Abstract:

Vordergründige Argumente (A) waren Bewegungsbehinderungen, Aktivitätseinschränkungen und Schmerzen.

Abstract:
Abstract The stock market participation rate among homeowners is twice as high as among renters. This paper builds a life-cycle portfolio choice model with endogenous housing tenure choice. A stylized form of preference heterogeneity generates a substantial difference in participation rates. A majority of households have a large savings motive and choose to be homeowners and participate. A minority of households have a small savings motive and find it less worthwhile to participate. Fewer of these households become homeowners. Difference-in-difference regressions on panel data do not find evidence of a crowding-out effect of homeownership on participation, supporting the message that preference heterogeneity matters. Abstract:
Abstract Motivation Post-translational modifications (PTMs) of proteins are associated with many significant biological functions and can be identified in high throughput using tandem mass spectrometry. Many PTMs are associated with short sequence patterns called ‘motifs’ that help localize the modifying enzyme. Accordingly, many algorithms have been designed to identify these motifs from mass spectrometry data. Accurate statistical confidence estimates for discovered motifs are critically important for proper interpretation and in the design of downstream experimental validation. Results We describe a method for assigning statistical confidence estimates to PTM motifs, and we demonstrate that this method provides accurate P-values on both simulated and real data. Our methods are implemented in MoMo, a software tool for discovering motifs among sets of PTMs that we make available as a web server and as downloadable source code. MoMo re-implements the two most widely used PTM motif discovery algorithms—motif-x and MoDL—while offering many enhancements. Relative to motif-x, MoMo offers improved statistical confidence estimates and more accurate calculation of motif scores. The MoMo web server offers more proteome databases, more input formats, larger inputs and longer running times than the motif-x web server. Finally, our study demonstrates that the confidence estimates produced by motif-x are inaccurate. This inaccuracy stems in part from the common practice of drawing ‘background’ peptides from an unshuffled proteome database. Our results thus suggest that many of the papers that use motif-x to find motifs may be reporting results that lack statistical support. Availability and implementation The MoMo web server and source code are provided at http://meme-suite.org. Supplementary information Supplementary data are available at Bioinformatics online.

Abstract Motivation Motif-HMM (mHMM) scanning has been shown to possess unique advantages over standardly used sequence-profile search methods (e.g. HMMER, PSI-BLAST) since it is particularly well-suited to discriminate proteins with variations inside conserved motifs (e.g. family subtypes) or motifs lacking essential residues (false positives, e.g. pseudoenzymes). Results In order to make mHMM widely accessible to a broader scientific community, we developed Leitmotif, an mHMM web application with many parametrization options easily accessible through intuitive interface. Substantial improvement of performance (ROC scores) was obtained by using two novel parameters. To the best of our knowledge, Leitmotif is the only available mHMM application. Abstract:
The development of chromatin immunoprecipitation sequencing (ChIP-seq) technology has revolutionized the genetic analysis of the basic mechanisms underlying transcription regulation and led to accumulation of information about a huge amount of DNA sequences. There are a lot of web services which are currently available for de novo motif discovery in datasets containing information about DNA/protein binding. An enormous motif diversity makes their finding challenging. In order to avoid the difficulties, researchers use different stochastic approaches. Unfortunately, the efficiency of the motif discovery programs dramatically declines with the query set size increase. This leads to the fact that only a fraction of top “peak” ChIP-Seq segments can be analyzed or the area of analysis should be narrowed. Thus, the motif discovery in massive datasets remains a challenging issue. Argo_Compute Unified Device Architecture (CUDA) web service is designed to process the massive DNA data. It is a program for the detection of degenerate oligonucleotide motifs of fixed length written in 15-letter IUPAC code. Argo_CUDA is a full-exhaustive approach based on the high-performance GPU technologies. Compared with the existing motif discovery web services, Argo_CUDA shows good prediction quality on simulated sets. The analysis of ChIP-Seq sequences revealed the motifs which correspond to known transcription factor binding sites.

