Abstract:

Abstract:

Park, Hyun Jung, Sung Won Lee, Luc Van Kaer та Seokmann Hong. "CD1d-Dependent iNKT Cells Control DSS-Induced Colitis in a Mouse Model of IFNγ-Mediated Hyperinflammation by Increasing IL22-Secreting ILC3 Cells". International Journal of Molecular Sciences 22, № 3 (2021): 1250. http://dx.doi.org/10.3390/ijms22031250.

Full text
Abstract:
We have previously shown that CD1d-restricted iNKT cells suppress dysregulated IFNγ expression and intestinal inflammation in Yeti mice on the C57BL/6 background. Since type 3 innate lymphoid cells (ILC3s) in mesenteric lymph nodes (MLN) protect against intestinal inflammation in a CD1d-associated manner, we investigated whether crosstalk between iNKT cells and MLN ILC3s controls IFNγ-mediated intestinal inflammation in Yeti mice. We found that Yeti mice display increased levels of ILC3s and that iNKT cell deficiency in Yeti/CD1d KO mice decreases levels of IL22-producing ILC3s during DSS-induced colitis. This finding indicates that iNKT cells and ILC3s cooperate to regulate intestinal inflammation in Yeti mice. Yeti iNKT cells displayed a pronounced anti-inflammatory (IL4- or IL9-producing) phenotype during colitis. Their adoptive transfer to iNKT cell-deficient animals induced a significant increase in IL22 production by ILC3s, indicating that crosstalk between iNKT cells and ILC3s plays a critical role in modulating colitis in Yeti mice. Moreover, we showed that the IL9-producing subset of iNKT cells potently enhances IL22-producing ILC3s in vivo. Taken together, our results identify a central role of the iNKT cell-ILC3 axis in ameliorating IFNγ-mediated intestinal inflammation.

Abstract:
We have previously shown that a deficiency of CD1d-restricted invariant natural killer T (iNKT) cells exacerbates dextran sulfate sodium (DSS)-induced colitis in Yeti mice that exhibit IFNγ-mediated hyper-inflammation. Although iNKT cell-deficiency resulted in reduced Foxp3 expression by mesenteric lymph node (MLN) CD4+ T cells in DSS-treated Yeti mice, the cellular mechanisms that regulate Foxp3 expression by CD25+CD4+ T cells during intestinal inflammation remain unclear. We found that Foxp3−CD25+CD4+ T cells expressing Th1 and Th17 phenotypic hallmarks preferentially expanded in the MLNs of DSS-treated Yeti/CD1d knockout (KO) mice. Moreover, adoptive transfer of Yeti iNKT cells into iNKT cell-deficient Jα18 KO mice effectively suppressed the expansion of MLN Foxp3−CD25+CD4+ T cells during DSS-induced colitis. Interestingly, MLN dendritic cells (DCs) purified from DSS-treated Yeti/CD1d KO mice promoted the differentiation of naive CD4+ T cells into Foxp3−CD25+CD4+ T cells rather than regulatory T (Treg) cells, indicating that MLN DCs might mediate Foxp3+CD25+CD4+ T cell expansion in iNKT cell-sufficient Yeti mice. Furthermore, we showed that Foxp3−CD25+CD4+ T cells were pathogenic in DSS-treated Yeti/CD1d KO mice. Our result suggests that pro-inflammatory DCs and CD1d-restricted iNKT cells play opposing roles in Foxp3 expression by MLN CD25+CD4+ T cells during IFNγ-mediated intestinal inflammation, with potential therapeutic implications.

Abstract:
Abstract. This paper outlines the development and operation of Yeti, a bottom-up traffic emission inventory framework written in the Python 3 scripting language. A generalized representation of traffic activity and emission data affords a high degree of scalability and flexibility in the use and execution of Yeti, while accommodating a wide range of details on topological, traffic, and meteorological data. The resulting traffic emission data are calculated at a road-level resolution on an hourly basis. Yeti is initially applied to traffic activity and fleet composition data provided by the senate administration for the city of Berlin, which serves as the region of interest, where the Yeti-calculated emissions are highly consistent with officially reported annual aggregate levels, broken down according to different exhaust and non-exhaust emission modes. Diurnal emission profiles on select road segments show not only the dependence on traffic activities but also on road type and meteorology. These road-level emissions are further classified on the basis of vehicle categories and Euro emission classes, and the results obtained confirmed the observations of the city of Berlin and subsequent rectifications.

