Full textENGLISH ABSTRACT: Phenolic compounds are important quality indicators of a red wine, as they can contribute to the colour of a young red wine, colour stability during ageing as well as astringency, bitterness, body and overall mouthfeel properties. Wine composition is commonly influenced by winemaking and viticultural practices. In South Africa it often happens that vines are excessively vigorous, resulting in canopies that are too dense, which in turn could have a negative effect on the quantity and quality of the grapes produced. Viticultural practices such as judicious canopy management and irrigation are designed to control vine vigour and yield, thus improving fruit ripening and colour development. Artificial shading and water deficit have been reported to have an influence on the sensory properties of red wine as well as on the flavonoid composition. These effects are dependent on a number of factors, however, including the season, cultivar, light intensity, and the extent and timing of water deficit. There is limited research on the possible interactive effects of grapevine water deficits and canopy manipulation on grape and wine flavonoid composition in Shiraz, as well as the relationships between berry and wine composition. We thus investigated the effect of canopy reduction in combination with water deficit on the phenolic and colour composition of Shiraz grapes at different levels of ripeness, and in their corresponding wines after alcoholic and malolactic fermentation as well as after six months’ ageing. This study found that it is possible to improve the phenolic composition of grapes and wine by shoot removal, and some of the tendencies in the wines were also observed after the ageing period. If the shoot removal is not performed at a very early stage, sunburn damage can occur and this will result in berries with a lower mass and volume at harvest due to excessive exposure without the berry having adapted to the imposed conditions. Harvesting at different ripeness levels also affected the chemical and phenolic composition of the grapes and resulting wines. The water deficit effect on most phenolic parameters measured in the grapes and wine was not as prominent as that of the canopy manipulation treatment. This study improved our understanding of how an improvement in the canopy microclimate of Shiraz could be reflected in the phenolic composition of wines, along with a potentially important effect of harvesting date. On this basis it may be possible to attain a specific wine style. Harvesting at a ripe stage, for example, could result in the production of wines with higher colour density and astringency, while unripe grapes could result in wines with higher levels of perceivable fresh berry attributes. In particular, canopy reduction could increase the astringency and body of wines made from grapes subjected to water deficit.
AFRIKAANSE OPSOMMING: Fenoliese verbindings is belangrike kwaliteitsparameters van rooiwyn, aangesien dit kan bydra tot die kleur van ‘n rooiwyn, kleurstabiliteit tydens veroudering sowel as frankheid, bitterigheid en mondgevoel. ‘n Wyn se fenoliese samestelling word algemeen bepaal deur wynmaak- en wingerdkundige praktyke. In Suid-Afrika gebeur dit gereeld dat wingerde uitermatig groeikragtig is, wat lei tot te digte lower wat sodoende ‘n negatiewe effek op kwantiteit en kwaliteit van die druiwe wat geproduseer word, het. Wingerdkundige praktyke soos oordeelkundige lowerbestuur en besproeiing is ontwerp om wingerdstokke se groeikrag en opbrengs te beheer, en sodoende vrugrypwording en kleurontwikkeling te verbeter. Kunsmatige beskaduwing en waterstres is gerapporteer om ‘n invloed te hê op die sensoriese eienskappe van rooiwyn sowel as op die flavonoïedsamestelling. Full textENGLISH ABSTRACT: The microclimate environment around the bunch is complex. The spatial distribution of leaves as well as bunch position and morphology impact on the path of direct radiation received by the berries. Canopy microclimate is largely determined by the meteorological conditions (air temperature, solar radiation, wind speed and direction, relative humidity, and precipitation) as well as management practices (trellis/training system, canopy height, vine spacing, row orientation, canopy management practices, irrigation and soil variability and management). The fact that the grapevine continuously responds to its environment, adds to the complexity and dynamic nature of the microclimate that the bunches experience. Field studies involving the effect of the natural bunch environment (i.e. light and temperature conditions) on berry composition, are therefore a challenge, due to the difficulties in quantifying meteorological elements such as temperature and light, which can be hugely variable. There are different sensors available to assess bunch and berry temperature and it can be deployed in different ways within the grapevine canopy, but the difficulty remains in studying the variability that exists within a bunch. This study investigated the value of available sensor technology to measure bunch/berry temperature as well as the spatial and temporal variability on a bunch. Differences in temperature on an intra-berry level were assessed whereas the impact of canopy configuration and bunch orientation on the different sensor levels was also investigated. The contribution of macro- and mesoclimate on bunch and berry temperature was addressed by measuring at two locations (Robertson and Stellenbosch). The potential long term differences in temperature within a bunch with regard to thermal accumulation are discussed. Issues around sensor placement and some technical difficulties related to the sensors are discussed. The results indicated how the effects of mesoclimate were transferred through to the different sensors. A dominating effect of the sea breeze in Stellenbosch was found. Canopy configuration/architecture affected the light regime in the canopy, consequently impacting on bunch temperature variability, specifically in Stellenbosch where a "Ballerina" trellising system was used. Bunch orientation resulted in differences in the temporal variability of bunch/berry temperature and little variability was observed in temperature within the berry. Temperatures of berries situated at the back of the bunch were judged more optimal compared to exposed berries. Direct radiation caused extreme temperatures in exposed berries, which may be detrimental to berry composition and wine quality. This emphasized the importance of the canopy (trellis/training system and management practices) in protecting the bunch from extreme conditions. The large on-bunch spatial variability, observed from measurements with the thermal imager, demonstrated the importance of sensor placement in quantifying the bunch temperature regime; this is also relevant for the future development of berry temperature modelling. Thermal accumulation through the season also illustrated the variability that existed within a bunch, suggesting a potential long term effect on the berry composition. This study proved, in conditions similar to those that may prevail in the South African wine industry, that sensor type and positioning need to be carefully considered in any viticultural/oenological study where bunch microclimate and grape temperatures are assessed.

Die ruimtelike verspreiding van blare sowel as trosposisie en -morfologie het 'n impak op die pad waarlangs direkte straling ontvang word deur die korrels. Lowermikroklimaat word grootliks bepaal deur die meteorologiese kondisies (lugtemperatuur, sonstraling, windspoed en -rigting, relatiewe humiditeit en reënval) sowel as bestuurspraktyke (prieel/opleistelsel, lowerhoogte, wingerdstokspasiëring, ry-oriëntasie, lowerbestuurspraktyke, besproeiing asook grondvariasie en bestuur). Die feit dat die wingerdstok voortdurend reageer op sy omgewing dra by tot die kompleksiteit en dinamiese aard van die mikroklimaat wat die trosse ervaar. Veldstudies gemoeid met die effek van die natuurlike trosomgewing (d.w.s. lig- en temperatuurkondisies) op korrelsamestelling is daarom 'n uitdaging. Die rede hiervoor is dat dit problematies is om meteorologiese elemente soos temperatuur en lig, wat baie veranderlik kan wees, te kwantifiseer. Verskillende sensors is beskikbaar waarmee tros- en korreltemperatuur bepaal kan word en dit kan op verskillende wyses binne die wingerdstoklower aangewend word. Die bestudering van die variasie wat bestaan binne 'n tros is egter steeds problematies. Hierdie studie het die waarde ondersoek van die beskikbare sensortegnologie vir die meting van tros/korreltemperatuur en die ruimtelike en tydsvariasie op 'n tros. Verskille in temperatuur op 'n intra-korrelvlak is bepaal terwyl die impak van lowerkonfigurasie en trosoriëntasie op die verskillende sensorvlakke ook ondersoek is. Die bydrae van makro- en mesoklimaat tot tros- en korreltemperatuur is ondersoek deur te meet by twee verskillende liggings (Robertson en Stellenbosch). Die potensiële langtermyn verskille in temperatuur binne-in 'n tros met betrekking tot temperatuur akkumulasie word bespreek. Kwessies rakende sensorplasing en sommige tegniese probleme wat verband hou met sensors word bespreek. Die resultate het aangedui hoedat die effekte van mesoklimaat oorgedra is na die verskillende sensors. 'n Dominerende effek van die seebries is waargeneem in Stellenbosch. Temperature van korrels wat voorkom aan die agterkant van die tros is beoordeel as meer optimaal vergeleke met blootgestelde korrels. Direkte straling het uiterste temperature in blootgestelde korrels veroorsaak wat nadelig kan wees vir korrelsamestelling en wynkwaliteit. Hierdeur is die belang van die lower (prieel/opleistelsel en bestuurspraktyke) om die tros te beskerm teen uiterste kondisies beklemtoon. Die groot ruimtelike variasie op 'n tros, soos waargeneem in metings met die termiese kamera, het die belangrikheid van sensorplasing in die kwantifisering van die trostemperatuur regime beklemtoon. Dit is ook relevant vir die toekomstige ontwikkeling van korreltemperatuur modellering. Termiese akkumulasie gedurende die seisoen is ook geïllustreer deur die veranderlikheid wat voorkom binne 'n tros, wat dui op 'n potensiële langtermyn effek op die korrelsamestelling. Hierdie studie het bewys, in kondisies wat algemeen voorkom in die Suid-Afrikaanse wynbedryf, dat sensortipe en -plasing sorgvuldig in ag geneem moet word in enige wingerd/wynkundige studie waar trosmikroklimaat en druiftemperature bepaal word.

Full textENGLISH ABSTRACT: In an attempt to reduce the pH of juice and wine, different fertiliser applications and canopy management practices were evaluated in a field trial. Fertiliser treatments consisted of no, CaS04, Ca(OH)2, and MgS04 fertilisation. Canopy management was as follows: suckering (leaving only two shoots per bearer), tipping, vertical shoot positioning and removal of lateral shoots and yellow leaves in the bunch zone (Canopy 1); suckering (leaving three shoots per bearer), vertical shoot positioning as well as topping (Canopy 2); vertical shoot positioning and topping (Canopy 3). The field trial was conducted in the Paardeberg region on the farms Meerlus and Kersfontein. The vineyard at Meerlus was Cabernet franc/R99 with a high canopy density and a good root distribution, established on a sandy loam soil of granite origin, with a low subsoil pH and a high K content. The vineyard at Kersfontein was Cabemet Sauvignon/101-14 Mgt with a lower canopy density and a less extensive root distribution, also established on a sandy loam soil of granite origin, but with a low top- and subsoil pH and a higher K content. Fertilisation had no significant influence on the K content of juice and wine. Fertilisation with Ca and Mg reduced the pH of juice significantly in the case of Meerlus. In contrast, Mg fertilisation increased the pH of juice significantly at Kersfontein. Lastly, fertilisation had no significant effect on the pH of the wine. The K content of the juice at Meerlus was significantly reduced by Canopy 3 in comparison with Canopy 1 and 2. However, in contrast with Canopy 1 and 3, Canopy 2 significantly increased the pH of juice at Meerlus. The K content of the juice at Kersfontein was significantly reduced by Canopy 1, compared to Canopy 2 and 3, with no significant effect on the pH of the juice. Canopy management had no significant effect on wine pH. It appears to be possible to reduce the pH of juice in the case of Cabernet franc/99R, situated on granite soils, by means of Ca and Mg fertilisation. Because Ca(OH)2 and MgS04 fertilisation increased the maturity of Cabernet Sauvignon/101-14 Mgt grapes, it was impossible to evaluate the effect of fertilisation on the pH of juice at Kersfontein. The general phenomenon that a high canopy density results in a high pH of juice and wine, was not observed in this field trial. The possible reasons for this were the low canopy density of the control plots, as well as the difference in maturity between canopy management treatments.
AFRIKAANSE OPSOMMING: In 'n poging om die pH van sap en wyn te verlaag, is die effek van verskillende bemesting- en lowerbestuursfaktore in 'n veldproef geêvalueer. Bemesting het bestaan uit geen, CaS04, Ca(OH)2 en MgS04 toediennings. Lowerbestuur was: suier tot twee lote per draer, tip, vertikale lootposisionering, verwydering van sylote en geel blare in trossone (Lower 1); suier tot drie lote per draer, top en vertikale lootposisionering (Lower 2); top en vertikale lootposisionering met geen suier nie (Lower 3). Die veldproef is op twee plase nl. Meerlus en Kersfontein, in die Paardeberg omgewing uitgevoer. Die wingerd by Meerlus was Cabernet franc/R99 met 'n hoë lowerdigtheid en goeie wortelverspreiding, wat op 'n sandleemgrond van graniet oorsprong met In lae ondergrond-pH en hoë K-inhoud gevestig is. Die wingerd by Kersfontein het bestaan uit Cabernet Sauvignon/101-14 Mgt met 'n laer lowerdigtheid en swakker wortelverspreiding, wat op In sandleemgrond van graniet oorsprong met In deurgaans lae grond-pH en baie hoë K-inhoud gevestig is. Bemesting het geen betekenisvolle invloed op die K-inhoud van sap en wyn gehad nie. By Meerlus het Ca- en Mg-bemesting egter die pH van sap betekenisvol verlaag. In teenstelling hiermee het Mg-bemesting die sap-ph by Kersfontein betekenisvol verhoog. Bemesting het verder geen betekenisvolle invloed op die pH van wyn gehad nie. Lower 3 het die K-inhoud van sap by Meerlus betekenisvol verlaag in vergelyking met Lower 1 en Lower 2. By Kersfontein was die K-inhoud van sap by Lower 1 betekenisvollaer as by Lower 2 en Lower 3. Teenoor Lower 1 en Lower 3 het Lower 2 'n betekenisvol hoër sap-pH by Meerlus tot gevolg gehad. Lowerbehandelings het egter geen betekenisvolle invloed op die pH van wyn gehad nie. Die moontlikheid bestaan dus om die sap-pH van Cabernet franc/R99 op granietgrond betekenisvol m.b.v. Ca- en Mg-bemesting te verlaag. Aangesien Ca(OH)2- en MgS04- bemesting die rypheidsgraad van Cabernet Sauvignon/101-14 Mgt se druiwe betekenisvol verhoog het, was dit onmoontlik om die effek van bemesting op sap-pH by Kersfontein te evalueer. Die algemene verskynsel dat 'n hoë lowerdigtheid tot hoë pH's in sap en wyn lei, is nie in die proef ondervind nie. Die lae lowerdigtheid van die kontrole persele en die verskil in rypheidsgraad tussen lowerbehandelings kan moontlik as rede hiervoor aangevoer word.