Abstract:
This article focuses on Wilfried Steiner’s 2003 novel Der Weg nach Xanadu / The Way To Xanadu which appears to be an Austrian campus novel owing to the setting of the Austrian world of academia in its first part. Owing to its lack of local coloring, however, the Vienna-based plot of the first part does not feature a (stereo)typically ‘Austrian’ genius loci. Although this part of the text echoes features of the international campus novel tradition, it may be definitely not considered an Austrian campus novel. The second part of the novel is set in the Lake District, focuses on Coleridge’s Romantic poetry and the Doppelgänger-motif. Whereas the first part may be vaguely contextualized within the international campus novel tradition, the second part is deeply imbued with the Romantic tradition of the Künstlerroman or artist’s novel.

AbstractSummaryUnderstanding the regulatory roles of non-coding genetic variants has become a central goal for interpreting results of genome-wide association studies. The regulatory significance of the variants may be interrogated by assessing their influence on transcription factor binding. We have developed atSNP Search, a comprehensive web database for evaluating motif matches to the human genome with both reference and variant alleles and assessing the overall significance of the variant alterations on the motif matches. Convenient search features, comprehensive search outputs and a useful help menu are key components of atSNP Search. atSNP Search enables convenient interpretation of regulatory variants by statistical significance testing and composite logo plots, which are graphical representations of motif matches with the reference and variant alleles. Existing motif-based regulatory variant discovery tools only consider a limited pool of variants due to storage or other limitations. In contrast, atSNP Search users can test more than 37 billion variant-motif pairs with marginal significance in motif matches or match alteration. Computational evidence from atSNP Search, when combined with experimental validation, may help with the discovery of underlying disease mechanisms.Availability and implementationatSNP Search is freely available at http://atsnp.biostat.wisc.edu.Supplementary informationSupplementary data are available at Bioinformatics online.

Abstract:
Abstract The interaction between RNA and RNA-binding proteins (RBPs) has a key role in the regulation of gene expression, in RNA stability, and in many other biological processes. RBPs accomplish these functions by binding target RNA molecules through specific sequence and structure motifs. The identification of these binding motifs is therefore fundamental to improve our knowledge of the cellular processes and how they are regulated. Here, we present BRIO (BEAM RNA Interaction mOtifs), a new web server designed for the identification of sequence and structure RNA-binding motifs in one or more RNA molecules of interest. BRIO enables the user to scan over 2508 sequence motifs and 2296 secondary structure motifs identified in Homo sapiens and Mus musculus, in three different types of experiments (PAR-CLIP, eCLIP, HITS). The motifs are associated with the binding of 186 RBPs and 69 protein domains. The web server is freely available at http://brio.bio.uniroma2.it.

Abstract:
Sport event tourism is a major component of sport related tourism in many countries. Sport event organizations should strive to develop Internet marketing communication that features event information relevant to potential sport tourists. Using the Psychological Continuum Model (PCM) as its theoretical framework, this article presents two studies examining information requirements for sport event Web sites and evaluating the impact of Web site communications on consumer motivation and attitudes toward the event. Study 1 first used an open-ended response listing exercise to identify 15 information themes that should be accessible on a sport event Web site (N= 54) and then demonstrated in a between-subjects experimental design that providing these information themes increased satisfaction with the Web site (N= 40). Study 2 used a within-subjects experimental design to reveal that provision of these information themes had no impact on travel motives, but did increase favorable attitudes toward a sport event and intention to attend the event (N= 39). This research provides evidence that Web site marketing communication does activate attitude change within consumers, as well as empirical support for attitude change within the PCM framework. Findings highlight the potential strategic use of Web site communication for sport event organizers to enhance consumer attitudes toward the event and increase attendance.

Abstract The article deals with the question how the Torah can become a relevant topic in learning and teaching processes in religious education. In doing so, the motif of „godly and humanly commemoration“ is used to elaborate on the idea that learning is commemoration in a two-fold sense: „It is the commemoration of god and the commemoration of the human being, in which the shared memory of the human being and god form a oneness.“ Derived from this idea the thought is developed, that such a learning remembrance is on the one hand aimed at the interpretation of the present, but on the other hand also aimed at creating perspectives for the future. Commemoration aims at concretion in the present with a glance at the future.

