nature.

The topic of this diploma thesis is the application of certain operation research methods on a practical example. The main objective is to evaluate and compare hypermarkets of selected retail company on the basis of data from cosmetics department for the year 2013. In the thesis are firstly described theoretical assumptions necessary for understanding the decision theory and DEA models, next part is then focused on selected company and its history, current market position and the different types of store formats. Following practical part shows the application of multicriteria evaluation methods (WSA, TOPSIS, ELECTRE III) and DEA models (CCR and BCC) on concrete intern data and results which are then commented, compared and completed with recommendations, how to eliminate store or stores weaknesses, if needed. All calculations in the practical part of this thesis were done by the supplement of MS Excel -- Sanna and optimalization software LINGO.

This diploma thesis deals with the real use of operation research method in conjunction with logistics Cross-Docking technology. It is possible to solve this conjunction with either routing problems along with its modifications or simulations methods. The theoretical part describes routing problems, routing problems with time windows, simulations methods with their possibilities of real use and logistics technologies. A model of a Cross-Docking terminal that was created in the program Simul8 can be found in the practical part of this thesis. The goal of the simulation was to ensure smooth flow of products through the terminal without any long queues in the terminal. Routing problems designed to minimize the length of its routes can be found in the second half of the practical part. Other possible real application is routing problems on the real problem. The solution for this task was found using the modelling system MPL for Windows.

This thesis deals with methods of recruitment, which play an important role
for the proper functioning of organization. Employees are an important element
of any society because they are carriers of knowledge. The aim is to identify and evaluate methods of recruitment in the selected organization. In case of deficiencies are identified, suggest appropriate changes that will lead to the improvement of the methods
of recruitment.
The theoretical part was processed by the methods of the analysis of primary
and secondary sources of information, relevant to the issue. The views of the individual authors focused on the issue were compared and then using the method of the synthesis processed in the theoretical background of the thesis. The practical part was prepared
based on the study and analysis of primary and secondary sources and methods of personal and written questioning, was therefore carried out quantitative research (questionnaire)
and qualitative research (semi-structured interview). On the basis of a synthesis
of the results of this research and the previous outputs of the thesis, proposals
and recommendations were processed. The main weakness in the studied area
is underestimating the methods of the recommendation by the company, even though
93% of employees would recommend to a friend, to run for the job in the organization.

The company is fully dependent on the actions of people who work in it, because people make up a very significant element of the company.Recruitment methods applied to each company, when he tries to get the best potential candidates. The target of the thesis was to evaluate methods of recruitment at Schneider Electric, a. s., and a staffing agency Manpower, in case of deficiencies suggest better proposals for recruitment. To obtain the information needed to develop the work was analyzed literature and internal documents of the company. The results were achieved using qualitative and quantitative research. It was applied so. Interrogation method using a questionnaire survey among employees, and structured interviews with the personnel department of Schneider Electric, a. s. And a staffing agency Manpower. From the survey results, it was found that Schneider Electric, a. s. Uses modern methods of recruitment. The findings are recommended suggestions to improve methods of recruitment. In the future the company should expand cooperation with educational institutions to participate in the job fair, organize teambuilding and hold regular meetings, to change the company's official website - a regular update of jobs, improve cooperation with the staffing agency Manpower, and expand advertising jobs from across the web website.

In chapter 1 the work deals with the theoretical background of this method and shows the principles of a desorption process influenced by subsurface diffusion. Chapter 2 first shows application of TDS for detection of surface molecules and determination of binding energy.Experiments were mainly focused on ditermination of surface adsorbents and impurities on Si wafers. The second part of chapter 2 describes desorption of atoms of a Ga layer on Si surface and their subsurface diffusion. A Ga diffusion process was also observed by with secondary ion mass spectrometry (SIMS) and numerically simulated.

This thesis is focused on analyzing the process of an online request for a consumer loan. It targets its weak points and creates a set of recommendations that should lead to more user - friendly environment. In the theoretical part, the thesis covers issues of consumer loans and explains topics from user testing and usability. It also describes methodology of the research. The practical part of this thesis is centered around a general simulation model, which was created based on the previous analysis. With this model and using selected methods the thesis identifies critical points common to most tested participants. Based on the results of the user testing, recommendations are made to resolve these issues. The general model was modified according to the validity of these recommendations and then subjected to re-testing. This thesis is unique for the use of an eye-tracking camera among other methods.

