Abstract:

Abstract:

Abstract:
Abstract Weibel-Palade bodies (WPBs) are secretory granules that contain von Willebrand factor and P-selectin, molecules that regulate hemostasis and inflammation, respectively. The presence of CD63/LAMP3 in the limiting membrane of WPBs has led to their classification as lysosome-related organelles. Many lysosome-related organelles contain intraluminal vesicles (ILVs) enriched in CD63 that are secreted into the extracellular environment during cell activation to mediate intercellular communication. To date, there are no reports that WPBs contain or release ILVs. By light microscopy and live-cell imaging, we show that CD63 is enriched in microdomains within WPBs. Extracellular antibody recycling studies showed that CD63 in WPB microdomains can originate from the plasma membrane. By cryo-electron tomography of frozen-hydrated endothelial cells, we identify internal vesicles as novel structural features of the WPB lumen. By live-cell fluorescence microscopy, we directly observe the exocytotic release of EGFP-CD63 ILVs as discrete particles from individual WPBs. WPB exocytosis provides a novel route for release of ILVs during endothelial cell stimulation.

Abstract:
Colloidal semiconductor quantum dots (QDs) have gained substantial interest as adjustable, bright and spectrally tunable fluorophores in the past decades. Besides their in-depth analyses in the scientific community, first industrial applications as color conversion and color enrichment materials were implemented. However, stability and processability are essential for their successful use in these and further applications. Methods to embed QDs into oxides or polymers can only partially solve this challenge. Recently, our group introduced the embedding of QDs into ionic salts, which holds several advantages in comparison to polymer or oxide-based counterparts. Both gas permeability and environmental-related degradation processes are negligible, making these composites an almost perfect choice of material. To evaluate this new class of QD-salt mixed crystals, a thorough understanding of the formation procedure and the final composites is needed. The present work is focused on embedding both aqueous-based and oil-based metal-chalcogenide QDs into several ionic salts and the investigations of their optical and chemical properties upon incorporation into the mixed crystals. QDs with well-known, reproducible and high-quality synthetic protocols are chosen as emissive species. CdTe QDs were incorporated into NaCl as host matrix by using the straightforward "classical" method. The resulting mixed crystals of various shapes and beautiful colors preserve the strong luminescence of the incorporated QDs. Besides NaCl, also borax and other salts are used as host matrices. Mercaptopropionic acid stabilized CdTe QDs can easily be co-crystallized with NaCl, while thioglycolic acid as stabilizing agent results in only weakly emitting powder-like mixed crystals. This challenge was overcome by adjusting the pH, the amount of free stabilizer and the type of salt used, demonstrating the reproducible incorporation of highest-quality CdTe QDs capped with thioglycolic acid into NaCl and KCl salt crystals. A disadvantage of the "classical" mixed crystallization procedure was its long duration which prevents a straightforward transfer of the protocol to less stable QD colloids, e.g., initially oil-based, ligand exchanged QDs. To address this challenge, the "Liquid-liquid-diffusion-assisted-crystallization" (LLDC) method is introduced. By applying the LLDC, a substantially accelerated ionic crystallization of the QDs is shown, reducing the crystallization time needed by one order of magnitude. This fast process opens the field of incorporating ligand-exchanged Cd-free QDs into NaCl matrices. To overcome the need for a ligand exchange, the LLDC can also be extended towards a two-step approach. In this modified version, the seed-mediated LLDC provides for the first time the ability to incorporate oil-based QDs directly into ionic matrices without a prior phase transfer. The ionic salts appear to be very tight matrices, ensuring the protection of the QDs from the environment. As one of the main results, these matrices provide extraordinary high photo- and chemical stability. It is further demonstrated with absolute measurements of photoluminescence quantum yields (PL-QYs), that the PL-QYs of aqueous CdTe QDs can be considerably increased upon incorporation into a salt matrix by applying the "classical" crystallization procedure. The achievable PL enhancement factors depend strongly on the PL-QYs of the parent QDs and can be described by the change of the dielectric surrounding as well as the passivation of the QD surface. Studies on CdSe/ZnS in NaCl and CdTe in borax showed a crystal-induced PL-QY increase below the values expected for the respective change of the refractive index, supporting the derived hypothesis of surface defect curing by a CdClx formation as one main factor for PL-QY enhancement. The mixed crystals developed in this work show a high suitability as color conversion materials regarding both their stability and spectral tunability. First proof-of-concept devices provide promising results. However, a combination of the highest figures of merit at the same time is intended. This ambitious goal is reached by implementing a model-experimental feedback approach which ensures the desired high optical performance of the used emitters throughout all intermediate steps. Based on the approach, a white LED combining an incandescent-like warm white with an exceptional high color rendering index and a luminous efficacy of radiation is prepared. It is the first time that a combination of this highly related figures of merit could be reached using QD-based color converters. Furthermore, the idea of embedding QDs into ionic matrices gained considerable interest in the scientific community, resulting in various publications of other research groups based on the results presented here. In summary, the present work provides a profound understanding how this new class of QD-salt mixed crystal composites can be efficiently prepared. Applying the different crystallization methods and by changing the matrix material, mixed crystals emitting from blue to the near infrared region of the electromagnetic spectrum can be fabricated using both Cd-containing and Cd-free QDs. The resulting composites show extraordinary optical properties, combining the QDs spectral tunability with the rigid and tight ionic matrix of the salt. Finally, their utilization as a color conversion material resulted in a high-quality white LED that, for the first time, combines an incandescent-like hue with outstanding optical efficacy and color rendering properties. Besides that, the mixed crystals offer huge potential in other high-quality applications which apply photonic and optoelectronic components.

