Full texthowever, the control of the cell organization and cell behavior to create fully functional 3-D constructs has not yet been achieved. To overcome these, activities have been concentrated on the development of multi-functional tissue engineering scaffolds capable of delivering the required bioactive agents to initiate and control cellular activities. The aim of this study was to prepare tissue engineered constructs composed of polymeric scaffolds seeded with mesenchymal stem cells (MSCs) carrying a nanoparticulate growth factor delivery system that would sequentially deliver the growth factors in order to mimic the natural bone healing process. To achieve this, BMP-2 and BMP-7, the osteogenic growth factors, were encapsulated in different polymeric nanocapsules (poly(lactic acid-co-glycolic acid) (PLGA) and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV)) with different properties (degradation rates, crystallinity) and, therefore, different release rates to achieve the early release of BMP-2 followed by the release of BMP-7, as it is in nature. Initially, these nanoparticulate delivery systems were characterized and then the effect of single, simultaneous and sequential delivery of BMP-2 and BMP-7 from these delivery systems was studied in vitro using rat bone marrow MSCs. The effect of using these two growth factors in a sequential manner by mimicking their natural bioavailability timing was shown with maximized osteogenic activity results. BMP-2 loaded PLGA nanocapsules were subcutaneously implanted into Wistar rats and according to initial results, their biocompatibility as well as the positive effect of BMP-2 release on the formation of osteoclast-like cells was shown. To complete the construction of the bioactive scaffold, this nanoparticulate sequential delivery system was incorporated into two different types of polymeric systems

-caprolactone) (PCL)). 3-D fibrous scaffolds were produced using these materials by wet spinning and 3-D plotting. Incorporation of nanocapsules into 3-D chitosan scaffolds was studied by two different methods: incorporation within and onto chitosan fibers. Incorporation into 3-D PCL scaffolds was achieved by coating the nanocapsules onto the fibers of the scaffolds in an alginate layer. With both scaffold systems, incorporation of nanocapsule populations capable of delivering BMP-2 and BMP-7 in single, simultaneous and sequential fashion was achieved. As with free nanocapsules, the positive effect of sequential delivery on the osteogenic differentiation of MSCs was shown with both scaffold systems, creating multi-functional scaffolds capable of inducing bone healing.

Full textBono, the lead singer of U2, has built a reputation as a champion of political issues, such as publicly taking a pacifist stance against the Troubles in Northern Ireland, and as a prominent social activist, especially known for working with Amnesty International. However, Bono’s lobbying to save the continent of Africa from its economic downward spiral bridged social justice with politics. Bono headed the American branch of the Drop the Debt/Jubilee 2000 campaign, co-founded his own non-profit group DATA, and is responsible for the success of the ONE campaign in the United States. As a celebrity, Bono uses his connections to influence his fanbase using U2 lyrics and performances. He also politicks with government officials and impresses them with his expert knowledge, tuning his argument to suit his audience. These tactics, combined with ceaseless passion and a grassroots movement, have made Bono the most effective social and political activist of the 21st century. Bono inspires others to take up a crusade and defend it using all of their power. If the rest of humanity were to follow his example, social justice would be a realized necessity in this new millennium

Full textOver the past few years’ competition between Swedish and international companies have increased. This puts requirements on how Swedish companies must act to survive. To handle these external changes, continuous work with internal efficiency is a solution for competitive advantage. The studied company Bong in Kristianstad has recently had problems concerning measurements of their internal efficiency, more precisely their production efficiency. A company that does not measure their production efficiency will have a hard time prioritizing the right areas, processes, activities, and solutions for improvement work. The purpose of this study is to identify relevant parameters with the intention of calculating and visualizing production efficiency within a manufacturing industry. The goal of the study is to develop a model for calculation and visualization of production efficiency that can be used as a basis for improvement work. The goal is also to be able to use this model in the purpose of identifying different areas that can be improved and to create suggestions for improvement for these areas. Knowledge was obtained by doing a qualitative case study concerning the company's work methods and their own interpretation of production efficiency. Parameters were identified, a model was created, calculations were done, and the results were analyzed to be able to visualize and determine the company's actual production efficiency based on the OEE/TAK-method. The analysis and conclusion showed that the studied company’s method for calculating their production efficiency was inadequate and non-representative. It was also proved why the current method was inadequate. Areas for improvement were identified where setup times constituted the biggest loss for the company's production efficiency. Three suggestions for improvement work were generated and implemented theoretically to prove what positive economic and psychosocial effects they have on the business.