Full textENGLISH ABSTRACT: South African red wine is often acknowledged world wide as being full bodied and deep in colour. This is often the result of high temperatures that is experienced during the important growth stages of grapes especially post véraison. In the Robertson area in South Africa however, temperatures often exceeds the range for optimal anthocyanin development during these growth stages. The distinction between grapes being technologically ripe and being ripe on a phenolic level is also accepted as an important determining factor for the perfect time to pick grapes. In co-operative wineries such as Robertson Winery (RW) where grapes are delivered from a large area and different producers, it is difficult to individualise grape blocks when it comes to ripeness level in terms of sugar or phenolic ripeness. In most circumstances a generalised set of parameters for deeming grapes ripe or acceptable for delivery is the best substitute. The levels of these parameters are based on research literature that is available for the area as well as data collected through years of maintaining the vineyards of that area. The grape parameters that are currently being used by RW for ripeness and quality are pH, titratable acidity (TA) and sugar level. In recent years RW in conjunction with the Department of Viticulture and Oenology, Stellenbosch University, decided to investigate more parameters to determine the quality of grapes at the time of harvest. Most importantly for the grape growers this quality is connected to a price point and therefore compensation. Two important quality parameters of red wine are the red colour and mouth feel of wine. Anthocyanin and tannins are respectively connected to these two quality attributes and are both widely accepted as quality indicators. Wine with high anthocyanin and tannin content often originates from grapes with a high colour and phenolic profile. The existence of a correlation between grape and wine anthocyanin and tannin content is therefore the basis of attempting to use these parameters in the grape to predict end wine’s colour and phenolic quantity. Determination of anthocyanin and tannin content of grapes has already become part of some private owned wineries’ standard set of determinations. However, sample preparations, extractions and consumables needed are all factors that need to be reduced to make the measurement and therefore the use of these parameters more viable in a co-operative cellar laboratory, where large volumes of grapes are received during harvest. The first objective of this work was to determine the levels of anthocyanin and tannin in red grapes from different vineyard blocks from the producers of RW from three successive vintages. This would give insight as to what can be seen as a low and high anthocyanin and tannin content for grapes received at the cellar. For this purpose, blocks of the most important red wine cultivars for RW was selected and analysed for these compounds. The ranges and average levels of anthocyanin and tannin content were determined using measurement techniques that could be used by any winery. The average mono flavanol and total colour level of the grapes were found to be lower than those often reported in literature, with total grape flavanols being higher. However, a wide range of values for these compounds were found that correlated with those found in other studies. The possible reasons for differences in levels of occurrence of these compounds were discussed and mostly pertain to differences in cultivar, micro climatic and season. The second objective was to determine the correlation between levels of colour and phenolic compounds in grapes and their corresponding wines. Such correlations will form the foundation for the use of phenolic content to predict the colour and phenolic potential of the wine and possibly wine quality as well. When the grape and wine colour and phenolic data were correlated for all seasons and cultivars inclusive it was found that grape and wine colour showed better correlations than for instance total phenols and tannins. This was especially true for total colour pigments in red grapes, measured with HPLC, when correlated with certain spectrophotometric analysis of wine colour. Cultivar and season as well as the synergism between the two were further investigated for its role in affecting correlations. When these relationships were further differentiated by season and by cultivar the resulting correlations varied. This work contributed a great deal of information to support the use of grape colour and phenolic compounds for the prediction of end wine colour and phenolic composition. The third objective was to investigate near infrared spectroscopy (FT-NIR) as a viable option to rapidly measured anthocyanins, tannins and total phenolics in red grapes. If proven successfully, this could be employed by a large cellar such as RW. FT-NIR has been used with success on grape extracts and in this instance the focus was to establish a calibration on the grape homogenate itself. Preliminary results showed that FT-NIR could be applied for the use of determination of anthocyanin and tannin levels in red grapes originating from RW. The prediction of total phenols was not found to be as accurate, but this could also be due to the reference method that was used. This work brought some interesting, practical information not only of importance for RW, but all wineries that are concerned with improving the basis on which grape quality is determined. The use of aerial data mapping for indicating areas regarding important grape colour and phenolic parameters was used in this study and is a very visual way of showing the distribution of certain ripeness parameters over a large area. Correlations between the grape and wines of such a large amount of red grape blocks for a specific area have not also been reported in South Africa before. The use of FT-NIR to determine anthocyanins and tannin concentrations in grape homogenates is also novel for its use in South African wineries. This work may assist grape and wine producers as well as analysts on the phenolic and colour profile of grapes and wines from RW.
AFRIKAANSE OPSOMMING: Suid-Afrikaanse rooiwyn word wêreld-wyd geken aan ‘n dieprooi kleur en vol struktuur. Die grootste rede vir hierdie verskynsel is hoë temperature wat ervaar word tydens rypwording en veral na véraison. In die Robertson wynstreek is temperature egter tydens rypwording dikwels vêr bo dit was as optimaal vir antosianien ontwikkeling beskou word. Die gepaste tyd om druiwe te pluk word nie net gedryf deur die tegnologiese rypheidsvlak nie, maar ook deur fenoliese rypheid. In ‘n koöperatiewe kelder omgewing soos Robertson Wynkelder (RW) word ‘n hoë lading druiwe elke dag ontvant vanaf verskillende produsente oor ‘n breë streek. Dit maak dit moeilik om te bepaal watter druiwe werklik beide tegnologies en fenolies ryp is. Die beste manier om hiervoor te vergoed is om ‘n standaard te stel vir ‘n reeks voorafbepaalde parameters. Die vlakke van die gekose parameters is, word bepaal deur navorsinguitsette sowel as die geskiedkunde data wat ingesamel is vanaf elkeen van die bepaalde blokke. Die parameters wat tans in gebruik is by RW om oesdatum en kwaliteit by inname te bepaal is pH, titreerbare suur (TA) en suiker vlak. Die tekortkoming hier is dat kwaliteit van druiwe beswaarlik met slegs hierdie informasie kan bepaal word, maar dat dit die betaling van die produsent by aflewering wesenlik kan beïnvloed. Dit het RW genoop om in samewerking met die Departement van Wingerd en Wynkunde, Universiteit van Stellenbosch nog parameters te ondersoek wat hierdie rypheid- en kwaliteitsbepaling by inname sou kon versterk. Twee belangrike faktore wat kwaliteit van rooiwyn bepaal is die kleur en struktuur. Antosianiene en tanniene is onderskeidelik verantwoordelik vir hierdie kwaliteits eienskappe van wyn. Wyn wat bestempel word as hoog in kleur en tannien inhoud word dikwels verbind met druiwe wat hoog is in hierdie faktore. Die moontlike korrelasie tussen die antosianien en tannien inhoud van druiwe en die wyn wat daarvan berei word is dus die basis waarop die potensiële toepassing van hierdie parameters berus. Die bepaling van antosianien en tannien vlakke word reeds in sommige laboratoriums gedoen. Die monster voorbereidings tyd, ekstraksies, toerusting en verbruikbare items nodig om hierdie tipe analieses te doen is egter hoog. Die analiese van hierdie komponente is meer lewensvatbaar in groot laboratoriums (soos in ‘n koöperatiewe kelder) waar groter volume druiwe ingeneem word gedurende parstyd. Die eerste doelwit van hierdie studie was om te bepaal teen watter vlakke antosianiene en tanniene in druiwe voorkom, spesifiek van die Robertson area. Die het behels ‘n wye verskeidenheid van blokke, verspreid oor die hele streek wat oor 3 seisoene gemonitor is in terme van veral kleur en tanniene maar ook ander belangrike parameters. Die idee hier is om insig te kry rakende watter vlakke bestempel kan word as laag en hoog in terme van antosianien en tanniene vir die Robertson streek. Daarvoor is slegs die mees aangeplantste rooi kultivars gebruik. Die verspreiding en gemiddelde vlakke waarteen antosianien en tanniene voorkom was bepaal deur gebruik te maak van metodes wat as relatief algemeen in laboratoria gebruik word. Die gemiddelde mono-flavonoïed en totale kleur pigment inhoud van die druiwe was laer as van die vlakke wat in die literatuur beskikbaar is, met totale flavanole wat hoër was. Die wyer verspreiding van die waardes het egter beter gekorreleer met die waardes soos beskryf in die literatuur. Die moontlike redes vir die verskillende vlakke word in die studie bespreek en word waarskynlik bepaal deur verskille in kultivar, mikro-klimaat en seisoen. Die tweede doelwit was om te bepaal of daar ‘n korrelasie te vinde is tussen die kleur en tannien inhoud van die druiwe en ooreenstemmende wyne. Sulke tipe korrelasies sal die basis vorm om antosianien en tannien inhoud van wyn reeds in die druiwe fase te kan voorspel. Nadat die ingesamelde druif en wyn data as ‘n geheel beskou was, was dit sigbaar dat die wynkleur parameters beter korrelasies bied as meeste tannien en totale fenole. Dit was veral waar in die geval van totale kleur pigmente soos gemeet met die HPLC teenoor die wynkleur parameters gemeet met spektrofotometriese metodes. Verdere ondersoeke in terme van die impak wat die kultivar en seisoenale kan hê het tot variërende korrelasies gelei.. Hierdie werk het ‘n groot bydrae gelewer om voorspellings van wyn kleur en fenoliese inhoud reeds met sukses vanaf die druif te bepaal. Derdens het die werk fourier transformasie naby infrarooi skandering (FT-NIR) ondersoek as ‘n lewensvatbare metode vir die bepaling van antosianien, tannien en totale fenoliese inhoud van druiwe en wyn. FT-NIR word reeds oor ‘n wye reeks wyne en druiwe ekstraksiemonsters toegepas en die doelwit hier was om druiwe homogenaat as matriks te kalibreer. Voorlopige resultate het bevind dat antosianien en tannien vlakke in druiwe van RW gemeet kan word met die FT-NIR, maar dat die kalibrasie vir totale fenole nog verbeter kan word. Hierdie werk het ‘n wye reeks interessante en prakties bruikbare informasie na vore gebring wat van onskatbare belang is vir RW en ander kelders wat besorgd is oor die verbetering van algemene druifkwaliteit. Geografiese kaarte wat belangrike druifkleur en fenoliese parameters aandui is in hierdie studie gebruik en wys hoe data visueel voorgestel kan word om die geheelindruk van gekose parameters oor ‘n groot area te vergelyk. Korrelasies tussen druiwe en wyn van so ‘n groot hoeveelheid druiwe blokke is nog nooit voorheen in Suid-Afrika getoon nie. Dieselfde geld vir die gebruik van FT-NIR vir die meet van kleur en fenoliese parameters in druiwe homogenate. Hierdie werk kan druiwe- en wynproduseerders sowel as analiste assisteer in terme van die kleur en fenoliese profiel van druiwe en wyn van RW.