Lemon, Kiersty. "The Infectious Monster: Borders and Contagion in Yeti and Lágrimas en la lluvia." BYU ScholarsArchive, 2015. https://scholarsarchive.byu.edu/etd/5734.

Full text
Abstract:
Monsters are disruptive characters, who cross boundaries and blend categories. They come in various kinds: Non-human monsters, such as Dracula, created-by-human monsters like Frankenstein, human monsters like Hitler, and more-than-human monsters such as the X-men. These monsters can either be dangerous or helpful to humanity. Dangerous monsters appear as infectious, viral forces, while helpful monsters are inoculative forces for positive change. In either case, they penetrate the borders set up between normatively separate categories. Critics and authors have long realized the connection between heroes and monsters, often portraying them as necessary to one another, as two sides of a single coin. However, this analogy is lacking, because it does not allow for the possibility that a single character can display varying degrees of both heroism and monstrosity. Mario Yerro and Bruna Husky present such characteristics in Yeti and Lágrimas en la lluvia, as evidenced by their physical appearance, their relations to scapegoats, the porosity of species and other boundaries, and the decisions they make in regards to the Other.

Orton, Richard J. "Visualising and exploring linked functional genomic data sets in the Yeast Exploration Tool Integrator (YETI)." Thesis, University of Edinburgh, 2006. http://hdl.handle.net/1842/12741.

Full text
Abstract:
This PhD project is split into two parts: (1) The development of the Yeast Exploration Tool Integrator (YETI), a novel bioinformatics tool for the integrated visualisation and analysis of S. <i>cerevisiae </i>functional genomic data sets; and (2) Using YETI for data analysis purposes. Part 1: YETI consists of a MySQL database for the storage and management of data and a Java program for visualisation and analysis. YETI uses publicly available data from both online databases and published scientific studies to populate its own database and scripts have been written to automatically retrieve and process all relevant data to keep the database up-to-date. YETI consists of four inter linked sections: (1) A Genome Section for the informative display of the <i>S. cerevisiae </i>genome, its chromosomes, known and predicted genes and associated annotations; (2) A Transcriptome Section for the integration, visualisation and analysis of gene expression microarray data; (3) A Proteome Section for the effective visualisation of protein-protein interactions on a dynamic graphical panel; and (4) An Analysis Section providing a graphical interface to the database with a complex query search system. Part 2: A number of additional data analysis tools have been developed for use in conjunction with YETI. These tools have been used for the statistical analyses of individual data sets and for genome scale correlation analysis between data sets: 1) Genome vs Proteome: investigating if genes located near each other on the genome result in proteins that interact together in the cell. 2) Genome vs Transcriptome: investigating if genes located near each other on the genome are co-expressed under a number of different conditions. 3) Transcriptome vs Proteome: investigating if genes that are co-expressed under a number of different conditions result in proteins that interact together in the cell.