This thesis discusses the financial, commercial, political and legal risks of international trade operations involving International Bank Guarantees. It starts by defining the most usual international trade instruments and, then, defining Standby Letters of Credit, Performance Bonds and Bid Bonds issued according to the ISP 98, URGD 758 or UCP 600 as International Bank Guarantees. Later, it explores each specific risk. The financial risks are divided into foreign exchange risk, applicant's default risk and bank reputation risk. It is considered as a commercial risks frauds and low quality products. Legal risks are legal battles and local legislation. There is a topic that discusses specifically the U.S. related risks, as the country has a very particular legislation when it comes to International Bank Guarantees. The political risks are related to commercial embargoes and sanctions, changes in foreign policy and to the deterioration of commercial reations. The following section discusses how to mitigate each of the risks discussed. It shows whether or not this risk can be mitigated and if it also affects internation trade operations that do not use International Bank Guarantees. Lastly, the thesis discusses about the ideal international trade instrument. At the moment, International Bank Guarantees are the best instrument, still not the perfect one as it has some limitations.
Esta tese debate os riscos financeiros, comerciais, políticos e legais relacionados a operações de comércio internacional. A tese começa definindo os principais instrumentos usados em transações de comércio internacional. Em seguida, define como Garantia Bancária Internacional as Standby Letters of Credit, Performance Bonds e os Bid Bonds, emitidos de acordo com a ISP 98, URGD 758 ou UCP 600. Depois, a tese explora os riscos específicos das Garantias Bancárias Internacionais. São considerados riscos financeiros o risco cambial, o risco de calote da parte que solicitou a garantia e o risco de reputação dos bancos envolvidos na transação. Riscos comerciais são as fraudes e produtos de má qualidade. Os riscos legais analisados são referentes a batalhas judiciais e legislação local. Além disso, há uma parte da tese dedicada a discutir especificamente os riscos relacionados a legislação americana, que é muito diferente dos outros países em relação a Garantias Bancárias Internacionais. São considerados riscos políticos embargos e sanções comerciais, mudanças na política externa e a deterioração das relações comerciais entre dois países. A última seção da tese discute como mitigar os riscos involvidos em transações com Garantias Bancárias Internacionais. Nesta parte, discute-se quais riscos podem ou não ser mitigados eq quais riscos são inerentes a transações comerciais internacionais, independentemente do uso de Garantias Bancárias Internacionais. Conclui-se que, no momento, Garantias Bancárias Internacionais ainda são o melhor instrumento para assegurar transações comerciais internacionais. Contudo, estes intrumentos ainda não são perfeitos e estão sujeitos a muitos riscos e limitações.

Full textThere are several possibilities for those with an eating disorder, who wishes to get treatment and recover to get help and support. But for those who are not ready to recover, there are not any help to receive. Pro-ana is a youth culture where individuals who want to achieve the anorectic ideal help and support each other. The purpose of this study was to describe and analyse the content and the need for the Pro-ana community on Internet, among young people with eating disorders. More specifically the aim was to find out how young Swedish people uses the Pro-ana communities on Internet. The study was an ethnography based on concealed observations of 6 different Pro-ana websites on Internet. During the observations five themes has been used: anonymity, thinspiration, life choice or disease, help or no help and identity. The analysis was based on Goffmans (2004) The presentation of self in everyday life, and the authors thesis about how humans represents them self in the accompany of others. Pro-ana websites consists of diary notes about weight, food, exercise and how to cope with an eating disorder. The Pro-ana culture online has become kind of a support group to which young people turn when society has no knowledge and no resources to meet them and social work are very limited on these sites. When not dare to tell friends and family about their situation, they turn to Pro-ana, which has been described as a sanctuary away from society’s judgements.