Full textENGLISH ABSTRACT: In recent years South African wines have been under the spotlight due to references in the international wine media to a distinctive ‘burnt rubber’ character. Many winemakers and wine experts argued that the peculiar character could be ascribed to winemaking errors linked to mismanaged fermentation. An alternative possible source of the taint was identified in the coal tar creosote used as a wood preservative in vineyard trellis systems. South African regulations allow for the use of creosoted utility poles in agricultural land, but in Europe and USA their usage has been highly regulated and even banned for certain applications. Contamination of foodstuff by polycyclic aromatic hydrocarbons (PAHs) is one of the major motives for the banning of creosote in Europe and USA. Some of the compounds in the creosote mixture display very strong odour characteristics and for this reason it became the focus of attention for the present study. The overall aim of this study was to determine if vines trellised with creosoted posts could accumulate or absorb the various malodorous compounds deriving from the wood treatment onto the grape berries. These compounds could then be extracted from the grape berries into the wine during alcoholic fermentation, creating quality and sensory problems. Chapter 2 of this thesis gives an overview of the extensive literature that deals with methods of analysis of PAHs and malodorous phenols using both Gas Chromatography (GC), as well as High Performance Liquid Chromatography (HPLC). New methods for sample preparation, as well as analysis of PAHs using HPLC-Diode Array Detector (DAD), were developed and the results reported in Chapter 3. It was demonstrated that Sauvignon blanc experimental wines contained only chrysene at very low levels. The concentrations of chrysene found in the experimental wines are within the prescribed parameters as established by The Commission of the European Communities. Since no other PAH compounds were found in the samples analysed, it was concluded that the experimental wines were safe for human consumption and complied with EU regulations. The effect of vineyard posts on the sensorial characteristics of wine is discussed in Chapter 4. Creosoted poles were found to be responsible for an off-flavour described as ‘burnt rubber’ and ‘tarry’ in Merlot wines produced from grapes grown in close proximity to the posts. Following some of the reported findings, new guidelines have been introduced in the Integrated Production of Wine certification, which advise against the use of creosoted poles for vineyard trellising. This preliminary but important guideline will bring the South African wine industry a step closer to the fulfilling the obligations for food safety as required by the legislation of our major export partners. Future investigations are recommended to completely understand and evaluate the cumulative effect of creosoted posts in a fully trellised vineyard.
AFRIKAANSE OPSOMMING: Oor die afgelope paar jaar is Suid-Afrikaanse wyne onder die soeklig geplaas as gevolg van verwysings in die internasionale wynmedia na ‘n duidelike ‘gebrande rubber’-karakter. Baie wynmakers en wyndeskundiges het aangedui dat hierdie besonderse karakter toegeskryf kan word aan wynbereidingsfoute wat verband hou met gisting wat wanbestuur is. ‘n Alternatiewe moontlike oorsprong van die smaak is geïdentifiseer in die koolteer wat as ‘n houtpreserveermiddel in wingerdopleistelsels gebruik word. Suid-Afrikaanse regulasies maak voorsiening vir die gebruik van kreosoteerde nutspale in landbougrond, hoewel hulle gebruik in Europa en die VSA hoogs gereguleerd en in sommige gevalle selfs verbied is. Die besmetting van kossoorte deur polisikliese aromatiese koolwaterstowwe (polycyclic aromatic hydrocarbons (PAHs)) is een van die vernaamste redes vir die verbanning van kreosoot in Europa en die VSA. Sommige van die verbindings in die kreosootmengsel het baie sterk geurkenmerke en daarom is dit die fokus van die huidige studie. Die oorhoofse doelwit van hierdie studie was om te bepaal of wingerde wat op kreosoteerde pale opgelei is, die verskillende onwelriekende verbindings afkomstig van die houtbehandeling in die druiwekorrels kan akkumuleer of absorbeer. Hoofstuk 2 van hierdie tesis verskaf ‘n oorsig van die breedvoerige literatuur wat handel oor metodes om PAH’s en onwelriekende fenole met behulp van beide gaschromatografie (GC) en hoëdrukvloeistofchromatografie (HPLC) te analiseer. Nuwe metodes is ontwikkel om monsters voor te berei en om PAH’s met behulp van ‘n HPLC-diode array detector (DAD) te analiseer. Die resultate word in Hoofstuk 3 gerapporteer. Daar is aangetoon dat die eksperimentele Sauvignon blanc-wyne chriseen teen baie lae vlakke bevat het. Die konsentrasies van chriseen wat in die eksperimentele wyne gevind is, is binne die voorgeskrewe parameters van die Kommissie van die Europese Gemeenskap. Aangesien daar nie ander PAHverbindings in die geanaliseerde monsters gevind kon word nie, is daar tot die gevolgtrekking gekom dat die eksperimentele wyne veilig is vir menslike verbruik en aan die EG-regulasies voldoen. Die effek van wingerdpale op die sensoriese kenmerke van wyn word in Hoofstuk 4 bespreek. Kreosoteerde pale is gevind wat verantwoordelik is vir ‘n wangeur in Merlot-wyne afkomstig van druiwe wat naby die pale gegroei het en wat as ‘gebrande rubber’ en ‘teeragtig’ beskryf is. Op grond van sommige van die gerapporteerde bevindings, is nuwe riglyne ingesluit in die sertifisering vir die Geïntegreerde Produksie van Wyn, wat aanbeveel dat kreosoteerde pale nie vir wingerdoplei gebruik word nie. Hierdie voorlopige, maar belangrike riglyn sal die Suid-Afrikaanse wynbedryf al beter in staat stel om te voldoen aan die voedselveiligheid regulasies wat vereis word deur die wetgewing van ons belangrikste uitvoervennote. Toekomstige ondersoeke moet aangewend word om die kumulatiewe effek van kreosoteerde pale in volledig opgeleide wingerde ten volle te verstaan en te evalueer.

Full textENGLISH ABSTRACT: Grapevine performance and wine quality are influenced by various factors, two of the most important being the availability and quality of irrigation water. In relatively dry countries such as South Africa the conservation and effective use of water is of utmost importance. Expected increases in temperature and decreases in rainfall in the future due to climate change impacts highlights the importance of water conservation. This inspired investigations into possible alternative irrigation water sources and therefore the possibility of vineyard irrigation using winery wastewater is of utmost importance for the sustainability of the wine industry. Winery wastewater contains higher concentrations of certain elements other than water generally used for vineyard irrigation, the most important differences being Na and K levels. Furthermore, winery wastewater contains larger populations of microorganisms such as yeasts, lactic acid bacteria and acetic acid bacteria, typical associated with wine production. If irrigation using winery wastewater affects the uptake of certain elements or alters grapevine water status, it may affect grapevine growth, juice and wine composition. Furthermore, if juice and wine composition is affected wine composition and sensorial quality may be affected. Cabernet Sauvignon/99R grapevines, growing in a sandy soil in the Breede River Valley, were subjected to eight irrigation treatments using augmented winery wastewater in addition to irrigation using raw river water as control. The study was carried out during the 2010/11 and 2011/12 seasons. The various wastewater irrigation treatments were made up by augmenting winery wastewater with raw river water to obtain a target chemical oxygen demand (COD) concentration. In this study, the level of COD in the irrigation water is a direct indication of water quality, the two being indirectly proportional. The eight wastewater irrigation treatments ranged from 100 mg/L COD up to 3000 mg/L COD. The first objective of the study was to determine the effect of irrigation using augmented winery wastewater on grapevine response, with regards to vegetative growth, berry development and berry composition. The various wastewater irrigation treatments did not affect grapevine vegetative growth or reproductive growth, including yield, throughout berry development up to harvest. Berry sugar accumulation and evolution in acid concentrations were also not affected. An increase in berry juice pH was observed with an increase in the level of COD in the augmented winery wastewater only in the second season. The amount of elements, ions and heavy metals in juice was not affected by wastewater irrigation, indicating that there was no absorption by the grapevines. Berry skin thickness, colour and phenolic content as well as yield and its associated components were not affected by irrigation using augmented winery wastewater. The second objective of the study was to determine the effect of irrigation using augmented winery wastewater on wine microbial and chemical composition, fermentation performance and wine sensorial characteristics. The natural yeast and bacteria flora of juice was not affected by the various wastewater irrigation treatments. In addition, the ability of the inoculated yeast and lactic acid bacteria strains to conduct their respective fermentation processes were not affected. With the exception of total titratable acidity (TTA) and pH, irrigation using augmented winery wastewater did not affect wine chemical composition with regards to basic wine parameters as well as colour, phenolic and tannin composition. Similar to juice, phosphorus and selected ions were not affected. None of the measured wine sensorial characteristics were affected by irrigation using augmented winery wastewater. The third objective of the study was to investigate the effect of direct contact between berries and winery wastewater on wine sensorial characteristics. The study focussed on the transference of off-flavours from the wastewater into the wine and the occurrence of off-flavours as a response to contact with winery wastewater. Wine colour and general sensory wine descriptives were not affected by direct contact with winery wastewater. The presence of a winery wastewater-like off-odour and volatile acidity was, however, more detectable in wines made from berries that were in contact with the most concentrated wastewater. Therefore, it may be possible for off-odours to be transferred from the winery wastewater into the wines, or that off-odours are formed as a direct or indirect result of contact with winery wastewater. Under the given conditions, results obtained in this two seasons of the study suggest that irrigation using augmented winery wastewater does not affect grapevine performance or wine quality substantially. The major impact that was observed was an increase in wine pH and a decreasing trend in TTA. Both these parameters could be rectified by simply adding acid to the wines. Therefore, irrigation using augmented winery wastewater may be considered as a possible future alternative source for vineyard irrigation. It is, however, important to remember that some of the effects of wastewater irrigation may be cumulative and could possibly arise only after several years. Furthermore, different field conditions and cultivars may respond differently.
AFRIKAANSE OPSOMMING: Wingerd prestasie en wyngehalte word deur verskeie faktore beïnvloed waarvan twee van die belangrikste die beskikbaarheid en gehalte van besproeiingswater is. In relatiewe droë lande soos Suid Afrika is waterbesparing en die effektiewe benutting van water hulpbronne van uiterste belang. Die verwagte toename in temperatuur en afname in reënval in die toekoms as gevolg van klimaatsveranderinge plaasdieklem op op die belangrikheid van waterbesparing. Dit het navorsing om moontlike alternatiewe vorme van besproeiingswater te ontdek geïnspireer. Na aanleiding van hierdie faktore word daar toenemend gefokus op navorsing oor die moontlikheid om kelder afvalwater as alternatiewe bron van besproeiings water vir wingerde te benut. Kelder afvalwater bevat hoër konsentrasie van sekere elemente as water wat onder normale omstandighede gebruik word vir die besproeiing van wingerde, die belangrikste verskille was die vlakke van Na en K. Benewens die hoër konsentrasie van sekere elemente bevat kelder afvalwater ook groot populasies van mikroörganismes soos giste, melksuurbakterieë en asynsuurbakterieë, tipies geassosieerd met wynbereiding. Indien besproeiing met kelder afvalwater die opname van sekere elemente of die plant water status beϊnvloed, mag wingerd groei, sap en wyn samestelling beϊnvloed word. Daar benewens, indien die mikrobiese samestelling van die sap en wyn beϊnvloed word sal die samestelling en sensoriese gehalte van die wyn moontlik beϊnvloed word. Cabernet Sauvignon/99R wingerde, geleë in sanderige grond in die Breede Rivier Vallei, is onderwerp aan besproeiing met agt verskillende konsentrasies van verdunde kelder afvalwater, bykomend tot besproeiing met onbehandelde rivier water wat as kontrole gedien het. Hierdie studie is uitgevoer gedurende die 2010/11 en 2011/12 seisoene. Die teiken besproeiings konsentrasies is verkry deur kelder afvalwater met onbehandelde rivier water te verdun tot ‘n sekere chemiese suurstofbehoefte (CSB) konsentrasie bereik is. Die CSB is in hierdie studie ‘n direkte aanduiding van watergehalte, die twee was indirek eweredig tot mekaar. Die agt CSB konsentrasies waarteen die afvalwater besproei is wissel tussen 100 mg/L CSB en 3000 mg/L CSB. Die eerste doelwit van die studie was om te bepaal wat die effek van besproeiing met verdunde kelder afvalwater op wingerdprestasie, met spesifieke verwysing na vegetatiewe groei, korrelontwikkeling en korrelsamestelling is. Wingerd vegetatiewe en reproduktiewe groei, insluitende opbrengs, is op geen stadium tydens korrelontwikkeling tot en met oes beïnvloed nie. Die laai van suikers gedurende rypwording, sowel as verskuiwings in suurkonsentrasie, is nie deur besproeiing met kelder afvalwater beïnvloed nie. In die tweede seisoen is ‘n toename in sap pH waargeneem soos die CSB konsentrasie van die besproeiings water toegeneem het. Die element, ioon en swaar metaal samestelling van sap was nie beïnvloed deur besproeiing met afvalwater nie wat aandui dat daar geen opname was deur die wingerd nie. Die dikte, kleur en fenoliese samestelling van druifdoppe is ook nie beïnvloed nie. Die tweede doelwit van die studie was om te bepaal wat die effek van besproeiing met verdunde kelder afvalwater op wyn mikrobiese en chemiese samestelling, fermentasie effektiwiteit en wyn sensoriese eienskappe is. Die verskeie afvalwater besproeiings behandelings het geen effek op die natuurlike gis of bakterieë flora van die sap gehad nie. Die vermoë van die geïnokuleerde gis en melksuurbakterieë om hul afsonderlike fermentasie prosesse te voltooi is ook nie beïnvloed nie. Met die uitsondering van totale titreerbare suur (TTS) en pH, is die chemiese samestelling van wyne met betrekking tot basiese wyn parameters, kleur, fenole en tanniene nie beïnvloed nie. Soortgelyk aan sap is wyn fosfor en geselekteerde ioon samestelling nie geaffekteer nie. Die sensoriese karakteristieke was eenders vir wyne van alle behandelings. Die derde doelwit van die studie was om te bepaal wat die effek wat direkte kontak van kelder afvalwater met druiwekorrels op wyn sensoriese eienskappe het. Hierdie studie het gefokus op die oordrag van afgeure vanaf kelder afvalwater na die wyne sowel as die voorkoms van afgeure as ‘n reaksie op kontak met kelder afvalwater. Wyn kleur en algemene sensoriese eienskappe is nie geaffekteer deur kontak tussen druiwe en kelder afvalwater nie. Kelder afvalwater-geassosieerde afgeure en vlugtige suur was meer duidelik waarneembaar in wyne wat gemaak is van druiwe wat in kontak was met die meer gekonsentreerde afvalwater. Dit mag dus moontlik wees dat afgeure vanaf kelder afvalwater oorgedra word na wyne, of dat sekere afgeure gevorm word as ‘n direkte of indirekte reaksie op kontak met kelder afvalwater. Onder die gegewe toestande oor die twee jaar studie periode het resultate getoon dat besproeiing met verdunde kelder afvalwater nie wingerdprestasie en wyn gehalte aansienlik beïnvloed nie. Die grootste impak wat afvalwater besproeiing gehad het, was om ‘n toename in wyn pH en ‘n tendens tot afname in TTS te veroorsaak. Deur eenvoudig suur by die wyn te voeg kan albei hierdie probleme reg gestel word. Op grond van hierdie bevindings kan besproeiing met verdunde kelder afvalwater moontlik as toekomstige bron vir addisionele wingerdbesproeiing dien. Dit is egter belangrik om te onthou dat die effekte van besproeiing met kelder afvalwater mootlik kumulatief kan wees en dat probleme moontlik eers na etlike jare na vore kan kom. Ander kultivars en veldkondisies mag ook lei tot ander resultate.