Abstract:

Abstract:
The Second Opium War concluded in 1860 with Anglo-French forces looting and sacking the Summer Palace at Yuanmingyuan. Commentators such as Victor Hugo delighted that these incidents occurred under the leadership of Lord Elgin, whose father instigated the Parthenon Sculptures controversy. Memoirists and journalists show that the Summer Palace incident was divisive: looting posed a threat to military discipline, and the wanton destruction occasioned a debate over whether Britain was civilized or barbaric. To some, the melancholy victory evoked the Aeneid. Inevitably debates over repatriation of Summer Palace treasures have invoked discussion of the Parthenon Sculptures. Yet commentators like artist Ai WeiWei show that sculptures that the Chinese Communist Party made emblematic of National Humiliation are not really Chinese and were probably not removed by Europeans. Chinese efforts to retrieve the sculptures demonstrate that modern China, like Victorian Britain, reaches to the cultural past for stability amidst bewildering change.

Abstract:
This chapter introduces the reader to Tony Harrison’s audiovisual poetry, composed for and broadcast on British television through the late 1980s and 1990s. Harrison has to date made twelve full documentary film-poems, and one film-poem feature, entitled Prometheus (1998). They all brim with the darker side of European history, current affairs and class politics, and explore what limits of what poetry might do, and how it might do it, in the televisual age. The chapter sets Harrison’s film poetry in its cultural context by paying attention to the poet’s major influences in the experimental medium (John Grierson’s GPO film unit and early Soviet filmmaking), before homing in on his 1992 film-poem The Gaze of the Gorgon as a species of social psychotherapy. The act of gazing at the Medusa-like and thus usually petrifying image of twentieth-century atrocity is offered here not as a ‘dope’ to mankind, but a stimulant towards its cure. The film-poem draws explicitly on Nietzsche’s concept of ‘Dionysiac art’ and Simone Weil’s pacifist notion of ‘force’, and Robert Jay Lifton’s work on trauma and the holocaust. Harrison’s hugely ambitious and uniquely accessible film-poems are currently publically unavailable. Are they the ‘missing link’ in the evolution of contemporary film and video poetry?

