Abstract:

Abstract:

Abstract:
Abstract Weibel-Palade bodies (WPBs) are secretory granules that contain von Willebrand factor and P-selectin, molecules that regulate hemostasis and inflammation, respectively. The presence of CD63/LAMP3 in the limiting membrane of WPBs has led to their classification as lysosome-related organelles. Many lysosome-related organelles contain intraluminal vesicles (ILVs) enriched in CD63 that are secreted into the extracellular environment during cell activation to mediate intercellular communication. To date, there are no reports that WPBs contain or release ILVs. By light microscopy and live-cell imaging, we show that CD63 is enriched in microdomains within WPBs. Extracellular antibody recycling studies showed that CD63 in WPB microdomains can originate from the plasma membrane. By cryo-electron tomography of frozen-hydrated endothelial cells, we identify internal vesicles as novel structural features of the WPB lumen. By live-cell fluorescence microscopy, we directly observe the exocytotic release of EGFP-CD63 ILVs as discrete particles from individual WPBs. WPB exocytosis provides a novel route for release of ILVs during endothelial cell stimulation.

Abstract:
Abstract The angiopoietins Ang-1 and Ang-2 have been identified as ligands with opposing functions of the receptor tyrosine kinase Tie-2 regulating endothelial cell survival and vascular maturation. Ang-1 acts in a paracrine agonistic manner, whereas Ang-2 appears to act primarily as an autocrine antagonistic regulator. To shed further light on the complexity of autocrine/paracrine agonistic/antagonistic functions of the angiopoietin/Tie-2 system, we have studied Ang-2 synthesis and secretion in different populations of wild-type and retrovirally Ang-2–transduced endothelial cells. Endogenous and overexpressed endothelial cell Ang-2 is expressed in a characteristic granular pattern indicative of a cytoplasmic storage granule. Light and electron microscopic double staining revealed Ang-2 colocalization with von Willebrand factor, identifying Ang-2 as a Weibel-Palade body molecule. Costaining with P-selectin showed that storage of Ang-2 and P-selectin in Weibel-Palade bodies is mutually exclusive. Stored Ang-2 has a long half-life of more than 18 hours and can be secreted within minutes of stimulation (eg, by phorbol 12-myristate 13-acetate [PMA], thrombin, and histamine). Collectively, the identification of Ang-2 as a stored, rapidly available molecule in endothelial cells strongly suggests functions of the angiopoietin/Tie-2 system beyond the established roles during angiogenesis likely to be involved in rapid vascular homeostatic reactions such as inflammation and coagulation.

Abstract:
The past is never dead. It’s not even past“ – wir haben William Faulkner im Kopf). Through The Skin Of Our Teeth“.

Abstract:
ABSTRACT Fast radio bursts (FRBs) are bright, short-duration radio transients with very high brightness temperatures implying highly coherent emission. We suggest that the FRBs are caused by the self-focusing of an electron beam interacting with an ambient plasma right beyond the light cylinder radius of a neutron star. The magnetic field at the light cylinder radius is relatively high that can accommodate both young Crab-like systems and old millisecond pulsars addressing the diverse environments of FRBs. At the first stage, the intense pulsed-beam passing through the background plasma causes instabilities such that the trapped particles in local Buneman-type cavitons saturate the local field. The beam is then radially self-focused due to the circular electric field developed by the two-stream instability that leads to Weibel instability in the transverse direction. Finally, the non-linear saturation of the Weibel instability results in the self-modulational formation of solitons due to plasmoid instability. The resonant solitary waves are the breather-type solitons hosting relativistic particles with self-excited oscillations. The analytical solutions obtained for non-linear dispersion and solitons suggest that, near the current sheets, the relativistic bunches are accelerated/amplified by klystron-like structures due to self-excited oscillations by the induced local electric field. Boosted coherent radio emission propagates through a narrow cone with strong focusing due to radial electric field and magnetic pinching. The non-linear evolution of solitons and the stimulated emission are associated with the Buneman instability and the possibility of the presence of nanosecond shots in FRBs are investigated.

Abstract:
Most prosthetic hands adopt an under-actuated mechanism to achieve dexterous motion performance with a lightweight and anthropomorphic design. Many have been verified in laboratories, and some have already been commercialized. However, a trade-off exists between the dexterity and the light weight of such prosthetic hands. In general, current commercially available prosthetic hands usually consider one aspect at the expense of the other, such as obtaining diversiform hand motions but an increased weight, or achieving lightweight design but with limited motion functions. This study attempts to attain a balance between the two factors, by realizing diversiform hand motions while reducing the weight as far as possible. An anthropomorphic prosthetic hand is proposed with only three servomotors embedded in a human-sized palm, with multiple functions, such as a stable/adaptive grasp and passive hyperextension. The proposed hand can achieve 13 grasp types with over 80% of the grasp motions under the Cutkosky taxonomy, while it weighs only 132.5 g, at less than 36% of the prosthesis weight limitation based on the study of Kay et al.