Full textENGLISH ABSTRACT: Highly valued Sauvignon blanc wines, with the distinctive cultivar-typical aromas, reminiscent of grassy, green pepper or asparagus-like, are produced in some South African regions. Quite often, however, neutral and sulphur-like, low quality Sauvignon blanc wines are produced and this phenomenon is of great concern to wine producers and consumers, and affects our competition on overseas markets, negatively. The aim of this study was to investigate the effect of region, ascorbic acid/S02 treatments and yeast strain on Sauvignon blanc wine aroma and quality. Wines were produced from grapes obtained from the warmer Robertson and the relatively cooler Stellenbosch regions (1998 season). The juices were treated with different combinations of ascorbic acid/S02 treatments [commercially available ascorbic acid/meta preparate, S02 (control), pure ascorbic acid/S02] and Saccharomyces cerevisiae yeast strains (Vin 13, VL3C, NT 116). The wines were analysed for esters, higher alcohols, monoterpenes and 2-methoxy-3-isobutylpyrazine (ibMP). The wines were also sensorially evaluated for wine aroma intensities (fruity/ester, sulphur-like, grassy/green pepper) and overall quality. Additionally, the synergistic action of ibMP and the sulphur-containing component, 4-mercapto-4-methylpentan-2- one (MMP), considered to be the most important impact components of Sauvignon blanc, was studied. The two components were added, separately and in combinations at increasing concentrations, to different media. The nuances perceived, varied from dusty, grassy to green pepper for ibMP and from guava, sulphur-like to cat urine or "conifer" for MMP. Significant differences were observed between the wines treated with the different combinations of ascorbic acid/S02 treatments and fermented with different yeast strains, irrespective of region. The highest quality, cultivar-typical Sauvignon blanc wines were produced from the pure ascorbic acid/S02 treatment in combination with yeast strains Vin 13 and NT 116. This coincided with high ester and low higher alcohol concentrations, which did not overpower the typical Sauvignon blanc character. The treatments had, in some cases, a significant effect on monoterpene levels, but it was concluded that these differences were not big enough to affect wine quality. Levels of ibMP were too low and could not be reliably measured. Low quality wines, with prominent, undesirable sulphur-like aromas, were produced from juices, treated with the commercially available ascorbic acid/meta preparate and the French yeast strain, VL3C. Techniques, followed to identify the aroma components causing the sulphur-like offflavours, as MMP or as other sulphur-containing components, were gas chromatography/mass spectrometry, solid phase microextraction and sniffing. However, these tests were not successful and studies to identify these off-flavours should be continued. It was succeeded in this study to produce Sauvignon blanc wines without the undesirable, sulphur-like aromas. Although this investigation showed that a newly developed, commercially available ascorbic acid/meta preparate did not yield any sulphur-like off-flavours, quite often Sauvignon blanc wines with such off-flavours are still produced. Further research is needed to clarify this phenomenon.
AFRIKAANSE OPSOMMING: Hoë kwaliteit Sauvignon blanc wyne, met kenmerkende kultivar-tipiese, gras-, groenrissie- of aspersie-agtige aromas, word in sekere streke van Suid Afrika geproduseer. Die gereelde produksie van neutrale, en swawelagtige lae kwaliteit Sauvignon blanc wyne, wek egter nie net groot kommer by wynprodusente en verbruikers nie, maar het ook 'n negatiewe impak op kompetisie met oorsese markte. Die doel van hierdie studie was dus om die effek van streek, askorbiensuur/Sóbehandelings en gisras op Sauvignon blanc wynaroma en kwaliteit vas te stel. Wyne is berei met druiwe wat verkry is van die warmer Roberston en relatief koeler Stellenbosch streke (1998 seisoen). Verskillende kombinasies askorbiensuur/So, behandelings [kommersieel-beskikbare askorbiensuur/meta preparaat, S02 (kontrole), suiwer askorbiensuur/So-l en Saccharomyces cerevisiae gisrasse (Vin 13, VL3C, NT 116) is gebruik tydens die wynbereidingsproses. Spesifieke ester-, hoër alkohol-, monoterpeen- en 2-metoksi-3-isobutiel metoksipirasienkonsentrasies (ibMP) is in die wyne bepaal. Full textENGLISH ABSTRACT: Technological advances made over the past century have had a major impact on traditional wineries. Software solutions for management issues are widely available and give rise to the prospect of computerized decision support systems assisting in various aspects of managing a winery. The most popular applications seem to be concerned with supply chain management or harvest scheduling. Such projects are under way all over the globe and great success has been achieved to this e ect. However, prior to this study no such project has been considered in South Africa. The phrase active cellar scheduling problem refers to the assignment of grape batches to processors inside the cellar where bottlenecks often occur during the busy harvesting period. The phrase harvest scheduling problem, on the other hand, refers to selecting the best possible dates to harvest the respective vineyard blocks in order to preserve grape quality. A mixed integer programming model for the active cellar scheduling problem is derived in this thesis, but proves to be too time consuming to solve exactly via the branch-and-bound method. A meta-heuristic tabu search approach is therefore designed to solve the problem approximately instead. When applied to a small, ctitious cellar, it is found that the tabu search method often solves the problem optimally. The computer processing time associated with the tabu search approach also constitutes a signi cant (often thousand-fold) improvement over that of the branch-and-bound approach for realistically sized problem instances. A generic tabu search is also designed to solve the over-arching harvest scheduling problem for a general winery. This schedule is found by referring to the smaller tabu search of the active cellar scheduling in order to verify the impact that harvesting moves have on activities in the cellar. One harvesting schedule is considered a better schedule than another when it has a lower harvest evaluation score, determined by the placement of the vineyard blocks in the harvesting schedule. The harvest evaluation score takes into account the combination of vineyard blocks selected for harvesting on the same day (and their e ect on the active cellar) as well as the ripeness and quality of the grapes. Both tabu searches are nally included in a exible, computerized decision support system, called VinDSS. This system is found to produce good harvesting schedules when compared to an actual ve day schedule during the 2009 harvesting period at Wamakersvallei, a winery serving as case study for this thesis.