Abstract:
This afterword opens with an analysis of two of Gertrud Kolmar’s poems, one prior to the Nazi seizure of power and one prior to Kristallnacht, in which the Jewish and the Animal Questions intersect. It then turns to Gombiner’s lament that the human treatment of animals “is an eternal Treblinka” in I. B. Singer’s “The Letter Writer” (1968) and how it had been preceded by its converse in the experiences of the intended victims of the Holocaust and in memoirs, poems, and autobiographical novels of survivors (e.g., Primo Levi and Uri Zvi Greenberg). The Bestiarium Judaicum and its uses provided them with a language for understanding the dehumanization, the transformation into the suffering, killable Jew-Animal, to which they, “like sheep to the slaughter,” were being subjected. The relationship between dehumanization and objectification is parsed through an examination of Jiri Weil’s depiction of Prague’s deportation site as a “circus.”

This chapter considers the importance of Arendt and Weil's interpretations of the Homeric world, and of The Iliad in particular. This is the case because it is a question to which they both return on a number of occasions, as if the return itself were decisive for the formulation of their own categories. But, above all, it is the case because their interpretations uncover the phenomenon of “concordant dissonance” or of “dissonant concordance.” Homer also evokes another word, which binds them in an affirmative sense. This is the question of the justice, impartiality, or equity of the poet who unites both victors and vanquished in light of the dignity of two adversarial peoples.

This chapter examines one of the largest interventions in computer-based learning currently underway, the One Laptop per Child (OLPC) project. Started in 2005 by people from the MIT Media Lab, this project distributed more than 2.5 million laptops worldwide, over 80 percent of them in Latin America. Drawing on 2010 and 2013 fieldwork investigating a project in Paraguay with 10,000 of OLPC’s “XO” laptops, the chapter explores the ways in which the children who were meant to be the primary beneficiaries interpreted leisure laptop use as “learning.” It also shows that the most captivating uses of the laptops were not “productive” or programming-centric, as OLPC’s developers hoped, but “consumptive” and media-centric, focused on music, videos, and video games. It discusses the learning benefits and drawbacks of this use, as understood by participants and in light of education research, and in light of the broader context of transnational corporations interested in marketing to these children. In the process, it weighs OLPC’s utopian dreams against the interests of the child beneficiaries, concerns of media imperialism, and a potential shift in the meaning of computers.

Abstract:
This chapter argues that Weil's overturning of Arendt's views is not limited to the judgment on Rome. The very Christianity that Arendt situates as the commencement of the drift toward the modern constitutes for Weil both its internal rampart and its principle source of contention simultaneously. Indeed, for Weil Christianity is the spiritual thread that allows modernity to continue advancing in light of its originary inspiration. This does not mean that the two interpretative horizons are openly contradictory. They even coincide at one point precisely in their severe judgment of historical Christianity, which, paradoxically, Weil blames for the de-Christianization of our times. However, their two perspectives diverge once again as Arendt observes that this phenomenon surely occurs because Christianity is lacking in mundane, public figures. In contrast, Weil believes that it is precisely Christianity's spirituality that makes it the unique, authentic “continuation” of Greece.

Abstract:
Existing published works on classic Chinese watercolor-and-ink animation (shuimo donghua) tend to focus on the propaganda-nationalistic aspects of its making. This essay, however, draws attention to the philosophical side of the story-telling images and narratives found principally in the animated works directed by the late animator, Te Wei (1915-2010), who was a founding member of the Shanghai Animation Studio. The animated work, Cowherd’s Flute (1963), is the main focus in this essay. The essay argues that the spiritual yoke of Te Wei’s watercolor-and-ink animation is profoundly Daoist, reflecting a traditional Chinese native philosophy in a subtle technological setting. It advances the view that such generic animation has rekindled a laden strand of Chinese heritage thought by giving life and movement to its aesthetic and philosophical elements. The author also theorizes the concept of the animated self in the light of the historical-political environment in which the animation was made.