Abstract:
Imagine a theater. Build one in your mind. I do not care if it is the largest theater in the universe or if it is the size of a shoebox. The theater can be an elaborate construction with gilded cherubs and priceless jewels. If you would prefer the theater to be a minimal design, imagine it that way. Maybe the theater is made out of glass or crystal. You can be all alone in the theater or maybe you are seated next to an alien, a cave man, or a robot. As the curtain rises an enormous pipe organ is spewing out J.S. Bach’s Toccata and Fugue in D minor. The sound is colossal. The stage is designed to look like a filthy and bleak art studio. Sculptures that look like decaying flesh hang from the ceiling. Papers and dirty painting rags are scattered all over the floor. A roach crawls towards some moldy food in the wastebasket. A piece of beef jerky sits next to a pile of paint tubes and paintbrushes. Canvases are stacked against the wall. Dried paint is splattered everywhere. A woman enters stage left. As she begins to speak, the music fades… “My most recent paintings are meant to serve as gateways between daily life and an exotic utopian fantasy. These strange psychological spaces function as portals into a new frontier inhabited with the most stupendous creatures. Active volcanoes, carnivorous plants, terrible lizards, pink Chihuahuas, flesh-eating insects, unidentified flying objects, and a host of other amazing things coexist in this cruel tropical paradise. “Everything I do is a self-portrait and a disguise. I imagine the characters in my paintings to be mutant clones. The act of painting is like looking in the mirror. I like to spend most of my time doing one or the other. I am driven by an insatiable urge to see what I truly look like. I suffer from an unfulfilled desire to meet myself. I am frustrated with impressions and reflections. These confessions reveal my work as possibly the most disgusting display of narcissism in the entire history of art. “The act of painting is an absurd and self-indulgent enterprise. I paint with the ridiculous and frivolous purpose of delighting the eye. Nevertheless, my hope is that my paintings capture an important aspect of my utopian fantasy with their sumptuous surroundings embellished with rainbows, natural disasters, butterflies, exotic grasses and bizarre costumes. “My paintings can be read on many different levels from surface to deep allegory. The finished product is the result of hours upon hours of painting, pondering, and dreaming. Below the surface lies a nauseating abyss. “My work is a delicious feast for the aesthetic consumer. I am a villainous mad scientist with a hunger for immortality. My eyes are gluttonous fools. I am a mystery and a deception. Vermillion is my opulent fetish. Lemon yellow is my nemesis.” The music grows louder again. The woman begins to cackle. She then takes a deep breath and a deeper and more dramatic bow. She exits stage right--you find that you want to applaud--but unfortunately, this story is not over. I suggest that you get yourself a snack like some popcorn, chocolate, or gummy bears before reading any further. The curtain slowly and elegantly comes down.

Abstract:
Although there are no apparent structural links between these nine stories, they were conceived of as belonging to a particular mood or being variations of a particular theme. This theme, I suppose, is South African Disappointment. The scope of this abstract does not allow a full explain what I mean by this term, but Stephen Watson's immortal essay A Version of Melancholy expresses it beautifully; as does some of Damon Galgut's writing, Don Maclennan's poetry, Johannes Kerkorrel's music, and David Goldblatt's photography. A serviceable definition might be that feeling of anxious exhaustion one feels when dealing with a world that refuses to live up to one's best expectations of it. My aim with this collection is to give this feeling its first sustained South African fictional rendition and to show how ordinary people may endure it with their dignity still intact.

Alatas, Irem. "Fragmented Yet United: Alevis." Master's thesis, METU, 2011. http://etd.lib.metu.edu.tr/upload/12613545/index.pdf.

Full text
Abstract:
The present thesis aims to recount the current situation of the Alevi community in the urban Turkish setting. The data were collected during eight months of ethnographic field research from February 2009 to October 2009 in a complex preferred to be called as the Dikmen Alevi Community Center, in Ankara. I present different ways adopted by various groups while explaining what Alevism is and I stress the fact that there is a certain degree of fragmentalization within the community due to such diverse descriptions. Thenceforth, I emphasize the reasons behind the existence of divergent classifications and analyze Alevis&rsquo<br>migration from rural to urban areas during 1970s and 1980s as it relates to the changes in the institutions. After offering an evaluation of the changes accompanying migration, I accentuate the competitive sharing of the city as a religious space between Alevis and Sunnis concentrating on Alevis&rsquo<br>perception of religious space and providing a comparison between Sunni and Alevi perceptions regarding the places of worship. Subsequently, I attempt to show that there is a competitive sharing relationship present within the community giving the example of Dikmen Alevi Community Center after the establishment of the Alevi Institute for Research, Documentation and Application. I conclude that this kind of a relationship and the current state of affairs are the results of the struggle to adapt to a changing environment, which in turn alters the individuals themselves.