The thesis is focused on the research method of Mystery Shopping, used for anonymous and impartial evaluation of service quality and customer service. In the theoretical part I will examine how and why the method is used, what is the procedure for its application and how the results are then interpreted. An important part is the ethical codes that are specific to this method. In the practical part I will use theoretical areas of knowledge which will then be applied into practice. In the selected company measures of the level of customer service are examined with the just-mentioned method. After the research I was able to reveal the company's management strengths and weaknesses in customer care, and advise them on which areas should be addressed in the next period. I also suggested ways to optimize methods for the mystery shopping company to give better information about the company's management level customer service to stores.

The author applies marketing research methods, in order to work out a recommendation for sales growth on the background of an IT services company looking for expansion in the hotel sector in the Czech Republic. Through the application of qualitative and quantitative research methods it is established that the anonymized company needs to focus on the segment of three to five star hotels in Prague that do not have their HQ outside of the Czech Republic. Furthermore, a clear product focus on cost saving utilisation of product like virtualization and office automatization is advised, whilst expanding the portfolio on basic hotel needs like camera and entertainment systems. Finally, the analysis recommends to establish professional behaviour and appearance guidelines for in the field service technicians.

Grafomotorické dovednosti (GA) představují skupinu psychomotorických procesů, které se zapojují během kreslení a psaní. GA jsou nutnou prerekvizitou pro zvládání základních školních schopností, konkrétně psaní. Děti v první a druhé třídě mohou mít potíže s prováděním jednoduchých grafomotorických úkolů (GD) a později ve třetí a čtvrté třídě také se samotným psaním (HD). Narušení procesů spojených se psaním je obecně nazýváno jako vývojová dysgrafie (DD). Prevalence DD v České republice se pohybuje kolem 3–5 %. V současné době je DD hodnocena subjektivně týmem psychologů a speciálních pedagogů. V praxi stále chybí objektivní měřicí nástroj, který by umožňoval hodnocení GD a HD. Z tohoto důvodu se tato disertační práce zabývá identifikováním symptomů spojených s grafomotorickou neobratností u dětí školního věku a vývojem nových parametrů, které je budou kvantifikovat. Byl vytvořen komplexní GA protokol (36 úloh), který představuje prostředí, ve kterém se mohou projevit různé symptomy spojené s GD a HD. K těmto symptomům bylo přiřazeno 76 kvantifikujících parametrů. Dále byla navrhnuta nová škála grafomotorických obtíží (GDRS) založena na automatizovaném zpracování online píma. Nakonec byla prezentována a otestována nová sada parametrizačních technik založených na Tunable Q Factor Wavelet Transform (TQWT). Parametry TQWT dokážou kvantifikovat grafomotorickou obratnost nebo nedostatečný projev v jemné motorice. GDRS přestavuje nový, moderní a objektivní měřící nástroj, který doposud chyběl jak v České republice, tak v zahraničí. Použití škály by pomohlo modernizovat jak diagnostiku DD, tak reedukační/remediační proces. Další výzkum by tento nástroj mohl adaptovat i do jiných jazyků. Navíc, tato metodologie může být použita a optimalizována pro diagnostiku dalších nemocí a poruch, které ovlivňují grafomotorické dovednosti, například pro autismus, poruchu pozornosti s hyperaktivitou (ADHD) nebo dyspraxii (DCD).

In the theoretical part I summarized nowadays Framework educational program at schools in the Czech republic and nowadays educational methods to teaching plants. Also, the thesis describes basic characteristics of plants, methods of plant preservation and research methods that have been used during writing the thesis. The research part indicates the results of the research conducted at grammar-schools in Ústí region and indicates the level of the pupils' knowledge of plants. Furthermore, I have tried to suggest how to make the plant identification tests more intriguing and comprehensible. The thesis uses information obtained from interviews with teachers and survey research carried out among the pupils of Ústí region grammar-schools. Based on the research results I have laid out an optimal number of plant species for teaching the plant identification. Unfortunately, there is a limited number of publications, so I exploited the knowledge and experience of Biology teachers at grammar-schools, and I followed their advice when creating worksheets.