Abstract:
This work presents the design, fabrication, and testing of a novel lightweight yet sturdy cage for micro aerial vehicles. Fabricated from a polymer infused 12k carbon-fiber using a smart liquid silicone rubber (LSR) molding process, the cage weighs only 3g and is capable of sustaining impacts at speeds up to 6m/s. We also quantitatively and qualitatively characterize the rigidity and stiffness of the cage using a universal testing machine. This paper subsequently describes the fabrication methodology employed for designing the cage to achieve the high strength-weight ratio. This involves 3D printing, silicon rubber molding, plaster mold rotocasting, wet layup of carbon fiber and finally autoclaving. Finally we demonstrate a 23g autonomous, pico quadrotor capable of sustaining stable flight with collisions with the environment.

Abstract:
The project and construction of the German School of Valencia took place between 1957 and 1961 by the architects Pablo Navarro Alvargonzalez and Julio Trullenque (supervised by Rolf Dieter Weisse and Peter Müller) who fully adapted to the principles of Modern architecture. In addition, typical influences of constructive and architectural practices in the German context are detected due to the collaborative work between technicians of both nationalities (Spanish and German). This is the case of the modern conception of the building as a block arranged on a free floor and its exposed concrete structure, combined with independent enclosure walls, taking special care of the finishing. Furthermore, it is remarkable the detailed development of the structural project, and the widespread use of reinforced concrete, whose pouring and curing processes were rigorously controlled. This represented an innovation in Valencia since constructive quality from postwar period on was quite precarious. These issues bring to light the need to delve into the building and its construction once again, given the significance and early chronology as a participatory work of the architectural modenity of the 20th Century.

Abstract:
The active magnetic regenerator (AMR) is at the heart of the thermo-magnetic Brayton cooling cycle. It consists of a porous matrix heat exchanger whose solid phase is a magnetocaloric material (solid refrigerant) that undergoes a reversible magnetic phase transition when subjected to a changing magnetic field. The cooling capacity of the cycle is proportional to the mass of solid refrigerant, operating frequency, volumetric displacement of the working fluid (generally an aqueous solution) and regenerator effectiveness. AMRs can be modeled via a porous media approach and a model has been developed to simulate the time-dependent fluid flow and heat transfer processes. Gadolinium (Gd) is usually adopted as a reference material for magnetic cooling at near room temperature and, in this study, its magnetic temperature change and physical properties were accounted for using a combination of experimental data and the Weiss-Debye-Sommerfeld theory. In this paper, the influence of the applied magnetic field waveform and of demagnetizing effects on the AMR performance is investigated numerically. An evaluation of the model is also carried out in the light of a comparison against experimental data for a regenerator containing spherical Gd particles.

Abstract:
The culture of human endothelial cells is largely used for vascular research. The possibility of developping long term culture of human endothelial cells (EC) raised the question regarding the identity after several passages. To further investigate this aspect we have cultured human umbilical vein EC until the 12th passage on fibronectin coated dishes supplemented with ECGF. We have studied the EC morphology by light and electron microscopy, the reactivity with 51Cr labelled platelets, and prostacyclin synthesis. Until the 6th passage no major change could be noted, except the occurence of rare large EC and a reduction in the doubling time between 2nd and 5th passage. After the 7th passage up to the 10th EC became more elongated and did not grow in strict monolayer. The number of vacuoles and mitochondria increased as well as the doubling time. After the 12th passage the EC were still viable but proliferated very slowly. The adhesion of radiolabelled platelets dramatically increased (150%) and PGI2 production significantly decreased (6 Keto PGF1α : 1st passage 13±2.5 ng; 6th passage 0.33±0.27 ng/106 EC). In our culture conditions EC kept most of their original characteristics up to the 6th passage but then lost some of them. At any passage EC contained Weibel Palade bodies and von Willebrand factor. We can conclude that after the 7th passage EC in culture are different from the original cells and could possibly represent an in vitro model of EC ageing.