Sagteware-oplossings wat besluitsteun tot bestuursaangeleenthede bied, is algemeen beskikbaar en het gelei tot die rekenaarmatige implementering van besluitsteunstelsels vir wynkelders. Dit blyk dat die mees popul^ere besluitsteuntoepassings in die wynindustrie te make het met besluite rakende van voorsieningskettings en oes-skedulering. Sulke besluitsteunprojekte is w^ereldwyd onderweg en het alreeds groot sukses behaal. Daar is egter tot dusver geen so 'n projek in Suid-Afrika onderneem nie. Die frase aktiewe kelderskeduleringsprobleem verwys na die toekenning van druifvragte aan masjiene binne die kelder waar bottelnekke algemeen tydens die besige parstydperk voorkom. Die frase oes-skeduleringsprobleem, daarenteen, verwys na die seleksie van bes moontlike oesdatums vir elk van die wingerdblokke om sodoende druifkwaliteit te verseker. 'n Gemengde heeltallige programmeringsmodel is vir die aktiewe kelderskeduleringsprobleem ontwikkel, maar die rekenaaroplossingstyd van hierdie benadering blyk te lank te wees om die probleem eksak deur middel van 'n vertak-en-begrens metode op te los. So 'n oes-skedule word gevind deur na die kleiner tabu soektog vir die aktiewe kelderskedulering te verwys om sodoende die e ekte van veranderinge in die oesskedule op die prosesse binne die aktiewe kelder na te speur. Een oes-skedule word beter as 'n ander skedule beskou wanneer dit met 'n beter oes-evalueringswaarde gepaard gaan, soos deur die plasing van die wingerdblokke in die skedule bepaal. Die oes-evalueringswaarde neem die moontlike kombinasies van wingerblokke wat op dieselfde dag geoes word, in ag (en ook die e ek wat dit op aktiwiteite in die kelder het), asook die rypheid en kwaliteit van die druiwe. Beide tabu soektogte word in 'n plooibare, rekenaar-ge mplementeerde besluitsteunstelsel, bekend as VinDSS, ingesluit. Full textENGLISH ABSTRACT: Due to increased consumer demand for products labelled by origin, and the requirement that these labels are a guarantee of both quality and product character, there is an increasing global focus on delimitation of denominations of origin. The integrity of denominations of origin and their defensibility can be ensured through the use of terroirs as a basis for delimitation. The aims of this study were to establish the dominant environmental criteria that affect the viticultural behaviour and wine character of two important cultivars (Cabernet Sauvignon and Sauvignon blanc) in the Stellenbosch Wine of Origin District, to use an appropriate methodology to identify viticultural terroirs in this district based on these criteria and with the use of a geographic information system, and finally to use these viticultural terroirs to identify denominations of origin within the same area. A terroir can be defined as a grouping of homogenous environmental units, or natural terroir units, based on the typicality of the products obtained. Identification and characterisation of terroirs depends on knowledge of environmental parameters, the functioning of the grapevine and the characteristics of the final product. Field studies, resulting in point data, are necessary to investigate the functioning of the grapevine but in order for this information to be of use within zoning studies it must be placed in a spatial context. As a first phase in data acquisition, the Stellenbosch Wine of Origin District was characterised and natural terroir units were identified using existing digital data and a geographic information system. A natural terroir unit (NTU) can be defined as a unit of land that is characterised by relatively homogenous topography, climate, geological substrate and soil. A total of 1389 NTUs were identified in the Stellenbosch Wine of Origin District (84 537 ha). The identified NTUs were homogenous with respect to terrain morphological unit, altitude, aspect and soil type. Each of the identified units was further described with respect to the extent of the expected sea breeze effect and, for certain of the soil types, the associated parent material. As a second phase of data acquisition, a network of plots of Sauvignon blanc and Cabernet Sauvignon were delimited in commercial vineyards in proximity to weather stations and their viticultural and oenological response monitored for a period of seven years. Regression tree analyses were performed on the complete data set and the relative importance of the environmental and management related variables determined for each dependent variable. Excepting for scion clone, which had a high relative importance for bunch mass of Sauvignon blanc and yield to pruning mass index of Cabernet Sauvignon, no other non-environmental variable included in the analyses appeared to have a strong effect on grapevine performance and wine character. The performance of Cabernet Sauvignon was affected by the potassium content of the subsoil and the climate of the season. The performance of Sauvignon blanc appeared to be related to soil texture, wind exposure and temperature, both during the green berry growth stage and the month prior to ripening. From the results presented, it appears that environmental parameters have an overriding effect on the performance of both Cabernet Sauvignon and Sauvignon blanc, but that these two cultivars react differently to environmental stimuli. A knowledge-driven model used the rules generated in the regression tree analyses to directly classify natural terroir units with respect to expected response of Cabernet Sauvignon and Sauvignon blanc in the Stellenbosch Wine of Origin District. The natural terroir units were thus grouped into terroir units that were homogenous with respect to predicted viticultural and oenological response for each cultivar. The use of representative sites to determine the response of the grapevine to its environment is time consuming and costly and limits terroir studies to research related investigations. Vineyard managers were therefore surveyed with respect to the functioning of established Sauvignon blanc vineyards in the Stellenbosch Wine of Origin District in an attempt to obtain the necessary data. Comparison of the data generated with these questionnaires to measured data in commercial vineyards suggested that the vineyard managers were able to characterise the performance of vineyards with respect to vigour, signs of drought stress and yield. Each vineyard was mapped and the responses were linked to modelled environmental variables. Classification and regression trees were used to construct decision trees, which could be applied to environmental data in a geographic information system to determine viticultural terroirs for production of Sauvignon blanc. These terroirs, although fewer, were comparable to those generated using field data. Data gathered during terroir studies, and the identified viticultural terroirs for Cabernet Sauvignon and Sauvignon blanc, were used to revisit the boundaries of the Stellenbosch Wine of Origin District and the Simonsberg-Stellenbosch ward. Modifications were proposed based on expected wine characteristics. Boundaries for two new wards in the Helderberg basin were proposed. It was also possible to identify vineyards within a ward for the production of terroir specific wines.
AFRIKAANSE OPSOMMING: Toenemende verbruikersaanvraag vereis produkte waarvan die etikette nie net die oorsprong aandui nie, maar ook kan dien as ‘n waarborg vir die produk se kwaliteit en kenmerkendheid. Hierdie tendens verklaar die toenemende wereldwye fokus op afgebakende areas van oorsprong. Die integriteit van die afgebakende areas van oorsprong sowel as hul verdedigbaarheid kan gewaarborg word deur terroirs as basis vir afbakening te gebruik. Klassifikasie en regressiebome is gebruik om besluitnemingsmodelle saam te stel wat toegepas kon word op omgewingsdata in ‘n geografiese inligtingstelsel om terroirs vir die produksie van Sauvignon blanc te bepaal. Full textENGLISH ABSTRACT: Cabernet Sauvignon is the most planted red cultivar in South Africa and the second most planted red cultivar in the Olifants River region. The cultivar is prone to vigorous growth with low yields. Excessive irrigation could accentuate these cultivar characteristics. Considering the foregoing, the aim of the study was to describe how Cabernet Sauvignon will react to climate, soil type (texture) and irrigation within the Lower Olifants River wine region to enable growers to make the right decisions regarding long term as well as short term cultivation practices. This study is part of a project carried out by the ARC Infruitec-Nietvoorbij at Stellenbosch to determine the effects of soil type and atmospheric conditions on yield and wine quality of Cabernet Sauvignon in different grape growing regions of South Africa. Similar studies are being carried out in the Orange River, Stellenbosch and Swartland regions of South Africa. The Lower Olifants River region could be divided into three climatic regions. Furthermore, two climatic regions is evident regarding the formation of grape wine colour and aromas. Proximity to the Atlantic Ocean would play an important role in a cultivar establishment policy. The variation in stem water potential (ΨS) could be related to soil water status expressed in terms of matric potential (ΨM). In the case of sandy soils, ΨS decreased substantially more as the ΨM decreased compared to the sandy loam soils. The reason could be that the unsaturated hydraulic conductivity in sandy soils decreased more rapidly as the ΨM decreased compared to the heavier soils. Thus could explain why the grapevines in the sandy soils experienced more water stress than the ones in the sandy loam soils at a given ΨM. Climate had a strong influence on grapevine water status with grapevines nearer to the ocean experiencing less water stress compared to the ones further inland. This was especially true for grapevines in the sandy soils. Vegetative growth and yield of grapevines in the sandy soils were more sensitive to water deficits compared to the ones in the sandy loam soils. For deficit irrigated grapevines in the sandy soils, vegetative growth and yield decreased by ca. 30% when ca. 55% less water was applied from flowering to harvest. Yield reduction were ca. 15% with no or very little influence on vegetative growth with ca. 80% reduction in water applied from flowering to harvest for grapevines in the sandy loam soils. The influence of soil texture on wine quality and style were evident under intensive irrigation as well as over different climatic regions. Overall sensorial potential wine quality of grapevines in sandy soils tended to be higher compared to the ones in the sandy loam soils. Deficit irrigation tended to increase wine colour intensity, irrespective of soil texture. Furthermore, deficit irrigation in sandy loam soils tended to increase wine fullness and the berry characteristics of the wine. Berry characteristics of wines from the sandy soils tended to be higher compared to the ones from the sandy loam soils. Too severe water deficits in sandy soils could be detrimental to wine quality. Climate tended to have an influence on wine style of grapevines in the sandy soils with wines produced further away from the ocean tended to have higher berry characteristics. Irrigation management could be a powerful tool to manipulate the grapevine in sandy soils. For grapevines the sandy loam soils in addition to regulated deficit irrigation, additional canopy management practices could be needed to improve wine quality.
AFRIKAANSE OPSOMMING: Cabernet Sauvignon is die mees aangeplante rooidruif kultivar in die Suid-Afrikaanse wynbedryf. In die Olifantsriver streek is dit naas Shiraz, die tweede mees aangeplante rooidruif kultivar. Cabernet Sauvignon is bekend as ‘n groeikragtige skaamdraer. Indien oorbesproei word, kan hierdie potensiële nadelige eienskappe nog meer na vore tree. Die doel van die studie is om die invloed van die klimaat, grond en besproeiing op Cabernet Sauvignon se vegetatiewe groei, produksie en wyngehalte in die Benede Olifantsrivier streek te bepaal. Hierdie inligting kan produsente help om ingeligte kortsowel as langtermyn besluite te maak rakende die verbouing van Cabernet Sauvignon. Hierdie studie vorm deel van ‘n breër studie in die Suid-Afrikaanse wynbedryf, gedryf deur die Landbou Navorsingsraad (LNR) Infruitec-Nietvoorbij, Stellenbosch om die invloed van atmosferiese toestande en grond op die produksie en wyngehalte van Cabernet Sauvignon te bepaal. Soortgelyke projekte word uitgevoer in die Oranjerivier, Stellenbosch en Swartland wynstreke. Die Benede Olifantsrivier streek kan verdeel word in drie klimaatstreke op grond van temperatuurdata. In terme van die ontwikkeling van druifkleur en aromas, kan die streek verdeel word in twee klimaatstreke. Die afstand vanaf die Atlantiese Oseaan kan ‘n belangrike rol speel in die ontwikkeling van ‘n kultivarriglynplan vir die streek. Grondwaterstatus, uitgedruk as die matrikspotensiaal (ΨM), kan aanleiding gee tot variasie in middag blaarwaterpotensiaal (ΨS) lesings. Die ΨS van die sand gronde verlaag vinniger soos die ΨM verlaag invergelyke met die sandleem gronde. Dit kan moontlik wees as gevolg van die verskil in die grond onversadigde hidroliese konduktiwiteit. Sand gronde se hidroliese konduktiwiteit verlaag vinniger soos die ΨM verlaag, invergelyke met sandleem gronde. Dit verklaar waarom wingerde in sand gronde by dieselfde ΨM, meer waterspanning ondervind as wingerde in sandleem gronde. Klimaat het ‘n invloed op die waterstatus van die wingerdstok. Wingerde nader aan die see het minder waterspanning ondervind invergelyke met wingerde wat verder in die binneland geleë is. Dit was veral die geval met wingerde in die sand gronde. Vegetatiewe groei en produksie van wingerde in die sand gronde is meer sensitief vir waterspanning as wingerde in die sandleem gronde. Tekortbesproeiing in die sand gronde het die groei asook produksie met ongeveer 30% verlaag deur ongeveer 55% minder water toe te dien vanaf blom tot oes. In teenstelling daarmee is die produksie van wingerde in die sandleem gronde met ongeveer 15% verlaag met geen tot baie min verlaging in die groeikrag. Ongeveer 80% minder water is toegedien vanaf blom tot oes. Grondtekstuur kan wyngehalte en -styl beïnvloed ten spyte van intensiewe besproeiing en klimaatsverskille. Sensoriese potensiële wyngehalte van wingerde in die sand gronde was beter invergelyke met dié van die sandleem gronde. Die wyne vanaf die sand gronde het ook geneig om oor meer bessie intensiteit te beskik as wyne vanaf die sandleem gronde. Tekortbesproeiing neig om die wynkleur intensiteit te verhoog, ongeag van grondtekstuur. Tekortbesproeiing in die sandleem gronde kan ook die volheid van die wyne verbeter, asook die bessie intensiteit van die wyn verhoog. Te hoë waterspanning in die sand gronde kan wyngehalte nadelig beïnvloed. Klimaat kan ook die wynstyl vanaf sand gronde beïnvloed met wyne verder vanaf die see wat oor meer bessie intensiteit beskik as wyne nader aan die see. Beheerde tekortbesproeiing kan as ‘n kragtige hulpmiddel gebruik word om wingerde in die sand gronde te manipuleer. Vir wingerde in die sandleem gronde, addisioneel tot beheerde tekortbesproeiing en normale loofbestuurspraktyke, kan ekstra loofbestuurspraktyke bv. die verwydering van sylootlote, dalk nodig wees om wyngehalte te verbeter.

In order to avoid economic losses due to spoilage, the wine industry is seeking feasible techniques to possibly reduce the levels of S02 in wine. The purpose of this study was, therefore to determine the efficacy of ultraviolet radiation (UV)-C (254 nm) as an alternative technology to inactivate microorganisms in white and red grape juices and wines.

With increasing population and industrial production growth, there is an increasing amount of waste coming from these transformations. The focus of this study will be on the co-products of the winemaking process, known as grape marc. Every year 60 million tons of grape are produced, 80% of which are used in the winemaking process, generating tons of underutilized organic co-products. This is characterized by its high concentrations of phenolic compounds which can be reused as antioxidants. Thus, the main goal of this project was to present the influence of different extraction methods on chemical composition and antioxidant activity of these extracts/fractions. For this, the extractions were performed by microwave, ultrasound and liquid-liquid, with different polarities solvents (ethanol, water, methanol, hexane, chloroform and ethyl acetate). Moreover, the extract was fractionated with the aid of an open column using silica as stationary phase and different polarities solvents as mobile phase. The chemical characterization was performed by High Performance Liquid Chromatography with UV-Vis detector (CLAE) and High Resolution Mass Spectrometry (HRMS). Among the main compounds identified are phenolics such as rutin, gallic acid, catechin, and some carboxylates such as linoleic acid and stearic acid. In the extracted extractions we can identify that the ethanol solvent, in all extraction methods, obtained better results regarding the qualification and quantification of chemical compounds in analysis with HPLC-UV. The evaluation of antioxidant capacity was carried out using the 2,2-diphenyl-1-picrylhydrazyl (DPPH •) method. The data obtained by microwave extraction showed a higher percentage of scanning (2,44%±36,64), when the solvent used was ethanol. In the extraction by sonography, the highest values 2,07%±74,22 were observed using ethyl acetate. In the liquid-liquid extraction the highest percentage of sweep (0,61±33,62) was observed for the solvent ethyl acetate. In general, the extracts obtained by extraction with microwaves presented higher levels of phenolic compounds (0,04±13,18 a 6,02mg±107,03 GAE/100g). The results obtained in this project indicate that this co-product can be better utilized for the application as a source of functional ingredients, as food supplements or in the cosmetic industry, because they contain important compounds (sic).

This paper presents a case study for the use of a Hazard and Critical Control Points System (HACCP) in the wine industry, considering all elaboration stages as well as cares and necessary evaluations. HACCP is a method that provides technical evaluation of products and regarding to winemaking, it aims to detect possible contamination points and their control, taking into account all agents involved on the elaboration process. Therefore its implementation follows rules based upon documented research. Tracing the system foundations in details ensures its effective application. These guidelines follow regulated rules, already used and accepted worldwide. The outcome of such systematization is a higher quality product, with food safety accepted by the consumer. This generates increased profitability, loyalty towards the brand and places the company within the market of wines accepted and appreciated for their valid provenance and safety certification. On this study it was made a general systematization of the HACCP for wineries, taking into account all stages of winemaking, from harvesting and transporting the grapes up to the final product storage. After the general analysis, Vinícola Larentis, placed on Vale dos Vinhedos, State of Rio Grande do Sul, Brazil, was chosen to carry out the investigation. The first step was accomplished in the winery, determining the compliance degree with the prerequisites envisaged by the HACCP. The second step was to plan the system for Merlot wine vinification. With the analysis divided into areas, it was observed that hazards may occur in almost all of them and eventually, corrective actions were proposed. Therefore, the HACCP was considered and discussed, as it lessens the hazards of the winemaking process, decreases production costs and consequently improves the final product quality.