Abstract:
229 Background: Estimates of the 1 year implementation delay of ICD-10-CM (I10) range upto $6.6 billion with the majority of cost attributable to I10 implementation. Little attention has been paid to post implementation costs and management strategies for data accuracy within longitudinal cancer related databases spanning both ICD-9-CM (I9) and I10. The objective of our study was to quantify financial impact and compare data quality of I10 transition in cancer clinical trials through backward I10 to I9 mapping via two tools. Methods: Cancer related original research articles published in NEJM, Blood and JCO between Jan 1st 2013 and Dec 31st 2013 were reviewed for I9 utilization. Each I9 code was mapped forward to I10 via the Motif web portal tool. The I10 codes were manually reviewed with additional clinically relevant I10 codes added. The final I10 code list was mapped backward to I9 using two tools, the Motif web portal and CMS GEMs. Generated I9 codes were compared to the original I9 list to identify ‘lost’ codes. To assess financial implications of lost data, lost I9 codes were assessed in 2010 Illinois Medicaid data. Results: Of 1,567 original articles reviewed, 7 articles were included and yielded 412 I9 codes which mapped to 1437 I10 codes. Upon manual review, 416 I10 codes were added. Backward mapping by Motif web and CMS GEMs respectively yielded 652 and 551 I9 codes; 16 of the original I9codes were absent from the CMS GEMs list while Motif tool yielded 100% of original codes. Of these 16, 12 codes were present in 2010 Illinois Medicaid data, and accounted for 7,490 patient encounters (0.52%) and $1.1 million (0.35%) of total Medicaid reimbursements. Conclusions: We report significant differences in data quality of I9 generated by backward coding via CMS GEMs and the Motif web tool, with major financial impact. Longitudinal data analyses upto 5 years post ICD-10-CM transition will require backward coding, and comparison of ICD-9-CM codes for accuracy. The bridging methodology and analytic tools presented can be replicated by organizations and clinical researchers to increase data fidelity spanning the upcoming transition.

Abstract:
Salmonella enterica serovar Typhimurium is a facultative intracellular pathogen causing disease in several hosts. These bacteria use two distinct type III secretion systems that inject effector proteins into the host cell for invasion and to alter maturation of the Salmonella-containing vacuole. Members of the Salmonella translocated effector (STE) family contain a conserved N-terminal translocation signal of approximately 140 aa. In this study, the STE family member SifA was examined using deletion strategies. Small deletions (approx. 20 residues long) throughout SifA were sufficient to block its secretion and/or translocation into host cells. Transfection of HeLa cells with a GFP-SifA fusion was previously shown to be sufficient to induce formation of Sif-like tubules resembling structures present in Salmonella-infected cells. The present study showed that both N- and C-terminal domains of SifA are required for this phenotype. Furthermore, both domains could induce aggregation of Lamp1-positive compartments, provided they were coupled to the minimal C-terminal membrane-anchoring motif of SifA. Mutation or deletion of the conserved STE N-terminal WEK(I/M)xxFF translocation motif of SopD2 disrupted its association with Lamp1-positive compartments, implicating these residues in both effector translocation and subcellular localization. Interestingly, one GFP-SifA deletion mutant lacking residues 42–101, but retaining the WEK(I/M)xxFF motif, targeted the Golgi apparatus. In addition, short peptides containing the signature WEK(I/M)xxFF motif derived from the N-termini of Salmonella effectors SopD2, SseJ and SspH2 were sufficient to localize GFP to the Golgi. These studies suggest that Salmonella effectors contain multifunctional motifs or domains that regulate several effector traits, including protein secretion/translocation, localization and subversion of host cell systems. Conditions that perturb the tertiary structure of effectors can influence their localization in host cells by liberating cryptic intracellular targeting motifs.