Abstract:
Liver cells were derived from cadaveric organ donors. Pieces of human liver 5 to 50 grams were minced, washed, and incubated in collagenase at 37 degrees C. After washing, the cell suspension was plated into culture vessels that had been briefly pre-treated with an extract derived from human liver. A mixed population of liver cells, including endothelial cells, hepatocytes, and Kupffer cells, attached within hours. At the end of 2 to 3 weeks there developed clusters of densely packed cells of two types. The most numerous cells were initially fusiform but grew as a monolayer even when densely packed. As density increased they assumed a polygonal form; cells with this morphological appearance stained immunocytochemically for von Willebrand factor antigen. They were relatively small and resembled cells derived from human umbilical vein except that the cytoplasm was more filmy in appearance. The second prominent cell type was significantly larger and likewise replicated to form clusters. These large cells sometimes contained multiple nuclei, exhibited a relatively low nuclear to cytoplasmic ratio, and immunocytochemically stained for human fibrinogen. A more distinct nuclear membrane and prominent nucleoli were characteristics of hepatocytes that were useful light microscopically in distinguishing these cells from sinusoidal endothelial cells. Ultrastructurally, endothelial cells were characterized by small size, holes in and among the cells that probably were the in vitro analogue of fenestrae, and numerous Weibel-Palade bodies in the cytoplasm, which otherwise was relatively bland. Hepatocytes, by contrast, had an active appearing cytoplasm containing more organelles. Canaliculi and typical tight junctions formed between adjacent hepatocytes. Levels of vWF and fibrinogen increased in a time dependent manner in media overlying this mixed population of cells. Human factor VIII has not yet been detected in the media overlying these mixed cells derived from human liver, and factor VIII antigen has not yet been demonstrable immunocytochemically in either cell type.

Abstract:
Hobie Cat is one of the most popular sailboat production companies to ever exist, and the Hobie 16 is the most popular boat in the Hobie Cat line-up. The key to Hobie’s success was largely in part to advancements in strong and lightweight materials. This advancement allowed Hobie to achieve record breaking performance in their particular class of boat. The mast of the Hobie 16 is one of the heavier pieces of the craft. The overall mass and center of mass is greatly affected by this component, which begs to question if there may possibly be room for improvement. A sailboat mast is defined as a tall vertical spar, sometimes sectioned, that rises from the keel or deck of a sailing vessel to support the sails and the standing and running rigging. Sailboat masts are long, slender structures. They must be designed to be able to hold the thrust loads that are acting on them from the sails which will differ based on wind conditions. A mast must also be made of a material that will allow it to go through the varying environmental conditions without fatiguing. When designing a sailboat mast a few of the main goals are to have a low center of gravity, be light weight, and also aerodynamically efficient. During this research a Hobie 16 mast design was taken as a baseline design for testing. The current mast on the market weighs between 50–60 lbs. While this is not extremely heavy, the center of gravity is approximately 13.5 feet from the end of the mast. This high center of gravity makes it tough to raise and lower the mast when launching the boat. One important objective in this work is to create a mast that will be easier for the user to raise and lower without sacrificing the structural integrity of the mast. Three masts have been created in solid modeling software, and their performance characteristics have been tested using finite element analysis software ANSYS. Two of the designs are original works, while one is a copy of the original Hobie Cat design. The original design was analyzed, providing baseline data for which to compare the new designs with. In addition to the new designs, all designs have been constructed from three different materials as Aluminum Alloy 6061, Aluminum Alloy 7075 and Acrylic. This allows for comparison of designs and possible new materials. After an in depth analysis of all masts, the recommendation is made that Hobie 16 users would benefit greatly from the use of a tapered mast made of aluminum alloy 7075.
