Abstract:
Abstract Introduction: The effect of trastuzumab in targeted therapy for HER2-positive gastric cancer (GC) is limited by the eventual development of resistance within patients. Although numerous studies have been published on this topic, both the mechanism of innate and acquired trastuzumab resistance are still not fully understood. In this study, we compare the mutational landscape of paired solid tumor samples before and after treatment in an effort to identify acquired events associated with resistance. Methods: We performed whole-exome sequencing on 46 tumor and 23 whole blood samples from patients with stage IV gastric cancer were obtained from before treatment (n = 23) and at disease progression (PD) (n = 23). HER2 status was checked using immunohistochemistry and validated with florescence in situ hybridization (FISH). Correlation analysis was then conducted between the mutational profile and clinical data of patients. Results: Comparisons between innate resistance samples and acquired resistance samples at baseline revealed three copy number variation events (CDK12 amplification, ERBB2 amplification, MECOM amplification) that occurs in significantly different proportions (p < 0.05). In addition, for baseline samples, MECOM was shown to be associated with poor progression-free survival (PFS) (Hazard Ratio (HR), 12; 95% CI, 1.6 - 85; p = 0.027). Commonly acquired events (i.e., lacking in before treatment samples and present at PD) within patients include TP53 mutation, MYC amplification, MCL1 amplification, CDKN2A/2B deletion, and CCNE1 amplification. Of these, patients with MCL1 gene amplification had worse survival (HR, 4.8; 95% CI, 1.2 - 20; p = 0.050) than patients with wild-type MCL1. In addition, the NOTCH pathway acquired mutations in 4 of 23 patients and was found to be correlated with a worse PFS (HR, 5.1; 95% CI, 1.4 - 19; p = 0.023), indicating that this pathway is possibly involved with trastuzumab resistance to some degree. Conclusion: Overall, our results show that comparisons of samples obtained before treatment and at PD can give insight about innate and acquired resistance mechanisms to trastuzumab in stage IV, HER2-positive GC patients. Abstract:
We carefully discuss the finiteness of SUSY YM N=4 in the light cone gauge, first at the one loop level by directly exhibiting the relevant terms of the lowest order Green functions and then at any loop order by using a recent treatment of the renormalization of general Yang-Mills theories in the light cone gauge. We point out the existence of a set of divergent Green functions which however do not contribute to observable quantities, thereby recovering consistency with formulations in other gauges.

Abstract:
Han Shu is our first history book which narrates the story by dynasty. It carries a lot of valuable information of the Han dynasty, both historically and literarily. However, because of the long-ago history, a lot of language, pronunciation, naming system, names of officials and places are now difficult to read and understand. Throughout the history, a number of people had analyzed Han Shu. These people have solved a lot of queries. Yang Shuda has high achievements in analyzing Han Shu. He is not only good at etymology and phonology but also analyzing parts of speech and sentence patterns. In terms of the use of materials, not only does he compare different versions of literature, but also uses inscriptions and even cultural data such as marriage and death for analysis. All these contribute to his unique analysis. The analysis of Han Shu has opened up a new field as more and more materials are excavated. Bronze, lacquer and even lute and currency are used in research. This has broadened the academic horizons and solved many mysteries. In 1983, in Hubei province, a lot of Han bamboo slips were excavated, which have provided tremendous help to our understanding of the medicine, law, mathematics and others of the Han dynasty. By the time when current books of Han analysis such as Yang Shuda’s Han Shu Kui Guan were punished, bamboo clips of Zhangjiashan were not yet unearthed. It is unable to invoke this important information, resulting in some doubtful content. I hereby try to make use of these bamboo slips to supplement previous comments, provide new research directions, or affirm previous results. This dissertation is divided to six parts. Each uses the notes of current analysts as the starting point while incorporating Zhangjiashan Han bamboo slips. My analysis is based on Yang Shuda Han Shu Kui Guan, together with the analysis of some current analysts.

Full text
Abstract:

Abstract:
AbstractThe conservative versus the liberal conflict currently seems to be prevalent in Western societies. This chapter tests the association between conservative and liberal worldviews and basic human values: conservation values (CONS) versus values we term “liberal attitude values” (LA). We explore their association to each other, to religiosity, and to social policy attitudes. We first explore these associations in the full Young Adults and Religion in a Global Perspective (YARG) sample, and then focus on three samples from countries that have recently experienced a liberal-conservative political polarization: Poland, Israel (Jewish sample), and the United States. The contribution of the YARG data is in the ability to present a more nuanced view of this dimension, with an emphasis on its relation to religiosity. Our intention is to unpack the conservative and the liberal worldviews in order to explore the elements that each of them promotes and to identify variance within national contexts, shifting the liberal-conservative dichotomy to a culturally-nuanced spectrum.