Full textThe assessments of grape and wine quality are complex issues and the wine industry needs more objective analysis of grape and wine quality. The standard quality assessment protocol for grading grapes at most wine cellars in South Africa is based on viticultural practices and the determination of chemical parameters such as ºBrix, pH and titratable acidity (TA). Grape juice indices calculated by formulae such as ºBrix/pH, TA/pH, ºBrix/TA, ºBrix x (pH)2 have been used in the past but these approaches have had limited success. It was shown that the total anthocyanins and total phenolics of red grapes correlate with wine quality and provide additional objective measures of grape quality. Most methods for the quantification of total anthocyanins and total phenolics are complex and time consuming and therefore not easily implemented in the routine laboratory environment. Fourier transform infrared spectroscopy (FT-IR) is widely used in South African laboratories for the routine quantification of wine and grape parameters but the commercial calibration models supplied for the quantification of grape total anthocyanins and phenolics are not satisfactory. The focus of this study was to develop new FT-IR calibration models for the quantification of total anthocyanins and phenolics of grapes and to use the generated data during a preliminary evaluation of the implementation of these parameters as part of the grape quality control protocol at a commercial winery in South Africa. The potential of Fourier transform infrared spectroscopy (FT-IR) for the rapid quantification of total anthocyanins and total phenolics in red grapes was investigated and evaluated for prediction accuracy with independent validation sets. The design of calibration sets aimed at capturing most of the variation due to vintage (2004 and 2005), cultivar (Cabernet Sauvignon, Merlot, Pinotage, and Shiraz) and sugar concentration. Best prediction accuracies were obtained for calibration sets using grapes from a single vintage or cultivar or approximately the same sugar concentration. The highest prediction accuracies were obtained for total anthocyanins calibration sets of grapes with sugar concentrations ≥ 23.5ºBrix (SEP = 0.13 mg/g; R2 validation set = 0.77) and for total phenolics calibration sets of grapes with sugar concentrations < 23.5ºBrix (SEP = 0.13 OD280/g; R2 validation set = 0.74). Strong correlations were found between the spectral data and the total anthocyanins (SEP = 0.12 mg/g; R2 validation set = 0.84) and total phenolics concentration data (SEP = 0.10 OD280/g; R2 validation set = 0.76) for 2005 Merlot calibration sets indicating that the FT-IR spectra captured most of the variation. Overall the RPD (ratio of the standard deviation of the reference data to the standard error of prediction) values of all calibration models were below 3 indicating that calibration models are fit for screening purposes. Spectroscopic absorbance at 280 nm is not specific enough for the quantification of total phenolics and the use of an alternative reference method such as high performance liquid chromatography (HPLC) will be considered in the future. Principal component analysis (PCA) revealed that the major sources of variation in the FT-IR spectra of grapes could be ascribed to vintage and grape sugar concentration and this had an effect on the accuracy of the analytical data generated when using FT-IR spectroscopy. This report is the first to our knowledge where FT-IR has been used for the quantification of total anthocyanins and phenolics of grapes. The evaluation of the reference laboratory protocol for the quantification of total anthocyanins and total phenolics in grapes were evaluated in Chapter 4 and emphasized the importance of meticulous laboratory practices to obtain reliable reference data for calibration purposes. This large scale investigation of the total anthocyanins and phenolics concentrations in grapes is the first of its kind in South Africa and a quantitative database containing analytical data of the anthocyanins and total phenolics concentrations of 692 grape samples representing a wide range of grape maturities of Vitis vinifera cultivars Cabernet Sauvignon, Merlot, Pinotage and Shiraz from the 2004 and 2005 vintages was established based on the reference values. The data were used in a preliminary investigation of the implementation of total anthocyanins and total phenolics concentrations as part of grape quality classification at a commercial South African winery (Chapter 5). The results showed that the total anthocyanins and total phenolics concentration in grapes increased with increasing grape maturity (measured as sugar concentration). ANOVA and post-hoc analysis (Bonferroni testing) revealed significant differences between the total anthocyanins and total phenolics concentrations of the four Vitis vinifera cultivars investigated. Grapes harvested earlier in the season had significantly higher (p≤0.05) total anthocyanins and total phenolics concentrations than grapes harvested later in the season. This implies that grapes harvested earlier in the harvest season could produce wines with higher quality. Grapes from regularly irrigated vineyards had lower total anthocyanins and total phenolics concentrations compared to dryland vineyards. The current grape grading system in use at the industrial cellar did not correlate well with the total anthocyanins and total phenolics concentrations of grapes which highlighted the need for the inclusion of more objective measures during grape grading. The information captured in the database can be used as a starting point to establish profiles of the typical anthocyanins and total phenolics of South African grapes and data from more vintages should be included and continually updated. These findings highlight the important contribution of the results obtained in this preliminary study for the incorporation of total anthocyanins and phenolics concentrations as objective parameters of grape quality. Finally multivariate data analysis of the FT-IR spectra revealed important information regarding factors (both physical and chemical) that contribute to the variation of the spectra. The main variation between the 2004 and 2005 samples can probably be interpreted in terms of the water content of the samples.

Every year, the worldwide wine production generates about 20 million tons of waste, which usually end up being discarded in the environment. Previous data showed that it is possible to obtain flavan-3-ol compounds from different winery wastes with significant antioxidant activity. Thus, the aim of this work was to investigate the neuroprotective, hepatoprotective, genotoxic, mutagenic and cytotoxic activities of aqueous winery seeds extract from Vitis labrusca, cv. Bordo. The results showed that the extract minimizes the oxidative damage to lipids and proteins in the cerebral cortex, cerebellum, hippocampus and liver of rats. Also, the extract did not alter locomotor behavior, and it was non-genotoxic (comet assay) or nonmutagenic in lymphocytes from rats, and in Saccharomyces cerevisiae cells, respectively. The treatment with the extract was able to prevent depletion of mitochondrial eletron transfer chain complex I proteins in neuroblastoma cells (SH-SY5Y) treated with H2O2. At doses of 0.1 and 1mg/mL, the extract was found to be cytotoxic to liver (HepG2) and breast (MCF-7) cancer cell lines. This effect was accompanied with an increase in nitric oxide production, increase in the expression of p53, and increase in DNA damage index. A significant decrease in the expression levels of total PARP was observed in HepG2 cell line. In MCF-7 cells, V. labrusca extract treatment increased the expression levels of Bax and AIF proteins, and decreased the expression levels of total PARP. The expression levels of Her-2 protein were found to be decreased in HepG2 and MCF-7 cells after V. labrusca extract treatment. These results show significant biological effects of V. labrusca winery wastes extract, cv. Bordo.

Full textENGLISH ABSTRACT: Glutathione (GSH), a tripeptide consisting of glutamate, cysteine and glycine, is the most ubiquitous non‐protein intracellular thiol in a large variety of organisms, including plants, animals and fungi. The thiol moiety of the cysteine residue confers unique redox and nucleophilic properties. In plant cells, GSH fulfils an indispensible role in the antioxidant system, sulphur metabolism and detoxification of xenobiotics. Upon grape crushing, GSH is extracted into the juice where it exerts several protective effects during the vinification process. In must, it reacts with oxidized phenolic compounds to form the colourless grape reaction product (GRP) which limits must browning to a certain extent. During wine ageing, GSH impedes the decrease of important aroma compounds, including certain esters, terpenes and volatile thiols, while at the same time preventing the development of atypical ageing off‐flavours. GSH may also inhibit the yellowing of wine during ageing. It is thus evident that elevated GSH levels in wine, in particular white wine which is more sensitive to oxidation, may be highly valuable for wine quality. The reductive crushing and pressing of white grape varieties, which limits oxidation and the consequent incorporation of GSH into GRP, promotes higher GSH levels in the juice. The reductive handling of juice also limits the formation of oxidized glutathione (GSSG). However, during alcoholic fermentation and maturation, levels generally decrease as a result of assimilation by the yeast Saccharomyces cerevisiae, and inevitable oxidation that takes place during the vinification process. The principal focus of this study was to gain a better understanding of the fate of glutathione during alcoholic fermentation and to establish whether certain oenological applications could result in elevated wine GSH levels. The application studied, included choice of yeast strain, extended lees contact, nitrogen supplementation and supplementation with glutathione enriched inactive dry yeast preparations (GHS‐IDYs). In addition, the need for a rapid analytical method for the simultaneous quantification of both GSH and GSSG in must and wine which does not involve derivatization or require extensive sample preparation, led to the development of a novel UPLC‐MS/MS method. The method was also employed to determine intracellular GSH and GSSG contents of the yeast S. cerevisiae and was studied for the first time in winemaking conditions. It was shown that the GSH levels fluctuated during alcoholic fermentation, suggesting the uptake and release by yeast. At the end of alcoholic fermentation, levels were generally lower than those initially present in grape juice, but in some cases, concentration increases were also observed. This finding indicates that, in some cases, endogenously‐produced GSH may be secreted into must during alcoholic fermentation, contributing to higher GSH levels in wine. Albeit small, significant differences in GSH content could be seen in wines fermented with different yeast strains, implying that yeast strain may to a certain extent influence wine GSH levels. While the effects of lees ageing and nitrogen supplementation seem to be insignificant in contributing to higher GSH levels in wine, the supplementation of must with GSH‐IDYs could result in increased wine GSH levels, provided the supplementation is done early during fermentation. This study has broadened our knowledge of several oenological factors, influencing GSH levels in wine and provided a new baseline for future research studies.
AFRIKAANSE OPSOMMING: Glutatioon (GSH), ’n tripeptied bestaande uit glutamaat, sisteïen en glisien, is die mees algemene nieproteïenagtige intrasellulêre tiool in ‘n wye verskeidenheid organismes, insluitende plante, diere en fungi. Die tioolfunksiegedeelte van die sisteïenresidu verleen unieke redoks‐ en nukleofiliese eienskappe. GSH vervul ‘n onmisbare rol in die antioksidantsisteem, swaelmetabolisme en die ontgiftiging van xenobiotika in plantselle. Tydens die maal van druiwe word glutatioon in die sap geëkstraeer waar dit verskeie beskermende effekte tydens die wynbereidingsproses uitoefen. GSH reageer met geöksideerde fenoliese verbindings om die kleurlose druifreaksieproduk (DRP) te vorm wat die verbruining van mos in ‘n sekere mate beperk. GSH verminder ook die afname van belangrike aromaverbindings tydens wynveroudering, insluitende sekere esters, terpene en vlugtige tiole, terwyl dit terselfdertyd die vorming van atipiese verouderingswangeure belemmer. So ook kan GSH die vergeling van wyn tydens veroudering inhibeer. Dit is dus voor die hand liggend that verhoogde GSH‐vlakke in wyn, in die besonder witwyn, wat meer oksidasie‐sensitief is, van waarde kan wees vir wynkwaliteit. Die reduktiewe maal en pers van witdruifvariëteite wat oksidasie en gevolglike inkorporasie van GSH in DRP beperk, bevorder hoër GSH‐vlakke in sap. So ook beperk die reduktiewe behandeling van sap die vorming van geoksideerde glutatioon (GSSG). Gedurende alkoholiese gisting en veroudering neem GSH‐vlakke egter af as gevolg van assimilasie deur die gis, Saccaromyces cerevisiae, asook onvermydelike oksidasie wat gedurende die wynbereidingsproses plaasvind. Die hooffokus van die studie was om ‘n beter begrip van die lot van glutatioon tydens alkoholiese gisting te verkry en om vas te stel of sekere wynkundige praktyke verhoogde GSH‐vlakke in wyn tot gevolg kan hê. Die studie het gisraskeuse, verlengde gismoerkontak, stikstofaanvulling en aanvulling met glutatioon‐verrykte, onaktiewe droëgis ingesluit. Daarbenewens het die behoefte aan ‘n vinnige analitiese metode vir die gelyktydige kwantifisering van sowel GSH as GSSG in mos en wyn wat nie derivatisering of uitgebreide monstervoorbereiding vereis nie, gelei tot die ontwikkeling van ‘n nuwe UPLC‐MS/MS metode. Hierdie metode is ook gebruik om die intrasellulêre GSH‐ en GSSG‐inhoud van die gis S. cerevisiae te bepaal wat vir die eerste keer in wynbereiding bestudeer is. Daar is bewys dat GSH‐vlakke tydens alkolholiese gisting fluktueer, wat dui op die opname en vrystelling daarvan deur die gis. Die vlakke aan die einde van alkoholiese gisting was oor die algemeen laer as vlakke aanvanklik teenwoordig in die sap. In sommige gevalle is konsentrasietoenames egter ook waargeneem. Hierdie bevinding dui daarop dat intrasellulêr‐vervaardige GSH, in sommige gevalle, in die mos uitgeskei kan word, wat tot hoër GSH‐vlakke in wyn lei. Klein, dog beduidende verskille in GSHinhoud is waargeneem in wyne wat met verskillende gisrasse berei is, wat daarop dui dat gisras in ‘n sekere mate die GSH‐vlakke in wyn kan beïnvloed. Alhoewel die effek van gismoerveroudering en stikstofaanvulling onbeduidend is, kan die aanvulling van mos met glutatioon‐verrykte, onaktiewe droëgis tot verhoogde GSH‐vlakke in wyn lei, mits die aanvulling vroeg tydens alkoholiese gisting gedoen word. Hierdie studie verbreed ons kennis van verskeie wynkundige praktyke wat GSH‐vlakke in wyn beïnvloed en vorm ‘n nuwe basis vir toekomstige navorsingstudies.