Abstract:
Immunogold staining of primary cultures of human brain microvessel endothelial cells demonstrated the presence of Factor VIII-related antigen within cytoplasmic vesicles in close association with the rough endoplasmic reticulum and Golgi apparatus. Immunoperoxidase staining, at the light microscopic level, revealed a similar granular, perinuclear staining. The morphology and location of these vesicular profiles indicate that they are part of the trans-Golgi region where terminal processing and short-term storage of Factor VIII-related antigen takes place. Weibel-Palade bodies, specific storage organelles for von Willebrand factor in large vessel endothelium, were not observed in cerebral microvessel endothelium. The release of Factor VIII-related antigen from the cytoplasmic vesicles was influenced by some of the factors known to stimulate or inhibit the regulated pathway of secretion from Weibel-Palade bodies. Thus, stimulation of endothelial cells with calcium ionophore A23187 resulted in almost complete loss of staining, while addition of EGTA to the culture medium led to slight increase of intracellular pools of Factor VIII-related antigen. Pre-incubation of monolayers with interferon-gamma was associated with significant increase in the number of labeled vesicles, suggesting an additional role of this cytokine in the localized immune reaction within the central nervous system.

Abstract:
Given the socio-political context of Latin American countries and their leftist and center-leftist parties that had governed some countries since the beginning of the 2000’s, this essay will present Simone Weil’s anthropology of a rooted people towards social justice guided by a spirituality of supernatural justice. This anthropology supports her criticism of political parties that this essay will use to examine the Brazilian context and its Workers’ Party that governed the country for over thirteen years and ended its tenure after a process of impeachment. Although Simone Weil seems to be pessimistic as to whether multiparty or monoparty political systems are able to promote and maintain a social order able to lead people to find their rootedness, she develops a critical anthropology that allows us to understand when a political party abandons its commitment to develop social policies for the poor and working class. Consequently, the political party assumes an agenda of maintaining power, no matter the means that must be used to achieve this goal.

Abstract:
This paper reports the mineral compositions and geochemical characteristics of the No. 5-2 high-sulfur coal (Taiyuan Formation) from Dongpo Mine, Weibei Coalfield, Shaanxi, Northern China via transmitted and reflected light microscopy, scanning electron microscope equipped with an energy-dispersive X-ray spectrometer (SEM-EDS), X-ray diffraction (XRD), inductively coupled plasma mass spectrometry (ICP-MS), and X-ray fluorescence spectrometry (XRF). We also confirmed the input of intermediate-felsic volcanic ashes into the Taiyuan Formation coals in Dongpo Mine, Weibei Coalfield. The results show that Dongpo coals are enriched in Ga and Li compared to the average values for world hard coals, and they are depleted in Al2O3 compared to Chinese coals. The coal low temperature ash contains kaolinite, illite, quartz, calcite, pyrite, and to a lesser extent, chlorite, plagioclase, dolomite, ankerite, and apatite. The concentration anomalies of Ga and Li in No. 5-2 high-sulfur coal were not caused by the Benxi Formation Bauxite, but by the influence of multiple geological factors. The Middle Proterozoic moyite from the Yinshan Oldland led to the slightly higher Ga and Li contents of the No. 5-2 coal than those in world hard coals. Abstract:
It has now become a fashion among many historians, notably from the West, to denounce Gandhi’s fast and the Poona Pact of 1932 as a great betrayal of the ‘Untouchables’. This view essentially overlooks the constant effort of British imperialism to divide the Indian people into a number of special-interest groups at loggerheads with each other and so weaken the National Movement. This paper weighs the critics’ assertions in the light of the British Government’s own statements and those of the leaders of the Depressed Castes. It is also forgotten that the Poona Pact greatly increased Depressed Castes’ representation; and that no separate electorates have been ever established in Western democracies to avoid majoritarian rule.

Woman and Development in the Maghreb. A Socio-Cultural Approach. The under-development problem of the Third World can not exclusively be attributed to economic backlogging, political crises or supernatural events. The perseverance of old ancestral values, cultural, social and psychological, still remain obstructions to any social evolution in a country. The woman’s role in society in these countries, is restricted to house-keeping and the bearing of children. A situation which weighs heavy on their economies. This article will shed light on the essential elements which hinder active woman participation in development of their economies, in particular in the Maghreb-countries. Discussed is their personal status, women's position in education and in the work-force.