The diploma thesis deals with evaluation of subjective well-being of elementary school teachers with use of anchoring vignettes. General goal of the thesis is to research actual subjective well-being of teachers and find out what factors are affecting it. Theoretical part characterizes subjective well-being and its components, its interconnection with life satisfaction, quality of life and happiness. Theoretical part also consists of explanation of the anchoring vignettes method including description of its application in particular researches. Empirical part of the thesis uses an existing survey method called Diener's satisfaction with life scale supplemented by anchoring vignettes. The research refers to high level of subjective well-being among teachers. Some variables (eg. family status, work in city or village) which make differences between levels of satisfaction were found. Application of anchoring vignettes method confirmed results of self-assessing questions in most cases. The theoretical part describes chosen methods and techniques of active teaching within the context of the educational process. An experiment was carried out in the practical part and it verified the influence of these methods on the development of essay-writing potential of the pupils. The popularity of the chosen methods and techniques of active teaching was being researched by a questionnaire survey. The methods of active teaching were discovered to facilitate the success rate of the essay-writing and a statistically significant increase in the number of sentences and syntactic pairs was found in the experimental group.

This paper analyzes methods of therapeutic effect's research, its advantages and limitations. It describes the theory of therapeutic change, medical model of measurements and randomized clinical trials. Based on this we suggest a three-level evaluation of change, where clinical, statistical and comparative (effect size) view is used. We conclude that the high effectiveness of therapy has been proven, but it has not been determined so far, which therapy is the most effective and for which diagnosis. The techniques of therapies are far less relevant than the capabilities of the therapist and client and the common factors. In the empirical part we measure the effect of sevenweeks- long outpatient therapeutic community program at Horni Palata. In a non-RCT study with 162 clients we found, that the change was positive and statistically significant. The evaluation differs much with the tool (SCL-90, DZS, MMPI-2) and method that is used. The percentage of clinically improved ranges form 25 to 68, the effect size is between 0.33 and 0.66. The effect doesn't differ with sex, age, medication, diagnosis and therapist, but it correlates with the pre-treatment scores.

Author of these thesis researches the ability Paired comparison method to predict the results in the presidential elections during January 2018 ( the first and the second round). The pair comparison method is based on the assessment of individual candidates in combinations (pairs) where two candidates stand against each other and there is a judge who compares them in all combinations. The author carried out an investigation using three standardized questionnaires. She has applied the method in the first questionnaire and surveyed the actual choice of the respondents in the other two questionnaires. There were 392 respondents who participated in the complete questionnaire surfy and their answers were analyzed and compared with their results of Paired comparison method. The thesis also shows the accuracy of the method and the suitability of the application, as well as the ability to predict the results of undecided voters. The Paired comparison method in prediction of the results of the first round, but was able to estimate the results of the second round. The author concludes that Paired comparison method should be used in several round types of elections or those where two candidates stand against each other. There are also examined the disadvantages and risks of the method.

Title of Doctoral Thesis: Using of separation methods for clinical research This doctoral thesis deals with development and validation of a chromatographic determination of liposoluble vitamins and with optimization of biological materials preparation. The first part of this work is focused on vitamins D, A and E assessment in serum. To separate studied analytes two different methods were developed successively. 1) Separation employing monolithic columns - two different monolithic columns Chromolith Performance RP-18e, 100×4.6 mm and SpeedRod RP-18e, 50×4.6 mm were connected in series. Vitamins 1,25(OH)2D3, 25(OH)D3, retinol, tocol (internal standard), cholecalciferol, ergocalciferol and α-tocopherol were separated within 6.5 minutes using step gradient elution. 2) Separation exploiting a core-shell column - Ascentis Express RP-Amide 75×3.0 mm. To decrease the separation time a linear gradient was employed. The determination of 25(OH)D2, 25(OH)D3, tocol (internal standard) α-tocopherol took 8.5 minutes within separation of impurities.