Full textENGLISH ABSTRACT: Wine production involves complex interactions between a variety of yeasts and bacteria. Conventional microbiological methods can be used to identify the different microorganisms present in wine, but prove to be time-consuming and certain microbial species may not grow on synthetic isolation media. The aim of this study was to evaluate the microbial population present in two South African red wines, Pinotage and Merlot, as well as five spoilt commercial South African wines by using a non-culturable approach, polymerase chain reaction (PCR)-based denaturing gradient gel electrophoresis (DGGE). The results from the non-culturable approach were compared to conventional platings. Unique PCR-based DGGE fingerprints were obtained for the Bacteria and yeasts present in the South African Pinotage and Merlot wines. Using yeast specific primers the Pinotage wine showed the presence of non-Saccharomyces yeasts at the beginning of the alcoholic fermentation, while Saccharomyces cerevisiae was present until the completion of the malo-lactic fermentation (MLF). This yeast was also identified during both the alcoholic fermentation and MLF of the Merlot wine using PCR-based DGGE and conventional plating. Using Bacteria specific primers, Lactobacillus plantarum and Lactobacillus sp. was identified in the Pinotage wine using PCR-based DGGE, while Lactobacillus brevis were isolated from Merlot wine using conventional platings. Although the presence of S. cerevisiae is expected during wine fermentation, the presence of this microbe in bottled wine could lead to spoilage. Four of the spoilt commercial wine samples (RW1, RW2, RoW1 and WW1) were found to be spoilt by S. cerevisiae, while a fifth wine sample (RW3) was found to be spoilt by an Acetobacter sp. using PCR-based DGGE. Members of the family Enterobacteriaceae were identified from all the wines using PCR-based DGGE, while Enterobacter sakazakii was identified from RW1 using PCR-based DGGE and conventional plating. The members of the family Enterobacteriaceae could possibly have contributed to the spoilage of the wine by producing undesirable secondary metabolites. PCR-based DGGE proved to be an alternative to conventional microbiological methods for the identification of the microbial species in South African red grape must and wine. This method also proved to be useful in the identification of spoilage microbes in spoilt commercial South African wines.
AFRIKAANSE OPSOMMING: Die produksie van rooi wyn behels komplekse interaksies tussen ‘n verskeidenheid van giste en bakterieë. Konvensionele mikrobiologiese metodes kan gebruik word om die verskillende mikro-organismes wat in rooi wyn teenwoordig is te identifiseer, maar dit blyk tydrowend te wees, terwyl sekere mikro-organismes nie groei op sintetiese media nie. Die doel van hierdie studie was om die mikrobiologiese populasie wat in twee Suid-Afrikaanse rooi wyne, Pinotage en Merlot, en vyf bederfde kommersiële wyne teenwoordig is, te evalueer met die gebruik van ‘n kultuur-onafhanklike benadering, polimerase ketting-reaksie (PKR)-gebaseerde denaturerende gradiënt jel elektroforese (DGJE). Die resultaat van die kultuur-onhafhanklike benadering was vergelyk met konvensionele uitplating tegnieke. Unieke, ongeëwenaarde PKR-gebaseerde DGGE vingerafdrukke was verkry van die Bakterieë en giste aanwesig in die Pinotage en Merlot wyne. Deur gebruik te maak van gis-spesifieke inleiers het die Pinotage wyn die teenwoordigheid van nie-Saccharomyces giste getoon, terwyl Saccharomyces cerevisiae teenwoordig was tot en met die afhandeling van die appel-melksuur gisting (AMG). Hierdie gis is ook geïsoleer gedurende beide die alkoholiese gisting en AMG van die Merlot wyn deur gebruik te maak van PKR-gebaseerde DGGE en konvensionele uitplating tegnieke. Met Bakterieë-spesifieke inleiers, was Lactobacillus plantarum en Lactobacillus sp. geïdentifiseer in die Pinotage wyn deur gebruik te maak van PKR-gebaseerde DGGE, terwyl Lactobacillus brevis geïsoleer is uit Merlot wyn deur gebruik te maak van konvensionele uitplatings. Alhoewel die teenwoordigheid van S. cerevisiae verwag word gedurende wynfermentasie, kan die teenwoordigheid van hierdie mikrobe in gebottelde wyn tot bederwing lei. Vier van die bedorwe kommersiële wynmonsters (RW1, RW2, RoW1 en WW1) was bederf deur S. cerevisiae, terwyl ‘n vyfde wynmonster (RW3) bederf was deur ‘n Acetobacter sp. deur die gebruik van PKR-gebaseerde DGGE. Hierdie metode het ook die bruikbaarheid in die identifikasie van mikrobes wat kommersiële Suid-Afrikaanse wyne bederf, bewys.

ENGLISH ABSTRACT: Grape and wine phenols consist of flavanols which is the building blocks for tannins. These building blocks are called monomers which consist of catechins, epicatechins, epigallocatechins and epicatechin-gallate. Tannin is important in wine as it contributes to bitterness, mouth feel (astringency) and maturation potential of the wine. Futhermore it has a health benefit as an antioxidant. Anthocyanins are responsible for the colour of red wine. The anthocyanins combine with tannins to form stable polymeric pigments. Due to the importance of tannins and anthocyanins in wine, it is imperitative that different winemaking techniques are used to extract as much of these components as possible and that the analysis is done quickly and accurately. The aim of this study was to evaluate different winemaking techniques and their extraction of tannins and anthocyanins into the wine. Too much tannin extraction can have a negative effect on the sensory quality of the wine. Therefore a second aim was to evaluate the mouth feel properties of a Shiraz wine. A third aim was to compare the two tannin precipitation methods in terms of time efficiency, repeatability and the ease of practice. To investigate the amount of tannin concentration extracted by different winemaking techniques, two cultivars (Cabernet Sauvignon and Shiraz) were used. These treatments included the addition of an enzyme during fermentation [E], cold maceration [CM], post maceration [PM] and the combination of cold and post maceration [CM+PM]. The grapes were harvested in two different climatic areas during the 2008 and 2009 vintages. The two climatic areas were classified according to the Winkler scale as a III (Morgenster) and a IV (Plaisir de Merle). The grapes were harvested at two different ripeness levels in order to evaluate the effect of the different winemaking processes on the extraction of tannins and anthocyanins. One harvest was before (LB) and the other after (HB) the commercial harvest. The results of this study showed significant differences in the phenolic composition of the wines. It was found that the warmer area showed higher tannin concentrations than the cooler area for both cultivars. In the 2008 Cabernet Sauvignon the CM extracted higher concentrations of tannin from the cooler area at both ripeness levels. In the warmer area, CM extracted the highest tannin concentration HB, but the CM+PM extracted the highest tannin concentration from Cabernet Sauvignon at the LB and CM at the HB of the warmer area. In 2009 the PM extracted the highest concentration of tannin at the lower ripeness level, while the E treatment extracted the highest concentration from the warmer area. In the cooler area the CM+PM extracted the highest concentration of tannin at a lower ripeness level, while there were no siginicant differences between the different treatments at the higher ripeness level. The highest anthocyanin concentration was found in the cooler area. The CM treatment was found to have no effect on anthocyanin extraction. Different methods are available to quantify the tannin concentration in wine. Two of the most popular tannin analytical methods are the bovine serum albumin (BSA) and the methyl cellulose precipitable tannin (MCP) methods. The BSA method is a very complex method which uses at least 3 times more reagents than the MCP method. The MCP method only analyzes tannins, while the BSA method analyzes tannins, monomeric pigments (MP), small polymeric pigments (SPP) and large polymeric pigments (LPP). In this study a good correlation was found between the two tannin precipitation methods (R2 – 0.88). There is controversy regarding the variability of these methods. Some scientists found that the two methods show a good correlation with HPLC, while others found that there was no such correlation between the precipitation methods and the HPLC. The MCP method had a practical advantage as it could be performed in half the time required for the BSA method. This has a significant impact in scenarios where a high sample throughput is required although it only measures total tannin. The phenolic composition and mouth feel of the wine was strongly influenced by the climatic area. In the warmer area the effect of tannin concentration on mouth feel was much less than in the cooler area. The wine made of riper grapes, was more grippy, bitter and numbing than the wines made from greener grapes. The E treatment was especially associated with a dry, grippy sensation.
AFRIKAANSE OPSOMMING: Druif en wyn fenole bestaan uit flavanole wat weer die boublokke is van tanniene. Hierdie boublokke, wat bekend staan as monomere, betsaan uit katesjiene, epikatesjiene, epigallokatesjiene an epikatesjien-gallaat. Tanniene is belangrik in wyn aangesien dit bydra tot bitterheid, mondgevoel (vrankheid) asook die verouderingspotensiaal van wyn. As antioksidante hou dit ook gesondheidsvoordele in. Antosianiene dra by tot die kleur van rooiwyn. Antosianiene kombineer met tanniene om meer stabiele polimeriese pigmente te vorm. As gevolg van die belangrikheid van tanniene en antosianiene is dit van uiterse belang dat verskillende wynmaak tegnieke gebruik word om ekstraksie in die wyn te bevoordeel en dat die analitiese metode so vinnig en akkuraat as moontlik gedoen word. Die eerste doel van hierdie studie was om die ekstraksie van tanniene en antosianiene deur middel van verskillende wynmaak tegnieke te evalueer. Te veel tanniene in die wyn kan negatiewe sensoriese kwaliteit tot gevolg het. Daarom is die tweede doel om die sensoriese kwaliteit van Shiraz wyn te evalueer. Die derde doel van hierdie studie was die twee tannien presipitasie metodes met mekaar te vergelyk in terme van die moeilikheidsgraad van die metode, tyd doeltreffendheid en herhaalbaarheid. Verskillende wynmaak tegnieke (ensiem byvoegings [E], koue maserasie [CM], verlengde dopkontak [PM] en ‘n kombinasie van koue maserasie en verlengde dopkontak [CM+PM]) is vergelyk ten opsigte van tannien en antiosianien ekstraksie. In 2008 en 2009 is twee kultivars (Cabernet Sauvignon en Shiraz) in twee verskillende klimatologiese areas gepars. Hierdie areas is geklassifiseer in die Winklerskaal as ‘n IV (Plaisir de Merle) en ‘n III (Morgenster). Om die effek van die verskillende wynmaak tegnieke op die ekstraksie van antosianiene en tanniene te vergelyk, is hierdie twee kultivars by twee verskillende rypheidsgrade geoes. Full textThis thesis acts as a series of 'snapshots' into the meaning of 'wine tourism'. Each chapter of my main body of work looks at a different segment of wine tourism in the Western Cape: a fast growing industry that inherits attributes from both the wine and tourism industries. Themes of landscape and the tourist experience track through these separate snapshots, linking them together. A passion for wine and the drinking of wine would seem to have been an enjoyable pastime passed down from epochs of wine lovers and producers that stood before us in the 'winescapes' of time. While this conception of the wine drinking tradition may be presented to us today, it should be remembered that this might not have been the case in times gone by. Looking back to South Africa and the wine industry in the 1950s where 'wine consciousness' was a real concern for the marketers and makers of wine, we find no such traditions in place. Obstacles to the integration of wine into everyday living came in the form of an avid temperance movement concerned with drunkenness and alcoholism. Over time these obstacles heeded to the power of the wine industry so that increasing emphasis was placed on the role of publicity and marketing of wine. It was perhaps a natural development that wine tourism came to hold particular potential and interest for South African wine producers. The history of wine tourism of the Western Cape is inherently connected to the establishment of our first wine route in Stellenbosch. With a concern for the superiority of the European wine making tradition and landscape, it was only in 1970 that we saw a change in interest to the wine regions and heritage landscapes of our own country. The SteHenbosch wine route was a concept inspired by European example but grounded in local landscape. The significance of the mapping out of this landscape of space into place was a real concern for the wine makers of the regions whose freedom to market and export their wine overseas was severely restricted by legal prohibitions established by the KWV in the 1960s. With the defining of distinct wine regions, came the emphasis of difference of place within the winelands of the Western Cape. Each region has a formula for difference based on some combination of breathtaking scenery, quality wines, first class cuisine, and with increasing frequency the heritage of European roots. The construction of place and landscape identities gives us a sense of the perspective of the marketer and promoter of the wine region. I found it important to explore how this construction of identity of place came to be experienced by and presented to tourists in the present day.