Abstract:
Since the 1990s, the Schwaz-Brixlegg mining district in the Lower Inn Valley, North Tyrol, Austria, features excavations on mostly Late Bronze (LBA) to Early Iron (EIA) Age sites, focusing on the reconstruction of metallurgic activities and of all aspects related to it. This paper reviews the Schwaz-Brixlegg archaeozoological materials and compares them with those from contemporaneous mining (copper and salt) sites on the Eastern Alps, to assess diet and subsistence strategies of the early alpine, geo-resource-centered, communities. The faunal remains from Schwaz-Brixlegg document a change in diet for the Lower Inn Valley area from the LBA to the EIA exemplified by a shift from a pig-based economy to another one based on cattle and occasionally small ruminants. These species were most often brought whole to the sites and only occasionally as meat cuts (in particular, ribs). Age and sex profiles indicate that miners consumed high-quality meat. As is also documented on prehistoric mining sites from the Eastern Alps, butchery marks evidence a standardized slaughtering process car- ried out by professional butchers. At Weißer Schrofen, pig was the main meat provider during the LBA, whereas cattle and sheep/goat were more important as dairy products and wool/skin providers. This pattern changed in the EIA, when sheep became the dominating meat supplier at the site of Bauernzeche. This shift may reflect an adaptation to climate changes, which determined the amount of fodder available for stocks, and/or to the impact of cultural and economic developments taking place during the Final Bronze Age. Variations on the faunal assemblages might also reflect agents such as topography and altitude. All in all, a logistic balance between miners (consumers) and peasants (producers) is revealed although more information is required (e.g. archaeobotany), to shed more light on the major changes recorded in the EIA. Based on gnawing marks from Weißer Schrofen, some of the dogs there must have been large-sized. Although this may constitute an exceptional case for the Bronze Age, similar results were reported from the EBA Brixlegg settlement at Mariahilfbergl. Future research is needed to elucidate the possible functional role of dogs in the context of early mining activities.

AbstractThe radio emission from supernovae (SNe) is nonthermal synchrotron radiation of high brightness temperature, with a “turn-on” delay at longer wavelengths, power-law decline after maximum with index β, and spectral index α asymptotically decreasing with time to a final, optically thin value. Radio supernovae (RSNe) are best described by the Chevalier (1982) “mini-shell” model, with modifications by Weiler et al. (1990). RSNe observations provide a valuable probe of the SN circumstellar environment and constraints on progenitor masses. We present a progress report on a number of recent RSNe, as well as on new behavior from RSNe 1979C and 1980K, and on RSNe as potential distance indicators. In particular, we present updated radio light curves for SN 1993J in M81.

The photoelectric payload could complete the reconnaissance tasks above the target area (including visible and infrared). Because of its light weight, small size, full-featured, high integrated, the constraints of the UAV platform carrying the payload will be reduced a lot, which helps the payload suit for more extensive using occasions. Abstract:
The idea that Husserl’s phenomenology is a kind of reflective philosophy inspired by the Cartesian tradition has become a commonplace in the philosophical literature. Heidegger was one of the first thinkers who criticized the Husserlian emphasis on reflection and transcendental phenomenology. Since then it is easy to find the affirmation that Husserl and Heidegger developed two different, even antagonistic concepts of phenomenology. Here is not the place to continue embracing this discussion. Based on the complex development process of Heidegger’s hermeneutic phenomenology in the course of his early lectures in Freiburg, the present paper weighs up some of Heidegger’s critical remarks regarding the reflective nature of Husserlian phenomenology in the light of important textual evidences ignored not only by Heidegger, but also by a surprising number of specialists in the fields of philosophy, cognitive sciences, and philosophy of mind.

Abstract:
Abstract In endothelial cells, von Willebrand factor (VWF) multimers are packaged into tubules that direct biogenesis of elongated Weibel-Palade bodies (WPBs). WPB release results in unfurling of VWF tubules and assembly into strings that serve to recruit platelets. By confocal microscopy, we have previously observed a rounded morphology of WPBs in blood outgrowth endothelial cells transduced to express factor VIII (FVIII). Using correlative light-electron microscopy and tomography, we now demonstrate that FVIII-containing WPBs have disorganized, short VWF tubules. Whereas normal FVIII and FVIII Y1680F interfered with formation of ultra-large VWF multimers, release of the WPBs resulted in VWF strings of equal length as those from nontransduced blood outgrowth endothelial cells. After release, both WPB-derived FVIII and FVIII Y1680F remained bound to VWF strings, which however had largely lost their ability to recruit platelets. Strings from nontransduced cells, however, were capable of simultaneously recruiting exogenous FVIII and platelets. These findings suggest that the interaction of FVIII with VWF during WPB formation is independent of Y1680, is maintained after WPB release in FVIII-covered VWF strings, and impairs recruitment of platelets. Apparently, intra-cellular and extracellular assembly of FVIII-VWF complex involves distinct mechanisms, which differ with regard to their implications for platelet binding to released VWF strings.