Background: Obesity is reaching epidemic proportions globally, with a threefold increase from 1975 to 2016. The percentage of obese adolescents aged 12-19 years increased from 5% to 21% over the same period. Social Cognitive Theory can determine human behavior in limiting food intake (diet) and regular physical activity. A valid and accurate measuring instrument for the factors influencing obesity is essential. This study aimed to determine the reliability of measuring instruments for diet, physical activity, social support, and Social Cognitive Theory on overweight and obesity in adolescent girls. Subjects and Method: This study used primary data. As many as 15 study subjects were selected by simple random sampling. The value of item-rest correlation and Cronbach’s alpha were examined using split half method run on Stata 14. Results: Dietary intake questionnaire haditem-rest correlation ≥0.20 with Cronbach alpha= 0.77. Physical activity questionnaire had item-rest correlation ≥0.20 with Cronbach alpha= 0.78. Social support questionnaire had item-rest correlation ≥0.20 with Cronbach alpha= 0.88. Social cognitive theory questionnaire had item-rest correlation ≥0.20 with Cronbach alpha= 0.89. Conclusion: Dietary intake, physical activity, and social cognitive questionnaires are reliable to be used. Keywords: reliability, item-rest correlation, Cronbach alpha Correspondence: Yeti Rahelli. Abstract:
The National Science Foundation operates stations on the ice sheets of Antarctica and Greenland to investigate Earth’s climate history, life in extreme environments, and the evolution of the cosmos. Understandably, logistics costs predominate budgets due to the remote locations and harsh environments involved. Currently, manual ground-penetrating radar (GPR) surveys must preceed vehicle travel across polar ice sheets to detect subsurface crevasses or other voids. This exposes the crew to the risks of undetected hazards. We have developed an autonomous rover, Yeti, specifically to conduct GPR surveys across polar ice sheets. It is a simple four-wheel-drive, battery-powered vehicle that executes autonomous surveys via GPS waypoint following. We describe here three recent Yeti deployments, two in Antarctica and one in Greenland. Our key objective was to demonstrate the operational value of a rover to locate subsurface hazards. Yeti operated reliably at −30 ◦C, and it has good oversnow mobility and adequate GPS accuracy for waypoint-following and hazard georeferencing. It has acquired data on hundreds of crevasse encounters to improve our understanding of heavily crevassed traverse routes and to develop automated crevasse-detection algorithms. Importantly, it helped to locate a previously undetected buried building at the South Pole. Yeti can improve safety by decoupling survey personnel from the consequences of undetected hazards. It also enables higher-quality systematic surveys to improve hazard-detection probabilities, increase assessment confidence, and build datasets to understand the evolution of these regions. Yeti has demonstrated that autonomous vehicles have great potential to improve the safety and efficiency of polar logistics.

Abstract:
The neoclassical market model still has a decisive influence on important economic policy decisions today. A central role in this model is played by the formation of equilibrium prices, where aggregate supply functions and aggregate demand functions meet. We examine whether equilibrium prices are actually formed. For this purpose, we analyse 2,217 prices for homogeneous products that were collected by students between October 2020 and May 2022 in stationary and online retail. In 143 of 146 cases, no equilibrium price is found. The percentage price range is regularly over 100%. The presumed steering function of an equilibrium price does not materialise. The establishment of market mechanisms for the efficient solution of economic problems must therefore be questioned.

Abstract:
The aim of the study: analysis of the current state and prospects for the development of augmented reality in Ukraine in business and education. The objectives of the study: to analyze the experience of using the augmented reality in advertising, marketing, education of Ukraine; to investigate the problems existing in this direction. The object of the study: the process of using augmented reality in advertising, marketing, education. The subject of the study: specific projects using the augmented reality in advertising, marketing, education. The used method of study was theoretical that included analysis of articles and materials of conferences on the research problem. The results of the study: nowadays, the augmented reality is used primarily in the field of advertising and marketing of Ukraine. As an example is the advertisement of Kyivstar (virtual tour around Ukraine, augmented reality quest), some of the Ukrainian companies have certain results in in this direction, for example, Augmented Pixels, Simo AR (in the development of a browser with augmented reality, the Kontramarka ticket service is implemented), Live Animations (such projects as Wonderland AR, My Yeti, Live Coloring, Gapchinska, Live Photo are already implemented). Among the problems that exist with the introduction of these technologies in education, first of all, we should note the shortage of specialists in the preparation of such educational projects and the uncoordinated actions of business and education in this direction. Main conclusions and recommendations: in order to disseminate research results it is necessary to hold thematic events of the all-Ukrainian level.