Full textENGLISH ABSTRACT: Chenin blanc wine is of economic importance to South Africa and a range of diverse dry and semi-dry wines are locally produced in this genre. Currently, the use of three distinctly different style names, each aimed at providing consumers with information about the flavour of the wines, is encouraged by the South African (SA) wine industry. The styles are fresh and fruity (FF), rich and ripe unwooded (RRUW) and rich and ripe wooded (RRW). Feedback from retail sectors over the past few years, however, repeatedly suggested that the style names are perceived as confusing by SA consumers. This master study was undertaken to re-evaluate the FF, RRUW and RRW style classification, based on both the volatile fermentation-derived aroma composition and the sensory attributes of a set of wines containing all the styles under investigation. For the purposes of chemical profiling, a set of 105 commercial Chenin blanc wines, selected to be representative of these three styles and originating from the major SA wine producing areas, were analysed by Gas Chromatography (GC) to quantify fermentation-derived volatile aroma compounds in the wines. ANOVA performed on the chemical data showed that 29 compounds represent significant differences between at least two of the 3 styles (FF, RRUW and RRW). Principal component analysis (PCA) of the volatile compounds showed a large degree differentiation between FF and RRW wine styles, however, RRUW wine styles overlapped with the other two styles. Considering vintage effects, ANOVA indicated no significant differences within FF (vintages 2009 and 2010) and RRW (vintages 2008 and 2009) styles, whereas only 2 esters and 4 terpenes showed significant differences between the three wine producing regions investigated for this purpose, Paarl/Wellington, Breede River and Stellenbosch. Volatile aroma compounds generated for Chenin blanc were included in the Winetech database consisting of the most important cultivars of South Africa. Combining the data for the volatiles for Chardonnay and Sauvignon blanc from this database and the data for Chenin blanc obtained in this study, a PCA indicated a clear separation between Chenin blanc and the other two white cultivars. Sensory evaluation of the style classification was done by two separate sensory tests. Firstly, a sorting task was performed by wine industry experts to categorise 21 Chenin blanc wines (FF, RRUW and RRW) based on their similarity. The results showed a differentiation between FF and RRW styles, however, RRUW was mostly classified together with FF wines. This indicated a possible continuum between the three styles, as opposed to three distinct different categories, currently suggested by the style names. The second sensory analysis test, Descriptive Sensory Analysis (DSA), was performed by a trained panel to generate sensory profiles for 42 wines. ANOVA of the flavour attribute intensities between different styles once again showed significant differences between FF and RRW, with RRUW wines forming a continuum between the FF and RRW styles. These results provide valuable information that could be used by the wine industry for labelling purposes.

Eerstens is 21 wyne (met al drie style verteenwoordig) deur wynindustrie eksperts gesorteer op grond van die waargenome eendersheid van die onderskeie wyne en die resultate is grafies geprojekteer. Die resultate het getoon dat daar ‘n duidelike verskil waargeneem is deur die assessors tussen die vars en vrugtige styl en ryk en ryp gehoute styl. Die ryk en ryp ongehoute wyne het in die analises meer met die vars en vrugtige style geassosieer, as die ryk en ryp gehoute wyne. Die tweede sensoriese metode is uitgevoer deur sensoriese paneel wat vir die doel van hierdie studie opgelei is om die geur eienskappe van 42 wyne (al drie style verteenwoordig) te profileer. Analise van statistiese beduidende verskille tussen die voorkoms van die geurkomponente en hul intensiteite vir elke styl, het weereens aangedui dat daar ‘n kontinuum bestaan tussen die style. Hierdie resultate kan van waarde vir die wynindustrie wees in besluite rakende etikettering.

Full textENGLISH ABSTRACT: Key research areas in modern enology are related to the production of wine of consistent quality, as well as to the improvement of existing wine quality through the enhancement of the sensory properties of wine. The formation of glycerol during alcoholic fermentation is highly relevant to both these issues. Since the early years of the zo" century, glycerol has been positively associated with the quality of wine and it is thought to impart important mouth-feel sensations such as "viscosity", "smoothness" and "body". In general, it is considered that glycerol concentrations higher than those normally found in wine, can contribute towards the improvement of wine quality. It has also been suggested that increased concentrations of glycerol can enhance the aroma of wine. On the basis of these perceptions, several strategies have been developed to favour the production of glycerol during the fermentation process and over a period of years, a large volume of data has been collected that relates to various aspects regarding glycerol production during alcoholic fermentation. To date, however, several aspects regarding the relationship between glycerol and wine quality remain unclear. The reasons for this situation can mainly be ascribed to the lack of reliable analytical data to serve as a basis for investigating the relationship between glycerol and wine quality, as well as the preponderance of empirical and anecdotal evidence. Despite numerous opinions regarding optimal glycerol concentrations in wine, glycerol is indeed seldom assayed on a routine basis and targets with respect to specific wine grape cultivars and glycerol concentrations have largely remained unspecified. To date, very little information regarding glycerol concentrations in South African wines has been published. The analytical techniques that are most frequently used for the quantification of glycerol in grape juice, fermenting must and wine are not easily automated and this aspect placed severe limitations on the generation of large volumes of analytical data on glycerol concentrations in these matrices. This project was undertaken with the aim to holistically address some of the unresolved issues relating to the relationship between glycerol and wine quality. This also implied the development and optimisation of analytical techniques suitable for the rapid and accurate determination of glycerol in fermentation media, as well as in finished wine. In the first stage of this project a quantitative database was established that contained the analytical data on the glycerol concentrations of a statistically significant number of wines of adjudged quality, as well as additional information for each wine regarding the geographic origin, vintage, routine chemical analyses and the yeast strain(s) used for the production of the wine. The relevance of glycerol in wine for the modern South African winemaker was evaluated through the establishment of a quantitative database that contained the opinions of an expert panel of 15 South African winemakers, enologists and wine chemists on topics relating to glycerol in wine. In the second stage of the project the data captured in the databases were used to investigate aspects regarding the relationship between glycerol and wine quality. From the data captured in the qualitative database, it was clear that the topic regarding glycerol in wine was important to the South African winemakers and it was also evident that there was a need for the development and optimisation of methods suitable for the routine analysis of the glycerol concentrations in grape juice, fermenting must and wine. The opinions of the panel members also highlighted the issue that the mouth-feel property of wine was considered to be an area where the quality of some wines could be further improved. The quantitative database contained the information on the glycerol concentrations of 450 commercial South African table wines of adjudged quality. The premium cultivars Chardonnay, Chenin blanc, Sauvignon blanc, Cabernet Sauvignon, Merlot, Shiraz and Pinotage were used for the purpose of investigating the relationship between glycerol concentration and wine quality. The wines represented a wide variety of wine styles, including dry white, off-dry white, dry red and late harvest wines. The average glycerol concentration was significantly associated with the wine style. In white wines the average glycerol concentration was much lower than in the red wines (6.82 gIL versus 10.49 gIL, respectively). No significant relationship between the final glycerol concentration and the geographic origin, vintage and the yeast strain used for the fermentation was found. Wine quality could not be significantly associated with glycerol concentration in the red wines. In the white wines, the relationship between glycerol concentration and wine quality was significant, but due to the very small differences in the average glycerol concentrations of the wines of different quality ratings, the statistical significance is probably of little practical value. The effect of glycerol on the volatility of a selection of esters and higher alcohols was also investigated. Solid-phase microextraction, followed by gas chromatography, was used to analyse the composition of the headspace at equilibrium between the liquid phase and the gas phase of a model wine, and a dry white wine that contained a basal concentration of 5.4 gIL glycerol. Results showed that incremental increases in the glycerol concentrations over a range of 1 -10 gIL in the model wine and in a Chenin blanc wine, were not accompanied by a proportional increase or decrease in the abundance of the aroma components in the headspace. The volatile components tested were isoamyl acetate, ethyl butyrate, ethyl valerate, ethyl lactate, ethyl hexanoate, hexyl acetate, isoamyl alcohol and isobutanol. For all the volatile components tested, the difference between the headspace composition of samples containing the lowest glycerol concentration, and those containing the highest glycerol concentration, was not significant. However, sufficient experimental evidence was obtained to indicate that increasing glycerol concentrations had an effect on the volatility of aroma components, and that the effect is of a complex and non-linear nature. In the third phase of the work Fourier transform infrared spectroscopy (FT-IR) was used to establish and optimise methods for the accurate and rapid quantification of glycerol in wine. For this purpose calibrations were developed for the quantification of glycerol in dry wine and late harvest wines. The accuracy of prediction was evaluated by means of the standard error of prediction that was 0.38 gIL for the dry wines and 0.65 gIL for the sweet wines. Large variations are introduced in the FT-IR spectra of wine by factors such as process technology, cultivar and geographic origin, and this variation can have an effect on the accuracy of the analytical data generated when employing FT-IR spectroscopy. Using glycerol prediction in wine as a model system, principal component analysis of the FT-IR spectra was done in order to establish quality control measures for the detection of poorly predicted, or outlier samples. A classification model, based on principal component analysis, was established that enabled the interpretation and classification of the outlier samples in the data set in 100% of the cases tested. This work forms the basis for expanding the quality control measures for the detection of wines of which the FT-IR spectra are highly unnatural, as well as for establishing quality control measures to ensure that accurate analytical data are generated when FT-IR is used. FT-IR spectroscopy was also used to develop a rapid screen for the evaluation of the fermentation profiles of wine yeasts. For this purpose, a selection of wine yeasts, which included commercial wine yeasts frequently used in winemaking in South Africa, as well as a selection of hybrid Saccharomyces cerevisiae yeasts, that were obtained through a selective breeding strategy aimed at increasing glycerol concentrations were used. Calibrations necessary for the accurate quantification of glycerol, volatile acidity, ethanol, reducing sugar and glucose, in Chenin blanc must and a synthetic must were developed and optimised. This work forms the basis upon which the scope of the analysis, both in terms of the number of components that can be measured, as well as the medium in which the yeasts are being evaluated, can be enlarged. This would be valuable for future applications in both the research as well as the industrial environment. The method that was developed serves to illustrate how this application can play a supportive role in yeast development programmes, through the speeding up of the initial stages of yeast strain evaluation.
AFRIKAANSE OPSOMMING: Navorsing in moderne wynkunde is sterk gefokus op die produksie van wyn waarvan die kwaliteit van In volhoubare goeie peil is, sowel as die verdere verbetering van bestaande wynkwaliteit, deur In verhoging van die sensoriese eienskappe van wyn. Die vorming van gliserol tydens alkoholiese fermentasie het betrekking op beide hierdie aspekte. Die invloed van gliserol op die vlugtigheid van "n seleksie van hoëralkohole en esters is ondersoek in "n model wyn sowel as "n wit wyn, waarvan die basiese gliserolkonsentrasie 5.4 gIL was. Soliede-fase mikroekstraksie van die gasfase van wyn is opgevolg met "n gaschromatografiese analise. Nietemin het die resultate getoon dat gliserol wel "n effek het op die vlugtigheid van die aroma komponente wat in hierdie ondersoek gebruik is, maar dat die aard van die effek kompleks en nie-liniê is. Fourier-transformasie-infrarooispektroskopie (FT-IR) is gebruik om die metodes vir die analise van gliserol in wyn sodanig te optimiser, dat vinnige en akkurate bepalings op "n roetine basis in wyn gedoen kan word. Kalibrasies is ontwikkel vir die kwantifisering van gliserol in droëwyn en laatoeswyn. Die standaard voorspellingsfout van die kalibrasies wat ontwikkel is, was 0.38 gIL in droëwyn en 0.65 gIL in die geval van laatoeswyn. Hierdie werk vorm die basis waarop kwaliteitsbeheer verder uitgebrei kan word sodat wyne met onnatuurlike spektra gëidentifiseer kan word, sowel as om te verseker dat akkurate data gegenereer word as FT-IR as analitiese metode gebruik word vir die kwantifisering van gliserol in wyn. FT-IR spektroskopie is ook gebruik om "n vinnige skandeerproses te ontwikkel waarmee die fermentasieprofiele van wyngiste geëvalueer kan word. Die seleksie giste wat vir hierdie doel gebruik is, sluit kommersiële Suid-Afrikaanse wyngiste in, sowel as hibriede Saccharomyces cerevisia-giste wat normaalweg hoër vlakke van gliserol tydens die fermentasieproses produseer. Kalibrasies is ontwikkel vir die akkurate kwantifisering van gliserol, vlugtige suur, alkohol, reduserende suiker en glukose, in Chenin blanc-mos sowel as in 'n sintetiese medium. Hierdie werk vorm die basis waarop verdere uitbreidings gedoen kan word, sodat meer metaboliete gemeet kan word en 'n groter verskeidenheid van fermentasiemedia gebruik kan word. Hierdie ontwikkeling is waardevol vir toekomstige toepassings in die navorsings- sowel as die industriële omgewing. Die metode wat ontwikkel is illustreer ook hoe hierdie toepassing 'n ondersteunende rol kan speel in wyngisontwikkelingsprogramme deur die aanvanklike evalueringsproses van die giste te versnel.