Abstract:
Shape memory alloys (SMAs) are smart materials used in robotics because of its light weight and high force-to-weight ratio. The low energy efficiency, up to 5%, has limited their use for large actuators. However, they have shown advantages in the design of mini-robots because of the limited volume required for the actuation system. The present study reports the design and construction of a mini compliant joint (MCJ) with a 2 degrees of freedom (DOFs) intersecting axis. The MCJ prototype has a 20 mm external diameter surrounding a cavity of 8 mm, weighs 2 g, is 20 mm high and can perform an angle rotation of 30 ∘ in less than 260 ms. It uses SMA NiTi wires in antagonistic configuration and springs to reduce the energy consumption and minimise heat production. The design methods and experimental results of the manufactured prototype are reported and discussed.

Abstract:
SummaryRegulated secretion provides a means by which endothelial cells (EC) can rapidly and selectively alter the microenvironment of individual vascular beds, and modulate the interrelated processes of coagulation, fibrinolysis, and inflammation. The rapid release of high molecular weight multimers of von Willebrand factor (VWF) and surface expression of P-selectin in response to a wide variety of stimuli have been well documented, and are the result of exocytosis of Weibel-Palade bodies (WPB). The regulated release of other EC secretory proteins, including tissue plasminogen activator (tPA), interleukin-8, endothelin-1, and multimerin, has also been described. New light is being shed on how secretory proteins are selectively targeted to storage granules in EC, and on the molecular and cellular events that comprise regulated secretion. Knowledge of the mechanisms of sorting and secretion from EC storage granules may provide basis for new strategies for treating inflammatory and coagulation disorders.

Abstract This essay examines the human-nonhuman proximity emerging from Beckett’s representation of a deconstructed human being and his encounters with nonhuman animals in the “The Expelled,” “The Calmative,” “The End” and “First Love.” With reference to Simone Weil’s categories from The Need for Roots, I show how Beckett’s narrator is lacking physical, psychological, socio-political and philosophical aspects associated with normative human being, which result in a precarious, imprecise identity. In light of this dehumanisation, I close read passages featuring nonhuman animals to argue that while they emphasise the narrator’s marginalisation from human community, they also reveal profound alienation from other animals too. The destabilisation of specific identity, I argue, initiates a reevaluation of the narrator’s place among living beings in general and prefigures the multispecies connectedness advocated in twenty-first century ecocritical reviews of the human-nonhuman divide, such as Donna Haraway’s ‘chthulucene.’

The goal of this contribution is to bring to light some systemic applications of organizational power that occur within the child protection system in Iceland. The specific focus is on the ways in which notifications are made to the child protection system concerning allegations of parental neglect on the part of parents with disabilities and a criticism of some specific mechanisms by which these notifications may contribute to unjust custody deprivation proceedings. We suggest that accusations of bias or prejudice within the child protection system need to be grounded upon the demonstration of concrete mechanisms which illustrate this bias in practice. In this contribution, we chose to focus on the power of notifications and to explore the institutional logic and practices that can result in unjust custody deprivation decisions. A critical analysis of the knowledge production of custody deprivation is required to fully understand these process as systematic applications of power and to recognize the problematic nature of child protection work that weighs heavily against marginalized parents.

Abstract. Costs for maintenance of infrastructure in municipalities with soft soil underground conditions, are estimated to be almost 40 % higher than in others. As a result, these municipalities meet financial problems that cause overdue maintenance. In some cases municipalities are even afraid to be unable to offer a minimum service level in future. In common, traditional practice, roads and sewerage systems have been constructed in trenches that consist of sandy material that replaces the upper meters of the soft soil. Under influence of its weight, this causes accelerated settlements of the construction. A number of alternative constructions have been developed, e.g. using light-weight materials to limit settlement velocity. In order to limit future maintenance costs, improvement of maintenance strategies is desired. Tools have been and will be developed to support municipalities in improving their maintenance strategies and save money by doing that. A model (BALANS) that weighs the attractiveness of alternative solutions under different soil, environmental and economic circumstances, will be presented.