The goal on this dissertation performs both a description and an analysis on human and nonhuman relation based on Floresta's countryside, a town located in Pernambuco State. I have described this relation from some different point of view, yet their particular way of breeding (criação na solta) has been the main focus on my analysis. The intensive field research gave rise to the sketching of interspecies relations in that area. It had occurred in the period of three months when took place conversations and interviews with local husbandman who breeds both in the countryside (no mato) and the streets (na rua). In particular, the issue draws upon a description about how five families from the riparian zone (Cachoeira, Pocinhos, Quebra-Unha, Capim e Riacho do Meio) deal with and understand their relations with goats. At first, my analysis looks forward to the contrast between laboro as a husbandman daily activity and both the concept of work and and the extensive production method. The laboro is a set of very specific procedure and skill – a part of what is called criação na solta – and they are a condition of possibility for what I intend to describe and understand. Signs belong to the set of skills: they are a knife cutting made at the goat's ear which concurrently symbolize and identify the animal's owner and its family. The laboro and the signs are ways of realization and accomplishment of family affiliation. The laboro relies upon the animal's intense acquaintance with the caatinga which brings together a particular husbandry expertise; just a few men knows the terrain and how to breed goats. The concept of domestication can be drawn from the nexus relating breeder, livestock and caatinga in the manner of their mutual relation. Hence, from the standpoint of breeders and their families I describe how this practical way of breeding give rise to an understanding on interspecies relation in a given region. Those who knows the veredas, the goat-tracks traced as daily over the years, knows as well the ground pattern and footprint which deliver a way of being which is proper to husbandman, caatinga and the goat.
O propósito desta dissertação é descrever e analisar relações tramadas entre humanos e não humanos na zona rural de Floresta, município localizado no sertão de Pernambuco. Essas relações são caracterizadas por mim a partir de distintas perspectivas, mas admitem como eixo condutor da análise a prática da criação na solta. A pesquisa de campo intensiva, com duração de três meses, somada aos diálogos e entrevistas com moradores da região, criadores do mato e da rua, permitiram que relações interespecíficas daquele local pudessem ser delineadas. Mais especificamente, trata-se de descrever como famílias residentes em cinco ribeiras (Cachoeira, Pocinhos, Quebra-Unha, Capim e Riacho do Meio) lidam e compreendem suas relações com cabras e bodes. A princípio, por meio de uma análise contrastiva demonstro como o laboro, atividade diária dos criadores, pode ser pensada em contraposição à noção de trabalho e a um modo de produção extensivo. O laboro é um conjunto de técnicas e procedimentos muito específicos que fazem parte desse modo de criação na solta, ambos condição de possibilidade para a compreensão das relações que pretendo descrever. Parte dessas técnicas são os sinais, recortes feitos a faca nas orelhas da criação, que, ao mesmo tempo, simbolizam e identificam o proprietário do animal e a família a que pertence. O laboro e os sinais são meios de efetivação e visualização de relações de parentesco. O laboro, que pressupõe um convívio intenso entre animais e caatinga, permite que os criadores produzam e conservem um conhecimento particular, uma expertise. Apenas alguns conhecem o mato e sabem criar cabras e bodes. É por meio do nexo constituído entre criadores, criação e caatingas, dos seus afetos e das afecções de seus corpos que foi possível pensar a ambivalência da noção de domesticação. Portanto, partindo do ponto de vista dos criadores e de suas famílias, descrevo como um modo de criação e suas práticas, possibilitam compreender as relações interespecíficas em uma determinada região. Aqueles que conhecem as veredas, traçadas diariamente e ao longo de anos por cabras e bodes, conhecem as impressões e marcas na terra de um modo de existência de sertanejos, caatinga e bodes.