Abstract:

Abstract:

Abstract:
An extensive experimental study on sprays from an injector for gasoline direct injection (GDI) engines has been performed. Spatial and temporal evolution measurements of a large cone-angle jet, emerging from a high pressure swirled injector, have been carried out in an optically accessible vessel. The spray has been lightened, both along the spray axis and in cross sections perpendicular to it, by a 532 nm Nd-YAG pulsed laser sheet, 80 mm thickness and 12 ns duration. The scattered light has been collected at 90° with respect to the sheet direction by a digital CCD camera with a frame grabber synchronized with the injection command and the laser pulse. A digital delay system has provided a fine temporal shift (up to microseconds range) of the images acquisition with respect to the start of the injection (SOI). Finally, a digital image processing system has provided analysing the images collected by the CCD camera. The emerging spray has been acquired with three spatial scales, providing both the global and local spray behaviour, and with a detailed temporal resolution to characterize the early stage of the jet formation. The initial phase of the spray is characterized by a strong axial component of the velocity with respect to the radial one, resulting in a pre-spray or slug phase. It produces a cylindrical shape of the jet with the characteristic mushroom shape. Large droplets with high momentum are produced, travelling downstream in advance to the main spray. At later time the radial velocity component controls the process and it gives up to the classical hollow-cone shape with a strong interaction with the gas in the vessel. The images give evidence in time of the collapse of the hollow-cone structure hence resulting in a full cone spray. This behaviour is confirmed by the cross section measurements through the spray, carried out in the range 10–40 mm from the nozzle tip. These measurements make evidence of the refilling of the cone with the presence of strong vortexes on the boundary of the jet. The effects of the fuel injection pressure, injection duration and air-flow field interaction on the structure and evolution of the spray have been studied in details. The digital image processing system also has allowed to reconstruct the spray profile and to determine a refilling index.

Abstract:
Abstract A large share of passenger vehicles are currently running on gasoline, which is considered the major contributor to carbon emissions from the transportation sector. In gasoline direct injection (GDI), the fuel is introduced directly into the combustion chamber in a spark ignition (SI) engine. GDI has the advantage of operating at a higher compression ratio due to the charge cooling effect, resulting in better fuel utilization and lower pollutant emissions. The spray formed from fuel injection under different engine running conditions governs the charge (air-fuel) characteristics. The present work focuses on experiments using a GDI system with a multi-hole injector for gasoline fuel sprays in a constant volume chamber. The saturation pressure of the fuel present within the nozzle increases with rising temperature (80 to 90°C) from heat transfer due to the placement of the injector directly into the combustion chamber. At part-load running conditions, when the heated fuel injected into an environment has a pressure sufficiently lower (achieved in early injection) than its saturation pressure, it attains the state of superheating and undergoes flash boiling. The abrupt vaporization of injected fuel results from flash boiling and affects spray characteristics. The ratio of chamber pressure to saturation pressure (Pch/Psat) indicates the propensity of flashing for a given super-heated operating condition. Macroscopic analysis of GDI sprays is reported in this study under flashing and non-flashing conditions through different experimental techniques to provide a comprehensive understanding. Experimental studies reported in this kind of literature typically adopt a specific technique to elucidate the details of spray evolution. A Diffused Back Illumination (DBI) imaging technique with a low-speed Nd: YAG laser has been carried out to analyse the liquid phase of the spray, such as penetration, cone angle, and width. Additionally, a novel Structured Laser Illumination Planar Imaging (SLIPI) technique has been adopted to identify the relatively dense spray regions by addressing the difficulty due to multiple scattering of light illuminating from surrounding scatterer media (droplets). SLIPI is used to suppress the effect of the surrounding light artefacts, helping explore the structure in a new dimension. A multi-plume spray is expected to have voids, dense, and dilute portions at different locations, affecting the fuel mixture, and interestingly, it appears after subduing the effects of multi-scattering from a modulated laser sheet using SLIPI. An LED-based spray illumination was also used to compare the DBI and SLIPI results to better understand the utility and effectiveness of different imaging techniques in a densely populated and highly transient spray phenomenon.