The Bt spore count reaches up to 3.15x109cell/ml and its sporulation rate achieves 99% after incubation for 72 h at 28±2°C. Under aerobic with light condition with a PSB inoculum: FW ratio of 1:3, the proliferated PSB cells weighs 13.5 g/ l after being incubated for 72 h at 28±2°C. The treatments of FW with Bt followed by PSB significantly reduces the chemical oxygen demand, total protein, total nitrogen, total phosphorus, and salinity. Their total removal rates are 91.1%, 78.8%, 89.6%, 76.3% and 21.0%, respectively. The ammonia nitrogen increases by 46.3% .The rapid nutrient removal and cell proliferation may make Bt and PSB applicable in treating FW to reduce environmental pollution and produce valuable bacteria.

AbstractThe comment expands the logic of the critique of the ‘judicialisation’ in the global era and suggests that arguments in support of this development often engage in confirmatory research which weighs the ‘evidence’ in light of our wishes and political projects. The talk about ‘learning’ and ‘dialogue’ cannot sustain this form of judicial paternalism (at best) as an instantiation of emancipation or celebrate it as a victory for law by dispensing with politics. It is just a politics by other means. But in this politics some traditional remedies for insuring the accountability of the ‘rulers’ (or rule-handlers) have been weakened. The comment adds several critical observations about the practices of discourse, law, politics and judging which cannot camouflage the problem of power and its legitimisation. Thus we had better consider also a political alternative which relies on a variety of different institutional solutions where courts have to compete with other institutions without fixed hierarchies and where different sources of legitimacy stand in tension with each other.

Abstract:
Traditional philosophy of religion has tended to focus on the doxastic dimension of religious life, which although a vitally important area of research, has often come at the cost of philosophical engagements with religious practice. Focusing particularly on Christian traditions, this essay offers a sustained reflection on one particular model of embodied Christian practice as presented in the work of Søren Kierkegaard. After a discussion of different notions of practice and perfection, the paper turns to Kierkegaard’s conception of the two churches: the Church Triumphant and the Church Militant. Then, in light of Kierkegaard’s defense of the latter and critique of the former, it is shown that Kierkegaard’s specific account gets appropriated and expanded in Dietrich Bonhoeffer’s account of “costly grace” and “religionless Christianity,” and Simone Weil’s conception of “afflicted love.” Ultimately, it is suggested that these three thinkers jointly present a notion of “militant liturgies” that offers critical and constructive resources for contemporary philosophy of religion.

Additionally, the device must be targeted at effective human-machine interaction. In this paper, we present and test such a device based upon commercial inertial measurement units: it weighs 575 g in total, lasts up to 10.5 h of continual operation, can be donned and doffed in under a minute and costs less than 290 EUR. We assess the attainable performance in terms of error in an online trajectory-tracking task in Virtual Reality using the device through an experiment involving 10 subjects, showing that an average user can attain a precision of 0.66 cm during a static precision task and 6.33 cm while tracking a moving trajectory, when tested in the full peri-personal space of a user.

Background and Objectives: Pain is one of the most common symptoms that weighs on life’s quality and health expenditure. In a reality where increasingly personalized therapies are needed, the early use of genetic tests that highlights the individual response to analgesic drugs could be a valuable help in clinical practice. The aim of this preliminary study is to observe if the therapy set to 5 patients suffering of chronic or acute pain is concordant to the Pharmacogenetic test (PGT) results. Materials and Methods: This preliminary study compares the genetic results of pharmacological effectiveness and tolerability analyzed by the genetic test Neurofarmagen Analgesia®, with the results obtained in clinical practice of 5 patients suffering from acute and chronic pain. Results: Regarding the genetic results of the 5 samples analyzed, 2 reports were found to be completely comparable with the evidences of the clinical practice, while in 3 reports the profile of tolerability and effectiveness were partially discordant. Conclusion: In light of the data not completely overlapping with results observed in clinical practice, further studies would be appropriate in order to acquire more information on the use of Neurofarmagen in routine clinical settings.

Abstract:
Abstract Weibel-Palade bodies (WPBs) are elongated secretory granules of endothelial cells that are packed with tubules composed of von Willebrand factor (VWF), a multimeric protein required for hemostasis. Disruption of tubular packing prevents the orderly secretion of VWF multimers and blocks the subsequent binding of platelets. The cigar-like shape and tubular cross section of WPBs are conserved in all vertebrates, but little is known about how VWF specifies this packing arrangement. Starting from recombinant 82 kDa VWF propeptide (domains D1D2) and 114 kDa disulfide-bonded D’D3 dimer, we now have assembled tubules reversibly in vitro with the same dimensions as VWF tubules in WPBs. Assembly was induced at pH 6.2, reversed at pH 7.4, and required Ca2+. Recombinant D’D3 dimers did not self-associate at pH 7.4 or pH 6.2, with or without Ca2+. Without Ca2+, VWF propeptide did not bind to D’D3 dimers. At pH 7.4, with Ca2+, VWF propeptide formed noncovalent 160 kDa dimers and, when mixed with D’D3 dimers, assembled a 280 kDa complex of two propeptides and one D’D3 dimer as shown by gel filtration chromatography and multi-angle light scattering. Lowering the pH to 6.2 caused the formation of >3 MDa aggregates with the same stoichiometry, which dissociated upon adding EDTA or raising the pH to 7.4. Quick-freeze deep-etch EM showed that the large aggregates are hollow right-handed tubular helices. The iterative helical real space reconstruction method was used to make 3D reconstructions of the tubules at 22 Å resolution from negative stain EM images (Figure, left). Tubules consist of a right-handed helix with axial rise of 26.2 Å and twist of 85.6 degrees per subunit, or 4.2 subunits per 11 nm turn. The dimensions (outside diameter 25 nm, inside diameter 12 nm) are similar to those of tubules in WPBs in thin sections of endothelial cells by transmission EM (Figure, right and its insert). Each subunit contains one D’D3 dimer flanked by two D1D2 propeptides (Figure, center). Each D’D3 dimer makes a total of six contacts with D1D2 domains. Each D1D2 propeptide makes three contacts with D’D3 and just one end-to-end homotypic contact. The spatial arrangement of these building blocks and inter-domain contacts in tubules suggest a model by which decreasing pH along the secretory pathway coordinates the formation of intersubunit disulfide bonds with the tubular packaging of VWF multimers. Within the WPB, Ca2+-dependent and pH-dependent binding of D1D2 to D’D3 domains stabilizes the packing of VWF multimers into tubules, which behave as constrained springs. Upon secretion, the increased pH weakens these constraints and permits the helical tubules to unfurl into flowing blood without tangling. Figure Figure

CFRP (Carbon Fiber Reinforced Plastics) of the advanced composite materials as structural materials for vehicles, has a wide application in light weighs structural materials of airplanes, ships and automobiles because of high strength and stiffness. However, there is a design variable to be considered in practical application of the laminate composite material, that is, this material is vulnerable to transverse impact. This paper aims at evaluating the effects of curvature and oblique impact on the penetration characteristics of composite laminate shells. They are stacked to [02/902]S, [03/903]S and [02/903/0]S, [0/902/0]S and their interlaminar number two and four. Those specimens were prepared to varied curvature radius (R=100, 150, 200mm and ∞). They are cured by heating to the appropriate hardening temperature (130oC) by means of a heater at the vacuum bag of the autoclave. Test specimens were prepared with dimensions 100mm×140mm. When the specimen is subjected to transverse impact by a steel ball, the velocity of the steel ball was measured both before and after impact by determining the time for it to pass two ballistic-screen sensors located a known distance apart. In general, the critical penetration energy increased as the curvature increased, interface decreased and slope angle on the impact surface increased.

Abstract: Residential waste contributes up to 55% of the total volume of urban waste such as in Jakarta. It makes the transport process of residential waste to temprorary landfill site (TPS) that has been existed, less efficient. Transporting waste process with garbage cart is depended on the manpower, while with the garbage truck, the vehicle’s width and the use of fossil fuels are the weaknesses. Thus, the design concept of a waste transport vehicle using electric motor as driving source is made which has the same payload capacity as a garbage cart in general. Garbage payload along with the main components must be strongly supported by the vehicle frame, which incidentally serves as the support vehicle. In this study, discussed the optimization of electric waste transport vehicle with the help of 3-dimensional CAD modelling, then simulated with static loading to obtain the results of the stress, displacement with the maximum value and location, and can assign the payload distribution position. With variable of the beam profile shape and its thickness, then obtained the optimal order forms, which frame weighs as light as possible but still robust in supporting given loads, which is frame with a cylindrical hollow profile with diameter of 60.3 mm and thickness of 4 mm .

Abstract:
The goal of this paper is to develop and experimentally test portable vibration-based electromagnetic energy harvesters which are fit for extracting low frequency kinetic energy. Based on a previous study on fixed vibration-based electromagnetic energy harvesters, three kinds of portable energy harvesters (prototype I, prototype II, and prototype III) are developed and tested. To obtain the related parameters of the energy harvesters, an experimental platform used to measure the vibrational systems electrical power at the resonant frequency and other fixed frequencies is also established. Based on the research work of vibration theory, a low frequency vibration-arm mechanism (prototype III) which is easily in resonance with a walking tempo is developed. Here, a strong magnet fixed to one side of the vibration-arm along with a set of wires placed along the vibrating path will generate electricity. The circular device has a radius of 180 mm, a width of 50 mm, and weighs 200 grams. Because of its light mass, it is easy to carry and put into a backpack. Experimental results reveal that the energy harvester (prototype III) can easily transform kinetic energy into electrical power via the vibration-based electromagnetic system when walking at a normal speed. Consequently, electrical energy reaching 0.25 W is generated from the energy harvester (prototype III) by extracting kinetic energy produced by walking.

Abstract:
This article revisits earlier proposals that language learning is, in essence, the learning of formulaic sequences and their interpretations; that this occurs at all levels of granularity from large to small; and that the language system emerges from the statistical abstraction of patterns latent within and across form and function in language usage. It considers recent research in individual differences, the psycholinguistics of language processing, and longitudinal studies of first (L1) and second (L2) language acquisition. The first section reviews studies of individual differences in phonological short-term memory (PSTM) and working memory (WM) and their correlations with vocabulary and grammar acquisition in L2. The second section summarizes evidence that language processing is sensitive to the statistical properties of formulaic language in terms of frequency and transitional probability. The third section examines the definition of formulas and formulaicity using different statistical metrics. The fourth section evaluates longitudinal research in L1 and L2 into the putative developmental sequence commonly proposed in usage-based approaches, from formula to low-scope pattern to creative construction. The final section weighs the implications of the statistical distributions of formulaicity in usage for developmental sequences of language acquisition. Zipf's law and the “phrasal teddy bear” explain the paradox whereby formulas seed language acquisition and yet learner language is formula-light in comparison to native norms.

Abstract:
Abstract Abstract 325 The liver is generally recognized as the major site of coagulation factor (F)VIII synthesis. However, there is now increasing evidence that FVIII can also be synthesized in specific endothelial cells where it is stored with its natural carrier protein von Willebrand factor (VWF) in the Weibel-Palade bodies (WPBs). WPBs have a typical cigar-shaped appearance that most likely originates from the macromolecular organization of VWF multimers into tubules. The tubular storage of VWF is thought to be essential for orderly secretion of VWF strings during activation of endothelial cells. Recently we have shown that expression of FVIII with VWF changes the WPB morphology to spherical vesicles. This finding suggests alterations in the biochemical properties of stored VWF. We now studied in detail the effect of FVIII co-expression on the VWF molecule using a combination of innovative techniques, including correlative light-electron microscopy (CLEM), and live-cell fluorescence microscopy under flow conditions. Analysis of human blood outgrowth endothelial cells (BOECs) expressing human B-domain deleted FVIII-GFP by CLEM revealed that FVIII containing WPBs were electron-dense, spherical structures. These structures contained disorganized short VWF tubules, which was confirmed in 3D by electron tomography. Double immunogold labelling with VWF and GFP antibodies showed that the spherical FVIII containing structures were always positive for VWF. These observations imply that FVIII blocks the expansion of VWF tubules, possibly by binding to the N-terminal VWF domains. As the N-terminal domains are also implicated in the formation of multimers, we therefore investigated whether FVIII affects VWF multimer size. Indeed, multimer analysis showed that VWF secreted by FVIII-GFP transduced BOECs was multimerized to a lesser extent when compared to VWF secreted by non-transduced BOECs. The combined absence of high molecular weight (HMW) VWF multimers and long VWF tubules made us question whether these cells could still release ultra-large VWF (UL-VWF) strings. UL-VWF strings play a key role in bleeding arrest, as platelets adhere to the released VWF string which ultimately leads to the formation of a platelet plug. We examined the release of UL-VWF strings under shear stress from BOECs expressing FVIII-GFP employing live-cell confocal imaging. This technique allowed us to follow FVIII release during exocytosis of WPBs in real-time as well. When we stimulated FVIII-transduced BOECs with histamine, these cells were equally able to release VWF strings as non-transduced BOECs. Although spherical WPBs lacked long VWF tubules and did not secrete HMW multimers, released VWF strings were of similar length as strings secreted by non-transduced BOECs. Surprisingly, released VWF strings were completely covered with FVIII which remained attached to the strings throughout the whole experiment. Another remarkable observation was that platelet binding to the FVIII-covered VWF strings was almost completely absent. We hypothesize that FVIII either shields the A1 domain for platelet binding or causes a conformational change in the VWF strings that prevents platelets from binding to the strings. Our results demonstrate that FVIII co-trafficking with VWF has a major impact on properties of VWF as it reduces the degree of multimerization, shortens tubules and prevents platelets from adhering to strings. This leads us to the conclusion that the macromolecular structure of VWF is considerably altered when FVIII is present in WPBs. Disclosures: No relevant conflicts of interest to declare.

Abstract:
Magnetic surveying is a widely used and cost-efficient remote sensing method for the detection of subsurface structures at all scales. Traditionally, magnetic surveying has been conducted as ground or airborne surveys, which are cheap and provide large-scale consistent data coverage, respectively. However, ground surveys are often incomplete and slow, whereas airborne surveys suffer from being inflexible, expensive and characterized by a reduced signal-to-noise ratio, due to increased sensor-to-source distance. With the rise of reliable and affordable survey-grade Unmanned Aerial Vehicles (UAVs), and the developments of light-weight magnetometers, the shortcomings of traditional magnetic surveying systems may be bypassed by a carefully designed UAV-borne magnetometer system. Here, we present a study on the development and testing of a light-weight scalar field UAV-integrated magnetometer bird system (the CMAGTRES-S100). The idea behind the CMAGTRES-S100 is the need for a high-speed and flexible system that is easily transported in the field without a car, deployable in most terrain and weather conditions, and provides high-quality scalar data in an operationally efficient manner and at ranges comparable to sub-regional scale helicopter-borne magnetic surveys. We discuss various steps in the development, including (i) choice of sensor based on sensor specifications and sensor stability tests, (ii) design considerations of the bird, (iii) operational efficiency and flexibility and (iv) output data quality. The current CMAGTRES-S100 system weighs ∼5.9 kg (including the UAV) and has an optimal surveying speed of 50 km/h. The system was tested along a complex coastal setting in Brittany, France, targeting mafic dykes and fault contacts with magnetite infill and magnetite nuggets (skarns). A 2.0 × 0.3 km area was mapped with a 10 m line-spacing by four sub-surveys (due to regulatory restrictions). The sub-surveys were completed in 3.5 h, including >2 h for remobilisation and the safety clearance of the area. A noise-level of ±0.02 nT was obtained and several of the key geological structures were mapped by the system.

Abstract:
The bipolar plate is one of the most significant components of a polymer electrolyte membrane (PEM) fuel cell, and contributes substantially to the cost structure and the weight of the stacks. A number of graphite polymer composites with different fabrication techniques have been reported in the literature. Graphite composites show excellent electromechanical properties and chemical stability in acidic environments. Compression and injection molding are the most common manufacturing methods being used for mass production. In this study, a competitive bipolar plate design and fabrication technique is adopted in order to develop a low-cost and light-weight expanded graphite (EG) polymer composite bipolar plate for an air-breathing PEM fuel cell. Cutting molds are designed to cut fuel flow channels on thin expanded graphite (EG) sheets (0.6 mm thickness). Three separate sheets, with the flow channel textures removed, are glued to each other by a commercial conductive epoxy to build a single bipolar plate. The final product has a density of 1.79 g/cm3. A bipolar plate with a 20 cm2 active area weighs only 11.38 g. The manufacturing cost is estimated to be 7.77 $/kWe, and a total manufacturing time of 2 minutes/plate is achieved with lab-scale fabrication. A flexural strength value of 29 MPa is obtained with the three-point bending method. A total resistance of 22.3 milliohms.cm2 is measured for the three-layer bipolar plate. We presume that the suggested design and fabrication process can be a competitive alternate for the small-scale, as well as mass production of bipolar plates.

Abstract:
Introduction: To present our findings of the porcine ocular surface that were obtained with an ultra-compact hand-held microscope that weighs less than 500 g, we examined the corneal epithelial cells with this hand-held microscope. Methods: This device is equipped with an automatic focusing mechanism that enabled us to observe living cells in macro to micro magnifications with a series of operations. The focus is semi-automatically adjusted by the infrared and ultrasonic distance sensor. The instrument has a commercially-available microscope objective lens of 20x or 40x magnification and has a high-resolution 2K Complementary Metal-Oxide-Semiconductor (CMOS) camera. The theoretical spatial resolution is around 300 nm with a higher Numerical Aperture (high-NA) lenses. The widefield reflectance-based imaging system is equipped with three-color visible Light-Emitting Diodes (LEDs) for use in bright environments and an infrared LED for dark environments. Ten normal and two injured porcine corneas were examined with this hand-held microscope. Results: Our observations showed that the corneal and conjunctival epithelial cells could be continuously observed. The epithelial cells of the central cornea, limbus, and conjunctiva were clearly seen. The epithelial cells on the injured corneal surface were also easily and clearly observed. Conclusion: This hand-held microscopic imaging device allows medical health care workers such as ophthalmologists and endoscopists to obtain real-time in vivo optical biopsies without collecting tissues and cells. Our system enables us to observe single cells in the superficial layers without any fluorescein or other dyes.
